Mg-air batteries are explored as the next-generation power systems for wearable and implantable electronics as they could work stably in neutral electrolytes and are also biocompatible. However, high corrosion rate and low utilization of Mg anode largely impair the performance of Mg-air battery with low discharge voltage, poor specific capacity and low energy density. Here, to the best of our knowledge, we first report a dual-layer gel electrolyte to simultaneously solve the above two problems by preventing the corrosion of Mg anode and the production of dense passive layer, respectively. The resulting Mg-air batteries produced an average specific capacity of 2190 mAh g-1 based on the total Mg anode (99.3 % utilization rate of Mg anode) and energy density of 2282 Wh kg-1 based on the total anode and air electrode, both of which are the highest among the reported Mg-air batteries. Besides, our Mg-air batteries could be made into a fiber shape, and they were flexible to work stably under various deformations such as bending and twisting.

Plasmopara viticola, the causal organism of grapevine downy mildew, secretes a vast array of effectors to manipulate host immunity. Previously, several cell death‐inducing PvRXLR effectors have been identified, but their functions and host targets are poorly understood. Here, we investigated the role of PvRXLR111, a cell death‐inducing RXLR effector, in manipulating plant immunity. When coexpressed with other PvRXLR effectors, PvRXLR111‐induced cell death was prevented. Transient expression of PvRXLR111 in Nicotiana benthamiana suppressed bacterial flagellin peptide flg22‐elicited immune responses and enhanced Phytophthora capsici infection. PvRXLR111 induction in Arabidopsis increased susceptibility to Hyaloperonospora arabidopsidis. PvRXLR111 expression in Pseudomonas syringae promoted bacterial colonization. By immunoprecipitation‐mass spectrometry analysis, yeast two‐hybrid, pull‐down, and bimolecular fluorescence complementation assays, it was shown that PvRXLR111 interacted with Vitis vinifera putative WRKY transcription factor 40 (VvWRKY40), which increased VvWRKY40 stability. Transient expression of VvWRKY40 in N. benthamiana inhibited flg22‐induced reactive oxygen species burst and enhanced P. capsici infection and silencing NbWRKY40 attenuated P. capsici colonization. These results suggest VvWRKY40 functions as a negative regulator in plant immunity and that PvRXLR111 suppresses host immunity by stabilizing VvWRKY40., PvRXLR111 from Plasmopara viticola suppresses PTI to promote pathogen virulence by stabilizing VvWRKY40, which functions as a negative regulator in plant immunity.

Taro (Colocasia esculenta (L.), Schott), from the Araceae family, is one of the oldest crops with important edible, medicinal, nutritional and economic value. Taro is a highly polymorphic species including diverse genotypes adapted to a broad range of environments, but the taro genome has rarely been investigated. Here, a high-quality chromosome-level genome of C. esculenta was assembled using data sequenced by Illumina, PacBio and Nanopore platforms. The assembled genome size was 2,405 Mb with a contig N50 of 400.0 kb and a scaffold N50 of 159.4 Mb. In total, 2,311 Mb (96.09%) of the contig sequences was anchored onto 14 chromosomes to form pseudomolecules, and 2,126 Mb (88.43%) was annotated as repetitive sequences. Of the 28,695 predicted protein-coding genes, 26,215 genes (91.4%) could be functionally annotated. On the basis of phylogenetic analysis using 769 genes, C. esculenta and Spirodela polyrhiza were placed on one branch of the tree that diverged approximately 73.23 million years ago. The synteny analyses showed that there have been two whole-genome duplication events in C. esculenta separated by a relatively short gap. According to comparative genome analysis, a larger number (1,189) of distinct gene families and long terminal repeats were enriched in C. esculenta. Our high-quality taro genome will provide valuable resources for further genetic, ecological and evolutionary analyses of taro or other species in the Araceae.

Modulation of the electronic structure of metal catalysts is an effective approach to optimize the electrocatalytic activity. Herein, we show a surprisingly strong activation effect of black phosphorus (BP) on platinum (Pt) catalysts to give greatly enhanced catalytic activity in the hydrogen evolution reaction (HER). The unique and negative binding energy between BP and Pt leads to spontaneous formation of Pt-P bonds producing strong synergistic ligand effects on the Pt nanoparticles. No Pt-P bonds are formed with red phosphorus which is another allotrope of P. By controlling the number of Pt-P bonds, 3.5-fold enhancement in the HER activity can be achieved from the BP-activated Pt catalyst and the activity is 6.1 times higher than that of the state-of-the-art commercial Pt/C catalyst. The BP-activated Pt catalyst exhibits a current density of 82.89 mA cm-2 with only 1 μg of Pt in 1 m KOH at an overpotential of 70 mV.

This study investigated whether and how fetal malnutrition would influence endogenous melatonin synthesis, and whether such effect of fetal malnutrition would transmit to the next generation. We enrolled 2466 participants and 1313 of their offspring. The urine 6-hydroxymelatonin sulfate and serum melatonin rhythm were measured. Methylation microarray detection and bioinformatics analysis were performed to identify hub methylated sites. Additionally, rat experiment was performed to elucidate mechanisms. The participants with fetal malnutrition had lower 6-hydroxymelatonin sulfate (16.59 ± 10.12 μg/24 hours vs 24.29 ± 11.99 μg/24 hours, P .001) and arear under curve of melatonin rhythm (67.11 ± 8.16 pg/mL vs 77.11 ± 8.04 pg/mL, P .001). We identified 961 differentially methylated sites, in which the hub methylated sites were locating on protein kinase C alpha (PRKCA) and cAMP response element-binding protein (CREB1) promoters, mediating the association of fetal malnutrition with impaired melatonin secretion. However, such effects were not observed in the offspring (all P .05). Impaired histomorphology of pineal, decreased melatonin in serum, pineal, and pinealocyte were also found in the in vivo and in vitro experiments (P .05 for the differences of the indicators). Hypermethylation of 10 CpG sites on the PRKCA promoter and 8 CpG sites on the CREB1 promoter were identified (all P .05), which down-regulated PRKCA and CREB1 expressions, leading to decreased expression of AANAT, and then resulting in the impaired melatonin synthesis. Collectively, fetal malnutrition can impair melatonin synthesis through hypermethylation of PRKCA and CREB1 promoters, and such effects cannot be transmitted to the next generation.

In this study, we aimed to explore the mechanism of glutathione peroxidase 3 (GPX3) in the growth of malignant melanoma (MM) cells by hypoxia-inducible factor-1α (HIF1-α) and HIF2-α regulating the metabolism through reactive oxygen species (ROS). The messenger RNA and protein expression of GPX3, HIF1-α, HIF2-α in tissues, and cell lines were measured by reverse transcription-quantitative PCR and Western blot analysis. A375 cells were transfected with GPX3 overexpression plasmid, small interfering RNA (siRNA) targeting GPX3, or siRNA targeting HIF1-α/HIF2-α to upregulate or downregulate the expression of GPX3 or HIF1-α/HIF2-α. The effects of H2 O2 and N-acetylcysteine (NAC) on the levels of HIF1-α and HIF2-α after overexpression of GPX3 were studied. The cell viability was detected by Cell Counting Kit-8. The levels of ROS, glucose uptake and lactic acid production, oxidative phosphorylation, and glycolysis of cells were measured for assessment of cellular metabolism. The expression of GPX3 decreased, while ROS, HIF1-α, and HIF2-α increased in MM tissues and cells. Overexpression of GPX3 inhibited the viability of MM cells and the growth of melanoma xenografts. The overexpression of GPX3 reduced the glucose uptake, extracellular lactic acid content, and extracellular acidification rate and increased the oxygen consumption rate level. Overexpression of GPX3 could reduce the levels of HIF1-α and HIF2-α, which could regulate metabolic levels. GPX3 reduced ROS level in MM to inhibit HIF1-α and HIF2-α. The addition of H2 O2 increased while NAC reduced the protein levels of HIF1-α and HIF2-α in the cells overexpressing GPX3. Our study demonstrates that GPX3 inhibits the growth of MM cells through its inhibitory effect on cell metabolic disorder by inhibiting HIF1-α via regulating ROS.

To develop wearable and implantable bioelectronics accommodating the dynamic and uneven biological tissues and reducing undesired immune responses, it is critical to adopt batteries with matched mechanical properties with tissues as power sources. However, batteries available cannot reach the softness of tissues due to high Young's moduli of components (e. g., metals, carbon materials, conductive polymers or composite materials). The fabrication of tissue-like soft batteries thus remains a challenge. Here we report the first ultrasoft batteries totally based on hydrogels. The ultrasoft batteries exhibited Young's moduli of 80 kPa, perfectly matching skin and organs (e. g., heart). The high specific capacities of 82 mAh·g-1 in all-hydrogel lithium-ion batteries and 370 mAh·g-1 in all-hydrogel zinc-ion batteries at a current density of 0.5 A·g-1 were achieved. Both high stability and biocompatibility of the all-hydrogel batteries has been demonstrated upon the applications of wearable and implantable. Aliphatic polyester, poly(3-hydroxyvalerate) (PHV), is commonly produced as a granular component in bacterial cells of various species. Based on 16S rDNA gene sequence analysis, strain PJC48 was identified as a Bacillus species. The current study is aimed to screen for a high-yield strain that can produce PHV efficiently and to increase PHV product yield by optimizing the fermentative process. We identified a high-producer strain based on Nile red staining. Characterization of the PHV produced by PJC48 by nuclear magnetic resonance spectroscopy revealed that it consisted of (R)-3-hydroxyvalerate monomers. The suggested model was validated by response surface methodology. Optimization of the PHV yield resulted in an increase of 32.75% compared to control, with a maximum production of 1.64 g/L after 48 H.

Zinc (Zn) metal is considered the promising anode for “post-lithium” energy storage due to its high volumetric capacity, low redox potential, abundant reserve, and low cost. However, extravagant Zn is required in present Zn batteries, featuring low Zn utilization rate and device-scale energy/power densities far below theoretical values. The limited reversibility of Zn metal is attributed to the spontaneous parasitic reactions of Zn with aqueous electrolytes, that is, corrosion with water, passive by-product formation, and dendrite growth. Here, a new ion-selective polymer glue coated on Zn anode is designed, isolating the Zn anode from the electrolyte by blocking water diffusion while allowing rapid Zn2+ ion migration and facilitating uniform electrodeposition.
