Soft tissue injuries (STIs) are the most common cause of extremity pain and motion dysfunction. Persistent inflammatory activation of immune cells characterized by senescence‐associated secretory phenotype (SASP) and mitochondrial stress are considered the primary causes of STIs, a pathological process also termed inflammaging. Meanwhile, scavenging excessive "cellular waste" in the inflammaging microenvironment and further activating tissue repair processes remain elusive. Herein, an anti‐inflammaging photothermal hydrogel microneedle patch for treating STIs is developed. Taurine‐loaded Prussian blue nanoparticles (Taurine@PB) are encapsulated in a methacrylate‐based hyaluronic acid hydrogel (HAMA) and further fabricated into taurine@PB@HAMA@microneedles (TPH@MN) patches. The acidic microenvironment of chronic inflammation and mild photothermal effects promote taurine release and anti‐inflammaging immunomodulation, inhibiting mitochondrial stress via the SIRT3‐NF‐κB axis to promote glycolytic metabolic microenvironment of neutrophils reprogramming toward oxidative phosphorylation metabolism. Furthermore, TPH@MN activates macrophage efferocytosis and initiates the process of tissue repair. In mouse models of chronic diabetic wounds and tibialis anterior (TA) muscle injury, TPH@MN inhibits SASP expression and promotes STIs healing through thermosensitized anti‐inflammaging immunomodulation. In summary, TPH@MN circumvents the side effects of systemic administration, providing new translatable options in the treatment modalities for patients suffering from STIs worldwide. Radical difunctionalization of aliphatic alkynes provides direct access to valuable multi‐substituted alkenes, but achieving a high level of chemo‐ and stereo‐control remains a formidable challenge. Herein a novel photoredox neutral alkyne di‐functionalization is reported through functional group migration followed by a radical‐polar crossover and energy transfer‐enabled stereoconvergent isomerization of alkenes. In this sequence, a hydroxyalkyl and an aryl group are incorporated concomitantly into an alkyne, leading to diversely functionalized E‐allyl alcohols. The scope of alkynes is noteworthy, and the reaction tolerates aliphatic alkynes containing hydrogen donating C─H bonds that are prone to intramolecular hydrogen atom transfer. The protocol features broad functional group compatibility, high product diversity, and exclusive chemo‐ and stereoselectivity, thus providing a practical strategy for the elusive radical di‐functionalization of unactivated alkynes. Seed dispersal by frugivorous birds facilitates plant invasions, but it is poorly known how invasive plants integrate into native communities in fragmented landscapes. We surveyed plant–frugivore interactions, including an invasive plant (Phytolacca americana), on 22 artificial land‐bridge islands (fragmented forests) in the Thousand Island Lake, China. Focusing on frugivory interactions that may lead to seed dispersal, we built ecological networks of studied islands both at the local island (community) and at landscape (metacommunity) levels. On islands with P. americana, we found that P. americana impacted local avian frugivory networks more on islands with species‐poor plant communities and on isolated islands. Moreover, as P. americana interacted mainly with local core birds (generalists), this indicates reduced seed dispersal of native plants on invaded islands. At the landscape level, P. americana had established strong interactions with generalist birds that largely maintain seed‐dispersal functions across islands, as revealed by their topologically central roles both in the regional plant–bird trophic network and in the spatial metanetwork. This indicates that generalist frugivorous birds may have facilitated the dispersal of P. americana across islands, making P. americana well integrated into the plant–frugivore mutualistic metacommunity. Taken together, our study demonstrates that the impact of plant invasion is context‐dependent and that generalist native frugivores with high dispersal potential may accelerate plant invasion in fragmented landscapes. These findings highlight the importance of taking the functional roles of animal mutualists and habitat fragmentation into account when managing plant invasions and their impact on native communities. The agricultural subsidy is an important policy instrument that aims to maintain food security and promote agricultural development in several countries. China introduced its first nationwide agricultural subsidy (i.e., the direct grain subsidy) for farmers in 2004. This study examines the impacts of direct grain subsidies on rural residents' food consumption patterns and nutritional intake on an individual basis. Based on a nationally representative sample of rural households from the National Rural Fixed Observation Points Survey data collected by the Ministry of Agriculture and Rural Affairs of China between 2003 and 2015, high‐dimensional fixed‐effects estimates show that the direct grain subsidy has boosted rural residents' grain consumption but reduced aquatic products, eggs, and dairy consumption in China. Specifically, an increase of 100 Chinese Yuan in per capita subsidies is associated with a rise of 14.94 g in daily grain consumption but a decrease of 0.57, 0.46, and 0.90 g in aquatic products, eggs, and dairy consumption, respectively. Furthermore, direct grain subsidies intensified rural residents' dietary imbalance and declined their dietary diversity. Observed dietary imbalance can be primarily attributed to the subsidy encouraging households' self‐sufficient grain consumption, leading to excessive cereal consumption and relatively lower consumption of meat, eggs, and milk. Our findings suggest that direct grain subsidies emphasizing the production of staple grains may inadvertently skew farmers' food consumption and nutritional intake, which may require consideration when evaluating the overall costs and benefits of agricultural subsidy policies. Aim: Mitochondria is one of the important organelles involved in cell energy metabolism and regulation and also play a key regulatory role in abnormal cell processes such as cell stress, cell damage, and cell canceration. Recent studies have shown that mitochondria can be transferred between cells in different ways and participate in the occurrence and development of many central nervous system diseases. We aim to review the mechanism of mitochondrial transfer in the progress of central nervous system diseases and the possibility of targeted therapy. Methods: The PubMed databank, the China National Knowledge Infrastructure databank, and Wanfang Data were searched to identify the experiments of intracellular mitochondrial transferrin central nervous system. The focus is on the donors, receptors, transfer pathways, and targeted drugs of mitochondrial transfer. Results: In the central nervous system, neurons, glial cells, immune cells, and tumor cells can transfer mitochondria to each other. Meanwhile, there are many types of mitochondrial transfer, including tunneling nanotubes, extracellular vesicles, receptor cell endocytosis, gap junction channels, and intercellular contact. A variety of stress signals, such as the release of damaged mitochondria, mitochondrial DNA, or other mitochondrial products and the elevation of reactive oxygen species, can trigger the transfer of mitochondria from donor cells to recipient cells. Concurrently, a variety of molecular pathways and related inhibitors can affect mitochondrial intercellular transfer. Conclusion: This study reviews the phenomenon of intercellular mitochondrial transfer in the central nervous system and summarizes the corresponding transfer pathways. Traumatic heterotopic ossification (THO) represents one of the most prominent contributors to post‐traumatic joint dysfunction, which currently lacks an effective and definitive preventative approach. Inflammatory activation due to immune dyshomeostasis during the early stages of trauma is believed to be critical in initiating the THO disease process. This study proposes a dual‐homeostatic modulation (DHM) strategy to synergistically prevent THO without compromising normal trauma repair by maintaining immune homeostasis and inducing stem cell homeostasis. A methacrylate‐hyaluronic acid‐based hydrogel spray device encapsulating a curcumin‐loaded zeolitic imidazolate framework‐8@ceric oxide (ZIF‐8@CeO2, CZC) nanoparticles (CZCH) is designed. Photo‐crosslinked CZCH is used to form hydrogel films fleetly in periosteal soft tissues to achieve sustained curcumin and CeO2 nanoparticles release in response to acidity and reactive oxygen species (ROS) in the inflammatory microenvironment. In vitro experiments and RNA‐seq results demonstrated that CZCH achieved dual‐homeostatic regulation of inflammatory macrophages and stem cells through immune repolarization and enhanced efferocytosis, maintaining immune cell homeostasis and normal differentiation. These findings of the DHM strategy are also validated by establishing THO mice and rat models. In conclusion, the CZCH hydrogel spray developed based on the DHM strategy enables synergistic THO prevention, providing a reference for a standard procedure of clinical operations. Oxford unicompartmental knee arthroplasty (UKA) has been particularly prevalent because the concept of knee preservation is deeply rooted in people's minds. Mobile bearing UKA is a surgical type of UKA with considerable advantages. This note describes some surgical techniques, including patient position, surgical field exposure, selection of the size of the prosthesis, sagittal tibial osteotomy, placement of the femoral prosthesis and gap balance, to assist surgeons with less experience in performing these operations successfully. The techniques described in this note have been used in over 500 Oxford UKA cases, and nearly 95% patients achieved good prosthesis position and satisfactory postoperative outcome. We hope that the empirical summaries from numerous cases will help surgeons to learn Oxford UKA quickly and effectively, driving the spread of the technique and benefiting more patients. This article discusses the challenges and potential solutions for transforming agri-food systems to improve nutrition and health in emerging economies. The triple burden of malnutrition, which includes hunger, hidden hunger, and overweight/obesity, is a significant issue in these countries due to shifts in dietary patterns towards Western and processed foods. The article presents a collection of papers that examine various aspects of this transformation, including the impact of agricultural and food policies, internet access, homestead gardening, and access to fast food restaurants. The efficiency of drug biosynthesis depends on different transcriptional regulatory pathways in Streptomyces, and the protein degradation system adds another layer of complexity to the regulatory processes. AtrA, a transcriptional regulator in the A‐factor regulatory cascade, stimulates the production of daptomycin by binding to the dptE promoter in Streptomyces roseosporus. Using pull‐down assays, bacterial two‐hybrid system and knockout verification, we demonstrated that AtrA is a substrate for ClpP protease. Furthermore, we showed that ClpX is necessary for AtrA recognition and subsequent degradation. Bioinformatics analysis, truncating mutation, and overexpression proved that the AAA motifs of AtrA were essential for initial recognition in the degradation process. Finally, overexpression of mutated atrA (AAA‐QQQ) in S. roseosporus increased the yield of daptomycin by 225% in shake flask and by 164% in the 15 L bioreactor. Fire is a common disturbance in many biomes, with both beneficial and detrimental effects on soil biology, which largely depend on fire intensity. However, little is known about the impact of fire on soil nematode communities in terrestrial ecosystem. In the present study, we investigated the effects of short‐term prescribed fire on soil nematode communities and soil properties in an old‐field grassland in Northern China. The results showed that burning significantly increased soil nematode abundance by 77% and genus richness by 49% compared to the control. Burning also decreased taxon dominance by 45% (Simpson's D) and increased nematode diversity by 31% (Shannon‐Weaver H'). However, burning increased plant parasites (particularly genera Cephalenchus and Pratylenchus) and shifted community to more bacterial‐feeding genera (i.e., decreased Channel Index). Generally, burning increased soil bio‐available nitrogen (NH4+–N and NO3−–N) content, which would be the main drivers causing nematode community to flourish via a "bottom‐up" effect. These results suggest that prescribed fire increases nematode diversity and alters community composition toward more plant parasites and bacterial feeders. Our findings highlight the importance of prescribed fire management in shaping short‐term nematode community structure and function, but the long‐term effects and impacts of these changes on soil nutrient and carbon cycling remain unknown. Pressure injury is a serious and preventable problem in intensive care units. Translating guidelines into clinical practice can reduce the incidence of pressure injury. Identifying clinical status, barriers and facilitators contribute to guideline implementation. To identify the knowledge, attitudes, and practices of pressure injury prevention in Chinese critical care nurses. Secondary data were extracted from a multicentric clinical trial. Knowledge and attitudes toward pressure injury prevention were assessed by a fourteen‐item questionnaire. The observed practices were recorded using a case report form. The report complies with the STROBE statement. A total of 950 critical care nurses in 15 hospitals from six provinces of China were investigated. A total of 53.1% of nurses received a median score of 6 points or less. Knowledge regarding the repositioning procedure, risk assessment, and heel pressure injury prevention was insufficient. Over 99% of nurses strongly or somewhat agreed that pressure injury prevention was very important and that they were willing to take measures to prevent pressure injury. A total of 27 781 patient days of pressure injury prevention practice were recorded. Repositioning was the most commonly used prevention measure, followed by support surfaces and prophylactic dressings. A combination of repositioning, support surface, and prophylactic dressing was lacking. Chinese critical nurses showed a low level of knowledge and a positive attitude toward pressure injury prevention. Practices of pressure injury prevention were unsatisfactory. There is a clear gap between the guidelines and clinical practices. The barrier (low‐level knowledge) and facilitator (positive attitude) were identified in this study. The present work investigated the effects of high‐pressure processing (200 and 400 MPa, 5 min) combined with chitosan‐tea polyphenol (1.5% and 0.5% [w/v], respectively) coating to improve the quality and stability of shrimp (Penaeus monodon) during 28 days of storage. The chemical (pH, TVB‐N, TBARS), microbiological, textural, chromatic characteristics, protein oxidation, and endogenous enzyme activities of shrimps were regularly evaluated. Results showed that the combination treatment exerted a better intense antimicrobial effect, stabilized shrimp's freshness, and resulted in lower pH and TVB‐N than the control sample. Also, combined treated samples had better oxidative stability than a single treatment until the end of shelf life. Although combination treatment had no significant effect on endogenous proteases, the combined use of 400 MPa high‐pressure and chitosan‐tea polyphenol coating was most effective in inhibiting the bacteria and improved the hardness and chromatic characteristics of shrimp within the storage. The thermoelectric properties of dually substituted Cu12−xInxSb4S12.8Se0.2 tetrahedrites are studied in temperature range from 300 to 723 K. The results indicate that dual substitution of In for Cu and Se for S not only causes the enhancement of thermopower S but also gives rise to a significant decrease in lattice thermal conductivity κL. An ultralow κL ≈0.2 W m−1 K−1 is obtained at 723 K in Cu11.95In0.05Sb4S12.8Se0.2 sample due to the enhanced phonon scattering mainly by point defects. Consequently, a high figure of merit ZTmax ≈1.0 is achieved in Cu11.95In0.05Sb4S12.8Se0.2 sample, which is ≈56% larger than that of pristine Cu12Sb4S13, indicating that dual substitution is effective in boosting the thermoelectric properties of tetrahedrites. Background: An isolated bone fragment from the posterolateral tibial plateau retrieved from the patellofemoral compartment is a rarely seen Schatzker type II tibial plateau fracture and is prone to misdiagnosis. To the best of our knowledge, this injury mechanism has not been previously described. Case presentation: A 63‐year‐old female sustained left knee pain and activity limitation after falling off an electric bicycle. Local hospital ignored the intra‐articular bone fragment and failed to provide effective treatment. This case described an uncommon Schatzker type II tibial plateau fracture with an isolated bone fragment, its physical examination and radiological findings, the potential injury mechanism, and surgical protocol. Conclusions: Combining the physical examination and radiological findings to evaluate the potential injury mechanism is important for developing an appropriate surgical protocol. Cresols are valuable phenolic chemicals which are mainly separated from coal tar. The separation of cresols using sustainable technology is of importance for comprehensive utilization of coal tar. In this work, for extracting o‐, m‐, and p‐cresol from the model coal tar, an ionic liquid 1‐ethyl‐3‐methylimidazolium dicyanamide ([Emim][DCA]) was prepared. The influence of ionic liquid dosage, extraction time, and temperature on separating o‐, m‐, and p‐cresol was explored. The extraction efficiency was determined from the experimental results to assess the extraction ability of [Emim][DCA]. With the optimal conditions, o‐, m‐, and p‐cresol were extracted efficiently from the model coal tar and the extraction efficiencies of o‐cresol, m‐cresol, and p‐cresol were up to 99.50%, 99.65%, 98.38%, respectively. In the meantime, the extraction mechanism was determined by calculation of σ‐profile, interaction energy, and electron density. The results showed that [Emim][DCA] and cresols formed the complexes by hydrogen bond. The formation of hydrogen bond was verified by the FTIR spectra, which is helpful for designing the related ionic liquids for recovering the phenolic compounds from coal tar. High nitrogen (N) fertilizer inputs accelerate soil acidification and degradation in tea plantations, thus posing a threat to soil microbial diversity, species composition, and ecosystem service functions. The effects of organic fertilizer and biochar applications on improving soil fertility have been extensively studied on cropland; however, little is known about their effectiveness in promoting soil multifunctionality on rapidly expanding acidic soils in tea plantations. In this study, we conducted a two‐year field experiment in a subtropical tea plantation to investigate the effects of organic fertilizer substitution and biochar amendment on soil microbial communities and multifunctionality. The results showed that soil multifunctionality was enhanced in plots amended with organic fertilizer and biochar. Soil multifunctionality was significantly and positively correlated with alpha‐diversity of bacteria but not fungi. We also found that organic fertilizer substitution and biochar amendment improved soil multifunctionality by altering the abundance of keystone species. The abundance of keystone species classified as module hubs in the bacterial co‐occurrence network contributed significantly and positively to soil multifunctionality. In contrast, the keystone species categorized as module hubs in the fungal co‐occurrence network negatively affected soil multifunctionality. Soil pH was a key driver of soil microbial community composition, indicating that the increase in soil pH under organic fertilizer and biochar amendment had a crucial role in biological processes. These results suggest that organic substitution and biochar amendment are beneficial in preventing soil degradation and maintaining soil multifunctionality in subtropical tea plantations. Although arboreal camera trapping is a growing field, it has rarely been used for monitoring plant‐frugivore interactions in the trees. Frugivore foraging behavior generally occurs in trees, hence arboreal camera trapping can be a potentially useful tool for frugivory research. We developed a camera trap sampling method to monitor plant‐frugivore interactions during mature fruiting periods. We used this method to monitor 318 individuals (camera sites) of 18 fleshy‐fruit plant species on 22 subtropical land‐bridge islands in the Thousand Island Lake, China. We recorded a total of at least 52 frugivorous animals, including a ground‐foraging bird species (Lophura nycthemera) and several mammals with foraging behaviors in the trees. We also recorded 4399 independent interaction events, including 275 unique plant‐bird interactions. We proposed a framework to classify interaction types and performed a sampling completeness test. We found that a sampling strategy that covered approximately a third of the fruit maturation period when most fruits were ripe was sufficient to sample plant‐frugivore interactions. Our results demonstrated that our sampling method with camera transects is reliable to monitor plant‐frugivore interactions in a fragmented landscape. This study helps to lay the methodological foundation for building networks of plant‐frugivore interactions with arboreal camera trapping on large spatial/temporal scales. As a non‐invasive, labor‐saving, and largely unbiased sampling method, the field application of arboreal camera trapping in different regions can advance the technology of biodiversity monitoring and lead to more accurate biodiversity inventories in arboreal environments. Tree mortality is an important ecological process influencing multiple functions of forest ecosystems. Previous studies have shown two basic size–mortality patterns, including a competition‐driven declining and a disturbance‐driven increasing mortality rate with tree size. Subtropical forests, which have a high species diversity and subject to frequent monsoon disturbances, are widely distributed in eastern Asia. However, the tree size–mortality pattern in the mature subtropical forests remains unclear.Here we analysed patterns of size‐dependent mortality from tree species to forest community using a 5‐year inventory data from 117 species and 163,612 individuals in a 20‐ha forest dynamic plot in a mature subtropical monsoon evergreen forest in eastern China. To explain the spatial variability in mortality patterns, two major biotic drivers (competition and tree size) and multiple local‐scale environmental factors were further analysed.Our results showed that tree size was the best predictor of tree mortality at the scales of both species and community. A species‐level analysis identified four size–mortality patterns that are shaped by species‐specific attributes such as maximum size and life form. For 27 out of 92 species that comprised 59% of tree individuals, the relationship between size and mortality exhibited a U‐shaped pattern of a first decline followed by an increase. An overall community‐scale size‐dependent mortality also showed a U‐shaped pattern.Tree mortality was also influenced by the competition and environmental conditions, but the relative importance varied widely across tree sizes and species. The competition showed significant correlations with the mortality of small trees while the effect of environmental conditions on mortality was strongest for large trees. A principal component analysis showed that a combination of biotic and abiotic factors explained 42.3% of the spatial variation in mortality at large sizes.Synthesis. Our results reveal four identifiable size‐dependent mortality patterns that differ across diverse species, jointly leading to a U‐shaped size–mortality pattern at the community level. This finding calls for the need to establish the details of every potential size–mortality pattern with consideration of the different effects of biotic and abiotic factors on tree mortality of specific size. DNA methylation is an epigenetic mark important for genome stability and gene expression. In Arabidopsis thaliana, the 5‐methylcytosine DNA glycosylase/demethylase DEMETER (DME) controls active DNA demethylation during the reproductive stage; however, the lethality of loss‐of‐function dme mutations has made it difficult to assess DME function in vegetative tissues. Here, we edited DME using clustered regularly interspaced short palindromic repeats (CRISPR) /CRISPR‐associated protein 9 and created three weak dme mutants that produced a few viable seeds. We also performed central cell‐specific complementation in a strong dme mutant and combined this line with mutations in the other three Arabidopsis demethylase genes to generate the dme ros1 dml2 dml3 (drdd) quadruple mutant. A DNA methylome analysis showed that DME is required for DNA demethylation at hundreds of genomic regions in vegetative tissues. A transcriptome analysis of the drdd mutant revealed that DME and the other three demethylases are important for plant responses to biotic and abiotic stresses in vegetative tissues. Despite the limited role of DME in regulating DNA methylation in vegetative tissues, the dme mutants showed increased susceptibility to bacterial and fungal pathogens. Although Co-IP has been widely used to examine I in vivo i protein-protein interactions, especially for strong and stable protein interactions, it needs to be performed with great experimental caution for weak or transient interactions, such as those that occur between a kinase and its substrates. Under normal conditions, TOR kinase phosphorylates ABA receptors (PYR1/PYL/RCARs) to abolish their ABA binding ability, thereby preventing stress signaling activation and promoting plant growth. In the presence of abscisic acid or environmental stress, activated SnRK2s transiently phosphorylate Raptor1B, a regulatory component of the TOR complex, to inhibit plant growth. As phosphorylation of hundreds of proteins has been shown to be dependent on SnRK2s (Umezawa et al., 2013; Wang et al., 2013, 2018, 2020), it is not surprising there are other kinases/signal components that can mediate stress adaptation processes downstream of SnRK2s. Vegetation carbon stock (Cveg) in global forests, which is important for C cycle‐climate feedbacks, commonly increases with forest age. Due to the allometric growth of plants, the nonlinear increase in Cveg with woody fraction (fw) is expected across space. However, it remains unclear whether such a nonlinear relationship between Cveg and fw can be constrained by observations and further used to benchmark Earth system models (ESMs). Here, based on the in situ measurements at 1,145 forest sites, we found that the nonlinear relationship between Cveg and fw followed an exponential equation (i.e., Cveg=bea·fw). Then, we showed that such an exponential dependence of Cveg on fw also exists in ESMs of CMIP5 and CMIP6 (all P < 0.01), even though age‐dependent processes have not been incorporated in most models. However, the exponential Cveg‐fw relationship varied greatly among the models, and the coefficient b was systematically lower in the ESMs (0.08 ± 0.11; mean ± SD) than the observations (0.28). Based on a compiled forest age data set, we further found that the observed nonlinear increase of Cveg with forest age across the Northern Hemisphere (>30°N) was not captured by ESMs. These findings reveal a high disagreement on the spatially nonlinear relationship between vegetation carbon stock and woody fraction in current ESMs. The exponential relationship based on observations provides one useful benchmark for ESMs when they implement the age‐dependent processes in the future. The nonlinear Cveg‐fw relationship was then used to benchmark the age impacts on Cveg predictions in Earth system models (ESMs). Combined with a global forest age data set, we show that current ESMs divergently represent the relationship between Cveg and forest age. High‐throughput gene sequencing has identified various genetic variants as the culprits for some common hereditary cancers. However, the heritability of a substantial proportion of cancers remains unexplained, which may result from rare deleterious mutations hidden in a myriad of nonsense genetic variations. This poses a great challenge to the understanding of the pathology and thus the rational design of effective treatments for affected patients. Here, whole genome sequencing is employed in a representative case in which one monozygotic twin is discordant for lung inflammatory myofibroblastoma to disclose rare tumor‐related mutations. A missense single nucleotide variation rs61955126 T>C in the lysine methyltransferase SETD8 (accession: NM_020382, SETD8C302R) is exposed. It is shown that SETD8 is vital for genomic integrity by promoting faithful DNA replication, and its C302R mutation downregulates the p53/p21 pathway. Importantly, the SETD8C302R mutation significantly increases the sensitivity of cancer cells to WEE1 inhibition. Given that WEE1 inhibitors have shown great promise for clinical approval, these results impart a potential therapeutic approach using WEE1 inhibitor for cancer patients carrying the same mutation, and indicate that genome sequencing and genetic functional studies can be integrated into individualized therapies. In eukaryotic cells, gene expression is greatly influenced by the dynamic chromatin environment. Epigenetic mechanisms, including covalent modifications to DNA and histone tails and the accessibility of chromatin, create various chromatin states for stress‐responsive gene expression that is important for adaptation to harsh environmental conditions. Recent studies have revealed that many epigenetic factors participate in abiotic stress responses, and various chromatin modifications are changed when plants are exposed to stressful environments. In this review, we summarize recent progress on the cross‐talk between abiotic stress response pathways and epigenetic regulatory pathways in plants. Our review focuses on epigenetic regulation of plant responses to extreme temperatures, drought, salinity, the stress hormone abscisic acid, nutrient limitations and ultraviolet stress, and on epigenetic mechanisms of stress memory. Glycyrrhizin is a bioactive triterpenoid saponin extracted from a traditional Chinese medicinal herb, glycyrrhiza, and has been reported to protect the organs such as liver and heart from injuries. However, there is no report about the effects of glycyrrhizin on atrophic age‐related macular degeneration (AMD). This study investigated the effects of glycyrrhizin on retinal pigment epithelium (RPE) in vitro and retina of mice in vivo treated with sodium iodate (SI). Glycyrrhizin significantly inhibited SI‐induced reactive oxygen species (ROS), and decreased apoptosis of RPE in vitro. The underlying mechanisms included increased phosphorylation of Akt, and increased expression of nuclear factor erythroid 2‐related factor2 (Nrf‐2) and HO‐1, thereby protecting RPE from SI‐induced ROS and apoptosis. Furthermore, glycyrrhizin significantly decreased the apoptosis of retinal cells in vivo, resulting in the inhibition of thinning of retina, decreasing the number of drusen and improving the function of retina. In addition to the known antitumour effects of ursolic acid (UA), increasing evidence indicates that this molecule plays a role in cardiac protection. In this study, the effects of ursolic acid on the heart in mice treated with doxorubicin (DOX) were assessed. The results showed that ursolic acid improved left ventrical fractional shortening (LVFS) and left ventrical ejection fraction (LVEF) of the heart, increased nitrogen oxide (NO) levels, inhibited reactive oxygen species (ROS) production and decreased cardiac apoptosis in mice treated with doxorubicin. Mechanistically, ursolic acid increased AKT and endothelial nitric‐oxide synthase (eNOS) phosphorylation levels, and enhanced eNOS expression, while inhibiting doxorubicin induced eNOS uncoupling through NADPH oxidase 4 (NOX4) down‐regulation. These effects of ursolic acid resulted in heart protection from doxorubicin‐induced injury. Summary: Aims: Glioblastoma (GBM) is a lethal disease of the central nervous system with high mortality, and novel therapeutic targets and strategies for GBM are urgently needed. Caveolae‐associated protein 1 (CAVIN1) is an essential caveolar component‐encoding gene and has been poorly studied in glioma. To this end, in this study, we evaluated CAVIN1 expression in glioma tissue as well as the correlation between CAVIN1 expression and prognosis in glioma patients using the data collected from clinical samples or from the Cancer Genome Atlas (TCGA), Chinese Glioma Genome Atlas (CGGA), Rembrandt, and Gene Expression Omnibus (GEO) data sets. Methods: Survival analysis was performed with the Kaplan‐Meier curve and log‐rank test. The predictive role of CAVIN1 in progressive malignancy in glioma was evaluated by using a receiver operator characteristic (ROC) curve. Gene ontology (GO), Gene set enrichment analysis (GSEA), and gene set variation analysis (GSVA) methods were used to interpret the functions of CAVIN1 in GBM. Results: CAVIN1 expression was elevated in GBM compared with that in low‐grade glioma and nontumor brain samples and was correlated with unfavorable outcomes in glioma patients. Additionally, CAVIN1 could serve as an independent predictive factor for progressive malignancy in GBM. Furthermore, CAVIN1 was associated with disrupted angiogenesis and immune response in the tumor microenvironment of GBM. Aims: Immune checkpoint blockade has made breakthroughs in immunotherapy for glioma. However, current immunotherapy has therapeutic benefits only in a subset of patients and accompanied by immune‐related side effects. SLAMF8 is a costimulatory molecule that affects the activation of macrophages in inflammation. The study of SLAMF8 may provide new information for immunological research and treatment of glioma. Methods: CGGA and TCGA cohorts of 946 patients with RNA sequencing data and full clinical information were analyzed using R language and GraphPad Prism 7. Results: SLAMF8 was overexpressed along with malignancy progression and was a biomarker of mesenchymal subtype. As an independent prognostic factor, high SLAMF8 conferred reduced overall survival and chemotherapy resistance. SLAMF8 implied lower proportion of cancer cells along with increasing enrichment of monocytic lineage, myeloid dendritic cells. Functional analysis showed higher SLAMF8 indicated activation of antigen processing and presenting and the IFN‐γ/TNF/TLR‐mediated signaling. Meanwhile, coexpressing with classical checkpoint SLAMF8 aggravated immunosuppression and enhanced inflammation response. Conclusion: Our study highlighted the important role of SLAMF8 in malignancy progression, shortened survival, and immune disorders. Glioblastomas (GBMs) are the most common of both benign and malignant primary brain tumours, in which the inflammatory and immunologic abnormalities are involved. Interleukin‐17A (IL‐17A) plays an important role in various inflammatory diseases and cancers. Several recent studies revealed that the expression of IL‐17A was overexpressed in human GBMs tissue. However, the accurate role of IL‐17A in GBMs remains unclear. In this study, we aimed to explore the effect of IL‐17A on cell migration and invasion of GBMs and the mechanism by which the effects occurred. We found that exogenous IL‐17A promoted significantly cell migration and invasion abilities in two GBMs cell lines (U87MG and U251) in a time‐dependent manner. In addition, the protein expressions of PI3K, Akt and MMP‐2/9 were increased in the GBMs cells challenged by IL‐17A. Furthermore, a tight junction protein ZO‐1 was down‐regulated but Twist and Bmi1 were up‐regulated. Treatment with a PI3K inhibitor (LY294002) significantly reduced the abilities of both migration and invasion in U87MG and U251 cells. LY294002 treatment also attenuated the IL‐17A causing increases of protein levels of PI3K, AKT, MMP‐2/9, Twist and the decreases of protein level of ZO‐1 in the U87MG and U251 cells. Taken together, we concluded that IL‐17A promotes the GBM cells migration and invasion via PI3K/AKT signalling pathway. Summary: Apple (Malus × domestica) trees are vulnerable to freezing temperatures. However, there has been only limited success in developing cold‐hardy cultivars. This lack of progress is due at least partly to lack of understanding of the molecular mechanisms of freezing tolerance in apple. In this study, we evaluated the potential roles for two R2R3 MYB transcription factors (TFs), MYB88 and the paralogous FLP (MYB124), in cold stress in apple and Arabidopsis. We found that MYB88 and MYB124 positively regulate freezing tolerance and cold‐responsive gene expression in both apple and Arabidopsis. Chromatin‐Immunoprecipitation‐qPCR and electrophoretic mobility shift assays showed that MdMYB88/MdMYB124 act as direct regulators of the COLD SHOCK DOMAIN PROTEIN 3 (MdCSP3) and CIRCADIAN CLOCK ASSOCIATED 1 (MdCCA1) genes. Dual luciferase reporter assay indicated that MdCCA1 but not MdCSP3 activated the expression of MdCBF3 under cold stress. Moreover, MdMYB88 and MdMYB124 promoted anthocyanin accumulation and H2O2 detoxification in response to cold. Taken together, our results suggest that MdMYB88 and MdMYB124 positively regulate cold hardiness and cold‐responsive gene expression under cold stress by C‐REPEAT BINDING FACTOR (CBF)‐dependent and CBF‐independent pathways. Purpose To describe a new robot-assisted surgical system for retinal vascular bypass surgery ( RVBS) and to compare the success rate with freehand RVBS. Methods A robot-assisted system for retinal microsurgery was constructed to include two independent robotic arms. A 23-gauge light probe and an intraocular forceps were affixed to the arm end effectors to perform the intraocular manipulation. Harvested porcine eyes were introduced to be established animal models of closed-sky eyeballs after that pars plana vitrectomy using temporary keratoprosthesis was performed by a skilful surgeon. Retinal vascular bypass surgery (RVBS) was performed by an inexperienced ophthalmologist to test the ease of use. A stainless steel wire (45- μm pipe diameter) was used as an artificial vessel. Before RVBS, the wires were prepositioned at the retinal surface of the eyes. The Control group ( n = 20) underwent freehand RVBS, and the Experimental group ( n = 20) underwent robot-assisted RVBS. To create the simulated bypass, the distal end of the wire was inserted into the selected vessel and advanced ~4 mm away from the optic disc. If successful, then the proximal wire end was inserted and advanced ~2 mm towards the optic disc. The difference in the success rate for the freehand and robot-assisted procedures was analysed by the chi-square test. Results The success rate for the freehand RVBS was 5% (1/20 eyes). In contrast, the robot-assisted success rate was 35% (7/20) of eyes (p < 0.05). Conclusion This study demonstrated the feasibility of robot-assisted RVBS in ex vivo porcine eyes. The robotic system increased the accuracy and stability of manipulation by eliminating freehand tremor, leading to a higher surgical success rate. [ABSTRACT FROM AUTHOR]

Transcription factors play a key role to enable plants to cope with abiotic stresses. DREB2 regulates the expression of several stress-inducible genes and constitutes major hubs in the water stress signalling webs. We cloned and characterized a novel gene encoding the Fp DREB2A transcription factor from Fraxinus pennsylvanica, and a yeast activity assay confirmed its DRE binding and transcription activation. Overexpression of Fp DREB2A in R. pseudoacacia showed enhanced resistance to drought stress. The transgenic plant survival rate was significantly higher than that of WT in soil drying and re-watering treatments. Transgenic lines showed a dramatic change in root architecture, and horizontal and vertical roots were found in transgenic plants compared to WT. The vertical roots penetrated in the field soil to more than 60 cm deep, while horizontal roots expanded within the top 20-30 cm of the soil. A physiological test demonstrated that chlorophyll contents were more gradually reduced and that soluble sugars and proline levels elevated more sharply but malondialdehyde level stayed the same ( P < 0.05). Plant hormone levels of abscisic acid and IAA were higher than that of WT, while gibberellins and zeatin riboside were found to be lower. The root transcriptomes were sequenced and annotated into 2011 differential expression genes ( DEGs). The DEGs were categorized in 149 pathways and were found to be involved in plant hormone signalling, transcription factors, stimulus responses, phenylalanine, carbohydrate and other metabolic pathways. Esophageal cancer and gastric cancer have shared risk factors and inherited susceptibility. Recent genome-wide association studies have identified multiple genetic loci associated with gastric cancer risk, which may also involve in the development of esophageal cancer. Herein, we evaluated the relationship of gastric cancer risk-related variants at 1q22, 3q13.3, 5p13.1, and 8q24 with the risk of esophageal squamous cell carcinoma ( ESCC) in a Chinese population with a case-control study (2139 cases and 2273 controls). We found that the T allele of rs2294008, an intronic variant of the PSCA gene at 8q24 that was previously associated with an increased risk of gastric cancer, was inversely associated with a decreased risk of ESCC (odds ratio = 0.90; 95% confidence interval, 0.81-0.99; P = 0.034). Of interest, the association of rs2294008 with ESCC was consistent with that observed in esophageal adenocarcinoma and ESCC in Caucasian populations. However, no significant associations were observed for the other three variants at 1q22 (rs4072037), 3q13.31 (rs9841504), and 5p13.1 (rs13361707). Summary Staphylococcus epidermidis strains are diverse in their pathogenicity; some are invasive and cause serious nosocomial infections, whereas others are non-pathogenic commensal organisms. To analyse the implications of different virulence factors in Staphylococcus epidermidis infections, the complete genome of Staphylococcus epidermidis strain ATCC 12228, a non-biofilm forming, non-infection associated strain used for detection of residual antibiotics in food products, was sequenced. This strain showed low virulence by mouse and rat experimental infections. The genome consists of a single 2499 279 bp chromosome and six plasmids. The chromosomal G + C content is 32.1% and 2419 protein coding sequences (CDS) are predicted, among which 230 are putative novel genes. Compared to the virulence factors in Staphylococcus aureus, aside from δ-haemolysin and β-haemolysin, other toxin genes were not found. In contrast, the majority of adhesin genes are intact in ATCC 12228. Most strikingly, the ica operon coding for the enzymes synthesizing interbacterial cellular polysaccharide is missing in ATCC 12228 and rearrangements of adjacent genes are shown. No mec genes, IS256, IS257, were found in ATCC 12228. It is suggested that the absence of the ica operon is a genetic marker in commensal Staphylococcus epidermidis strains which are less likely to become invasive. [ABSTRACT FROM AUTHOR]

The chloride mass balance (CMB) method for estimating groundwater recharge is economic and effective, provided that the hydrological conditions for its applications are met and the modeling parameters are known. However, modeling parameters such as precipitation and Cl− deposition rates vary temporally, most notably as a result of the climatic changes from late Pleistocene to Holocene. The temporal variability of atmospheric Cl− input and annual precipitation were considered in this study by using a discrete steady state CMB model with different parameters for late Pleistocene and Holocene. Cl− deposition rates, estimated from 36Cl data, were lower in late Pleistocene than Holocene at Yucca Mountain, Nevada, but higher in late Pleistocene than Holocene at Black Mesa, Arizona. Paleoclimate proxies at both Yucca Mountain and Black Mesa point to higher precipitation rates in late Pleistocene than Holocene. The resulting average recharge estimates for Black Mesa are 9 ± 5 mm/yr for Holocene and 35 ± 22 mm/yr for late Pleistocene. Local recharge rates at Yucca Mountain were estimated from the 36Cl/Cl ratios and Cl− concentrations in perched waters. The estimated recharge for Yucca Mountain is 5 ± 1 mm/yr for Holocene and 15 ± 5 mm/yr for late Pleistocene. These recharge rates are comparable to results of independent numerical groundwater flow models and watershed-scale infiltration models at Black Mesa and Yucca Mountain, respectively. Background Case Presentation Conclusions Mobile bearing fracture is a rare long‐term complication of unicompartmental knee arthroplasty (UKA), and relevant reports are sparse. Hence, its treatment options need further exploration.This study presents the case of fracture of a polyethylene insert that occurred 12 years after mobile bearing medial UKA in a 75‐year‐old overweight woman who then underwent surgical intervention at our institution. However, we encountered significant challenges in removing the fragments from the broken bearing, resulting in retention of the remaining one‐third of the fragment. We solved this problem by replacing the fractured insert with thicker mobile bearing. During the 1‐month postoperative follow‐up, the patient achieved good range of motion and excellent satisfaction, with no reported complications and a Knee Society Score of 90. Additionally, we reviewed the literature on the treatment for mobile bearing fractures after UKA.Bearing fracture is a rare cause of failure of mobile bearing UKA. This case highlights the challenges of UKA fracture bearing retrieval and underscores that mobile bearing replacement can be an effective intervention. The case we report shows that when removal of a residual meniscal bearing in a posterior dislocation is difficult to achieve, compromise may be an appropriate option because it does not cause additional complaints to the patient. This case emphasizes the importance of the surgeon having a thorough preoperative understanding of the location and potential pitfalls of fracture fragments in such situations. Objective Methods Results Conclusion Total hip arthroplasty (THA) combined with proximal femoral reconstruction is a novel osteotomy technique developed to address severe hip deformities. There is a paucity of robust clinical and radiological evidence regarding the outcomes of this novel osteotomy technique. This study aimed to evaluate the clinical and radiological outcomes of THA combined with proximal femoral reconstruction during the early follow‐up.This is a retrospective case series of 63 hips who underwent THA combined with proximal femoral reconstruction at a single institution between January 2020 and July 2023. The mean age of patients was 39.6 ± 12.6 years. The mean follow‐up was 25.6 ± 3.8 months. Surgical characteristics and perioperative variables were evaluated to assess the efficacy of this technique. Harris hip score (HHS) was utilized to evaluate hip function. Leg length discrepancy (LLD) was evaluated in X‐ray. The incidence of major adverse events including deep vein thrombosis (DVT), osteolysis, nonunion of the osteotomy, intraoperative femoral fracture, and infection was also evaluated. Paired‐samples t‐test was used to compare preoperative and postoperative HHS and LLD.The mean operative time was 125.1 min. The mean size of the acetabular components used was 45.2 mm, and the stem size was 7.5. The primary friction interface was ceramic‐on‐ceramic, accounting for 92.1% of cases. The average length of hospital stay was 8.5 days. The mean cost of treatment was 46,296.0 Yuan. There was a significant improvement in postoperative HHS (p < 0.001) and LLD (p < 0.001) compared to preoperative values. The incidence of deep venous thrombosis was 4.8%; osteolysis rates for the cup and stem were 4.8% and 6.4%, respectively. The non‐union and dislocation rates were 1.6% and 3.2%, respectively. There was no incidence of postoperative infection.The novel osteotomy surgical procedure yields reliable and impressive clinical and radiological outcomes, with minimal complications. ABSTRACT Deep sowing is a traditional method for drought resistance in maize production, and mesocotyl elongation is strongly associated with the ability of maize to germinate from deep soil. However, little is known about the functional genes and mechanisms regulating maize mesocotyl elongation. In the present study, we identified a plant‐specific SIMILAR TO RCD‐ONE (SRO) protein family member, ZmSRO1e, involved in maize mesocotyl elongation. The expression of ZmSRO1e is strongly inhibited upon transfer from dark to white light. The loss‐of‐function zmsro1e mutant exhibited a dramatically shorter mesocotyl than the wild‐type in both constant light and darkness, while overexpression of ZmSRO1e significantly promoted mesocotyl elongation, indicating that ZmSRO1e positively regulates mesocotyl elongation. We showed that ZmSRO1e physically interacted with ZmbZIP61, an ortholog of Arabidopsis ELONGATED HYPOCOTYL 5 (HY5) and showed a function similar to that of HY5 in regulating photomorphogenesis. We found that ZmSRO1e repressed the transcriptional activity of ZmbZIP61 toward target genes involved in the regulation of cell expansion, such as ZmEXPB4 and ZmEXPB6, by interfering with the binding of ZmbZIP61 to the promoters of target genes. It is challenging to manufacture large‐area, ultrathin, flexible/stretchable electronics on an industrial scale. Recent ground‐breaking advances in the manufacture of flexible electronics are based on powerful laser processes. Laser irradiation at an internally absorbing interface through a transparent substrate will bring various physical changes and chemical reactions at the interface, accompanied by distinct phenomena. Numerous techniques derived from these phenomena with the unique ability to fabricate materials, structures, and devices on flexible substrates with the advantages of noncontacting processing, high efficiency, adjustable coverage from micro‐ to macroscale, and compatibility with organic and inorganic materials have been developed. The latest impressive progress in these laser‐based techniques for the production of flexible electronics is reviewed, and the key developments in laser lift‐off, laser‐assisted printing, and laser‐assisted transfer printing techniques are highlighted. The fundamental principles of these techniques are discussed along with their basic mechanisms, followed by an exploration of the latest progress and future prospects in the field. The unique features of these techniques and state‐of‐the‐art applications related to flexible electronics are also highlighted. Finally, the challenges and future prospects of these techniques are explored, from the essential mechanics, engineering efforts, and novel approaches, to comprehensive combinations with innovative fabrication concepts and device structures.
