We investigated the effects of the presence of a selective insecticide, pyridalyl, in aqueous solutions of honey as food for adults of diamondback moth ( DBM) Plutella xylostella (L.) and its larval parasitoid Cotesia vestalis (Halliday) on their performances. We used a commercial formulation of pyridalyl which contained 10% pyridalyl. Survival times of DBMs reared with honey solution with pyridalyl at 10 000-fold dilution were not significantly different from those of DBMs reared with pure honey solution. However, at 1000-fold and 100-fold dilutions of pyridalyl in honey solution, survival times were significantly shorter than those with honey solution alone. By contrast, survival times of C. vestalis reared with honey solution with pyridalyl at 1000-fold and 100-fold dilution were not significantly different from those of C. vestalis reared with pure honey solution. Offering honey solution with pyridalyl at 100-fold dilution to C. vestalis did not affect its parasitization ability or offspring sex ratio. The effects of the insecticides etofenprox, malathion, diazinon, methomyl, alanycarb, clothianidin and emamectin benzoate on the foraging behaviour and survival of Cotesia vestalis (Hymenoptera: Braconidae), a larval parasitoid of the diamondback moth Plutella xylostella (Lepidoptera: Plutellidae) were investigated in the laboratory. The period required for C. vestalis females to reach P. xylostella-infested Komatsuna ( Brassica rapa) plants treated with etofenprox, methomyl or malathion was significantly longer than that to infested plants treated with emamectin benzoate or clothianidin; and the period to reach alanycarb or diazinon-treated plants was intermediate between them. The period to reach emamectin benzoate- or clothianidin-treated plants was not significantly different from that to distilled water-sprayed plants, suggesting that etofenprox, methomyl and malathion have strong inhibitory effects on the flight response, whereas the inhibitory effects of alanycarb and diazinon are relatively weak. By contrast, emamectin benzoate and clothianidin showed no inhibitory effect. The time of residency on the clothianidin-treated plants was significantly longer than that on other insecticide-treated plants (etofenprox, methomyl, alanycarb, malathion or diazinon), with the time of residency on emamectin benzoate-treated plants being intermediate between them. These results suggest that etofenprox, methomyl, alanycarb, malathion, and diazinon have a strong inhibitory effect on host-searching behaviour, while emamectin benzoate and clothianidin have a relatively weak effect. Furthermore, the mortality of wasps after foraging on the clothianidin-treated plants was significantly higher than after foraging on other insecticide-treated plants (etofenprox, methomyl or malathion), with subsequent mortality on alanycarb-, diazinon- and emamectin benzoate-treated plants being intermediate between them. Although it is well known that the application of broad-spectrum synthetic insecticides reduces the effectiveness of natural enemies, the details of the actual mechanisms, including the lethal and sublethal effects of this reduction, are not fully understood. The inhibitory effects of a pyrethroid insecticide (permethrin), Adion 20% EC on the flight responses, host-searching behaviour and foraging behaviour of Cotesia vestalis (Hymenoptera: Braconidae), a larval parasitoid of the diamondback moth, Plutella xylostella (Lepidoptera: Plutellidae), were investigated under laboratory conditions. In choice trials, the wasps showed significant preference for P. xylostella-infested Komatsuna plants over insecticide-treated plants, suggesting an inhibitory effect of the insecticide on the flight response of C. vestalis. When offered a pair of plants, the wasps showed a significant preference for P. xylostella-infested plants compared to uninfested plants. However, significantly more wasps were attracted to infested permethrin-treated plants than to uninfested plants, suggesting that the wasps are attracted to the volatile infochemicals from the infested plants, even if treated with permethrin. The searching time was significantly shorter and the mortality of C. vestalis adults on the insecticide-treated plants significantly higher than in the control plants treated with distilled water. These results suggest that the application of the insecticide had an inhibitory effect on the wasps’-searching behaviour and consequently reduced the effectiveness of C. vestalis as a biological control agent against P. xylostella. In addition, the strength of the inhibitory effect of permethrin on the attraction of the wasps to the plants is critical to the survival of C. vestalis. Aims: To clarify the distribution and significance of the oesophageal and gastric cardiac mucosae at the oesophago–gastric junction (EGJ). Methods and results: Oesophagectomy specimens from 131 consecutive patients with middle and upper thoracic oesophageal cancer were examined. The surgically resected specimens including the EGJ were cut into 5 mm thick serial sections and examined histopathologically for the length of the oesophageal and gastric cardiac mucosae and the incidence of columnar epithelial islands (CEIs). We also determined the presence of short-segment Barrett's oesophagus (SSBE) and goblet cell metaplasia in SSBE. Oesophageal cardiac mucosa was found in 125 cases (95%) and gastric cardiac mucosa was found in all cases. The mean length of the oesophageal and gastric cardiac mucosa was 4 mm (range 1–26 mm) and 13 mm (range 2–64 mm), respectively. CEIs were found in 75 cases (57%). SSBE was found in 70 cases (53%), among which goblet cell metaplasia was found in 28 cases (21%). No long-segment Barrett's oesophagus was found. The mean length of oesophageal cardiac mucosa (6 mm) and gastric cardiac mucosa (17 mm) in SSBE was significantly greater than that (3 mm and 8 mm, respectively) in non-SSBE cases ( P < 0.0001 and P < 0.0001). The incidence (69%) of CEIs in SSBE was significantly higher than that (44%) in non-SSBE cases ( P = 0.005). Conclusions: Oesophageal and gastric cardiac mucosae were found frequently. Summary Background The cysteinyl leukotriene receptor 1 (cysLTR1) antagonists are useful for oral treatment of bronchial asthma. The underlying mechanism of cysLTR1 antagonists on inhibition of inflammatory cytokine production is yet to be determined. Objective The present study was designed to determine the effect of pranlukast, a cysLTR1 antagonist, on production of inflammatory cytokines by allergen-stimulated peripheral blood monocytes (PBM) from atopic asthmatics. Methods PBM were obtained from normal control (n = 10) and Dermatophagoides farinae (Der f) allergen-sensitized atopic asthmatics (n = 12), and were cultured in the presence of Der f allergen. The production of TNF-α and nuclear-translocation of nuclear factor kappa B (NF-κB) was determined. In atopic asthmatics, pranlukast, tacrolimus or dexamethasone was added before stimulation by Der f. The additive effect of pranlukast and dexamethasone was also determined. Results PBM from atopic asthmatics cultured with Der f exhibited a significant increase in TNF-α production and nuclear translocation of NF-κB compared with normal control (P < 0.01). Pranlukast, tacrolimus and dexamethasone significantly inhibited production of TNF-α and nuclear-translocation of NF-κB in PBM of atopic asthmatics (P < 0.01). An additive effect of pranlukast on low-dose dexamethasone was also demonstrated. However, LTD4 did not induce TNF-α production or NF-κB nuclear translocation. Conclusion Our results suggest that pranlukast may inhibit TNF-α production via suppression of NF-κB activation through pathways distinct from cysLTR1 antagonism. [ABSTRACT FROM AUTHOR]

Background: About 70% of childhood asthmatics become free of asthma-related symptoms during adolescence. Little is known about bronchial hyperresponsiveness (BHR) and airway inflammation in young adults with “outgrown” childhood asthma. Methods: We studied 61 nonsmoking medical students (18 intermittent mild asthmatics, 23 students with outgrown childhood asthma but free of asthma-related symptoms for 10 years (asymptomatic asthmatics) and 20 healthy students). BHR and lung function were measured, and induced sputum samples analyzed for eosinophil count, eosinophilic cationic protein (ECP), granulocyte-macrophage colony stimulating factor (GM-CSF), and tumor necrosis factor-α (TNF-α). Results: BHR was still present in most asymptomatic asthmatics, but it was milder compared with healthy students. Only three subjects with previous asthma had no BHR and no signs of airway inflammation. Percentages of eosinophil, and ECP, TNF-α and GM-CSF concentrations in induced sputum of mild asthmatics and asymptomatic asthma groups were higher than in the healthy group. In asymptomatic asthmatics group, the duration of asthma, sputum eosinophil percentage, and the level of TNF-α in sputum correlated significantly with BHR. Conclusions: Only a few subjects with longstanding asymptomatic asthma could be considered as cured; most asymptomatic asthmatics continued to exhibit BHR and signs of airway inflammation. Unlabelled: The Aral Sea region is a natural area seriously polluted by human activities. In addition to the increased prevalence of diverse chronic diseases in children, the risk of developing urolithiasis is reported to be high in this region. This study was undertaken to clarify the prevalence of hypercalciuria in children of the Aral Sea region. A group of 205 children living in Kazalinsk, close to the Aral Sea, and a group of 187 children living in Zhanakorgan, far from the Aral Sea, were screened for hypercalciuria. Urinary sodium excretion (sodium per creatinine, uNa/Cr) in addition to calcium excretion (calcium per creatinine, uCa/Cr) was also calculated for each subject. Mean uCa/Cr (mmol/mmol) and uNa/Cr (mmol/mmol) excretions were significantly higher in Kazalinsk than in Zhanakorgan (uCa/Cr: 0.75 +/- 0.74 and 0.33 +/- 0.30; uNa/Cr: 3.54 +/- 2.27 and 2.89 +/- 1.69, respectively, mean +/- SD, p < 0.01). Hypercalciuria regarded as an uCa/Cr of more than 0.703 was observed in 79 out of 205 Kazalinsk children (38.6%) while this was seen in only 24 out of 187 Zhanakorgan children (12.8%). Linear regression analysis revealed a direct positive correlation between urinary calcium and sodium excretion (p < 0.01) in Kazalinsk children.Conclusion: The prevalence of hypercalciuria in children around the Aral Sea region is extremely high. This may be associated with excessive intake of calcium and sodium, or due to impaired renal tubular function caused by toxic chemicals. Infraorbital dark circles and wrinkles of the lower eyelids are a cosmetic problem, especially with age.To determine whether a gel containing 2% phytonadione, 0.1% retinol and 0.1% vitamins C and E is effective in reducing dark under-eye circles and wrinkles of the lower eyelids in healthy Japanese adults.Fifty-seven adult Japanese volunteers with dark under-eye circles and wrinkles were enrolled in an open label study. The gel formulation was applied twice daily to the lower eyelid site for 8 weeks. Haemostasis, pigmentation and wrinkles were evaluated by a physician and by the patients themselves, using a digital camera and a visual analogue scale respectively, after 4 and 8 weeks of treatment.Topical application of the gel decreased not only haemostasis but also wrinkles after 8 weeks of treatment. Of 57 patients, 27 (47%) had reductions in haemostasis. Wrinkles were also decreased in some patients. However, pigmentation was not clearly removed by this gel.Topical application of the gel containing 2% phytonadione, 0.1% retinol, 0.1% vitamin C and 0.1% vitamin E was fairly or moderately effective in reducing dark under-eye circles, especially in cases of haemostasis, over a short treatment period in healthy Japanese adults. Background: Allergic bronchopulmonary aspergillosis (ABPA) develops in the presence or absence of asthma, either atopic or nonatopic. We have tried to explore the essential components in the pathogenesis of the disease, which are either consistent and variable according to the presence and type of asthma., Methods: Non-cystic fibrosis ABPA cases satisfying Asano's criteria were extracted from a prospective registry of ABPA and related diseases in Japan between 2013 and 2023. According to the type of preceding asthma, ABPA was classified into three groups: ABPA sans asthma (no preceding asthma), ABPA with atopic asthma, and ABPA with nonatopic asthma. Exploratory and confirmatory factor analyses were performed to identify the components that determined the clinical characteristics of ABPA., Results: Among 106 cases of ABPA, 25 patients (24%) had ABPA sans asthma, whereas 57 (54%) and 24 (23%) had ABPA with atopic and nonatopic asthma, respectively. Factor analysis identified three components: allergic, eosinophilic, and fungal. Patients with atopic asthma showed the highest scores for the allergic component (p < .001), defined by total and allergen-specific IgE titers and lung opacities, and the lowest scores for the fungal component defined by the presence of specific precipitin/IgG or positive culture for A. fumigatus. Eosinophilic components, including peripheral blood eosinophil counts and presence of mucus plugs/high attenuation mucus in the bronchi, were consistent among the three groups., Conclusion: The eosinophilic component of ABPA is considered as the cardinal feature of ABPA regardless of the presence of preceding asthma or atopic predisposition., (© 2023 The Authors. Background: Great variability exists in repair strategies for the tetralogy of Fallot. While transannular patching, as introduced by Kirklin, was a breakthrough for primary repair, pulmonary regurgitation and a need for reoperation have led to the development of methods that preserve the natural morphology of the right ventricular outflow tract., Methods: State-of-the-art details of tetralogy of Fallot repair are explained from the standpoint of architectural preservation, especially with regard to sparing native tissue such as the annulus, valve, or infundibulum. Particular attention is given to the latest technical details of each approach, benefits and disadvantages, and any long-term data available., Results: The choice of procedure is complex and unique to each case as transannular patching alone may carry long-term pulmonary risks. Modifications that spare the annulus, valves, or infundibulum may thus be essential as preservation of natural morphology has resulted in excellent mid-term results., Conclusions: The complexity of tetralogy of Fallot repair demands constant attention to clinical presentation and vigilance against long-term sequelae. Background and Aims: The results of a randomized controlled study and meta-analysis study have recently proved that Helicobacter pylori eradication has a preventive effect against the development of metachronous and primary gastric cancer. However, gastric cancer is sometimes detected after successful eradication. There is a lack of study about gastric cancers in eradicated patients. To clarify the characteristics of gastric cancers detected after H. pylori eradication, we analyzed the clinicopathological features of these cancers., Methods: The subjects were 18 early-stage gastric cancer specimens resected from 17 patients who had received successful eradication of H. pylori from February 1995 to March 2009. The control group consisted of 36 specimens from noneradicated patients with persistent H. pylori infection who were matched with the subjects in age, sex, and depth of invasion. Clinicopathological features and mucin phenotypes of gastric cancer were clinically and immunohistologically evaluated., Results: The average diameter of gastric cancer was smaller and Ki-67 index was lower in the eradication group. The morphological distribution of depression types was significantly lower in the control group. Immunohistochemical phenotyping revealed that 72.2% of the lesions in the eradicated group were complete gastric type or gastric predominant mixed type, whereas the percentages of gastric type and intestinal type in the control group were similar., Conclusion: Our findings indicate that the clinicopathological characteristics of gastric cancers detected after H. pylori eradication are different from those of gastric cancers in patients with persistent H. pylori infection. We describe a case of an 82-year-old Japanese woman with basal cell carcinoma (BCC) on the leg with secondary chronic lymphedema due to treatment for uterine cancer. Sparse tumor nests with remarkable edema of the dermis in the nodule appeared to be influenced by the chronic lymphedema. However, it remains inconclusive whether or not the tumorigenesis of the BCC was associated with chronic lymphedema in this case.

We have recently segregated a new line of rabbit, named TGH, with severely high levels of plasma triglyceride and cholesterol. The aim of the present study was to investigate the progression of atherosclerosis and haemodynamic parameters in TGH rabbits. 2. Japanese white (JW) and TGH rabbits (24-27 months old) were anaesthetized with ketamine and xylazine. Plasma concentrations of triglyceride were 63.1 8.0 and 446.0 35.2 mg/dL in JW and TGH rabbits, respectively. Blood pressure was measured by a catheter implanted in the femoral artery. Histological examinations were performed using haematoxylin-eosin and elastica-Masson trichrome staining to detect atherosclerotic lesions. 3. The JW rabbits had no atherosclerotic lesions. In TGH rabbits, severe atherosclerotic lesions were observed throughout the aorta, especially in the aortic arch. Basal femoral arterial pressure was not significantly different between JW and TGH rabbits. However, the basal pulse pressure in TGH rabbits (48.3 4.5 mmHg) was significantly greater than that of JW rabbits (28.0 5.6 mmHg). Intravenous infusion of N(G)-nitro-L-arginine methyl ester (L-NAME; 26.9 mg/kg) increased the blood pressure of TGH and JW rabbits. There was no significant difference in the response to L-NAME between the two rabbit strains. 4. The present study shows that severe atherosclerotic changes develop in TGH rabbits and suggests that the hyperlipidaemia combined with hypercholesterolaemia and hypertriglyceridaemia is an important factor for promoting atherosclerosis in TGH rabbits. The greater pulse pressure in TGH rabbits may be due to the increased vascular stiffness with atherosclerosis. 5. This newly developed TGH rabbit line of heritable hypertriglyceridaemia with hypercholesterolaemia will become a useful animal model for studies on the role of hyperlipidaemia in the progression of atherosclerosis and in many atherosclerosis-related diseases.
