We have explored orally effective thyrotropin-releasing hormone (TRH) mimetics, showing oral bioavailability and brain penetration by structure–activity relationship (SAR) study on the basis of in vivo antagonistic activity on reserpine-induced hypothermia in mice. By primary screening of the synthesized TRH mimetics, we found a novel TRH mimetic: l-pyroglutamyl-[3-(thiazol-4-yl)-l-alanyl]-l-prolinamide with a high central nervous system effect compared with TRH as a lead compound. Further SAR optimization studies of this lead compound led to discovery of a novel orally effective TRH mimetic: 1-{N-[(4S,5S)-(5-methyl-2-oxooxazolidine-4-yl)carbonyl]-3-(thiazol-4-yl)-l-alanyl}-(2R)-2-methylpyrrolidine trihydrate (rovatirelin hydrate), which was selected as a candidate for clinical trials.

Polyethylene terephthalate (PET) fiber-reinforced polymer (FRP) composites are characterized by a large rupture strain (LRS) (usually larger than 5%) but a low elastic modulus compared to conventional FRP materials made of carbon, glass, and aramid fibers. Previous studies have proved that the use of PET FRP sheets as jacketing materials for plain concrete or reinforced concrete (RC) columns could efficiently enhance their ductility. This paper presents a combined experimental and analytical study on the shear strengthening of corroded RC columns with stirrups having different levels of corrosion before strengthening. The shear resistance contributions from the substrate column and PET FRP sheets at various volume ratios were carefully examined and their interaction mechanisms were discussed. Design equations following a conventional truss analogy were then proposed to predict the shear strength of PET FRP-wrapped columns considering the effect of stirrup corrosion, and the predictions were verified against the test results.

Summary When Escherichia coli grows in the presence of DNA-damaging agents such as methyl methanesulphonate (MMS), absence of the full-length form of Translation Initiation Factor 2 (IF2-1) or deficiency in helicase activity of replication restart protein PriA leads to a considerable loss of viability. MMS sensitivity of these mutants was contingent on the stringent response alarmone (p)ppGpp being at low levels. While zero levels (ppGpp0) greatly aggravated sensitivity, high levels promoted resistance. Moreover, M+ mutations, which suppress amino acid auxotrophy of ppGpp0 strains and which have been found to map to RNA polymerase subunits, largely restored resistance to IF2-1- and PriA helicase-deficient mutants. The truncated forms IF2-2/3 played a key part in inducing especially severe negative effects in ppGpp0 cells when restart function priB was knocked out, causing loss of viability and severe cell filamentation, indicative of SOS induction. Even a strain with the wild-type infB allele exhibited significant filamentation and MMS sensitivity in this background whereas mutations that prevent expression of IF2-2/3 essentially eliminated filamentation and largely restored MMS resistance. The results suggest different influences of IF2-1 and IF2-2/3 on the replication restart system depending on (p)ppGpp levels, each having the capacity to maximize survival under differing growth conditions.

Interview and questionnaire surveys were conducted with bus drivers in Japan, with the goal of developing an educational program for better control of emotions among bus drivers. The interviews aimed at identifying stressors and ways in which stress negatively influenced bus services. The questionnaire survey. which was being developed as a self-diagnosis tool, further provided bus drivers with the opportunity to understand their own emotional tendencies. Factor analysis identified six factors underlying work-related stress: anger at unsafe behaviours of nearby road users, irritation caused by complaints from passengers, time pressures, anxiety about traffic accidents, impatience with slow passengers, and resentment of bad-mannered passengers. The influence of stress on the drivers comprised four factors: cognitive failure, sullen behaviour, abrupt acceleration/deceleration, and aggressive driving. Moreover, drivers with lower stress were relatively older and more experienced. 本研究では，T字型無信号交差点内に敷設された台形ハンプが，非優先道路から優先道路へ右折する車両に及ぼす影響を観察調査により分析した．5つの時期（ハンプ敷設前，敷設直後，3ヶ月後，半年後，1年後）に記録された2740台のデータを分析したところ，ハンプは通行車両の速度を低下させる効果があるものの，その有効性は時間経過とともに小さくなることが示された．また，ハンプ敷設直後から3ヶ月後にかけては，ドライバーの左右確認回数が一時的に増加し，正の波及効果が認められた．その一方，一時停止率はハンプ敷設前が最も高く，負の波及効果が生じた．これらの結果から，道路安全対策の有効性評価に際しては，一時的な効果だけでなく持続的な効果測定が必要であり，また狙いとする側面以外への影響についてもリスク補償行動の観点から検証することが重要と言える．

Summary Conversion of bacteriophage Mu repressor to ClpXP-sensitive form correlates with induced local flexibility at the ClpX recognition motif located at the C-terminal end. Changing the C-terminal valine to an alanine (RepV196A) caused the degradation tag to be constitutively active like that of mutant repressors called Vir, which have a dominant ClpXP-sensitive conformation. However, unlike Vir, RepV196A was unable to convert wild-type repressor (Rep) to the ClpXP-sensitive form. In mixtures with Rep, only RepV196A was rapidly degraded by ClpXP. Unlike Rep, RepV196A was ClpXP sensitive without induced C-terminal flexibility. And unlike adaptor proteins that tether and deliver substrates to ClpX for trans-targeting, Vir promoted rapid degradation of Rep by ClpX deleted for the tethering site that binds adaptor proteins. Therefore, Rep's ClpX recognition motif has regulable properties, allowing an alternative trans-targeting mechanism in which an inactive degradation tag is turned on by induced conformational changes.

建設コスト縮減のために，連続合成桁が見直され，様々な開発・研究が行われている．それらの設計・施工においては，床版の耐久性を向上するために，中間支点上の橋軸方向プレストレス導入を目的としたジャッキアップダウン工法や，内・外PC 鋼材を用いたプレストレス導入工法が主流となっている．しかしながら，これらの工法では，使用限界状態が改善されるものの，終局限界状態における耐荷力は全く，または，ほとんど改善され得ない．そこで，加熱した補強鋼板を鋼桁に高力ボルトで取り付け，鋼桁を補強すると同時にプレストレス力を導入する熱プレストレス工法によって，使用限界状態と同時に，終局限界状態の耐荷力を向上させる研究を試みた．本稿では，その実験，および解析結果を報告するものである．

Kinoshita and Nakai (Celest. Mech. Dyn. Astr. 75, 125–147, 1999) gave the analytical solution of the Kozai mechanism. In this solution the eccentricity and the inclination of a disturbed body take any value, but the argument of the pericenter is restricted to take 0° or 90°. In this paper, we derive the general solution that can be applied for any value of the argument of the pericenter.

近年，過去の高度経済成長期に建設された多くの道路橋が，高齢化の時期を迎えている．したがって，これらの橋梁を，貴重な社会資本として，延命化させることが重要な課題である．既設鋼橋の補強工法としては，単なる添接補強材工法と外ケーブル工法とが挙げられるが，その補強効果に，長短がある．筆者らは，既設鋼橋の新しい補強・補修工法として，熱プレストレス工法を提案してきた．この工法は，耐荷力が不足している橋梁でも，B活荷重対応橋梁として，格上げすることができる，同時に，耐久陸も向上させることができる．本文では，この熱プレストレス工法による既設連続桁の補強・補修効果について検討した結果を報告する．

Primosome assembly protein PriA functions in the assembly of the replisome at forked DNA structures. Whereas its N-terminal DNA binding domain (DBD) binds independently to DNA, the affinity of DBD protein for forked structures is relatively weak. Although the PriA helicase domain (HD) is required for high affinity fork binding, HD protein had very low affinity for DNA. It had only low levels of ATPase activity, and it hydrolyzed ATP when DNA was absent whereas PriA did not. HD catalyzed unwinding of a minimal substrate composed of a duplex with a 3' single-stranded tail. Single-strand binding protein (SSB) bound to the tail of this substrate inhibited this reaction by full-length PriA but enhanced the reaction by HD. SSB stabilized binding of PriA but not of DBD or HD to duplexes with a 5' or 3' single-stranded tail. On forked substrates SSB enhanced helicase action on the lagging-strand arm by PriA but not by HD. The results indicate that synergy of the DBD and HD allows stable binding at the interface between duplex and single-stranded DNA bound by SSB. This mode of binding may be analogous to fork binding, which orients the helicase to act on the lagging-strand side of the fork.

Novel 15-membered macrolides possessing the dilactone skeleton, diolides 13a and 13b, have been synthesized in our research program aimed at finding new antibacterial macrolides. Key strategic elements of the approach include the ring-expanding reaction of 13-membered dilactones, prepared from erythromycin A (Ery-A), to 15-membered dilactones via intramolecular translactonization. The absolute configuration at the regenerated C-8 position of the new diolides was determined by chemical transformation, leading to the corresponding lactam analogues, whose stereochemistry is known in the literature. For further confirmation, X-ray analysis was performed. The X-ray structure determination of 13a revealed a backbone conformation similar to that of Ery-A. Novel 15-membered diolide 13a and the 11,12-diol 18 exhibited antibacterial activities comparable to that of Ery-A.

Two novel compounds, stachyflin and acetylstachyflin, have been isolated by solid-state fermentation of Stachybotrys sp. RF-7260. The structures of both metabolites, determined by detailed NMR analyses and X-ray crystallographic analysis, are novel with a pentacyclic moiety including cis-fused decalin. The absolute stereochemistry of stachyflins was determined by circular dichroism analysis. Stachyflin showed antiviral activity against influenza A virus (H1N1) in vitro with an IC50 value of 0.003 microM. Acetylstachyflin was about 77-fold less active than stachyflin.

The efficiency of applying composite members as an alternative of improving the compressive strength of steel structures has already been proved by several researches and experiments. However, not much has been done concerning the creep phenomenon in such structures. The present paper reports on a series of experiments that was carried out in an attempt to clarify the time-dependent behavior of concrete filled steel members through a viscoelastic model. Creep coefficients thus obtained were then compared to those recommended by the Japanese Specifications for Highway Bridges. Comments on the effects of concrete shrinkage observed during the experimental measurements were also added.

The squalene synthase inhibitor bisabosqual A was isolated from the culture broth of Stachybotrys sp. RF-7260, and its structure was determined on the basis of spectroscopic methods including detailed 2D NMR analyses. The structures of bisabosquals B, C and D isolated from Stachybotrys ruwenzoriensis RF-6853 were determined by spectroscopic methods and chemical reactions. The absolute stereochemistry of bisabosquals A, B and D was determined by X-ray crystallographic analysis. They have novel cis-fused tetracyclic structures with a bisabolane-type sesquiterpene and phenol moieties.

連続構造上路式吊床版橋では, 活荷重載荷に伴い吊床版に生じる水平力による橋脚の変形を抑えることで, 吊床版取付部の曲げモーメントや吊床版のたわみを低減させることができる. この目的で, 上床版端部を橋台と弾性結合する構造を考案したが, 本論文では, この弾性結合構造を有する連続構造上路式吊床版橋の構造特性について示すとともに, 端部ばね定数を簡略的に設定する手法を提案した. また, 構造特性を把握するために行った解析の妥当性を, 実橋における静的・動的載荷試験結果により確認した.

本論文では, 開口部を有する圧縮補剛板の終局強度特性を明らかにし, 効果的な補強方法について検討を加え, 簡便な耐荷力算定法を提案している. まず, 実鋼製橋脚20基の初期たわみを実測するとともに, 縮尺模型の残留応力を計測している. そして, この種の補剛板の終局強度に影響を及ぼす初期不整の特性を調べている. つぎに, 4種類の実験供試体による耐荷力実験, および, その実験供試体を対象とした解析モデルを用いた弾塑性有限変位解析を行い, 対象とする補剛板の終局強度特性について検討している. また, 多数の弾塑性有限変位解析を行うことによって, 開口部の補強方法, および開口部を有する補剛板の耐荷力の一算定法を提案している.

After the shock of the Hyogo-Ken Nambu Earthquake, a newly revised Japanese Seismic Design Method for Bridge Structures requires that steel bridge piers should not suffer serious damage due to such strong earthquakes. As one of the suitable methods to meet this requirement, a method of inserting an additional steel tube into the inside of a steel bridge pier can be considered. In this method, the ductility of the bridge pier is significantly enhanced, if the cross section is designed in such a manner that the axial compressive load, caused by the dead load of the superstructure, is mainly carried by the inner steel tube and the local buckling of the outer steel pier as well as the column buckling of the inner steel tube are prevented by the encased concrete between them. To investigate the ultimate strength and ductility of these concrete-filled steel box columns under strong earthquakes, an experimental study is executed.

本研究では, コンクリートを充填した薄肉円形断面の鋼製部材にねじり, 曲げ, および, それらの組合せ断面力が作用する場合の剛性, 降伏・終局強度, および変形性能について検討するために, 耐荷力実験を実施した. すなわち, 径厚比D/tが30～40の比較的コンパクトな断面の鋼管部材, および, それにコンクリートを充填し合成部材とした実験供試体を各4体の計8体作製した. そして, それらに一様な分布をするねじりモーメントと三角形分布の曲げモーメントとを単独に, あるいは同時に載荷する耐荷力実験を実施した. その結果, 鋼管によって閉じ込められた充填コンクリートに作用する側圧を考慮した実際的な充填コンクリートの圧縮強度と, せん断強度とを基準とした合成断面の全塑性相関曲線を用いれば, この種の合成断面の終局強度を精度よく推定できることがわかった.

Investigated in this paper is a new hybrid cable-wire consisting of outer new Fiber Reinforced Plastic (FRP) and a steel wire as a core. First of all, a design method of cross section of the new hybrid cable-wire is proposed on the basis of the safety factors of 1.7, 3.0 and 1.1 against the yield limit state of the core steel wire, the ultimate limit state of the hybrid wire and the hazard state corresponding to the breaking of the outer FRP by accidents from standpoint of fail safe, respectively. Next, the mechanical properties of carbon-steel hybrid wires are dealt with, which consist of outer CFRP containing parallel carbon fibers and a core steel wire, and are dimensioned by the design method and produced by a method called the pultrusion method for trial. Results of a static tensile test, bending test, lateral pressure test and fatigue test prove the possibility of their application to structural cables and the necessity of further study for developing reliable anchorage systems of hybrid cables. A corrosion test emphasizes the importance of investigation on countermeasures against galvanic corrosion of carbon-steel hybrid wires.

著者らは, 文献6)～10) において, 無補剛・補剛薄肉箱形断面短柱に圧縮, 曲げ (2軸曲げを含む), および, ねじりがそれぞれ単独に, あるいは, それらが組み合わさって作用する場合の終局強度特性を, 実験, ならびに解析を通して検討し, 各断面力を単独に受ける薄肉箱形断面短柱の耐荷力曲線, および, これらが組み合わさって作用する場合の終局強度相関曲線を提案している. 本論文では, これらの耐荷力曲線, および終局強度相関曲線を, 弾塑性有限変位理論にもとづくパラメトリック解析を通して再検討し, これまでの研究成果をまとめたものである. さらに, 本論文では, 薄肉箱形断面短柱の終局強度をより精度よく推定すべく, 数々の検討も行っている.

本論文では, 薄肉箱形断面を有する単独柱形式の鋼製橋脚柱の耐震性を向上ための以下の方法について検討するために, 7体の実験供試体を用いて, 仮動的実験 (ハイブリッド実験), および漸増変位の繰返し載荷実験を実施している. すなわち, (1) 断面の構成板パネルの幅厚比を小さくして局部座屈を防止する方法, (2) コンクリートを断面内部へ部分的に充填するときの充填方法, および (3) 柱基部を三角リブで補強する方法を, 主に着目している. そして, それらの方法を用いれば, 橋脚柱の耐震性をどの程度向上させることができるのかについて検討し, それらの方法を用いる際の留意点についてまとめている.

Herniography was performed to improve diagnosis prior to surgery in a 9-year-old male patient, who presented a left groin bulging mass not typical for indirect hernia. Under local anesthesia, peritoneal needle-puncture was performed with ultrasonographic guidance. Herniograms were obtained after an injection of 50-ml non-ionic contrast medium under fluoroscopy. The total examination time was approximately 20 minutes. No complication was encountered. Scrotal hernia in the left side and patent processus vaginalis in the right side were clearly demonstrated. Both sides were then properly repaired simultane-ously in consideration of those herniographic findings. Herniography can be performed simply and safely even on children, who can tolerate abdominal puncture under local anesthesia. This examination provides useful information for selection of proper surgical method preoperatively, especially in cases with diagnostic difficulties. It is supposed to be indicated selectively for older children whose clinical manifestations are not typical for congenital inguinal hernias.

During transposition bacteriophage Mu transposase (MuA) catalyzes the transfer of a DNA strand at each Mu end to target DNA and then remains tightly bound to the Mu ends. Initiation of Mu DNA replication on the resulting strand transfer complex (STC1) requires specific host replication proteins and host factors from two partially purified enzyme fractions designated Mu replication factors alpha and beta (MRFalpha and beta). Escherichia coli ClpX protein, a molecular chaperone, is a component required for MRFalpha activity, which removes MuA from DNA for the establishment of a Mu replication fork. ClpX protein alters the conformation of DNA-bound MuA and converts STC1 to a less stable form (STC2). One or more additional components of MRFalpha (MRFalpha2) displace MuA from STC2 to form a nucleoprotein complex (STC3), that requires the specific replication proteins and MRFbeta for Mu DNA synthesis. MuA present in STC2 is essential for its conversion to STC3. If MuA is removed from STC2, Mu DNA synthesis no longer requires MRFalpha2, MRFbeta and the specific replication proteins. These results indicate that ClpX protein activates MuA in STC1 so that it can recruit crucial host factors needed to initiate Mu DNA synthesis by specific replication enzymes.

In order to clarify the metabolic fate of MCI-186 in rats, blood and plasma levels, distribution, metabolism and excretion were investigated after a single intravenous administration of 14C-MCI-186 to male and female rats. 1) After intravenous administration of 14C-MCI-186 (0.3, 2, 5.2 mg/kg), blood levels of radioactivity decreased relatively rapidly, and the areas under the concentration time curves (AUC0→∞) correlated well with doses (r=0.987). 2) Plasm a levels of radioactivity and the unchanged MCI-186 decreased rapidly and biphasically after intravenous administration of 14C-MCI-186 (2 mg/kg). In plasma, sulfate of MCI-186, unchanged MCI-186 and glucuronide of MCI-186 were present in higher levels and their AUC0→∞ accounted for 30 ?? 33%, 13 ?? 15% and 6 % of the AUC0→∞ of plasma radioactivities, respectively. 3) Distribution of radioactivity to tissues was rapid, and disappearance of radioactivity was also rapid except from the blood vessel (aorta). Disappearance of radioactivity from blood vessel was very delayed. And sex difference was observed in these disappearance, that is, in female rats radioactivities in tissues decreased more rapidly than in male rats. Concentrations of radioactivity in brain at 5 min after administration were low corresponding to 1/16 ?? 1/18 of the concentrations in plasma, and site specific distribution in brain was not observed. Most of radioactivity in brain consisted with unchanged MCI-186, and most of radioactivity in kidney consisted of sulfate of MCI-186. 4) Excretion of radioactivity was rapid and complete in both male and female rats. Eighty six to 92% of the dosed radioactivity was excreted to urine, therefore main excretion route was urinary excretion. Biliary excretion was 4.6% of the dose and it is possible that most of it was subjected to entero-hepatic circulation.

Pluto's motion is chaotic in the sense that the maximum Lyapunov exponent is positive and the Lyapunov time (the inverse of the Lyapunov exponent) is about 20 million years (Myr). We have carried out the numerical integration of Pluto over the age of the solar system (5.7 billion years towards the past and 5.5 billion years towards the future), which is about 280 times of the Lyapunov time. Our integration does not show any indication of gross instability in the motion of Pluto. The time evolution of Keplerian elements of a nearby trajectory of Pluto at first grow linearly with the time and then start to increase exponentially. These exponential divergences stop at about 420 Myr and saturate. The exponential divergences are suppressed by the following three resonances that Pluto has: (1)Pluto is in the 3:2 mean motion resonance with Neptune and the libration period of the critical argument is about 20000 years.(2)The argument of perihelion librates around 90 degrees and its period is 3.8 Myr.(3)The motion of the Pluto's orbital plane referred to the Neptune's orbital plane is synchronized with the libration of the argument of perihelion (a secondary resonance). The libration period associated with the second resonance is 34.5 Myr.We briefly discuss the motions of Kuiper belt objects in a 3:2 mean motion resonance with Neptune and several possible scenarios how Pluto evolves to the present stable state.

During and after a single or repeated intravenous administration of 14C-MCI-186 (2 mg/kg/day) for 7, 14 and 21 days to male rats, blood and plasma levels, distribution, metabolism, excretion and accumulation of MCI-186 were investigated. 1) Tissue levels of radioactivity at 5 min after each dosing during repeated administration were constant except in the blood vessel (aorta). Tissue levels of radioactivity at 24 hr increased to 2 ?? 9 times by 21 days repeated administration, but they were saturated after 7 or 14 days repeated administration. Transfer rates to blood cells also increased by repeated administration. Decrease of radioactivity from tissues was delayed after 21 days repeated administration, especially from the blood vessel (aorta). 2) Concentrations of the unchanged MCI-186 and metabolites in plasma, brain, liver and kidney at 5 min after each dosing during repeated administration were not significantly changed by repeated administration, however concentrations of the unchanged MCI-186 in the liver showed a tendency to decrease by repeated administration. 3) During 21 days repeated administration, radioactivities were excreted to urine and feces constantly, and until 192 hr after final dosing of repeated administration, 88% and 10% of the total dose were excreted to urine and feces, respectively. Total excretion extent reached 99% of the dose, therefore remaining fraction in the rat body was suggested to be minimal. Urinary excretion of the unchanged MCI-186 and metabolites were also not affected by repeated administration, except the excretion rates of MCI-186 glucuronide those seemed to decrease by repeated administration.

In order to clalify the species difference in metabolism of MCI-186, the identification of metabolites, urinary excretion rates of unchanged MCI-186 and its metabolites, and in vitro metabolism were investigated in rats, dogs and human. 1. Main metabolites of MCI-186 were sulfate and glucuronide of MCI-186 in all species. In the urine of rats and dogs, excretion rates of sulfate were higher than those of glucuronide, however in the urine of human, excretion rate of glucuronide was higher than that of sulfate, therefore species difference was observed in urinary excretion. 2. Urinary excretion was the main excretion route in all species. Excretion rates of the unchanged MCI-186 to urine were less than 3 % of dose. In dogs and human, urinary excretion can be mostly explained by excretion of unchanged MCI-186, and its sulfate and glucuronide. However in rats, unknown metabolites were present at higher rates of 20 ?? 27% of the urinary radioactivities. And sex difference in urinary excretion was not observed mostly in rats. 3. Activities of formation of MCI-186 sulfate were higher than those of MCI-186 glucuronide at in vitro metabolism of MCI-186 with liver S9 of rats and dogs. And both activities were higher in male rats than in female rats.

Three planets are detected around ν Andromedae. The stability of Upsilon Andromedae Planetary system is maintained by the co rotation of the pericenters of the two outer planets. If the pericenters of the two outer planets move independently, the planetary system becomes unstable. The corotation of the pericenters is explained by the secular perturbation theory. This corotation is not a secular resonance.

本研究は, 圧縮, 曲げ, および, ねじりの代表的な組合せ断面力を受ける薄肉補剛箱形断面の終局状態に至るまでの挙動, ならびに座屈・終局強度特性について, 実験的な立場から検討を加えたものである. また, 別途, 理論的研究によって導かれた圧縮, 曲げ, および, ねじりが単独に作用した場合のそれぞれの耐荷力曲線, ならびに, これらの組合せ断面力が作用したときの終局強度相関曲線の妥当性を実験データをもとにして検討している. さらに, 道路橋示方書に基づく許容強度曲線, および許容強度相関曲線, ならびに上述の耐荷力曲線, また終局強度相関曲線を安全率1.7で縮尺した許容強度曲線, および許容強度相関曲線と実験結果とを比較し, これらの曲線の妥当性についても若干の考察を行っている.

本論文は, 曲線箱桁橋の耐荷力を, 弾塑性有限変位解析によるものと, 弾性微小変位解析により推定されるものとを比較・検討し, 弾性微小変位解析でも十分な精度で推定できることを明らかにした. さらに, 弾性微小変位解析理論のプログラムに, 薄肉曲線箱桁断面としての局部座屈を考慮した終局強度の判定条件を付加して, 曲線箱桁橋の耐荷力の推定方法について示したものである.
