Background: Experimental and clinical data suggest that reducing inflammation without affecting lipid levels may reduce the risk of cardiovascular disease. Yet, the inflammatory hypothesis of atherothrombosis has remained unproved. Methods: We conducted a randomized, double-blind trial of canakinumab, a therapeutic monoclonal antibody targeting interleukin-1β, involving 10,061 patients with previous myocardial infarction and a high-sensitivity C-reactive protein level of 2 mg or more per liter. The trial compared three doses of canakinumab (50 mg, 150 mg, and 300 mg, administered subcutaneously every 3 months) with placebo. The primary efficacy end point was nonfatal myocardial infarction, nonfatal stroke, or cardiovascular death. RESULTS: At 48 months, the median reduction from baseline in the high-sensitivity C-reactive protein level was 26 percentage points greater in the group that received the 50-mg dose of canakinumab, 37 percentage points greater in the 150-mg group, and 41 percentage points greater in the 300-mg group than in the placebo group. Canakinumab did not reduce lipid levels from baseline. At a median follow-up of 3.7 years, the incidence rate for the primary end point was 4.50 events per 100 person-years in the placebo group, 4.11 events per 100 person-years in the 50-mg group, 3.86 events per 100 person-years in the 150-mg group, and 3.90 events per 100 person-years in the 300-mg group. The hazard ratios as compared with placebo were as follows: in the 50-mg group, 0.93 (95% confidence interval [CI], 0.80 to 1.07; P = 0.30); in the 150-mg group, 0.85 (95% CI, 0.74 to 0.98; P = 0.021); and in the 300-mg group, 0.86 (95% CI, 0.75 to 0.99; P = 0.031). The 150-mg dose, but not the other doses, met the prespecified multiplicity-adjusted threshold for statistical significance for the primary end point and the secondary end point that additionally included hospitalization for unstable angina that led to urgent revascularization (hazard ratio vs. placebo, 0.83; 95% CI, 0.73 to 0.95; P = 0.005). Canakinumab was associated with a higher incidence of fatal infection than was placebo. There was no significant difference in all-cause mortality (hazard ratio for all canakinumab doses vs. placebo, 0.94; 95% CI, 0.83 to 1.06; P = 0.31). Conclusions: Antiinflammatory therapy targeting the interleukin-1β innate immunity pathway with canakinumab at a dose of 150 mg every 3 months led to a significantly lower rate of recurrent cardiovascular events than placebo, independent of lipid-level lowering. 21. Correlated crust and mantle strain fields in Tibet

Vertical to subvertical planes of shear within the active crustal deformation field in Tibet align with fast directions of shear-wave polarization. This observation suggests that the present-day velocity gradient tensor field within the crust of Tibet correlates with the lithospheric mantle velocity gradient tensor field beneath Tibet. This inference requires the following. (1) The [100] axes of olivine are aligned within established zones of lithospheric mantle fabric or anisotropy, built up through finite strain. (2) Present-day shear in the mantle lithosphere is occurring within these established zones of fabric, parallel to the direction of olivine [100] axis alignment. (3) The zones of mantle lithospheric fabric, or zones of mantle shear, are aligned with zones of crustal shear (faults). The correlation of crustal and mantle strain fields most simply results from the fact that both crust and mantle lithosphere are under the influence of similar velocity boundary conditions. Furthermore, the observations confirm the distributed nature of lithospheric mantle deformation beneath east-central Tibet and suggest that left-lateral shear has been the dominant component of finite shear there. - Abstract
Mars’ polar regions are covered with ice-rich layered deposits that potentially contain a record of climate variations. The sounding radar SHARAD on the Mars Reconnaissance Orbiter mapped detailed subsurface stratigraphy in the Promethei Lingula region of the south polar plateau, Planum Australe. Radar reflections interpreted as layers are correlated across adjacent orbits and are continuous for up to 150 kilometers along spacecraft orbital tracks. The reflectors are often separated into discrete reflector sequences, and strong echoes are seen as deep as 1 kilometer. In some cases, the sequences are dipping with respect to each other, suggesting an interdepositional period of erosion. In Australe Sulci, layers are exhumed, indicating recent erosion.

- Abstract
The study of gene function in testis and sperm has been greatly assisted by transgenic mouse models. Recently, an alternative way of expressing transgenes in mouse testis has been developed that uses electroporation to introduce transgenes into the male germ cells. This approach has been successfully used to transiently express reporter genes driven by constitutive and testis-specific promoters. It has been proposed as an alternative method for studying gene function in testis and sperm, and as a novel way to create transgenic animals. However, the low levels and transient nature of transgene expression that can be achieved using this technique have raised concerns about its practical usefulness. It has also not been demonstrated in mammals other than mice. In this study, we show for the first time that in vivo gene transfer using electroporation can be used to express a fluorescent transgene in the testis of a mammal other than mice, the Syrian golden hamster. Significantly, for the first time we demonstrate expression of a transgene in epididymal sperm using this approach. We show that expression of the transgene can be detected in sperm for as long as 60 days following gene transfer. Finally, we provide the first systematic demonstration that this technique does not lead to any significant long-term adverse effects on testicular integrity and sperm quality. This technique therefore offers a novel way to study gene function during fertilization in hamsters and may also have potential as a way of creating transgenic versions of this important model species.

- Abstract
In mammals, fertilization and early pre-implantation development occur in the oviduct. Previous results obtained in our laboratory have identified specific molecules in the oviduct that affect porcine sperm-egg interactions. The aim of the present study was to determine whether the contact between oocytes and oviductal fluid also affect embryo development, quality, and gene expression. In vitro matured porcine oocytes were exposed to bovine oviductal fluid (bOF) for 30 min prior to fertilization. Cleavage and blastocyst development rates were significantly higher from bOF-treated oocytes than from untreated oocytes. Blastocysts obtained from bOF-treated oocytes had significantly greater total cell numbers than those obtained from untreated oocytes. Using real-time PCR, grade 1 (very good morphological quality) and grade 2 (good morphological quality) blastocysts were analyzed for gene transcripts related to apoptosis (BAX, BCL2L1), mitochondrial DNA (mtDNA) transcription/replication (POLG, POLG2, and TFAM), blastomere connection and morula compaction (GJA1), and blastocyst formation and pluripotency (POU5F1). We found that the entire set of genes analyzed was differentially expressed between grade 1 and 2 blastocysts. Furthermore, bOF treatment reduced the ratio of BAX to BCL2L1 transcripts and enhanced the abundance of TFAM transcripts in grade 2 blastocysts. Not only do these findings demonstrate that factors within the bOF act on porcine oocytes both quickly and positively, but they also suggest that such factors could promote embryo development and quality by protecting them against adverse impacts on mtDNA transcription/replication and apoptosis induced by the culture environment. © 2009 Society for Reproduction and Fertility.

Positron lifetime measurements were made in copolyimides synthesized from linear 4,4 prime-bis(3,4-dicarboxyphenoxy) diphenylsulfide dianhydride (BDSDA)/4,4 prime-diaminodiphenyl (ODA) and BDSDA/1,3-diaminobenzene (m-phenylene diamine) homopolymers. The probability of positronium formation as well as its subsequent lifetime are lower in the BDSDA/ODA/MPD (50-50) copolyimide, indicating the presence of a transition molecular architecture characterized by higher electron density and stronger bonds which permit both chemical as well as physical entry of water molecules into it. The presence of this transition region imparts unique physical and mechanical properties to the copolyimide.

Positron annihilation time spectra have been measured in nylon-6 samples as a function of their moisture content. The measured average long life component lifetime values are: 1722 + or - 47 ps (dry), 1676 + or - 40 ps (14.6 percent saturation value), 1719 + or - 26 ps (29.3 percent saturation value), 1720 + or - 35 ps (50 percent of saturation value), 1857 + or - 35 ps (78.1 percent saturation value), and 1936 + or - 57 ps (saturated). It appears that nylon-6 has a special affinity for water at low concentration levels where H2O molecules enter between the (C = O - H-N) chemical bonds between nylon molecular chains. As the water concentration increases beyond a critical level, nylon-6 specimens start trapping H2O molecules in other bond sites or potential wells. The trapped water increases the free volume in the test specimens and reduces Ps atom formation as well as its subsequent decay rate.

In the course of developing improved moisture-resistant epoxy resins, two different types of epoxy resins containing variable mole ratios of chromium ions per polymer repeat unit were developed. Positron annihilation characteristics have been investigated in these resins as a function of their metal ion content. In both cases, the presence of metal ions reduces the lifetime as well as the intensity of the long life component. The long life component intensity reduction is considerably more pronounced than the lifetime reduction. These results have been discussed in terms of increased unpaired electron density at Ps formation sites due to the presence of chromium ions in the matrix.

As expected, the lifetime values with magnetically analyzed positron beams in uniform targets are similar to those obtained with conventional positron sources. Positron lifetime measurements were made in two thermoplastic polyimide materials recently developed at Langley. The long component lifetime values in polyimidesulfone samples are 847 + or - 81 Ps (dry) and 764 + or - 91 Ps (saturated). The corresponding values in LARC thermoplastic imides are 1080 + or - 139 Ps (dry) and 711 + or - 96 Ps (saturated). Clearly, the presence of moisture has greater effect on positron lifetime in LARC thermoplastic imides than in the case of polyimidesulfones. This result is consistent with the photomicrographic observations made on frozen water saturated specimens of these materials.

Positron annihilation rates have been studied in polymers and graphite-polymer composites as a function of their moisture content. The annihilation rates have been found to increase linearly with increasing moisture content in epoxies and polyamides, whereas no definite trends have been observed in polyimides. These experimental results have been used as the basis for the calculation of moisture content of several polymeric test specimens. For example, the directly measured moisture content of a Kevlar specimen was 45.5 + or - 5.0% of saturation value, whereas the moisture content on the basis of the decrease in positron lifetime was calculated to be 46.5 + or - 3.5%. Similarly, the directly measured moisture content of a graphite-epoxy composite (55 v/o fiber) was 19.2 + or - 0.6% of saturation value as opposed to a calculated value of 16.0 + or - 5.0%.

The positron lifetime technique of monitoring absorbed moisture is applied to several composites, including graphite/polymides which are candidates for high-temperature (over 260 C) applications. The experimental setup is a conventional fast-slow coincidence system wherein the positron lifetime is measured with respect to a reference time determined by the detection of a nuclear gamma ray emitted simultaneously with the positron. From the experiments, a rate of change of positron mean lifetime per unit mass of water can be determined for each type of specimen. Positron lifetime spectra are presented for a graphite/polyimide composite and for a pure polyimide.

En este trabajo se estudió la variabilidad en la supervivencia del semen porcino de diferentes individuos al congelado-descongelado. En primer lugar, se empleó un sistema estandarizado de congelación ( pellets en CO2) para comprobar la existencia de individuos cuyo semen es relativamente resistente (llamados good frezers) o muy susceptible ( bad freezers) al proceso de congelación-descongelación. En segundo lugar, se evaluó el efecto de la tasa de congelación en la criosobrevivencia espermática entre individuos y entre eyaculados, por medio de criomicroscopía. Los resultados indican que la susceptibilidad entre individuos parece ser más importante que la tasa de congelación en la criosobrevivencia espermática. Se observó además, por medio de criomicroscopía con tinciones fluorescentes, que la membrana plasmática del espermatozoide se mantiene intacta durante el congelado pero al descongelado se manifiesta la ruptura de esta estructura. Diferencias en la composición química de la membrana espermática podrían explicar la variabilidad individual al congelado-descongelado.
