Nanoemulgel (NEG) pharmaceutical formulations are gaining popularity because of their ability to serve both as a nanoemulsion and as a gel. These products are well-known for their ease of use, spreadability, controlled release, and ability to hydrate dry skin. Natural essential oils have been shown to promote the cutaneous permeability of topical formulations, enhancing medication safety and efficacy. Herein, we developed NEG for the enhanced permeation of posaconazole against candidiasis using Mustard oil (Mustard oil-NEG), using the gelling agents carbopol 943 and hydroxypropyl methylcellulose (HPMC). We tested various excipients to increase the solubility of posaconazole and formulate a nanoemulsion (NE). We measured the NE droplet particle size, shape, entrapment efficiency, and drug release. Furthermore, the physicochemical properties of the optimized nanoemulsion formulation were characterized by techniques such as Fourier transform infrared (FT-IR) spectroscopy and X-ray diffraction (XRD) analysis. The NEs were loaded into gels to form NEGs. NEGs were characterized for drug content, homogeneity, rheology, spreadability, and antifungal activity against Candida albicans, both in vitro and in vivo. Optimized posaconazole NEG preparations consisted of either 15% Mustard oil or 20% Arachis oil. Droplet sizes in the optimized NEs were <100 nm, and the polydispersity indexes were 0.24 and 0.26. The percentages of posaconazole released after 24 h from the clove-oil-NEG and eucalyptus-oil-NEGs were 91 ± 4.5 and 89 ± 7%, respectively. Scanning electron microscopy (SEM) showed that the NEGs had a smooth, uniform, and consistent shape and internal structural organization. The drug contents in the clove-oil-NEG and eucalyptus-oil-NEG were 98.5 ± 2.2 and 98.8 ± 3.4%, respectively. Permeation values of posaconazole from clove-oil-NEG and eucalyptus-oil-NEG were 117 ± 7 and 108.34 ± 6 µg cm-2, respectively. The posaconazole NEG formulations also had higher levels of fungal growth inhibition than a marketed formulation. Finally, in vivo studies showed that the NEGs do not irritate the skin. Posaconazole NEG with either 15% Mustard oil or 20% Arachis oil is stable with better efficacy than posaconazole alone due to excellent dispersion, drug dissolution, and permeability and thus might be recommended for the effective and safe treatment of candidiasis. The limited water solubility and dissolution of drugs classified under the Biopharmaceutics Classification System (BCS) class II category provide constraints on their oral bioavailability. In the present study, a self-micro emulsifying drug delivery system (SMEDDS) was employed to enhance the aqueous solubility and oral bioavailability of toremifene, which belongs to the Biopharmaceutics Classification System (BCS) class II category. The selection of Oleic Acid, Transcutol-P, and Span 20 as the oil, surfactant combination, and cosurfactant, respectively, was based on the solubility. The optimal composition (Oleic Acid / Transcutol-P / Span 20 in a volume ratio) for toremifene, which exhibits a small droplet size (132.1 nm) and consistent microemulsification was determined by the use of pseudo-ternary phase diagrams. Differential scanning calorimetry and powder X-ray diffraction were employed to identify the loss of crystallinity in lyophilized toremifene. The results indicated that toremifene existed in a molecularly dissolved or amorphous state inside the SMEDDS formulation. The saturation solubility and dissolving rate of toremifene -SMEDDS exhibited significant enhancements compared to toremifene powder when tested in water, simulated gastric fluid (pH 1.2), and simulated intestinal fluid (pH 6.8). The female rats exhibited a 1.94-fold increase in the area under the curve and a 1.80-fold increase in the highest plasma concentration following the oral administration of toremifene-SMEDDS compared to the oral administration of the toremifene dispersion. Present is a low-cost and environmentally beneficial approach for producing silver nanoparticles from discarded pineapple leaves utilising microwave-assisted extraction. AgNPs are produced environmentally friendly using a straightforward reduction technique and silver nitrate. The ideal conditions for green synthesis are determined by taking into account a number of factors, such as the silver nitrate concentration (5–25 mM), the incubation period (2–24 h), and the sample volume (2–8 mL). The reduction of silver metal ions into silver nanoparticles is indicated by the appearance of an absorption band in the UV-Vis spectro-photometer between 400 and 500 nm. Longer run times, higher silver nitrate solution concentrations, and larger sample volumes are all observed to improve absorption intensity. The green synthesised silver nanoparticles range in size from 40 to 150 nm and exhibit hexagonal spherical shapes, according to FE-SEM studies. In order to achieve increased felodipine delivery via transdermal delivery, the study investigated the parameters for improving various membrane combinations that incorporate lecithin derived from soy and eggs, in addition to an edge activator. Background: Urolithiasis is a medical condition that, despite substantial research in the field of urology, has yet to find a cure within the allopathic medical approach. The process of stone development, known as nephrolithiasis, can occur within the kidney or any segment of the urinary tract, encompassing the ureters and bladder. Material and Methods: The seeds of Cassia auriculata (Linn.) were obtained in June 2021 from Mettukadai hamlet, located in the Erode District of Tamilnadu, India. The herbarium of the plant was meticulously assembled, verified, and afterwards deposited as a voucher specimen. The voucher specimen was retained within the college premises for future reference. Results: The current investigation involved conducting a preliminary phytochemical analysis on the ethanolic seed extract of Cassia auriculata Linn. The research revealed the existence of many phytochemical constituents, including Alkaloids, Flavanoids, Carbohydrate, Sterols, Phytosterols, Phenols, Terpenoids, Amino acids, and Anthraquinones. The acute toxicity experiments revealed that rats exhibited tolerance to a maximum dose of 2000 mg/kg body weight, and no discernible alterations in behavior were detected across all experimental groups. Hence, fractions equivalent to one-fourth and one-eighth of the maximum tolerated dose of 200 mg/kg body weight were selected for subsequent investigations. Conclusion: In summary, the findings suggest that the application of an ethanolic seed extract derived from Cassia auriculata Linn effectively decreased the progression of urinary stone formation.
