A polyvinylimidazole-based cryogel is presented as a pioneering solution for efficient extraction and release of partially water-soluble polyphenols from olive byproducts. Specifically, oleuropein was used as model molecule to evaluate its recovery from water. The material merges the properties of interconnected cryogel structure in adsorbing molecules via fast diffusion flux, with the strong electrostatic interactions acted by imidazole moiety. Such cryogel achieves effective oleuropein binding likely through hydrogen bonding and π–π interactions. Comprehensive assessments of static adsorption kinetics, isotherms, and desorption kinetics underscore the cryogel's efficacy in oleuropein extraction and release, highlighting its pivotal role in valorizing olive wastewater through sustainable biotechnological applications. Advances in materials science and technology have prompted researchers to look to nature for new high-performance, low-cost materials. Among these, cyclodextrins have been widely used as a material in industrial applications. Inspired by previous work by our research group that led to the functionalization of cucurbit[6]uryl and its conversion into supramolecular nanospheres with good CO2 adsorption capacity, this work aims to improve the ability of cyclodextrins to capture CO2 by functionalizing them with amide groups. Carbon dioxide adsorption experiments on functionalized cyclodextrins showed an adsorption capacity similar to that of BEA zeolite, a material currently used in the industry for gas adsorption. Moreover, these adsorption properties could also be exploited to improve the adsorption capacity of drugs, a field in which cyclodextrins are widely used. The new cyclodextrin molecules were characterized by nuclear magnetic resonance spectroscopy and mass spectrometry, thanks to which we could determine the degree of functionalization of the new macrocycles. In addition, using Fourier-transform infrared spectroscopy, we demonstrated the presence and interaction of carbon dioxide adsorbed by the material, whereas an in silico study confirmed the chemisorption as the principal adsorption process, as experimentally inferred using the pseudo-second-order (PSO) kinetic model. Fatty acid binding protein (FABP4) inhibitors are of synthetic and therapeutic interest and ongoing clinical studies indicate that they may be a promise for the treatment of cancer, as well as other diseases. As part of a broader research effort to develop more effective FABP4 inhibitors, we sought to identify new structures through a two-step computing assisted molecular design based on the established scaffold of a co-crystallized ligand. Novel and potent FABP4 inhibitors have been developed using this approach and herein we report the synthesis, biological evaluation and molecular docking of the 4-amino and 4-ureido pyridazinone-based series. Herein, a proof of concept is reported, concerning the formulation of methacrylic and styrene-based hydrogels with adsorption properties versus the emerging pollutants (dyes, pesticides, and pharmaceuticals). The synthesis of the hydrogels was achieved by functionalizing the HEMA monomer with specifically chelating groups, such as amino acids (lysine and histidine), cyclodextrins, and meglumine. The as-prepared monomers were polymerized in water at different temperatures to obtain macroporous samples. All the samples were characterized by chemical–physical and morphological analyses confirming the success of the reactions. The resulting systems were successfully tested to adsorb 2, 4 D, methylene blue, and lomefloxacin. In addition, up to five cycles of regeneration tests were performed, confirming the aptitude of the samples to be used several times without losing efficiency and maintaining their mechanical properties. Cisplatin-based chemotherapy is the standard treatment of choice for head and neck squamous cell carcinoma (HNSCC). The efficiency of platinum-based therapies is directly influenced by the development of tumor resistance. Multiple signaling pathways have been linked to tumor resistance, including activation of nuclear factor kappa B (NFκB). We explore a novel mechanism by which NFκB drives HNSCC resistance through histone modifications. Post-translational modification of histones alters chromatin structure, facilitating the binding of nuclear factors that mediate DNA repair, transcription, and other processes. We found that chemoresistant HNSCC cells with active NFκB signaling respond to chemotherapy by reducing nuclear BRCA1 levels and by promoting histone deacetylation (chromatin compaction). Activation of this molecular signature resulted in impaired DNA damage repair, prolonged accumulation of histone γH2AX and increased genomic instability. We found that pharmacological induction of histone acetylation using HDAC inhibitors prevented NFκB-induced cisplatin resistance. Furthermore, silencing NFκB in HNSCC induced acetylation of tumor histones, resulting in reduced chemoresistance and increased cytotoxicity following cisplatin treatment. Collectively, these findings suggest that epigenetic modifications of HNSCC resulting from NFκB-induced histone modifications constitute a novel molecular mechanism responsible for chemoresistance in HNSCC. Therefore, targeted inhibition of HDAC may be used as a viable therapeutic strategy for disrupting tumor resistance caused by NFκB. [ABSTRACT FROM AUTHOR]

Bacterial involvement in cancer's development, along with their impact on therapeutic interventions, has been increasingly recognized. This has prompted the development of novel strategies to disrupt essential biological processes in microbial cells. Among these approaches, metal-chelating agents have gained attention for their ability to hinder microbial metal metabolism and impede critical reactions. Nanotechnology has also contributed to the antibacterial field by offering various nanomaterials, including antimicrobial nanoparticles with potential therapeutic and drug-delivery applications. Halloysite nanotubes (HNTs) are naturally occurring tubular clay nanomaterials composed of aluminosilicate kaolin sheets rolled multiple times. The aluminum and siloxane groups on the surface of HNTs enable hydrogen bonding with biomaterials, making them versatile in various domains, such as environmental sciences, wastewater treatment, nanoelectronics, catalytic studies, and cosmetics. This study aimed to create an antibacterial material by combining the unique properties of halloysite nanotubes with the iron-chelating capability of kojic acid. A nucleophilic substitution reaction involving the hydroxyl groups on the nanotubes' surface was employed to functionalize the material using kojic acid. The resulting material was characterized using infrared spectroscopy (IR), thermogravimetric analysis (TGA), energy-dispersive X-ray spectroscopy (EDX), and scanning electron microscopy (SEM), and its iron-chelating ability was assessed. Furthermore, the potential for drug loading-specifically, with resveratrol and curcumin-was evaluated through ultraviolet (UV) analysis. The antibacterial assay was evaluated following CLSI guidelines. The results suggested that the HNTs-kojic acid formulation had great antibacterial activity against all tested pathogens. The outcome of this work yielded a novel bio-based material with dual functionality as a drug carrier and an antimicrobial agent. This innovative approach holds promise for addressing challenges related to bacterial infections, antibiotic resistance, and the development of advanced therapeutic interventions.

In this work, we report on the in vitro and in vivo pharmacological properties of LP1 analogs to complete the series of structural modifications aimed to generate compounds with improved analgesia. To do that, the phenyl ring in the N -substituent of our lead compound LP1 was replaced by an electron-rich or electron-deficient ring and linked through a propanamide or butyramide spacer at the basic nitrogen of the (-)- cis - N -normetazocine skeleton. In radioligand binding assays, compounds 3 and 7 were found to display nanomolar binding affinity for the μ opioid receptor (MOR) (K i = 5.96 ± 0.08 nM and 1.49 ± 0.24 nM, respectively). In the mouse vas deferens (MVD) assay, compound 3 showed an antagonist effect against DAMGO ([D-Ala 2 , N-MePhe 4 , Gly-ol]-enkephalin), a highly selective MOR prototype agonist, whereas compound 7 produced naloxone reversible effect at MOR. Moreover, compound 7 , as potent as LP1 and DAMGO at MOR, was able to reduce thermal and inflammatory pain assessed by the mouse tail-flick test and rat paw pressure thresholds (PPTs) measured by a Randall-Selitto test.

The data have been obtained from FABP4 inhibitor molecules previously published. The 120 compounds were used to build a 3D-QSAR model. The development of the QSAR model has been undertaken with the use of Forge software using the PM3 optimized structure and the experimental IC 50 of each compound. The QSAR model was also employed to predict the activity of 3000 new isosteric derivatives of BMS309403. The isosteric replacement was also validated by the synthesis and the biological screening of three new compounds reported in the related research article "3D-QSAR assisted identification of FABP4 inhibitors: An effective scaffold hopping analysis/QSAR evaluation" (Floresta et al., 2019).
