Abstract This study aimed to elucidate the influence of miR-483-3p on human renal tubular epithelial cells (HK-2) under high glucose conditions and to understand its mechanism. Human proximal tubular epithelial cells (HK-2) were exposed to 50 mmol/L glucose for 48 h to establish a renal tubular epithelial cell injury model, denoted as the high glucose group (HG group). Cells were also cultured for 48 h in a medium containing 5.5 mmol/L glucose, serving as the low glucose group. Transfection was performed in various groups: HK-2 + low glucose (control group), high glucose (50 mM) (HG group), high glucose + miR-483-3p mimics (HG + mimics group), high glucose +miR-483-3p inhibitor (HG + inhibitor group), and corresponding negative controls. Real-time quantitative polymerase chain reaction (qPCR) assessed the mRNA expression of miR-483-3p, bax, bcl-2, and caspase-3. Western blot determined the corresponding protein levels. Proliferation was assessed using the CCK-8 assay, and cell apoptosis was analyzed using the fluorescence TUNEL method. Western blot and Masson’s staining were conducted to observe alterations in cell fibrosis post miR-483-3p transfection. Furthermore, a dual-luciferase assay investigated the targeting relationship between miR-483-3p and IGF-1. The CCK8 assay demonstrated that the HG + mimics group inhibited HK-2 cell proliferation, while the fluorescent TUNEL method revealed induced cell apoptosis in this group. Conversely, the HG + inhibitor group promoted cell proliferation and suppressed cell apoptosis. The HG + mimics group upregulated mRNA and protein expression of pro-apoptotic markers (bax and caspase-3), while downregulating anti-apoptotic marker (bcl-2) expression. This study presents the case of a 40-year-old male patient after renal transplantation. The CT scan revealed a large mass in the lower abdomen and pelvis, with a branch of the right external iliac artery intersecting the growth. After a comprehensive examination, it was shown that the mass originated from the transplanted kidney, and a radical nephrectomy (including the mass) was performed. We document a case of atypical angiomyolipoma (AAM) occurring in a transplanted kidney. This article reports the case study and a brief literature review of the clinical presentation, diagnosis and treatment of AAM.

In the Junggar Basin plague focus, Rhombomys opimus and Meriones meridianus live together, and their parasitic fleas have a similar community structure. However, R. opimus has significantly higher positive rates of Yersinia pestis and anti-F1 antibody compared with M. meridianus. In this study, Y. pestis- and antiF1 antibody-negative R. opimus and M. penicilliger were collected in Qitai county, Fukang city and Mulei county of the eastern part of the Junggar Basin. The genomic DNA was extracted from their spleen tissues, and RT1-Db1*exon1 was amplified through PCR procedure and then sequenced. Sequence analysis was performed and molecular diversity parameters were calculated and compared. The results showed that there were significant differences in nucleotide composition, amino acid composition, number and distribution of single nucleotide polymorphism (SNP) sites and number of haplotypes between R. opimus and M. penicilliger. The nucleotide diversity (π) for R. opimus was 0.00420 ± 0.00139, the haplotype diversity (h) was 0.833 ± 0.086, and the average number of nucleotide differences (K) was 2.02564. The π for M. penicilliger was 0.06569 ± 0.02524, and the h was 1.000 ± 0.045, and the K was 10.4444. The fixation index (FST) value between R. opimus and M. penicilliger was 0.9207. Furthermore, the FST value within R. opimus (0.0275) was significantly lower than that within M. penicilliger (0.2106), indicating a greater genetic variation of M. penicilliger compared with R. opimus. In conclusion, the genetic diversity analysis based on RT1-Db1*exon1 showed that M. penicilliger had higher gene polymorphism and greater genetic differentiation compared with R. opimus in the Junggar Basin plague focus, which might be associated with the low infection rate of Y. pestis.

IntroductionThe precise associations between temperature-related indices and mental and behavioral disorders (MBDs) have yet to be fully elucidated. Our study aims to ascertain the most effective temperature-related index and assess its immediate impact on emergency ambulance dispatches (EADs) due to MBDs in Shenzhen, China.MethodsEADs data and meteorological data from January 1, 2013, to December 31, 2020, in Shenzhen were collected. Distributed lag non-linear models (DLNMs) were utilized to examine the non-linear and lagged effects of temperature-related indices on EADs due to MBDs. The Quasi Akaike Information criterion (QAIC) was used to determine the optimal index after standardizing temperature-related indices. After adjusting for confounding factors in the model, we estimated the immediate and cumulative effects of temperature on EADs due to MBDs.ResultsThe analysis of short-term temperature effects on EADs due to MBDs revealed Humidex as the most suitable index. Referring to the optimal Humidex (3.2th percentile, 12.00°C), we observed a significant effect of Humidex over the threshold (34.6th percentile, 26.80°C) on EADs due to MBDs at lag 0–5. The cumulative relative risks for high temperature (90th percentile, 41.90°C) and extreme high temperature (99th percentile, 44.20°C) at lag 0–5 were 1.318 (95% CI: 1.159–1.499) and 1.338 (95% CI: 1.153–1.553), respectively. No significant cold effect was observed on EADs due to MBDs.ConclusionHigh Humidex was associated with more EADs due to MBDs in subtropical regions. Health authorities should implement effective measures to raise public awareness of risks related to high temperature and protect vulnerable populations.

The condition and hazard monitoring of bridges play important roles in ensuring their service continuity not only throughout their entire lifespan but also under extreme conditions such as those of earthquakes. Advanced structural health monitoring (SHM) systems using vision-based technology, such as surveillance, traffic, or drone cameras, may assist in preventing future impacts due to structural deficiency and are critical to the emergence of sustainable and smart transportation infrastructure. This study evaluates several feature detection and tracking algorithms and implements them in the vision-based SHM of bridges along with their systematic procedures. The proposed procedures are implemented via a two-span accelerated bridge construction (ABC) system undergoing a large-scale shake-table test. The research objectives are to explore the effect of refined matching algorithms on blob-based features in improving their accuracies and to implement the proposed algorithms on large-scale bridges tested under seismic loads using vision-based SHM. The procedure begins by adopting blob-based feature detectors, i.e., the scale-invariant feature transform (SIFT), speeded-up robust features (SURF), and KAZE algorithms, and their stability is compared. The least medium square (LMEDS), least trimmed square (LTS), random sample consensus (RANSAC), and its generalization maximum sample consensus (MSAC) algorithms are applied for model fitting, and their sensitivity for removing outliers is analyzed. The raw data are corrected using mathematical models and scaled to generate displacement data. Finally, seismic vibrations of the bridge are generated, and the seismic responses are compared. The data are validated using target-tracking methods and mechanical sensors, i.e., string potentiometers. The results show a good agreement between the proposed blob feature detection and matching algorithms and target-tracking data and reference data obtained using mechanical sensors.

Abstract Background Low-grade gliomas (LGG) are a type of brain tumor that can be lethal, and it is essential to identify genes that are correlated with patient prognosis. In this study, we aimed to use CRISPR-cas9 screening data to identify key signaling pathways and develop a genetic signature associated with high-risk, low-grade glioma patients. Methods The study used CRISPR-cas9 screening data to identify essential genes correlated with cell survival in LGG. We used RNA-seq data to identify differentially expressed genes (DEGs) related to cell viability. Moreover, we used the least absolute shrinkage and selection operator (LASSO) method to construct a genetic signature for predicting overall survival in patients. We performed enrichment analysis to identify pathways mediated by DEGs, overlapping genes, and genes shared in the Weighted correlation network analysis (WGCNA). Finally, the study used western blot, qRT-PCR, and IHC to detect the expression of hub genes from signature in clinical samples. Results The study identified 145 overexpressed oncogenes in low-grade gliomas using the TCGA database. These genes were intersected with lethal genes identified in the CRISPR-cas9 screening data from Depmap database, which are enriched in Hippo pathways. A total of 19 genes were used to construct a genetic signature, and the Hippo signaling pathway was found to be the predominantly enriched pathway. The signature effectively distinguished between low- and high-risk patients, with high-risk patients showing a shorter overall survival duration. Differences in hub gene expression were found in different clinical samples, with the protein and mRNA expression of REP65 being significantly up-regulated in tumor cells. The study suggests that the Hippo signaling pathway may be a critical regulator of viability and tumor proliferation and therefore is an innovative new target for treating cancerous brain tumors, including low-grade gliomas. Conclusion Our study identified a novel genetic signature associated with high-risk, LGG patients. We found that the Hippo signaling pathway was significantly enriched in this signature, indicating that it may be a critical regulator of tumor viability and proliferation in LGG. Targeting the Hippo pathway could be an innovative new strategy for treating LGG.

Ecosystems in arid areas are under pressure from human activities and the natural environment. Long-term monitoring and evaluation of arid ecosystems are essential for achieving the goal of sustainable development. The Xinjiang Uygur Autonomous Region (Xinjiang) is a typical arid region located in Northwest China with a relatively sensitive ecosystem. Under the support of the Google Earth Engine (GEE) cloud platform’s massive data collection, the remote sensing ecological index (RSEI) from 2000 to 2020, both in summer and spring, is established, and the variation trend of the ecological quality in Xinjiang is evaluated by coefficient of variation (CV), Sen’s slope analysis, Mann–Kendall trend test (M–K test) and Hurst index. In addition, a partial correlation analysis is processed between RSEI and selected climatic factors, including precipitation and temperature, to find out the mode of correlation between ecological quality and the natural climate. In the last two decades the following has become apparent: (1) The RSEI values of Xinjiang have been relatively low and unstable both in summer and spring, with a trend toward increasing; (2) The distribution characteristics of RSEI levels both in summer and spring have been similar; low levels were concentrated in the desert and wilderness, while high levels were concentrated around the oasis; (3) The ecological quality in Xinjiang has been relatively stable, with a trend of sustained increase both in summer and spring. There was also a small area of sustained decrease around the Junggar Basin and Turpan Basin in summer and a small area of significant decrease in the center of the Taklamakan Desert in spring; (4) In summer, the precipitation has obviously positively correlated in the Southwest. The temperature has obviously positively correlated in the northwestern part; in spring, the precipitation has obviously positively correlated in the Western part; the temperature has obviously positively correlated in the oasis around the Yili River Basin and Tarim Basin.

Abstract Diabetic retinopathy attracts considerable research interest due to the number of diabetic patients increasing rapidly in recent years. Diabetic retinopathy is a common symptom of retinopathy, which damages the patient's eyesight and even causes the patient to lose sight. The authors propose a novel attention mechanism named block attention mechanism to actively explore the role of attention mechanisms in recognizing retinopathy features. Specifically, the block attention mechanism contributions are as follows: (1) The relationship between the blocks in the entire feature map is explored, and the corresponding coefficients are assigned to different blocks to highlight the importance of blocks. (2) Furthermore, the relationship between the edge elements of the feature map and the edge elements is explored, and corresponding coefficients are assigned to the elements at different positions on the feature map to highlight the importance of the elements in the feature map. Experimental results show that the proposed framework outperforms the existing popular attention‐based baselines on two public retina datasets, OCT2017 and SD‐OCT, achieving a 99.64% and 96.54% accuracy rate, respectively.

Transition-metal-doped clusters have long been attracting great attention due to their unique geometries and interesting physical and/or chemical properties. In this paper, the geometries of the lowest- and lower-energy CoKn (n = 2–12) clusters have been screened out using particle swarm optimization and first principles relaxation. The results show that except for CoK2 the other CoKn (n = 3–12) clusters are all three-dimensional structures, and CoK7 is the transition structure from which the lowest energy structures are cobalt atom-centered cage-like structures. The stability, the electronic structures, and the magnetic properties of CoKn clusters (n = 2–12) clusters are further investigated using the first principles method. The results show that the medium-sized clusters whose geometries are cage-like structures are more stable than smaller-sized clusters. The electronic configuration of CoKn clusters could be described as 1S1P1D according to the spherical jellium model. The main components of petal-shaped D molecular orbitals are Co-d and K-s states or Co-d and Co-s states, and the main components of sphere-like S molecular orbitals or spindle-like P molecular orbitals are K-s states or Co-s states. Co atoms give the main contribution to the total magnetic moments, and K atoms can either enhance or attenuate the total magnetic moments. CoKn (n = 5–8) clusters have relatively large magnetic moments, which has a relation to the strong Co-K bond and the large amount of charge transfer. CoK4 could be a magnetic superatom with a large magnetic moment of 5 μB.

In a software development project, aspects of software quality are fundamental; all stakeholders expect high-quality software. To ensure the quality of software products, it is necessary to ensure the software quality process. A software process is essential to be assessed from their quality. In the software development process, the developer needs guidance in carrying out every aspect of it. The goals to achieve and the procedure to measure for each aspect's goals performance must be determined. One method that can be used is the Extended Goal Question Metric method. This method determines what aspects must be achieved for each development process. A few goals to measure are defined for each aspect. For each goal, one or more goals determine one or more relevant questions. For each question, an appropriate metric is determined. The next step is mapping between G to Q and Q to M. The measurement was conducted by calculating the goal value obtained from the metric calculation. From this metric, each goal's value could be obtained, whether it is achieved or not. The tests were carried out on the software process to develop the academic Directorate of Technology and Information System Development of Institut Teknologi Sepuluh Nopember Surabaya, Indonesia. Each goal's value exceeded 0.51 (for a scale of 0-1), which achieved the Software development process's quality. The total average score was 0.889.

Tetrahydroisoquinolines have been widely investigated for the treatment of arrhythmias. 1−(3′−bromophenyl)−heliamine (BH), an anti−arrhythmias agent, is a synthetic tetrahydroisoquinoline. This study focuses on the pharmacokinetic characterization of BH, as well as the identification of its metabolites, both in vitro and in vivo. A UHPLC−MS/MS method was developed and validated to quantify BH in rat plasma with a linear range of 1–1000 ng/mL. The validated method was applied to a pharmacokinetic study in rats. The maximum concentration Cmax (568.65 ± 122.14 ng/mL) reached 1.00 ± 0.45 h after oral administration. The main metabolic pathways appeared to be phase-I of demethylation, dehydrogenation, and epoxidation, and phase II of glucuronide and sulfate metabolites. Finally, a total of 18 metabolites were characterized, including 10 phase I metabolites and 8 phase II metabolites. Through the above studies, we have gained a better understanding of the absorption and metabolism of BH in vitro and in vivo, which will provide us with guidance for future in-depth studies on this compound.

Acid secretion inhibitors, such as proton pump inhibitors (PPIs) and potassium competitive acid blockers (PCABs), are used to treat ulcers after endoscopic submucosal dissection (ESD) for early gastric cancer. These drugs can influence serum gastrin and pepsinogen (PG) levels; however, their definite effects remain unclear. This open-label, randomized study investigated the effect of acid secretion inhibitors on the serum gastrin and pepsinogen levels. In total, 76 patients were enrolled in the study. They underwent gastric ESD and received a PPI (n=21) or PCAB (n=55). Changes in the serum gastrin and PG levels before and 4 weeks after administration were examined. Patient factors associated with the alteration of serum PG or gastrin levels were identified. The median serum levels of gastrin, PGI, and PGII before the administration of the acid secretion inhibitors were 110.5 pg/mL, 36.4 ng/mL, and 8.9 ng/mL, respectively; after administration, the levels increased to 300 pg/mL, 64.7 ng/mL, and 15.8 ng/mL, respectively (P

Abstract Background CYP2A6 is an enzyme involved in oxidation of a number of environmental chemicals, including nicotine, pro-carcinogenic nitrosamines and polycyclic aromatic hydrocarbons (PAHs). The whole gene deletion of CYP2A6 (CYP2A6*4) is prevalent in East Asian population. Whether or not CYP2A6*4 associates with cancer is still controversial. Methods We undertook an association study to determine whether deletion of CYP2A6 gene associates with total cancer and major cancer types employing data of consecutive autopsy cases registered in the Japanese single-nucleotide polymorphisms for geriatric research (JG-SNP) database. The presence of cancer were inspected at the time of autopsy and pathologically confirmed. Genotyping for CYP2A6 wild type (W) and deletion (D) was done by allele specific RT-PCR method. Results Among 1373 subjects, 826 subjects (60.2%) were cancer positive and 547 subjects (39.8%) were cancer negative. The genotype frequency in the whole study group for WW, WD and DD were 65.0, 30.6 and 4.4%, respectively, which obeyed the Hardy-Weinberg equilibrium (p = 0.20). Total cancer presence, as well as major cancers including gastric, lung, colorectal, and blood cancers did not show any positive association with CYP2A6 deletion. When male and female were separately analyzed, CYP2A6 deletion associated with decreased gastric cancer risk in female (OR = 0.49, 95%CI: 0.25–0.95, p = 0.021, after adjustment for age, smoking and drinking). When smoker and non-smoker were separately analyzed, CYP2A6 deletion associated with decreased total cancer in female nonsmokers (OR = 0.67, 95%CI: 0.45–0.99, p = 0.041 after adjustment). On the other hand, CYP2A6 deletion associated increase blood cancers in smokers (OR = 2.05, 95%CI: 1.19–3.53, p = 0.01 after adjustment). Conclusion The CYP2A6 deletion may not grossly affect total cancer. It may associate with individual cancers in sex and smoking dependent manner. Further studies with larger sample size are warranted to confirm our results.

Abstract Background A link between lipid metabolism and disease has been recognized in cats. Since hepatic lipidosis is a frequent disorder in cats, the aim of the current study was to evaluate liver and plasma lipid dimorphism in healthy cats and the effects of gonadectomy on lipid profiling. From six female and six male cats plasma and liver lipid profiles before and after spaying/neutering were assessed and compared to five cats (three neutered male and two spayed female) diagnosed with hepatic lipidosis. Results Intact female cats had a significantly lower level of plasma triacylglycerides (TAG) and a higher liver level of the long chain polyunsaturated fatty acid arachidonic acid (AA) compared to their neutered state. Both male and female cats with lipidosis had a higher liver, but not plasma TAG level and an increased level of plasma and liver sphingomyelin compared to the healthy cats. Conclusion Although lipid dimorphism in healthy cats resembles that of other species, intact female cats show differences in metabolic configuration that could predispose them to develop hepatic lipidosis. The increased sphingomyelin levels in cats with lipidosis could suggest a potential role in the pathogenesis of hepatic lipidosis in cats.

Pentacyclic triterpenoids are one of the main functional components in Dracocephalum heterophyllum. In this study the optimal process, the fairly simple and accessible extraction and purification of triterpenoids of D. heterophyllum, was developed by a remaceration method. Remaceration is characterized by minimal loss of biologically active compounds on diffusion, which contributes to the largest depletion of raw materials. The triterpenoid yield was 2.4% under optimal conditions which was enhanced to 98.03% after purification. The triterpenoid profiles and their anticancer and antidiabetic activities were further analyzed. GC-MS analysis of triterpenoidal extract of D. heterophyllum resulted ursolic acid (71.9%) and oleanolic acid (18.1%) as the major components. Additionally, total purified triterpenoid contents of D. heterophyllum and its main components were shown to possess significant cytotoxic activity against three human breast cancer cell lines (SK-Br-3, T47D, and MCF-7). The purification of triterpenoids influenced their biological activity. The antidiabetic effect, as measured by inhibition of protein-tyrosine phosphatase (PTP-1B), of the purified fraction of triterpenoids of D. heterophyllum increased by five-fold against the enzyme. The results provide important guidance for the industrial application of D. heterophyllum confirming the prospect of developing plant extracts into effective drugs and health foods for human applications.

Hepatic stellate cell (HSC) activation is a critical step in the development of chronic liver disease. During activation, HSCs lose their lipid droplets (LDs) containing triacylglycerol (TAG), cholesteryl esters (CEs), and retinyl esters (REs). Here we aimed to investigate which enzymes are involved in LD turnover in HSCs during activation in vitro. Targeted deletion of the Atgl gene in mice HSCs had little effect on the decrease of the overall TAG, CE, and RE levels during activation. However, ATGL-deficient HSCs specifically accumulated TAG species enriched in PUFAs and degraded new TAG species more slowly. TAG synthesis and levels of PUFA-TAGs were lowered by the diacylglycerol acyltransferase (DGAT)1 inhibitor, T863. The lipase inhibitor, Atglistatin, increased the levels of TAG in both WT and ATGL-deficient mouse HSCs. Both Atglistatin and T863 inhibited the induction of activation marker, α-smooth muscle actin, in rat HSCs, but not in mouse HSCs. Compared with mouse HSCs, rat HSCs have a higher turnover of new TAGs, and Atglistatin and the DGAT1 inhibitor, T863, were more effective. Our data suggest that ATGL preferentially degrades newly synthesized TAGs, synthesized by DGAT1, and is less involved in the breakdown of preexisting TAGs and REs in HSCs. Furthermore a large change in TAG levels has modest effect on rat HSC activation.

Semakin baik pelayanan gizi yang diberikan oleh rumah sakit, maka semakin baik pula standar akreditasi rumah sakit tersebut. Tujuan penelitian ini dilakukan untuk menganalisis beban kerja tenaga gizi di Instalasi Gizi RSUD Banjarbaru. Penelitian ini merupakan penelitian observasional yang bersifat deskriptif kualiatif. Populasi dalam penelitian ini adalah seluruh tenaga gizi di RSUD Banjarbaru yang berjumlah 9 orang. Analisis data disajikan dalam bentuk deskriptif. Hasil penelitian ini Penggunaan waktu produkif tenaga gizi di ruang rawat inap 60,6% sedangkan di ruang penyelenggaraan 94%, waktu kerja yang tersedia bagi tenaga gizi di ruang rawat inap sebesar 34,25 jam/minggu sedangkan di ruang penyelenggaraan makanan 35 jam/minggu. Standar beban kerja tenaga gizi pertahun menunjukkan setiap kegiatan membutuhkan waktu dalam setiap standar beban kerja sesuai kegiatan dari hari kerja yang tersedia. Proporsi waktu beban kerja tenaga gizi di ruang rawat inap kurang dari standar sedangkan di ruang penyelenggaraan makanan melebihi standar waktu produktif dan waktu kerja yang tersedia tenaga gizi belum sesuai. Diharapkan dapat melanjutkan penelitian selanjutnya untuk menentukan standar kelonggaran dan kebutuhan tenaga kerja di rumah sakit lain atau ditempat lainnya.

Chlorogenic acid (CGA), the ester formed between caffeic acid and l-quinic acid, is a widespread phenolic compound. It is part of the human diet, found in foods such as coffee, apples, pears, etc. CGA is also was widely used in cosmetics, but the effects of CGA on melanogenesis are unknown. In this study, we analyzed the effects of CGA on cell proliferation, melanin content and tyrosinase of B16 murine melanoma cells. Additionally, the enzymatic reactions of CGA in B16 melanoma cells lytic solution were detected by UV spectrophotometry. Results showed CGA at 30 and 60 μM significantly suppresses cell proliferation. 8-MOP at 100 μM significantly promotes cell proliferation, but CGA can counter this. Incubated for 24 h, CGA (500 μM) improves melanogenesis while suppressing tyrosinase activity in B16 melanoma cells or 8-methoxypsoralen (8-MOP) co-incubated B16 melanoma cells. After 12 h, B16 melanoma cell treatment with CGA leads to an increase in melanin accumulation, however, after 48 h there is a decrease in melanin production which correlates broadly with a decrease in tyrosinase activity. CGA incubated with lytic solution 24 h turned brown at 37 °C. The formation of new products (with a maximum absorption at 295 nm) is associated with reduction of CGA (maximum absorption at 326 nm). Therefore, CGA has its two sidesroles in melanogenesis of B16 melanoma cells. CGA is a likely a substrate of melanin, but the metabolic product(s) of CGA may suppress melanogenesis in B16 melanoma cells by inhibiting tyrosinase activity.

(1) Background: Almost 500 million people worldwide are suffering from diabetes. Since ancient times, humans have used medicinal plants for the treatment of diabetes. Medicinal plants continue to serve as natural sources for the discovery of antidiabetic compounds. Prangos pabularia Lindl. is a widely distributed herb with large reserves in Tajikistan. Its roots and fruits have been used in Tajik traditional medicine. To our best knowledge, there are no previously published reports concerning the antidiabetic activity and the chemical composition of the essential oil obtained from roots of P. pabularia. (2) Methods: The volatile secondary metabolites were obtained by hydrodistillation from the underground parts of P. pabularia growing wild in Tajikistan and were analyzed by gas chromatography (GC) and gas chromatography-mass spectrometry (GC-MS). Protein tyrosine phosphatase 1B (PTP-1B) inhibition assay and molecular docking analysis were carried out to evaluate the potential antidiabetic activity of the P. pabularia essential oil. (3) Results: The main constituents of the volatile oil of P. pabularia were 5-pentylcyclohexa-1,3-diene (44.6%), menthone (12.6%), 1-tridecyne (10.9%), and osthole (6.0%). PTP-1B inhibition assay of the essential oil and osthole resulted in significant inhibitory activity with an IC50 value of 0.06 ± 0.01 and 0.93 ± 0.1 μg/mL. Molecular docking analysis suggests volatile compounds such as osthole inhibit PTP-1B, and the results are also in agreement with experimental investigations. (4) Conclusions: Volatile secondary metabolites and the pure isolated compound (osthole) from the roots of P. pabularia exhibited potent antidiabetic activity, twenty-five and nearly two times more than the positive control (3-(3,5-dibromo-4-hydroxybenzoyl)-2-ethylbenzofuran-6-sulfonic acid-(4-(thiazol-2-ylsulfamyl)-phenyl)-amide)) with an IC50 value of 1.46 ± 0.4 μg/mL, respectively.

Background: Central Asia is the center of origin and diversification of the Artemisia genus. The genus Artemisia is known to possess a rich phytochemical diversity. Artemisinin is the shining example of a phytochemical isolated from Artemisia annua, which is widely used in the treatment of malaria. There is great interest in the discovery of alternative sources of artemisinin in other Artemisia species. Methods: The hexane extracts of Artemisia plants were prepared with ultrasound-assisted extraction procedures. Silica gel was used as an adsorbent for the purification of Artemisia annua extract. High-performance liquid chromatography with ultraviolet detection was performed for the quantification of underivatized artemisinin from hexane extracts of plants. Results: Artemisinin was found in seven Artemisia species collected from Tajikistan. Content of artemisinin ranged between 0.07% and 0.45% based on dry mass of Artemisia species samples. Conclusions: The artemisinin contents were observed in seven Artemisia species. A. vachanica was found to be a novel plant source of artemisinin. Purification of A. annua hexane extract using silica gel as adsorbent resulted in enrichment of artemisinin.

Plants or plant-derived products have been routinely used in several traditional medicine systems for vitiligo treatment. It is well-known that melanogenesis can be promoted by certain flavonoid compounds isolated from the traditional Uyghur medicinal plant, Kaliziri. Therefore, Chalcones, one class of flavonoid compounds, has become an interesting target for the development of anti-vitiligo agents. A series of novel isoxazole chalcone derivatives have been designed, synthesized, and evaluated for biological activities by our group. Among them, derivative 1-(4-((3-phenylisoxazol-5-yl)methoxy)phenyl)-3-phenylprop-2-en-1-one (PMPP) was identified as a potent tyrosinase activator with better activity and lower toxicity than the positive control 8-methoxypsoralen (8-MOP) in this study. Further investigations revealed that Akt and GSK3β were the signaling pathways involved in the hyperpigmentation of PMPP. Overall, these studies may provide a convenient and novel approach for the further development of anti-vitiligo agents.

The root of Geranium collinum Steph is known in Tajik traditional medicine for its hepatoprotective, antioxidant, and anti-inflammatory therapeutic effects. The present study was conducted to evaluate of potential antidiabetic, antioxidant activities, total polyphenolic and flavonoid content from the different extracts (aqueous, aqueous-ethanolic) and individual compounds isolated of the root parts of G. collinum. The 50% aqueous-ethanolic extract possesses potent antidiabetic activity, with IC50 values of 0.10 μg/mL and 0.09 μg/mL for the enzymes protein-tyrosine phosphatase (1B PTP-1B) and α-glucosidase, respectively. Phytochemical investigations of the 50% aqueous-ethanolic extract of G. collinum, led to the isolation of ten pure compounds identified as 3,3′,4,4′-tetra-O-methylellagic acid (1), 3,3′-di-O-methylellagic acid (2), quercetin (3), caffeic acid (4), (+)-catechin (5), (–)-epicatechin (6), (–)-epigallocatechin (7), gallic acid (8), β-sitosterol-3-O-β-d-glucopyranoside (9), and corilagin (10). Their structures were determined based on 1D and 2D NMR and mass spectrometric analyses. Three isolated compounds exhibited strong inhibitory activity against PTP-1B, with IC50 values below 0.9 μg/mL, more effective than the positive control (1.46 μg/mL). Molecular docking analysis suggests polyphenolic compounds such as corilagin, catechin and caffeic acid inhibit PTP-1B and β-sitosterol-3-O-β-d-gluco-pyranoside inhibits α-glucosidase. The experimental results suggest that the biological activity of G. collinum is related to its polyphenol contents. The results are also in agreement with computational investigations. Furthermore, the potent antidiabetic activity of the 50% aqueous-ethanolic extract from G. collinum shows promise for its future application in medicine. To the best of our knowledge, we hereby report, for the first time, the antidiabetic activity of G. collinum.

The condition and hazard monitoring of bridges play important roles in ensuring their service continuity not only throughout their entire lifespan but also under extreme conditions such as those of earthquakes. Advanced structural health monitoring (SHM) systems using vision-based technology, such as surveillance, traffic, or drone cameras, may assist in preventing future impacts due to structural deficiency and are critical to the emergence of sustainable and smart transportation infrastructure. This study evaluates several feature detection and tracking algorithms and implements them in the vision-based SHM of bridges along with their systematic procedures. The proposed procedures are implemented via a two-span accelerated bridge construction (ABC) system undergoing a large-scale shake-table test. The research objectives are to explore the effect of refined matching algorithms on blob-based features in improving their accuracies and to implement the proposed algorithms on large-scale bridges tested under seismic loads using vision-based SHM. The procedure begins by adopting blob-based feature detectors, i.e., the scale-invariant feature transform (SIFT), speeded-up robust features (SURF), and KAZE algorithms, and their stability is compared. The least medium square (LMEDS), least trimmed square (LTS), random sample consensus (RANSAC), and its generalization maximum sample consensus (MSAC) algorithms are applied for model fitting, and their sensitivity for removing outliers is analyzed. The raw data are corrected using mathematical models and scaled to generate displacement data. Finally, seismic vibrations of the bridge are generated, and the seismic responses are compared. The data are validated using target-tracking methods and mechanical sensors, i.e., string potentiometers. The results show a good agreement between the proposed blob feature detection and matching algorithms and target-tracking data and reference data obtained using mechanical sensors. Purpose: To propose an efficient collimator angle optimization method by combining island blocking (IB) and parked gap (PG) problem to reduce the radiotherapy dose for normal tissue. The reduction will be done with single‐isocenter multi‐lesion volumetric modulated arc therapy (VMAT) for the stereotactic body radiation therapy (SBRT) of liver cancer. Methods: A novel collimator angle optimization algorithm was developed based on the two‐dimensional projection of targets on a beam's eye view (BEV) plane as a function of gantry and collimator angle. This optimization algorithm minimized the sum of the combined IB and PG (IB & PG) areas from all gantry angles for each arc. For comparison, two SBRT plans were respectively generated for each of the 20 retrospective liver cancer cases with multiple lesions. One plan was optimized using the IB & PG algorithm, and the other plan was optimized with a previously reported optimization algorithm that only considered the IB area. Plans were then evaluated and compared using typical dosimetric metrics. Results: With the comparable target coverage, IB & PG plans had significantly lower D500cc, D700cc, mean dose (Dmean), and V15 of normal liver tissues when compared to IB plans. The median percent reductions were 3.32% to 5.36%. The D1cc, D5cc, and Dmean for duodenum and small intestine in IB & PG plans were significantly reduced in a range from 7.60% up to 16.03%. Similarly, the median integral dose was reduced by 3.73%. Furthermore, the percentage of normal liver Dmean sparing when IB & PG plans compared to IB plans, was found to be positively correlated (ρ = 0.669, P = 0.001) with the inter‐target distance. Conclusion: The proposed IB & PG algorithm has been demonstrated to outperform the IB algorithm in almost all normal tissue sparing, and the magnitude of liver sparing was positively correlated with inter‐target distance. Ecosystems in arid areas are under pressure from human activities and the natural environment. Long-term monitoring and evaluation of arid ecosystems are essential for achieving the goal of sustainable development. The Xinjiang Uygur Autonomous Region (Xinjiang) is a typical arid region located in Northwest China with a relatively sensitive ecosystem. Under the support of the Google Earth Engine (GEE) cloud platform's massive data collection, the remote sensing ecological index (RSEI) from 2000 to 2020, both in summer and spring, is established, and the variation trend of the ecological quality in Xinjiang is evaluated by coefficient of variation (CV), Sen's slope analysis, Mann–Kendall trend test (M–K test) and Hurst index. In addition, a partial correlation analysis is processed between RSEI and selected climatic factors, including precipitation and temperature, to find out the mode of correlation between ecological quality and the natural climate. In the last two decades the following has become apparent: (1) The RSEI values of Xinjiang have been relatively low and unstable both in summer and spring, with a trend toward increasing; (2) The distribution characteristics of RSEI levels both in summer and spring have been similar; low levels were concentrated in the desert and wilderness, while high levels were concentrated around the oasis; (3) The ecological quality in Xinjiang has been relatively stable, with a trend of sustained increase both in summer and spring. There was also a small area of sustained decrease around the Junggar Basin and Turpan Basin in summer and a small area of significant decrease in the center of the Taklamakan Desert in spring; (4) In summer, the precipitation has obviously positively correlated in the Southwest. The temperature has obviously positively correlated in the northwestern part; in spring, the precipitation has obviously positively correlated in the Western part; the temperature has obviously positively correlated in the oasis around the Yili River Basin and Tarim Basin. Transition-metal-doped clusters have long been attracting great attention due to their unique geometries and interesting physical and/or chemical properties. In this paper, the geometries of the lowest- and lower-energy CoKn (n = 2–12) clusters have been screened out using particle swarm optimization and first principles relaxation. The results show that except for CoK2 the other CoKn (n = 3–12) clusters are all three-dimensional structures, and CoK7 is the transition structure from which the lowest energy structures are cobalt atom-centered cage-like structures. The stability, the electronic structures, and the magnetic properties of CoKn clusters (n = 2–12) clusters are further investigated using the first principles method. The results show that the medium-sized clusters whose geometries are cage-like structures are more stable than smaller-sized clusters. The electronic configuration of CoKn clusters could be described as 1S1P1D according to the spherical jellium model. The main components of petal-shaped D molecular orbitals are Co-d and K-s states or Co-d and Co-s states, and the main components of sphere-like S molecular orbitals or spindle-like P molecular orbitals are K-s states or Co-s states. Co atoms give the main contribution to the total magnetic moments, and K atoms can either enhance or attenuate the total magnetic moments. CoKn (n = 5–8) clusters have relatively large magnetic moments, which has a relation to the strong Co-K bond and the large amount of charge transfer. CoK4 could be a magnetic superatom with a large magnetic moment of 5 μB. [ABSTRACT FROM AUTHOR]
