The project aim was to develop an innovative biosensor with a high sensitivity, to detect low abundant protein biomarkers in small volumes of blood ( 1 pg/mL, thus overcoming the current standard limits. Current clinical diagnostic criteria for AD require a patient to have clear symptoms of dementia which are excluded from other brain disorders, such as memory loss and confusion, inability to learn new things, difficulty with language and so on.

Live cells act as biological lenses and can be employed as real-world optical components in bio-hybrid systems. Imaging at nanoscale, optical tweezers, lithography and also photonic waveguiding are some of the already proven functionalities, boosted by the advantage that cells are fully biocompatible for intra-body applications. So far, various cell types have been studied for this purpose, such as red blood cells, bacterial cells, stem cells and yeast cells. White Blood Cells (WBCs) play a very important role in the regulation of the human body activities and are usually monitored for assessing its health. WBCs can be considered bio-lenses but, to the best of our knowledge, characterization of their optical properties have not been investigated yet. Here, we report for the first time an accurate study of two model classes of WBCs (i.e., monocytes and lymphocytes) by means of a digital holographic microscope coupled with a microfluidic system, assuming WBCs bio-lens characteristics. Thus, quantitative phase maps for many WBCs have been retrieved in flow-cytometry (FC) by achieving a significant statistical analysis to prove the enhancement in differentiation among sphere-like bio-lenses according to their sizes (i.e., diameter d) exploiting intensity parameters of the modulated light in proximity of the cell optical axis. The most recent discoveries in the biochemical field are highlighting the increasingly important role of lipid droplets (LDs) in several regulatory mechanisms in living cells. LDs are dynamic organelles and therefore their complete characterization in terms of number, size, spatial positioning and relative distribution in the cell volume can shed light on the roles played by LDs. Until now, fluorescence microscopy and transmission electron microscopy are assessed as the gold standard methods for identifying LDs due to their high sensitivity and specificity. However, such methods generally only provide 2D assays and partial measurements. Furthermore, both can be destructive and with low productivity, thus limiting analysis of large cell numbers in a sample. Here we demonstrate for the first time the capability of 3D visualization and the full LD characterization in high-throughput with a tomographic phase-contrast flow-cytometer, by using ovarian cancer cells and monocyte cell lines as models. A strategy for retrieving significant parameters on spatial correlations and LD 3D positioning inside each cell volume is reported. The information gathered by this new method could allow more in depth understanding and lead to new discoveries on how LDs are correlated to cellular functions.

In recent years, intracellular LDs have been discovered to play an important role in several pathologies. Therefore, detection of LDs would provide an in-demand diagnostic tool if coupled with flow-cytometry to give significant statistical analysis and especially if the diagnosis is made in full non-invasive mode. Here we combine the experimental results of in-flow tomographic phase microscopy with a suited numerical simulation to demonstrate that intracellular LDs can be easily detected through a label-free approach based on the direct analysis of the 2D quantitative phase maps recorded by a holographic flow cytometer. In fact, we demonstrate that the presence of LDs affects the optical focusing lensing features of the embracing cell, which can be considered a biological lens. The research was conducted on white blood cells (i.e., lymphocytes and monocytes) and ovarian cancer cells. Results show that the biolens properties of cells can be a rapid biomarker that aids in boosting the diagnosis of LDs-related pathologies by means of the holographic flow-cytometry assay for fast, non-destructive, and high-throughput screening of statistically significant number of cells.

The goal of the SensApp FET-Open project is to develop an innovative super-sensor that will be able to detect Alzheimer’s disease (AD) biomarkers (β-amyloid, Tau and pTAU) in peripheral blood. Considering that nowadays an accurate diagnosis of AD requires the highly invasive withdrawal and analysis of cerebrospinal fluid, SensApp will represent a breakthrough in the field of AD diagnosis thanks to the ability to detect the early stage of the disease by a simple blood collection. We call Droplet-Split-and-Stack (DSS) the new technology that will emerge from SensApp. The achievement of SensApp goal is enabled by the interdisciplinary cooperation between different research institutions and one company involved in the key fields of the project, Vrije Universiteit Brussels, VTT Technical Research Centre of Finland, University of Linz, Ginolis Ltd, IRCCS Centre “Bonino Pulejo”, under the coordination of CNR-Institute of Applied Sciences and Intelligent Systems. This communication will illustrate the progress of the activities.

The effective detection of low-concentrated molecules in small volumes represents a significant challenge in many sectors such as biomedicine, safety, and pollution. Here, we show an easy way to dispense liquid droplets from few mu l volume (0.2-0.5 mu l) of a mother drop, used as reservoir, by using a pyro-electrohydro-dynamic jetting (p-jet) dispenser. This system is proposed for multi-purpose applications such as printing viscous fluids and as a biosensor system. The p-jet system is based on the pyroelectric effect of polar dielectric crystals such as lithium niobate (LN). The electric field generated by the pyroelectric effect acts electro-hydrodynamically on the sample of liquid, allowing the deposition of small volumes. The p-jet approach allows to obtain the dispensing of drops of very small volumes (up to tenths of a picoliter) avoiding the use of syringes and nozzles generally used in standard technologies. The reliability of the technique as a biosensor is demonstrated both in the case of oligonucleotides and in a sample of clinical interest, namely gliadin. The results show the possibility of detecting these biomolecules even when they are low abundant, i.e. down to attomolar. The results show a marked improvement in the detection limit (LOD) when compared with the conventional technique (ELISA). Moreover, it has been presented the possibility of using the p-jet as a useful tool in the detection of biomarkers, present in the blood but currently not detectable with conventional techniques and related to neurodegenerative diseases such as Alzheimer.

Graphene family materials (GFMs) have large perspectives for drug-delivery applications, but their internalization in live cells is under investigation in a wide variety of studies in order to assess the best conditions for efficient cellular uptake. Here we show that mild oxidation of graphene nanoplatelets produces nanographene oxide (nGO) particles, which are massively internalized into the cell cytoplasm. This remarkable uptake of nGO in NIH-3T3 cells has never been observed before. We performed vitality tests for demonstrating the biocompatibility of the material and analyzed the internalization mechanism under different oxidation degrees and concentrations. Moreover, we evaluated quantitatively, for the first time, the cell volume variation after nGO internalization in live cells through a label-free digital holographic imaging technique and in quasi-real-time modality, thus avoiding the time-consuming and detrimental procedures usually employed by electron-based microscopy. The results demonstrate that nGO formulations with a tailored balance between the exposed surface and content of functional groups are very promising in drug-delivery applications.

The demand for sensors capable of measuring low-abundant collagen in human fluids has highly increased in recent years. Indeed, collagen is expected to be a biomarker for chronic diseases and could monitor their progression. Here we show detection of highly diluted samples of collagen at picogram level thanks to an innovative pyro-electrohydrodynamic jet (p-jet) system. Through the intense electric fields generated by the pyroelectric eect in a ferroelectric crystal, the collagen solution was concentrated on a small area of a slide that was appropriately functionalized to bind proteins. The collagen molecules were labeled by an appropriate fluorophore to show how the number of tiny droplets influences the limit of detection of the technique. The results show that the p-jet is extremely promising for overcoming the current detection limits of collagen-based products in human fluids, performing 10 times better than the enzyme-linked immunosorbent assay (ELISA) and thus paving the way for the early diagnosis of related chronic diseases.
