Thermal conductivity is one of the most crucial physical properties of matter when it comes to understanding heat transport, hydrodynamic evolution, and energy balance in systems ranging from astrophysical objects to fusion plasmas. In the warm dense matter regime, experimental data are very scarce so that many theoretical models remain untested. Here we present the first thermal conductivity measurements of aluminum at 0.5-2.7 g/cc and 2-10 eV, using a recently developed platform of differential heating. A temperature gradient is induced in a Au/Al dual-layer target by proton heating, and subsequent heat flow from the hotter Au to the Al rear surface is detected by two simultaneous time-resolved diagnostics. A systematic data set allows for constraining both thermal conductivity and equation-of-state models. Simulations using Purgatorio model or Sesame S27314 for Al thermal conductivity and LEOS for Au/Al release equation-of-state show good agreement with data after 15 ps. Discrepancy still exists at early time 0-15 ps, likely due to non-equilibrium conditions.

We report observation of kHz-pulsed-laser-accelerated electron energies up to 3 MeV in the -klaser (backward) direction from a 3 mJ laser interacting at normal incidence with a solid density, flowing-liquid target. The electrons/MeV/s.r. >1 MeV recorded here using a mJ-class laser exceeds or equals that of prior super-ponderomotive electron studies employing lasers at lower repetition-rates and oblique incidence. Focal intensity of the 40-fs-duration laser is 1.5 · 10 18 W cm -2 , corresponding to only ∼80 keV electron ponderomotive energy. Varying laser intensity confirms electron energies in the laser-reflection direction well above what might be expected from ponderomotive scaling in normal-incidence laser-target geometry. This direct, normal-incidence energy spectrum measurement is made possible by modifying the final focusing off-axis-paraboloid (OAP) mirror with a central hole that allows electrons to pass, and restoring laser intensity through adaptive optics. A Lanex-based, optics-free high-acquisition rate (>100 Hz) magnetic electron-spectrometer was developed for this study to enable shot-to-shot statistical analysis and real-time feedback, which was leveraged in finding optimal pre-plasma conditions. 3D Particle-in-cell simulations of the interaction show qualitative super-ponderomotive spectral agreement with experiment. The demonstration of a high-repetition-rate, high-flux source containing >MeV electrons from a few-mJ, 40 fs laser and a simple liquid target encourages development of future ≥kHz-repetition, fs-duration electron-beam applications.

When a GC/MS system is coupled with a pressurized reactor, the separation efficiency and the retention time are directly affected by the reactor pressure. To keep the GC column flow rate constant irrespective of the reaction pressure, a restrictor capillary tube and an open split interface are attached between the GC injection port and the head of a GC separation column. The capability of the attached modules is demonstrated for the on-line GC/MS analysis of catalytic reaction products of a bio-oil model sample (guaiacol), produced under a pressure of 1 to 3 MPa.

We describe the first demonstration of plasma mirrors made using freely suspended, ultra-thin films formed dynamically and in-situ. We also present novel particle-in-cell simulations that for the first time incorporate multiphoton ionization and dielectric models that are necessary for describing plasma mirrors. Dielectric plasma mirrors are a crucial component for high intensity laser applications such as ion acceleration and solid target high harmonic generation because they greatly improve pulse contrast. We use the liquid crystal 8CB and introduce an innovative dynamic film formation device that can tune the film thickness so that it acts as its own antireflection coating. Films can be formed at a prolonged, high repetition rate without the need for subsequent realignment. High intensity reflectance above 75% and low-field reflectance below 0.2% are demonstrated, as well as initial ion acceleration experimental results that demonstrate increased ion energy and yield on shots cleaned with these plasma mirrors.

We report on the recently completed 400 TW upgrade to the Scarlet laser at The Ohio State University. Scarlet is a Ti:sapphire-based ultrashort pulse system that delivers >10  J in 30 fs pulses to a 2 μm full width at half-maximum focal spot, resulting in intensities exceeding 5×10 21   W/cm 2 . The laser fires at a repetition rate of once per minute and is equipped with a suite of on-demand and on-shot diagnostics detailed here, allowing for rapid collection of experimental statistics. As part of the upgrade, the entire laser system has been redesigned to facilitate consistent, characterized high intensity data collection at high repetition rates. The design and functionality of the laser and target chambers are described along with initial data from commissioning experimental shots.

Efficient coupling of intense laser pulses to solid-density matter is critical to many applications including ion acceleration for cancer therapy. At relativistic intensities, the focus has been mainly on investigating various laser beams irradiating initially overdense flat interfaces with little or no control over the interaction. Here, we propose a novel approach that leverages recent advancements in 3D direct laser writing (DLW) of materials and high contrast lasers to manipulate the laser-matter interactions on the micro-scales. We demonstrate, via simulations, that usable intensities ≥10(23) Wcm(-2) could be achieved with current tabletop lasers coupled to micro-engineered plasma lenses. We show that these plasma optical elements act as a lens to focus laser light. These results open new paths to engineering light-matter interactions at ultra-relativistic intensities.

A novel and simple interface for capillary gas chromatography-mass spectrometry (GC-MS) was developed using a piece of deactivated stainless-steel tubing. This interface eliminated the need to vent the MS ion source when changing columns. Various chromatographic performance indicators, such as inertness, and thermal and chemical stability, were confirmed to be unaffected by using this interface at an elevated temperature of around 300°C. The new interface should facilitate the characterization of polymeric materials using analytical pyrolysis techniques in which frequent switching is required in the measuring mode, such as evolved gas analysis-MS and flash pyrolysis-GC-MS.

A thermal desorption (TD)-GC/MS method was applied for the determination of phthalates in plastic children's toys. The method, which does not need any cumbersome sample pretreatment such as solvent extraction, filtration and derivatization, has been proved to be useful not only as a sensitive analytical method to evaluate phthalate concentration in the toys, but also as a rapid and practical screening method to check whether the concentration exceeds the regulatory limit.

The development of ultra-intense lasers has facilitated new studies in laboratory astrophysics and high-density nuclear science, including laser fusion. Such research relies on the efficient generation of enormous numbers of high-energy charged particles. For example, laser-matter interactions at petawatt (10(15) W) power levels can create pulses of MeV electrons with current densities as large as 10(12) A cm(-2). However, the divergence of these particle beams usually reduces the current density to a few times 10(6) A cm(-2) at distances of the order of centimetres from the source. The invention of devices that can direct such intense, pulsed energetic beams will revolutionize their applications. Here we report high-conductivity devices consisting of transient plasmas that increase the energy density of MeV electrons generated in laser-matter interactions by more than one order of magnitude. A plasma fibre created on a hollow-cone target guides and collimates electrons in a manner akin to the control of light by an optical fibre and collimator. Such plasma devices hold promise for applications using high energy-density particles and should trigger growth in charged particle optics.

A molybdenum/silicon multilayer-coated 1:1 ring-field optic with a numerical aperture of 0.0835 is used to carry out soft-x-ray projection imaging with undulator radiation at 12.9 nm. An ideal optic of this type should be able to image 0.1-µm features with a contrast exceeding 90% at this wavelength. The useful resolution of our ring-field optic is experimentally found to be approximately 0.2 µm, probably because of the presence of substrate figuring errors.

Soft-x-ray projection imaging is demonstrated by the use of 14-nm radiation from a laser plasma source and a single-surface multilayer-coated ellipsoidal condenser. Aberrations in the condenser and the Schwarzschild imaging objective are characterized and correlated with imaging performance. A new Schwarzschild housing, designed for improved alignment stability, is described.

There exists in the mouse a family of I-region-controlled (Ia) antigens which carry carbohydrate-defined determinants. These antigens appear in serum as glycolipids and seem to be actively secreted by antigen-activated T-cells. This paper describes the ability of selected viral, bacterial, and protozoal infections of mice to markedly alter the serum levels of these Ia antigens. All the infectious agents examined induced substantial augmentation or suppression of serum Ia concentrations or both. Lymphocytic choriomeningitis (LCM) virus first enhanced and then suppressed serum Ia levels during the course of acute infection. Enhancement occurred during the time of ongoing virus replication and splenic lymphoproliferation while suppression coincided with the peak of the cytotoxic T-cell response and virus clearance. Listeria monocytogenes infection induced a substantial reduction in Ia levels at a time just after marked depletion of T-cells in the spleen. In contrast, Brucella abortus caused a significant increase in Ia levels 7 days postinfection, which correlates with the appearance of peak numbers of bacteria in tissues. Finally, Plasmodium yoelii, a nonlethal malarial parasite which stimulates prolonged T-cell proliferation, augmented serum Ia levels, whereas P. berghei, a lethal parasite which tends to inhibit. T-cell division, suppressed Ia secretion. Possible interpretations of these different results are presented.

The response of BALB/c mice to immunization with a 230,000 mol. wt protective antigen purified from the blood stages of Plasmodium yoelii was analysed. The protective response to immunization was adjuvant-dependent, and saponin was found to be the most effective adjuvant tested. The intraperitoneal route was found to be superior to the subcutaneous route for protective immunization. Maximal protection was achieved using two immunizations with greater than 12.5 micrograms of antigen plus saponin, but even under these conditions challenge infection developed normally for 5 days, followed by the appearance of crisis forms and rapid clearance of parasites. Effective immunization induced high antibody titres, but serum from immunized mice was not protective on passive transfer. On the other hand, hyperimmune serum from mice recovered and rechallenged with live P. yoelii was protective on passive transfer, even though the titre of antibody specific for the 230,000 mol. wt antigen in this serum was the same as that in the serum from mice immunized with the purified antigen. It was concluded that the 230,000 mol. wt antigen may provide protection against P. yoelii via the cell-mediated immune effector pathway described by Playfair et al. (1979), and that hyperimmune serum contains protective antibodies specific for P. yoelii antigens other than the 230,000 mol. wt antigen.

A cDNA library of P. falciparum was constructed. Using size-selected mRNA as a probe several clones were isolated which hybridized to mRNAs larger than 5 kilobases (kb). The cDNA insert of pFC 17, which hybridizes to 5.6-kb mRNA was expressed by fusion to anthranilate synthetase I in a plasmid expression vector. The expressed fusion protein was shown to contain epitopes of a 195-kDa protein which is the precursor to 3 major surface antigens of P. falciparum merozoites.

A 195,000 mol wt Plasmodium falciparum protein and processing fragments derived from it have been purified by monoclonal antibody affinity chromatography. A polyvalent antiserum has been raised against the purified protein and used to identify the terminal processing products associated with the merozoite. Three unique fragments of 83,000, 42,000, and 19,000 mol wt are present and they represent the major surface antigens of P. falciparum merozoites.

The numbers of 'background' plaque-forming cells (PFC) secreting IgM specific for either sheep erythrocytes or horse erythrocytes were found to be elevated in the spleens of BALB/c mice during Plasmodium berghei and P. yoelii infection. 'Background' PFC numbers were similarly elevated in the spleens of uninfected mice injected with high speed supernatants of lysates of parasitized red blood cells. The active factor (or factors) in the supernatants was (were) non-dialysable and stable at 56 degrees C, but was (were) destroyed by heating to 100 degrees C.

A technique was developed for obtaining high yields of naturally released Plasmodium falciparum merozoites from synchronous cultures of parasitized erythrocytes. The cultured erythrocytes were treated with trypsin to prevent reinvasion (6), and the released merozoites that accumulated extracellularly were harvested by differential centrifugation. The total biosynthetically labeled proteins of schizonts and merozoites, and those immunoprecipitated by human immune serum were analyzed and compared. The surface antigens of free merozoites, labeled by lactoperoxidase-catalyzed iodination, were also described. A monoclonal antibody, specific for a 195,000 mol wt schizont protein, and processing fragments derived from it (3) were used in immunoprecipitation and Western transfer analyses to determine which of the processing fragments are associated with merozoites and which of them are located on the merozoite surface. It was found that processing of the 195,000 mol wt precursor down to an 83,000 mol wt fragment is complete in free merozoites, and that this fragment is expressed as one of the major surface antigens of P. falciparum merozoites.

Changes in the proportions and total numbers of splenic Thy-1.2+ cells, Ig+ cells and normoblasts were analysed during fatal Plasmodium berghei and non-fatal P. yoelii infections in mice. Thy-1.2+ and Ig+ cells were identified by rosetting techniques, and normoblasts by morphological criteria. The splenomegaly observed during these infections was found to be caused mainly by proliferation of normoblasts. An early increase in the numbers of Thy-1.2+ and Ig+ cells was detected in both infections, but in P. berghei infections these responses were subsequently suppressed. In P. yoelii infections Thy-1.2+ and Ig+ cell numbers were maintained at four to five-fold above normal levels until the mice had completely recovered. During the acute phase of P. yoelii infection it appeared that most splenic T-cells expressed surface immunoglobulin.

A purified 230,000 mol wt protein antigen from the lethal mouse malaria parasite Plasmodium yoelii YM which had previously been shown to be highly effective as a vaccine, was tested for its ability to stimulate specific helper T cells and T cells responsible for delayed hypersensitivity. Strong stimulation was found in both assays, but larger doses were required for delayed hypersensitivity, correlating well with the requirements for protection. It is suggested that T stimulation may be a requirement for effective protection by purified antigens in malaria.

The structures of the major merozoite surface antigen of Plasmodium falciparum and the gene encoding it were indistinguishable for the Wellcome strain and the Thai clone T9/94 but different for clones T9/96, T9/98, and T9/101. The central portion of the gene is subject to the greatest variation in structure. The protein from all five lines was found to be posttranslationally modified by covalent addition of both carbohydrate and fatty acid.

Immunity to malaria is mediated, at least in part, by antibody. Resistance to infection has been passively transferred with immune serum or its immunoglobulin fraction in human, simian and rodent malaria. However, because of the structural and antigenic complexity of the malaria parasites, it has proved difficult to identify and characterise those parasite antigens against which protective antibody is directed. We have produced several hybrid cell lines secreting monoclonal antibodies against the rodent malaria parasite, Plasmodium yoelii, and we now report that, of the antibodies tested, only those specific for antigens exclusive to the merozoite were protective in passive transfer experiments. Other anti-P. yoelii monoclonal antibodies, apparently recognising antigens in the membrane of the infected erythrocyte, were not protective on passive transfer. The protective monoclonal antibodies should be useful in the isolation of the important antigens of this parasite.

Stage-specific protein synthesis by the erythrocytic forms of the malaria parasite Plasmodium falciparum was investigated by pulse labeling synchronous parasite cultures with [35S]methionine at 6-h intervals during a complete 48-h developmental cycle. About 40 labeled parasite proteins could be immunoprecipitated with human immune serum, and most of these were associated with the schizont stage of development. In particular, one schizont protein was a 195,000-mol wt species against which a murine monoclonal antibody was produced. This monoclonal antibody, 89.1 reacted with the parasite membrane in schizonts and also with the surface of free merozoites in the indirect immunofluorescence test. In addition to the 195,000-mol wt protein, antibody 89.1 immunoprecipitated a series of lower-molecular weight polypeptides from extracts of labeled asynchronous P. falciparum parasite cultures. These were shown to be related to the 195,000-mol wt protein by peptide mapping. Pulse-chase labeling of synchronized cultures, and immunoprecipitation with antibody 89.1, showed that specific processing of the 195,000-mol wt polypeptide to the lower-molecular-weight products in concomitant with schizont maturation and merozoite release. It is suggested that this P. falciparum protein may be analogous to a similarly processed 230,000-mol wt protective antigen of the rodent malaria parasite, P. yoelii.

Atomic clusters containing from two to several hundred atoms offer the possibility of studying the transition from molecules to crystalline solids. The covalent group IV elements carbon, silicon, and germanium are now being examined with this long-range objective. These elements are particularly interesting because of the very different character of their crystalline solids and because they are intermediate between metals and insulators in the nature of their bonding. Small mass-selected atom cluster ions are formed by pulsed laser techniques and identified by time-of-flight methods. Laser photoexcitation is used to study the relative stability of these clusters and their modes of fragmentation. These modes for C(n)(+) clusters, which tend to fragment with a characteristic loss of a neutral C(3), are found to be different from the modes for Si(n)(+) and Ge(n)(+) clusters, which tend to fragment to "magic" clusters such as Si(4)(+), Si(6)(+) and Si(10)(+). These experimental results can be accounted for by recent theoretical calculations of the ground-state structure and stability of small silicon and carbon clusters. Several theoretical approaches give consistent results, showing that small silicon clusters are compact and different from small fragments of the bulk crystal. Calculations show that carbon clusters change from linear structures toward cyclic structures as the cluster size increases, but with significant odd-even differences.
