The polyadenosine-diphosphate-ribose polymerase 14 (PARP14) has been implicated in DNA damage response pathways for homologous recombination. PARP14 contains three (ADP ribose binding) macrodomains (MD) whose exact contribution to overall PARP14 function in pathology remains unclear. A medium throughput screen led to the identification of N-(2(-9H-carbazol-1-yl)phenyl)acetamide (GeA-69, 1) as a novel allosteric PARP14 MD2 (second MD of PARP14) inhibitor. We herein report medicinal chemistry around this novel chemotype to afford a sub-micromolar PARP14 MD2 inhibitor. This chemical series provides a novel starting point for further development of PARP14 chemical probes.

We present descriptions of the in situ instrumentation, calibration procedures, intercomparison efforts, and data filtering methods used in a 39-yr record of continuous atmospheric carbon dioxide (CO2) observations made at Baring Head, New Zealand. Located on the southern coast of the North Island, Baring Head is exposed to extended periods of strong air flow from the south with minimal terrestrial influence resulting in low CO2 variability. The site is therefore well suited for sampling air masses that are representative of the Southern Ocean region. Instrumental precision is better than 0.015 ppm (1-σ) on 1-Hz values. Comparisons to over 600 co-located flask samples, as well as laboratory based flask and cylinder comparison exercises, suggest that over recent decades compatibility with respect to the Scripps Institution of Oceanography (SIO) and World Meteorological Organisation (WMO) CO2 scales has been 0.3 ppm or better.

Introducción: Streptococcus agalactiae es el principal agente causal de sepsis perinatal precoz. La adquisición de políticas de prevención ha traído consigo la utilización frecuente de antimicrobianos intra-parto. La vigilancia de resistencia antimicrobiana se hace indispensable para definir el fármaco de elección y alternativas en dicha profilaxis. Nuestro centro realiza tamizaje universal desde hace 10 años. Objetivos: Determinar la evolución de la resistencia antimicrobiana de S. agalactiae de colonización materna, a los antimicrobianos utilizados en la prevención de sepsis neonatal, entre 2002 y 2008. Métodos: Se estudiaron 100 muestras vaginales-anales positivas para S. agalactiae de mujeres embarazadas, con edad gestacional de 35 a 37 semanas. Se realizó estudio de susceptibilidad in vitro por discos a penicilina, ampicilina, cefazolina, eritromicina y clindamicina, según método estandarizado por Clinical and Laboratory Standards Institute (CLSI). Resultados: Se analiza la susceptibilidad de 99 cepas. Diecisiete fueron resistentes a eritromicina (17,1%) y 13 eran resistentes a clindamicina (13,1%). De las 17 cepas resistentes a eritromicina, 13 eran fenotipo MLS y 4 del fenotipo M. Dentro del fenotipo MLS, la resistencia fue constitutiva en nueve cepas e inducible en cuatro cepas (test D positivo). En comparación con el año 2002, hubo un aumento significativo de resistencia a clindamicina (de 3,2 a 13,1% p < 0,002) y a eritromicina (de 1,09 a 17% p < 0,001). Introduction: Streptococcus agalactiae (GBS) is the main causative agent of early perinatal sepsis. The acquisition of prevention policies has led to frequent use of intrapartum antibiotics. Surveillance of antimicrobial resistance is indispensable for defining drugs of choice and alternatives for such prophylaxis. Objectives: To determine the evolution of antimicrobial resistance of GBS from maternal colonization to drugs used in the prevention of neonatal sepsis, between 2002 and 2008. Methods: We studied 100 GBS positive vaginal and anal samples from pregnant women. Disc diffussion susceptibility method was performed for penicillin, ampicillin, cefazolin, erythromycin and clindamycin according to the Clinical and Laboratory Standards Institute (CLSI). Results: We analyzed the susceptibility of 99 strains. Seventeen were resistant to erythromycin (17.1%) and 13 were resistant to clindamycin (13.1%). Thirteen of the 17 strains resistant to erythromycin had the MLS phenotype (resistance to erythromycin and clindamycin) and 4 had the M phenotype (resistance to erythromycin only). Within the MLS phenotype, resistance was constitutive in 9 strains, and induced in 4 strains (positive D test). Compared with 2002 there was a significant increase in resistance to clindamycin (from 3.27% to 13.1% p < 0.002) and erythromycin (1.09% to 17% p < 0.001). 100% GSB remained sensitive to penicillin and ampicillin. Conclusions: GBS remains highly susceptible to drugs of choice for prevention of perinatal sepsis. There is a significant increase in antimicrobial resistance to clindamycin and erythromycin. Therefore, it is necessary to request susceptibility testing in GBS from third trimester of pregnancy screening in patients allergic to penicillin.

This paper examines locative relatives in Durban Zulu. We show that locative relatives differ from nominal relatives crucially in prosodic phrasing as well as in resumptive pronoun marking. We propose that the best way to account for locative relatives in Zulu is to resort to the old style adjunction analysis of relative clauses, with an empty operator. This thesis investigates the syntax of object marking in Sambaa and the Bantu languages in general, with particular focus on Swahili and Haya, as points of comparison. Object marking is approached from the perspective of Minimalist syntax. The central claim is that object marking in Sambaa and related languages can be analysed as Agree (in the sense of Chomsky 2000, 2001), with certain modifications. These modifications have implications for the Agree mechanism in general. Object marking is discussed in the context of a range of syntactic environments: simple affirmative clauses, wh-questions, relative clauses and coordination structures. Based on this broad set of data, it is shown that Bantu languages cannot, as has been proposed, be divided into two types, namely those with object agreement and those with pronominal object marking (Bresnan and Mchombo 1987; Byarushengo et al. 1976, 1977 and Baker 2007). Rather, the Agree analysis can account for the object markings patterns in all languages examined. It is further shown that Bonet’s (1991, 1994) Person Case Constraint (PCC) holds for Bantu. The data discussed strongly support Bonet’s distinction between a “weak” and a “strong” PCC, as the languages discussed obey the weak but not the strong version of the PCC. Moreover, the PCC is shown to apply not only to object marking but to all ditransitive constructions in Bantu. This thesis is of relevance to syntacticians interested in agreement, object marking and the interaction of verbs and objects more generally, and to linguists interested in Bantu syntax, and in particular Sambaa, Swahili and Haya.

The rhizosphere-associated bacterium Serratia plymuthica HRO-C48 is not only able to suppress symptoms caused by soil-borne pathogens but is also able to stimulate growth of plants. Detailed knowledge about the underlying mechanisms and regulation are crucial for the application in biocontrol strategies. To analyse the influence of N-acyl homoserine lactone (AHL)-mediated communication on the biocontrol activity, the AHL-degrading lactonase AiiA was heterologously expressed in the strain, resulting in abolished AHL production. The comparative analysis of the wild type and AHL negative mutants led to the identification of new AHL-regulated phenotypes. In the pathosystem Verticillium dahliae-oilseed rape, the essential role of AHL-mediated signaling for disease suppression was demonstrated. In vitro, the regulatory function of AHLs in the synthesis of the plant growth hormone indole-3-acetic acid is shown for the first time. Additionally, swimming motility was found to be negatively AHL regulated. In contrast, production of extracellular hydrolytic enzymes is shown to be positively AHL-regulated. HRO-C48 emits a broad spectrum of volatile organic compounds that are involved in antifungal activity and, interestingly, whose relative abundances are influenced by quorum sensing (QS). The diagnostic difficulties of brucellosis makes the evaluation of new diagnostic tests necessary.Evaluation of different commercial tests in the serological diagnosis of brucellosis by ELISA and immunocapture antibodies in a clinical series of patients with brucellosis of the Health Network of the Catholic University of Chile.All the serums received in the Laboratory of Microbiology for suspicion of brucellosis during five years were studied. Two groups were obtained, one that fulfilled diagnostic criteria for brucellosis [clinical evidence, and/or positive blood culture and/or seroagglutination test (SAT) in titers1/160] and the control group. Each serum sample was analyzed using irnmunocapture-agglutination test (Brucellacapt), ELISA IgM and IgG.Of 10 patients with brucellosis, the serologic results were: 8/10 positives for ELISA IgG, 7/10 for Brucellacapt and SAT, and 5/10 for ELISA IgM.ELISA IgG alone was superior than SAT. The combination ELISA IgG/ Brucellacapt reaches the best detection performance (9/10) and can be an alternative to SAT.

Metodos: Estudio retrospectivo de pacientes cuyos sueros fueron recibidos en el laboratorio de Microbiologia para estudio serologico de brucelosis. Se obtuvieron 2 grupos, aquellos que cumplian criterios diagnosticos de brucelosis [cuadro clinico compatible, y/o hemocultivo positivo y/o serologia por seroaglutinacion en tubo (SAT) en titulos > 1/160] y el grupo control. Todos los sueros se analizaron mediante aglutinacion con inmunocaptura (Brucellacapt®), ELISA IgM y ELISA IgG. Resultados: De 10 pacientes con brucelosis, los resultados serologicos fueron: 8/10 positivos para ELISA IgG, 7/10 para Brucellacapt® y SAT y 5/10 para ELISA IgM. Discusion: ELISA IgG por si solo fue la mejor prueba para el diagnostico de brucelosis. Streptococcus grupo anginosus, y sus especies constellatus, anginosus e intermedius, causan infecciones supuradas. Su identificación microbiológica es compleja pues pueden ser beta-hemolíticos y comparten antígenos de Lancefield con otros estreptococos. Si bien morfológicamente son colonias pequeñas y presentan un olor característicos, si sólo se realiza test de látex para aglutinación del carbohidrato C pueden ser clasificados erróneamente. El objetivo de nuestro trabajo fue evaluar un algoritmo que utiliza 3 pruebas bioquímicas, conocer su susceptibilidad a antimicrobianos y describir las características clínicas de infecciones producidas por Streptococcus grupo anginosus. Se estudiaron cepas de Streptococcus grupo C, G y F, determinando sus características morfológicas de colonia, para identificar cepas de colonia pequeña que se incluyeron en el algoritmo de bajo costo propuesto (b-glucuronidasa y el test Voges-Proskauer) comparándose al método API Rapid ID 32 Strept®, establecido como estándar de oro. Se estudiaron 219 cepas, de las cuales 101 presentaron las características morfológicas compatibles con grupo anginosus, identificándose S. constellatus en 58% (59/101), S. anginosus en 37% (37/101) y S. intermedius en 5% (5/101). Las cepas aisladas procedían principalmente de infecciones piogénicas (61%, 62/101), en especial de foco abdominal. Se observó 98% de susceptibilidad a penicilina y cefotaxima y resistencia a eritromicina y clindamicina en 6,8 y 5%, respectivamente. La edad promedio de los pacientes fue de 30 años (2-85), con distribución similar por sexo y predominio de pacientes hospitalizados. Los resultados expuestos demuestra la efectividad del algoritmo de bajo costo para la detección de S. grupo anginosus. En nuestra experiencia este grupo bacteriano conserva su susceptibilidad a penicilina y cefotaxima, siendo necesario vigilar clindamicina y eritromicina

Its microbiological identification is complex because it can be beta-hemolytic and shares Lancefield antigens with other Streptococci. This group grows as small colonies and presents a characteristic smell, but if only latex test for carbohydrate C agglutination is used it can be erroneously classified. The aims of this publication were to evaluate an algorithm that includes three biochemical tests, and to describe in vitro susceptibility to antibiotics and clinical characteristics of infections caused by Streptococcus anginosus group. Streptoccoccal strains belonging to groups C, G and F were evaluated, determining its colony characteristics in order to select small colony strains that were included into the proposed low cost algorithm (beta-glucurosidase and Voges Proskauer tests) comparing them with API Rapid ID 32 Strept® method, considered the gold standard. Two hundred nineteen strains were studied, 101 had a morphological profile compatible with the anginosus group, identifying S. constellatus in 58% (59/101), S. anginosus in 37% (37/101) and S. intermedius in 5% (5/101). Strains were isolated mainly from pyogenic infections (61%, 62/101), specially from abdominal origin. High grade of penicillin and cefotaxime susceptibility (98%) and low grade resistance to erithromycin and clindamycin (6,8 and 5% respectively) were observed. Mean patient age was 30 years old (2-85), a similar sex distribution with predominance of inpatients. These results document the usefulness of the proposed low cost algorithm for detection of S. anginosus group. In our experience this bacterial group maintains good susceptibility to penicillin and cefotaxime, and we recommend a continous surveillance of clindamycin and erythromycin susceptibility
