Abstract Maize rough dwarf disease (MRDD) threatens maize production globally. The P7-1 effector of the rice black-streaked dwarf virus (RBSDV) targets maize Rab GDP dissociation inhibitor alpha (ZmGDIα) to cause MRDD. However, P7-1 has difficulty recruiting a ZmGDIα variant with an alternative helitron-derived exon 10 (ZmGDIα-hel), resulting in recessive resistance. Here, we demonstrate that P7-1 can recruit another maize protein, gibberellin 2-oxidase 13 (ZmGA2ox7.3), which also exhibits tighter binding affinity for ZmGDIα than ZmGDIα-hel. The oligomerization of ZmGA2ox7.3 is vital for its function in converting bioactive gibberellins into inactive forms. Moreover, the enzymatic activity of ZmGA2ox7.3 oligomers increases when forming hetero-oligomers with P7-1/ZmGDIα, but decreases when ZmGDIα-hel replaces ZmGDIα. Viral infection significantly promotes ZmGA2ox7.3 expression and oligomerization in ZmGDIα-containing susceptible maize, resulting in reduced bioactive GA1/GA4 levels. This causes an auxin/cytokinin imbalance and ultimately manifests as MRDD syndrome. Conversely, in resistant maize, ZmGDIα-hel counters these virus-induced changes, thereby mitigating MRDD severity.

Background Microalbuminuria is a sensitive indicator of early renal microvascular damage and an independent risk factor for cardiovascular diseases. Previous studies have shown that long-term chronic inflammation plays an important role in the occurrence and development of microalbuminuria. However, the correlation between blood inflammatory markers and microalbuminuria has been less frequently reported. Objective To investigate the correlation of systemic immune inflammatory index (SII) and systemic inflammatory response index (SIRI) with microalbuminuria in a middle-aged and elderly physical examination population. Methods This was a cross-sectional study in which 2 105 medical examiners over the age of 40 years who underwent health check-ups from April to July 2023 at the Department of Health Medicine, General Hospital of the Eastern Theatre of Operations were selected as study subjects. The study subjects were divided into two groups based on the urinary microalbumin/urinary creatinine ratio (UACR) values: the non-microalbuminuric group (1 857 cases, UACR0.05) ; age, BMI, systolic blood pressure, diastolic blood pressure, fasting blood glucose, 2 h blood glucose, glycosylated hemoglobin, triglycerides, homocysteine, glomerular filtration rate erythrocyte sedimentation rate, SII, and SIRI levels were higher in the microalbuminuria group than those in the non-microalbuminuria group, and high density lipoprotein cholesterol level was lower than that in the non-microalbuminuria group (P

Abstract Hu sheep (HS), a breed of sheep carrying the FecB mutation gene, is known for its “year-round estrus and multiple births” and is an ideal model for studying the high fecundity mechanisms of livestock. Through analyzing and comparing the genomic selection features of Hu sheep and other sheep breeds, we identified a series of candidate genes that may play a role in Hu sheep’s high fecundity mechanisms. In this study, we conducted whole-genome resequencing on six breeds and screened key mutations significantly correlated with high reproductive traits in sheep. Notably, the CC2D1B gene was selected by the fixation index (F ST) and the cross-population composite likelihood ratio (XP-CLR) methods in HS and other five breeds. It was worth noting that the CC2D1B gene in HS was different from that in other sheep breeds, and seven missense mutations have been identified. Furthermore, the linkage disequilibrium (LD) analysis revealed a strong linkage disequilibrium in this specific gene region. Subsequently, by performing different grouping based on FecB genotypes in Hu sheep, genome-wide selective signal analysis screened several genes related to reproduction, such as BMPR1B and PPM1K. Besides, F ST analysis identified functional genes related to reproductive traits, including RHEB, HSPA2, PPP1CC, HVCN1, and CCDC63. Additionally, a missense mutation was found in the CCDC63 gene and the haplotype was different between the high reproduction (HR) group and low reproduction (LR) group in HS. In summary, we discovered genetic differentiation among six distinct breeding sheep breeds at the whole genome level. Additionally, we identified a set of genes which were associated with reproductive performance in Hu sheep and visualized how these genes differed in different breeds. These findings laid a theoretical foundation for understanding genetic mechanisms behind high prolific traits in sheep.

Abstract Background Interventional therapy, in conjunction with tyrosine kinase inhibitors (TKIs), has shown promising outcomes for treating hepatocellular carcinoma (HCC) with portal vein tumor thrombosis (PVTT). With the advent of immunotherapy, the combined use of immune checkpoint inhibitors (ICIs) has attracted great attention due to their potential effectiveness in advanced HCC. This study aims to compare the efficacy and safety of a triple therapy regimen (Interventional therapy, TKIs and ICIs, IT-TKI-ICI) with a dual therapy regimen (Interventional therapy and TKIs, IT-TKI) in the treatment of HCC and PVTT (HCC-PVTT). Methods A comprehensive search was carried out in PubMed, Web of Science, Embase, Scopus, and the Cochrane Library databases. Primary outcome measures were overall survival (OS) and progression-free survival (PFS), while secondary outcomes included tumor response rate, adverse event incidence as well as downstaging surgery rate. Statistical analysis was conducted using Revman 5.4 software. Results The meta-analysis finally included 6 cohort studies. The triple therapy group demonstrated significantly prolonged OS and PFS compared to the dual therapy group. Meanwhile, the former exhibited significantly higher rates of objective response rate (ORR), disease control rate (DCR) and better downstaging effects with a higher salvage surgery rate without significantly increasing adverse events. Conclusion In comparison to dual therapy, the triple therapy with interventional therapy, TKIs, and ICIs demonstrates superior efficacy and equivalent safety for HCC-PVTT.

BackgroundRNA editing, especially A-to-I editing sites, is a common RNA modification critical for stem cell differentiation, muscle development, and disease occurrence. Unveiling comprehensive RNA A-to-I editing events associated with myogenesis of the skeletal muscle satellite cells (MuSCs) is essential for extending our knowledge of the mechanism underpinning muscle development.ResultsA total of 9,632 RNA editing sites (RESs) were screened in the myoblasts (GM), myocytes (DM1), and myotubes (DM5) samples. Among these sites, 4,559 A-to-I edits were classified and further analyzed. There were 3,266 A-to-I sites in the protein-coding region, out of which 113 missense sites recoded protein. Notably, five A-to-I sites in the 3′ UTR of four genes (TRAF6, NALF1, SLC38A1, ENSCHIG00000019092) altered their targeted miRNAs. Furthermore, a total of 370 A-to-I sites with different editing levels were detected, including FBN1, MYH10, GSK3B, CSNK1D, and PRKACB genes. These genes were predominantly enriched in the cytoskeleton in muscle cells, the hippo signaling pathway, and the tight junction. Furthermore, we identified 14 hub genes (TUFM, GSK3B, JAK2, RPSA, YARS1, CDH2, PRKACB, RUNX1, NOTCH2, CDC23, VCP, FBN1, RARS1, MEF2C) that potentially related to muscle development. Additionally, 123 stage-specific A-to-I editing sites were identified, with 43 sites in GM, 25 in DM1, and 55 in DM5 samples. These stage-specific edited genes significantly enriched essential biological pathways, including the cell cycle, oocyte meiosis, motor proteins, and hedgehog signaling pathway.ConclusionWe systematically identified the RNA editing events in proliferating and differentiating goat MuSCs, which was crucial for expanding our understanding of the regulatory mechanisms of muscle development.

With increasing concern about mental health issues and active lifestyles among university students, understanding the interplay between different physical activity motivations and various dimensions of psychological well-being is important. The present study aims to explore the canonical relationship between physical activity motivations based on self-determination theory and psychological well-being according to Ryff’s model in university students. Nine hundred and sixty-six Chinese university students participated in this study. A canonical correlation analysis was conducted using six variables of motivations as predictors of six variables of psychological well-being. The canonical correlation analysis yielded two canonical functions. The first canonical function, which was primary, indicated that intrinsic motivation, integrated regulation, identified regulation, and introjected regulation contributed the most to psychological well-being. The second canonical function indicated that a decrease in external motivation and amotivation accounted for an increase in personal growth. This study underscores the importance of elucidating the underlying motivations driving physical activity behaviors in order to enhance psychological well-being in this population.

Understanding the colonization and change patterns of gut microbiota is pivotal for comprehending host health. As a newly cultured breed, the studies on the gut microbiota of Tianfu goats remain limited. This study aimed to address this gap by analyzing the microbial composition and colonization patterns of fecal samples collected from goat kids from birth to weaning. Fecal samples were collected on days 0, 7, 14, 21, 28, 35, 42, 49, 53, 55, 57, and 64, and the changes and colonization patterns of microorganisms were analyzed through high-throughput 16S rRNA sequencing. The results showed that the abundance of fecal microbiota in goat kids gradually increased over time, followed by a decrease after weaning and stabilization, with reduced individual differences. The colonization of fecal microorganisms mainly presented three different stages: days 0–14, days 21–49, and days 53–64. During the suckling period, the relative abundance of Proteobacteria (72.34%) was the highest, followed by Firmicutes (21.66%). From 21 days old, the microbiota in goat kids gradually to be diverse, with Lachnospiraceae and Ruminococcaceae being dominant. In this paper, we carry out an investigation into the semileptonic decays Ds+→K0ℓ+νℓ with ℓ=(e,μ) by employing the QCD light-cone sum rules approach. The vector transition form factor (TFF) f+Ds+K0(q2) for Ds+→K0 decay is calculated while considering its next-to-leading order contribution. Subsequently, we briefly introduce the twist-2, 3 kaon distribution amplitudes, which are calculated by using QCD sum rules within the framework of the background field theory. At the large recoil point, the TFF has f+Ds+K0(0)=0.692−0.026+0.027. Then, we extrapolate f+Ds+K0(q2) to the whole physical q2-region via the simplified z(q2,t)-series expansion, and the behavior of TFF f+Ds+K0(q2) is exhibited in the numerical results part, including the theoretical and experimental predictions for comparison. In addition, we compute the differential branching fraction B(Ds+→K0ℓ+νℓ) with the electron and muon channels, which are expected to be B(Ds+→K0e+νe)=3.379−0.275+0.301×10−3 and B(Ds+→K0μ+νμ)=3.351−0.273+0.299×10−3 as well as contained other results for comparison. Our results show good agreement with the BESIII measurements and theoretical predictions. Furthermore, we present our prediction with respect to the CKM matrix element |Vcd| by using the B(Ds+→K0e+νe) result from BESIII Collaboration, yielding |Vcd|=0.221−0.010+0.008. Finally, we provide the ratio between Ds+→K0e+νe and Ds+→ηe+νe channels, i.e. RK0/ηe=0.144−0.020+0.028.

Abstract The current‐generation adenine base editor (ABE) ABE8e, which has evolved from the prokaryotic evolution system, exhibits high efficiency in mediating A‐to‐G conversion and is presumed to be promising for gene therapy. However, its much wider editing window and substantially higher off‐target editing activity restricted its applications in precise base editing for therapeutic use. This study uses a library‐assisted protein evolution approach using eukaryotic cells to generate ABE variants with improved specificity and reduced off‐target editing while maintaining high activity in human cells. The study generated an expanded set of ABEs with efficient editing activities and chose four evolved variants that offered either similar or modestly higher efficiency within a narrower editing window of protospacer position ≈4–7 compared to that of ABE8e in human cells, which would enable minimized bystander editing. Moreover, these variants resulted in reduced off‐target editing events when delivered as plasmid or mRNA into human cells. Finally, these variants can install both disease‐suppressing mutations and disease‐correcting mutations efficiently with minimal undesired bystander editing making them promising approaches for specific therapeutic edits. In summary, the work establishes a mutant‐library‐assisted protein evolution method in eukaryotic cells and generates alternative ABE variants as efficient tools for precise human genome editing.

The ignition issue of the hypersonic aircraft remains a challenging task due to the extremely short residual time of milliseconds. To study the potential candidate of hydrogen peroxide for ignition enhancement, the ignition delay times of pyrolysis gas with the addition of 0–5000 ppm hydrogen peroxide were experimentally measured and quantitatively analyzed at temperatures of 833.8–962.1 K, equivalence ratios of 0.5–1.5, pressures of 31.9–48.8 bar, and the dilution of 45 vol% Ar. A novel injection system based on a Laval nozzle was designed to well atomize and evaporate the liquid hydrogen peroxide. The experimental results showed that the enhancing effect of hydrogen peroxide was proceeded by three stages, in which the decreasing efficiency gradually weakened with the increase of additive fraction. The ignition delay times of pyrolysis gas dramatically shortened by 66.1 % with 1000 ppm of hydrogen peroxide. Only a decrement of 19.1 % was obtained as the additive fraction increased from 1000 to 4000 ppm. The decline at 4000–5000 ppm even reduced to 2.0 %. It is suggested to add no more than 1000 ppm of hydrogen peroxide from the perspective of the highest acceleration efficiency on the ignition delay times of pyrolysis gas.

Abstract Background The small YABBY plant-specific transcription factor has a prominent role in regulating plant growth progress and responding to abiotic stress. Results Here, a total of 16 PvYABBYs from switchgrass (Panicum virgatum L.) were identified and classified into four distinct subgroups. Proteins within the same subgroup exhibited similar conserved motifs and gene structures. Synteny analyses indicated that segmental duplication contributed to the expansion of the YABBY gene family in switchgrass and that complex duplication events occurred in rice, maize, soybean, and sorghum. Promoter regions of PvYABBY genes contained numerous cis-elements related to stress responsiveness and plant hormones. Expression profile analysis indicated higher expression levels of many PvYABBY genes during inflorescence development and seed maturation, with lower expression levels during root growth. Real-time quantitative PCR analysis demonstrated the sensitivity of multiple YABBY genes to PEG, NaCl, ABA, and GA treatments. The overexpression of PvYABBY14 in Arabidopsis resulted in increased root length after treatment with GA and ABA compared to wild-type plants. Conclusions Taken together, our study provides the first genome-wide overview of the YABBY transcription factor family, laying the groundwork for understanding the molecular basis and regulatory mechanisms of PvYABBY14 in response to ABA and GA responses in switchgrass.

Emotional intelligence is considered as an important factor impacting on sports motivation of students-athletes. Meanwhile the role of culture in the development of emotional intelligence is still insufficiently studied in sports psychology. The purpose of the study included comparing the indicators of emotional intelligence of student-athletes in China and Russia, identifying the relationship between emotional intelligence, sports motivation and physical activity, as well as studying the prognostic effect of emotional intelligence on autonomous motivation. The research was done among 474 student- athletes. In Chinese students sample (N = 281), the 163 men and 118 women. In the Russian student sample (N = 193), there were 64 men and 129 women. The following research methods were used: emotional intelligence scale, the sport motivation scale-6, the international physical activity questionnaire. The results of the study have showed that the level of emotional intelligence of Chinese student-athletes is higher than that of Russian students-athletes. Positive correlations between emotional intelligence, autonomous motivation and physical activity were found in both samples. At the same time, the correlation coefficient in the Russian sample was lower. A predictive relationship between emotional intelligence and autonomous motivation has been established; meanwhile the self-assessment of emotions and emotion regulation make the greatest contribution to autonomous motivation in the Chinese sample, while in the Russian sample there is only the use of emotions. Considering the cultural context can contribute to the preparation effective programs for the development of emotional intelligence and autonomous motivation for physical activity.

Chimeric antigen receptor (CAR)-T cell therapy has been confirmed improving remission rates in refractory patients or relapsed B-cell acute lymphoblastic leukemia (R/R B-ALL). However, the added benefits of undergoing allogeneic hematopoietic stem cell transplantation (allo-HSCT) following CAR-T therapy remain a subject of debate. In this research we investigated the efficiency and long-term outcomes of CD19 CAR-T bridging with allo-HSCT in R/R B-ALL patients. A total of 42 patients were brought into the cohort studies. Our findings revealed that patients who appected CAR-T followed by HSCT had a 1-year overall survival (OS) rate of 70 % and a 1-year leukemia-free survival (LFS) rate of 95 %. Moreover, patients who underwent this combined treatment had higher OS and LFS rates compared to those who received CAR-T therapy alone. In conclusion, the results of this clinical trial provide compelling evidence for the safety and efficacy of using CAR-T therapy as a bridging strategy to allo-HSCT in patients with R/R B-ALL.

Accurate processing and analysis of non-human primate (NHP) brain magnetic resonance imaging (MRI) serves an indispensable role in understanding brain evolution, development, aging, and diseases. Despite the accumulation of diverse NHP brain MRI datasets at various developmental stages and from various imaging sites/scanners, existing computational tools designed for human MRI typically perform poor on NHP data, due to huge differences in brain sizes, morphologies, and imaging appearances across species, sites, and ages, highlighting the imperative for NHP-specialized MRI processing tools. To address this issue, in this paper, we present a robust, generic, and fully automated computational pipeline, called non-human primates Brain Extraction and Segmentation Toolbox (nBEST), whose main functionality includes brain extraction, non-cerebrum removal, and tissue segmentation. Building on cutting-edge deep learning techniques by employing lifelong learning to flexibly integrate data from diverse NHP populations and innovatively constructing 3D U-NeXt architecture, nBEST can well handle structural NHP brain MR images from multi-species, multi-site, and multi-developmental-stage (from neonates to the elderly). We extensively validated nBEST based on, to our knowledge, the largest assemblage dataset in NHP brain studies, encompassing 1,469 scans with 11 species (e.g., rhesus macaques, cynomolgus macaques, chimpanzees, marmosets, squirrel monkeys, etc.) from 23 independent datasets. Compared to alternative tools, nBEST outperforms in precision, applicability, robustness, comprehensiveness, and generalizability, greatly benefiting downstream longitudinal, cross-sectional, and cross-species quantitative analyses. We have made nBEST an open-source toolbox (https://github.com/TaoZhong11/nBEST) and we are committed to its continual refinement through lifelong learning with incoming data to greatly contribute to the research field.

This article examines sport/discipline-specific market concentrations in the context of the eight Summer Olympic Games held between 1992 and 2020. Three market concentration-related debates underpin this study: (1) the number of competing firms; (2) market size; and (3) time. A normalized version of the Herfindahl-Hirschman Index (HHIN) is calculated for gold medal and medal concentrations for all 42 sports/disciplines. The analyses included panel data models and Pearson correlations. The main findings are that (1) market concentrations for (gold) medals within most sport/disciplines have not significantly declined over time; and (2) neither an increased number of competing National Olympic Committees (NOCs) nor increased market size is likely to have impacted market concentration within a sport/discipline. The findings advance the understanding of market concentration trends and have implications for elite sport practitioners including the International Olympic Committee (IOC) and International Federations (IFs) in terms of an evaluation of existing measures introduced to promote competitive balance, and NOCs in terms of resource distribution among sports/disciplines and prioritization strategies.

Abstract The $$\eta ^{(\prime )}$$ η ( ′ ) -mesons in the quark-flavor basis are mixtures of two mesonic states $$|\eta _{q}\rangle =|{\bar{u}} u+{\bar{d}} d\rangle /\sqrt{2}$$ | η q ⟩ = | u ¯ u + d ¯ d ⟩ / 2 and $$|\eta _{s}\rangle =|{\bar{s}} s\rangle .$$ | η s ⟩ = | s ¯ s ⟩ . In previous work, we have made a detailed study on the $$\eta _{s}$$ η s leading-twist distribution amplitude by using the $$D^+_s$$ D s + meson semileptonic decays. As a sequential work, in the present paper, we fix the $$\eta _q$$ η q leading-twist distribution amplitude by using the light-cone harmonic oscillator model for its wave function and by using the QCD sum rules within the QCD background field to calculate its moments. The input parameters of $$\eta _q$$ η q leading-twist distribution amplitude $$\phi _{2;\eta _q}$$ ϕ 2 ; η q at the initial scale $$\mu _0\sim 1$$ μ 0 ∼ 1 GeV are fixed by using those moments. The QCD sum rules for the $$0_{\textrm{th}}$$ 0 th -order moment can also be used to fix the magnitude of $$\eta _q$$ η q decay constant, giving $$f_{\eta _q}=0.141\pm 0.005$$ f η q = 0.141 ± 0.005 GeV. As an application of $$\phi _{2;\eta _q},$$ ϕ 2 ; η q , we calculate the transition form factors $$B(D)^+ \rightarrow \eta ^{(\prime )}$$ B ( D ) + → η ( ′ ) by using the QCD light-cone sum rules up to twist-4 accuracy and by including the next-to-leading order QCD corrections to the leading-twist part, and then fix the related CKM matrix element and the decay width for the semi-leptonic decays $$B(D)^+ \rightarrow \eta ^{(\prime )}\ell ^+ \nu _\ell .$$ B ( D ) + → η ( ′ ) ℓ + ν ℓ .

ObjectivesAs the ship side wet exhaust system is widely equipped on many stealth frigates, it is necessary to study its infrared signature suppression (IRSS) effect.MethodsIn this paper, the IRSS principle of the wet exhaust system is analyzed, a mathematical model of the system based on an exporting vessel is built, the exhaust system outlet temperature is simulated and a real vessel infrared signature test is carried out.Results The simulation results show that the outlet temperature is reduced to 60 ℃. Compared to popular funnel IRSS devices, the infrared signature of the 3–5 μm wave band can be reduced. The real vessel infrared signature test results show that the outlet temperature is reduced to 66.7–70.0 ℃ and the infrared signature intensity is 1.39 W/s.ConclusionsThe results of this study can provide useful references for the application of the ship side wet exhaust system in the naval ship IRSS field.

Abstract Selective defluorinative functionalization of trifluoromethyl group (–CF3) is an attractive synthetic route to the pharmaceutically privileged fluorine-containing moiety. Herein, we report a strategy based on photoexcited copper catalysis to activate the C–F bond of di- or trifluoromethylated arenes for divergent radical C–N coupling with carbazoles and aromatic amines. The use of different ligands can tune the reaction products diversity. A range of substituted, structurally diverse α,α-difluoromethylamines can be obtained from trifluoromethylated arenes via defluorinative C-N coupling with carbazoles, while an interesting double defluorinative C-N coupling is ready for difluoromethylated arenes. Based on this success, a carbazole-centered PNP ligand is designed to be an optimal ligand, enabling a copper-catalyzed C–N coupling for the construction of imidoyl fluorides from aromatic amines through double C-F bond functionalization. Interestingly, a 1,2-difluoroalkylamination strategy of styrenes is also developed, delivering γ,γ-difluoroalkylamines, a bioisostere to β-aminoketones, in synthetically useful yields. The DFT studies reveal an inner-sphere electron transfer mechanism for Cu-catalyzed selective activation of C(sp3)–F bonds.

Skeletal muscle development is spotlighted in mammals since it closely relates to animal health and economic benefits to the breeding industry. Researchers have successfully unveiled many regulatory factors and mechanisms involving myogenesis. However, the effect of N6-methyladenosine (m6A) modification, especially demethylase and its regulated genes, on muscle development remains to be further explored. Here, we found that the typical demethylase FTO (fat mass- and obesity-associated protein) was highly enriched in goats’ longissimus dorsi (LD) muscles. In addition, the level of m6A modification on transcripts was negatively regulated by FTO during the proliferation of goat skeletal muscle satellite cells (MuSCs). Moreover, a deficiency of FTO in MuSCs significantly retarded their proliferation and promoted the expression of dystrophin-associated protein 1 (DAG1). m6A modifications of DAG1 mRNA were efficiently altered by FTO. Intriguingly, the results of DAG1 levels and its m6A enrichment from FB23-2 (FTO demethylase inhibitor)-treated cells were consistent with those of the FTO knockdown, indicating that the regulation of FTO on DAG1 depended on m6A modification. Further experiments showed that interfering FTO improved m6A modification at site DAG1-122, recognized by Insulin-like growth factor 2 mRNA-binding protein 1 (IGF2BP1) and consequently stabilized DAG1 transcripts. Our study suggests that FTO promotes the proliferation of MuSCs by regulating the expression of DAG1 through m6A modification. This will extend our knowledge of the m6A-related mechanism of skeletal muscle development in animals.

The process of skeletal muscle development is intricate and involves the regulation of a diverse array of genes. Accurate gene expression profiles are crucial for studying muscle development, making it essential to choose the right reference genes for real-time quantitative PCR (RT-qPCR). In the present study, eight candidate reference genes were identified from our previous transcriptome sequencing analysis of caprine skeletal muscle satellite cells (MuSCs), and two traditional reference genes (ACTB and GAPDH) were assessed. The quantitative levels of the candidate reference genes were determined through the RT-qPCR technique, while the stability of their expression was evaluated utilizing the GeNorm, NormFinder, BestKeeper, and RefFinder programs. Furthermore, the chosen reference genes were utilized for the normalization of the gene expression levels of PCNA and Myf5. It was determined that conventional reference genes, including ACTB and GAPDH, were not appropriate for normalizing target gene expression. Conversely, RPL14 and RPS15A, identified through RNA sequencing analysis, exhibited minimal variability and were identified as the optimal reference genes for normalizing gene expression during the proliferation and differentiation of goat MuSCs. Our research offers a validated panel of optimal reference genes for the detection of differentially expressed genes in goat muscle satellite cells using RT-qPCR.

Compared to conventional irrigation and fertilization, the Water-fertilizer coupling can significantly enhance the efficiency of water and fertilizer utilization, thereby promoting crop growth and increasing yield. Targeting the challenges of poor crop growth, low yield, and inefficient water and fertilizer utilization in the arid region of northwest China under conventional irrigation and fertilization practices. Therefore, a two-year on-farm experiment in 2022 and 2023 was conducted to study the effects of water-fertilizer coupling regulation on pumpkin growth, yield, water consumption (ET), and water and fertilizer use efficiency. Simultaneously the comprehensive evaluation of multiple objectives was carried out using principal component analysis (PCA) methods, so as to propose an suitable water-fertilizer coupling regulation scheme for the region. The experiment was set up as a two-factor trial using water-fertilizer integration technology under three irrigation volume (W1 = 37.5 mm, W2 = 45.5 mm, W3 = 52.5mm) and three organic fertilizer application amounts (F1 = 3900-300 kg ha-1, F2 = 4800-450 kg·ha-1, F3 = 5700-600 kg·ha-1), with the traditional irrigation and fertilization scheme from local farmers as control treatments (CK). Brain extraction and image quality assessment are two fundamental steps in fetal brain magnetic resonance imaging (MRI) 3D reconstruction and quantification. However, the randomness of fetal position and orientation, the variability of fetal brain morphology, maternal organs around the fetus, and the scarcity of data samples, all add excessive noise and impose a great challenge to automated brain extraction and quality assessment of fetal MRI slices. Conventionally, brain extraction and quality assessment are typically performed independently. However, both of them focus on the brain image representation, so they can be jointly optimized to ensure the network learns more effective features and avoid overfitting. To this end, we propose a novel two-stage dual-task deep learning framework with a brain localization stage and a dual-task stage for joint brain extraction and quality assessment of fetal MRI slices. Specifically, the dual-task module compactly contains a feature extraction module, a quality assessment head and a segmentation head with feature fusion for simultaneous brain extraction and quality assessment. Besides, a transformer architecture is introduced into the feature extraction module and the segmentation head. We utilize a multi-step training strategy to guarantee a stable and successful training of all modules. Finally, we validate our method by a 5-fold cross-validation and ablation study on a dataset with fetal brain MRI slices in different qualities, and perform a cross-dataset validation in addition. Experiments show that the proposed framework achieves very promising performance.

Summary: Glioblastoma stem cells (GSCs) reside in hypoxic periarteriolar niches of glioblastoma micro-environment, however, the crosstalk of GSCs with macrophages on regulating tumor angiogenesis and progression are not fully elucidated. GSCs-derived exosomes (GSCs-exos) are essential mediators during tumor immune-microenvironment remodeling initiated by GSCs, resulting in M2 polarization of tumor-associated macrophages (TAMs) as we reported previously. Our data disclosed aberrant upregulation of miR-374b-3p in both clinical glioblastoma specimens and human cell lines of GSCs. MiR-374b-3p level was high in GSCs-exos and can be internalized by macrophages. Mechanistically, GSCs exosomal miR-374b-3p induced M2 polarization of macrophages by downregulating phosphatase and tensin expression, thereby promoting migration and tube formation of vascular endothelial cells after coculture with M2 macrophages. Cumulatively, these data indicated that GSCs exosomal miR-374b-3p can enhance tumor angiogenesis by inducing M2 polarization of macrophages, as well as promote malignant progression of glioblastoma. Targeting exosomal miR-374b-3p may serve as a potential target against glioblastoma.

Abstract Cotton maps (10 m) of Xinjiang (XJ_COTTON10), which is the largest cotton production region of China, were produced from 2018 to 2021 through supervised classification. A two-step mapping strategy, i.e., cropland mapping followed by cotton extraction, was employed to improve the accuracy and efficiency of cotton mapping for a large region of about 1.66 million km2 with high heterogeneity. Additionally, the time-series satellite data related to spectral, textural, structural, and phenological features were combined and used in a supervised random forest classifier. The cotton/non-cotton classification model achieved overall accuracies of about 95% and 90% on the test samples of the same and adjacent years, respectively. The proposed two-step cotton mapping strategy proved promising and effective in producing multi-year and consistent cotton maps. XJ_COTTON10 agreed well with the statistical areas of cotton at the county level (R2 = 0.84–0.94). This is the first cotton mapping for the entire Xinjiang at 10-meter resolution, which can provide a basis for high-precision cotton monitoring and policymaking in China.

Abstract Background Increasing evidence suggests that immunotherapy, especially immune checkpoint inhibitors (ICIs), has the potential to facilitate long-term survival in various cancer besides prostate cancer. Emerging evidence indicated that pyroptosis, an immunogenic form of cell death, could trigger an anti-tumor immune microenvironment and enhance the effectiveness of immunotherapy. Nevertheless, the mechanism underlying the regulation of pyroptosis signaling in prostate cancer remains unclear. Methods The differential expression of human E3 ligases in prostate cancer was integratedly analyzed from five independent public datasets. Moreover, the immunohistochemistry analysis of a tissue microarray derived from prostate cancer patients confirmed the results from the bioinformatic analysis. Furthermore, prostate cancer cell lines were evaluated via the next-generation RNA sequencing to assess transcriptomic profile upon CDC20 depletion. Next, qRT-PCR, Western blotting, cycloheximide assay, immunoprecipitation, and ubiquitination assay were employed to explore the correlation and interaction between CDC20 and GSDME. Both immune-deficient and immune-competent murine models were utilized to examine the anti-tumor efficacy of CDC20 inhibition with or without the anti-PD1 antibodies, respectively. To analyze the immune microenvironment of the xenografts, the tumor tissues were examined by immunohistochemistry and flow cytometry. Results The analysis of multiple prostate cancer cohorts suggested that CDC20 was the most significantly over-expressed E3 ligase. In addition, CDC20 exerted a negative regulatory effect on the pyroptosis pathway by targeting GSDME for ubiquitination-mediated proteolysis in a degron-dependent manner. Knockdown of CDC20 leads to increased GSDME abundance and a transition from apoptosis to pyroptosis in response to death signals. Furthermore, in our syngeneic murine models, we found that depletion of CDC20 significantly enhances the anti-tumor immunity by promoting the infiltration of CD8+ T lymphocytes dependent on the existence of GSDME, as well as reducing myeloid immune cells. More importantly, Apcin, a small molecular inhibitor that targets CDC20, exhibited synergistic effects with anti-PD1-based immunotherapy in murine models of prostate cancer. Conclusions Overall, these findings provide new insights into the upstream regulation of GSDME-mediated pyroptosis by CDC20, which specifically interacts with GSDME and facilitates its ubiquitination in a degron-dependent manner. Importantly, our data highlight novel molecular pathways for targeting cellular pyroptosis and enhancing the effectiveness of anti-PD1-based immunotherapy.

Domestic animals have multiple phenotypes of skin and coat color, which arise from different genes and their products, such as proteins and metabolites responsible with melanin deposition. However, the complex regulatory network of melanin synthesis remains to be fully unraveled. Here, the skin and tongue tissues of Liangshan black sheep (black group) and Liangshan semi-fine-wool sheep (pink group) were collected, stained with hematoxylin–eosin (HE) and Masson–Fontana, and the transcriptomic and metabolomic data were further analyzed. We found a large deposit of melanin granules in the epidermis of the black skin and tongue. Transcriptome and metabolome analysis identified 744 differentially expressed genes (DEGs) and 443 differentially expressed metabolites (DEMs) between the pink and black groups. Gene ontology (GO) and Kyoto encyclopedia of genes and genomes (KEGG) enrichment analyses revealed the DEGs and DEMs were mainly enriched in the pathways of secondary metabolic processes, melanin biosynthesis processes, melanin metabolism processes, melanosome membranes, pigment granule membranes, melanosome, tyrosine metabolism, and melanogenesis. Notably, we revealed the gene ENSARG00020006042 may be a family member of YWHAs and involved in regulating melanin deposition. Furthermore, several essential genes (TYR, TYRP1, DCT, PMEL, MLANA, SLC45A2) were significantly associated with metabolite prostaglandins and compounds involved in sheep pigmentation. These findings provide new evidence of the strong correlation between prostaglandins and related compounds and key genes that regulate sheep melanin synthesis, furthering our understanding of the regulatory mechanisms and molecular breeding of pigmentation in sheep.

Aiming at the problems of large error and redundancy in the multi-node data acquisition of multi-greenhouse photo growth environmental information, a three-level fusion algorithm based on adaptive weighting, an LMBP network, and an improved D-S theory is proposed. The box-and-line graph method recognizes the original data and then replaces it based on the mean value method; the air temperature, humidity, and light intensity measurements are unbiased estimations of the true value to be estimated, so the first level of fusion chooses the adaptive weighted average algorithm to find the optimal weights of each sensor under the condition of minimizing the total mean-square error and obtains the optimal estimation of the weights of the homogeneous sensors of a greenhouse. The Levenberg–Marquardt algorithm was chosen for the second level of fusion to optimize the weight modification of the BP neural network, i.e., the LMBP network, and the three environmental factors corresponding to “suitable”, “uncertain” and “unsuitable” potato growth environments were trained for the three environmental factors in the reproductive periods. The output of the hidden layer was converted into probability by the Softmax function. The third level is based on the global fusion of evidence theory (also known as D-S theory), and the network output is used as evidence to obtain a consistent description of the multi-greenhouse potato cultivation environment and the overall scheduling of farming activities, which better solves the problem of the difficulty in obtaining basic probability assignments in the evidence theory; in the case of a conflict between the evidence, the BPA of the conflicting evidence is reallocated, i.e., the D-S theory is improved. Example validation shows that the total mean square error of the adaptive weighted fusion value is smaller than the variance of each sensor estimation, and sensors with lower variance are assigned lower weights, which makes the fusion result not have a large deviation due to the failure of individual sensors; when the fusion result of a greenhouse feature level is “unsuitable”, the fusion result of each data level is considered comprehensively, and the remote control agency makes a decision, which makes full use of the complementary nature of multi-sensor information resources and solves the problem of fusion of multi-source environmental information and the problem of combining conflicting environmental evaluation factors. Compared with the traditional D-S theory, the improved D-S theory reduces the probability of the “uncertainty” index in the fusion result again. The three-level fusion algorithm in this paper does not sacrifice data accuracy and greatly reduces the noise and redundancy of the original data, laying a foundation for big data analysis.

A more reasonable sum rule formula for $$\xi $$ ξ -moments $$\langle \xi ^n\rangle _{2;K_0^*}$$ ⟨ ξ n ⟩ 2 ; K 0 ∗ is suggested, which eliminates the influence brought by the fact that the sum rule of $$\langle \xi ^0_p\rangle _{3;K_0^*}$$ ⟨ ξ p 0 ⟩ 3 ; K 0 ∗ cannot be normalized in whole Borel region. More accurate values of the first ten $$\xi $$ ξ -moments, $$\langle \xi ^n\rangle _{2;K_0^*} (n = 1,2,\ldots ,10)$$ ⟨ ξ n ⟩ 2 ; K 0 ∗ ( n = 1 , 2 , … , 10 ) , are evaluated. A new light-cone harmonic oscillator (LCHO) model for $$K_0^*(1430)$$ K 0 ∗ ( 1430 ) leading-twist DA is established for the first times. By fitting the resulted values of $$\langle \xi ^n\rangle _{2;K_0^*} (n = 1,2,\ldots ,10)$$ ⟨ ξ n ⟩ 2 ; K 0 ∗ ( n = 1 , 2 , … , 10 ) via the least squares method, the behavior of $$K_0^*(1430)$$ K 0 ∗ ( 1430 ) leading-twist DA described with LCHO model is determined. Further, by adopting the light-cone QCD sum rules, we calculate the $$B_s,D_s \rightarrow K_0^*(1430)$$ B s , D s → K 0 ∗ ( 1430 ) transition form factors and branching fractions of the semileptonic decays $$B_s,D_s \rightarrow K_0^*(1430) \ell \nu _\ell $$ B s , D s → K 0 ∗ ( 1430 ) ℓ ν ℓ . The corresponding numerical results can be used to extract the Cabibbo-Kobayashi-Maskawa matrix elements by combining the relative experimental data in the future.

The excessive use of antibiotics has resulted in the contamination of the environment with antibiotic resistance genes (ARGs), posing a significant threat to public health. Wastewater treatment plants (WWTPs) are known to be reservoirs of ARGs and considered to be hotspots for horizontal gene transfer (HGT) between bacterial communities. However, most studies focused on the distribution and dissemination of ARGs in hospital and urban WWTPs, and little is known about their fate in industrial WWTPs. In this study, collected the 15 wastewater samples containing N,N-dimethylformamide (DMF) from five stages of the anaerobic anoxic aerobic (AAO) process in an industrial WWTPs. The findings revealed a stepwise decrease in DMF and chemical oxygen demand (COD) content with the progression of treatment. However, the number and abundances of ARGs increase in the effluents of biological treatments. Furthermore, the residues of DMF and the treatment process altered the structure of the bacterial community. The correlation analysis indicated that the shift in bacterial community structures might be the main driver for the dynamics change of ARGs. Interestingly, observed that the AAO process may acted as a microbial source and increased the total abundance of ARGs instead of attenuating it. Additionally, found that non-pathogenic bacteria had higher ARGs abundance than pathogenic bacteria in effluents. The study provides insights into the microbial community structure and the mechanisms that drive the variation in ARGs abundance in industrial WWTPs.

BackgroundRecent studies have suggested a relationship between gut microbiota and non-alcoholic fatty liver disease (NAFLD)/nonalcoholic steatohepatitis (NASH). However, the nature and direction of this potential causal relationship are still unclear. This study used two-sample Mendelian randomization (MR) to clarify the potential causal links.MethodsSummary-level Genome-Wide Association Studies (GWAS) statistical data for gut microbiota and NAFLD/NASH were obtained from MiBioGen and FinnGen respectively. The MR analyses were performed mainly using the inverse-variance weighted (IVW) method, with sensitivity analyses conducted to verify the robustness. Additionally, reverse MR analyses were performed to examine any potential reverse causal associations.ResultsOur analysis, primarily based on the IVW method, strongly supports the existence of causal relationships between four microbial taxa and NAFLD, and four taxa with NASH. Specifically, associations were observed between Enterobacteriales (P =0.04), Enterobacteriaceae (P =0.04), Lachnospiraceae UCG-004 (P =0.02), and Prevotella9 (P =0.04) and increased risk of NAFLD. Dorea (P =0.03) and Veillonella (P =0.04) could increase the risks of NASH while Oscillospira (P =0.04) and Ruminococcaceae UCG-013 (P=0.005) could decrease them. We also identified that NAFLD was found to potentially cause an increased abundance in Holdemania (P =0.007) and Ruminococcus2 (P =0.002). However, we found no evidence of reverse causation in the microbial taxa associations with NASH.ConclusionThis study identified several specific gut microbiota that are causally related to NAFLD and NASH. Observations herein may provide promising theoretical groundwork for potential prevention and treatment strategies for NAFLD and its progression to NASH in future.

Mulching is an important practice to reduce soil water loss and improve productivity in agriculture. To investigate the influence of dual mulching with crushed wheat straw padding and plastic film combined with different irrigation amounts on soil nutrients and the growth of edible sunflower, a field experiment was conducted in Minqin Experimental Station of Gansu Institute of Hydraulic Sciences from 2016 to 2018. The experiment adopted a two-factor design with four mulching methods: no mulching (N), straw mulching (S) at 4500 kg ha−1, plastic film mulching (F), crushed wheat straw padding and plastic film mulching (SF) at 4500 kg ha−1, and three irrigation levels: high (H=900 m3 ha−1), medium (M=750 m3 ha−1) and low (L=600 m3 ha−1). Background BANF1 is well known as a natural opponent of cyclic GMP-AMP synthase (cGAS) activity on genomic self-DNA. However, the roles of BANF1 in tumor immunity remain unclear. Here, we investigate the possible impact of BANF1 on antitumor immunity and response to immunotherapy.Methods The Cancer Genome Atlas public data were analyzed to evaluate the relevance of the expression of BANF1, patients’ survival and immune cell infiltration. We monitored tumor growth and explored the antitumor efficacy of targeting tumor-intrinsic BANF1 in combination with anti-programmed cell death protein-1 (PD-1) in MC38 or B16F10 tumor models in both immunocompetent and immunodeficient mice. Flow cytometry, immunofluorescence and T cells depletion experiments were used to validate the role of BANF1 in tumor immune microenvironment reprogramming. RNA sequencing was then used to interrogate the mechanisms how BANF1 regulated antitumor immunity.Results We show that upregulated expression of BANF1 in tumor tissues is significantly associated with poor survival and is negatively correlated with immune cell infiltration. Deficiency of BANF1 in tumor cells markedly antagonizes tumor growth in immunocompetent but not immunocompromised mice, and enhances the response to immunotherapy in murine models of melanoma and colon cancer. In the immunotherapy clinical cohort, patients with high BANF1 expression had a worse prognosis. Mechanistically, BANF1 knockout activates antitumor immune responses mediated by cGAS-synthase-stimulator of interferon genes (cGAS-STING) pathway, resulting in an immune-activating tumor microenvironment including increased CD8+ T cell infiltration and decreased myeloid-derived suppressor cell enrichment.Conclusions BANF1 is a key regulator of antitumor immunity mediated by cGAS-STING pathway. Therefore, our study provides a rational that targeting BANF1 is a potent strategy for enhancing immunotherapy for cancer with BANF1 upregulation.

While utilizing hydrothermal resources, it is necessary to reinject wastewater into the reservoir through reinjection wells to extract heat without mining groundwater. Chemical clogging is a serious problem in the process of reinjection. The precipitation of minerals can lead to reservoir clogging and the reduction of permeability. Therefore, to study the effect of chemical clogging on permeability, the weakly consolidated sandstone of the Neogene Guantao Formation geothermal reservoir in northern Shandong (Eastern China) was taken as the research object. A long-term thermal-hydro-mechanical-chemistry (THMC) coupling reinjection experiment was carried out. The results showed that when the temperature of wastewater was higher than 45 °C, there was a temporary phase of permeability enhancement in the first 10 min of reinjection. However, wastewater with higher temperatures would cause more chemical clogging eventually. XRD and ion analysis results showed that the precipitation of minerals was mainly potash feldspar, illite, calcite, and other carbonate minerals during reinjection. According to the characteristics of low-TDS wastewater in the Guantao Formation, it is recommended to adopt low-temperature wastewater reinjection and reduce the concentration of Ca2+ and Mg2+ in wastewater before reinjection. Collaborative filtering is a widely used technique in current recommendation systems.It leverages the similarity between different users or items to retrieve interactive information between users and items and recommends new items for target users.In recent years,graph learning has gradually become an emerging recommendation paradigm due to its excellent perfor-mance and scalability in graph representation learning.This paper systematically reviews the most recent research on recommendation field from the perspective of graph learning.First,we provide a taxonomy that groups the current recommendation scenarios into two categories according to the data type used,including recommendation systems based on interactive information that leverage user-item interaction data as the main data source and auxiliary information-enhanced recommendation systems that incorporate social information associated with users and items as well as the knowledge graph information.Then,we review the main approaches,fundamental algorithms and critical difficulties of current recommendation models from the perspectives of random walk,graph representation learning and graph neural networks.Finally,we summarize the main challenges of graph learning methods in the field of recommendation system and outline the possible future research directions.

Abstract Background Adaptive thermogenesis by brown adipose tissue (BAT) is important to the maintenance of temperature in newborn mammals. Cold exposure activates gene expression and lipid metabolism to provide energy for BAT thermogenesis. However, knowledge of BAT metabolism in large animals after cold exposure is still limited. Results In this study, we found that cold exposure induced expression of BAT thermogenesis genes and increased the protein levels of UCP1 and PGC1α. Pathway analysis showed that cold exposure activated BAT metabolism, which involved in cGMP-PKG, TCA cycle, fatty acid elongation, and degradation pathways. These were accompanied by decreased triglyceride (TG) content and increased phosphatidylcholine (PC) and phosphatidylethanolamine (PE) content in BAT. Conclusion These results demonstrate that cold exposure induces metabolites involved in glycerolipids and glycerophospholipids metabolism in BAT. The present study provides evidence for lipid composition associated with adaptive thermogenesis in goat BAT and metabolism pathways regulated by cold exposure.

Abstract Background The pigmentation phenotype diversity is rich in domestic goats, and identification of the genetic loci affecting coat color in goats has long been of interest. Via the detections of selection signatures, a duplication upstream ASIP was previously reported to be a variant affecting the Swiss markings depigmentation phenotype in goats. Results We conducted a genome-wide association study using whole-genome sequencing (WGS) data to identify the genetic loci and causal variants affecting the pigmentation phenotype in 65 Jintang black (JT) goats (i.e., 48 solid black vs. 17 non-classic Swiss markings). Although a single association peak harboring the ASIP gene at 52,619,845–72,176,538 bp on chromosome 13 was obtained using a linear mixed model approach, all the SNPs and indels in this region were excluded as causal variants for the pigmentation phenotype. We then found that all 17 individuals with non-classic Swiss markings carried a 13,420-bp duplication (CHI13:63,129,198–63,142,617 bp) nearly 101 kb upstream of ASIP, and this variant was strongly associated (P = 1.48 × 10− 12) with the coat color in the 65 JT goats. The copy numbers obtained from the WGS data also showed that the duplication was present in all 53 goats from three European breeds with Swiss markings and absent in 45 of 51 non-Swiss markings goats from four other breeds and 21 Bezoars, which was further validated in 314 samples from seven populations based on PCR amplification. The copy numbers of the duplication vary in different goat breeds with Swiss markings, indicating a threshold effect instead of a dose-response effect at the molecular level. Furthermore, breakpoint flanking repeat analysis revealed that the duplication was likely to be a result of the Bov-B-mediated nonallelic homologous recombination. Conclusion We confirmed that a genomic region harboring the ASIP gene is a major locus affecting the coat color phenotype of Swiss markings in goats. Although the molecular genetic mechanisms remain unsolved, the 13,420-bp duplication upstream of ASIP is a necessary but not sufficient condition for this phenotype in goats. Moreover, the variations in the copy number of the duplication across different goat breeds do not lead to phenotypic heterogeneity.

Image instance segmentation is an important part of image processing and computer vision technology about image understanding.With the development of deep learning and deep convolutional neural network,image instance segmentation method based on deep convolutional neural network has made great progress.Instance segmentation task is actually the combination of target detection and semantic segmentation,which can complete the task of recognizing the target contour in the image at thepixel level.Instance segmentation can not only locate the position of the object in the image,segment all the objects from the pixel level,but also mark different individuals of the same category in the image,which is not only the pixel level segmentation of the image,but also the instance level understanding.Firstly,the reason of image segmentation and the function of deep convolution neural network are described.Then,according to the process and characteristics of image instance segmentation methods,the research progress of image instance segmentation is introduced from two-stage and single-stage perspectives,and the advantages and disadvantages of the two methods are described in detail.Then,the design ideas of region,feature extraction and mask are summarized.In addition,the performance evaluation criteria and common public data sets of image instance segmentation methods are summarized,and on this basis,the segmentation accuracy of mainstream image instance segmentation models is compared and evaluated.Finally,it points out the problems and solutions of the current image instance segmentation,summarizes the development of image instance segmentation and prospects for the future.
