Background: Breast cancer has become a major public health problem in the current society, and its incidence rate ranks the first among Chinese female malignant tumors. This paper once again confirmed the efficacy of lncRNA in tumor regulation by introducing the mechanism of the diagnosis of breast cancer by the MIR497HG/miR-16-5p axis. Methods: The abnormal expression of MIR497HG in breast cancer was determined by RT-qPCR method, and the correlation between MIR497HG expression and clinicopathological characteristics of breast cancer patients was analyzed via Chi-square test. To understand the diagnostic potential of MIR497HG in breast cancer by drawing the receiver operating characteristic curve (ROC). The overexpressed MIR497HG (pcDNA3.1-MIR497HG) was designed and constructed to explore the regulation of elevated MIR497HG on biological function of BT549 and Hs 578T cells through Transwell assays. Additionally, the luciferase gene reporter assay and Pearson analysis evaluated the targeting relationship of MIR497HG to miR-16-5p. Results: MIR497HG was decreased in breast cancer and had high diagnostic function, while elevated MIR497HG inhibited the migration and invasion of BT549 and Hs 578T cells. In terms of functional mechanism, miR-16-5p was the target of MIR497HG, and MIR497HG reversely regulated the miR-16-5p. miR-16-5p mimic reversed the effects of upregulated MIR497HG on cell biological function. Conclusions: In general, MIR497HG was decreased in breast cancer, and the MIR497HG/miR-16-5p axis regulated breast cancer tumorigenesis, providing effective insights for the diagnosis of patients. A study published in the Plant Biotechnology Journal introduces a novel CRISPR/Cas9 system called the fluorescence marker and pollen killer-assisted CRISPR/Cas9 system (FMPKC) for efficient isolation of transgene-free edited plants in rice. The FMPKC system utilizes a DsRed2 fluorescence marker and a pollen killer to distinguish transgenic offspring and increase the proportion of non-transgenic seeds. The system was used to edit the internode length gene EUI1 and pollen fertility gene PTC1 in rice, resulting in the production of transgene-free plants. The FMPKC system shows promise for multi-target or multi-gene editing and has potential applications in rice functional genomics and breeding improvement. Harvesting sunlight into cost-effective electricity presents an enticing prospect for self-powered wearable applications. The photothermal materials with an extensive absorption are fundamental to achieve optical and thermal concentration of the sunlight for efficiency output electricity of wearable solar thermoelectric generators (STEGs). Here, we synthesize an organic charge-transfer (CT) cocrystal with a flat absorption from ultraviolet to second near-infrared region (200 to 1950 nanometers) and a high photothermal conversion efficiency (PCE) of 80.5%, which is introduced into polyurethane toward large-area nanofiber membrane by electrospinning technology. These corresponding membranes demonstrate a high PCE of 73.7% under the strain more than 80%. Sandwiched with carbon nanotube-based thermoelectric fibers, the membranes as stretchable solar absorbers of STEGs could supply a notably increase temperature gradient, processing a maximum output voltage density of 23.4 volts per square meter at 1:00 p.m. under sunlight. Background: Extracellular vesicles (EVs) from human umbilical cord mesenchymal stem cells (hUMSCs) are widely considered to be the best mediators for cell-free therapy. An understanding of their composition, especially RNA, is particularly important for the safe and precise application of EVs. Up to date, the knowledge of their RNA components is limited to NGS sequencing and cannot provide a comprehensive transcriptomic landscape, especially the long and full-length transcripts. Our study first focused on the transcriptomic profile of hUMSC-EVs based on nanopore sequencing. Methods: In this study, different EV subtypes (exosomes and microvesicles) derived from hUMSCs were isolated and identified by density gradient centrifugation. Subsequently, the realistic long transcriptomic profile in different subtypes of hUMSC-EVs was systematically compared by nanopore sequencing and bioinformatic analysis. Results: Abundant transcript variants were identified in EVs by nanopore sequencing, 69.34% of which transcripts were fragmented. A series of full-length and long transcripts was also observed and showed a significantly higher proportion of intact or near-complete transcripts in exosomes than that in microvesicles derived from hUMSCs. Although the composition of RNA biotypes transported by different EV subtypes was similar, the distribution of transcripts and genes revealed the inter-heterogeneity and intra-stability between exosomes and microvesicles. Further, 85 different expressed transcripts (56 genes) and 7 fusion genes were identified. Pathway enrichment analysis showed that upregulated-expressed genes in microvesicles were mainly enriched in multiple neurodegenerative diseases, while upregulated-expressed genes in exosomes were mainly enriched in neutrophil extracellular trap formation, suggesting different functional tendencies of EV subtypes. Conclusions: This study provides a novel understanding of different types of hUMSC-EVs, which not only suggests different transcriptome sorting mechanisms between exosomes and microvesicles, but also shows that different EV subtypes from the same source have different physiological functions, suggesting distinct clinical application prospects. Metal nanoparticles play an outstanding role in the field of wound healing due to their excellent properties, and the significance of iron, one of the most widely used metals globally, cannot be overlooked. The purpose of this review is to determine the importance of iron nanoparticles in wound-healing dressings. Prolonged, poorly healing wounds may induce infections; wound infections are a major cause of chronic wound formation. The primary components of iron nanoparticles are iron oxide nanoparticles, which promote wound healing by being antibacterial, releasing metal ions, and overcoming bacterial resistance. The diameter of iron oxide nanoparticles typically ranges between 1 and 100 nm. Magnetic nanoparticles with a diameter of less than 30 nm are superparamagnetic and are referred to as superparamagnetic iron oxide nanoparticles. This subset of iron oxide nanoparticles can use an external magnetic field for novel functions such as magnetization and functionalization. Iron nanoparticles can serve clinical purposes not only to enhance wound healing through the aforementioned means but also to ameliorate anemia and glucose irregularities, capitalizing on iron's properties. The rampant spread of explicit content across social media can leave a damaging mark on our society. Hence, the need to be vigilant in detecting and curtailing sexually explicit content cannot be overstated. As such, it becomes paramount to discern and manage sexually explicit material to curb its dissemination and safeguard our digital communities from its harmful effects. In this article, we propose a unique technique entitled attention‐enabled pooling (ABP) embedded Swin transformer‐based YOLOv3 (ASYv3) for the detection of obscene areas present in the images with a bounding box around the offensive regions. ASYv3 employs a unique two‐step approach for enhanced performance in obscene detection. In the first step, a scalable and efficient Swin transformer block is integrated, utilizing self‐attention and model parallelism to train massive models effectively. In the second phase, the embedding layer of the Swin transformer is replaced with ABP, mitigating disruption of feature context. ABP allows for the projection of raw‐valued features into linear form with proper attention to feature context information at specified locations, resulting in optimized feature extraction. The proposed ABP embedded Swin transformer‐based YOLOv3 (ASYv3) was trained with annotated obscene images (AOI) dataset. The proposed ASYv3 model surpassed the state‐of‐the‐art methods by achieving 97% testing accuracy, 96.62% precision, 97.40% sensitivity, 3.48% FPR rate, 97.37% NPV values, and 95.59% mAP values, respectively. As a hearing disorder, sensorineural hearing loss (SNHL) can be effectively detected by magnetic resonance imaging (MRI). However, the manual detection of MRI scanning is subjective, time‐consuming, and unpredictable. An accurate and automatic computer‐aided diagnosis system is proposed for SNHL detection, providing reliable references for professionals. The system first employs a wavelet entropy layer to extract features of MRI images. Then, a neural network layer is proposed as the classifier consisting of a feedforward neural network (FNN) and an adaptive‐probability PSO (APPSO) algorithm. The authors prove the rotation‐variant property of the basic particle swarm optimization (PSO) by the algebraic property of matrix transformation. The property is unsuitable for optimising parameters of neural networks. Thus, in APPSO, the authors integrate the new update rules based on all‐dimensional variation and adaptive‐probability mechanism into the basic PSO, which can improve its searching ability without losing population diversity. The authors compare APPSO‐NN with FNN trained by five popular evolutionary algorithms. The simulation results show that APPSO performs best in training FNN. The method also compares with six state‐of‐the‐art methods. The simulation results show that the best performance in sensitivity and overall accuracy of hearing loss classification, which proves that the method is effective and promising for SNHL detection. Countless cybercrime instances have shown the need for detecting and blocking obscene material from social media sites. Deep learning methods (DLMs) outperformed in recognizing obscene content flooded on many online platforms. However, these contemporary DLMs primarily treat the recognition of obscene content as a simple task of binary classification, rather than focusing on the labelling of obscene areas. Hence, many of these methods could not pay attention to the fact that misclassification samples are so diverse. Therefore, this paper focuses on two aspects (i) developing a deep learning model that could classify and label the obscene portion, and (ii) generating a labelled obscene image dataset with a wide variety of obscene samples to minimize the risks of inaccurate recognition. We have proposed a method named S3Pooling based bottleneck attention module (BAM) embedded MobileNetV2‐YOLOv3 (SBMYv3) for automatic detection of obscene content using an attention mechanism and a suitable pooling strategy. The key contributions of our article are: (i) generation of a well‐labelled obscene image dataset with a variety of augmentation strategies using Pix‐2‐Pix GAN (ii) modifications to the backend architecture of YOLOv3 using MobileNetV2 and BAM to ensure focused and accurate feature extraction, and (iii) selection of an optimal pooling strategy, that is, S3Pooling strategy, while taking the design of the feature extractor into account. The tremendous personal and economic burden worldwide caused by low back pain (LBP) has been surging in recent years. While intervertebral disc degeneration (IVDD) is the leading cause of LBP and vast efforts have been made to develop effective therapies, this problem is far from being resolved, as most treatments, such as painkillers and surgeries, mainly focus on relieving the symptoms rather than reversing the cause of IVDD. However, as stem/progenitor cells possess the potential to regenerate IVD, a deeper understanding of the early development and role of these cells could help to improve the effectiveness of stem/progenitor cell therapy in treating LBP. Single-cell RNA sequencing results provide fresh insights into the heterogeneity and development patterns of IVD progenitors; additionally, we compare mesenchymal stromal cells and IVD progenitors to provide a clearer view of the optimal cell source proposed for IVD regeneration. In this work, the mechanical behavior and energy absorption characteristics of flexible fabric under hypervelocity impact (HVI) were investigated. Basalt fabric, ultra-high molecular weight polyethylene (UHMWPE) fabric, and aluminum alloy (Al) plate were chosen to be the sample materials for their excellent mechanical properties and applicative prospect in spacecraft shielding. HVI experiments had been conducted with the help of a two-stage light-gas gun facility, wherein Al projectile with 3.97 mm diameter was launched at velocities in the range 4.1~4.3 km/s. Impact conditions and areal density were kept constant for all targets. The microstructural damage morphology of fiber post-impact was characterized using a scanning electron microscope (SEM). Analysis results show that a brittle fracture occurred for Basalt fiber during HVI. On the contrary, the ductile fractures with large-scale plastic deformation and apparent thermal softening/melting of the material had happened on the UHMWPE fiber when subjected to a projectile impact. According to the HVI shielding performance and microstructural damage analysis results, it can be inferred that ductile fractures and thermal softening/melting of the material were the prevailing energy absorption behaviors of UHMWPE fabric, which leads to absorbing more impact energy than Basalt fabric and eventually, contributes the superior shielding performance. Back-arc extension has been well documented in subduction plate tectonic regimes. However, the reasons why back-arc extensions are associated with some subduction systems but not others have remained elusive. Here, spatio-temporal variations in the composition of Cenozoic basalts in the northeast Asian continental margin are used to constrain the mechanism for episodic back-arc extensions. Using geochemical data sensitive to tectonic affinity, we show that typical volcanic arc compositions are located in the eastern margin of northeast Asia, whereas coeval intraplate volcanic compositions are located in the western part of northeast Asia, and that the intraplate and arc volcanism exhibit two eastward shifts, from 52 Ma to 33 Ma and from 33 Ma to 21 Ma. Intraplate basalts dated at ca. 11 Ma display a weak, arc-like geochemical signature, which suggests that the upwelling of asthenospheric mantle resulted in the remelting of previously melt-extracted lithospheric mantle modified by slab-derived fluids and the cessation of back-arc extensions. Thus, we propose that the eastward mantle flow resulted in eastward shifts of back-arc extensions that led to the development of extensive Cenozoic arc and intraplate volcanism in the northeast Asian continental margin. Background: Tracheal stents can be placed in a narrow position in the human trachea to ensure smooth breathing. And the stent will deform during service by the influence of the physiological environment or random excitations, such as coughing. Methods: This paper divides the vibration into periodic and random vibrations according to the different pressures. And a coupling vibration model was established by analyzing the contact relationship between the stent and the trachea tissue. And this study discusses the influence of tracheal diameter, respiratory pressure, and frequency on the stent vibration characteristics through Ansys simulation. In addition, the nonlinear equations were solved by the Matlab numerical analysis method, which could help analyze the influence of cough intensity on the stability of the tracheal stent system. Results: The results showed that when tracheal stenosis occurred in the trachea's more significant grade, the trachea stent was more likely to fall off when treated with a tracheal stent. With the increase in respiratory frequency and pressure, the deformation of the tracheal stent is more considerable. Moreover, the frequency of normal cough hardly affects the stability of the stent system, while the excitation force and damping coefficient value greatly influence the system. When the excitation force of the cough exceeds the critical importance of 20 N, the tracheal stent is prone to fall off. This study comprehensively obtained the forced vibration characteristics of the stent under service conditions, which could make up for the shortage of the vibration theory of the stent. Different nutrient supply brings about changes in leaf stoichiometry, which may affect growth rate and primary production of plants. Invasion of alien plants is a severe threat to biodiversity and ecosystem worldwide. A pot experiment was conducted by using three stoloniferous alien plants Wedelia trilobata, Alternanther philoxeroides and Hydrocotyle vulgaris to investigate effects of nutrient supply on their leaf stoichiometry and relative growth rate. Different nitrogen or phosphorus supply was applied in the experiment (N1:1 mmol L-1, N2:4 mmol L-1, and N3:8 mmol L-1, P1:0.15 mmol L-1, P2:0.6 mmol L-1 and P3:1.2 mmol L-1). Nitrogen and phosphorus concentrations in leaves of the three alien plants significantly increased with increase of nitrogen supply. With increase of phosphorus supply, nitrogen or phosphorus concentration of leaf was complex among the three alien plants. N:P ratio in leaf of the three alien plants subjected to different levels of nutrient supply was various. A positive correlation between relative growth rate and N:P ratio of the leaf is observed in W. trilobata and A. philoxeroides suffering from N-limitation. A similar pattern was not observed in Hydrocotyle vulgaris. We tentatively concluded that correlations between relative growth rate and N: P ratio of the leaf could be affected by species as well as nutrient supply. It is suggested that human activities, invasive history, local abundance of species et al maybe play an important role in the invasion of alien plants as well as relative growth rate. Protein–protein interactions (PPIs) are fundamental to many biological processes. The coevolution-based prediction of interacting residues has made great strides in protein complexes that are known to interact. A multiple sequence alignment (MSA) is the basis of coevolution analysis. MSAs have recently made significant progress in the protein monomer sequence analysis. However, no standard or efficient pipelines are available for the sensitive protein complex MSA (cpxMSA) collection. How to generate cpxMSA is one of the most challenging problems of sequence coevolution analysis. Although several methods have been developed to address this problem, no standalone program exists. Furthermore, the number of built-in properties is limited; hence, it is often difficult for users to analyze sequence coevolution according to their desired cpxMSA. In this article, we developed a novel cpxMSA approach (cpxDeepMSA. We used different protein monomer databases and incorporated the three strategies (genomic distance, phylogeny information, and STRING interaction network) used to join the monomer MSA results of protein complexes, which can prevent using a single method fail to the joint two-monomer MSA causing the cpxMSA construction failure. We anticipate that the cpxDeepMSA algorithm will become a useful high-throughput tool in protein complex structure predictions, inter-protein residue-residue contacts, and the biological sequence coevolution analysis. To make full use of the flexible adjustment capability of DC tie-lines in multiarea AC/DC systems and to coordinate the generation resources and load demand of multiarea AC/DC systems, this paper presents an intraday two-stage hierarchical optimal scheduling model for multiarea AC/DC systems based on analytical target cascading (ATC). To avoid repeated adjustment and overadjustment of DC tie-lines after wind power integration, a two-stage rolling coordinated scheduling model for the area subsystem based on model predictive control (MPC) is proposed. The two-stage rolling coordinated scheduling method takes into consideration the influence of the predicted value in the future finite time domain and the latest measured DC tie-line power on the current scheduling state. Based on the ATC and the area decomposition criterion of the AC/DC grid, an optimal scheduling model for the upper-level system is proposed that takes into consideration the DC tie-line adjustment constraints and the area coupling constraints. The optimal scheduling model for the upper-level system formulates the two-stage DC tie-line plan for the multiarea AC/DC system, and the two-stage rolling coordinated scheduling model of the area subsystem solves the subproblems of the generation plan for each area subsystem in a parallel manner considering the area coupling constraints. The proposed method can achieve intraday two-stage decoupling scheduling of multiarea AC/DC systems and promote the cross-area consumption of large-scale wind power through flexible adjustment of the DC tie line. This approach also reduces the communication burden between the area subsystems and ensures the efficiency of the solution algorithm. • Based on analytical target cascading technology (ATC), we integrate the hierarchical control structure of decomposition-coordination into the model predictive control (MPC) method to achieve decentralized dispatch for multiarea AC/DC systems. • A two-stage rolling coordinated scheduling model for the area subsystem considering the area coupling constraints based on the MPC is proposed. • Based on the ATC and the area decomposition criterion of the AC/DC grid, an optimal scheduling model for the upper-level system is proposed. • Flexible adjustment of the DC tie-line and decoupling scheduling of multiarea AC/DC systems are achieved. • The hierarchical optimal scheduling model reduces the communication burden between the subsystems and ensures the efficiency of the solution algorithm. Background: Uniparental disomy (UPD) is a condition in which both chromosomes are inherited from the same parent, except for imprinting disorders. Uniparental isodisomy (UPiD) may result in a homozygous variant contributing to an autosomal recessive disorder in the offspring of a heterozygous carrier. Junctional epidermolysis bullosa intermediate (JEB intermediate) is an autosomal recessive inherited disease that is associated with a series of gene variants, including those of COL17A1. Case presentation: We report the first case of complete paternal UPiD of chromosome 10 harbouring a novel homozygous variant in COL17A1: c.1880(exon23)delG (p.G627Afs*56). This variant led to the clinical phenotype of junctional epidermolysis bullosa intermediate in a 5-year-old child. Trio-whole exome sequencing (Trio-WES) and in silico data analysis were used for variant identification, Sanger sequencing was performed for variant validation, and pathological examination was performed as the gold standard for phenotype confirmation. Conclusions: We recommend the use of WES as a first-tier test for the diagnosis of epidermolysis bullosa, especially for paediatric patients. Galectins are a family of proteins with at least one carbohydrate-recognition domain. Galectins are present in various tissues and organs and participate in different physiological and pathological molecular reactions in vivo. Wound healing is the basic process of traumatic disease recovery. Wound healing involves three overlapping stages: inflammation, proliferation, and remodelling. Furthermore, a comparison of wound healing with the tumour microenvironment revealed that galectin plays a key role in the wound healing process. The current review describes the role of galectin in inflammation, angiogenesis, re-epithelialisation, and fibrous scar formation and evaluates its potential as a therapeutic drug for wounds. Background: The principal objective of this project was to investigate the Epidermal Growth Factor Receptor (EGFR) gene mutation characteristics of lung cancer patients, which can provide a molecular basis for explaining the clinicopathological features, epidemiology and use of targeted therapy in lung cancer patients in the coal-producing areas of East Yunnan. Methodology: We collected 864 pathologically confirmed lung cancer patients' specimens in First People's Hospital of Qujing City of Yunnan Province from September 2016 to September 2021. We thereafter employed Next Generation Sequencing (NGS) technology to detect all exons present in the EGFR gene. Results: The overall mutation frequency of the EGFR gene was 47.22%. The frequency of EGFR gene mutations in the tissue, plasma, and cytology samples were found to be 53.40%, 23.33%, and 62.50%, respectively. Univariate analysis indicated that the coal-producing areas and Fuyuan county origin were significantly associated with relatively low EGFR gene mutation frequency. Female, non-smoking history, adenocarcinoma, non-brain metastasis, and tissue specimens were found to be related to high EGFR gene mutation frequency. Multivariate logistic regression analysis suggested the lung cancer patients in the central area of Qujing City, stage Ia, non-coal-producing areas, non-Fuyuan origin, and non-Xuanwei origin were more likely to develop EGFR gene mutations. The most common mutations were L858R point mutation (33.09%) and exon 19 deletion (19-del) (21.32%). Interestingly, the mutation frequency of G719X (p = 0.001) and G719X + S768I (p = 0.000) in the coal-producing areas were noted to be more significant than those in non-coal-producing regions. Conclusion: This findings of this study might be important in establishing the correlation between routine using NGS for EGFR gene mutation diagnosis and clinical practice in the lung cancer patients. Effect of solution treatment on microstructure and mechanical properties of Al-12Si-4Cu-2Ni-0.8Mg-0.2Gd alloy was investigated. Results show the Si particles become stable and more intermetallic compounds dissolve in the matrix after solution treatment at 500 °C for 2 h followed by 540 °C for 3 h (T4). The skeleton-like Al3CuNi develops into two parts in the T4 alloy: one is Al3CuNi which has the framework shape; the other is intermetallics including the Al3CuNi (size: 5–10 µm) and AlSiCuNiGd phases (size: ≤5 µm) with complex structure. Adding 0.2% Gd can improve the mechanical properties of the alloys after two-step solution treatment (500 °C/2 h followed by 540 °C/3 h), the hardness of the alloy increases from 130.9 HV to 135.8 HV compared with the alloy with one-step solution treatment (500 °C/2 h), the engineering strength increases from 335.45 MPa to 352.03 MPa and the fracture engineering strain increases from 1.44% to 1.67%. Remote sensing of surface pressure from space is critical; differential absorption LIDAR and differential absorption radar are only two kinds of remote sensing instruments with this potential. The differential absorption LIDAR works in integral path mode from the satellite in low-Earth orbit. It measures the differential optical depth of the Oxygen A-band, and the surface pressure is thereafter obtained by performing circle-iterative calculation. Performance evaluation of the differential absorption LIDAR model was conducted with respect to the advanced system parameters of the space instrument, Low echo pulse energy at ocean surface and the challenging calculation of repetitive cumulative average of echo on uneven land surface yielded random errors in surface pressure measurement. On the other hand, uncertain atmospheric temperature and water vapor mixture profiles resulted in systematic error of surface pressure. Consequently, controlling the error of surface pressure within 0.1% proved challenging. Under a strict implementation of the error budget, the time resolution is 6.25 s and along-orbit distance resolution is 44km, and the results showed that 765.6735/765.4637 nm is suitable as the working wavelength pair. Further, error could be expected to within 0.2-0.3% for the cumulative average of 625 ocean surface laser pulse echoes, cumulative average of more than 144 pulse echoes on land, and observation from the 400km orbit. Aim: Fruit category classification is important in factory packing and transportation, price prediction, dietary intake, and so forth. Methods: This study proposed a novel artificial intelligence system to classify fruit categories. First, 2D fractional Fourier entropy with rotation angle vector grid was used to extract features from fruit images. Afterwards, a five‐layer stacked sparse autoencoder was used as the classifier. Results: Ten runs on the test set showed our method achieved a micro‐averaged F1 score of 95.08% for an 18‐category fruit dataset. Within the framework of nonrelativistic QCD (NRQCD) factorization, we calculate the next-to-leading-order (NLO) perturbative corrections to the radiative decay Υ → ηc(χcJ) + γ. Both the helicity amplitudes and the helicity decay widths are obtained. It is the first computation for the processes involving both bottomonium and charmonium at two-loop accuracy. By employing the Cheng-Wu theorem, we are able to convert most of complex-valued master integrals (MIs) into real-valued MIs, which makes the numerical integration much efficient. Our results indicate the O α s corrections are moderate for ηc and χc2 production, and are quite marginal for χc0 and χc1 production. It is impressive to note the NLO corrections considerably reduce the renormalization scale dependence in both the decay widths and the branching fractions for χcJ, and slightly improve that for ηc. With the NRQCD matrix elements evaluated via the Buchmüller-Tye potential model, we find the decay width for ηc production is one-order-of-magnitude larger than χcJ production, which may provide a good opportunity to search for Υ → ηc + γ in experiment. In addition, the decay width for χc1 production is several times larger than those for χc0,2. Finally, we find the NLO NRQCD prediction for the branching fraction of Υ → χc1 + γ is only half of the lower bound of the experimental data measured recently by Belle. Moreover, there exists serious contradiction between theory and experiment for Υ → ηc + γ. The discrepancies between theory and experiment deserve further research efforts. [ABSTRACT FROM AUTHOR]

We report a molecular switching ensemble whose states may be regulated in synergistic fashion by both protonation and photoirradiation. This allows hierarchical control in both a kinetic and thermodynamic sense. These pseudorotaxane-based molecular devices exploit the so-called Texas-sized molecular box (cyclo[2]-(2,6-di(1Himidazol- 1-yl)pyridine)[2](1,4-dimethylenebenzene); 14+, studied as its tetrakis-PF6 2 salt) as the wheel component. Anions of azobenzene- 4,40-dicarboxylic acid (2H+•2) or 4,40-stilbenedicarboxylic acid (2H+•3) serve as the threading rod elements. The various forms of 2 and 3 (neutral, monoprotonated, and diprotonated) interact differently with 14+, as do the photoinduced cis or trans forms of these classic photoactive guests. The net result is a multimodal molecular switch that can be regulated in synergistic fashion through protonation/deprotonation and photoirradiation. The degree of guest protonation is the dominating control factor, with light acting as a secondary regulatory stimulus. The present dual input strategy provides a complement to more traditional orthogonal stimulus-based approaches to molecular switching and allows for the creation of nonbinary stimulus-responsive functional materials. The mechanism for the radiation efficiency local minimum in antennas, especially for open-slot antennas and T-shaped antennas, has been investigated. It is found that the local minimum in those antennas always corresponds to an ideal differential mode. Furthermore, the frequency position of the ideal differential mode is decided by the parasitic parts of the antennas. Simple methods for estimating the local minimum position are proposed based on the mechanism we have found. In addition, the duality relation between those two types of antennas is also discussed. The Qinghai–Tibetan Plateau (QTP) played a crucial role in shaping the biodiversity in Asia during the Cenozoic. However, fossil records attributed to insects are still scarce from the QTP, which limits our understanding on the evolution of biodiversity in this large region. Fulgoridae (lanternfly) is a group of large planthopper in body size, which is found primarily in tropical regions. The majority of the Fulgoridae bear brilliant colors and elongated heads. The fossil records of Fulgoridae span from the Eocene to Miocene in the Northern Hemisphere, and only a few fossil species from Neogene deposits have been reported in Asia so far. Here, we report a new fossil record of Fulgoridae from the middle Eocene Lunpola Basin, central QTP. The specimen is in lateral compression, with complete abdomen, thorax, and part of the wings preserved, while most of the head is missing. It belongs to the "lower Fulgoroidea" judging by several strong lateral spines on the hind tibia and a row of teeth at the apex of the second metatarsomere. This fossil specimen is assigned to Fulgoridae by comparison with nine families of the "lower Fulgoroidea". The specimen represents the earliest Fulgoridae fossil record in Asia and was considered a new morphotaxon based on the peculiar legs and wings. Based on the modern distribution of fulgorid and other paleontological evidence, we suggest a warm climate with relatively low elevation during the middle Eocene in the central QTP. Therefore, this new fossil record not only provides important information on insect diversity in the middle Eocene, but also gives new evidence on the paleoenvironment in the core area of the QTP from the perspective of an insect. Mutations in the DNAJB6 gene have been identified as rare causes of myofibrillar myopathies. However, the underlying pathophysiologica mechanisms remain elusive. DNAJB6 has two known isoforms, including the nuclear isoform DNAJB6a and the cytoplasmic isoform DNAJB6b, which was thought to be the pathogenic isoform. Here, we report a novel recessive mutation c.695_699del (p. Val 232 Gly fs*7) in the DNAJB6 gene, associated with an apparently recessively inherited late onset distal myofibrillar myopathy in a Chinese family. Notably, the novel mutation localizes to exon 9 and uniquely encodes DNAJB6a. We further identified that this mutation decreases the mRNA and protein levels of DNAJB6a and results in an age-dependent recessive toxic effect on skeletal muscle in knock-in mice. Moreover, the mutant DNAJB6a showed a dose-dependent anti-aggregation effect on polyglutamine-containing proteins in vitro. • The impact of precipitation, slope, and swamp wetland ratio on water yield is not affected by spatial scale and watershed. • Identifying key areas for ecosystem services combined with multiple scenarios can help to take ecological restoration. • The water yield in the Baihe basin is more sensitive on climate change, while the water yield in the Heihe basin is more easily affected by land use changes. • The economic development and ecosystem services can both be ensured by wetland ecological restoration. Water supply is one of the core ecosystem services functions for wetland. Due to the important status of Zoige wetland, assessment of water supply in Zoige wetland is significant for ecological security all over the world. In this paper, Heihe and Baihe basins in Zoige wetland were taken as research objects, the water yield module of the Integrated Valuation of Ecosystem Services and Trade-offs (InVEST) model was used to spatially quantify water supply, with the aims of revealing the driving mechanism of water supply under different basins and spatial scale. We observed the spatial pattern change of water yield under baseline and eleven future scenarios and used the ANOVA of repeated measures and multiple linear regression model to explore the patterns and driving factors of water supply in Heihe and Baihe rivers basins. The research results were as follow. (1) The precipitation is main factor to improve water yield, but this intensity is different in different basins and spatial scale. Topography, climate, vegetation, and percentage of wetland conditions have important impacts on water yield services. However, the impacts of these conditions on water yield were various significantly across different watersheds and spatial scales (excluding factors such as precipitation, slope, and percentage of swamp wetland). (2) The spatial pattern of water yield main was affected by land use change and the amount of water yield main was affected by future climate change. (3) Water yield in the Baihe basin was more sensitive to climate change, while the Heihe basin is more sensitive to land use changes. Wetland ecological restoration strategies can effectively improve regional water yield, especially in the Heihe basin. (4) In all scenarios, the southeast regions of the study region were both the main hot spots of water yield. Traditional lateral flow immunoassays (LFIA) suffer from insufficient sensitivity, difficulty for quantitation, and susceptibility to complex substrates, limiting their practical application. Herein, we developed a polyethylenimine (PEI)-mediated approach for assembling high-density Au nanoshells onto Fe 3 O 4 nanoclusters (MagAu shell) as LFIA labels for integrated enrichment and photothermal/colorimetric dual-mode detection of SARS-CoV-2 nucleocapsid protein (N protein). PEI layer served not only as "binders" to Fe 3 O 4 nanoclusters and Au nanoshells, but also "barriers" to ambient environment. Thus, MagAu shell not only combined magnetic and photothermal properties, but also showed good stability. With MagAu shell , N protein was first separated and enriched from complex samples, and then loaded to the strip for detection. By observation of the color stripes, qualitative detection was performed with naked eye, and by measuring the temperature change under laser irradiation, quantification was attained free of sophisticated instruments. The introduction of Fe 3 O 4 nanoclusters facilitated target purification and enrichment before LFIA, which greatly improved the anti-interference ability and increased the detection sensitivity by 2 orders compared with those without enrichment. Moreover, the high loading density of Au nanoshells on one Fe 3 O 4 nanocluster enhanced the photothermal signal of the nanoprobe significantly, which could further increase the detection sensitivity. The photothermal detection limit reached 43.64 pg/mL which was 1000 times lower than colloidal gold strips. Moreover, this method was successfully applied to real samples, showing great application potential in practice. Plasmonic physical unclonable function (PUF) labels have been proven to be an effective anti-counterfeiting strategy, but their high tunability and multi-encoded capabilities remain to be explored for higher encoding capacity (EC). Herein, the PUF labels with high security level are prepared, which are constructed by the micro-domains of plasmonic nanostructures. The micro-domains are formed by mask-assisted ultraviolet (UV) exposure and 50 nm gold nanoparticles (Au NPs) electrostatic adsorbed on the unexposed area. And a robotic wet-chemical system is invented to modulate the in situ epitaxial growth of Au NPs by programming the growth conditions for human-free preparation and mass production. Three random processes, including mask-assisted UV photochemical reaction, electrostatic self-assembly, and in situ epitaxial growth, make sure multiscale random features. Thus, two-dimensional views with different resolutions can be obtained by three random optical imaging signals-bright field (BF), Raman imaging (RI), and dark field (DF), and the random signals will change with the modulation methods, leading to higher EC. Through analyzing the random features, a robust image authentication system is built, and the EC is calculated as 1.8 × 104966 in a 50 × 50 μm2 domain for each modulation method. A nonlinear device is presented for simultaneous vibration absorption and energy harvesting. This device is composed of a nonlinear energy sink and a piezoelectric energy harvester (NES-EH), and its core component is the simply-supported bi-stable piezoelectric composite laminate (BPCL). Its outstanding advantages are light weight, simple and reliable structure. The theoretical model is developed to model a harmonically excited linear oscillator coupled with NES-EH. The Runge–Kutta method and the harmonic balance method (HBM) are used to solve the theoretical model numerically and analytically. The vibration suppression efficiency is utilized to quantify the vibration absorption performance, while the energy harvesting performance is assessed by bandwidth for energy harvesting and maximum voltage within the bandwidth. The effects of the excitation amplitude, the concentrated mass of NES-EH, the length and width of BPCL, and the load resistance are considered on the performances of NES-EH. The vibration absorption and energy harvesting performances of NES-EH can be improved by adjusting these parameters. In addition, the proposed NES-EH can achieve the goal of higher power density and normalized power density under low-amplitude excitation and simultaneously have a good vibration absorption effect. Clonal integration of defense or stress signal induced systemic resistance in leaf of interconnected ramets. However, similar effects of stress signal in root are poorly understood within clonal network. Clonal fragments of Centella asiaticas with first-young, second-mature, third-old and fourth-oldest ramets were used to investigate transportation or sharing of stress signal among interconnected ramets suffering from low water availability. Compared with control, oxidative stress in root of the first-young, second-mature and third-old ramets was significantly alleviated by exogenous ABA application to the fourth-oldest ramets as well as enhancement of antioxidant enzyme (SOD, POD, CAT and APX) activities and osmoregulation ability. Surface area and volume in root of the first-young ramets were significantly increased and total length in root of the third-old ramets was significantly decreased. POD activity in root of the fourth-oldest and third-old ramets was significantly enhanced by exogenous ABA application to the first-young ramets. Meanwhile, total length and surface area in root of the fourth-oldest and third-old ramets were significantly decreased. Ratio of belowground to aboveground biomass in the whole clonal fragments was significantly increased by exogenous ABA application to the fourth-oldest or first-young ramets. It is suggested that transportation or sharing of stress signal may induce systemic resistance in root of interconnected ramets. Specially, transportation or sharing of stress signal against phloem flow was observed in the experiment. Possible explanation is that rapid recovery of foliar photosynthesis in first-young ramets subjected to exogenous ABA application can partially reverse phloem flow within clonal network. Neuroimaging fusion can achieve higher temporal and spatial resolution, enhance contrast, correct imaging distortions, and bridge physiological and cognitive information. In this study, we analyzed over 450 references from PubMed, Google Scholar, IEEE, ScienceDirect, Web of Science, and various sources published from 1978 to 2020. We provide a review that encompasses (1) an overview of current challenges in multimodal fusion (2) the current medical applications of fusion for specific neurological diseases, (3) strengths and limitations of available imaging modalities, (4) fundamental fusion rules, (5) fusion quality assessment methods, and (6) the applications of fusion for atlas-based segmentation and quantification. Overall, multimodal fusion shows significant benefits in clinical diagnosis and neuroscience research. Background and Purpose: 9-Phenanthrol, known as a specific inhibitor of the transient receptor potential melastatin 4 (TRMP4) channel, has been shown to modulate cardiac electrical activity and exert antiarrhythmic effects. However, its pharmacological effects remain to be fully explored. Here, we tested the hypothesis that cardiac sodium current inhibition contributes to the cardioprotective effect of 9-phenanthrol.Experimental Approach: Single ventricular myocytes (VMs) and Purkinje cells (PCs) were enzymatically isolated from rabbits. Arterially perfused rabbit wedge preparations were also used, and transmural electrocardiogram and endocardial action potentials (APs) were simultaneously recorded. Wild-type and mutated human recombinant SCN5A were expressed in HEK293 cells. Anemonia toxin II (ATX-II) was used to amplify the late sodium current (INaL ) and induce arrhythmias. Whole-cell patch clamp technique was used to record APs and ionic currents.Key Results: 9-Phenanthrol (10-50 μM) stabilized ventricular repolarization and abolished arrhythmias induced by ATX-II in both isolated VMs, PCs and wedge preparations. Further study revealed that 9-phenanthrol modulated the gating properties of cardiac sodium channels and dose-dependently inhibited INaL and peak sodium current (INaP ) in VMs with an IC50 of 18 and 71.5 μM respectively. Its ability to inhibit INaL was further confirmed in PCs and HEK293 cells expressing SCN5A mutations.Conclusions and Implications: Our results indicate that 9-phenanthrol modulates the gating properties of cardiac sodium channels and inhibits INaL and INaP , which may contribute to its antiarrhythmic and cardioprotective effects. Although various previous studies have reported that the experimentally induced refractive errors interfered with postural control, few studies have demonstrated the optical correction effect of wearing glasses. The purpose of this study was to investigate whether wearing full corrected glasses to correct myopia and hyperopia can have a positive effect on postural stability. To this end, a total of 34 subjects (19 males and 15 females) of an average age of 22.38 ± 2.41-years-old participated in this study. After measuring the full corrected powers of refractive errors of subjects through subjective refraction, updated glasses were provided to 17 myopic subjects and first time glasses were provided to 17 hyperopic subjects as full corrected glasses, respectively. Postural evaluation was carried out using the TETRAX biofeedback system, after which we compared and analyzed the postural instability index and sway power index before and after wearing full corrected glasses. When updated and old glasses for correcting myopia were worn, the postural instability index was significantly reduced, and the sway power index was statistically decreased only in the mid-high frequency region associated with the somatic system, compared to the no glasses state, respectively. However, after wearing first time glasses for hyperopia correction, no significant difference was found in the postural instability index or sway power index. We suggest that providing optimal visual information through the optical correction of myopic refractive error is a useful approach that can lead to synergistic effects of somatic functions involved in postural control. Consequently, we demonstrated that wearing glasses to fully correct the refractive errors has a positive effect on increasing postural control in static posture. Background: The safety of hypoglycemic drugs should be paid more attention to in elderly patients with type 2 diabetes mellitus due to their concomitant diseases, physiological decline of liver and kidney function and cognitive decline. The aim of this study was to evaluate the efficacy and safety of DPP-4 inhibitors in elderly patients with type 2 diabetes mellitus. Methods: From January 2010 to November 2018, 300 patients with type 2 diabetes mellitus who were over 60 years old were enrolled in the outpatient clinic of Geriatric Medical Center. Their medication records and follow-up medical records were used for retrospective analysis. The duration of treatment with DPP-4 inhibitors was more than 3 months. The changes of fasting blood glucose (GLU), glycosylated hemoglobin (HbA1C), body weight, body mass index (BMI) and liver and kidney function were compared before and after treatment. Results: The average age of 300 patients (212 males and 88 females) was 73.7 ± 9.1 years old, BMI was 26.5 ± 2.8 kg/m2 and the duration of diabetes was 10.7 ± 8.2 years. The results of retrospective analysis showed that HbA1C decreased by 0.27% after treatment (P < 0.001). In the group of DPP-4 inhibitors used for less than 12 months, there was no difference in liver transaminase (ALT and AST) between before and after treatment, whereas in the group of DPP-4 inhibitors used formore than 12 months, liver transaminase decreased statistically compared with after treatment (P < 0.001). The incidence of fatty liver in elderly diabetic patients decreased after using DPP-4 inhibitors. There was no significant change in serum creatinine level and creatinine clearance rate in elderly patients with type 2 diabetes mellitus after treatment of DPP-4 inhibitor. In addition, the body weight and BMI of the patients decreased significantly (P < 0.001). No hypoglycemic reaction and gastrointestinal discomfort were found in the medical records. Conclusion: After DPP-4 inhibitors were used in elderly patients with type 2 diabetes mellitus, the elevated glycosylated hemoglobin could be controlled with improved safety of liver and kidney, and might have the effect of weight loss. Lung adenocarcinoma (LUAD) is a common malignancy; however, the majority of its underlying molecular mechanisms remain unknown. In the present study, weighted gene co-expression network analysis was applied to construct gene co-expression networks for the GSE19804 dataset, in order to screen hub genes associated with the pathogenesis of LUAD. In addition, with the aid of the Database for Annotation, Visualization and Integrated Discovery, Gene Ontology, and Kyoto Encyclopedia of Genes and Genomes, pathway enrichment analyses were performed on the genes in the selected module. Using the GSE40791 dataset and The Cancer Genome Atlas database, the hub genes were identified. It was discovered that the turquoise module was the most significant module associated with the tumor stage of LUAD. After performing functional enrichment analyses, it was indicated that the turquoise module was mainly enriched in signal transduction. Additionally, at the transcriptional and translational level, nine hub genes were identified and validated: Carbonic anhydrase 4 (CA4), platelet and endothelial cell adhesion molecule 1 (PECAM1), DnaJ member B4 (DNAJB4), advanced glycosylation end-product specific receptor (AGER), GTPase, IMAP family member 6 (GIMAP6), chromosome 10 open reading frame 54 (C10orf54), dedicator of cytokinesis 4 (DOCK4), Golgi membrane protein 1 (GOLM1) and platelet activating factor acetylhydrolase 1b catalytic subunit 3 (PAFAH1B3). CA4, PECAM1, DNAJB4, AGER, GIMAP6, C10orf54 and DOCK4 were expressed at lower levels in the tumor samples, whereas GOLM1 and PAFAH1B3 were highly expressed in tumor samples. In addition, all hub genes were associated with prognosis. In conclusion, one module and nine genes were recognized to be associated with the tumor stage of LUAD. Various microRNAs (miRNAs) are of importance in the development of colon cancer, but most of the mechanisms of the miRNAs are still unclear. In order to clarify the hub miRNAs and their roles in colon cancer development, GSE98406 was used to screen hub miRNAs by bioinformatics analysis. 46 DE-miRNAs (14 were upregulated and 32 were downregulated) and 1738 target genes of DE-miRNAs were ascertained. miRNAs-gene-networks and miRNAs-GO-networks were built to get more knowledge about the function of candidate miRNAs. After validation, three miRNAs (miR-17-5p, miR-182-5p and miR-200a-3p) were recognized to be hub miRNAs associated with the progression of colon cancer. Objective: Using three-dimensional speckle-tracking echocardiography (3D-STE), we aimed to evaluate left ventricular (LV) function in type 2 diabetes mellitus (T2DM) patients with non-alcoholic fatty liver disease (NAFLD). Methods: In total, 97 T2DM patients were categorized into three groups based on hepatic ultrasonography: group A (those without NAFLD, n=30), group B (those with mild NAFLD, n=32), and group C (those with moderate-to-severe NAFLD, n=35). Our conventional echocardiographic parameters included transmitral peak early and late diastolic velocity (E and A), septal and lateral early (e') mitral annular diastolic tissue velocities, and left atrial maximum volume index (LAVImax). LV end-diastolic and -systolic volume, LV mass index (LVMI), and LV ejection fraction were measured using real-time three-dimensional echocardiography. The 3D-STE parameters included LV global radial strain (GRS), global longitudinal strain (GLS), global area strain (GAS), and global circumferential strain (GCS). Results: Our results showed that in group C, GCS, GRS, GLS, GAS, and septal and lateral e' velocity decreased, whereas average E/e' and LAVImax increased compared to groups B and A (p<0.05). Multiple linear regression analysis showed that NAFLD is independently associated with 3D-STE parameters, and glycosylated hemoglobin also has negative impacts on all LV 3D strains. Conclusion: When combined with conventional echocardiography, 3D-STE can assess LV function effectively in T2DM patients with NAFLD. The present paper deals with the taxonomy of Selaginella from Nepal based on the examination of herbarium collections housed in major herbaria of Europe and Asia (with additional collections from virtual herbaria). In this paper, an outer rotor interior permanent magnet synchronous machine (IPMSM) with hybrid permanent magnet (PM) material is proposed. The key is to adopt the outer rotor structure with two-layer crescent barriers and hybrid PM material to maximize the output torque and reduce the usage of high-cost rare-earth PM. The ferrite PMs are filled in the middle of the crescent barriers with thicker thickness, while the rare-earth PMs are filled into the bilateral of the crescent barriers with thinner thickness. To verify the effectiveness of the proposed rotor structure, optimizations are first carried out among the conventional rotor structures with single kind of PM material and the proposed rotor structure with hybrid PMs. Then, detailed comparisons have been investigated. Results indicate that the proposed machine has higher torque density than the machine with only ferrite PM and can reduce almost half the usage of rare-earth PM in comparison to the machine with only rare-earth PM. Abstract Percolation behavior of electrical conduction that is related to the formation of conductive networks has been well demonstrated in conductive polymer composites (CPC). However, the electromagnetic interference (EMI) shielding of CPC is normally very low at the percolation threshold due to the different mechanism. Here, we first predicted the percolation behavior of EMI shielding existing in CPC because the EMI shielding effectiveness (SE) was mainly dependent on the conductivity of CPC if assuming the constantly magnetic permeability. In practice, we also found the percolation behavior of EMI shielding in the multi-walled carbon nanotube (MWCNT) filled poly(l -lactide) (PLLA) with good dispersion and isotactic polypropylene (iPP) with poor dispersion. Furthermore, the percolation threshold of EMI shielding was higher than the percolation threshold of electrical conduction. The percolation thresholds of EMI shielding and electrical conduction were ∼2.00 and ∼0.40 vol% for PLLA/MWCNT nanocomposites, and ∼5.40 and ∼1.40 vol% for iPP/MWCNT nanocomposites, respectively. The SEM and TEM image shows the sparse and dense MWCNT network near the percolation threshold of electrical conduction and EMI shielding, respectively, indicating that the sparse network of MWCNT can create high electrical conductivity as long as the formation of conductive paths. Ocular parameters have been applied to ophthalmic lens designs in order to satisfy individual wearers. An axial length (AL) of them can be used in individual ophthalmic lens designs. Our aim was to propose a reliable formula that predicts an individual’s AL using the corneal radius and refractive error, and to demonstrate the applicability of this formula. A total of 348 subjects underwent keratometry, objective and subjective refraction, and AL measurement. The formula of calculated AL for prediction obtained from the original Gullstrand simplified schematic eye: calculated AL = (24.00 × aveK/7.80)—(SE × 0.40), where aveK and SE denote average corneal radius and spherical equivalent, respectively. Calculated AL was 24.50 ± 1.83 mm, which was 0.18 ± 0.47 mm longer than the measured value of 24.32 ± 1.73 mm (p < 0.001). The proportion showing the differences between the calculated and measured ALs were 284 eyes (40.8%) for 0.00–0.25 mm, 525 eyes (75.4%) for less than 0.50 mm, 665 eyes (95.5%) for less than 1.00 mm, and 31 eyes (4.5%) for more than 1.01 mm. Correlation coefficient showed a very high correlation between calculated and measured ALs (r = 0.967, p < 0.001), and higher in the myopic than in the hyperopic group. The mean difference was 0.18 mm; the 95% limit of agreement was +1.10—-0.75 mm in all groups. Agreement was better in hyperopic eyes than myopic eyes. Prediction from calculation of AL with a formula using the corneal radius and SE provides an alternative method to direct measurements of AL, especially in the restricted environment, which can’t use biometric equipment for personalized ophthalmic lens design. Long non-coding RNAs (lncRNAs) have been involved in occurrence and progression of multiple cancers. In the present study, we investigated the role of lncRNA Gm15290 in the proliferation and invasion of non-small cell lung cancer (NSCLC) cells. First, we found that lncRNA Gm15290 was markedly up-regulated in tumor tissues from NSCLC patients and NSCLC cell lines, compared with adjacent normal tissues and normal lung cell line HBE respectively. Then, different concentrations of pcDNA-Gm15290 expression vector and Gm15290 siRNA were respectively transfected into A549 NSCLC cells. Our results showed that overexpression of Gm15290 significantly increased the proliferation and invasion of A549 cells and suppressed cell apoptosis. Knockdown of Gm15290 suppressed A549 cell proliferation and invasion and promoted cell apoptosis. Subsequently, we explored the underlying mechanism through which Gm15290 promoted cell proliferation and invasion. The output of RNA hybrid bioinformatic tool revealed that Gm15290 potentially interacted with tumor suppressor miR-615-5p which displayed an opposite expression pattern in the cell lines and a strong negative correlation with the levels of Gm15290 in NSCLC patients (r2 = 0.9677, P<0.0001). The results of RNA pull-down assays confirmed that Gm15290 directly bound with miR-615-5p. Gm15290 negatively regulated the expression of miR-615-5p and increased the protein levels of miR-615-5p target genes, including IGF2, AKT2, and SHMT2. Background/Aims: Monoclonal antibodies (mAbs) are presently the most promising treatment against Ebola virus disease (EVD), and cocktail of two or more antibodies likely confers protection through complementary mechanisms. Zaire Ebolavirus (EBOV) glycoprotein (GP) and viral protein 40 (VP40) are targets for designing neutralizing antibodies. Currently, the antiviral therapeutics of mAb-cocktails are still limited solely to anti-GP antibodies，there is no Abs cocktail against Zaire EBOV GP and VP40, which both have important interactions with host cellular membrane. Methods: We used hybridoma technology to produce anti-Zaire EBOV GP mAb against GP receptor binding domain, and anti-Zaire EBOV VP40 mAbs against the N-terminal domain, the C-terminal domain, respectively; synthesized Zaire EBOV transcription and replication competent virus like particles (trVLPs), which model even all aspects of the EBOV life cycles in order to evaluate the anti-viral effect of mAbs. Then, we characterized the anti- Zaire EBOV trVLPs effect of anti-GP and VP40 mAbs in vitro by real time-PCR, immunofluorescence assay and western blot analysis. Results: Our results demonstrate that anti-GP or anti-VP40 mAbs effectively inhibit trVLPs replication. The cocktails of anti-GP and anti-VP40 mAbs, or between anti-VP40 mAbs, had synergistic anti-trVLPs effect. Meanwhile, the detailed DNA and amino acid sequences of the mAbs were checked. Conclusion: The study verifies neutralizing efficacy of anti-GP or anti-VP40 mAb, report promising cocktail of anti-GP and anti-VP40 mAb, or cocktail of two anti-VP40 mAbs. Background: Acyl-CoA dehydrogenase long chain (ACADL) was revealed to have a correlation with malignant progression of cancer. However, whether ACADL plays a role in clinical therapy remains unclear. The clinicopathological role of ACADL in esophageal squamous cell carcinoma (ESCC) will be discussed in this study. Materials and methods: The expression of ACADL was analyzed via real-time PCR and Western blotting to assess mRNA and protein levels in ESCC cell lines and normal esophageal epithelial cells (NEECs), in six paired ESCC tumors and relative normal tissues. Furthermore, immunohistochemical staining was performed on 135 paraffin-embedded ESCC specimens to assess ACADL expression. The clinicopathological significance of ACADL expression was further investigated via survival analysis and Cox regression analysis. Results: ACADL was found to be markedly upregulated in ESCC cell lines when compared with NEECs. Moreover, various experiments such as quantitative real-time PCR, Western blot, and immunohistochemical analyses all revealed that ACADL expression was increased in all six paired ESCC tumors and matched normal tissues. Furthermore, immunohistochemical analysis revealed an increased level of ACADL protein expression in all 135 paraffin-embedded samples from ESCC patients, which increased with disease progression. Conclusion: We demonstrated that ACADL is overexpressed in ESCC, both in cell lines and clinical specimens. ACADL is found to be a vital regulator in ESCC progression and can predict a worse outcome for ESCC patients, suggesting that ACADL might be a valuable molecule to be targeted for clinical therapy of ESCC treatment. Anisotropic conductive composites with segregated conductive networks were constructed in poly( l- lactide)/poly(ε-caprolactone)/multi-walled carbon nanotubes/nickel (PLLA/PCL/MWCNT/Ni) composites by aligning Ni particles using a low magnetic field. Firstly, MWCNTs were melt-mixed with PLLA to form PLLA/MWCNT composites and then pulverized into microscale PLLA/MWCNT particles 425–850 μm in size. Later, the Ni particles were dispersed in PCL to yield PCL/Ni composites and then mixed with PLLA/MWCNT particles at 100 °C, a temperature lying in between the melting temperatures of PCL and PLLA. The coated PLLA/MWCNT particles were compressed to form PLLA/PCL/MWCNTs/Ni composites with segregated structures. A remarkable conductive anisotropy was observed in the segregated samples after magnetic alignment in a low magnetic field of ∼47.5 mT at 100 °C for 30 min. The electrical conductivity of the segregated samples diametrically increased in the direction parallel to the magnetic field, but decreased in the direction perpendicular to the magnetic field after the magnetic alignment of Ni particles in the PCL phase. Electrical conductivity in the parallel direction was almost eight orders of magnitude higher than that in the perpendicular direction at 3.0 wt% Ni and 0.7 wt% MWCNTs. Conductive anisotropy in the segregated systems could also be easily regulated by controlling the treatment time or changing the direction of the magnetic field. However, electrical conductivity could be maintained in both vertical and parallel directions in conventional composites after magnetic treatment because Ni particles preferably dispersed in the continuous PLLA phase, in which the Ni particles cannot be aligned at 100 °C. The recombinant adeno-associated virus human thioredoxin-PR39 (rAAV/hTRX-PR39) has been demonstrated to have a protective effect on hypoxic cells. The present study aimed to explore the potential effect of rAAV/hTRX-PR39 on acute cerebral infarction in rats. Middle cerebral artery occlusion (MCAO) model rats were produced and divided into three groups: Normal saline group, empty virus group (rAAV, without hTRX-PR39 cDNA) and rAAV/hTRX-PR39 group. Hematoxylin and eosin staining and electron microscopy observation were used to assess the morphological changes of ischemic brain tissue during different periods. Immunohistochemistry was employed to detect the expression of CD34 to reflect angiogenesis of ischemic brain tissue. Rats treated with rAAV/hTRX-PR39 showed an alleviated degree of ischemic brain edema relative to that in control groups, suggesting PR39 can ameliorate brain damage after cerebral ischemia. In the rAAV/hTRX-PR39 group, CD34-positive cells were significantly increased in ischemic brain tissues compared to control groups. Furthermore, CD34-positive cells were primarily observed around the perivascular in ischemic brain, indicating the angiogenesis role of PR39 in ischemic brain. The present findings suggest that PR39 could effectively ameliorate ischemic brain damage and promote angiogenesis, which may contribute to the treatment of acute cerebral infarction.
