Beauveria brongniartii is a fungal pathogen that infects the beetle Melolontha melolontha, a significant agricultural pest in Europe. While research has primarily focused on the use of B. brongniartii for controlling M. melolontha, the genomic structure of the B. brongniartii population remains unknown. This includes whether its structure is influenced by its interaction with M. melolontha, the timing of beetle‐swarming flights, geographical factors, or reproductive mode. To address this, we analysed genome‐wide SNPs to infer the population genomics of Beauveria spp., which were isolated from infected M. melolontha adults in an Alpine region. Surprisingly, only one‐third of the isolates were identified as B. brongniartii, while two‐thirds were distributed among cryptic taxa within B. pseudobassiana, a fungal species not previously recognized as a pathogen of M. melolontha. Given the prevalence of B. pseudobassiana, we conducted analyses on both species. We found no spatial or temporal genomic patterns within either species and no correlation with the population structure of M. melolontha, suggesting that the dispersal of the fungi is independent of the beetle. Both species exhibited clonal population structures, with B. brongniartii fixed for one mating type and B. pseudobassiana displaying both mating types. This implies that factors other than mating compatibility limit sexual reproduction. The European cockchafer Melolontha melolontha is an agricultural pest in many European countries. Populations have a synchronized 3 or 4 years life cycle, leading to temporally isolated populations. Despite the economic importance and availability of comprehensive historical as well as current records on cockchafer occurrence, population genomic analyses of M. melolontha are missing. For example, the effects of geographic separation caused by the mountainous terrain of the Alps and of temporal isolation on the genomic structure of M. melolontha still remain unknown. To address this gap, we genotyped 475 M. melolontha adults collected during 3 years from 35 sites in a central Alpine region. Subsequent population structure analyses discriminated two main genetic clusters, i.e., the South Tyrol cluster including collections located southeast of the Alpine mountain range, and a northwestern alpine cluster with all the other collections, reflecting distinct evolutionary history and geographic barriers. The "passo di Resia" linking South and North Tyrol represented a regional contact zone of the two genetic clusters, highlighting genomic differentiation between the collections from the northern and southern regions. Although the collections from northwestern Italy were assigned to the northwestern alpine genetic cluster, they displayed evidence of admixture with the South Tyrolean genetic cluster, suggesting shared ancestry. A linear mixed model confirmed that both geographic distance and, to a lower extent, also temporal isolation had a significant effect on the genetic distance among M. melolontha populations. These effects may be attributed to limited dispersal capacity and reproductive isolation resulting from synchronized and non‐synchronized swarming flights, respectively. This study contributes to the understanding of the phylogeography of an organism that is recognized as an agricultural problem and provides significant information on the population genomics of insect species with prolonged temporally shifted and locally synchronized life cycles. The mass occurrence of the sugar beet weevil (Asproparthenis punctiventris, previously Bothynoderes punctiventris) has been endangering sugar beet cultivation in Austria for centuries. Exacerbated by climatic and political changes (warmer, drier spring and limited access to chemical pesticides), new approaches are needed to counter the problem. The aim of our work was to test whether the bioinsecticide Metarhizium brunneum Ma 43 (formerly M. anisopliae var. anisopliae BIPESCO 5/F52) can be used as a sustainable plant protection product against the sugar beet weevil. Our goal was to control the pest in all its development stages through multiple applications. Therefore, GranMetTM-P, a granular formulation of M. brunneum Ma 43, was applied in spring to establish the fungus in the soil, whereas GranMetTM-WP, a liquid formulation of the production strain, was used in early summer on trap ditches and leaves to target the adult weevils. Soil and plant samples as well as weevils were collected during the planting season from the trial sites to evaluate the development of the fungus and the mycosis of the treated weevils. In addition, data on hibernating weevils and their emigration from untreated field sites was collected. In all field sites, the Metarhizium spp. abundance increased above the background level (<1000 CFU g−1 soil dry weight) after application of the product. With an increasing number of treatments per plot, and thus an increased contact possibility between pest and the fungus, a rise in the mycosis rate was observed. In conclusion, the various Metarhizium application strategies, which are already available or in testing, must be implemented to ensure control in both old and new sugar beet fields. Inundative mass application of Metarhizium brunneum BIPESCO 5 (Hypocreales, Clavicipitaceae) is used for the biological control of Diabrotica v. virgifera (Coleoptera, Chrysomelidae). Long-term field trials were performed in three Austrian maize fields—with different cultivation techniques and infestation rates—in order to evaluate the efficacy of the treatment to control the pest larvae. In addition, the indigenous Metarhizium spp. population structure was assessed to compare the different field sites with BIPESCO 5 mass application. Annual application of the product Granmet-PTM (Metarhizium colonized barley kernels) significantly increased the density of Metarhizium spp. in the treated soil above the upper natural background level of 1000 colony forming units per gram dry weight soil. Although a decrease in the pest population over time was not achieved in heavily infested areas, less damage occurred in treated field sites in comparison to control sites. The Metarhizium population structure was significantly different between the treated field sites. Results showed that inundative mass application should be repeated regularly to achieve good persistence of the biological control agent, and indicated that despite intensive applications, indigenous populations of Metarhizium spp. can coexist in these habitats. To date, crop rotation remains the method of choice for pest reduction in Europe, however continuous and preventive application of M. brunneum may also present an alternative for the successful biological control of Diabrotica. The European (Melolontha melolontha L.) and Forest (M. hippocastani F.) cockchafer are widespread pests throughout Central Europe. Both species exhibit a 3–5-year life cycle and occur in temporally shifted populations, which have been monitored and documented for more than 100 years. Visual identification of adults and larvae belonging to these morphologically similar species requires expertise and, particularly in the case of larvae, is challenging and equivocal. The goal of the study was the development of an efficient and fast molecular genetic tool for the identification and discrimination of M. melolontha and M. hippocastani. We established a collection of both species from Switzerland, Austria and Northern Italy in 2016, 2017 and 2018. An approximately 1550 bp long fragment of the cytochrome c oxidase subunit 1 (CO1) mitochondrial gene was amplified and sequenced in 13 M. melolontha and 13 M. hippocastani beetles. Alignment of the new sequences with reference sequences (NCBI GenBank and BOLDSYSTEMS databases) and subsequent phylogenetic analysis revealed consistent clustering of the two species. After the identification of M. melolontha and M. hippocastani species-specific single nucleotide polymorphisms (SNPs) in the CO1 alignment, we developed an effective SNP tool based on the ABI PRISM® SNaPshot™ Multiplex Kit for the rapid and accurate species discrimination of adults and larvae. Entomopathogenic fungi are used for biological control of insect pests. Metarhizium brunneum Petch (Hypocreales) has potential to control Diabrotica virgifera virgifera LeConte (Chrysomelidae), which is a major pest of maize in North America and has recently invaded Europe. The inundative application of an entomopathogenic fungal strain in biological control results in high densities of fungal propagules in the soil which can potentially affect soil microbial communities and their multiple functions in soil. The objective of the present study was to assess potential effects of M. brunneum on soil fungal and prokaryotic communities in a pot experiment over a time course of 4 months using high-throughput sequencing (HTS) of ribosomal markers. The application of M. brunneum formulated as fungus colonised barley kernels (FCBK) led to a significant increase of the applied strain in soil, as assessed by cultivation-dependent (plating on selective medium followed by genotyping of Metarhizium isolates) and cultivation-independent (HTS of ribosomal markers) approaches. Data revealed that soil fungal and prokaryotic community structures did not change after the application of M. brunneum. Temporal changes of the fungal and prokaryotic communities were observed and the prokaryotic communities showed minor changes to barley kernels (BK), the matrix of the formulation. Results of this study are in accordance with other investigations lacking any evidence for adverse effects on microbial communities caused by applied entomopathogenic fungi. [ABSTRACT FROM AUTHOR]

Selected biological control agents and conventional pesticides were used to critically review the applicability of a newly developed Risk Indicator (RI) system. Five basic components are proposed for the calculation of the overall environmental risk score: persistence of the active ingredient, dispersal potential, range of non-target organisms that are affected, and direct and indirect effects on the ecosystem. Several risk measurement systems were reviewed; risk categories in the proposed system were modified from a model developed for classical biocontrol agents. Additionally, one new category was included, to assess the risks to vertebrate non-target species. Besides a detailed discussion of the new RI model, the suitability of the model was demonstrated by calculating the risk scores for 17 selected products. It became obvious that the environmental risk score varied greatly within the assessed chemical products, and also within the group of biological products. The use pattern greatly influenced the estimated environmental risk posed by any given product. The overall environmental risk score varied between a very low risk score of 24 (Coniothyrium minitans, soil application) and a near maximum risk score of 4275 (high risk reference DDT, foliar spray). The proposed model can be used to communicate environmental risk and to design lower risk integrated pest management strategies. It is suggested that the proposed RI system may serve to define low risk and reduced risk pesticides. Abstract: Amending plant growing media with biocontrol agents is becoming an increasingly popular strategy for protecting plants against insect pests and diseases. Although this practice offers potential for pest and disease control, very little is known about the influence of introduced microbes on the indigenous microbial communities and vice versa. This study examines fungal and bacterial community structures of diverse horticultural growing media and evaluates their interactions with a plant Euonymus fortunei and/or the insect pathogenic fungus Metarhizium anisopliae. Complementary physiological (Biolog™) and molecular (automated ribosomal intergenic space analysis – ARISA) techniques were applied to analyse community structure and diversity over 1yr. These studies clearly showed distinct microbial communities in different plant growing media. Irrespective of media or any treatment, initial microbial diversity was very low but increased significantly over 1yr. A succession of new fungal and bacterial species was observed, with species originally present being displaced. This succession led to the establishment of distinct microbial community structures for each medium over 6–12 months. ARISA studies showed that bacterial and fungal community profiles were influenced by the presence of plant and M. anisopliae, but in complex ways depending on the medium. Certain microbes were specifically associated or inhibited by the presence of M. anisopliae and/or E. fortunei. Physiological studies, however, showed that neither M. anisopliae nor plant had any significant effect on the communities'' ability to decomposition a batch of carbon sources. Although diverse microbial communities were observed in different media, none of these communities influenced the efficacy of M. anisopliae. [Copyright &y& Elsevier]

Entomopathogenic fungi are promising alternatives to chemical insecticides. However, a major hurdle concerning the registration of these fungi as plant protection agents is the possible toxicity of secreted metabolites, especially secondary metabolites. This review summarizes data on specific secondary metabolites (destruxins, efrapeptins, oosporein, beauvericin and beauveriolides) produced by the important genera Beauveria, Metarhizium and Tolypocladium. The quantities of secondary metabolites produced by these fungi in vivo are usually much less than those secreted in nutrient rich liquid media. The "risk-society hypothesis" consists of two related parts. In its first part, the hypothesis views modem societies as being in a transition from "class societies" to "risk societies." In its second part, it states that modem societies undergo a process of "individualization." Essentially, Ulrich Beck and others elaborate both parts of the hypothesis with respect to industrial societies in general and to German society in particular. Critics argue that the hypothesis misjudges the relation between societal risk distribution, conflict, and social inequality. It fails to understand the main reasons for risk-related conflicts in society when it supposes that the basic conflicts characterizing risk societies arc different from the basic conflicts in class societies. It fails mainly because it ignores the possibility of causal attributions and risk perceptions directly related to antagonistic (class) positions. As far as the postulated individualization process is concerned, the risk society hypothesis represents a peculiar mixture of supposedly right and wrong assumptions. The hypothesis seems quite right in assuming changing modes of social integration: Not so much traditional ties as market and competitive mechanisms determine social life, often advancing to its most private corners. The hypothesis fails, however, in its structural implications. In particular, the view that the postulated trends question the "hierarchy model of social inequality" is neither theoretically convincing nor empirically tenable. Instead, neither exposition to global risks nor individualization is likely to make society more egalitarian. Furthermore, instead of assuming that risk-societies overcome class conflicts, the paper envisions the emergence of a new risk-related cleavage in society. [ABSTRACT FROM AUTHOR]
