Cronobacter sakazakii is a common foodborne pathogen that can tolerate various stress conditions. Acidic environment is a common stress condition encountered by bacteria in food processing and gastrointestinal digestion, including both inorganic and organic acids. In order to elucidate the Acid Tolerance Response (ATR) of C. sakazakii, we performed high-throughput RNA-seq to compare gene expression under hydrochloric acid and citric acid stresses. In this study, 107 differentially expressed genes (DEGs) were identified in both acids, of which 85 DEGs were functionally related to the regulation of acid tolerance. Multiple layers of mechanisms may be applied by C. sakazakii in response to acid stress: Firstly, in order to reduce excessive intracellular protons, C. sakazakii pumps them out through trans-membrane proteins or consumes them through metabolic reactions. Secondly, under acidic conditions, a large amount of reactive oxygen species and hydroxyl radicals accumulate in the cells, resulting in oxidative damage. C. sakazakii protects cells by up-regulating the antioxidant stress genes such as soxS and madB. Thirdly, C. sakazakii chooses energy efficient metabolic pathways to reduce energy consumption and maintain necessary processes. Finally, genes involved in chemotaxis and motility were differentially expressed to respond to different acidic conditions. The generation of pseudo-lignin as byproduct during the lignocellulose acidic pretreatment has been proposed for many years. However, the detailed formation mechanism is still unclear. Moreover, there is a lack of understanding in the initial reaction during the formation of humins (byproducts in furfural production) and acid-induced hydrothermal carbon (carbon material). In this work, the initial formation of these three substances were investigated. We first found the common feature of their formation process was that carbohydrate-hydrolyzed compounds could form black polymers by condensing in acidic media, but the difference was dependent on the reaction degree. Furthermore, the results revealed that oxidation was an accelerator for condensations during producing black polymers because oxidized compounds could enhance the acidity of the reaction system. However, condensations of oxidized compounds were more difficult to proceed. Meanwhile, during the initial stage, the dominating pathway was that furfural condensed with itself and isomerized xylose via aldol-condensation.

A feasible approach to produce xylooligosaccharides (XOS) using organic acids as catalysts by microwave-induced hydrolysis of different hemicelluloses was developed. The effects of different acids (oxalic acid, maleic acid, citric acid and sulfuric acid), acid concentration, reaction temperature and reaction time on the hemicelluloses hydrolysis were investigated. Results demonstrated that organic acid was more beneficial to the XOS production than the conventional sulfuric acid. Higher acid concentration, higher reaction temperature and longer reaction time accelerated the further depolymerization of XOS to form monosaccharide. Response surface methodology was employed to optimize the reaction conditions (temperature and time) for the production of XOS from beechwood xylan (BX), corncob hemicelluloses (CH) and recovered hemicelluloses from the industrial waste liquor of dissolving pulp (RH), respectively. The predicted highest XOS yields were achieved to 39.31% (126.54°C-7.95min), 27.29% (120.00°C-0min), 30.32% (122.63°C-15.85min), respectively, being close to the experimental value (39.42%, 27.46% and 30.89%) from BX, CH and RH, indicating the fitted models of XOS yield were in good agreement with the experimental results., (Copyright © 2016 Elsevier Ltd. Enzymatic hydrolysis of corn stover was studied at agitation speeds from 50 to 500rpm in a stirred tank bioreactor, at high solid concentrations (20% w/w dry solid/suspension), 50°C and 15.5mgprotein·gglucane(-1). Two empirical kinetic models have been fitted to empirical data, namely: a potential model and a fractal one. For the former case, the global order dramatically decreases from 13 to 2 as agitation speed increases, suggesting an increment in the access of enzymes to cellulose in terms of chemisorption followed by hydrolysis. For its part, the fractal kinetic model fits better to data, showing its kinetic constant a constant augmentation with increasing agitation speed up to a constant value at 250rpm and above, when mass transfer limitations are overcome. In contrast, the fractal exponent decreases with rising agitation speed till circa 0.19, suggesting higher accessibility of enzymes to the substrate., (Copyright © 2016 Elsevier Ltd. Selectively catalytic hydrothermal pretreatment of corncob into xylose and furfural has been developed in this work using solid acid catalyst (SO4(2-)/TiO2-ZrO2/La(3+)). The effects of corncob-to-water ratio, reaction temperature and residence time on the performance of catalytic hydrothermal pretreatment were investigated. Results showed that the solid residues contained mainly lignin and cellulose, which was indicative of the efficient removal of hemicelluloses from corncob by hydrothermal method. The prepared catalyst with high thermal stability and strong acid sites originated from the acid functional groups was confirmed to contribute to the hydrolysis of polysaccharides into monosaccharides followed by dehydration into furfural. Highest furfural yield (6.18 g/100g) could be obtained at 180°C for 120 min with 6.80 g/100g xylose yield when the corncob/water ratio of was 10:100. Therefore, selectively catalytic hydrothermal pretreatment of lignocellulosic biomass into important platform chemicals by solid acids is considered to be a potential treatment for biodiesel and chemical production., (Copyright © 2014 Elsevier Ltd. A new sucralose-derived solid acid catalyst (SUCRA-SO3H), containing -Cl and -SO3H functional groups, has been shown to be highly effective for hydrolyzing β-1,4-glucans, completely hydrolyzing cellobiose (1) into glucose (2) in 3 h and converting the microcrystalline cellulose pretreated by the ionic liquid into glucose (2) with a yield of around 55% and a selectivity of 98% within 24 h at a relatively moderate temperature (393K). The enhanced adsorption capacity that the catalyst has for glucan by virtue of the presence of chloride groups that act as cellulose-binding sites offers the possibility of resolving the existing bottleneck in heterogeneous catalysis to hydrolyze cellulose, namely, the low accessibility of cellulose to the reaction position in typical solid catalysts. The apparent activation energy for hydrolysis of cellobiose (1) with SUCRA-SO3H was 94 kJ/mol, which was much lower than that with sulfuric acid (133 kJ/mol) and the corresponding sucrose-derived catalyst (SUCRO-SO3H) without chlorine groups (114 kJ/mol).

Dilute sulfuric acid was used to pretreat eucalyptus chips prior to enzymatic hydrolysis. After both pretreatment and enzymatic hydrolysis processes. Attention is paid to sugar recovery. The maximum total sugars yield (combined xylose and glucose, 47.69g/100g raw material, representing 82% of total sugars in the eucalyptus biomass) was obtained at 160°C, 0.75% acid concentration and 10min residence time, which is consider to be the best reasonable conditions for the dilute acid pretreatment of eucalyptus, corresponding concentrations of acetic acid, HMF, and furfural in the prehydrolysate were about 2.01g/L, 0.13g/L and 1.37g/L, respectively. The presence of lignin impairs the utilization of the hemicelluloses dissolved in the pre-hydrolysis liquor (PHL) of the kraft-based dissolving pulp production process. In this paper, a novel process was developed by combining the acidification and poly ethylene oxide (PEO) flocculation concepts to improve the lignin removal. The results showed that the lignin removal was improved by the addition of PEO to the acidified PHL, particularly at a low pH of 1.5. The main mechanisms involved are the lignin/PEO complex formation and the bridging of the formed complexes. This hypothesis was supported by the turbidity, FTIR and particle size measurements. Interestingly, the hemicelluloses removal from the acidification/PEO flocculation was marginal, which would be beneficial for the down-stream ethanol production from the PHL. Rice hulls (RH) and bean hulls (BH) were subjected to prehydrolysis treatments, to define the optimum conditions for producing a high percentage of hydrolyzed hemicellulose with a small or moderate degradation of the cellulosic portion. The hydrolysis experiments were performed using hydrochloric and sulfuric acids in concentrations ranging from (0.5 to 5)% (w/w) at 120 degrees C for 90 min and 10% consistency. The effects of different temperatures (80 to 120 degrees C) and time (30 to 120 min) on acid hydrolysis of lignocellulosic materials were recorded. It was found that, the optimum condition to hydrolyze the lignocellulosic materials (RH) and (BH) are 2% (w/w) of mineral acid at 120 degrees C for 90 min and 10% consistency. The cellulose crystallinity index in the different types of lignocellulosic materials with and without acid treatment, were increased from 0.32 to 0.46 in case of RH and from 0.43 to 0.61 in case of BH. Due to the lignin depolymerization during the pretreatment process, the relative absorbency of the methoxyl group and the aromatic rings bands were lowered for the pretreated than the untreated lignocellulosic materials. The content and distribution of stilbenes and resin acids in Scots pine (Pinus sylvestris) and spruce (Picea abies), sampled in central Norway, have been examined. The contents of pinosylvin stilbenes in pine heartwood/living knots were 0.2-2/2-8 %(w/w). No stilbenes could be detected in spruce (Picea abies). The resin acid contents of pine sapwood/heartwood and knots were 1-4 and 5-10 % (w/w), respectively. Minor amounts of resin acids (<0.2/<0.04 %w/w) were identified in spruce wood/knots. The lignan content in knots of Norwegian spruce was 6.5 % (w/w). Diastereomerically pure hydroxymatairesinol (HMR, 84 % of total lignans) was readily isolated from this source since only minor quantities (2.6 % of total lignans) of the allo-HMR diastereomer was detected. Insignificant amounts of lignans were present in the sapwood. Lignans could not be detected in the sapwood or knots of Norwegian sallow (Salix caprea), birch(Betula pendula) or juniper (Juniperus communis).
