Objective: This study proposed to assess the effect of Cryptocarya moschata extract on single and mixed biofilms formed on denture base and reline acrylic resin., Materials and Methods: Single and mixed biofilms of Candida albicans and Streptococcus mutans were formed on the samples and treated with C. moschata extract; Nystatin solution at 100,000 IU/mL or Penicillin antibiotic solution at 100,000 IU/mL; or PBS solution. Antimicrobial activity was analyzed by counting colony-forming units, metabolism assay, assessment of protein components of the biofilm matrix, and of cell viability using confocal laser scanning microscopy (CLSM). Data were submitted to ANOVA and Tukey's post-test (α = 0.05)., Results: Cryptocarya moschata extract reduced cell viability of C. albicans and S. mutans single and mixed biofilms formed on samples. For all types of biofilms in the C. moschata group, there was a log reduction of the biofilm, proven by the Alamar Blue assay. Analyzing the extracellular matrix protein components, groups treated with the extract exhibited a lower level of fluorescence compared to the PBS groups. To evaluate the effect of antiseptic soap on single and dual-species biofilms of Candida albicans and Streptococcus mutans on denture base and reline resins. Samples of the resins were distributed into groups (n = 9) according to the prevention or disinfection protocols. In the prevention protocol, samples were immersed in the solutions (Lifebuoy, 0.5% sodium hypochlorite solution and PBS) for 7, 14 and 28 days before the single and dual-species biofilms formation. Overnight denture disinfection was simulated. In the disinfection protocol, samples were immersed in the same solutions during 8 hours after the single and dual-species biofilms formation. Antimicrobial activity was analyzed by counting colony-forming units (CFU/mL) and evaluating cell metabolism. Cell viability and protein components of the biofilm matrix were evaluated using confocal laser scanning microscopy (CLSM). Data were submitted to ANOVA, followed by Tukey's post-test (α = 0.05) or Dunnett's T3 multiple comparisons test. In the prevention protocol, Lifebuoy solution effectively reduced the number of CFU/mL of both species. In addition, the solution decreased the cell metabolism of the microorganisms. Regarding disinfection protocol, the Lifebuoy solution was able of reduce approximately of 2-3 logs for all the biofilms on the denture base and reline resin. Cellular metabolism was also reduced. The images obtained with CLSM corroborate these results. Purpose: To evaluate the influence of denture cleansers on the surface roughness, Candida albicans adhesion, and biofilm formation on denture base acrylic resins., Study Selection: Electronic databases and gray literature were searched using an individual search strategy. In vitro studies that evaluated the effects of immersion in denture cleansers on the surface roughness (µm) and antimicrobial activity (CFU/mL) on samples of heat-polymerized denture base acrylic resins were included., Results: After screening, 17 studies were included, and a qualitative synthesis was performed. After assessing the risk of bias, only nine studies were included in the meta-analysis. The meta-analysis results showed that the evaluated solutions (0.5% sodium hypochlorite, 1% sodium hypochlorite, alkaline peroxide, and natural substances) did not influence the roughness of the acrylic resin. However, in the qualitative analysis, it was not possible to confirm an association between roughness and C. albicans adhesion and biofilm formation on the acrylic resin samples., Conclusion: Denture cleansers did not affect the surface roughness of denture base acrylic resins.

Purpose: To evaluate some biological (Candida albicans biofilm formation and cytotoxicity) and mechanical (roughness and hardness) properties of a hard reline acrylic resin after immersion in liquid disinfectant soaps., Methods: Reline samples were made (14 mm in diameter and 1.2 mm in thickness) and immersed in different disinfectant solutions: DW: distilled water at 37°C (control group); DS: Dettol liquid soap; and LS: Lifebuoy liquid soap. After immersion in the different groups, the properties of C. albicans biofilm formation, cytotoxicity, hardness, and roughness were evaluated. Each sample was immersed in the disinfectant solutions for 8 hours for microbiological testing and for 0, 7, 14, 21, and 28 days, with daily immersion cycles, for cytotoxicity and mechanical testing. Data were tabulated and analyzed by Shapiro-Wilk and Levene statistical tests. Based on these results, the appropriate tests for finding statistically significant differences were applied (α= 0.05)., Results: There was a reduction of roughness after immersion of the samples in DS for 21 days (P= 0.0001) and LS for 28 days (P= 0.0218). Hardness values significantly increased after storage of samples in solutions (0.0007≥ P≥ 0.0076), especially considering the longer period of immersion (28 days). The viability and metabolic activity of the biofilm formed on the hard reline samples were significantly reduced (P< 0.05) after soaking in Lifebuoy liquid soap in relation to DW. In addition, the samples immersed in the different solutions were not cytotoxic, as they inhibited less than 25% in relation to the control group (100% of cell viability)., Clinical Significance: This study showed that, in general, immersion in liquid disinfectant soaps had no adverse effect on the physical properties of the hard reline acrylic resin evaluated. In addition, Lifebuoy liquid soap reduced the biofilm formed on the samples and the solutions did not affect the cytotoxicity of the material. Denture stomatitis triggered by Candida species requires better preventive measures. This study evaluated the physical and biological properties of a denture base acrylic resin after immersion in antiseptic soaps. Acrylic resin specimens were prepared and stored in distinct solutions for 0, 7, 14, 21, and 28 days. The solutions were as follows: DW: distilled water at 37°C (control group); DS: cycles of daily immersion in Dettol soap for 8 hours at room temperature, followed by immersion in distilled water for 16 hours at 37°C; PS: cycles of daily immersion in Protex soap, as described for the previous group; LS: cycles of daily immersion in Lifebuoy soap, as described for the DS group. The parameters evaluated at each time point were the following: biofilm formation capacity by Candida albicans and reduction of preformed fungal biofilms, cytotoxicity, surface roughness, hardness, and color change. For the fungal adhesion phase, the type of soap had a statistically significant effect (p = 0.0292), but after 24 hours, no differences were found between solutions or between storage times. Regarding the efficacy of biofilm reduction, there was a significant difference when the groups were compared to each other (p = 0.014). Dettol and Lifebuoy eliminated the preformed biofilm on the specimens. Moreover, all the soaps were classified as non-cytotoxic (on HaCaT cell line) because there was no difference in cell viability between the different groups, except after 21 days, when a decrease in cell viability occurred, regardless of the type of soap. Regarding the roughness, there was no statistically significant difference (p > 0.05) between the groups. Lifebuoy decreased resin hardness regardless of storage time (p = 0.003). After 21 and 28 days of storage, there was an increase in hardness value, regardless of the type of soap. The specimens' color, according to the National Bureau of Standards values, ranged from 0.27 to 0.58 (i.e., imperceptible or mild color changes). In general, the disinfectant soaps were not able to prevent biofilm formation, but all the soaps were effective in reducing the preformed biofilm. In addition, all soaps were non-cytotoxic and did not change surface roughness, hardness (except Lifebuoy), and color (except Lifebuoy). Therefore, immersion in two antiseptic soaps (Protex and Dettol) may be a cheap and easy procedure for preventing denture stomatitis., Competing Interests: The authors have declared that no competing interests exist.

Statement of Problem: The daily immersion of dentures in disinfectant solutions can cause the incorporation of toxic substances in the acrylic resins, and studies evaluating the cumulative effect of disinfectant solutions on cell culture are lacking., Purpose: The purpose of this in vitro study was to evaluate the cytotoxic potential of cell cultures of denture base and reline acrylic resins after immersion in disinfectant solutions., Material and Methods: Disk-shaped specimens (14×1.2 mm) were obtained and divided into groups (n=9) according to the disinfectant solutions (distilled water [control], 2% chlorhexidine digluconate, 3.8% sodium perborate, 0.5% sodium hypochlorite, and apple vinegar) and to the storage period (0, 1, 3, and 6 months). Solutions were changed daily. After the different storage periods, specimens were immersed in culture medium for 24 hours, and extracts were obtained. Human keratinocytes were cultivated, and the cellular metabolism was evaluated by using Alamar Blue. Data were submitted to 3-way analysis of variance and Games-Howell post hoc tests (α=.05)., Results: Both of the acrylic resins tested showed similar biocompatibility properties after immersion in different solutions (P=.400). Immersion in distilled water, 3.8% sodium perborate, and 0.5% sodium hypochlorite did not affect the cellular metabolism of the keratinocytes (P>.05), regardless of the immersion period and the type of acrylic resin (P>.05). Immersion in 2% chlorhexidine digluconate or apple vinegar resulted in high cytotoxicity over time, until the third month (P<.05), after which no changes were observed (P>.05)., Conclusions: The type of acrylic resin (base or reline) had no significant effect on the viability of cells. Vinegar and chlorhexidine digluconate solutions increased in cytotoxic effect over time, and were strongly cytotoxic after 6 months of immersion. Purpose: To investigate the efficacy of immersion and brushing with different cleansing agents in reducing the viability of multispecies biofilm on acrylic resins., Methods: Lucitone 550 (L) and Tokuyama Rebase Fast II (T) specimens (10 x 2 mm) were prepared, sterilized, and inoculated with a suspension of Candida albicans, Candida glabrata, and Streptococcus mutans. Specimens were incubated for 48 hours at 37 degrees C for biofilm formation. Then, they were divided into groups (n = 12) and subjected to brushing or immersion for 10 seconds in distilled water (W), 0.2% peracetic acid-Sterilife (Ac), 1% chlorhexidine digluconate (CHX), 1:1 water/dentifrice solution (D), 1% sodiumhypochlorite (NaOCl), and sodium perborate/Corega Tabs (Pb). Viable microorganisms were evaluated by the XTT assay and colony counts (cfu/mL). Data were performed by ANOVA and Tukey test with 5% significance level., Results: The multispecies biofilm on L and T were killed by brushing or immersion in Ac, CHX, and NaOCl for only 10 seconds.

Purpose: To investigate the cumulative effects of brushing (B) or immersion (I), using different cleansing agents, on the surface roughness, hardness and color stability of a heat-polymerized denture resin, Lucitone 550 (L), and a hard chairside reline resin, Tokuyama Rebase Fast II (T)., Methods: A total of 316 specimens (10 x 2 mm) were fabricated. The specimens (n = 9) were divided into brushing or immersion groups according to the following agents: dentifrice/distilled water (D), 1% sodium hypochlorite (NaOCl), Corega Tabs (Pb), 1% chlorhexidine gluconate (Chx), and 0.2% peracetic acid (Ac). Brushing and immersion were tested independently. Assays were performed after 1, 3, 21, 45 and 90 brushing cycles or immersion of 10 seconds each. Data were evaluated statistically by repeated measures ANOVA. Tukey's honestly significant difference (HSD) post-hoc test was used to determine differences between means (α = 0.05)., Results: For L there was no statistically significant difference in roughness, except a significant decrease in roughness by brushing with D. T showed a significant effect on the roughness after 90 immersions with Ac. Hardness values decreased for L when specimens were immersed or brushed in NaOCl and Pb. The hardness of T decreased with increases in the repetitions (immersion or brushing), regardless of the cleaning method. Values of color stability for L resin showed significant color change after brushing with and immersion in Ac and Pb. Brushing with D exhibited a higher incidence of color change. For T there were no significant differences between cleaning agents and repetitions in immersion. A color change was noted after three brushings with the Ac, Chx, and D. Brushing with dentifrice decreased roughness of L. Immersion in or brushing with NaOCl and Pb decreased the hardness of L. For T, hardness decreased with increases in immersions or brushing. Color changes after the immersion in or brushing with cleaning agents were clinically acceptable according to National Bureau of Standards parameters for both resins.

The colour stability of acrylic resin denture teeth in beverages was investigated. A spectrophotometer measured the colour (CIE-L*a*b* system) of all specimens after storage in distilled water/for 24 h at 37 degrees C (T0). Specimens were then immersed in various beverages. After 15 days (T1) and 30 days (T2), for each material, the mean deltaE values were calculated and compared by two-way ANOVA and Tukey intervals (alpha = 0.05). In the deltaT0T1 period, specimens stored in red wine were significantly discoloured, compared to distilled water (P = 0.003). There was no difference between immersion solutions in deltaET0T2 (P = 0.772) and in deltaET1T2 (P = 0.058), and no difference between materials in all immersion periods.

Purpose: The aim of this in vivo animal study was to investigate changes in the surface roughness of soft liners over time., Materials and Methods: Forty adult Wistar rats (Rattus norvergicus albinus) were fitted with acrylic custom-made palatal plates relined by dynamic impressions and tested with the following soft liners: Dentuflex (DF), Trusoft (TS), Dentusoft (DS), and Ufi Gel P (UG). Half of the animals for each tested material had the plates fitted during the material reline procedure. Their surface roughness was read immediately (IRa group, n = 5). The other half used the palatal plates for 14 days before roughness readings were performed (FRa group, n = 5). The surface roughness (Ra) of the inner surface from the relined dentures was recorded using a Surftest SJ-401 with eight readings per specimen, and mean values were obtained. Data (μm) were analyzed by two-way ANOVA and Tukey's test (α = 0.05)., Results: IRa means (2.92 ± 0.87 μm) and FRa means (3.35 ± 0.65 μm) were significantly different (p = 0.016). UG showed a lower (p = 0.01) Ra mean (2.1 ± 0.52 μm) than DF (3.94 ± 0.81 μm), TS (4.12 ± 0.64 μm), and DS (3.27 ± 0.64 μm)., Conclusions: Ufi Gel P showed the smoothest surface among the materials evaluated. This study evaluated the effect of experimental coatings, containing zwitterion or hydrophilic monomers, on the adherence of Candida albicans, Candida glabrata, and Streptococcus mutans to an acrylic resin. Acrylic samples (smooth or rough surfaces) were left untreated (control) or coated with one of the following experimental coatings: 3-hydroxypropylmethacrylate (HP) or sulfobetaine methacrylate (S), at concentrations of 25, 30, or 35%. Half of the specimens were coated with saliva. The adhesion test was performed by incubating specimens in C. albicans, C. glabrata, and S. mutans suspensions at 37°C for 90 min. The number of adhered microorganisms was determined by metabolic activity (XTT) and by cell viability (CFU). All coated specimens exhibited lower absorbance and CFU values compared to control specimens. Saliva and roughness did not promote microorganism adherence. An XPS analysis confirmed the modification in the chemical composition of the coatings in the experimental samples. These experimental coatings significantly reduced the adherence of C. albicans, C. glabrata and S. mutans to acrylic resin.

Unlabelled: In order to prolong the clinical longevity of resilient denture relining materials and reduce plaque accumulation, incorporation of antimicrobial agents into these materials has been proposed. However, this addition may affect their properties., Objective: This study evaluated the effect of the addition of antimicrobial agents into one soft liner (Soft Confort, Dencril) on its peel bond strength to one denture base (QC 20, Dentsply)., Material and Methods: Acrylic specimens (n=9) were made (75x10x3 mm) and stored in distilled water at 37 ºC for 48 h. The drug powder concentrations (nystatin 500,000 U--G2; nystatin 1,000,000 U--G3; miconazole 125 mg--G4; miconazole 250 mg--G5; ketoconazole 100 mg - G6; ketoconazole 200 mg--G7; chlorhexidine diacetate 5%--G8; and 10% chlorhexidine diacetate--G9) were blended with the soft liner powder before the addition of the soft liner liquid. A group (G1) without any drug incorporation was used as control. Specimens (n=9) (75x10x6 mm) were plasticized according to the manufacturers' instructions and stored in distilled water at 37 ºC for 24 h. Relined specimens were then submitted to a 180-degree peel test at a crosshead speed of 10 mm/min. Data (MPa) were analyzed by analysis of variance (α=0.05) and the failure modes were visually classified., Results: No significant difference was found among experimental groups (p=0.148). Cohesive failure located within the resilient material was predominantly observed in all tested groups., Conclusions: Peel bond strength between the denture base and the modified soft liner was not affected by the addition of antimicrobial agents.

Objective: To evaluate the weight loss and the surface roughness of acrylic resins after simulated brushing tests., Material and Methods: Ten specimens of each material (Tokuyama Rebase II-TR, New Truliner-NT, Ufi Gel Hard-UH and Lucitone 550-L) were made. The wear loss (mg) by weight and the surface roughness (Ra μm) of each specimen was determined before and after brushing. The specimens were placed on the brushing machine and a total of 20,000 brushing cycles was performed. The results of weight loss and roughness values were submitted to the anova followed by the Tukey's test (p = 0.05)., Results: The mean weight loss of material L was statistically higher (p < 0.001) than that of the relines TR, UH and NT. No significant differences were found among the roughness values of resins TR, UH and L (p > 0.05). Only for L, toothbrushing increased the surface roughness. After toothbrushing, there was no significant difference between the roughness values of materials L and NT. The highest mean weight loss during the simulated toothbrushing tests was observed for L. Objective: The aim of the study was to evaluate the effect of long-term disinfection procedures on the Vickers hardness (VHN) of acrylic resin denture teeth., Material and Methods: Five acrylic resin denture teeth (Vipi Dent Plus-V, Trilux-T, Biolux-B, Postaris-P and Artiplus-A) and one composite resin denture teeth (SR-Orthosit-O) were embedded in heat-polymerised acrylic resin within polyvinylchloride tubes. Specimens were stored in distilled water at 37°C for 48 h. Measurements of hardness were taken after the following disinfection procedures: immersion for 7 days in 4% chlorhexidine gluconate or in 1% sodium hypochlorite (CIm and HIm group, respectively) and seven daily cycles of microwave sterilisation at 650 W for 6 min (MwS group). In the WIm group, specimens were maintained in water during the time used to perform the disinfection procedures (7 days). Data were analysed with anova followed by the Bonferroni procedure (α = 0.01)., Results: Microwave disinfection decreased the hardness of all acrylic resin denture teeth (p < 0.001). Immersion for 7 days in 4% chlorhexidine gluconate or distilled water had significant effect on the hardness of the acrylic resin denture teeth A (p < 0.01), and 1% sodium hypochlorite on teeth T (p < 0.01). All disinfection procedures decrease the hardness of the composite resin denture teeth (p < 0.01). Purpose: Staining of prosthodontic materials may result in patient dissatisfaction and additional expense for replacement. This study aimed to determine the color stability of two heat-cured denture base acrylic (Lucitone 550, Vipi Cril) and one nylon denture base resin (Transflex) after immersion in beverages., Materials and Methods: Forty disks of each resin (20.0-mm diameter, 3.0-mm thick) were prepared and stored in distilled water for 24 hours at 37°C. During that time (T(0) ), the color of all specimens was spectrophotometrically measured. Each specimen was immersed in coffee, cola, red wine, and distilled water as a means of control. After 15-day (T(1) ) and 30-day (T(2) ) periods of immersion, the color of the specimens was measured again. The CIE (Commission Internationale de L' Eclairage) L*a*b* system was used to determine mean ΔE (color changes) values for each material and compared statistically with two-way ANOVA and Bonferroni intervals at 0.95., Results: In ΔET(0) T(1) and ΔET(0) T(2) the most severe staining was apparent with red wine (p < 0.001), followed by coffee (p < 0.01), when compared to the specimens stored in distilled water. Transflex also showed significant color change after immersion in cola (p < 0.01). In ΔET(1) T(2) only red wine promoted significant staining of all resins (p < 0.0001)., Conclusion: Chromatic changes were exhibited by specimens immersed in red wine, followed by coffee. For Transflex, cola also promoted color changes. Objective: The effect of disinfection by immersion in sodium perborate solution and microwave irradiation on surface roughness of one denture base resin (Lucitone 550 -L), 3 hard chairside reline resins (Tokuyama Rebase II-TR, New Truliner-NT, Ufi Gel hard-UH) and 3 resilient reline materials (Trusoft-T; Sofreliner-S, Dentusil-D) was evaluated., Material and Methods: Thirty specimens of each material were made and divided into 3 groups: Control - not disinfected; P - daily disinfection by immersing in sodium perborate solution (3.8%); MW - microwave disinfection (6 min/650 W). Roughness measurements were made after polymerization (baseline) and after 1, 3 and 28 days. Roughness differences relative to the baseline readings were analyzed by Student's t-test (P=0.05)., Results: At baseline, Trusoft showed the highest (P<0.001) mean surface roughness (3.54 µm), and its surface roughness was significantly reduced after 28 days of disinfection by immersion in sodium perborate (P=0.013). Roughness measurements of material Trusoft were not performed after microwave disinfection due to the severe alterations on the surface. In the 3 groups evaluated, changes in roughness were significant for materials Ufi Gel hard (from 0.11 to 0.26 µm; P<0.041) and New Truliner (0.19 to 0.76 µm; P<0.019). The roughness of materials Lucitone 550 (0.37 µm), Tokuyama Rebase II (0.37 µm), Sofreliner (0.49 µm) and Dentusil (0.38 µm) remained unaffected (P>0.05)., Conclusions: The roughness of the hard reline materials Ufi Gel hard and New Truliner was adversely affected by microwave disinfection, immersion in water or in sodium perborate. Microwave disinfection caused severe alterations on the surface of the resilient liner Trusoft.

Objectives: Microleakage is a pre-stage of debonding between hard chairside relines and denture base acrylic resins. Therefore, it is important to assess them with regard to the longevity of the relined denture. This study investigated the effect of thermal cycling on the microleakage at the interface of three hard chairside reline resins and three denture base resins., Material and Methods: Rectangular bars (12 mm × 3 mm × 3 mm) of Lucitone 550, Acron MC and QC 20 were made and relined with Kooliner, Tokuyama Rebase Fast II and Ufi Gel Hard, Lucitone 550, Acron MC and QC 20 resins. Specimens were divided into one control and two test groups (n=10). In specimens of the control group, the microleakage was performed after the reline procedure. In Test Group 1, the specimens were stored for 24 h in distilled water at room temperature and in Test Group 2; the specimens were thermal cycled from 5 to 55°C for 5000 cycles with a 30-s dwell time. Subsequently, all specimens were immersed in 50% silver nitrate solutions for 24h. All specimens were sectioned longitudinally into three fractions and the lateral sections were examined (n=20). Silver nitrate stain penetration was examined under a stereoscopic lens with ×30 magnification, and the images were captured. Leica Qwin image analysis software was used to determine microleakage at the interface of the materials. Data were analysed using the Kruskal-Wallis test at a 95% level of significance., Results: For all cycles, there were no statistically significant differences between thermal cycled and non-thermal cycled groups (p>0.05)., Conclusion: It can be concluded that thermal cycling had no effect on the microleakage., (© 2010 The Gerodontology Society and John Wiley & Sons A/S.)

Objectives: To evaluate the effect of microwave postpolymerization heat treatment and water storage time on the cytotoxicity of denture base and reline acrylic resins., Method and Materials: Sample disks of 6 acrylic resins were fabricated under aseptic conditions (Kooliner, GC; Tokuyama Rebase II, Tokuyama Dental; New Truliner, Bosworth; Acron MC, GC; Lucitone 550, Dentsply; QC 20, Dentsply). The denture base acrylic resin samples were stored in water for 24 and 48 hours at 37 degrees C. The fabricated samples were further subdivided into 2 groups: (1) samples without heat treatment and (2) samples treated in a microwave. The 3H-thymidine incorporation test was used to determine the cytotoxicity of the materials. The data were statistically analyzed using the incomplete 3-way ANOVA and Tukey HSD tests (P < .05)., Results: Microwave postpolymerization heat treatment improved the biocompatibility of Tokuyama Rebase II. Tokuyama Rebase II without heat treatment and Acron MC in both experimental groups were graded as slightly cytotoxic in the group without water storage. The other resins were graded as noncytotoxic. After 24 hours of immersion in water, all materials were graded as noncytotoxic. After water storage for 48 hours, Acron MC without heat treatment and QC 20 in both experimental groups were graded as slightly cytotoxic. Lucitone 550 was graded as noncytotoxic in all experimental groups., Conclusion: Microwave irradiation may be considered an alternative to reduce the cytotoxicity of Tokuyama Rebase II. Dental practitioners should choose Lucitone 550 processed with terminal boiling stage.

Background: Microwave energy has proved to be an effective method for disinfecting acrylic dentures. However, the effect of microwave heating on the porosity of autopolymerising denture reline resins has not been investigated., Objective: The purpose of the study was to determine the effect of microwave disinfection on the porosity of autopolymerised denture reline materials (Kooliner-K, New Truliner-NT, Tokuso Rebase Fast-TR and Ufi Gel Hard-UGH) and a conventional heat-polymerised denture base resin (Lucitone 550-L)., Material and Methods: Specimens (10 mm x 20 mm x 1 mm) were obtained from the impression surface of the palatal mucosa in a single person and divided into four groups (n = 5). The porosity was evaluated after polymerisation (C1), after two cycles of microwave disinfection (MW2), after seven cycles of microwave disinfection (MW7) and after 7 days storage in water at 37 degrees C (C2). Specimens from group MW7 were exposed to microwave disinfection daily being stored in water at 37 degrees C between exposures. All the replicas were sputter coated with gold and micrographs/digital images were taken of each replica using scanning electron microscopy at magnification x 100. The SEM micrographs were then examined using an image analyser to determine the number of pores. Comparison between materials and groups were made using Kruskal-Wallis tests., Results: MW7 resulted in a significant increase in the number from the pores of material K, but decreased in number in reline material TR and UGH reline resin. The number of pores in materials NT and L remained unaffected following microwave disinfection., Conclusion: Differences in the porosity amongst the materials and for different experimental conditions were observed following microwave disinfection.

Objective: The purpose of this study was to evaluate the effect of two post-polymerisation treatments and different cycles of polymerisation on the cytotoxicity of two denture base resins., Materials and Methods: The resins tested were Lucitone 550 and QC 20. Discs of resins were fabricated following the manufacturer's instructions. Lucitone 550 was processed by long cycle or short cycle. The resin QC 20 was processed by reverse cycle or normal cycle. The specimens were divided into groups: (i) post-polymerised in microwave for 3 min at 500 W; (ii) post-polymerised in water-bath at 55 degrees C for 60 min and (iii) without post-polymerisation. Eluates were prepared by placing three discs into a sterile glass vial with 9 ml of Eagle's medium and incubated at 37 degrees C for 24 hours. L929 cells were seeded into 96 well culture plates and DNA synthesis was assessed by (3)H-thymidine incorporation assay., Results: The results were submitted to two-way anova and Tukey HSD test. QC 20 specimens polymerised by the normal cycle and submitted to microwave post-polymerisation were graded as moderately cytotoxic. Similar results were observed for Lucitone 550 processed by long cycle without post-polymerisation. The other experimental groups were graded as not cytotoxic. After water-bath post-polymerisation, specimens of Lucitone 550 processed by long cycle produced significantly lower inhibition of DNA synthesis than the other groups., Conclusion: The long cycle increased the cytotoxicity of Lucitone 550 and water-bath post-polymerisation reduced the cytotoxicity of Lucitone 550 processed by long cycle.

Purpose: This study compared the effect of two postpolymerization heat treatments on the cytotoxicity of three denture base resins on L929 cells using 3H-thymidine incorporation and MTT assays., Materials and Methods: Sample disks of Lucitone 550, QC 20, and Acron MC resins were fabricated under aseptic conditions and stored in distilled water at 37 degrees C for 48 hours. Specimens were then divided into three groups: (1) heat treated in microwave oven for 3 minutes at 500 W; (2) heat treated in water bath at 55 degrees C for 60 minutes; and (3) no heat treatment. Eluates were prepared by placing three disks into a sterile glass vial with 9 mL of Eagle's medium and incubating at 37 degrees C for 24 hours. The cytotoxic effect from the eluates was evaluated using the 3H-thymidine incorporation and MTT assays, which reflect DNA synthesis levels and cell metabolism, respectively., Results: The components leached from the resins were cytotoxic to L929 cells when 3H-thymidine incorporation assay was employed. In contrast, eluates from all resins revealed noncytotoxic effects as measured by MTT assay. For both MTT assay and 3H-thymidine incorporation, the heat treatments did not decrease the cytotoxicity of the materials tested., Conclusion: Resins were graded by 3H-thymidine incorporation assay as slightly cytotoxic and by MTT assay as noncytotoxic. Cytotoxicity of the denture base materials was not influenced by microwave or water bath heat treatment.

Acrylic resins are widely used in the fabrication of denture bases and have been shown to be cytotoxic as a result of substances that leach from the resin. The primary eluate is residual monomer. Numerous reports suggest that residual monomer may be responsible for mucosal irritation and sensitization of tissues. This information is important, not only to assess the biologic effects of such materials, but also to enable a comparison among the different polymerization methods, thus assisting the clinician in selecting a material with minimal cytotoxicity. This article reviews the literature published from 1973 to 2000, selected by use of a Medline search, associated with cytotoxic effects usually ascribed to acrylic denture base materials.
