Brucellosis is a serious zoonosis caused by Brucella spp. infection, which not only seriously jeopardizes the health of humans and mammals, but also causes huge economic losses to the livestock industry. Brucella is a Gram-negative intracellular bacterium that relies primarily on its virulence factors and a variety of evolved survival strategies to replicate and proliferate within cells. Currently, the mechanisms of autophagy, inflammation, and apoptosis in Brucella -infected hosts are not fully understood and require further research and discussion. Importance: Gastrointestinal nematodiasis poses significant economic losses in the livestock industry due to mortality, morbidity, and decreased production. Objective: This study examined the prevalence of gastrointestinal nematodiasis in dairy cattle in Central Inner Mongolia, Northern China, the associated risk factors, and the effects on the pepsinogen and gastrin levels. Methods: Fecal samples (n = 590) were collected from four regions and analyzed using the standard floatation and sedimentation techniques. The serum gastrin and pepsinogen levels were also assessed as potential biomarkers for gastrointestinal nematodiasis. Results: Among 590 animals, 259 (43.9%; 95% confidence interval, 36.7–49.8) tested positive for at least one type of gastrointestinal nematode parasite. Among the positive animals, 38.6% were poly-parasitic, while 61.4% were mono-parasitic (p < 0.05). Cooperia was the predominant nematode among mono-infections, accounting for 10.8%, followed in order by Oesophagostomum (10.0%), Trichuris (9.6%), Trichostrongylus (8.5%), Dictyocaulus (6.9%), Ascarid (6.9%), Haemonchus (4.6%), and Strongyloides (3.9%). Among the poly-parasitic infected cattle, 22.7%, 9.3%, 4.2%, and 2.3% had two, three, four, and five parasite combinations, respectively. The risk factors, such as age, breed, area, physical condition, and presence of diarrhea, were significantly (p < 0.05) associated with the prevalence of gastrointestinal nematodiasis. Poly-parasitic infected cattle showed 49.1% higher (p = 0.013) serum pepsinogen levels than the control group, while the mono-parasitic ones exhibited a 28.33% increase (p = 0.030). Similarly, the serum gastrin levels increased by 99.2% (p = 0.004) and 71.3% (p = 0.015) in the poly-parasitic and mono-parasitic animals, respectively. Conclusions and Relevance: These findings demonstrate the high prevalence of gastrointestinal nematodiasis in Northern China, with an alarming rate of poly-parasitic infections. Simple Summary: The infection of animals and humans with Sarcocystis spp. causes public health problems and significant economic losses to the livestock industry. To date, more than 200 species in the genus Sarcocystis have been reported. Sarcocystis spp. can cause sarcocystosis, which leads to emaciation, abortion, and even death in sheep. However, to date, Sarcocystis spp. infection has not been reported in sheep (Ovis aries) in Shanxi Province. In the present study, 582 mutton samples collected from sheep in ten representative counties in this province were investigated for Sarcocystis spp. by PCR amplification of the mitochondrial cytochrome c oxidase subunit I (cox1) gene. The results showed that the prevalence of Sarcocystis spp. in sheep in Shanxi Province was 33.85% (197/582). Of them, 196 cox1 sequences showed a nucleotide similarity of 98.56–99.81% with those of S. tenella, and the remaining one cox1 sequence showed a nucleotide similarity of 99.71% with that of S. arieticanis. The present study reported the occurrence and prevalence of Sarcocystis spp. for the first time in sheep in Shanxi Province, China, which has important implications for the control and prevention of Sarcocystis infection in this province and elsewhere. Sarcocystis species are intracellular coccidian protozoans that can infect a range of animals and humans and cause public health problems and economically significant losses. Sarcocystosis in sheep (Ovis aries) can cause abortion, neurological symptoms, and even death and results in significant economic losses to the livestock industry. To date, however, it is yet unknown whether sheep in Shanxi Province, north China, are infected with Sarcocystis spp. The purpose of this study was to investigate the prevalence of Sarcocystis spp. in sheep in Shanxi Province. Thus, 582 muscle samples of sheep were purchased from farmers' markets from ten representative counties in Shanxi Province, north China, and examined for the presence and prevalence of Sarcocystis spp. by PCR amplification of the mitochondrial cytochrome c oxidase subunit I (cox1) gene. Of the examined 582 mutton samples, 197 samples (33.85%) were Sarcocystis-positive and were sequenced. Of the obtained 197 cox1 sequences, 196 sequences showed nucleotide similarity of 98.56–99.81% with those of S. tenella, and the remaining one cox1 sequence showed nucleotide similarity of 99.71% with that of S. arieticanis. Two representative cox1 sequences of S. tenella (accession nos. PQ189447 and PQ189448) have 99.52% and 99.61% identity with S. tenalla (KC209725) and S. tenalla (MK419984), respectively. The sequence of S. arieticanis (accession no. PQ165949) obtained in this study has 99.71% identity with S. arieticanis (MK419975). This present study documents the occurrence and prevalence of Sarcocystis spp. in sheep in Shanxi Province for the first time, which enriches the data on the distribution of Sarcocystis spp. in sheep in China and has implications for the control of sheep sarcocystosis. Haemonchus contortus (the barber's pole worm)—a highly pathogenic gastric nematode of ruminants—causes significant economic losses in the livestock industry worldwide. H. contortus has become a valuable model organism for both fundamental and applied research (e.g., drug and vaccine discovery) because of the availability of well-defined laboratory strains (e.g., MHco3(ISE).N1 in the UK and Haecon-5 in Australia) and genomic, transcriptomic and proteomic data sets. Many recent investigations have relied heavily on the use of the chromosome-contiguous genome of MHco3(ISE).N1 in the absence of a genome for Haecon-5. However, there has been no genetic comparison of these and other strains to date. Here, we assembled and characterised the mitochondrial genome (14.1 kb) of Haecon-5 and compared it with that of MHco3(ISE).N1 and two other strains (i.e., McMaster and NZ_Hco_NP) from Australasia. We detected 276 synonymous and 25 non-synonymous single nucleotide polymorphisms (SNPs) within Haecon-5. Between the Haecon-5 and MHco3(ISE).N1 strains, we recorded 345 SNPs, 31 of which were non-synonymous and linked to fixed amino acid differences in seven protein-coding genes (nad5, nad6, nad1, atp6, nad2, cytb and nad4) between these strains. Pronounced variation (344 and 435 SNPs) was seen between Haecon-5 and each of the other two strains from Australasia. The question remains as to what impact these mitogenomic mutations might have on the biology and physiology of H. contortus, which warrants exploration. The high degree of mitogenomic variability recorded here among these strains suggests that further work should be undertaken to assess the nature and extent of the nuclear genomic variation within H. contortus. Mycoplasma bovis (M. bovis) is a significant bovine pathogen associated with various diseases, including bovine bronchopneumonia and mastitis resulting in substantial economic losses within the livestock industry. However, the development of effective control measures for M. bovis is hindered by a limited understanding of its virulence factors and pathogenesis. Nucleomodulins are newly identified secreted proteins of bacteria that internalize the host nuclei to regulate host cell gene expression and serve as critical virulence factors. Although recent reports have initiated exploration of mycoplasma nucleomodulins, the efficiency of conventional techniques for identification is very limited. Therefore, this study aimed to establish high-throughput methods to identify novel nucleomodulins of M. bovis. Using a direct biotinylation (DB) approach, a total of 289 proteins were identified including 66 high abundant proteins. In parallel, the use of proximity-based biotinylation (PBB), identified 28 proteins. Finally, seven nucleomodulins were verified to be nuclear by transfecting the bovine macrophage cell line BoMac with the plasmids encoding EGFP-fused proteins and observed with Opera Phenix, including the known nucleomodulin MbovP475 and six novel nucleomodulins. The novel nucleomodulins were four ribosomal proteins (MbovP599, MbovP678, MbovP710, and MbovP712), one transposase (MbovP790), and one conserved hypothetical protein (MbovP513). Among them, one unique nucleomodulin MbovP475 was identified with DB, two unique nucleomodulins (MbovP513 and MbovP710) with PBB, and four nucleomodulins by both. Foot-and-mouth disease virus (FMDV) is a highly contagious virus that affects cloven-hoofed animals and causes severe economic losses in the livestock industry. Given that this high-risk pathogen has to be handled in a biosafety level (BSL)-3 facility for safety reasons and the limited availability of BSL-3 laboratories, experiments on FMDV call for more attention. Therefore, we aimed to develop an FMDV experimental model that can be handled in BSL-2 laboratories. The NanoBiT luciferase (Nano-luc) assay is a well-known assay for studying protein–protein interactions. To apply the NanoBiT split luciferase assay to the diagnosis and evaluation of FMD, we developed an inactivated HiBiT-tagged Asia1 Shamir FMDV (AS-HiBiT), a recombinant Asia1 shamir FMDV with HiBiT attached to the VP1 region of Asia1 shamir FMDV. In addition, we established LgBiT-expressing LF-BK cell lines, termed LgBit-LF-BK cells. It was confirmed that inactivated AS-HiBiT infected LgBiT-LF-BK cells and produced a luminescence signal by binding to the intracellular LgBiT of LgBiT-LF-BK cells. In addition, the luminescence signal became stronger as the number of LgBiT-LF-BK cells increased or the concentration of inactivated AS-HiBiT increased. Moreover, we confirmed that inactivated AS-HiBiT can detect seroconversion in sera positive for FMDV-neutralizing antibodies. This NanoBiT split luciferase assay system can be used for the diagnosis and evaluation of FMD and expanded to FMD-like virus models to facilitate the evaluation of FMDV vaccines and antibodies. (1) Background: Group A rotaviruses (RVAs) are the primary cause of severe intestinal diseases in piglets. Porcine rotaviruses (PoRVs) are widely prevalent in Chinese farms, resulting in significant economic losses to the livestock industry. However, isolation of PoRVs is challenging, and their pathogenicity in piglets is not well understood. (2) Methods: We conducted clinical testing on a farm in Jiangsu Province, China, and isolated PoRV by continuously passaging on MA104 cells. Subsequently, the pathogenicity of the isolated strain in piglets was investigated. The piglets of the PoRV-infection group were orally inoculated with 1 mL of 1.0 × 106 TCID50 PoRV, whereas those of the mock-infection group were fed with an equivalent amount of DMEM. (3) Results: A G5P[23] genotype PoRV strain was successfully isolated from one of the positive samples and named RVA/Pig/China/JS/2023/G5P[23](JS). The genomic constellation of this strain was G5-P[23]-I5-R1-C1-M1-A8-N1-T1-E1-H1. Sequence analysis revealed that the genes VP3, VP7, NSP2, and NSP4 of the JS strain were closely related to human RVAs, whereas the remaining gene segments were closely related to porcine RVAs, indicating a reassortment between porcine and human strains. Furthermore, infection of 15-day-old piglets with the JS strain resulted in a diarrheal rate of 100% (8 of 8) and a mortality rate of 37.5% (3 of 8). (4) Conclusions: The isolated G5P[23] genotype rotavirus strain, which exhibited strong pathogenicity in piglets, may have resulted from recombination between porcine and human strains. Foot-and-mouth disease (FMD) is a highly infectious disease affecting cloven-hoofed animals and causes significant economic losses to the livestock industry. The Type O PanAsia-2 (O PA-2) vaccine strain is protective against a wide range of serotype O FMD virus (FMDV) strains in East Asia, and A22 Iraq/24/64 (A22 IRQ) is the most widely used vaccine strain in FMD vaccine antigen banks. The aim of this study was to produce antigens from O PA-2 and A22 IRQ viruses using a 100 L bioreactor and evaluate the protective efficacy of varying antigen concentrations in pigs. More than 2 μg/mL of the antigen was recovered from the O PA-2 and A22 IRQ virus-infected supernatants. Further, inactivation of O PA-2 and A22 IRQ by binary ethyleneimine revealed that the viral titers decreased below 10−7 TCID50/mL within 13 h and 9 h, respectively. The O PA-2 and A22 IRQ vaccines, containing 10 μg and 5 μg of antigen, respectively, provided protection against homologous viruses in pigs. This is the first report demonstrating that the antigens obtained from the pilot-scale production of O PA-2 and A22 IRQ are viable candidate vaccines. Pimelea is a genus of about 140 plant species, some of which are well-known for causing animal poisoning resulting in significant economic losses to the Australian livestock industry. The main poisonous species/subspecies include Pimelea simplex (subsp. simplex and subsp. continua), P. trichostachya and P. elongata (generally referred to as Pimelea). These plants contain a diterpenoid orthoester toxin, called simplexin. Pimelea poisoning is known to cause the death of cattle (Bos taurus and B. indicus) or weaken surviving animals. Pimelea species are well-adapted native plants, and their diaspores (single seeded fruits) possess variable degrees of dormancy. Hence, the diaspores do not generally germinate in the same recruitment event, which makes management difficult, necessitating the development of integrated management strategies based on infestation circumstances (e.g., size and density). For example, the integration of herbicides with physical control techniques, competitive pasture establishment and tactical grazing could be effective in some situations. However, such options have not been widely adopted at the field level to mitigate ongoing management challenges. This systematic review provides a valuable synthesis of the current knowledge on the biology, ecology, and management of poisonous Pimelea species with a focus on the Australian livestock industry while identifying potential avenues for future research. The spotted alfalfa aphid (SAA, Therioaphis trifolii) (Hemiptera: Aphididae) is a destructive pest of cultivated alfalfa (Medicago sativa L.) that leads to large financial losses in the livestock industry around the world. Here, we present a chromosome-scale genome assembly of T. trifolii, the first genome assembly for the aphid subfamily Calaphidinae. Using PacBio long-read sequencing, Illumina sequencing, and Hi-C scaffolding techniques, a 541.26 Mb genome was generated, with 90.01% of the assembly anchored into eight scaffolds, and the contig and scaffold N50 are 2.54 Mb and 44.77 Mb, respectively. BUSCO assessment showed a completeness score of 96.6%. A total of 13,684 protein-coding genes were predicted. The high-quality genome assembly of T. trifolii not only provides a genomic resource for the more complete analysis of aphid evolution, but also provides insights into the ecological adaptation and insecticide resistance of T. trifolii. Microbial biofilms defined as extremely complex ecosystems are considered clinically important for humans. However, the concept and significant roles of microbial biofilms in the progression of disease have seriously lagged in veterinary medicine, when compared with human medicine. Although the importance of biofilms in animal health is just beginning to emerge, limited studies have paid attention that microbial biofilms are clinically important in the field of veterinary medicine, and lead to serious economic losses. In this review, the importance of microbial biofilms causing high economic losses in the livestock industry has been highlighted. Besides, the concept of microbial biofilm, their role in the pathogenesis of the animal diseases, as well as diagnosis approaches and possible therapeutic strategies needed to overcome their detrimental effects in veterinary medicine, have been discussed. High ambient temperature is one of the major causes of economic losses in the livestock industry. The poultry industry is an integral part of the livestock industry. It faces severe losses due to heat stress (HS). The adverse effects of HS can be seen on production performance, body temperature, intestinal health, appetite hormone regulation, immune responses and oxidative characteristics. It is important to monitor these parameters to identify the HS possessions during rearing so that timely action can be taken to minimize the adverse effects of high ambient temperature. Furthermore, the application of productive methods on farms is equally important. Several strategies have been suggested by researchers. Providing a suitable environment with selective rearing systems along with proper ventilation and hygiene is the basic requirement for all types of livestock reared for animal protein. Supplementation of appropriate feed additive could be useful for improving intestinal absorption and minimizing adverse effects of HS. Selection for breeding heat resistant birds also provide merits for improving the germplasm of the strains. Early age thermal conditioning also helps in developing resistance for HS. The most recent advancement is the supplementation of active substances during incubation. It is expected that these methods may have a potential impact on the poultry industry for creating thermotolerance in the newly hatched chicks. Coccidiosis is one of the most pathogenic intestinal diseases caused by different species of Eimeria spp. (Phylum:Apicomplexa), that cause important economic losses to the livestock industry. Given the importance of camel breeding and its products in the regional economy, in this study the risk factors of prevalence of camels Coccidiosis in the Turkestan region of Republic of Kazakhstan was investigated by oocyte excretion monitoring in the herd. The results revealed that the prevalence of Eimeria sp, was136 (42.5%); and three types of Eimeria spp. were identified: Eimeriabactriani, Eimeriacameli, Eimeria dromedarii. The statistical analysisdemonstratedthat the prevalence and intensity of infection in camel ≤1-year-old were 65.5% and 149.2 respectively, and they were more likely to be infected with Eimeria spp. compared with adult camels (22.5%, 5.7), (pvalue≤0.05). Another probable risk factor is seasons of the year as it seems the high and low prevalence andinfection intensity are observed in summer (60%, 102.1) and winter (20.6%, 21.25), respectively. Multivariate analysis of our data revealed that age and season were significant risk factors (p< 0.005) and adoption of hygienic measures and husbandry practice are needed among the high risk groups(in young camel and summer season) to minimize, control and prevent spread of the infection. Globally, climate change is an alarming threat to the livestock industry. Such changes in the climate can also adversely affect the returns of livestock farmers in Pakistan. Improvement in the production process could decrease the risk of losses. This study analyzes the efficacy of silage to abate the losses in livestock profitability resulting from the climate change. The study employed cross-sectional survey data of 492 livestock farmers collected from six districts of Punjab Province, Pakistan. The data are analyzed with endogenous switching regression, considering the possibility of selection bias and endogeneity in adopting silage as a climate change adaptation measure. The study findings show a significant difference in material well-being between silage users and conventional feed users. Furthermore, training programs such as silage training and livestock development programs could be meaningful provisions to abate climate change and improve food security. It is suggested that training and development programs should be incorporated in policy plans to improve the well-being of farmers in terms of their farm revenues. [ABSTRACT FROM AUTHOR]

Brucellosis has re‐emerged in China in recent years, resulting in an increasing health burden and economic losses for humans and the livestock industries. This study integrated data from human and livestock brucellosis surveillance systems to explore the changing epidemiology of brucellosis from 2004 to 2018 in China. A total of 524,980 human cases of brucellosis were reported, with the average annual incidence in humans being significantly higher for the period 2012–2018 than for 2004–2011 (3.3 vs. 1.9 per 100,000 residents). An autoregressive integrated moving average (ARIMA) model predicted an upward trend in the monthly incidence of brucellosis in humans in 2019 and 2020. Characteristics including being male, aged 45–54 years, working in the livestock industries, and residing in the northern provinces of China increased the risk of people contracting brucellosis. The percentage of provinces with infected people increased from 67.7% (21/31) in 2004 to all provinces in 2018. A total of 29,115 outbreaks were reported in livestock from 2004 to 2018, with 443,883 seropositive animals although only 381,224 (85.9%) of these were culled. The monthly incidence of brucellosis in humans was strongly positively correlated (r =.539, p <.001) with the number of outbreaks of brucellosis in livestock reported 3 months prior to the human cases. At the provincial level, the annual incidence of brucellosis in humans was significantly positively correlated with the sheep population (r =.786, p <.01). In conclusion, brucellosis in humans and livestock has been spreading in mainland China in the past decade. A more active surveillance of brucellosis in both livestock and humans in China should be coordinated and adjusted by adopting an evidence‐based 'One Health' approach, particularly in high‐risk regions and livestock industries. Livestock plays a significant role in the economy of a nation but its productivity can be hampered by numerous haemoparasites thereby leading to economic losses to the livestock industry. The prevalence of haemoparasite in cattle slaughtered at Jalingo abattoir was investigated. A total of four hundred blood samples were collected at the point of slaughter, processed, and screened for haemoparasites by examining Giemsa-stained thin blood smears. An overall prevalence of 12.25% was recorded. Four haemoparasites of cattle with prevalence rates of 5.0%, 6.75%, 0.25%, and 0.25% for Anaplasma, Babesia, Microfilaria and Trypanosoma respectively were observed. The prevalence of haemoparasite in relation to sex, revealed higher infection in females (13.75%) than in males (10.0%) which were not found statistically different (P > 0.05). All breeds encountered during the study were infected with haemoparasites with the highest prevalence of 13.91 % recorded in White Fulani, Red Bororo (10.94%), and Sokoto Gudali (10.00 %), while Adamawa Gudali had the least prevalence of 0.5%. There was no statistically significant difference in the prevalence of haemoparasite in relation to breeds (P > 0.05). High prevalence was observed in the young (14.29%) more than the adult (11.59%) and older (12.14%). The current study has revealed the haemoparasites status in cattle slaughtered at Jalingo abattoir. • A novel artificial bacterial consortium was constructed for DON biodegradation. • Paracoccus yeei A9 provides electron for Devosi a sp. A8 to degrade DON. • The DON degradation product was identified as 3-keto-DON. • Several trichothecenes could be degraded by the artificial bacterial consortium. Deoxynivalenol (DON) is a trichothecene mycotoxin mainly produced by various Fusarium species which cause Fusarium head blight in cereals. DON accumulation in grains not only poses a serious health threat to human and animals but also causes economic loss in the food and livestock industries. Here we reported the construction of an artificial bacterial consortium comprising of Devosia sp. A8 and Paracoccus yeei A9 isolated from an enriched bacterial culture, which shows effective degradation capability towards DON. The optimal conditions for temperature, pH and initial inoculum volume for DON degradation were 35 °C, pH 8–9 and 0.5 %, respectively. The degradation product was identified as 3-keto-DON by LC-MS/MS and 1H NMR spectra. Devosia sp. A8 played the major role in DON degradation, but the enhancement of the DON degradation activity by adding NADPH or riboflavin suggested that Paracoccus yeei A9 provided electrons for Devosia sp. A8 to degrade DON. Moreover, other trichothecenes including 15-acetyl DON, 3-acetyl DON and T-2 toxin were also degraded by the consortium. To the best of our knowledge, this is the first report about a Devosia species capable of degrading DON when co-cultured with Paracoccus yeei. The results demonstrated that the artificial bacterial consortium is a potential candidate for DON biodegradation as well as other trichothecene mycotoxins in a safe and eco-friendly manner. The article provides information on the degree of domestic livestock loss caused by natural hazard such as floods and typhoons in the Philippines during 1750 to 1898. The author discusses several natural calamities in the different areas in the country such as in Ilocos Norte, Bulacan and Leyte, wherein these tragic events destroyed both life and property. Archival sources made frequent reference to the dead animal bodies thrown up by the sea or trapped in town proper. Not only terrestrial animals were the victims of natural hazard, the marine creatures were also sometimes affected. In conclusion, the author states that the documentary evidence in the country showed that domesticated animals were at risk from natural hazards and died in large numbers during the nineteenth century.

Each year, foot-and-mouth disease leads to enormous economic losses to the livestock industry. Currently, the killed whole virus is widely using to control FMD. However, vaccination is constrained by lack of or incomplete protection. Therefore, along with vaccination, we need to find the antivirals against FMD. This study was conducted to investigate the antiviral potential of ivermectin against multiple serotypes of FMDV. Initially, an MTT assay was performed on the BHK-21 cell line to determine assay ivermectin cytotoxicity. Viral inhibition assays using the non-cytotoxic concentration of ivermectin were performed to check the antiviral potential of ivermectin on different stages of virus replication. At 2.5 μM and 5 μM concentrations of ivermectin, the virus titer was reduced significantly (p < 0.001) by two to three log in all three strains of viruses at both non-toxic concentrations (2.5 and 5 μM). The virus titer in strain O control was 106.0 TCID 50 /0.1 mL and was reduced to 104.1 TCID 50 /0.1 mL at a concentration of 2.5 μM and 103.10 TCID 50 /0.1 mL at 5 μM concentration. In the case of strain Asia-1, the virus titer was reduced to 103.8 TCID 50 /0.1 mL at 2.5 μM and 103.01TCID 50 /0.1 mL at 5 μM concentration. The titer of strain A was reduced from 105.8 TCID 50 /0.1 mL to 103.9 TCID 50 /0.1 mL at 2.5 μM concentration and 103.1 TCID 50 /0.1 mL at 5 μM concentration. Moreover, the virus titer was reduced more at the replication stage as compared to attachment and entry stages.
