Although garlic polysaccharides have been found to possess anti-inflammatory activities, anti-inflammatory study on small molecule water-soluble garlic polysaccharide (WSGP) is few. In this study, a novel WSGP with a molecular weight of 1853 Da was isolated by DEAE-52 and Sephadex G-100 column and the chemical composition was identified by monosaccharide composition and methylation analysis. Furthermore, the antioxidant effects of WSGP and the potential molecular mechanisms on LPS-induced inflammatory responses in RAW264.7 macrophage cells were investigated. The results showed that WSGP has strong antioxidant activity, such as DPPH, hydroxyl, superoxide anion, ABTS radical scavenging capacity, Fe2+ chelating ability and reducing power. Meanwhile, WSGP could considerably suppress the manufacturing of NO and the mRNA and protein expression degrees of IL-6, TNF-α, and IL-1β in LPS inspired RAW264.7 macrophages WSGP could significantly suppress the production of NO and the mRNA and protein expression levels of IL-1β, IL-6, and TNF-α in LPS stimulated RAW264.7 macrophage cells (p < 0.05). In addition, the phosphorylated IκB-α, p65, and STAT3 proteins were significantly increased in LPS-induced macrophages, while this trend was significantly reversed by WSGP treatment in a concentration-dependent manner (p < 0.05). Consequently, WSGP supplementation might reduce LPS-induced inflammatory responses by suppressing proinflammatory cytokines and NF-κB and STAT3 pathway activation. The finding of this research would give scientific guidelines for the judicious use of small molecular garlic polysaccharide in anti-inflammatory treatments.

Long-term non-healing diabetic wounds are always a serious challenge and a global healthcare burden that needs to be resolved urgently in the clinic. Prolonged inflammation and impaired angiogenesis are the main direct causes of diabetic wounds. With the development of polymer biomaterials, various wound dressings have been created, but a few of them have been applied to the clinical management of diabetic wounds. Here, we developed a mussel-inspired bioactive scaffold consisting mainly of collagen and hyaluronic acid, which are natural biopolymer materials contained in human tissues. First, we fabricated different polydopamine modified lyophilized collagen hyaluronic acid scaffolds under different concentrations of dopamine alkaline solutions, 0.5, 1, 2 ​mg/mL, so named CHS-PDA-0.5, CHS-PDA-1, CHS-PDA-2. After testing their physical and chemical properties, antioxidant effect, inflammation regulation, as well as drug loading and release capabilities, we obtained a bioactive endothelial growth factor (EGF)-loaded wound dressing, CHS-PDA-2@EGF, which can resist reactive oxygen species (ROS) and promote the regeneration of chronic wounds in diabetic rats by reducing inflammation. In addition, the scaffold showed excellent swelling ability, a certain coagulation effect and reasonable degradation. Therefore, the scaffold has great potential to be used in clinical diabetic wound treatment as a low-cost and easily available wound dressing to accelerate chronic wound healing.

Scorias spongiosa, as an edible fungus, has multiple health benefits. However, the effects of S. spongiosa on intestinal health are rarely explored. Hence, our study aims to elaborate on the influences of S. spongiosa polysaccharides (SSPs) on antioxidant, anti-inflammatory, and intestinal microflora in C57BL/6J mice. In the present study, 18 male mice were randomly distributed into three groups: (1) Control group (CON); (2) Low dose SSPs group (LSSP); (3) High dose SSPs group (HSSP). After 14-day administration, the jejunum and serum samples were collected for detection. The results showed that SSPs exert no effects on the growth performance of mice regardless of doses. Meanwhile, SSPs administration reduced the serum pro-inflammatory cytokines and elevated the anti-inflammatory cytokines. Moreover, the antioxidant capacity was elevated by SSPs administration, as evidenced by the increased contents of T-AOC, GSH-Px, and the decreased content of MDA. Mechanistically, the administration of SSPs enhanced the protein abundances of p-Nrf2, Keap1, and HO-1 in mice. The results of 16S rDNA demonstrated that the microbial community and composition were altered by SSPs administration. To summarize, SSPs benefit intestinal health in C57BL/6J mice via a mechanism that involves elevating antioxidant and anti-inflammatory activities and regulating intestinal microbiota.

• Polymorphism of formed NiO nanoparticles with size in 25-50 nm was observed. • High toxicity of nanoparticles was witnessed against MG-63 cancer cells. • NiO nanoparticles revealed high biocompatibility on HEK293 normal cells. • Nanoparticles showed bactericidal activity on various human pathogenic bacteria. The green synthesis of metal oxide nanoparticles is interestingly becoming familiar as the reason of its non-toxicity, economical, eco-friendly, and commerciality. The secondary metabolites and bioactive molecules in biomaterial extract play an important role of reducing, capping, stabilizing and chelating activities to form the nanoparticles as a substitute of hazardous chemical surfactants. Hence this present work concentrated on the biogenic synthesis of NiO nanoparticles using grains extract of Oryza longistaminata (red rice), and their toxicity assessment as well as biomedical applications. The synthesized nanoparticles were widely characterized by using several analytical and biomedical techniques. The reduction mechanism of Ni2+ ions using major active phytocompound in red rice grains extract was stated. UV–visible spectroscopic analysis revealed the formation of NiO nanoparticles by emerging the sharp surface Plasmon resonance band at 326 nm. The absorption peaks of red rice grains extract at 206, 260, 330 and 410 nm stated the presence of phytocompounds in them. X-ray diffraction pattern explicated the high crystallinity of NiO nanoparticles and their average crystallite size was found to be at 36 nm. The diffraction pattern of red rice grains revealed the amorphous characteristic. Fourier transform infrared analysis displayed the different vibrational functional groups of nanoparticles and red rice grains. Dynamic light scattering analysis showed the particle size distribution was in between 30-65 nm. High-resolution transmission electron microscopic analysis showed the hexagonal, rectangular, oval and spherical shaped polymorphism of NiO nanoparticles and their size was in 25-50 nm. Energy dispersive X-ray spectrum and mapping analyses validated the formation of NiO nanoparticles by the major existence of Ni and O elements. The high toxicity of nanoparticles towards MG-63 osteosarcoma cancer cells was revealed at 94.69% in 100 μg/mL concentration. The nanoparticles showed no severe toxicity against HEK293 normal cells and red blood cells till higher concentration. The biomedical analyses such as antioxidant, anti-inflammatory and anti-diabetic activities exposed the potential bio-drug activity of NiO nanoparticles with maximum activity of 86.22, 84.23 and 85.18% in the high concentration of 80 μg/mL. The antibacterial activity divulged the good bactericidal drug nature of NiO nanoparticles against both Gram-positive (S. aureus and B. Subtilis) and Gram-negative (S. typhi and K. pneumonia) bacteria with low MBC/MIC ratio. As per the assessment of results, the biosynthesized NiO nanoparticles have non-carcinogenic, high biocompatible and potent nature and it suggests that they can be used as therapeutic candidates in biomedical, clinical and pharmaceutical fields for treating various diseases.
