In this study, modified sea Cucumber Peptides (SCP) were prepared by reacting with xylooligosaccharide (XOS) and alginate oligosaccharides (AOS) via glycation. Free radical inhibitory and inhibition of oxidative stress of modified SCP was evaluated using human hepatocellular carcinoma (HepG2) cells and zebrafish embryos. LC-MS analysis revealed that SCPs mainly consist of 40 active peptides, with an average molecular weight of 1122.168 Da and an average length of 11 amino acid residues. For amino acid composition, L-Asparagine, L-Methionine, and L-Aspartic Acid were dominant amino acids in SCP. The result showed that the antioxidant ability of SCP against 2,2-Diphenyl-1-picrylhydrazyl (DPPH), superoxide anion radical (O -2 ), and Hydroxyl Radical (OH) was significantly improved after modification. In HepG2 cells, the modified SCP showed stronger protection than native SCP native against H 2 O 2 -induced oxidative stress by enhancing cell viability and reducing radical oxygen species (ROS) generation. The inhibition effect of SCP was increased after modification with XOS and AOS by 13 % and 19 % respectively. Further studies displayed that the activity of antioxidative enzymes, including Superoxide dismutase (SOD), Glutathione Peroxidase (GPx), and catalase (CAT), was remarkably enhanced, whereas malondialdehyde (MDA) level was reduced compared with native SCP and H 2 O 2 -treated groups, thus, improving the intracellular antioxidant defenses. The gene expression analysis showed that the mechanism underlying the modified SCP protective effect may be linked with the capability to regulate Nuclear factor-erythroid factor 2-related factor 2 (NRF2) gene expression. The protective effect of modified SCP against H 2 O 2 in vitro was confirmed in vivo by reduced toxicity in zebrafish embryos via improvement of mortality rate, hatching rate, heart beating rate, and deformities of the zebrafish model. However, SCPAOS conjugate displayed greater antioxidant potentials compared to the SCPXOS, the different effects between SCPAOS and SCPXOS could be due to their different antioxidant activity. Underutilized vegetables are currently studied not only for their nutrient values but also for their health-promoting components for protection against chronic diseases. The present study was performed to evaluate chemical compositions and antioxidant properties of underutilized vegetable palm hearts, namely, lalis (Plectocomiopsis geminiflora) and pantu (Eugeissona insignis). Additionally, the vegetable extracts were evaluated for their activities in the inhibition of digestive enzymes and effects on insulin secretion using BRIN BD11 pancreatic cell lines. Both vegetables contain valuable sources of dietary fiber, potassium, and zinc. For the first time, the phenolic compounds of the vegetables were identified and quantified using HPLC-DAD and LC-ESI-MS. Appreciable amounts of chlorogenic acid were found in the studied vegetables. The sample extracts exhibited potential antioxidant capacities through chemical and biological in vitro assays. High inhibition of α-amylase activity (>50%) was found from the extracts. Thus, it was suggested the vegetable consumption could fulfill the nutrient requirements among local communities.

Abstract: The optimum extraction conditions for highest recovery of total phenolics content (TPC) and antioxidant capacities (AC) were analysed for Mangifera pajang peels (MPP), using response surface methodology. The effects of ethanol concentration (X 1: 20–80%), extraction temperature (X 2: 30–65°C) and liquid-to-solid ratio (X 3: 20–50mL/g) on the recovery of total phenolics (Y 1) and antioxidant capacity (Y 2) were investigated. A second order polynomial model produced a satisfactory fitting of the experimental data with regard to total phenolic content (R 2 =0.9966, p <0.0001) and antioxidant capacity (R 2 =0.9953, p <0.0001). The optimum extraction conditions for TPC were 68%, 55°C and 32.7mL/g, and for AC were 68%, 56°C and 31.8mL/g, respectively. Predicted values for extraction of TPC and AC agreed well with the experimental values. Liquid chromatography–mass spectrometry of the optimally obtained extracts from MPP revealed the major phytochemicals as mangiferin, gallic acid, catechin and epicatechin. [Copyright &y& Elsevier]

Abstract: The composition of bambangan peel dietary fibre (DF) and several properties related to its nutritional quality were investigated. The physicochemical properties and antioxidant capacity for the fibre-rich powder (FRP) were investigated, and the chemical composition for soluble dietary fibre (SDF) and insoluble dietary fibre (IDF) were also studied. The FRP had a high amount of total dietary fibre (TDF; 72.3g/100g FRP) with a balanced SDF/IDF ratio (46.3/53.7%). The FRP had a high glucose retardation index, water-holding capacity (WHC), oil-holding capacity (OHC), and swelling. The antioxidant capacity of the FRP, as determined by the DPPH assay using an Elisa reader, exhibited a strong potency due to the presence of associated total polyphenols (98.3mg/g FRP). These FRP characteristics indicated that bambangan peels are a rich source of DF, antioxidants, and other bioactive compounds that can be incorporated with food products to improve the nutraceutical properties of these products. Abstract: Polysaccharides were isolated from the fibrous pulp of bambangan (Mangifera pajang Kort.). Neutral and acidic polysaccharides were separated using DEAE-Cellulose. Size exclusion chromatography analyses showed that the average molecular weight (MW) of the neutral M. pajang polysaccharides (F1) was approximately 7 kDa, and those of three acidic polysaccharides (F2, F3 and F4) were approximately 13, 24 and 9 kDa, respectively. The monosaccharide compositions of these polysaccharides were determined using high performance liquid chromatography. F1 contained erythrose, rhamnose, arabinose, mannose, fructose and glucose (5, 7, 21, 42, 4 and 21 mg/100 mg fraction respectively), F2 consisted of rhamnose, xylose and arabinose (33, 7 and 51 mg/100 mg fraction respectively), F3 consisted of fructose (14 mg/100 mg fraction) and glucose (72 mg/100 mg fraction), and F4 comprised arabinose, mannose, fructose and glucose (32, 36, 2 and 10 mg/100 mg fraction respectively). Results of Fourier transform infrared spectroscopy and the monosaccharide compositions suggested that the fibrous pulp of M. pajang fruit consisted of heteropolysaccharide and belonged to α and β–type of the pyran group. Additionally, crude polysaccharide and its fractions showed strong antioxidant activities. The acidic polysaccharides had the highest antioxidant activity and should be considered as a prospective antioxidant. [Copyright &y& Elsevier]
