Background: Active principles from natural sources, in the form of extracts and natural compounds, provide an infinite number of bioactive compounds with consummate disposal of chemical diversity. These compounds and active principles are of utmost importance in the discovery of drugs of biological origin particularly, from plants. Objective: Development of resourceful technology for the isolation and extraction of bioactive compounds of medicinal importance is considered as an important task for researchers. There are a number of extraction, isolation, and characterization techniques currently utilized; however, most are laborious and use toxic chemicals and huge quantities of raw materials with a very low output. There are a number of abiotic and biotic factors that affects the quality and the quantity of plants bioactive compounds. Considering this, the objectives of the current review are to discuss the various extraction and characterization techniques used to isolate the essential bioactive compounds from three plant species and the biotic and abiotic factors that affect the quantity and quality of the plants secondary metabolites. Methods: Many advanced technologies have been developed and tested for extraction, characterization, and their capacity for high yield products, and those requiring less application of toxic solvents are investigated continuously. Conclusion: In this context, the present review summarizes the different types of extraction and characterization techniques utilized commercially by the food, drug, and pharmaceutical industries for better output and environmentally- and healthbenefiting products with special reference to three industrially important plants: Leonotis leonurus (L.) R.Br. (Lamiaceae) and Santalum album L. (Santalaceae) and Aloe vera (L.) Burm. f. (Aloaceae or Asphodelaceae).

Nutritional supplementations are a form of nutrition sources that may help in improving the health complexities of a person throughout his or her life span. Being also categorized as food supplementations, nutraceuticals are products that are extracted from edible sources with medical benefits as well as primary nutritional values. Nutraceuticals can be considered as functional foods. There are evidences that nutraceutical supplementations can alter the commensal gut microbiota and help to prevent or fight against chronic non-communicable degenerative diseases in adults, including neurological disorders (Autism Spectrum Disorder [ASD], Parkinson’s disease [PD], Multiple sclerosis [MS]) and metabolic disorders (Type-II diabetes, obesity and non-alcoholic fatty liver disease). They can even lessen the complexities of preterm babies like extra-uterine growth restriction, necrotizing enterocolitis, infant eczema and allergy (during pregnancy) as well as bronchopulmonary dysplasia. Molecular perception of inflammatory and apoptotic modulators regulating the pathogenesis of these health risks, their control and management by probiotics and prebiotics could further emphasize the scientific overview of their utility. In this study, the pivotal role of nutraceutical supplementations in regulating or modulating molecular pathways in the above non-communicable diseases is briefly described. This work also gives an overall introduction of the sophisticated genome-editing techniques and advanced delivery systems in therapeutic activities applicable under these health risks.

Background Korean traditional food (KTF), originated from ancestral agriculture and the nomadic traditions of the Korean peninsula and southern Manchuria, is based on healthy food that balances disease prevention and treatment. Fermented foods that include grains, herbs, fruits, and mushrooms are also an important practice in KTF, providing high levels of Lactobacilli, which confer relevant health benefits, including antiviral properties. Some of these probiotics may also protect against the Influenza virus through the modulation of innate immunity. Scope and approach The emerging of the COVID-19 pandemic, in addition to other diseases of viral origin, and the problems associated with other respiratory disorders, highlight how essential is a healthy eating pattern to strengthen our immune system. Key Findings and Conclusions: The present review covers the information available on edible plants, herbs, mushrooms, and preparations used in KTF to outline their multiple medicinal effects (e.g., antidiabetic, chemopreventive, antioxidative, anti-inflammatory, antibacterial), emphasizing their role and effects on the immune system with an emphasis on modulating properties of the gut microbiota that further support strong respiratory immunity. Potential functional foods commonly used in Korean cuisine such as Kimchi (a mixture of fermented vegetables), Meju, Doenjang, Jeotgal, and Mekgeolli and fermented sauces, among others, are highlighted for their great potential to improve gut-lung immunity. The traditional Korean diet and dietary mechanisms that may target viruses ACE-2 receptors or affect any step of a virus infection pathway that can determine a patient's prognosis are also highlighted. The regular oral intake of bioactive ingredients used in Korean foods can offer protection for some viral diseases, through protective and immunomodulatory effects, as evidenced in pre-clinical and clinical studies.

In the search for novel, natural melanogenesis inhibitors, a new sesquiterpene, inularin, was isolated from the flowers of Inula britannica, and the structure was determined using spectroscopic and chemical methods. The antimelanogenic effects of inularin on B16F10 melanoma cells and zebrafish embryos were evaluated. Inularin dose-dependently reduced melanocyte-stimulating hormone-induced melanin production and L-DOPA oxidation in B16F10 cells. Zebrafish embryos were used to confirm the antimelanogenic activity. Inularin significantly decreased the pigmentation of embryos compared with untreated controls.

In the present study, we investigated the effects of Galla Rhois (GR) on obesity and gene expression. We prepared a GR extract and various solvent fractions and evaluated the degree to which they inhibited adipocyte differentiation and adipogenesis in vitro. Among them, the GR ethyl acetate fraction (GE) had the lowest EC50 for adipocyte differentiation and adipogenesis and thus was selected for in vivo experiments. We induced obesity in C57BL/6 mice by providing them a high-fat diet (HFD). Then, GE (10-40 mg/kg) or orlistat (positive control, 4 mg/kg) was orally administered daily for six weeks. Mean body weights and weight gain were significantly lower in the GE40 group (40 mg/kg of GE) compared with the HFD group (p < 0.05). The most significant changes in serum glucose, total cholesterol, and triglyceride levels were confirmed in the GE40 group (p < 0.05). Epididymal fat was weighed and stained for body fat measurement, and significant differences were recorded from GE10 to GE40 (p < 0.05). Finally, 3T3-L1 pre-adipocytes were treated with GE, and cDNA from these cells was used for microarray analysis and qRT-PCR. Microarray analysis revealed 13 genes up-regulated and 21 genes down-regulated by GE. From the qRT-PCR analysis, we found that GE altered the mRNA expression of eosinophil peroxidase, glucose-dependent insulinotropic polypeptide receptor, and apolipoprotein B. Based on this study, we suggest that GR could be developed as an anti-obesity therapeutic agent.

Background The synthesis of silver nanoparticles (AgNPs) using food waste materials and their biomedical applications have garnered considerable attention recently. Methods Here, we investigated the synthesis of AgNPs using an aqueous extract of outer peel of Pisum sativum under different lighting conditions using standard procedures and explored their antidiabetic, cytotoxicity, antioxidant, and antibacterial potential. Results Characterization of AgNPs was done by Ultra Violet (UV-VIS) spectroscopy that showed absorption maxima at 456 nm for the samples exposed to laboratory lighting and at 464 nm for the samples exposed to direct sunlight, by scanning electron microscopy and energy-dispersive X-ray analysis that showed the surface nature and their elemental composition with a strong peak at 3 keV that corresponded to Ag (61.85 wt%), by Fourier-transform infrared spectroscopy that predicted the functional groups involved, and by X-ray powder diffraction that showed the structural properties. The average diameter of the synthesized AgNPs was calculated to be in the range of 10-25 nm. AgNPs exhibited promising antidiabetic activity as determined by inhibition of α-glucosidase (95.29% inhibition at 10 µg/mL and IC50 value of 2.10 µg/mL) and cytotoxicity (IC50 value 4.0 µg/mL as calculated from the slope equation) against HepG2 cells. Furthermore, they also exhibited moderate antioxidant activity (50.17% reduction of 1,1-diphenyl-2-picrylhydrazyl at 100 µg/mL) and antibacterial activity against four human pathogenic bacteria (as indicated by 8.70-11.10 mm inhibition zones on agar plates). Conclusion In conclusion, the results confirm that food waste can be used in the synthesis of AgNPs and that the latter have the potential for applications in various fields including diabetic and cancer treatments as well as in biomedicine for the manufacture of antibacterial coatings in medical devices and instruments.

Background Ipomoea batatas (L.) Lam.(Ib) has high content of various beneficial nutrients which helps in improving and maintaining human health. It is well known as a functional food and also a valuable source of unique natural products. It contains various phenolic and flavonoid bioactive compounds.

Kenaf biomass (KB) was employed as feedstock for the synthesis of polyhydroxybutyrate (PHB) using Ralstonia eutropha to replace conventional petroleum-derived polymers. Various pretreatments followed by enzymatic saccharification were applied to release monomeric sugars from KB for PHB production. The effects of increasing concentration of Na2CO3 + Na2SO3 (NaC + NaS) pretreated KB hydrolysates (20–40 g/L) on PHB production were investigated. NaC + NaS pretreated KB hydrolysates (30 g/L) exhibited maximum 70.0% PHA accumulation, with PHB titers of 10.10 g/L and PHB yields of about 0.488 g/g of reducing sugar produced within 36 h of fermentation. PHA accumulation, PHB yield and R. eutropha growth performance using KB hydrolysates were found to be comparable with those of synthetic sugar mixture. Characterization of the produced PHB in terms of crystalline structure, and thermal properties was done using various analytical techniques and results coincide with standard PHB. Thus, green liquor pretreated KB hydrolysates deliver a promising and economically feasible carbon substrate for PHB production.

Background: Cancer remains a major health concern throughout history and is responsible for huge numbers of deaths globally. The sensitivity of cancer cells to anticancer drugs is a crucial factor for developing effective treatments. Methods: Pyrrolo[1,2-a]azepines coupled with benzothiazole and fluorinated aryl thiourea scaffolds have been synthesized, and their potential as cytotoxic agents was investigated against different cancer cell lines such as human ovarian cancer (SK-OV-3), cervical cancer (HeLa), colon adenocarcinoma (HT-29) and non-small-cell lung carcinoma (A549). Cytotoxicity of new compounds was confirmed using SRB assay against non-cancer MDCK cell line. In addition, free radical scavenging activity of new pyrrolo[1,2-a]azepines was examined by adopting DPPH and ABTS assays. Results: The results concluded that the presence and position of fluorine atom(s) on the thiourea unit played a significant role in order to gain anticipated efficacies. Results of the cytotoxic assay against non-cancer MDCK cells showed that these new derivatives are safe to study further. New structures were confirmed using spectral and elemental analyses. Conclusion: Pyrrolo[1,2-a]azepines endowed with a benzothiazole entity and fluorinated aryl thiourea substituents were derived aiming to furnish remarkable antioxidant and anticancer activities. New molecules generated showed interesting biological result correlated with the structure and substituent of the final derivatives. Specifically, numbers and position of fluorine atoms on the thiourea unit influenced the biological profile of the mentioned compounds.

Silver nanoparticles (AgNPs) have been synthesized by various ways but the green technology methods using food waste materials has been accepted now a days for their cost effectiveness, environmental friendly and effective biomedical application. In the present study an attempt has been made to synthesize AgNPs by using the outer accrescent fruiting calyx of Physalis peruviana fruits, a food waste material under different light source condition, and to investigate their cytotoxic activity against the HepG2 cells as well as their antibacterial and radical scavenging potential. The surface morphology and elemental composition of the biosynthesized AgNPs were evaluated by scanning electron microscopy, energy-dispersive X-ray analysis, and X-ray powder diffraction analysis. Fourier transform infrared spectroscopy of the sample extract and AgNPs was performed to determine the involvement of functional groups in the synthesis, capping, and stabilization process. The AgNPs showed promising cytotoxic activity against the HepG2 cells in a dose-dependent manner. The biosynthesized AgNPs also exhibited moderate antibacterial activity (8.14–10.22 mm inhibition zones) against two Gram-negative pathogenic bacteria with promising radical scavenging potential. Overall, the results highlight the effectiveness of the AgNPs for use in antibacterial wound dressing materials and other biomedical applications for the treatment of critical diseases such as cancer.

Valerianaceae, the sub-family of Caprifoliaceae, contains more than 300 species of annual and perennial herbs, worldwide distributed. Several species are used for their biological properties while some are used as food. Species from the genus Valeriana have been used for their antispasmodic, relaxing, and sedative properties, which have been mainly attributed to the presence of valepotriates, borneol derivatives, and isovalerenic acid. Among this genus, the most common and employed species is Valerianaofficinalis. Although valerian has been traditionally used as a mild sedative, research results are still controversial regarding the role of the different active compounds, the herbal preparations, and the dosage used. The present review is designed to summarize and critically describe the current knowledge on the different plant species belonging to Valerianaceae, their phytochemicals, their uses in the treatment of different diseases with particular emphasis on the effects on the central nervous system. The available information on this sub-family was collected from scientific databases up until year 2020. The following electronic databases were used: PubMed, Scopus, Sci Finder, Web of Science, Science Direct, NCBI, and Google Scholar. In this study, we have introduced newly synthesized substituted benzothiazole based berberine derivatives that have been analyzed for their in vitro and in silico biological properties. The activity towards various kinds of influenza virus strains by employing the cytopathic effect (CPE) and sulforhodamine B (SRB) assay. Several berberine–benzothiazole derivatives (BBDs), such as BBD1, BBD3, BBD4, BBD5, BBD7, and BBD11, demonstrated interesting anti-influenza virus activity on influenza A viruses (A/PR/8/34, A/Vic/3/75) and influenza B viral (B/Lee/40, and B/Maryland/1/59) strain, respectively. Furthermore, by testing neuraminidase activity (NA) with the neuraminidase assay kit, it was identified that BBD7 has potent neuraminidase activity. The molecular docking analysis further suggests that the BBD1–BBD14 compounds’ antiviral activity may be because of interaction with residues of NA, and the same as in oseltamivir.

In recent decades, interest in the Cordyceps genus has amplified due to its immunostimulatory potential. Cordyceps species, its extracts, and bioactive constituents have been related with cytokine production such as interleukin (IL)-1ß, IL-2, IL-6, IL-8, IL-10, IL-12, and tumor necrosis factor (TNF)-a, phagocytosis stimulation of immune cells, nitric oxide production by increasing inducible nitric oxide synthase activity, and stimulation of inflammatory response via mitogen-activated protein kinase pathway. Other pharmacological activities like antioxidant, anti-cancer, antihyperlipidemic, anti-diabetic, anti-fatigue, anti-aging, hypocholesterolemic, hypotensive, vasorelaxation, anti-depressant, aphrodisiac, and kidney protection, has been reported in pre-clinical studies. These biological activities are correlated with the bioactive compounds present in Cordyceps including nucleosides, sterols, flavonoids, cyclic peptides, phenolic, bioxanthracenes, polyketides, and alkaloids, being the cyclic peptides compounds the most studied. An organized review of the existing literature was executed by surveying several databanks like PubMed, Scopus, etc. using keywords like Cordyceps, cordycepin, immune system, immunostimulation, immunomodulatory, pharmacology, anti-cancer, anti-viral, clinical trials, ethnomedicine, pharmacology, phytochemical analysis, and different species names. This review collects and analyzes state-of-the-art about the properties of Cordyceps species along with ethnopharmacological properties, application in food, chemical compounds, extraction of bioactive compounds, and various pharmacological properties with a special focus on the stimulatory properties of immunity. The evaluation and confirmation of healing properties of several plant species of genus Terminalia based on their traditional uses and the clinical claims are of utmost importance. Genus Terminalia has received more attention to assess and validate the therapeutic potential and clinical approval due to its immense folk medicinal and traditional applications. Various species of Terminalia genus are used in the form of herbal medicine and formulations, in treatment of diseases, including headache, fever, pneumonia, flu, geriatric, cancer, to improve memory, abdominal and back pain, cough and cold, conjunctivitis, diarrhea, heart disorder, leprosy, sexually transmitted diseases, and urinary tract disorders. These are reported to possess numerous biological properties, counting: antibacterial, antifungal, antiinflammatory, antiviral, antiretroviral, antioxidant, and antipa7rasitic. This current research review aims to update the detailed biological activities, pre-clinical and clinical studies of various extracts and secondary metabolites from several plant species under the genus Terminalia, along with information on the traditional uses and chemical composition to develop a promising strategy for their potential applications in the form of medicine or use in modern drug formulations for treating diseases like pneumonia, flu, and other types of viral infections or controlling human contagions.

Grape pomace, a most abundant and renewable wine industry waste product was utilized as a suitable reducing, capping, and stabilizing biomolecules for green synthesis of silver nanoparticles (AgNPs). The physicochemical properties of biosynthesized grape pomace extract (GPE)-AgNPs were duly appraised via UV&ndash, Visible spectroscopy, X-ray diffractometer (XRD), Fourier-transform infrared spectroscopy (FTIR), and transmission electron microscopy. The analytical studies revealed that the GPE-AgNPs were well formed and stable in nature. The functional groups of organic molecules of GPE are present on the surface of AgNPs with average NPs diameter in the range of 20&ndash, 35 nm. GPE-AgNPs exhibited significant free radical scavenging activity mainly DPPH radical (IC50, 50.0 &plusmn, 2.25 &mu, g/mL) and ABTS radical (IC50, 38.46 &plusmn, 1.14 &mu, g/mL). Additionally, the synthesized AgNPs showed noticeable inhibition of carbohydrate hydrolyzing enzymes mainly, &alpha, amylase (IC50, 60.2 &plusmn, 2.15 &mu, g/mL) and &alpha, glucosidase (IC50, 62.5 &plusmn, 2.75 &mu, g/mL). The GPE fabricated AgNPs showed noteworthy antibacterial potential against infectious bacteria viz., Escherichia coli and Staphylococcus aureus. The reaction mechanism of antibacterial activity was studied by measuring the bacterial cell membrane breakage and cytoplasmic contents, mainly, nucleic acid, proteins, and reducing sugar. Therefore, this research attempt illustrated the potential of GPE as a novel source intended for the biosynthesis of AgNPs that may open up new horizons in the field of nanomedicine.

Background Berberine, a quaternary ammonium salt from the protoberberine group of benzylisoquinoline alkaloids has drawn high attention for its several biological potencies. Objective To furnish new rationalized derivatives based on berberine core which can deliver promising antioxidant and cytotoxic activities. Method The N-Mannich base of an isoquinoline alkaloid, berberine, bearing substituted benzothiazole moieties was obtained. Novel synthesized analogues were in vitro screened for antioxidant efficacy toward 2,2-diphenyl- 1-picrylhydrazyl (DPPH) and 2,2'-azino-bis(3-ethylbenzothiazoline-6-sulphonic acid) (ABTS) free radicals and in vitro cytotoxicity towards cervical cancer cell lines (HeLa and CaSki), an ovarian cancer cell line (SK-OV-3) and human renal cancer cell line (Caki-2). Cytotoxicity of the compounds toward normal cell lines was examined using the Madin-Darby canine kidney (MDCK) non-cancer cell line. Results Analogues bearing a methoxy functional group (5e), acid functionality (5c), and a cyano group (5m) showed remarkable radical scavenging potential in DPPH and ABTS bioassays. Potent cytotoxicity exhibited by berberine against the HeLa cell line was attributable to the presence of a 2-aminobenzothaizole moiety (5a) and its 6-chloro congener (5g) on the berberine core, and the 6-cyano group (5m) on the benzothiazole ring revealed strong sensitivity for the CaSki cell line, whereas subjected scaffolds demonstrated diminished activity against the SK-OV-3 cell line. In addition, the compound with a 2-aminobenzothaizole moiety (5a), compound with methoxy functional group (5e) and compound with cyano group appeared with the most significant cytotoxicity effect in Caki-2 cell line. Their structures have been elucidated by FT-IR, 1H NMR, 13C NMR, and elemental analyses (CHN) essential research. Conclusion N-Mannich bases of berberine were efficiently generated utilizing pharmacologically diverse substituted 2-aminobenzothiazole entities and final compounds were found remarkably active in antioxidant and cytotoxic assay. Hence, such types of compounds can be further studied or rationalized in future drug discovery studies.

Cherry blossom (Prunus yedoensis) petals are used as ingredients in many cosmetics. However, despite their use in numerous products, the exact function of cherry blossom petals in cosmetics is unclear. Therefore, we need evidence-based studies to support the labeling claims that are made in cherry blossom products in the cosmetics industry. We investigated the skin anti-aging potential of non-enzymatic softening cherry blossom extract (NES-CBE) in this study. The extract desalinated, to improve its quality such that it can be used as a functional material for the skin. The anti-wrinkle effect of NES-CBE was investigated on human keratinocytes (HaCaT cells) under solar UV (sUV) light exposure. We found that NES-CBE reduced the sUV-induced matrix metalloproteinase (MMP)-1 expression and modulated the transactivation of the activator protein (AP)-1. Furthermore, NES-CBE suppressed the phosphorylation of MEK1/2 and ERK proteins, indicating its regulation of sUV-induced MAPK signaling. Additionally, we observed NES-CBE reduced MMP-1 protein expression in a human skin equivalent model. Taken together, these results suggest that NES-CBE reduces sUV-induced MMP-1 protein expression through reducing AP-1 transactivation via regulation of the MEK1/2-ERK pathway.

This work evaluates lignocellulolytic enzymes production by a selected strain Streptomyces sp. MDS cultivated in various agricultural wastes under solid-state fermentation (SSF). Maximum activities (U/gds) of endoglucanase (132.6 ± 1.15), exoglucanase (14.6 ± 0.62), cellobiase (125.6 ± 1.75), filter paperase (19.7 ± 0.42), amylase (278.5 ± 3.53), and xylanase (342.5 ± 3.36) were produced with rice waste biomass (RWB). Operational parameters and supplementation of nitrogen sources, metal additives, and surfactants were systematically optimized with a view to maximizing enzyme production. The harvested enzyme exhibited good stability at high pH (5 to 8) and temperature (50 to 80 °C) and showed robust nature in the presence of organic solvents, surfactants, and commercial detergents. The potential of crude enzyme mixture for hydrolysis of mild alkaline pretreated RWB was demonstrated, resulting in equivalent saccharification (64.85 ± 1.11%) relative to commercial enzymes (69.41 ± 1.15%). Yeast co-culture of Saccharomyces cerevisiae and Kluyveromyces marxianus resulted in higher ethanol production (21.12 ± 0.82 g/L) and sugar consumption (88%) from enzymatic RWB hydrolysates (50 g/L) than monoculture. Finally, the leftover spent slurry from SSF effectively decolorized individual dyes and the actual textile wastewater, which increases the practical applicability of the developed bioprocess.

Microbial fuel cells (MFCs) are biocatalyzed systems which can drive electrical energy by directly converting chemical energy using microbial biocatalyst and are considered as one of the important propitious technologies for sustainable energy production. Much research on MFCs experiments is under way with great potential to become an alternative to produce clean energy from renewable waste. MFCs have been one of the most promising technologies for generating clean energy industry in the future. This article summarizes the important findings in electro-active biofilm formation and the role of exo-electrogens in electron transfer in MFCs. This study provides and brings special attention on the effects of various operating and biological parameters on the biofilm formation in MFCs. In addition, it also highlights the significance of different molecular techniques used in the microbial community analysis of electro-active biofilm. It reviews the challenges as well as the emerging opportunities required to develop MFCs at commercial level, electro-active biofilms and to understand potential application of microbiological niches are also depicted. Thus, this review is believed to widen the efforts towards the development of electro-active biofilm and will provide the research directions to overcome energy and environmental challenges.

Process byproducts from the fruit industry may represent a cheap and reliable source of green reducing agents to be used in current bio-nanosynthesis. This study reports the use of orange (Citrus × clementina) peel aqueous extract (OPE) for one-pot green synthesis of silver nanoparticles (AgNPs) with high effectiveness against various microbial pathogens as well as rat glial tumor C6 cells. The effects of various operational parameters on the synthesis of AgNPs were systematically investigated. The morphology, particle size, and properties of synthesized AgNPs were characterized using UV–visible spectroscopy, x-ray diffraction, x-ray photoelectron spectroscopy, field emission scanning electron microscopy, energy-dispersive x-ray spectroscopy, and Fourier transform infrared spectroscopy. High-resolution transmission electron microscopy shows that the nanoparticles are mostly spherical in shape and monodispersed, with an average particle size of 15–20 nm. Notably, the OPE-synthesized AgNPs were stable up to 6 months without change in their properties. Low doses of OPE-AgNPs inhibited the growth of human pathogens Escherichia coli, Bacillus cereus, and Staphylococcus aureus. The minimum inhibitory concentration and minimum bactericidal concentration of AgNPs against selected pathogenic bacteria were determined. OPE-AgNPs exhibited strong antioxidant activity in terms of ABTS (2,2′-azino-bis (3-ethylbenzothiazoline-6-sulfonic acid)) radical scavenging (IC50 49.6 μg/mL) and DPPH (1,1-diphenyl-2-picrylhydrazyl) radical scavenging (IC50 63.4 μg/mL). OPE-AgNPs showed dose-dependent response against rat glial tumor C6 cells (LD50 60 μg/mL) showing a promising potential as anticancer agents. Overall, the current investigation highlighted a cheap green technology route to synthesize AgNPs using OPE byproducts and could potentially be utilized in biomedical, cosmetic, and pharmaceutical industry.

The current investigation highlighted a novel cost-effective green synthesis of silver nanoparticles (AgNPs) using Argyreia nervosa leaves extract (ANE) as a potential reducing and capping agent. Surface plasmon resonance confirmed the formation of AgNPs with maximum absorbance at λ max = 435 nm. FTIR revealed the involvement of biological macromolecules of ANE in the synthesis and stabilization of AgNPs. HRTEM images showed that the size of the spherical AgNPs ranged between 5 and 40 nm with average particle size of about 15 nm. The ANE-AgNPs showed inhibition activity against carbohydrate digestive enzymes α-amylase and α-glucosidase, with EC50 of 55.5 and 51.7 µg/mL, respectively, indicating its antidiabetic potential. The in vitro antioxidant activity of ANE-AgNPs was evaluated in terms of ABTS (2,2′-azino-bis(3-ethylbenzothiazoline-6-sulphonic acid) and DPPH (1,1-diphenyl-2-picrylhydrazyl) free radicals scavenging assays with IC50 value of 44.3 and 55.9 µg/mL, respectively. The AgNPs displayed strong antibacterial activity against foodborne bacteria with zone of inhibition 16.0 and 12.5 mm for Escherichia coli and Staphylococcus aureus, respectively, and also exhibited strong synergistic antibacterial activity together with standard antibiotics. The biological activity in terms of antioxidant, antidiabetic and antibacterial potential could be useful in various bio-applications such as cosmetics, food, and biomedical industry.

Sesamol is a phenol derivative of sesame oil and a potent anti-oxidant, anti-inflammatory, anti-hepatotoxic, and anti-aging compound. We investigated the effects of sesamol on the molecular mechanisms of adipogenesis in 3T3-L1 preadipocytes. The intracellular lipid accumulation accompanied by increased extracellular release of free glycerol was decreased during differentiation on treating 3T3-L1 with sesamol. Sesamol treatment on 3T3-L1 inhibited adipogenic differentiation by down-regulating adipogenesis-related factors (C/EBPα, PPARγ, and SREBP-1). Lipid accumulation was repressed by decreasing fatty acid synthase and by up-regulating lipolysis-response genes (HSL and LPL). The molecular mechanisms of sesamol-induced inhibition in adipogenesis were mediated by increased levels of phosphorylated adenosine monophosphate-activated protein kinase and its substrate acetyl-CoA carboxylase. Sesamol treatment, in turn, modulated the different members of the mitogenactivated protein kinase family by suppressing phosphorylation of ERK 1/2 and JNK and by increasing the phosphorylation of p38. In summary, sesamol inhibits adipogenic differentiation by reducing phosphorylation levels of ERK 1/2 and JNK while inducing lipolysis by activating p38 and AMPK. Our results demonstrate that the molecular mechanisms of in vitro anti-obesity effects of sesamol are due to the combined effects of preventing both lipid accumulation and adipogenesis.

Cancer accounts for a number of deaths each year. Consequently, prevention of this deadly disease is more challenging and hence the invention of new anticancer agents is of utmost importance. The current review elaborates the importance of indole designs as patented in the form of anticancer druglike molecules targeting different cites of biological arena. Specific attention was given to kinases such as platelet-derived growth factor receptor, vascular endothelial growth factor receptor and fibroblast growth factor receptor, Bruton's tyrosine kinase, anaplastic lymphoma kinase, Janus kinase, cyclin-dependent kinase aurora kinases A, B and C, checkpoint kinases, protein kinase R, Pim kinases, phosphoinositide 3- kinase, altered proteins kinases, polo-like kinase and many more. Moreover, the article summarizes the mode of action through the particular functions of kinases and the inhibitory potential of indole derivatives toward specific kinase. Certain patents gathered in the existing review article suggest that indole core can be a versatile foundation to discover drug-like kinase inhibitor molecules and modification of substituents existing on the indole moiety may have important impact on the pharmacokinetic and pharmacodynamics aspects of the resultant scaffolds. The information presented here would gather a great deal of interest to identify the new molecular designs bearing indole nucleus presenting novel anticancer drugs with a wide variety of biological targets involved in cancer pathology focusing on the inhibition of tyrosine kinases, serine/threonine-specific protein kinases, cyclin-dependent kinases, lipid kinases and altered protein kinases.

Medicinal herbs are one of the imperative sources of drugs all over the world. Star anise (Illicium verum), an evergreen, medium-sized tree with star-shaped fruit, is an important herb with wide distribution throughout southwestern parts of the Asian continent. Besides its use as spice in culinary, star anise is one of the vital ingredients of the Chinese medicinal herbs and is widely known for its antiviral effects. It is also the source of the precursor molecule, shikimic acid, which is used in the manufacture of oseltamivir (Tamiflu®), an antiviral medication for influenza A and influenza B. Besides, several other molecules with numerous biological benefits including the antiviral effects have been reported from the same plant. Except the antiviral potential, star anise possesses a number of other potentials such as antioxidant, antimicrobial, antifungal, anthelmintic, insecticidal, secretolytic, antinociceptive, anti-inflammatory, gastroprotective, sedative properties, expectorant and spasmolytic, and estrogenic effects. This review aimed to integrate the information on the customary attributes of the plant star anise with a specific prominence on its antiviral properties and the phytochemical constituents along with its clinical aptness.

Cacao (Theobroma cacao) has a significant polyphenol content and has been reported to elicit anti-obesity effects. Previous studies have focused on the properties of cacao extract and procyanidins, while the potential mechanisms have not been fully elucidated. Here, we investigated the inhibitory effects of procyanidin metabolites on adipogenic cocktail-induced adipogenesis and lipogenesis in 3T3-L1 preadipocytes. It was observed that 5-(3′,4′-dihydroxyphenyl)-γ-valerolactone (DHPV), a major procyanidin metabolite, exhibited the greatest inhibitory effects on adipogenesis and lipogenesis. DHPV dose-dependently reduced the expression levels of proteins involved in adipogenesis including peroxisome proliferator-activated receptor γ (PPAR γ) and CCAT/enhancer-binding protein α (C/EBP α), as well as lipogenesis-related factors such as fatty acid synthase and acetyl-CoA carboxylase. These inhibitory effects were primarily due to G1 phase arrest and the suppression of cell proliferation during mitotic clonal expansion, the early stage of adipogenesis. In an extensive kinase array, DHPV directly suppressed activation of the CDK2/cyclin O complex, and inhibited the phosphorylation of C/EBP β, which is responsible for the induction of PPAR γ and C/EBP α. Taken together, these findings suggest that DHPV is a highly biologically active compound with potential anti-obesity effects and works by inhibiting the intracellular lipid content and cell differentiation.

Two series of sulfonylpiperazines linked [1,3]dioxolo[4,5-g]chromenones were synthesized featuring phenyl (7a-k) and chalcone (12a-k) bridge representing flavones or homoisoflavonoids core. New molecules are synthesized utilizing aldol condensation to inspect as antioxidants against DPPH and ABTS + and antiproliferative agents toward selected human cancer cell lines. Cytotoxicity of new compounds was confirmed using SRB assay against non-cancer MDCK cell line. The results concluded that both individual structures of 7 and 12 were vital for modulating pharmacological potencies and presence of different electron withdrawing and electron donating functional group(s) on the phenylsulfonyl entity yielded varied biological effects. Substituent h (OCF3) and j, k (OCH3) were found to play a crucial role scavenging DPPH and ABTS + as well as inhibiting cancer cell lines SK-OV-3 and HT-29. Moreover, molecules bearing halogen atom(s) such as substituent b-g expressed excellent inhibitory potential against HeLa and A-549 cancerous cell lines. Bioassay data displayed some interesting structure-activity relationships which are discussed in this paper. The results justified that tested derivatives are promising antioxidants and cytotoxic agents and warrant further structural optimization and bioassay studies. Spectroscopic techniques such as FT-IR, 1H NMR, 13C NMR and elemental analysis (CHN) were carried out to confirm the final structures.

Plants belonging to the subfamily Bombacoideae (family Malvaceae) consist of about 304 species, many of them having high economical and medicinal properties. In the past, this plant group was put under Bombacaceae; however, modern molecular and phytochemical findings supported the group as a subfamily of Malvaceae. A detailed search on the number of publications related to the Bombacoideae subfamily was carried out in databases like PubMed and Science Direct using various keywords. Most of the plants in the group are perennial tall trees usually with swollen tree trunks, brightly colored flowers, and large branches. Various plant parts ranging from leaves to seeds to stems of several species are also used as food and fibers in many countries. Members of Bombacoides are used as ornamentals and economic utilities, various plants are used in traditional medication systems for their anti-inflammatory, astringent, stimulant, antipyretic, microbial, analgesic, and diuretic effects. Several phytochemicals, both polar and non-polar compounds, have been detected in this plant group supporting evidence of their medicinal and nutritional uses. The present review provides comprehensive taxonomic, ethno-pharmacological, economic, food and phytochemical properties of the subfamily Bombacoideae.

There is much interest in the fishery industry in developing freshness indicator which can reflect the storage history of products and their quality. Freshness is one of the main considerations for the fishery industry and consumption. In this study, a freshness indicator of fish products, especially skate (Raja kenojei), was developed. The release of amines especially ammonia from decomposing fish is concentrated in headspace and they can be detected by pH-sensitive sensor unit. The sensor unit was organized with polymer matrix solution, which contains bromothymol blue-phenol red as pHsensitive dye to monitor visible color changes from yellow to purple. We investigated the sensor unit formulation, which responds to the ammonia formation in fish products. The color change of the sensor unit was evaluated as well as the selection of the proper weight of fish products and packaging material. Results will be useful to establish a basis for development of a freshness indicator and to determine the suitability of freshness indicator formulations for intelligent food packaging applications.

A gas freshness indicator consisting of the pH-sensitive dye bromothymol blue-phenol red (BTB-PR) was investigated for visible color changes of fish product quality based on the quantity of volatile amines. Chromaticity values of the gas indicator accurately tracked an increase in the ammonia content in the packaging headspace of fish products, especially skate (Raja kenojei). Gradual color changes of the gas indicator response correlated with the quality of fish including pH values during storage. Ammonia content was evaluated during the fish storage using headspace solid-phase microextraction analysis with gas chromatography-flame ionization detection. The validation results suggested that good linearity (R²=0.99) and recovery of 84.00%. The limits of detection and quantification were 8.70 and 26.30 mg/L, respectively. A pH values of skate and chromaticity of gas indicator were measured and the correlation with ammonia content was evaluated. Results of this study could prove the application of gas indicator as intelligent package for improvement of food safety.

Equisetum arvense is well known to hold numerous bioactive phytochemicals. In biosynthesis of nanoparticles (NPs), the bioactive compounds existing in natural materials like medicinal fern act as reducing and capping elements and this NPs synthesis process do not comprise of any toxic elements making them advantageous from other NPs synthesis process. After collection, identification and extraction of Equisetum arvense (Ea) aqueous extract, the biosynthesis of AgNPs was achieved followed by its characterization and multi-biopotential activity studies. The UV–visible spectroscopy, confirmed the biosynthesis of Ea-AgNPs. X-ray diffraction configurations (XRD) identified the crystalline nature of the NPs. The Elemental composition of the NPs was elucidated by the energy dispersive X-ray spectroscopy (EDX), and the scanning electron microscopy (SEM) revealed the structure of Ea-AgNPs. Bioactive compounds existing in Ea-extract accounting for Ag + ion reduction, capping and stabilization of NPs was detected by Fourier transform infrared spectroscopy (FTIR). The Dynamic Light Scattering (DLS) and the zeta potential was carried out to know the size and charge of Ea-AgNPs. The Ea-AgNPs exhibited high antidiabetic effect in terms of α-glucosidase inhibition, high cytotoxic effect against HepG2 cell lines along with antibacterial and antioxidant effect. This study reports biosynthesis of Ea-AgNPs using aqueous extract of Ea, its substantial anticancer, antidiabetic, antioxidant and antibacterial effects, which could be advantageous to pharmaceutical industries in the controlling of various diseases including diabetes, cancer, and antibacterial related diseases.

A potential natural melanogenesis inhibitor was discovered in the form of a sesquiterpene isolated from the flowers of Inula britannica, specifically 6-O-isobutyrylbritannilactone (IBL). We evaluated the antimelanogenesis effects of IBL on B16F10 melanocytes and zebrafish embryos. As a result, we found that 3-isobutyl-1-methylxanthine (IBMX)-induced melanin production was reduced in a dose-dependent manner in B16F10 cells by IBL. We also analyzed B16F10 cells that were and were not treated with IBMX, investigating the melanin concentration, tyrosinase activity, mRNA levels. We also studied the protein expressions of microphthalmia-associated transcription factor (MITF), tyrosinase, and tyrosinase-related proteins (TRP1, and TRP2). Furthermore, we found that melanin synthesis and tyrosinase expression were also inhibited by IBL through the modulation of the following signaling pathways: ERK, phosphoinositide 3-kinase (PI3K)/AKT, and CREB. In addition, we studied antimelanogenic activity using zebrafish embryos and found that the embryos had significantly reduced pigmentation in the IBL-treated specimens compared to the untreated controls.

2&alpha, Hydroxyeudesma-4,11(13)-dien-8&beta, 12-olide (HEDO), a eudesmane-type sesquiterpene lactone belonging to large group of plant terpenoids isolated from Inula britannica, displays cytotoxic activity against diffuse large B cell lymphoma cells in vitro. However, the molecular mechanism of the anticancer effect remains unclear. In this study, we showed that HEDO inhibits cell growth by inducing apoptosis in lymphoma cell lines through its antiproliferative activity. HEDO increases the depolarization of mitochondrial membrane potential and upregulated intracellular reactive oxygen species (ROS). Furthermore, we examined the cell cycle effect, and our results provided evidence that the arrest of the cell cycle at the SubG0/G1 phase plays an important role in the ability of HEDO to inhibit cell growth in Ontario Cancer Institute (OCI)-LY3 lymphoma cells by preventing nuclear factor-kappa B (NF-&kappa, B) signaling. In addition, HEDO induced apoptosis by instigating the activation of Bcl-2-associated X (BAX) and cleaved caspase-3, decreasing B-cell lymphoma 2 (BCL2), B-cell lymphoma-extra large (BCL-XL), and procaspase 3 expression levels. Based on these findings, we suggest that HEDO has potential as an anticancer drug of lymphoma by inducing ROS-dependent accumulation of SubG0/G1 arrest and apoptosis in OCI-LY3 cells.

In these studies, we analyzed substituted piperazine based berberine analogs conjugated through a pentyloxy side chain for their in vitro and in silico biological effects. All the final analogs were screened for their in vitro antiviral action against a collection of different influenza virus strains using the CPE assay and SRB assay. Moreover, their cytotoxicity towards non-cancer cell lines was examined employing Madin–Darby canine kidney (MDCK) cell lines. The anti-influenza activities of berberine-piperazine derivatives (BPD) were evaluated in the range from 35.16 μg/mL to 90.25 μg/mL of the IC50s along with cytotoxicity level which was observed in the range 44.8 μg/mL to 3890.6 μg/mL of CC50s towards MDCK cells. In an effort to know the mechanism of action of BPD1-BPD23, results of Neuraminidase inhibition assay and Molecular docking studies carried out against neuraminidase as the target enzyme revealed that titled compounds are potential neuraminidase inhibitors that merge to the active site of neuraminidase, with moderate to high binding energy.

Microorganism live inside the human body and thus the human are inimitable reservoir for diverse and vibrant group of microbes. These microbial species primarily modulates the host internal environment for the edification in the host health. This spectacular symbiotic relationship is under extensive research in recent years. The significance of these organisms in various metabolic activities inside the body is very crucial. Therefore any alterations in their diversity may results in several fatal diseases and disorders. The efficacy of the classical therapy is consecutively demising with the emergence of tolerance as well as resistance to these therapeutic drugs. Henceforth, a simple, cheaper, receptive and intrinsic means for health benefactor is a vital compulsion of the present era which could be achieved by the means of probiotics. Research have proved that probiotics supplementation against various enteric pathogens has shown promising results with their exceptional ability to compete with pathogenic microbes for various purposes by initiating the activation of specific genes of localized host cells. In the current chapter, the beneficial effect of probiotics consumption in human health and medical sectors has been discussed.

This study focuses on development of calcium peroxide (CaO2) pretreatment that removes major part of lignin but retaining most of sugar components of kenaf core powder (KCP) biomass. In chemical pretreatment, usually higher loss of biomass occurs which was less during this pretreatment strategy. Supplementation of inorganic salts; manganese sulfate (MnSO4) and cobalt chloride (COCl2) in CaO2 pretreatment resulted in maximum delignification of KCP relative to individual CaO2 pretreatment. Maximum glucose yield (98%) and hydrolysis yield (80.5%) was achieved after enzymatic hydrolysis (30 FPU/g of KCP) under optimized conditions. Analytical results proved effective lignin removal and significant destruction of KCP with this pretreatment strategy. Finally, utilization of KCP enzymatic hydrolysates by developed strain Klebsiella pneumoniae KMK05 resulted in maximum 2,3-butanediol (BDO) production (10.42 g/L) and BDO titer (0.385 g/g of sugar). BDO titer achieved with KCP derived sugars were found comparable with the mixture of standard sugars which is notable.

The effects of sesame oil (SO) on lipogenic gene expression and lipid metabolism were observed. One percent of SO was added into the high-fat diet (HFD) used to feed C57BL/6 male mice for 8 weeks. SO significantly lowered adipose tissue mass than that of the high-fat control group. In addition, serum and hepatic lipid contents were markedly lower than those of the high-fat control group. Especially, the ratio of HDL/LDL was much higher than that of the HFD group. The gene expression of key lipogenic enzymes, Ppar-γ, Srebp-1c, Scd-1, Fas, Acc, Lpl, and malic enzyme, was significantly downregulated in the liver and adipose tissue of mice fed sesame oil. This study suggests that high proportion of oleic acid in SO might be strongly associated with lowering serum and hepatic lipid profile and suppressing lipogenic gene expression.

Mercury is a well-known environmental pollutant that can cause nephropathic diseases, including acute kidney injury (AKI). Although quercetin (QC), a natural flavonoid, has been reported to have medicinal properties, its potential protective effects against mercury-induced AKI have not been evaluated. In this study, the protective effect of QC against mercury-induced AKI was investigated using biochemical parameters, new protein-based urinary biomarkers, and a histopathological approach. A 250 mg/kg dose of QC was administered orally to Sprague-Dawley male rats for 3 days before administration of mercury chloride (HgCl2). All animals were sacrificed at 24 h after HgCl2 treatment, and biomarkers associated with nephrotoxicity were measured. Our data showed that QC absolutely prevented HgCl2-induced AKI, as indicated by biochemical parameters such as blood urea nitrogen (BUN) and serum creatinine (sCr). In particular, QC markedly decreased the accumulation of Hg in the kidney. Urinary excretion of protein-based biomarkers, including clusterin, kidney injury molecule-1 (KIM-1), neutrophil gelatinase-associated lipocalin (NGAL), monocyte chemoattractant protein-1 (MCP-1), tissue inhibitor of metalloproteinases 1 (TIMP-1), and vascular endothelial growth factor (VEGF) in response to HgCl2 administration were significantly decreased by QC pretreatment relative to that in the HgCl2-treated group. Furthermore, urinary excretion of metallothionein and Hg were significantly elevated by QC pretreatment. Histopathological examination indicated that QC protected against HgCl2-induced proximal tubular damage in the kidney. A TUNEL assay indicated that QC pretreatment significantly reduced apoptotic cell death in the kidney. The administration of QC provided significant protective effects against mercury-induced AKI.

Mycobacterium tuberculosis is the causative agent of tuberculosis (TB), a major health issue of the present era. The bacterium inhabits the host macrophage and other immune cells where it modulates the lysosome trafficking protein, hinders the formation of phagolysosome, and blocks the TNF receptor-dependent apoptosis of host macrophage/monocytes. Other limitations such as resistance to and low bioavailability and bio-distribution of conventional drugs aid to their high virulence and human mortality. This review highlights the use of nanotechnology-based approaches for drug formulation and delivery which could open new avenues to limit the pathogenicity of tuberculosis. Moreover phytochemicals, such as alkaloids, phenols, saponins, steroids, tannins, and terpenoids, extracted from terrestrial plants and mangroves seem promising against M. tuberculosis through different molecular mechanisms. Further understanding of the genomics and proteomics of this pathogenic microbe could also help overcome various research gaps in the path of developing a suitable therapy against tuberculosis.

The progression of life in all forms is not only dependent on agricultural and food security but also on the soil characteristics. The dynamic nature of soil is a direct manifestation of soil microbes, bio-mineralization, and synergistic co-evolution with plants. With the increase in world’s population the demand for agriculture yield has increased tremendously and thereby leading to large scale production of chemical fertilizers. Since the use of fertilizers and pesticides in the agricultural fields have caused degradation of soil quality and fertility, thus the expansion of agricultural land with fertile soil is near impossible, hence researchers and scientists have sifted their attention for a safer and productive means of agricultural practices. Plant growth promoting rhizobacteria (PGPR) has been functioning as a co-evolution between plants and microbes showing antagonistic and synergistic interactions with microorganisms and the soil. Microbial revitalization using plant growth promoters had been achieved through direct and indirect approaches like bio-fertilization, invigorating root growth, rhizoremediation, disease resistance etc. Although, there are a wide variety of PGPR and its allies, their role and usages for sustainable agriculture remains controversial and restricted. There is also variability in the performance of PGPR that may be due to various environmental factors that might affect their growth and proliferation in the plants. These gaps and limitations can be addressed through use of modern approaches and techniques such as nano-encapsulation and micro-encapsulation along with exploring multidisciplinary research that combines applications in biotechnology, nanotechnology, agro biotechnology, chemical engineering and material science and bringing together different ecological and functional biological approaches to provide new formulations and opportunities with immense potential.

A number of wild crops remain unexplored in this world and among them some have excellent medicinal and nutritional properties. India is a harbor of biodiversity in general and phytodiversity in particular. The plant diversity is distributed from the Western Ghats to Eastern Ghats, along with the North-Eastern region and from the Greater Himalayas to the plain of Ganga. Among these distributed floral regions of the country, the Eastern Ghats are important due to their rich floral diversity. The forests of Odisha form a major part of Eastern Ghats in general and the Similipal Biosphere Reserve (SBR) in particular. The SBR is inhabited by many tribal communities. The food and medicinal habits of these communities are not fully explored even today. They are dependent on the forests of SBR for their food and medicine. Among their collections from forests, root and tuberous plants play a significant role. The tribal communities of SBR use about 89 types of tuberous plants for various purposes. Dioscorea is one such tuber, having maximum use among the tribal of SBR. However, less documentation and no specific reports are available on the food and medicinal values of the species available in this part of the World. Dioscorea species, popularly known as Yam worldwide and as Ban Aalu in Odisha, India, is a prime staple medicinal-food substitute for the majority of rural and tribal people of the state of India. Of the 13 Dioscorea species available in SBR, 10 species are known to be bitter in taste and unpalatable when taken raw. Since less documentation is available on the Dioscorea species of SBR and their traditional uses, the present study was focused on the ethnobotany, nutritional and pharmacological values of these species along its nutraceutical importance.

Gradual accumulation of the fishy-odor compound trimethylamine (TMA) from bacterial reduction of trimethylamine oxide (TMAO) is one of the characteristic chemical changes attributed to fish spoilage. Changes in TMA values were correlated with sensory testing results, storage temperature, storage time, and viable bacteria counts. TMA contents were determined under different storage conditions. Headspace solid-phase micro extraction analysis with gas chromatography using flame ionization detection was used with different storage times and temperatures. Mackerel samples were inoculated with Pseudomonas fragi to monitor changes in the TMA content and pH. The TMA content increased proportionally with time during storage, and pH values and the microbiological quality were evaluated to analyze correlations with the TMA content in fish products. The TMA content increased with an increasing number of P. fragi with good linearity (R2=0.997). Basic data are provided for developing a freshness indicator for fish spoilage.

A freshness indicator consisting of a polymer matrix solution containing the pH-sensitive dye bromocresol green was investigated for monitoring visible color changes of fish spoilage volatile compounds based on the quantity of volatile amines. Mackerel fillets were inoculated with Pseudomonas fragi and stored in incubators at 5, 12, 18, and 25°C. Chromaticity values of the freshness indicator accurately tracked an increase in the trimethylamine content in the packaging headspace. Gradual color changes of the freshness indicator response correlated with the quality of fish, including growth of P. fragi and pH values during storage. Results will be useful to establish a basis for development of a freshness indicator and to determine the suitability of freshness indicator formulations for intelligent food packaging applications.

Microtubule targeting chemicals are considered excellent antitumor drugs through their binding to tubulin, which affects the instability of microtubules resulting in arrest of cancer cells. The present study was designed to investigate the antitumor effects of viriditoxin (VDT) against human prostate cancer cells. VDT, isolated from Paecilomyces variotii fungus, which was derived from the jellyfish Nemopilema nomurai, offers a new approach for controlling resistant bacterial infections by blocking bacterial cell division proteins. VDT produced dose-dependent cytotoxicity against human prostate cancer cells. Treatment with VDT promoted both apoptosis and autophagy in LNCaP cells. Annexin V/FITC staining indicated that apoptosis occurred in VDT-treated LNCaP cells. DAPI staining revealed morphological changes in the cell nuclei indicative of mitotic catastrophe in LNCaP cells. VDT caused cell growth inhibition via G2/M phase arrest. Moreover, VDT also increased autophagic cell death in LNCaP cells by induction of several autophagy-related proteins such as LC3 II, Atg5, Atg7 and beclin-1 protein, which are essential for autophagy induction. These results were also confirmed by acridine orange staining. This study indicates that VDT could potentially be effective against prostate cancer by promoting multiple modes of growth arrest and cell death coupled with apoptosis and autophagy.

Chrysin-based sulfonylpiperazines 7a-k were synthesized and investigated for their in vitro free radical scavenging potential as well as cytotoxic efficacies against selected cancer cell lines. Cytotoxicity of the new compounds toward noncancer cells was confirmed using the SRB assay against Madin-Darby Canine Kidney cells. Reaction of piperazine with different substituted benzenesulfonyl chlorides in triethylamine furnished sulfonylpiperazines (3a-k), which were then allowed to react with 7-(4-bromobutoxy)-5-hydroxy-2-phenyl-4H-chromen-4-one (6) prepared reacting chrysin with 1,4-dibromobutane to give the final derivatives 7a-k. The results concluded that chrysin-sulfonylpiperazines exerted better antioxidant and anticancer efficacies than previously studied chrysin-piperazine precursors. For example, compounds 7h, 7j, and 7k with 4-OCF3 , 4-OCH3 , and 2,4-diOCH3 groups exhibited the best antioxidant potential against 2,2-diphenyl-1-picrylhydrazyl (DPPH) and 2,2'-azino-bis-3-ethylbenzothiazoline-6-sulfonic acid (ABTS) radicals. Moreover, halogenated analogues (7b, 7c, 7g, and 7h) demonstrated promising anticancer potential against SK-OV3, HeLa, and HT-29 cell lines, whereas those bearing a methoxy functional group (7j and 7k) had beneficial effects against the cell lines A-549 and HT-29. Thus, it can be confirmed from the bioassay results that the overall structural design as well as proper substitution is crucial to deliver the anticipated biological effects. Spectroscopic techniques such as FT-IR, 1 H NMR, 13 C NMR, mass and elemental analysis (CHN) were carried out to confirm the final structures.

This study was conducted to define the antioxidant properties and cytoprotective mechanisms of sea buckthorn fruit extract (SFE) against cellular oxidative stress in mouse embryonic fibroblast (MEF) cells. Cell viability of MEF cells damaged by H2O2 was significantly increased by addition of SFE in a concentration dependent manner. Cytoprotective effect of SFE against oxidative damage was observed to be co-related with regulation of cell cycle progression. Induction of cell cycle arrest in G2/ M checkpoint was mediated by oxidative stress, but significantly reduced by treatment of MEF cells with SFE. Analysis of key regulatory proteins involved in G2/M arrest showed that SFE treatment leads to down-regulation of both phosphorylated Chk1 and cyclin B, which play important roles in cell cycle arrest of oxidatively damaged cells. Effect of SFE on apoptosis was evaluated by morphological and flow cytometric analysis. Apoptotic cell accumulation occurred by H2O2 treatment was decreased by co-treatment of MEF cells with SFE. Early apoptotic process involved in DNA fragmentation and condensation was also inhibited by additional treatment with SFE. Overall results suggest that cytoprotective effect of SFE is mediated by effective radical scavenging activity as well as altered cell cycle regulation which prevent apoptotic cell death induced by cellular oxidative stress.

Different types of fermented foods such as chongkukjang, doenjang, ganjang, gochujang and kimchi are plentifully available and widely consumed in north eastern Asian countries including Korea. Among them, kimchi is one of the most popular Korean traditional food. It is prepared by fermenting the baechu cabbage together with other vegetables and lactic acid bacteria with functional potential. Many types of ingredients are added to kimchi to enhance its taste, flavor, nutritional value, texture etc. A number of bacteria are involved in the fermentation of kimchi, but lactic acid bacteria are the dominant species in the fermentation process. The addition of other sub ingredients and formation of different by-products during fermentation eventually leads to eradication of putrefactive and pathogenic bacteria, and also increase the functionalities, nutritional and nutraceutical potential of kimchi. Kimchi possesses anti-inflammatory, antibacterial, antioxidant, anticancer, antiobesity, probiotic properties, cholesterol reduction, and antiaging properties. In the present review an attempt has been made to review the different types of fermented foods found in the Korean peninsula with detailed scientific research regarding preparation, processing, structure of the microecosystem and health benefits of kimchi.

Endophytes are an endosymbiotic group of microorganisms that colonize in plants and microbes that can be readily isolated from any microbial or plant growth medium. They act as reservoirs of novel bioactive secondary metabolites, such as alkaloids, phenolic acids, quinones, steroids, saponins, tannins, and terpenoids that serve as a potential candidate for antimicrobial, anti-insect, anticancer and many more properties. While plant sources are being extensively explored for new chemical entities for therapeutic purposes, endophytic microbes also constitute an important source for drug discovery. This review aims to comprehend the contribution and uses of endophytes as an impending source of drugs against various forms of diseases and other possible medicinal use.

This study was conducted to evaluate the cytoprotective activity of lotus (Nelumbo nucifera Gaertner) seed extract (LSE) on mouse embryonic fibroblast (MEF) cells. The DPPH free radical scavenging activities of LSE increased in a concentration dependent manner. The cells, damaged by oxidative stress, decreased their viability following increasing concentration of H2O2, but the cotreatment of ethyl acetate fraction of LSE and H2O2 resulted in an increase in cell growth, by about 25%, compared to the cells treated with H2O2. The ethyl acetate fraction of LSE inhibited the cytotoxicity induced by H2O2 in a concentration dependent manner. The treatment of the n-butanol fraction of LSE on MEF cell was also examined by analyzing the DNA content and apoptotic rate, using flow cytometry. The oxidative damage to the cells, measured by apoptotic and necrotic cell accumulation, was similar with the addition of the ethyl acetate fraction of LSE to H2O2. These results suggest that LSE inhibited the cytotoxicity which is induced by H2O2, and has a protective effect on MEF cell against oxidative stress.

During lactic acid bacteria (LAB) transit through the gastrointestinal tract, ingested microorganisms were exposed to successive stress factors, including low pH in the human stomach and in bile acid. These stress factors can be used as criteria for the selection of a viable probiotic strain. Four such strains (Lactobacillus helveticus SGU 0011, Lactobacillus pentosus SGU 0010, Streptococcus thermophilus SGU 0021 and Lactobacillus casei SGU 0020) were isolated from raw milk. When the identified LAB were exposed to synthetic gastric juice, whereas L. casei SGU 0020 and S. thermophilus SGU 0021 exhibited a 0% survival rate, L. helveticus SGU 0011 and L. pentosus SGU 0010 exhibited 60% and 95% survival rates. L. casei SGU 0020 and S. thermophilus SGU 0021 could not be examined with regard to their tolerances to artificial bile juice, as they uniformly died upon exposure. However, L. helveticus SGU 0011 and L. pentosus SGU 0010 individually survived at rates of 39% and 93%. Also, all four of these strains were confirmed to be tolerant of ten different antibiotics.
