Kluyveromyces marxianus and Saccharomyces cerevisiae yeasts were compared for the production of Carboxypeptidase Y, a serine class glycoprotein exopeptidase (CPD-Y) using Bio calorimetry and Respirometric devices. The production medium components were optimized by Response Surface Methodology-based design of experiments. Bio calorimetric investigation combined with Respirometric studies suggested that the rate of heat evolution and respiration rate was directly linked to Dry Cell Weight (DCW), enzyme activity, and the consumption rate of the substrate. The Maximum yield obtained for K. marxianus and S. cerevisiae was 14 IU/mL and 8 IU/mL respectively which were threefold higher than the shake flask experiment. Yeast growth and nature of growth predicted using calorespirometric data fitted well with measured experimental results.
