Aims: Expression of the HER2 oncogene in breast cancer is associated with resistance to treatment, and Her2 may regulate bioenergetics. Therefore, we investigated whether disruption of the electron transport chain (ETC) is a viable strategy to eliminate Her2high disease. Results: We demonstrate that Her2high cells and tumors have increased assembly of respiratory supercomplexes (SCs) and increased complex I-driven respiration in vitro and in vivo. They are also highly sensitive to MitoTam, a novel mitochondrial-targeted derivative of tamoxifen. Unlike tamoxifen, MitoTam efficiently suppresses experimental Her2high tumors without systemic toxicity. Mechanistically, MitoTam inhibits complex I-driven respiration and disrupts respiratory SCs in Her2high background in vitro and in vivo, leading to elevated reactive oxygen species production and cell death. We aimed to define the taxonomic status of 40 haemolytic and/or proteolytic strains of the genus Acinetobacter which were previously classified into five putative species termed as genomic species 14BJ (n=9), genomic species 17 (n=9), taxon 18 (n=7), taxon 19 (n=6) and taxon 20 (n=9). The strains were recovered mostly from human clinical specimens or soil and water ecosystems and were highly diverse in geographical origin and time of isolation. Comparative analysis of the rpoB and gyrB gene sequences of all strains, and the whole-genome sequences of selected strains, showed that these putative species formed five respective, well-supported clusters within a distinct clade of the genus Acinetobacter which typically, although not exclusively, encompasses strains with strong haemolytic activity. The whole-genome-based average nucleotide identity (ANIb) values supported the species status of each of these clusters. Moreover, the distinctness and coherence of the clusters were supported by whole-cell profiling based on MALDI-TOF MS. Congruent with these findings were the results of metabolic and physiological testing. We conclude that the five putative taxa represent respective novel species, for which the names Acinetobacter courvalinii sp. nov. (type strain ANC 3623(T) =CCUG 67960(T) =CIP 110480(T) =CCM 8635(T)), Acinetobacter dispersus sp. nov. (type strain ANC 4105(T) =CCUG 67961(T) =CIP 110500(T) =CCM 8636(T)), Acinetobacter modestus sp. nov. (type strain NIPH 236(T) =CCUG 67964(T) =CIP 110444(T) =CCM 8639(T)), Acinetobacter proteolyticus sp. nov. (type strain NIPH 809(T) =CCUG 67965(T) =CIP 110482(T) =CCM 8640(T)) and Acinetobacter vivianii sp. nov. (type strain NIPH 2168(T) =CCUG 67967(T) =CIP 110483(T) =CCM 8642(T)) are proposed.

Background Diabetic Goto Kakizaki (GK) rats represent an established model of type 2 diabetes that exhibit an onset of pancreatic islet (PI) pathology characterized by islet hypertrophy with a decreased number of insulin-secreting β-cells. Among the remaining β-cells, oxidative phosphorylation (OXPHOS) and consequently glucose-stimulated insulin secretion (GSIS) are impaired, perhaps owing to a deficit in mitochondrial DNA (mtDNA). We sought to identify this abnormality. Methods β-Cells were obtained from Accutase-dissolved PI isolated from GK or Wistar rats and sorted based on the positive Zn2+ signal of Newport Green. The mtDNA copy number per cell was quantified as the amplicon ratio by polymerase chain reaction using specific primers against the rat ND5 mt gene and UCP2 nuclear gene. Results The 12-month-old GK rats exhibited drastically reduced copy numbers per remaining β-cell, from 7,400 ± 600 in 12-month old Wistar rats (100%) to 24 ± 4%; mtDNA content in heart and liver was 70 ± 25% and 60 ± 20%, respectively. Versus age-paired Wistar rats, 6- and 4-month-old GK rats showed reductions to 60 ± 15% and 50 ± 20%, respectively. Conclusions OXPHOS of remnant β-cells in diabetic GK was drastically impaired due to the lack of sufficient mtDNA levels. We suggest the use of mtDNA quantification to quickly assess PI quality before transplantation.

Mitochondrial calcium-independent phospholipase A2γ (iPLA2γ) belongs to a family of enzymes that participate in cellular signaling by simultaneously producing free fatty acids and lysophospholipids. We have shown previously that the activity of iPLA2γ is increased following the addition of oxidants, such as tert-butyl hydroperoxide (tBHP) or H2O2. We have also shown that iPLA2γ-liberated fatty acids induce H+ transport mediated by mitochondrial uncoupling proteins and mitochondrial adenine nucleotide translocase (ANT) in heart, lung and spleen mitochondria, leading to a decrease in the mitochondrial protonmotive force and subsequent decrease in mitochondrial superoxide production [1,2]. Here we tested the hypothesis that iPLA2γ serves similar antioxidant function in brain. Using brain mitochondria isolated from wild-type (WT) mice, we found that low micromolar concentrations of tBHP induced increase in respiration and a parallel decrease in mitochondrial superoxide and H2O2 production. These effects of tBHP were fully prevented by bovine serum albumin and inhibited by R-bromoenol lactone, a selective inhibitor of iPLA2γ, and were absent in brain mitochondria isolated from iPLA2γ-KO mice. Moreover, the tBHP-induced effects were fully inhibited by carboxyatractyloside, indicating the participation of ANT. In addition, we observed decrease in mRNA levels of selected antioxidant enzymes and increase in lipid peroxidation products in brain tissues homogenates of iPLA2γ-KO mice compared to the WT controls. Our results support the hypothesis of an antioxidant role of iPLA2γ in brain, consistent with iPLA2γ-dependent, ANT-mediated attenuation of mitochondrial superoxide production. Breast cancer cells can survive and proliferate under harsh conditions of nutrient deprivation, including limited oxygen and glucose availability. We hypothesized that such environments trigger metabolic adaptations of mitochondria, which promote tumor progression. Here, we mimicked aglycemia and hypoxia in vitro and compared the mitochondrial and cellular bioenergetic adaptations of human breast cancer (HTB-126) and non-cancer (HTB-125) cells that originate from breast tissue. Using high-resolution respirometry and western blot analyses, we demonstrated that 4 days of glucose deprivation elevated oxidative phosphorylation five-fold, increased the spread of the mitochondrial network without changing its shape, and decreased the apparent affinity of oxygen in cancer cells (increase in C 50 ), whereas it remained unchanged in control cells. The substrate control ratios also remained constant following adaptation. We also observed the Crabtree effect, specifically in HTB-126 cells. Likewise, sustained hypoxia (1% oxygen during 6 days) improved cell respiration in non-cancer cells grown in glucose or glucose-deprived medium (+ 32% and +38%, respectively). Conversely, under these conditions of limited oxygen or a combination of oxygen and glucose deprivation for 6 days, routine respiration was strongly reduced in cancer cells (−36% in glucose medium, −24% in glucose-deprived medium). The data demonstrate that cancer cells behave differently than normal cells when adapting their bioenergetics to microenvironmental conditions. The differences in hypoxia and aglycemia tolerance between breast cancer cells and non-cancer cells may be important when optimizing strategies for the treatment of breast cancer.

Oxidative stress of mitochondrial origin, i.e. elevated mitochondrial superoxide production, belongs to major factors determining aging and oxidative-stress-related diseases. Antioxidants, such as the mitochondria-targeted coenzyme Q, MitoQ(10), may prevent or cure these pathological conditions. To elucidate pro- and anti-oxidant action of MitoQ(10), we studied its effects on HepG2 cell respiration, mitochondrial network morphology, and rates of superoxide release (above that neutralized by superoxide dismutase) to the mitochondrial matrix (J(m)). MitoSOX Red fluorescence confocal microscopy monitoring of J(m) rates showed pro-oxidant effects of 3.5-fold increased J(m) with MitoQ(10). MitoQ(10) induced fission of the mitochondrial network which was recovered after 24h. In rotenone-inhibited HepG2 cells (i.e., already under oxidative stress) MitoQ(10) sharply decreased rotenone-induced J(m), but not together with the Complex II inhibitor thenoyltrifluoroacetone. Respiration of HepG2 cells and isolated rat liver mitochondria with MitoQ(10) increased independently of rotenone. The increase was prevented by thenoyltrifluoroacetone. These results suggest that MitoQ(10) accepts electrons prior to the rotenone-bound Q-site, and the Complex II reverse mode oxidizes MitoQ(10)H(2) to regenerate MitoQ(10). Consequently, MitoQ(10) has a pro-oxidant role in intact cells, whereas it serves as an antioxidant when Complex I-derived superoxide generation is already elevated due to electron flow retardation. Moreover, unlike mitochondrial uncoupling, MitoQ(10) exerted its antioxidant role when Complex I proton pumping was retarded by a hydrophobic amiloride, 5-(N-ethyl-N-isopropyl) amiloride. Consequently, MitoQ(10) may be useful in the treatment of diseases originating from impairment of respiratory chain Complex I due to oxidatively damaged mitochondrial DNA, when its targeted delivery to pathogenic tissues is ensured.

The recent knowledge on mitochondria as the substantial source of reactive oxygen species, namely superoxide and hydrogen peroxide efflux from mitochondria, is reviewed, as well as nitric oxide and subsequent peroxynitrite generation in mitochondria and their effects. The reactive oxygen species formation in extramitochondrial locations, in peroxisomes, by cytochrome P450, and NADPH oxidase reaction, is also briefly discussed. Conditions are pointed out under which mitochondria represent the major ROS source for the cell: higher percentage of non-phosphorylating and coupled mitochondria, in vivo oxygen levels leading to increased intensity of the reverse electron transport in the respiratory chain, and nitric oxide effects on the redox state of cytochromes. We formulate hypotheses on the crucial role of ROS generated in mitochondria for the whole cell and organism, in concert with extramitochondrial ROS and antioxidant defense. We hypothesize that a sudden decline of mitochondrial ROS production converts cells or their microenvironment into a "ROS sink" represented by the instantly released excessive capacity of ROS-detoxification mechanisms. A partial but immediate decline of mitochondrial ROS production may be triggered by activation of mitochondrial uncoupling, specifically by activation of recruited or constitutively present uncoupling proteins such as UCP2, which may counterbalance the mild oxidative stress.

Staphylococcus sciuri is a principally animal-associated bacterial species, but its clinical relevance for humans is increasing. Our study aimed to provide the first insight into the prevalence of this bacterium in a hospital environment. A 3-month surveillance was conducted in a hospital located in Belgrade, Serbia, and 1,028 samples taken from hands of medical personnel, medical devices, and various hospital surfaces were screened for S. sciuri presence. In total, 108 isolates were obtained, which resulted in a relatively high rate of colonization (10.5%). These isolates, along with 7 S. sciuri strains previously isolated in the same hospital ( n = 115), were phenotypically and genotypically characterized. Antimicrobial susceptibility testing revealed that 73% of the strains were resistant to one or more antibiotics, with 4.3% strains displaying multiresistance. Examination of 16S-23S ribosomal DNA intergenic spacer length polymorphism identified the strains at the subspecies level, and 74 (64.3%) strains of S. sciuri subsp. sciuri , 37 (32.2%) strains of S. sciuri subsp. rodentium , and 4 (3.5%) strains of S. sciuri subsp. carnaticus were established. Pulsed-field gel electrophoresis (PFGE) analysis showed 21 distinct pulsotypes, including 17 main types and 4 subtypes. One dominant cluster with 62 strains was found, while 19 (90.5%) of the PFGE types and subtypes identified had 5 or fewer strains. The predominance of small PFGE clusters suggests that the ubiquitous presence of S. sciuri in the outside environment presents the continuous source for colonization of the hospital environment. The presence of one dominant PFGE cluster of strains indicates that some S. sciuri strains may be capable for adaptation to hospital environment conditions and continuous existence in this environment.

The CO2 evolution of intact potato tubers (Solanum tuberosum, L., var. "Bintje") was analyzed during a 10-day period of their warm (25 +/- 2 degrees C) or cold (5 +/- 1 degrees C) storage, to evaluate cold-stress effects on expression and activities of plant uncoupling mitochondrial protein (PUMP) and alternative oxidase (AOX). CO2 evolution rates were analyzed at 20 degrees C, to reflect their possible capacities. The 20 degrees C CO2 production declined from 13 to 8 mg kg(-1) h(-1) after 2 days of warm storage and then (after 3 to 7 days) decreased from 8 to 6.5 mg kg(-1) h(-1). In contrast, 20 degrees C CO2 evolution did not change after the first day of cold storage, increased up to 14.5 mg kg(-1) h(-1) after 2 days, and decreased to about 12 mg kg(-1) h(-1) after 3 to 7 days of cold storage. Cold storage increased PUMP expression as detected by Western blots and led to elevated capacities of both PUMP (44%) and CN-resistant AOX (10 times), but not the cytochrome pathway. Since we found that cold storage led to about the same mitochondrial respiration of 40 nmol O2 min(-1) mg(-1) attributable to each of the respective proteins, we conclude that both AOX and PUMP equally contribute to adaptation of potato tubers to cold.

Two hundred and eighty-four genes of eight eukaryotic genomes for mitochondrial anion carriers were sorted into 43 (+18 single protein) subfamilies. Subfamilies differ by the number, nature, and locations of charges and polar residues in the transmembrane alpha-helices. Consequently, these residues and the rarely unique residues of the matrix and cytosolic segments most likely determine the different molecular phenotypes (functions). 'Common ancestral hydrophilic segments' were found in matrix and cytosolic segments, with interchangeable polar residues. Thus the hydrophobic microstructures of hydrophilic carrier parts are supposed to predetermine structure/conformation, whereas polar and charged microstructures should predetermine function, namely in the transmembrane spanning alpha-helices.

Cancer metabolic alterations result from complex genetic and epigenetic adjustments which include mitochondrial pathways glutaminolysis, reductive carboxylation (RC), and 2-hydroxy-glutarate (2HG) production. We studied regulatory mechanisms that promote anabolic function of mitochondrial isocitrate dehydrogenase 2 (IDH2) towards RC and/or 2HG production in breast cancer cell lines, especially the activity of mitochondrial deacetylase sirtuin SIRT3, which was previously implicated in the regulation of IDH2 enzymatic activity. We demonstrated that IDH2 is a direct substrate of SIRT3 and that distinct regulation by SIRT3 of oxidative vs. reductive IDH2 activity exists. The analysis of metabolite labeling with stable isotope shows that mitochondrial production of 2HG by wild-type IDH2 is largely dependent on SIRT3 presence and activity, as assumed by overexpression of wild-type SIRT3 and SIRT3-inactive mutant in breast cancer cells. Moreover, we demonstrate that IDH2 activity produces 2HG in vitro, as assayed by the product analysis of the enzyme reaction of the isolated recombinant wild-type IDH2. The observed 2HG-producing activity is regulated by SIRT3, as an acetylation surrogate IDH2 mutant K413Q tend to decrease levels of 2HG in vitro. Our findings impact understanding of breast cancer etiology, since breast cancer cell lines exhibit a broad range of IDH2 expression and metabolic phenotypes, including 2HG levels.

The presence of plant-uncoupling mitochondrial protein (PUMP), previously described by Vercesi et al. (1995), was screened in mitochondria of various organs or tissues of several plant species. This was done functionally, by monitoring purine nucleotide-sensitive linoleic acid-induced uncoupling, or by Western blots. The following findings were established: (1) PUMP was found in most of the higher plants tested; (2) since ATP inhibition of linoleic acid-induced membrane potential decrease varied, PUMP content might differ in different plant tissues, as observed with mitochondria from maize roots, maize seeds, spinach leaves, wheat shoots, carrot roots, cauliflower, broccoli, maize shoots, turnip root, and potato calli. Western blots also indicated PUMP presence in oat shoots, carnation petals, onion bulbs, red beet root, green cabbage, and Sedum leaves. (3) PUMP was not detected in mushrooms. We conclude that PUMP is likely present in the mitochondria of organs and tissues of all higher plants.

We have searched for the exclusivity of common sequence motifs of the mitochondrial uncoupling proteins (UCP1, UCP2, UCP3, UCP4, BMCP1, and plant UCP [PUMP]) within the gene family of mitochondrial anion carrier proteins. The UCP-specific sequences, "UCP signatures", were found in the first, second, and fourth alpha-helices. First: Ala/Ser-Cys/Thr/n-n/Phe-Ala/Gly-[negatively charged residue]-n/Phe-n/Cys-Thr-Phe/n; second: Gly/Ala-Ile/Leu-Gln/X-[positively charged residue]-NH-n/Cys-Ser/nphi/X-n/Ser-OH/Gly-n-[positively charged residue]-Ile/Met-Gly/Val-n/Thr; fourth: Pro-Asn/ Thr-n-X-[positively charged residue]-Asn/Ser/Ala-n-n-Ile/Leu-n-Asn/Val-Cys/n-n/Thr-[negatively charged residue]-n-n/Thr/Pro-OH/Val (n, nonpolar; phi, aromatic; (positively charged residue/negatively charged residue, charged residue). The second and part of the third signature are also present in the yeast dicarboxylate transporter. The UCP signature excluding BMCP1 was also found in the second matrix segment: [positively charged residue]-(Pro/ del-Leu/del)-[positively charged residue]-phi-X-Gly/Ser-Thr/n-X-NH/[negatively charged residue]-Ala-phi. These UCP signatures are thought to be involved in fatty acid anion binding and translocation.

Mycobacterium marinum, the cause of chronic systemic infections in fish, occasionally causes granulomatous skin and soft tissue lesions in humans. Cutaneous mycobacterial infection in two patients owing to unusual circumstances is presented in this report. The first patient was infected through improper hygienic behavior, while infection in the second patient was previously misdiagnosed as rheumatoid arthritis and treated with methylprednisolone for a period of three months, which resulted in a rare systemic spread ofM. marinuminto the bones of the hand, testis, and epididymis. Simultaneously, screening for possible sources ofM. marinuminfection in patients' aquaria revealed positive fish harboring VNTR profiles identical to those obtained for clinical isolates from patients.

Transport properties of plant mitochondria from potato tubers were investigated using the swelling technique and membrane potential measurements. Proton-dependent swelling of fatty acid-depleted mitochondria in potassium acetate with valinomycin was possible only in the presence of fatty acids (linoleic acid and 12-(4-azido-2-nitrophenylamino)dodecanoic acid) and was inhibited by various purine nucleotides including ATP, GDP, and GTP. Swelling representing uptake of hexanesulfonate was also inhibited by purine nucleotides. Also, the membrane potential of fatty acid-depleted potato mitochondria energized by succinate declined upon the addition of linoleic acid or 12-(4-azido-2-nitrophenylamino)dodecanoic acid, and this decrease was prevented by ATP and other purine nucleotides. These transport activities are identical to those reported for brown adipose tissue mitochondria and related to the uncoupling protein; therefore, we ascribed them to the plant mitochondrial uncoupling protein (PUMP). A major difference between plant and mammalian uncoupling protein is that PUMP transports small hydrophilic anions such as Cl- very slowly, if at all. We suggest that PUMP may play an important role in plant physiology, where a regulated uncoupling and thermogenesis can proceed during fruit and seed development.

Calcium-independent phospholipase A2γ (iPLA2γ) catalyses the hydrolysis of fatty acids from phospholipids with the consequent generation of lysophospholipids. iPLA2γ is targeted to peroxisomes and mitochondria and results from our laboratory have shown that the activity of mitochondrial iPLA2γ is increased by a redox-sensitive process [1]. Here we aimed to characterize the properties of isolated recombinant iPLA2γ and test the hypothesis that iPLA2γ is activated by thiol oxidation directly. We expressed His-tagged human iPLA2γ using HepG2 or HEK293 cell lines and reconstituted recombinant purified iPLA2γ into liposomes composed of L-α-Phosphatitylcholine and cardiolipin. Using selected specific fluorescent probes and GC/MS analysis, we observed H2O2-induced release of fatty acids in liposomes containing purified recombinant iPLA2γ but not in liposomes lacking the enzyme. The H2O2-activated enzyme released free fatty acids with a specific activity of about 20 nmol.min-1.mg-1. The activity increased several fold in the presence of bovine serum albumin or in mixed micelles containing non-ionic detergent, indicating a possible inhibition of iPLA2γ by the products of its enzymatic activities. H2O2 activated iPLA2γ with a half-maximal constant corresponding to about 1.5 mol H2O2 per mol of enzyme and the active enzyme was fully inhibited by thiol reducing compounds. These results indicate that the regulation of iPLA2γ is dependent on reversible oxidation of accessible cysteine residues and also on a feedback regulation by either fatty acids or lysophospholipids, which is consistent with the physiological participation of iPLA2γ in cellular redox signaling.Supported by the grant GA15-02051S to M.J. from the Grant Agency of the Czech Republic.1. During a 3-year study period, 32,741 urine samples were analyzed for the presence of members of the Staphylococcus sciuri group ( S. sciuri , S. lentus , and S. vitulinus ), and 13 isolates were identified. They presented 0.79% of the total number of coagulase-negative staphylococci isolated. One case of symptomatic urinary tract infection and five possible cases of asymptomatic bacteriuria caused by these bacteria were established. It is noteworthy, however, that over 50% of the isolates originated from hospitalized patients.

In this review we compare situations under which the major cellular role of mitochondria, oxidative phosphorylation (OXPHOS), is transiently suppressed. Two types of cellular bioenergetics exist, related to the predominance of glycolysis either disconnected or fully connected to OXPHOS: i) "glycolytic" phenotype, when the glycolytic end-product pyruvate is marginally used for OXPHOS; and, ii) OXPHOS phenotype with fully developed and active OXPHOS machinery consuming all pyruvate. A switch to glycolytic phenotype is typically orchestrated by gene reprogramming due to AMP-activated protein kinase, hypoxia-induced factor (HIF), NFkappaB, mTOR, and by oncogenes. At normoxia a continuous hydroxylation of HIF1alpha prolines by prolyl hydroxylase domain enzymes (PHDs) and asparagines by factor-inhibiting HIF (FIH) occurs, resulting in HIF1alpha polyubiquitination/degradation. A single mitochondrial network in the cell undergoes constant fission and fusion primarily depending on the local GTP gradients and the mitochondrial energetics. Here we overview the main properties and regulation of pro-fusion and pro-fission mitodynamins, i.e. dynamins-related GTPases responsible for mitochondrial shape-forming, such as pro-fusion mitofusins MFN1, MFN2, and the inner membrane-residing long OPA1 isoforms, and pro-fission mitodynamins FIS1, MFF, and DRP1 multimers required for scission. Notably, the OPA1 cleavage into non-functional short isoforms at a diminished ATP level (collapsed membrane potential) and the DRP1 recruitment upon phosphorylation by various kinases are overviewed. Possible responses of mitodynamins to the oxidative stress, hypoxia, and concomitant mtDNA mutations are also discussed. We hypothesize that the increased GTP formation within the Krebs cycle followed by the GTP export via the ADP/ATP carrier shift the balance between fission and fusion towards fusion by activating the GTPase domain of OPA1 located in the peripheral intermembrane space (PIMS). Since the protein milieu of PIMS is kept at the prevailing oxidized redox potential by the TOM, MIA40 and ALR/Erv1 import-redox trapping system, redox regulations shift the protein environment of PIMS to a more reduced state due to the higher substrate load and increased respiration. A higher cytochrome c turnover rate may prevent electron transfer from ALR/Erv1 to cytochrome c. Nevertheless, the putative links between the mitodynamin responses, mitochondrial morphology and the changes in the mitochondrial bioenergetics, superoxide production, and hypoxia are yet to be elucidated, including the precise basis for signaling by the mitochondrion-derived vesicles.

Vulnerability of mitochondrial Complex I to oxidative stress determines an organism's lifespan, pace of aging, susceptibility to numerous diseases originating from oxidative stress and certain mitopathies. The mechanisms involved, however, are largely unknown. We used confocal microscopy and fluorescent probe MitoSOX to monitor superoxide production due to retarded forward electron transport in HEPG2 cell mitochondrial Complex I in situ. Matrix-released superoxide production, the un-dismuted surplus (J(m)) was low in glucose-cultivated cells, where an uncoupler (FCCP) reduced it to half. Rotenone caused a 5-fold J(m) increase (AC(50) 2 microM), which was attenuated by uncoupling, membrane potential (DeltaPsi(m)), and DeltapH-collapse, since addition of FCCP (IC(50) 55 nM), valinomycin, and nigericin prevented this increase. J(m) doubled after cultivation with galactose/glutamine (i.e. at obligatory oxidative phosphorylation). A hydrophobic amiloride that acts on the ND5 subunit and inhibits Complex I H(+) pumping enhanced J(m) and even countered the FCCP effect (AC(50) 0.3 microM). Consequently, we have revealed a new principle predicting that Complex I produces maximum superoxide only when both electron transport and H(+) pumping are retarded. H(+) pumping may be attenuated by high protonmotive force or inhibited by oxidative stress-related mutations of ND5 (ND2, ND4) subunit. We predict that in a vicious cycle, when oxidative stress leads to higher fraction of, e.g. mutated ND5 subunits, it will be accelerated more and more. Thus, inhibition of Complex I H(+) pumping, which leads to oxidative stress, appears to be a missing link in the theory of mitochondrial aging and in the etiology of diseases related to oxidative stress.

Increased ATP/ADP ratio resulting from enhanced glycolysis and oxidative phosphorylation represents a plausible mechanism controlling the glucose-stimulated insulin secretion (GSIS) in pancreatic beta-cells. Although specific bioenergetics might be involved, parallel studies of cell respiration and mitochondrial membrane potential (DeltaPsi(m)) during GSIS are lacking. Using high resolution respirometry and parallel DeltaPsi(m) monitoring by two distinct fluorescence probes we have quantified bioenergetics in rat insulinoma INS-1E cells representing a suitable model to study in vitro insulin secretion. Upon glucose addition to glucose-depleted cells we demonstrated a simultaneous increase in respiration and DeltaPsi(m) during GSIS and showed that the endogenous state 3/state 4 respiratory ratio hyperbolically increased with glucose, approaching the maximum oxidative phosphorylation rate at maximum GSIS. Attempting to assess the basis of the "toxic" effect of fatty acids on insulin secretion, GSIS was studied after linoleic acid addition, which diminished respiration increase, DeltaPsi(m) jump, and magnitude of insulin release, and reduced state 3/state 4 dependencies on glucose. Its effects were due to protonophoric function, i.e. uncoupling, since without glucose, linoleic acid accelerated both state 3 and state 4 respiration by similar extent. In turn, state 3 respiration increased marginally with linoleic acid at 10-20mM glucose. We conclude that upon glucose addition in physiological range, the INS-1E cells are able to regulate the oxidative phosphorylation rate from nearly zero to maximum and that the impairment of GSIS by linoleic acid is caused by mitochondrial uncoupling. These findings may be relevant to the pathogenesis of type 2 diabetes.

Uncoupling proteins 1 (UCP1) and 2 (UCP2) belong to the family of mitochondrial anion transporters and share 59% sequence identity with each other. Whereas UCP1 was shown to be responsible for the rapid production of heat in brown adipose tissue, the primary function and transport properties of ubiquitously expressed UCP2 are controversially discussed. Here, for the first time, the activation pattern of the recombinant human UCP2 in comparison to the recombinant human UCP1 are studied using a well-defined system of planar lipid bilayers. It is shown that despite apparently different physiological functions, hUCP2 exhibited its protonophoric function similar to hUCP1--exclusively in the presence of long-chain fatty acids (FA). The calculated hUCP2 transport rate of 4.5 s(-1) is the same order of magnitude, as shown previously for UCP1. It leads to the conclusion that the differences in the activity of both proteins in living mitochondria are based exclusively on their different expression level. Both proteins are activated much more effectively by polyunsaturated than by saturated FA. The proton and total membrane conductances increased in the range palmiticoleiceicosatrienoiclinoleicretinoicarachidonic acids. The higher uncoupling protein (UCP)-dependent conductance in the presence of polyunsaturated FA is explained on the basis of the FA cycling hypothesis.

In order to assess the quality of freshly isolated and cultivated pancreatic islets designed for experimental transplantation in rats we combined the vitality staining test, in vitro measurement of insulin secretion capacity, and assessment of islet respiration. Oxygen consumption was measured using the respirometer Oxygraph 2K equipped with polarographic oxygen sensors. The results of oxymetry demonstrated a linear correlation between islet number and oxygen consumption. Respiration per unit of viable islet tissue was constant. Oxygen consumption tests were in good correlation with the results of insulin release assays, with a correlation coefficient of 0.82. We found no significant differences in all three vitality-testing methods performed with fresh and 24-hour cultivated islets (P > .05). We conclude that polarographic oxymetry provides a fast and easy evaluation test of islet quality. After appropriate standardization, the oxymetric technique can be used for routine clinical pretransplant islet quality testing. In addition, cell membrane integrity and mitochondrial function could be assessed after addition of specific respiration inhibitors or stimulators.

A total of 28 staphylococcal isolates from human clinical specimens belonging to the Staphylococcus sciuri group were identified and characterized. The API Staph and ID32 STAPH correctly identified S. sciuri and S. lentus but not S. vitulinus strains. Identification to the subspecies level was possible only by a PCR-based method.

The mammalian uncoupling protein (UCP-1) from the gene family of mitochondrial carriers is a dimer of identical 33 kDa subunits, each containing six membrane-spanning alpha-helices. Its expression, restricted to brown fat, occurs upon birth, cold acclimation and overfeeding. UCP-1 dissipates redox energy and thereby provides heat to the animal. Two additional isoforms have recently been discovered, 59% homologous UCP-2, widely expressed (heart, kidney, lung, placenta, lymphocytes, white fat); and UCP-3 (57% homologous), found in brown fat and skeletal muscle. Their physiological roles are unknown, but may include the regulation of body weight and energy balance, muscle nonshivering thermogenesis, fever, and defense against generation of reactive oxygen species. Consequently, great pharmacological potential is expected in revealing their biochemical and hormonal regulators. UCP-1 mediates a purine-nucleotide-sensitive uniport of monovalent unipolar anions, including fatty acids, that lead to fatty acid cycling and uncoupling. UCP-2 and UCP-3 are expected to share a similar mechanism.

Genes encoding staphylococcal enterotoxins ( sea to see , seg , and seh ), toxic shock syndrome toxin 1 ( tst ), and exfoliative toxins ( eta and etb ) were not detected in a large panel of 48 Staphylococcus sciuri group isolates tested. This strongly suggests that production of the staphylococcal exotoxins by these bacteria is highly unlikely.

We hypothesize that fatty acid-induced uncoupling serves in bioenergetic systems to set the optimum efficiency and tune the degree of coupling of oxidative phosphorylation. Uncoupling results from fatty acid cycling, enabled by several phylogenetically specialized proteins and, to a lesser extent, by other mitochondrial carriers. It is suggested that the regulated uncoupling in mammalian mitochondria is provided by uncoupling proteins UCP-1, UCP-2 and UCP-3, whereas in plant mitochondria by PUMP and StUCP, all belonging to the gene family of mitochondrial carriers. UCP-1, and hypotheticaly UCP-3, serve mostly to provide nonshivering thermogenesis in brown adipose tissue and skeletal muscle, respectively. Fatty acid cycling was documented for UCP-1, PUMP and ADP/ATP carrier, and is predicted also for UCP-2 and UCP-3. UCP-1 mediates a purine nucleotide-sensitive uniport of monovalent unipolar anions, including anionic fatty acids. The return of protonated fatty acid leads to H+ uniport and uncoupling. UCP-2 is probably involved in the regulation of body weight and energy balance, in fever, and defense against generation of reactive oxygen species. PUMP has been discovered in potato tubers and immunologically detected in fruits and corn, whereas StUCP has been cloned and sequenced from a potato gene library. PUMP is supposed to act in the termination of synthetic processes in mature fruits and during the climacteric respiratory rise.

Thermogenic uncoupling has been proven only for UCP1 in brown adipose tissue. All other isoforms of UCPs are potentially acting in suppression of mitochondrial reactive oxygen species (ROS) production. In this contribution we show that BAT mitochondria can be uncoupled by lauric acid in the range of ∼100 nM when endogenous fatty acids are combusted by carnitine cycle and β-oxidation is properly separated from the uncoupling effect. Respiration increased up to 3 times when related to the lowest fatty acid content (BSA present plus carnitine cycle). We also illustrated that any effect leading to more coupled states leads to enhanced H 2 O 2 generation and any effect resulting in uncoupling gives reduced H 2 O 2 generation in BAT mitochondria. Finally, we report doubling of plant UCP transcript in cells as well as amount of protein detected by 3 H-GTP-binding sites in mitochondria of shoots and roots of maize seedlings subjected to the salt stress.

Mersalyl, 5,5'-dithio-bis(2-nitrobenzoate) (Nbs2) and fluorescent Thiolyte DB react with SH groups in the H+ channel (SHc) of the uncoupling protein of brown adipose tissue mitochondria, as inferred from their inhibition of H+ transport. Cl- transport by the uncoupling protein was unaffected. Using these modifiers and N-ethylmaleimide (MalNEt), distinct SH groups (SHB) in the purine nucleotide binding site were identified. Nbs2 reacts more readily with the SHB than with the SHc groups, but mersalyl and Thiolyte DB are more reactive with the SHc groups. MalNEt reacts exclusively with the SHB. GDP inhibition is fully prevented after sufficient modification of the SHB. Pretreatment with p-diazobenzenesulfonate (N2PhSO2) suppresses only 20-25% of fluorescence of Thiolyte-DB-labeled uncoupling protein on SDS/PAGE gels, while MalNEt suppresses 66% and Nbs2 80-90%. Since N2PhSO2 also affects the GDP binding site, these results demonstrate that the N2PhSO2-reactive residue is not identical with the SHB.

The physiological role of monocarboxylate transport in brown adipose tissue mitochondria has been reevaluated. We studied pyruvate, α-ketoisovalerate, α-ketoisocaproate, and phenylpyruvate uniport via the uncoupling protein (UCP1) as a GDP-sensitive swelling in K+salts induced by valinomycin or by monensin and carbonyl cyanide- p-(trifluoromethoxy)phenylhydrazone in Na+salts. We have demonstrated that this uniport is inhibited by fatty acids. GDP inhibition in K+salts was not abolished by an uncoupler, indicating a negligible monocarboxylic acid penetration via the lipid bilayer. In contrast, the electroneutral pyruvate uptake (swelling in ammonium pyruvate or potassium pyruvate induced by change in pH) mediated by the pyruvate carrier was inhibited by its specific inhibitor α-cyano-4-hydroxycinnamate but not by fatty acids. Moreover, α-cyano-4-hydroxycinnamate enhanced the energization of brown adipose tissue mitochondria, which was monitored fluorometrically by 2-(4-dimethylaminostyryl)-1-methylpyridinium iodide and safranin O. Consequently, we suggest that UCP1 might participate in futile cycling of unipolar ketocarboxylates under certain physiological conditions while expelling these anions from the matrix. The cycle is completed on their return via the pyruvate carrier in an H+symport mode.

Upon certain insults and/or pathological states fragmented mitochondrial (mt) network persists in cells, with spheroid mt remnants containing clustered nucleoids of mtDNA (1).To image nucleoid distribution within mt network, we employed 3D super-resolution fluorescent photoactivable localization microscopy (FPALM)(2), in conjunction with photoconvertible fluorescent protein conjugates of marker proteins such as mitochondrial (mt) transcription factor A (TFAM) or mt single-stranded-DNA-binding protein (mtSSB). We have shown that nucleoids in hepatocellular HepG2 cells exhibit a rather wide size distribution ranging between 50 to 300 nm. This was confirmed using 3D TFAM/mtSSB immunocytochemistry in conjunction with direct stochastic optical reconstruction microscopy (dSTORM). Mitochondrial (mt) network undergoes locally frequent fragmentation and fusion events. When integrated over time, its basic morphology encompasses highly interconnected mt reticulum, a single mitochondrion within the cell (1). Upon certain insults and/or pathological states fragmented network persists. MtDNA is organized in nucleoids containing assessor proteins and recruited proteins of mt replication/ transcription machinery. It is debated on their uniform size and whether a single nucleoid contains a single mtDNA molecule or up to average 6 mtDNA molecules. To image nucleoid distribution within mt network, we employed 3D super-resolution fluorescent photactivable localization microscopy of Biplane schema (1). Mt network of hepatocellular carcinoma HepG2 cells was imaged first by its matrix space using mtEos2 or as outer mitochondrial membrane contour using Eos2-conjugates of truncated FIS1 protein (not inducing massive fission, Eos2-FIS1tr). Resulting 3D images confirm the existence of highly-connected mt network and unlike conventional confocal microscopy, 3D BiplaneFPALM distinguished a hollow character of mt reticulum tubules when visualized by Eos2-FIS1tr. Upon network fragmentation, hollow max ∼2 micrometer spheres occurred. Imaging of mt nucleoids confirmed the existence of ∼1000 nucleoids per cell with size distribution from 50 nm to 300 nm. Optimized dual transfection strategy had to be employed for simultaneous imaging of network (mtEos or Eos2-FIS1tr) and nucleoids (mtSSB-PSCFP2). Images revealed an equidistant nucleoid distribution of an average distance of ∼1 micrometer between nucleoids. Three-dimensional (3D) super-resolution microscopy, using a biplane detection scheme, termed biplane photo-activated localization microscopy (Biplane FPALM), enables imaging of volumes as thick as whole cells and could reveal unknown details of cellular organization. Hence, we attempted to visualize mitochondrial reticulum via the matrix space loaded with mitochondria-addressed Eos, while transfecting cells by lentiviral expression. Our 3D images of single Eos molecules in the matrix space have proven the continuous character of mitochondrial reticulum tubules, i.e., an existence of a highly interconnected major mitochondrial reticulum in insulinoma Ins1E and oxidative-phosphorylation-dependent glutaminolytic hepatoma HepG2 cells (Fig.1). Also, using Eos-conjugate of mitochondrial transcription factor-A (TFAM), we have imaged nucleoids of mitochondrial DNA (mtDNA) in which TFAM represents a major assessor protein. Using PA-CFP2-TFAM and mitochondria-addressed Eos, the first 3D two color super-resolution images were obtained for mitochondrial reticulum together with the distribution of mt nucleoids in it.
