However, clinical trials based on its oral administration have failed to provide significant effect on cardiac functionality. The main limitation of CoQ10 is based on its very low oral bioavailability and instability that limit dramatically its effects as a cardioprotective agent. Herein, we loaded CoQ10 in high bioavailable nano-emulsions (NEs) coated with chitosan or chitosan and hyaluronic acid in order to improve its performance.Methods: We tested cardioprotective and hepatoprotective effects of CoQ10-loaded nano-carriers against Doxorubicin and Trastuzumab toxicities in cardiomyocytes and liver cells through analysis of cell viability, lipid peroxidation, expression of leukotrienes, p65/NF-kB and pro-inflammatory cytokines involved in anticancer-induced cardio and hepatotoxicity.Results: Nano-carriers showed high stability and loading ability and increased cell viability both in hepatocytes and cardiomyocytes during anticancer treatments. We observed that these effects are mediated by the inhibition of lipid peroxidation and reduction of the inflammation. CoQ10-loaded nano-emulsions showed also strong anti-inflammatory effects reducing leukotriene B4 and p65/NF-κB expression and Interleukin 1β and 6 production during anticancer treatments.Discussion: Anthracyclines and Human epidermal growth factor receptor (HER2) inhibitors have shown significant anticancer effects in clinical practice but their use is characterized by cardiotoxicity and hepatotoxicity. Nano-carriers loaded with CoQ10 showed cardio and hepatoprotective properties mediated by reduction of oxidative damages and pro-inflammatory mediators. These results set the stage for preclinical studies of cardio and hepatoprotection in HER2+ breast cancer-bearing mice treated with Doxorubicin and Trastuzumab.Keywords: cardio-oncology, nano-medicine, Coenzyme Q10, doxorubicin, trastuzumab, inflammation

In cancer therapy, stimulus-responsive drug delivery systems are of particular interest for reducing side effects in healthy tissues and improving drug selectivity in the tumoral ones. Here, a strategy for the preparation of a photo-responsive cross-linked trilayer deposited onto an oil-in-water nanoemulsion via a layer-by-layer technique is reported. The system is made of completely biocompatible materials such as soybean oil, egg lecithin and glycol chitosan, with heparin as the polymeric shell. The oil core is pre-loaded with curcumin as a model lipophilic active molecule with anti-tumoral properties. The trilayer cross-linkage is performed via a photoinitiator-free thiol-ene ‘click’ reaction. In particular, the system is implemented with an o -nitrobenzyl group functionalized with a thiol moiety which can perform both the thiol-ene ‘click’ reaction and the cleavage meant for controlled drug release at two different wavelengths, respectively. So the preparation and characterization of a photo-responsive natural nanocarrier (PNC) that is stable under physiological conditions owing to the thiol-ene cross-linkage are reported. PNC performance has been assessed in vitro on melanoma cells as well as in vivo on xenograft tumour-induced mice.

Butyric acid (BA) has been reported to induce anticancer effects on hepatocellular carcinoma (HCC) cells both in vitro and in vivo. However, its delivery and release in cancer tissues must be optimized. On the basis of these requirements, we prepared liposomes coated with chitosan and uncoated liposomes and both types were loaded with BA through a thin-film hydration method. The liposomes coated or uncoated with chitosan had a mean hydrodynamic size of 83.5 and 110.3 nm, respectively, with a homogeneous size distribution of the particles. For evaluation of the biological effects of the nanoformulations, the hepatoblastoma (HB) HepG2 cell line was utilized. BA-loaded liposomes coated with chitosan showed a considerable higher cytotoxicity than both uncoated liposomes and free BA, with IC 50 values, after 72 h of incubation, of 7.5, 2.5 and 1.6 mM, respectively. Treatment of HepG2 cells for 5 h with the BA-loaded liposomes coated with chitosan at 5 mM lowered the extent of the increase in IL-8, IL-6, TNF-α and TGF-β expression of approximately 64, 58, 85 and 73.8%, respectively, when compared to the untreated cells. The BA-loaded liposomes coated with chitosan had marked capacity to be internalized in human HB cells showing an increased cytotoxic activity when compared with free BA and important anti-inflammatory effects by inhibiting production of cytokines with a central role in liver cell survival. This paper focused on the biodistribution of the cross-linked hyaluronic acid (HA-NPs) sub-micron particles in tumor-bearing mice. Solvent-non solvent method followed glutaraldehyde cross-linking utilized for the fabrication of HA-NPs. Size measurement and morphological analysis were performed by dynamic light scattering and electron microscopy, respectively and the size found to be in the range of 200–400 nm. In vitro viability in LNCaP cell line was assessed by water soluble tetrazolium assay after 24 h of exposure to sub-micron particles and no toxicity was found to higher concentration of 3 mg/mL. Internalization of particles in prostate cancer cell LNCaP were studied by confocal microscopy with FITC labeled submicron particles and involvement of hyaluronan receptor mediated uptake/endocytosis was confirmed by competitive assay. Biodistribution studies were performed in xenograft prostate cancer mice model with fluorophore labeled particles and monitored in tumoral parenchyma with strong fluorescence, meanwhile very less signal in liver, kidney and spleen while no fluorescence found in lung after 24 h of systemic administration; that shown ability of this HA based system to recognize cancer tissue. These result fetched that hyaluronic acid based system is selective for tumoral site and can be utilized to deliver bioactives in specific (targeting) and controlled (temporal) manner to cancerous tissue.

The fluidity of the hydrophobic interior of phospholipid vesicles after calcium-dependent binding of human annexin V (AVH) was studied using EPR spectroscopy. Vesicles (SUVs) composed of PC or PE and an acidic phospholipid (alternatively PS, PA, or CL) were probed at different bilayer depths by either phosphatidylcholine, or the accompanying acidic phospholipid, bearing a spin label probe at position C-5, C-12, or C-16 of the sn-2 acyl chain. Alternatively, the vesicle surface was probed with a polar head spin labeled PE (PESL). The EPR spectra of annexin-bound bilayer domain(s) were obtained by computer spectral subtraction. The order parameter values (S) from the resulting difference spectra revealed that the bilayer hydrophobic interior has a greatly altered fluidity gradient, with an increased rigidity up to the C-12 position. Thereafter, the rigidification progressively vanished. The effect is not linked to the phospholipid class, since all the acidic phospholipid spectra, as well as phosphatidylcholine, shared the same sensitivity to the bound protein. The observed membrane rigidification appears to parallel the "crystallizing" tendency of vesicle-bound annexin V, but may not be involved in the calcium channeling activity of this protein.

Sensitivity to digestion with pronase has been used to show that the precursor form of mitochondrial aspartate aminotransferase, the form lacking the N-terminal presequence, that with a deletion of the first 9 residues and mutants of the mature enzyme in which residue Cys-166 is mutated to alanine or serine, all retain unfolded conformations after synthesis in a reticulocyte lysate. In the presence of lysed mitochondria the various forms of mitochondrial aspartate aminotransferase retained their susceptibilities to pronase in a way that mirrored the efficiencies with which they are imported into intact mitochondria. The results are interpreted as showing that the presequence of mitochondrial aspartate aminotransferase is not uniquely required for interaction with cytosolic factors required to maintain the newly synthesised protein in a form competent for interacting with, and being imported into, mitochondria.

The properties of the binding of annexin V to variously composed phospholipid vesicles have been studied by applying a recently developed EPR method, using an annexin V spin label. By this approach, this protein is seen to bind to acidic phospholipid-containing vesicles, as reported, thus confirming the reliability of the method. In addition, binding of this annexin to cardiolipin-containing vesicles has been studied in more depth, and the protein has been shown to have a distinct affinity for this phospholipid. As a cardiolipin-rich natural membrane system, mitochondrial membranes and mitoplasts from rat liver were considered, and a strong binding of AV to these membranes was observed. Having compared this binding with that to phospholipid vesicles, cardiolipin-rich microdomains in the mitochondrial membranes are proposed as the putative mitochondrial binding sites for annexin V.

Glutamine transport in both normal and acidotic rat kidney mitochondria was investigated using both isotopic techniques and by spectroscopic measurements in which glutamine metabolism was allowed to occur. Widely used criteria for demonstrating the occurrence of carrier-mediated transport were successfully applied in both cases. Three transport mechanisms were found to occur, namely glutamine uniport, active only during acidosis and glutamine/glutamate and glutamine/malate antiports, active in both normal and acidotic mitochondria. Efflux of glutamate, via a glutamate/OH − translocator, following glutamine uptake by mitochondria was experimentally ruled out. Glutamine uniport in acidotic mitochondria and glutamine/glutamate and glutamine/malate antiports in both normal and acidotic mitochondria were investigated in detail: differences found in K m and V max values, in pH and temperature dependence, and in the pattern of inhibitor sensitivity of glutamine transport demonstrated the existence of five different translocators whose activities were found to fit with the physiological requirements of renal ammoniogenesis.

To gain further insight into the effect of He-Ne laser irradiation on the mitochondrial biosynthetic apparatus, DNA synthesis was measured in mitochondna, mitoplasts and mitochondrial matrix fraction irradiated with low power continuous wave He-Ne laser (energy dose 5 Joules/cm2 laser power 12 mW). As a result of irradiation, the amount of α-[32P]dATP incorporated in acid insoluble materials was found to increase. Electrophoretic analysis of in vitro synthesised DNA purified from mitochondria, mitoplasts and matrix fraction shows an increase of 50-60 % of DNA synthesis in the irradiated samples. These data show that He-Ne laser stimulates the replication of mitochondrial DNA in both intact organelles and soluble matrix fraction, thus suggesting an interaction of laser light with matrix soluble molecule/s.

To gain some insight into the role played by certain protein domains in the import of mitochondrial aspartate aminotransferase in isolated mitochondria, three protein mutants were constructed by using the plasmid pOTS-mAspAT, which contains the nucleotide sequence encoding for the mature form of this enzyme. Two mutant proteins in which Cys-166 was substituted with either serine or alanine and another protein lacking the nine N-terminal amino acids were all synthesized in a cell-free transcription/translation system. Comparison was made among the newly synthesized mutant proteins and the newly synthesized wild type aspartate aminotransferase with respect to their capability to enter mitochondria. All the mutant proteins proved to be able to enter mitochondria even though with a lower efficiency than the wild type enzyme. Interestingly the thiol reagent mersalyl proved to inhibit import of both wild type enzyme and serine mutant, whereas import of alanine mutant was found to be insensitive to mersalyl, thus showing that Cys-166 is the unique -SH group involved in import. Import of mitochondrial aspartate aminotransferase by mitochondria is shown to involve certain protein domains present in the mature protein, two of them being the Cys-166 and the N-terminal regions.

To gain some insight into the process by which both acetylCoA and NADPH, needed for fatty acid synthesis, are obtained, in the cytosol, from the effluxed intramitochondrial citrate, via citrate lyase and malate dehydrogenase plus malic enzyme respectively, the capability of externally added pyruvate to cause efflux of malate from rat liver mitochondria was tested. The occurrence of a pyruvate/malate translocator is here shown: pyruvate/malate exchange shows saturation features (Km and Vmax values, measured at 20 degrees C and at pH 7.20, were found to be about 0.25 mM and 2.7 nmoles/min x mg mitochondrial protein, respectively) and is inhibited by certain impermeable compounds. This carrier, together with the previously reported tricarboxylate and oxodicarboxylate translocators proved to allow for citrate and oxaloacetate efflux due to externally added pyruvate.

The nature of the inhibition of the dicarboxylate carrier by compounds reacting with SH groups has been investigated. 1. 1. Mersalyl and p-hydroxymercuribenzoate increase the KmKm without changing the V of malonate P i exchange, when they are added simultaneously with the dicarboxylate. If, on the other hand, the mitochondria are preincubated with SH reagents prior to the addition of malonate, the mersalyl inhibition of malonate P i exchange becomes predominantly non-competitive with respect to malonate. 2. 2. In the case of P i P i exchange, catalyzed by the dicarboxylate carrier, the mersalyl inhibition is competitive with respect to Pi (as indicated by Lineweaver-Burk plots), even when mersalyl is added before the substrate. Dixon plots of the rate of Pi uptake against mersalyl concentration are, however, non-linear, suggesting that the inhibition is partially competitive. 3. 3. Dicarboxylates and dicarboxylate analogous protect against SH reagent inhibition of both dicarboxylate and Pi uptake via the dicarboxylate carrier. The protectors are effective when added before, or together with the SH reagents, but do not reverse the inhibition once it has been established. Protection by substrate analogues progressively decreases, as the time of incubation with the SH reagent increases. 4. 4. The presence of Pi does not protect against the SH reagent inhibition of the Pi uptake. 5. 5. The rate of SH reagent inhibition of the dicarboxylate carrier is competively inhibited by dicarboxylates. 6. 6. It is concluded that SH reagents bind at or near the dicarboxylate specific binding site and distant from the Pi binding site. As a result of this reaction these inhibitors prevent dicarboxylate binding directly and decrease the affinity for Pi by an indirect conformational change.

Both the precursor and the mature form of mitochondrial aspartate aminotransferase were synthesized in a cell-free coupled transcription/translation system directed by the recombinant expression plasmid pOTS-pmAspAT and pOTS-mAspAT, respectively. Both newly synthesized forms of the protein were imported into isolated mitochondria, with the precursor correctly processed to the mature form. In both cases the import process showed resistance to externally added pronase and was abolished in mitochondria treated with the uncoupler carbonyl cyanide m-chlorophenylhydrazone. Moreover the imported products showed the same intramitochondrial localization as judged by a subfractionation procedure. In both cases import was time dependent and was completed in about 15 min. Finally a competitive inhibition of the import of the precursor of aspartate aminotransferase was found due to externally added purified aspartate aminotransferase.

Summary We have applied a double tagging system in order to study whether purified myelin basic protein is able to adhere to normal human peripheral T lymphocytes without the need to purify cells. Evaluation of myelin basic protein adherence to peripheral blood mononuclear cells was determined with biotinylated myelin basic protein and fluoresceinated avidin, and lymphocyte population was identified by the corresponding phycoerythrinated monoclonal antibody. The observed adherence of myelin basic protein to T lymphocytes was found to depend on protein conformation.

The effect of aging and treatment with acetyl-l-carnitine on the activity of cytochrome oxidase and adenine nucleotide translocase in rat heart mitochondria was studied. It was found that the activity of both these mitochondrial protein systems was reduced (by around 30%) in aged animals. Treatment of aged rats with acetyl-l-carnitine almost completely reversed this effect. Changes in the mitochondrial cardiolipin content appear to be responsible for these effects of acetyl-l-carnitine.

To gain insight into the uptake of mature aspartate aminotransferase by isolated mitochondria, the capability of certain cyanogen bromide peptides from mature beef heart mitochondrial aspartate aminotransferase to inhibit enzyme uptake was kinetically tested. N-terminal peptides (1–9 and 10–31) proved to inhibit the rate of aspartate aminotransferase uptake respectively in purely competitive and non-competitive ways, whereas other peptides distal from the N-terminus (203–217, 321–327 and 328–353) were found to be completely ineffective.

We show here that TPP→TMP conversion can take place in rat liver mitochondria. This occurs via the novel, putative TPP pyrophosphatase localised in the mitochondrial matrix, as shown both by digitonin titration and by an HPLC enzyme assay carried out on the mitochondrial matrix fraction. Certain features of the reaction, including the substrate and pH dependence, are reported. Additional evidence is given that externally added TMP can cross the mitochondrial membrane in a manner consistent with the occurrence of a carrier-mediated process. This can occur both via the TPP translocator and via a novel translocator, inhibited by CAT but different from the ADP/ATP carrier.

Rat heart mitochondrial membranes exposed to the free radicals generating system tert-butylhydroperoxide/Cu2+ undergo lipid peroxidation as evidenced by the accumulation of thyobarbituric acid reactive substances. Mitochondrial lipid peroxidation resulted in a marked loss of both cytochrome c oxidase activity and cardiolipin content. The alterations in the properties of cytochrome c oxidase were confined to a decrease in the maximal activity (Vmax) with no change in the affinity (Km) with respect to the substrate cytochrome c. Various lipid soluble antioxidants could prevent the lipid peroxidation reaction and the associated loss of cytochrome c oxidase activity. External added cardiolipin but no other phospholipids, nor peroxidized cardiolipin was able to prevent the loss of cytochrome oxidase activity induced by lipid peroxidation. These results establish a close correlation between oxidative damage to cardiolipin and alterations in the cytochrome oxidase activity and may prove useful in probing molecular mechanism of free radicals induced peroxidative damage of mitochondria which has been proposed to contribute to aging and to chronic degenerative diseases.

The effect of hyperthyroidism on fatty acid oxidation and on carnitine-acylcarnitine translocase activity in rat heart mitochondria has been studied. The rates of palmitoylcarnitine supported respiration as well as the carnitine-palmitoylcarnitine exchange reaction were both stimulated (approx. 36%) in heart mitochondria from hyperthyroid rats. Kinetic analysis of the carnitine-carnitine exchange reaction showed that thyroid hormone affects the Vmax of this process, while having no effect on the Km values. The level of cardiolipin was significantly higher (approx. 40%) in heart mitoplasts from hyperthyroid rats than from the control rats. It can be concluded that thyroid hormones produce a stimulation of heart mitochondrial carnitine translocase activity and that the basis of this effect is likely an increase in the cardiolipin content.

Different proteolytic enzymes were tested for their ability to degrade the myelin basic protein of the central nervous system, purified in two different forms, the lipid-free form and the lipid-bound form. As shown by SDS gel electrophoresis only clostripain, a thiol protease, was able to distinguish between the two MBPs since it degraded MBP only in the lipid-free form. The failure to degrade lipid-bound MBP by clostripain could not be ascribed to the presence of lipids, since the other proteolytic enzymes tested degraded both MBPs independently from lipids giving fragments with different size. These results may be related to different conformations of MBPs possibly relevant for the study of myelin structure and antigenic properties of the protein.

The Magnoliaceae are discussed as one of the key species at the root of the flowering plants. To obtain molecular information for one of these phylogenetically interesting plant species, we determined genomic and cDNA sequences of the mitochondrial cox3 gene in Magnolia grandifora . Twenty-two RNA editing events are identified to alter cytidines in the mRNA to uridines, all but one of which change the encoded amino acid identity. RNA editing in the cox3 coding region is thus more frequent in Magnolia than in other dicot or monocot plants investigated and almost as predominant as in some gymnosperms. The cox3 RNA editing frequency in Magnolia thus occupies an intermediate position between angiosperms and gymnosperms consistent with the phylogenetic position of the Magnoliales.

Mutant genes were constructed which coded for the precursor form of mitochondrial aspartate aminotransferase in which residue cysteine 166 was mutated to either serine or alanine and or forms of the protein lacking both the presequence and residues 1-9 of the mature protein but carrying the same cysteine mutations. The protein products of all of these mutant genes were imported into mitochondria that had been added to the expression system but with varying degrees of efficiency. The results showed that the effects of mutation of cysteine 166 and of deletion of residues 1-9 of the mature protein on sequestration into mitochondria were essentially cumulative, suggesting that these parts of the protein are involved in distinct steps on the recognition/uptake pathway.

Two fused genes were constructed which encode for two chimeric proteins in which either 10 or 191 N-terminal amino acids of mature mitochondrial aspartate aminotransferase had been attached to the entire polypeptide chain of cytosolic dihydrofolate reductase. The precursor and mature form of mitochondrial aspartate aminotransferase, dihydrofolate reductase and both chimeric proteins were synthesized in vitro and their import into isolated mitochondria was studied. Both chimeric proteins were taken up by isolated organelles, where they became protease resistant, thus indicating the ability of the N-terminal portion of the mature moiety of the precursor of mitochondrial aspartate aminotransferase to direct cytosolic dihydrofolate reductase into mitochondria.

The interaction of myelin basic protein (MBP) with zinc and phosphate ions has been studied by using the emission properties of the single tryptophan residue of the protein (Trp-115). The studies have been carried out by means of both static and time-resolved fluorescence techniques. The addition of either zinc to MBP in the presence of phosphate or phosphate to MBP in the presence of zinc resulted in an increase of fluorescence intensity and a blue shift of the emission maximum wavelength. Furthermore, a concomitant increase in the scattering was also detected. Anisotropy decay experiments demonstrated that these effects are due to the formation of MBP molecules into large aggregates. A possible physiological role for such interaction is discussed.

Proline transport in rat kidney mitochondria was investigated both by using isotopic techniques and by spectroscopic measurements, in which proline metabolism was essentially allowed to occur. Widely used criteria for demonstrating the occurrence of carrier-mediated transport were successfully applied in both cases. Differences found in the Km and Vmax values, in pH and temperature dependence of proline transport, and in the inhibitor sensitivity demonstrate the existence of two separate translocators for proline in rat kidney mitochondria, i.e., the proline uniporter and the proline/glutamate antiporter. Efflux of glutamate via glutamate/OH- translocator following proline uptake by mitochondria was experimentally ruled out. Discussion is also made of the possible role of such translocators in proline metabolism and in the putative proline/glutamate shuttle.

Hydroxyproline uptake by rat kidney mitochondria is here first shown by monitoring the reduction of the intramitochondrial pyridine nucleotides which occurs as a result of metabolism of imported hydroxyproline via hydroxyproline oxidase and 3-hydroxy-pyrroline-5-carboxylate dehydrogenase. Widely used criteria for demonstrating the occurrence of carrier-mediated transport were applied to this process. Hydroxyproline uptake shows saturation features (Km and Vmax values, measured at 20 degrees C and at pH 7.20, were found to be about 1.4 mM and 5 nmoles/min x mg mitochondrial protein, respectively) and proves to be inhibited by the impermeable compound phenylsuccinate, but insensitive to externally added methylglutamate. Difference found in the Km and Vmax values, a different inhibitor sensitivity and the failure of hydroxyproline to cause efflux of glutamate from the mitochondria show that hydroxyproline enters mitochondria by means of a translocator different from those which transport proline. The effect of hypothyroidism on kinetic characteristics of cytochrome oxidase in rat heart mitochondria was studied. Mitochondrial preparations from control and hypothyroid rats had equivalent Km values for cytochrome c, while the maximal activity of the oxidase was significantly decreased (more than 30%) in mitochondrial preparations from hypothyroid rats. This decrease is associated to a parallel decrease in state 3 respiration. The cytochrome aa3 content was slightly decreased (by around 15%) in mitochondria from hypothyroid rats. The Arrhenius plot characteristics differ for cytochrome oxidase activity in mitochondria from hypothyroid rats as compared with control rats in that the breakpoint of the biphasic plot is shifted to a higher temperature. Cardiolipin content was markedly decreased in the mitochondrial membrane from hypothyroid rats. No alterations were found in the pattern of cardiolipin fatty acid distribution of mitochondrial membrane from control and hypothyroid rats. The effects of the hypothyroid state on the activity of cytochrome oxidase, on cytochrome aa3 levels, and on cardiolipin contents were completely reversed by following the treatment of hypothyroid rats with thyroid hormone. The results support the conclusion that the depressed mitochondrial cytochrome oxidase activity in the hypothyroid state is due, at least in part, to a decrease in the cardiolipin content of the mitochondrial inner membrane.

Annexin p34 spin label has been used as a probe for studying annexin-membrane Ca-mediated interaction. The EPR spectrum of the purified probe is able to monitor the binding of the protein to phospholipid vesicles and isolated mitochondrial membranes, with increasing calcium concentration, as revealed by the calculated rotational correlation time. A novel mathematical approach is proposed to provide a direct estimation of annexin percentage binding to membranes, based on the correlation time calculated from experimental EPR spectra.

To gain some insight into the mechanism by which red light-biosystem interaction occurs, an investigation was made of certain features of purified glutamate dehydrogenase from beef liver (E.C. 1.4.1.3.) irradiated with either an HeNe laser (632.8 nm) or a red light-emitting diode (650±20 nm). In both cases the energy dose was 0.24 J cm −2 . Significant changes in the glutamate dehydrogenase extinction coefficient measured at 275 nm, the capability of the enzyme to bind the reduced form of nicotinamide adenine dinucleotide (NADH), certain kinetic parameters, the pH and temperature dependence and the sensitivity to guanosine 5 triphosphate (GTP) and adenosine diphosphate (ADP) were found, probably due to the interaction of lightwith a protein domain containing a metal ion or ions. HeNe laser and diode irradiation were found to differ with regard to their interaction with glutamate dehydrogenase. Interestingly, different effects were also found when an HeNe laser and a non-coherent XeHg lamp were used to irradiate glutamate dehydrogenase under the same experimental conditions. This confirms that non-coherent light at various power levels affects the isolated glutamate dehydrogenase.

Evidence is given in this paper that externally added flavin mononucleotide (FMN) induces flavin adenine dinucleotide synthesis in isolated rat liver mitochondria, as shown by fluorimetric, chromatographic, and enzymatic assays. FMN association with mitochondria is a process with hyperbolic features that is sensitive to externally added mersalyl.

Sodium chloride extracts obtained from purified bovine brain myelin were found to contain proteolytic activity capable of degrading isolated myelin basic protein as assessed by SDS gel electrophoresis. Using gels copolymerized with gelatin as substrate, two bands at about 54 and 117-125 KDa, respectively, were detected. Activity corresponding to the 54 KDa band was inhibited by zinc. Data presented in this article suggest that proteolytic activity can be released from the myelin sheath in water-soluble form and recognize MBP as substrate.

We have used the initiation-promotion model of MNU-induced hepatocarcinogenesis to test the hypothesis that alteration of the methylation status of DNA cytosines could be involved in the initiation of carcinogenesis. In fact cell proliferation plays a fundamental role in the initiation of liver carcinogenesis and hepatocytes in the S phase are more sensitive towards MNU initiation than at other times in the cycle. The molecular mechanisms involved in these processes, however, are still poorly understood and it seemed of value to monitor the DNA methylation status in this system. The results obtained indicate that MNU hepatocarcinogenic action might consist also of the inhibition of DNA hypomethylation biologically associated with cell proliferation.

The effect of aging and treatment with acetyl-L-carnitine on the activity of the phosphate carrier and on the phospholipid composition in rat heart mitochondria was studied. It was found that the activity of the phosphate carrier was reduced by aging. Treatment of aged rats with acetyl-L-carnitine reversed this effect. The mitochondrial level of cardiolipin was decreased with aging. Treatment of aged rats with acetyl-L-carnitine restored the level of cardiolipin to that of young rats. It is proposed that acetyl-L-carnitine may restore the correct phospholipid composition (cardiolipin level) of the mitochondrial membrane, altered by aging, thereby restoring the activity of the phosphate carrier.

An experiment was performed to isolate the small atypical mitochondria produced during the irradiation of normal mitochondria with an He-Ne laser. Rat liver mitochondria were irradiated with a low-power continuous-wave He-Ne laser (energy dose, 5 J cm-2), followed by isolation using a sucrose gradient. In the irradiated sample, two bands were observed, one corresponding to normal mitochondria and the other to atypical mitochondria. Certain biochemical features of the mitochondria were investigated: mitochondrial enzyme activity and the presence of DNA and RNA were demonstrated. Hybridization experiments carried out with labelled mitochondrial probes, containing the genes for cytochrome oxidase subunit I and 12S rRNA, confirmed the mitochondrial nature of the isolated RNA.

In the present study we have examined the effect of a single dose of the mitogen lead nitrate (75 μmol/kg body wt) on the methylation status of hepatic DNA in male Wistar rats. It was found that extensive hypomethylation of hepatic DNA occurs in mitogen-treated rat liver. This effect could be seen as early as 12 h after metal treatment and parallels the changes in liver weight. Probing with the methylation-sensitive enzymes Hpa II, Msp I, and Hae III confirmed HPLC analyses and showed that methylation at these sites was affected by lead treatment. DNA hypomethylation has already been found in regenerating rat liver and in hepatic (pre)malignant lesions when compared to normal nondividing liver. Thus the lowering of the DNA 5-methylcytosine content appears to be a property characteristic of cellular proliferation, regardless of whether it is caused by partial hepatectomy, carcinogen treatments, or mitogen administration.

To gain further insight into the ability of Photofrin II to photosensitize mitochondrial translocators, and to ascertain whether mitochondrial thiols are specific targets of Photofrin II, the activity of phosphate carrier was measured in isolated rat liver mitochondria irradiated with 365 nm light in the presence of Photofrin II. Photodynamic treatment decreased the maximum rate of phosphate uptake, without changing the phosphate affinity for its own carrier. The ability of the thiol reagent mersalyl (an inhibitor of phosphate, dicarboxylate and oxodicarboxylate carriers) to protect these carriers against Photofrin II photosensitization was also tested. Protection was observed, indicating the involvement of carrier thiols in mitochondrial photosensitization.

Bovine lung annexins p32 and p34 were spin labeled with an iodoacetamidoproxyl spin label, a reagent that reportedly couples with protein methionine residues. Labeling conditions and stoichiometry were studied with the radiolabeled analogue [1-14C]iodoacetamide. As judged by this method, carboxamidomethylation of both p32 and p34 occurred up to a 0.7 mol ratio after 60 h of reaction at 37 degrees C and at pH 4. The two proteins retained Ca2(+)-dependent phospholipid-binding ability both in radiolabeled and in spin-labeled forms. Electron resonance spectra of spin-labeled p32 and p34 showed the features of a partially immobilized spin probe, with rotational correlation time values of 1.15 and 1.25 ns, respectively, which definitely indicate successful spin labeling. Quantitation of ESR spectra by computer double integration indicated 70% spin labeling of both proteins, as anticipated by radiolabeling. The use of spin-labeled p32 and p34 in the study of Ca2(+)-dependent interaction of annexins with biomembranes is proposed.

The fact that thiamine pyrophosphate is synthesized in cytosol necessitates its uptake into mitochondria. The ability of mitochondria to take up externally added thiamine pyrophosphate was investigated by measuring the intramitochondrial thiamine pyrophosphate content using an enzymatic method. Thiamine pyrophosphate uptake by isolated rat liver mitochondria was found to occur in a time- and temperature-dependent manner. Uptake shows saturation characteristics with Km and Vmax values equal to about 20 μ m and 700 pmol/min × mg protein, respectively, and is inhibited by certain nonpenetrating compounds. The inhibition of thiamine uptake by thiamine pyrophosphate and the efflux of endogenous thiamine pyrophosphate, caused by externally added thiamine, suggest the existence of a thiamine pyrophosphate/thiamine antiporter which could play an active role in the turnover of intramitochondrial thiamine pyrophosphate linked enzymes.

The effect of aging and treatment with acetyl-L-carnitine on the lipid composition of rat plasma and erythrocytes was studied. It was found that aging increases the levels of free and esterified cholesterol. Fatty acid patterns in the plasma of aged rats show remarkable alterations when compared with control rats. These changes reverted to normal after three hours of acetyl-L-carnitine treatment. No significant differences in the erythrocyte lipid pattern of young and aged rats were observed. This study provides the first proof that acetyl-L-carnitine probably acts by lowering free and esterified cholesterol and arachidonic acid (20:4) levels in the plasma.

Incubation of intact mitochondria with aspartate aminotransferase results in efflux of malate dehydrogenase and vice versa. The export process is specific and rapid. It shows saturation kinetics with respect to the effector enzyme consistent with involvement of a receptor for the effector in the mitochondrial membrane system. Export is inhibited by both ?-mercaptoethanol and by the metal chelating agent bathophenanthroline; both substances inhibit release of malate dehydrogenase by aspartate aminotransferase competitively whereas for release of aspartate aminotransferase by malate dehydrogenase inhibition is non-competitive. The efflux process is dependent on a trans-membrane pH gradient. Exported enzymes differ from the native forms in their dependence of activity on pH. Export of both aspartate aminotransferase and malate dehydrogenase is effected by incubation of mitochondria with the newly-synthesised precursor of aspartate aminotransferase; this observation provides supporting evidence for the physiological significance of the other results reported here. It is speculated that exported enzymes are on a pathway to degradation, and that coupled uptake and export is involved in the co-ordination of synthesis and breakdown of mitochondrial proteins. © 1990 Elsevier Science Publishers B.V. Within 4 h following the addition of 3,3′,5 triiodo-L-thyronine to monolayer cultures of hepatocytes isolated from hypothyroid rats, a very distinct stimulation of fatty acid and cholesterol synthesis, measured as incorporation of either [1-14C]acetate or [3H]H2O into these lipid fractions, is observed. A smaller but significant increase in the rate of lipogenesis occurs in hepatocytes derived from euthyroid animals. These stimulatory effects of triiodothyronine are also observed in the presence of cycloheximide, indicating that the described early and direct stimulation of lipogenesis by the thyroid hormone is, at least in part, independent of protein synthesis.

In the present study we have investigated the transfer of phospholipids between vesicles and rat liver mitochondria. Transfer was measured by electron paramagnetic resonance spectroscopy using vesicles that contained spin-labeled phospholipids. A spontaneous transfer was observed which could be strongly inhibited by treating the mitochondria with the thiol reagent mersalyl. Transfer was also greatly reduced after a saline wash of the mitochondria; the transfer activity was then recovered in the wash. This activity was inhibited by tryptic digestion and mersalyl. By gel chromatography, enzyme immunoassay and immunoblotting it was demonstrated that the activity in the wash was due to the nonspecific lipid transfer protein (sterol carrier protein 2). We could estimate that up to 85% of the spontaneous phospholipid transfer between vesicles and rat liver mitochondria was mediated by this transfer protein.

Myelin basic protein isolated by a single step with the cationic detergent cethyltrimethylammonium bromide in a lipid-bound form is able to induce structural transition of lysophosphatydilcholine micelles into multilaminar vesicles. This finding, observed through electron microscopy, is discussed in the light of the assumed ability of the basic protein to organize myelin lipids.

Fatty acid synthetic capacity, investigated both in subcellular fractions and in vivo, is very active in brown adipose tissue of room temperature-acclimated rats. In hyperthyroid animals this tissue, analogously to the liver, exhibits an increased activity of acetyl-CoA carboxylase, fatty acid synthetase and microsomal fatty acid chain elongation, this last mechanism remaining unaffected in mitochondria. An enhancement of reducing capacities of a group of cytoplasmic NADP-dependent enzymes has also been observed in brown adipose tissue of hyperthyroid rats, probably due to a greater use of NADPH in lipogenesis under these conditions. An increase in palmitate oxidation and in polyenoic fatty acids was observed in mitochondria of brown adipose tissue from hyperthyroid animals. The latter increase is related to the importance of these compounds in the regulation of membrane fluidity and probably to an increased resistance to cold in the hyperthyroid state.

The transport of citric cycle intermediates from cytosol to the mitochondrial matrix in rat liver mitochondria was studied in order to explain transport mechanism and molecular structure of the carriers. Anion, pH and cation influence on dicarboxylate, tricarboxylate and oxoglutarate carrier activities was examined kinetically and metal complexing agents like bathophenanthroline were used as an investigation tool for molecular structure of the carrier. The following conclusions were reached: 1. The substrates bind themselves on the mitochondrial membrane to a metal ion present in carrier molecules. 2. The primary effect of pH on the initial rate of uptake is due to a rise of ΔpH across the mitochondrial membrane. 3. Membrane surface, surrounded by the carrier environment, is negatively charged, and general cation stimulation of the rate of uptake is due to the positive electric charge which neutralizes amphoteric negatively charge groups of the membrane.

Photoacoustic spectroscopy measurements reveal that HeNe low-power irradiation causes changes in mitochondrial optical properties. However, it has been found that this process damages neither the membrane of the mitochondria nor some of their enzymatic activities. The HeNe laser irradiation effect was investigated both with regard to the redox state change of intramitochondrial pyridine nucleotides, induced by addition to the mitochondria of oxaloacetate, oxoglutarate plus ammonia and cis-aconitate, and to the permeability of rat liver mitochondria to the same substrates. It was found that laser irradiation modifies some NADH-linked dehydrogenase reactions of the mitochondria.
