Abstract Background Phenotypic evolution is mainly explained by selection for phenotypic variation arising from factors including mutation and environmental noise. Recent theoretical and experimental studies have suggested that phenotypes with greater developmental stability tend to have a constant phenotype and gene expression level within a particular genetic and environmental condition, and this positively correlates with stronger evolutionary conservation, even after the accumulation of genetic changes. This could reflect a novel mechanism that contributes to evolutionary conservation; however, it remains unclear whether developmental stability is the cause, or whether at least it contributes to their evolutionary conservation. Here, using Japanese medaka lines, we tested experimentally whether developmental stages and gene expression levels with greater stability led to their evolutionary conservation. Results We first measured the stability of each gene expression level and developmental stage (defined here as the whole embryonic transcriptome) in the inbred F0 medaka population. We then measured their evolutionary conservation in the F3 generation by crossing the F0 line with the distantly related Japanese medaka line (Teradomori), followed by two rounds of intra-generational crossings. The results indicated that the genes and developmental stages that had smaller variations in the F0 generation showed lower diversity in the hybrid F3 generation, which implies a causal relationship between stability and evolutionary conservation. Conclusions These findings suggest that the stability in phenotypes, including the developmental stages and gene expression levels, leads to their evolutionary conservation; this most likely occurs due to their low potential to generate phenotypic variation. In addition, since the highly stable developmental stages match with the body-plan-establishment stage, it also implies that the developmental stability potentially contributed to the strict conservation of animal body plan.

Simple Summary: Size and shape are important features of most living beings, and the capacity of the organism to regulate size and shape during growth, containing the effects of developmental disturbances of different origins, is considered a key adaptation. Through a morphological study on the larvae of the cabbage butterfly Pieris brassicae, reared in conditions close to the natural environment, we found that the regulatory mechanisms for limiting the effects of developmental disturbances, known to be effective under strictly controlled, very stable laboratory conditions, are also effective during growth under more natural environmental conditions. Abstract Background Despite the morphological diversity of animals, their basic anatomical patterns—the body plans in each animal phylum—have remained highly conserved over hundreds of millions of evolutionary years. This is attributed to conservation of the body plan-establishing developmental period (the phylotypic period) in each lineage. However, the evolutionary mechanism behind this phylotypic period conservation remains under debate. A variety of hypotheses based on the concept of modern synthesis have been proposed, such as negative selection in the phylotypic period through its vulnerability to embryonic lethality. Here we tested a new hypothesis that the phylotypic period is developmentally stable; it has less potential to produce phenotypic variations than the other stages, and this has most likely led to the evolutionary conservation of body plans. Results By analyzing the embryos of inbred Japanese medaka embryos raised under the same laboratory conditions and measuring the whole embryonic transcriptome as a phenotype, we found that the phylotypic period has greater developmental stability than other stages. Comparison of phenotypic differences between two wild medaka populations indicated that the phylotypic period and its genes in this period remained less variational, even after environmental and mutational modifications accumulated during intraspecies evolution. Genes with stable expression levels were enriched with those involved in cell-cell signalling and morphological specification such as Wnt and Hox, implying possible involvement in body plan development of these genes. Conclusions This study demonstrated the correspondence between the developmental stage with low potential to produce phenotypic variations and that with low diversity in micro- and macroevolution, namely the phylotypic period. Whereas modern synthesis explains evolution as a process of shaping of phenotypic variations caused by mutations, our results highlight the possibility that phenotypic variations are readily limited by the intrinsic nature of organisms, namely developmental stability, thus biasing evolutionary outcomes.

Premise of research. Cotyledons have important functions in the early seedling stage and have important effects on later stages, but we know little about the relationships among developmental stability, canalization, and phenotypic plasticity in cotyledons. Methodology. We conducted a field experiment with five herbaceous species, subjecting them to contrasting light conditions and burial depths and measuring their cotyledon size and fluctuating asymmetry (random deviation from perfect bilateral symmetry, indicating developmental stability or instability), coefficient of variation, and plasticity of cotyledon size, to investigate the relationships among cotyledon developmental stability, canalization, and plasticity in response to shading and deep burial. Pivotal results. Pharbitis purpurea , Convolvulus arvensis , and Carpesium abrotanoides had increased cotyledon size in response to shading at both burial depths, and Abutilon theophrasti showed reduced cotyledon size in response to shading versus full light at shallow depth but greater cotyledon size in response to both shading and deep burial. Shading increased cotyledon fluctuating asymmetry of P. purpurea and C. abrotanoides , while deep burial decreased it. Cotyledon fluctuating asymmetry had positive correlations with coefficient of variation and plasticity in response to shading, with little correlation between coefficient of variation and plasticity. Conclusions. Results suggested that A. theophrasti may have greater tolerance for multiple stresses than the other species, and deep burial may improve shade tolerance of cotyledons through moderate levels of stress selection. Both developmental instability and decreased canalization may indicate the state of faster growth. Background: We compared skull shape and variation among genetically modified mice that exhibit different levels of connexin43 (Cx43) channel function, to determine whether Cx43 contributes to craniofacial phenotypic robustness. Specifically, we used two heterozygous mutant mouse models (G60S/+ and I130T/+) that, when compared to their wildtype counterparts, have an ~80% and ~50% reduction in Cx43 function, respectively. Results: Both mutant strains showed significant differences in skull shape compared to wildtype littermates and while these differences were more severe in the G60S/+ mouse, shape differences were localized to similar regions of the skull in both mutants. However, increased skull shape variation was observed in G60S/+ mutants only. Additionally, covariation of skull structures was disrupted in the G60S/+ mutants only, indicating that while a 50% reduction in Cx43 function is sufficient to cause a shift in mean skull shape, the threshold for Cx43 function for disrupting craniofacial phenotypic robustness is lower. Conclusions: Collectively, our results indicate Cx43 can contribute to phenotypic robustness of the skull through a nonlinear relationship between Cx43 gap junctional function and phenotypic outcomes. Key Findings: The G60S/+ and I130T/+ mutations both result in a shift in mean skull shape of newborn and 3‐month mice.The differences in skull shape are similarly localized in both mutant mice indicating that the mutations target the same processes of skull development and homeostasis.Cx43 can modulate phenotypic variation of the skull, but only if protein function is reduced beyond a relatively permissive threshold. Within-plant variation in seed size may merely reflect developmental instability, or it may be adaptive in facilitating diversifying bet-hedging, that is, production of phenotypically diverse offspring when future environments are unpredictable. To test the latter hypothesis, we analyzed patterns of variation in seed size in 11 populations of the perennial vine Dalechampia scandens grown in a common greenhouse environment. We tested whether population differences in the mean and variation of seed size covaried with environmental predictability at two different timescales. We also tested whether within-plant variation in seed size was correlated with independent measures of floral developmental instability and increased under stressful conditions. Populations differed genetically in the amount of seed-size variation occurring among plants, among infructescences within plants, and among seeds within infructescences. Within-individual variation was not detectably correlated with measures of developmental instability and did not increase under stress, but it increased weakly with short-term environmental unpredictability of precipitation at the source-population site. The genetic architecture of skull shape has been extensively studied in mice and the results suggest a highly polygenic and additive basis. In contrast few studies have explored the genetic basis of the skull variability. Canalization and developmental stability are the two components of phenotypic robustness. They have been proposed to be emergent properties of the genetic networks underlying the development of the trait itself, but this hypothesis has been rarely tested empirically. Here we use outbred mice to investigate the genetic architecture of canalization of the skull shape by implementing a genome-wide marginal epistatic test on 3D geometric morphometric data. The same data set had been used previously to explore the genetic architecture of the skull mean shape and its developmental stability. Here, we address two questions: (1) Are changes in mean shape and changes in shape variance associated with the same genomic regions? and (2) Do canalization and developmental stability rely on the same loci and genetic architecture and do they involve the same patterns of shape variation? We found that unlike skull mean shape, among-individual shape variance and fluctuating asymmetry (FA) show a total lack of additive effects. They are both associated with complex networks of epistatic interactions involving many genes (protein-coding and regulatory elements). Remarkably, none of the genomic loci affecting mean shape contribute these networks despite their enrichment for genes involved in craniofacial variation and diseases. We also found that the patterns of shape FA and individual variation are largely similar and rely on similar multilocus epistatic genetic networks, suggesting that the processes channeling variation within and among individuals are largely common. However, the loci involved in these two networks are completely different. This in turn underlines the difference in the origin of the variation at these two levels, and points at buffering processes that may be specific to each level.

Using the centipede model species Strigamia maritima as a subject of study, we illustrate the potential of geometric morphometrics for investigating the development and evolution of segmentation, with a specific focus on post-embryonic segmental patterning. We show how these techniques can contribute detailed descriptive data for comparative purposes, but also precious information on some features of the developmental system that are considered relevant for the evolvability of a segmented body architecture, such as developmental stability and canalization. Morphometric analyses allow to separately investigate several sources of phenotypic variation along a segmented body axis, like constitutive and random segment heteronomy, both within and among individuals. Specifically, in S. maritima , the segmental pattern of ventral sclerite shapes mirrors that of their bilateral fluctuating asymmetry and among-individual variation in associating the most anterior and most posterior segments in diverging from the central ones. Also, among segments, there seems to be a correlation between fluctuating asymmetry and shape variation among individuals, suggesting that canalization and developmental stability are somehow associated. Developmental stability and canalization describe the ability of developmental systems to minimize phenotypic variation in the face of stochastic micro-environmental effects, genetic variation and environmental influences. Canalization is the ability to minimize the effects of genetic or environmental effects, whereas developmental stability is the ability to minimize the effects of micro-environmental effects within individuals. Despite much attention, the mechanisms that underlie these two components of phenotypic robustness remain unknown. We investigated the genetic structure of phenotypic robustness in the collaborative cross ( CC) mouse reference population. We analysed the magnitude of fluctuating asymmetry ( FA) and among-individual variation of cranial shape in reciprocal crosses among the eight parental strains, using geometric morphometrics and a diallel analysis based on a Bayesian approach. Significant differences among genotypes were found for both measures, although they were poorly correlated at the level of individuals. An overall positive effect of inbreeding was found for both components of variation. The strain CAST/EiJ exerted a positive additive effect on FA and, to a lesser extent, among-individual variance. Sex- and other strain-specific effects were not significant. Neither FA nor among-individual variation was associated with phenotypic extremeness. Our results support the existence of genetic variation for both developmental stability and canalization. This finding is important because robustness is a key feature of developmental systems. Our finding that robustness is not related to phenotypic extremeness is consistent with theoretical work that suggests that its relationship to stabilizing selection is not straightforward. Abstract: Objective: Tooth formation normally progresses symmetrically between sides; the goal in this study was to test the clinical impression that left–right asymmetry in tooth formation is elevated in children with simple hypodontia. Materials and methods: Data from panoramic X-rays of American white children (5–14 years of age) with simple hypodontia (n =158) were compared to a comparable group from the same practises with all teeth present (n =206). Children with hypodontia were otherwise phenotypically normal, with no cleft or recognized syndrome. Crown–root formation of each tooth (ignoring third molars) was scored using an 11-grade scheme. Analysis relied on chi-square goodness-of-fit tests and odds ratios. Results: Hypodontia typically occurs unilaterally; it is more common in girls than boys; and it most frequently affects second premolars (omitting third molars). No evidence of a side preference was found as regards absence of the tooth or tooth formation. Tooth formation was decidedly more frequently asymmetric in those with hypodontia, though again the distribution by side was random in the sample. Summed over all tooth types, asymmetric formation occurred in 18.6% of cases with hypodontia compared to 11.9% in controls, and this is significant by chi-square (P =0.03), with an odds ratio of 1.43 (CL: 1.02, 2.04). All tooth types exhibit elevated developmental asymmetry in the hypodontic sample despite only one or a few teeth being agenic. Conclusions: Increased asymmetry suggests a breakdown in the rigour of developmental timing in cases with simple hypodontia. In concert with increased frequencies of other growth issues in such cases, such as side differences in size and morphology, hypodontia is best viewed as a symptom of an anatomically broad relaxation of developmental canalization between homologous structures, not an isolated dental feature. B chromosomes (Bs) are supernumerary chromosomes relative to the standard karyotype. The maintenance of Bs in the yellow-necked field mice (Apodemus flavicollis) was reconsidered by examining their effects on 3 components of cranial variability: canalization, developmental stability, and morphological integration. Bs do not disturb developmental homeostasis in their carriers. Moreover, Bs play a significant role in structuring cranial variation. We suggest that direct interactions between developmental pathways in mice without Bs might be a dominant mechanism for generating covariation of cranial traits, and integration of cranial traits in B carriers could be generated primarily by parallel variation of separate developmental pathways. Integration due to parallel variation is more predisposed to modifications by natural selection than integration caused by direct interactions, which could be beneficial to B carriers under variable environmental conditions. By contributing to the genetic variability of species possessing them, Bs provide themselves with long-term presence in populations. Environmental stress has been suggested to be a major evolutionary force, both through inducing strong selection and because of its direct impact on developmental buffering processes that alter the evolvability of organisms. In particular, temperature has attracted much attention because of its importance as an ecological feature and the relative ease with which it can be experimentally manipulated in the lab. Evolution Canyon, Lower Nahal Oren, Israel, is a well studied natural site where ecological parameters are suspected to drive evolutionary differentiation. In this study, using Drosophila melanogaster isofemale lines derived from wild flies collected on both slopes of the canyon, we investigated the effect of developmental temperature upon the different components of phenotypic variation of a complex trait: the wing. Combining geometric and traditional morphometrics, we find only limited evidence for a differentiation among slopes. Investigating simultaneously phenotypic plasticity, genetic variation among isofemale lines, variation among individuals and fluctuating asymmetry, we could not identify a consistent effect of the stressful conditions encountered on the south facing slope. The prevailing structuring effect is that of the experimentally manipulated temperature which clearly influences wing mean size and shape. Variability, in contrast, is not consistently affected by temperature. Finally, we investigated the specific relationship between individual variation and fluctuating asymmetry. Using metric multi-dimensional scaling we show that the related patterns of wing shape variation are not identical, supporting the view that the underlying developmental processes are to a certain extent different. Variability contrasts with variation in that variability describes the potential for variation, not simply the expressed variation. The power of studying variability lies in creating a conceptual framework around which the relationship between the genotype and phenotype can be understood. Here, we attempt to demonstrate the importance of phenotypic variability, how it structures variation, and how fundamental developmental processes structure variability. Given the broad scope of this topic, we focus on three widely studied properties of variability: canalization, developmental stability and morphological integration. We have organized the paper to emphasize the importance of differentiating between the theory surrounding these components of phenotypic variability, their measurement and the biological factors surrounding their expression. First, we define these properties of variability, how they relate to each other and to variability as a whole. Second, we summarize the common methods of measurement for canalization, developmental stability and morphological integration and the reasoning behind these methods. Finally, we focus on jaw development as an example of how the basic processes of development affect variability and the resultant variation, with emphasis on how processes at all levels of the organismal hierarchy interact with one another and contribute to phenotypic variability. The molecular chaperone protein Hsp90 has been widely discussed as a candidate gene for developmental buffering. We used the methods of geometric morphometrics to analyze its effects on the variation among individuals and fluctuating asymmetry of wing shape in Drosophila melanogaster. Three different experimental approaches were used to reduce Hsp90 activity. In the first experiment, developing larvae were reared in food containing a specific inhibitor of Hsp90, geldanamycin, but neither individual variation nor fluctuating asymmetry was altered. Two further experiments generated lines of genetically identical flies carrying mutations of Hsp83, the gene encoding the Hsp90 protein, in heterozygous condition in nine different genetic backgrounds. The first of these, introducing entire chromosomes carrying either of two Hsp83 mutations, did not increase shape variation or asymmetry over a wild-type control in any of the nine genetic backgrounds. In contrast, the third experiment, in which one of these Hsp83 alleles was introgressed into the wild-type background that served as the control, induced an increase in both individual variation and fluctuating asymmetry within each of the nine genetic backgrounds. No effect of Hsp90 on the difference among lines was detected, providing no evidence for cryptic genetic variation of wing shape. We tested whether directional selection on an index-based wing character in Drosophila melanogaster affected developmental stability and patterns of directional asymmetry. We selected for both an increase (up selection) and a decrease (down selection) of the index value on the left wing and compared patterns of fluctuating and directional asymmetry in the selection index and other wing traits across selection lines. Changes in fluctuating asymmetry across selection lines were predominantly small, but we observed a tendency for fluctuating asymmetry to decrease in the up-selected lines in both replicates. Because changes in fluctuating asymmetry depended on the direction of selection, and were not related to changes in trait size, these results fail to support existing hypotheses linking directional selection and developmental stability. Selection also produced a pattern of directional asymmetry that was similar in all selected lines whatever the direction of selection. Hybrids from crosses of different species have been reported to display decreased developmental stability when compared to their pure species, which is conventionally attributed to a breakdown of coadapted gane complexes. Drosophila subobscura and its close relative D, madeirensis were hybridized in the laboratory to test the hypothesis that genuine fluctuating asymmetry, measured as the within-individual variance between right and left wings that results from random perturbations in development, would significantly increase after interspecific hybridization. When sires of D. subobscura were mated to heterospecific females following a hybrid half-sib breeding design, F1 hybrid females showed a large bilateral asymmetry with a substantial proportion of individuals having an asymmetric index larger than 5% of total wing size. Such an anomaly, however, cannot be plainly explained by an increase of developmantal instability in hybrids but is the result of some aberrant developmental processes. Our findings suggest that interspecific hybrids are as able as their parents to buffer developmental noise, notwithstanding the fact that their proper bilateral development can be harshly compromised. Together with the low correspondence between the covariation structures of the interindividual genetic components and the within-individual ones from a Procrustes analysis, our data also suggest that the underlying processes that control (genetic) canalization and developmental stability do not share a common mechanism. Among the explanations for minimizing the effects of extraneous variation has been canalization and developmental stability. However, there is little agreement as to whether these two processes reflect a common set of mechanisms. This confusion is inflated due to the lack of consensus as to a precise definition of canalization. In this study, canalization in the sternopleural bristle system is used to investigate the relationships between measures of canalization and developmental stability by comparing how a panel of naturally derived lines responds to both genetic and environmental perturbations. No evidence for a common mechanism between the different measures of canalization was observed. Furthermore, a hypothesis regarding a common mechanism for environmental and genetic canalization was tested, and no evidence was found to support it. A/WySnJ mice are an inbred strain that develops cleft lip with or without cleft palate (CL/P) with a frequency of 25–30% and a predominantly unilateral expression pattern. As in humans, the pattern of incomplete penetrance, and variable and frequent unilateral expression suggests a role for altered regulation of variability (developmental stability, canalization and developmental integration) during growth. We compared both mean and variability parameters for craniofacial shape and size among A/WySnJ mice, a strain that does not develop CL/P (C57BL/6J) and their F1 cross. We show that adult A/WySnJ mice that do not express cleft lip exhibit decreased morphological integration of the cranium and that the co-ordination of overall shape and size variation is disrupted compared with both C57BL/6J mice and the F1 cross. The decrease in integration is most pronounced in the palate and face. The absence of this pattern in the F1 cross suggests that it is determined by recessive genetic factors. By contrast, the shape differences between the strains, which are thought to predispose A/WySnJ mice to CL/P, show a range of dominance which suggests a polygenic basis. Abstract Fluctuating asymmetry (FA) is frequently used as a measure of developmental instability (DI). Assuming a genetic basis to DI, many have argued that FA may be a good indicator of genetic quality to potential mates and to human managers of populations. Unfortunately FA is a poor indicator of DI, making it very difficult to verify this assertion. A recent review of the literature suggests that previous studies of the inheritance of FA and DI using half-sib covariances and parent–offspring regression have been unable to put meaningful limits on the heritability of FA and DI because of the extremely low power of the experiments performed. In this study, we consider the power of artificial selection on FA as an alternative approach to studying the inheritance of FA and DI. Using simulations, we investigate the efficacy of selection for both increased and decreased FA for detecting genetic variation. We find that selection for increased FA has much more power to detect the presence of genetic variation than does selection for decreased FA. These results hold when realistic sample sizes are employed. Artificial selection for increased FA is currently the most powerful approach for the detection of genetic variation in DI.
