This work reported a simple and ultrasensitive label-free method for the detection of hepatitis B virus (HBV) DNA by combining hyperbranched rolling circle amplification (HRCA) with dual-mode detection by inductively coupled plasma mass spectrometry (ICP-MS) and fluorescence using ruthenium complex [Ru(bpy)2dppz]2+ (bpy = 2,2'-bipyridine, dppz = dipyrido [3,2-a:2',3'-c] phenazine) as a dual functional probe. An HBV DNA-initiated HRCA system was designed to realize the highly efficient amplification of HBV DNA with the generation of a mass of dsDNA. Also, the [Ru(bpy)2dppz]2+ probe was then added to intercalate into the dsDNA products, resulting in strong fluorescence recovery of the probe for fluorescence detection. Meanwhile, using a biotin-modified primer in HRCA, the dsDNA-[Ru(bpy)2dppz]2+ complexes could be captured by the avidin-coated 96-well plates, and the captured [Ru(bpy)2dppz]2+ probe was later desorbed by acid for ICP-MS detection. The linear range of the proposed method was 3.5-200 amol L-1 and the limit of detection (LOD) was 1 amol L-1 for ICP-MS detection, while the linear range was 20-500 amol L-1 and the LOD was 9.6 amol L-1 for fluorescence detection. The developed method was applied to human serum sample analysis, and the analytical results coincided very well with those obtained by the real-time polymerase chain reaction (PCR) method. The developed dual-mode label-free detection method was ultrasensitive, simple, and accurate, showing great potential for therapeutic monitoring of HBV infection.

Studies show that two dimensional (2D) nanomaterial and its hybrid have a great promise in tribology for the special laminar microstructure. However, the majority of performed investigations about 2D graphitic carbon nitride (g-C3N4) nanosheets are most focused on energy storage, catalysis, adsorption, rarely tribology. In the present study, g-C3N4 supporting mono-dispersed Ag nanoparticle hybrid (g-C3N4/Ag) is prepared, and its microstructure and chemical composition are determined. More specifically, the tribological performance as the lubricating additive of poly phthalazinone ether sulfone ketone (PPESK) composite is investigated using the ball-on-disc friction tester. Moreover, the corresponding enhancement mechanism is well proposed based on the experimental analysis and theoretical simulation. Obtained results show that Ag nanoparticles with a size of about 10–20 nm are homogeneously anchored on g-C3N4 nanosheets, favoring for good compatibility between g-C3N4/Ag and PPESK. It is found that when 0.3 wt% of g-C3N4/Ag is added to PPESK, the friction coefficient and wear rate of PPESK decrease by 68.9% and 97.1%, respectively. These reductions are mainly attributed to the synergistic self-lubricating effect of Ag nanoparticles and g-C3N4 nanosheet, the formation of transfer film, as well as the limited effect of g-C3N4/Ag on the shear deformation of PPESK composite film. Furthermore, it is found that the proposed g-C3N4/Ag-PPESK composite still keeps reasonable friction-reducing and wear-resistant properties under heavy loads and high rotating speeds. The present study demonstrates that the proposed g-C3N4/Ag hybrid is an excellent lubricating additive for polymer composites.

It has been well established that the early-stage interactions of nanoparticles with cells are governed by the extracellular protein corona. However, after entering into the cells, the evolving protein corona is the key to subsequent processing of nanoparticles by cells. To identify the protein corona around intracellular nanoparticles, it is essential to maintain its original compositions during cell treatment. Herein, we develop a paraformaldehyde (PFA) cross-linking strategy to stabilize corona compositions when extracting protein coronas from cells, providing original information on protein coronas around intercellular gold nanoparticles (AuNPs). The stability of the protein corona after PFA cross-linking was carefully investigated with several characterization methods, and the results demonstrate that PFA cross-linking successfully prevents the dissociation and exchange of corona proteins. Then the recovered intracellular protein corona around AuNPs from living HepG2 cells with a PFA cross-linking strategy was subjected to nanoHPLC-MS/MS for proteomic analysis. It was found that the compositions of intracellular protein coronas are dominated by cell-derived proteins and undergo significant variation of protein species and amounts over time during internalization. Time-resolved analysis provides relevant proteins involved in nanoparticle cellular uptake and transportation, indicating that AuNPs are endocytosed mainly by a clathrin-mediated uptake mechanism and directed into an endolysosomal pathway toward their final destination. Such proteomic-based results are verified by pharmacological inhibition and TEM imaging analysis. This work provides a universal strategy to study compositions of protein corona around intercellular nanoparticles and could be a footstone to link the formation of protein corona around nanoparticles to their biological function in cells.

The difficulty in achieving high removal efficiency for contaminants in textile wastewater over a wide range of pH impedes the progress of its treatment technique greatly. Herein, a facile and sustainable strategy was adopted for constructing magnetic ordered mesoporous polymers (M-OMPs) without the assistance of organic solvent and catalyst. The prepared M-OMPs were endowed with high special surface area and good superparamagnetism simultaneously, and exhibited high removal efficiency (>99%) for Methylene Blue (MB) within a short time (10 min) at a concentration of 50 mg/L. What's more, high removal efficiency was achieved over a wide range of pH 2-12 and the adsorption capacity for MB on M-OMPs was substantially retained even after 5 adsorption-desorption cycles, further demonstrating the application potential of M-OMPs in the decontamination of textile wastewater.

Quantification of multiple disease-related microRNAs (miRNAs) is of great significance for clinical diagnosis. Based on the simultaneous multiple element detection ability of inductively coupled plasma-mass spectrometry (ICP-MS) and good specificity of multicomponent nucleic acid enzymes (MNAzymes), a novel and simple method based on the MNAzyme amplification strategy and lanthanide labeling coupled with ICP-MS detection was proposed for the sensitive and simultaneous detection of three miRNAs (miRNA-21, miRNA-155, and miRNA-10b). Specifically, a probe consisting of streptavidin-modified magnetic beads (SA-MBs) and three DNA substrates labeled with lanthanide tags (159Tb/165Ho/175Lu) was constructed. In the presence of target miRNAs, three pairs of MNAzymes were assembled where each pair was hybridized with the corresponding miRNA, and then the substrates on the SA-MBs were cleaved by the activated MNAzymes, continuously releasing the fragment with lanthanide tags. The released lanthanide tags in the supernatant were collected after magnetic separation and analyzed by ICP-MS, realizing the simultaneous quantification of multiple miRNAs. The correlation of the lanthanide tag signal with the miRNA concentration fitted well in a linear model in the range of 50-1000 pmol L-1 (miRNA-21) and 50-2000 pmol L-1 (miRNA-155 and miRNA-10b). The limits of detection for three miRNAs were 11-20 pmol L-1, with the relative standard deviations of 2.2-2.7%. The recoveries of target miRNAs in the human serum and HepG-2 cells were in the range of 87.2-111% and 93.3-111%, respectively. Overall, the method is ideal for the simultaneous quantification of multiple miRNAs with advantages of low spectral interference, high sensitivity, good selectivity, and strong resistance to the complex matrix.

Adenosine triphosphate (ATP), as a high-energy phosphate compound that stores and releases energy in living cells, has an irreplaceable role in many physiological processes and maintenance of biological functions, and can be used as an indicator of many diseases. In this work, a composite nanoprobe, silver nanocluster (AgNC) @ molybdenum disulfide (MoS2), was designed to achieve in situ fluorescence imaging and quantitative analysis of intracellular ATP in HeLa cells by fluorescence spectrometry and inductively coupled plasma mass spectrometry (ICP-MS). The probe was constructed based on the adsorption of DNA-AgNCs by MoS2 nanosheets, and the DNA-AgNCs were prepared with the ATP aptamer as a template, whose fluorescence was initially quenched by MoS2. When the probe was incubated into the cells, intracellular ATP recognized the aptamer sequence and caused the DNA-AgNCs to fall off the MoS2 nanosheets, resulting in fluorescence recovery. Here, AgNCs not only acted as a fluorescence label for imaging, but also as an element tag for quantitative analysis of intracellular ATP with the detection of 107Ag by ICP-MS. The ATP in HeLa cells detected by this method was 24.6 ± 1.7 nmol L−1, which was in good agreement with the test result of the ATP test kit (20.4 ± 0.8 nmol L−1). The proposed method has potential application in medical clinical diagnosis and evaluation of the body’s metabolic level via fluorescence imaging and ICP-MS detection of intracellular ATP.

Fructus Ligustri Lucidi (FLL) has been preclinically and clinically used to treat musculoskeletal diseases. However, whether and how FLL affect the canonical Wnt/β-catenin signaling in the management of osteoporosis remains largely unknown. To this end, ovariectomized (OVX) rats and primary osteoblasts were administrated with FLL aqueous extract and medicated serum, respectively. Supplement of FLL to OVX rats maintains bone quality by attenuating the reduction in bone mineral density, strength and microstructure. The maintenance may be associated with upregulating the expression of insulin-like growth factor-1, osteoprotegerin, phospho (p)-low-density lipoprotein receptor-related protein 6, p-glycogen synthase kinase 3 beta (GSK3β), β-catenin, Runx2 and c-Myc, and downregulating the expressions of sclerostin (SOST), dickkopf-related protein 1 (DKK1), GSK3β and p-β-catenin in rat femurs and tibias. In addition, the medicated serum promotes osteoblastic bone formation through activation of Wnt/β-catenin signaling via inhibition of DKK1 and SOST overexpression. Salidroside may be one of the active ingredients in FLL that are beneficial for bone homeostasis. In summary, our results suggest that FLL may preserve bone quality through induction of canonical Wnt/β-catenin signaling via inhibition of DKK1 and SOST overexpression. And FLL may offer a new source of the DKK1 or SOST inhibitors in protection against osteoporosis.

A spiral stir bar was proposed by using stainless steel spring as the extraction phase carrier to avoid the extraction phase friction and increase the amount of extraction phase for improving extraction efficiency. The extraction phase is filled in the cavity of the spring, resulting in a larger amount of the extraction phase than that conventionally coated on glass stir bar or stainless steel wire. Polyaniline-polydimethylsiloxane sol-gel packed spiral stir bar was prepared and evaluated for the extraction of five estrogens. The prepared spiral stir bar presented good extraction efficiency/preparation reproducibility and long lifetime (more than 150 reused times) for target estrogens. Based on it, a method of spiral stir bar sorptive extraction combined with high performance liquid chromatography coupled with ultra-violet detection was developed for the analysis of trace estrogens in environmental and food samples. The detection limit for five estrogens was 0.11-.31 µg/L, with the enrichment factors of 83.0-118-fold (maximal enrichment factor: 200-fold). The reproducibility evaluated with each estrogen of 5 µg/L (n = 5) was 5.8-8.9%. The method was successfully applied for the determination of estrogens in environmental water and animal-derived food samples.

Background: Radix Stephaniae Tetrandrine (RST), known as FangJi (Pinyin name) in Chinese, is the dried root of Stephania tetrandra S.Moore, and has been prescribed in combination with other herbs to treat cardiovascular diseases and breast cancer in traditional Chinese medicine (TCM) clinical trials. Objective: The aim of the review is to provide a comprehensive evaluation about the application of RST in breast cancer management in TCM clinical trials, its ingredients, and its action on preventing the development of breast cancer in vitro and in vivo studies. Methods: Literature sources used were Pubmed, CNKI.net, Cqvip.com, and the Web of Science. For the inquiry, keywords such as Fangji, breast cancer, clinical trials, Radix Stephaniae Tetrandrine, tetrandrine, and fangchinoline were used in various combinations. About 150 research papers and reviews were consulted. Results: In TCM, RST exhibited the anti-tumor ability through its action on the bladder and lungs through dispersing phlegm and blood stasis. 10 clinical trials were identified which used RST in combination with other herbs to treat breast cancer. On average, the trials were characterized by high efficacy (>85%) and low toxicity. However, most of the clinical trials are characterized as small patient samples, poor design, and different combinations of herbs in prescriptions. To date, more and more compounds have been isolated from this plant. RST exhibited anti-tumor activities by targeting reversing multidrug resistance, inhibiting cell proliferation, inducing apoptosis, preventing tumor angiogenesis, anti-oxidation, anti-inflammation, and enhancing the sensitization and attenuating the toxicity of radiotherapy. Conclusion: The successful applications of RST in TCM clinical trials and preclinical experiments to beating breast cancer will provide potent lead compounds in the identification of novel anti-cancer drugs, which further contributes to the scientific exploration of functions of RST in TCM.

Graphene oxide (GO) with abundant oxygen functional groups has been widely studied as friction-reduction additive in lubricating oil or directly as solid lubricant. However, the lubrication mechanisms of these groups in solid GO lubricants have not been well revealed yet. Herein, we report a new strategy to prepare hydroxyl-carboxyl-terminated GO, hydroxyl-terminated GO, less-hydroxyl-terminated GO, and reduced GO (rGO), and then employ them as a frictional research model to address this concern. The results reveal that a little amount of water confined in GO layers dominates the interlayer shearing resistance by confining hydrogen-bond interaction. The strongest confinement effect to hydrogen-bond is achieved in hydroxyl-terminated GO, leading to the lowest shearing resistance between GO layers and then the enhanced friction performances. This finding first reveals the synergistic lubrication mechanism of oxygen functional groups and confined interlayer water molecules, providing us a new design view to fabricate superior graphene-based solid lubrication materials.

It has been reported that lipid biosynthesis in plant host root cells plays critical roles in legume‐fungal or ‐rhizobial symbioses, but little is known about its regulatory mechanism in legume–rhizobia interaction. Soybean WRINKLED1 (WRI1) a and b, with their alternative splicing (AS) products a’ and b’, are highly expressed in developing seeds and nodules, but their functions in soybean nodulation are not known. GmWRI1a and b differently promoted triacylglycerol (TAG) accumulation in both Arabidopsis wild‐type and wri1 mutant seeds and when they ectopically expressed in the soybean hairy roots. Transcriptome analysis revealed that 15 genes containing AW boxes in their promoters were targeted by GmWRI1s, including genes involved in glycolysis, fatty acid (FA) and TAG biosynthesis. GmWRI1a, GmWRI1b and b’ differentially transactivated most targeted genes. Overexpression of GmWRI1s affected phospholipid and galactolipid synthesis, soluble sugar and starch contents and led to increased nodule numbers, whereas GmWRI1 knockdown hairy roots interfered root glycolysis and lipid biosynthesis and resulted in fewer nodules. These phenomena in GmWRI1 mutants coincided with the altered expression of nodulation genes. Thus, GmWRI1‐regulated starch degradation, glycolysis and lipid biosynthesis were critical for nodulation. GmWRI1 mutants also altered auxin and other hormone‐related biosynthesis and hormone‐related genes, by which GmWRI1s may affect nodule development. The study expands the views for pleiotropic effects of WRI1s in regulating soybean seed filling and root nodulation.

As one of nature clay minerals, sepiolite was used in various fields, such as photocatalytic, refractory, and dye adsorption, but rare in superhydrophobic application. In this study, sepiolite was modified by polysiloxane (Sepiolite@POS) and its microstructure, chemical composition, and hydrophobic property were investigated. Besides, its coatings were prepared by spray-coating the solution of Sepiolite@POS and epoxy (EP), which endowed various materials (wood blocks, steel sheets, A4 paper, and cotton) with excellent superhydrophobic property. In particular, as the mass ratio of Sepiolite@POS and EP was 7:5, the coating on glass slide presented a water contact angle above 160° and a sliding angle of 5°. And it could maintain superhydrophobicity after wear test or ultrasound treatment or immersion in solution with pH from 1 to 13, implying it had good mechanical and chemical stability. Besides, after being coated by Sepiolite@POS/EP, the surface possessed excellent self-cleaning property. Also, the Sepiolite@POS/EP coated melamine sponge could efficiently separate chloroform from water. All these results indicated that modified sepiolite showed great potential for application in the fields of waterproof, self-cleaning, and oil/water separation.

Increasing use of TiO2 nanoparticles (NPs) leads to an accumulation in environment and a risk to human. In this work, an on-line monolithic capillary microextraction (CME)-inductively coupled plasma mass spectrometry (ICP-MS) method was developed for sensitive determination of TiO2 NPs in environmental water samples. With the presence of citric acid, TiO2 NPs can be selectively adsorbed on the poly(acrylamide)-vinylpyridine-methylene bis-(acrylamide) (poly(AA-VP-Bis)) monolithic column at pH 1, whereas Ti ions cannot be adsorbed. Factors affecting the extraction efficiency such as sample pH, concentration of citric acid and elution conditions were carefully investigated. Under the optimum conditions, a detection limit of 0.63 μg L−1 for TiO2 NPs and a sample throughput of 6 h−1 were achieved. The method can be applied to the determination of TiO2 NPs in the size range of 5–100 nm. The developed method was then applied to the analysis of TiO2 NPs in the East Lake water and Yangtze River water, and recoveries of 84–109% were obtained for spiked samples. Compared with other reported methods for the determination of metal oxide NPs, the developed on-line monolithic CME-ICP-MS has the merits of simple operation, high throughput and no need for digestion before ICP-MS detection.

Archaea and Bacteria have evolved different defence strategies that target virtually all steps of the viral life cycle. The diversified virion morphotypes and genome contents of archaeal viruses result in a highly complex array of archaea-virus interactions. However, our understanding of archaeal antiviral activities lags far behind our knowledges of those in bacteria. Here we report a new archaeal defence system that involves DndCDEA-specific DNA phosphorothioate (PT) modification and the PbeABCD-mediated halt of virus propagation via inhibition of DNA replication. In contrast to the breakage of invasive DNA by DndFGH in bacteria, DndCDEA-PbeABCD does not degrade or cleave viral DNA. The PbeABCD-mediated PT defence system is widespread and exhibits extensive interdomain and intradomain gene transfer events. Our results suggest that DndCDEA-PbeABCD is a new type of PT-based virus resistance system, expanding the known arsenal of defence systems as well as our understanding of host-virus interactions., The prokaryote defence system Dnd relies on phosphorothioation (PT) of DNA backbone to distinguish between self and non-self DNA. Here, Xiong et al. describe a previously uncharacterized PT-based antiviral system that is independent of the canonical Dnd and is widespread in Archaea and Bacteria.

Zinc oxide (ZnO) nanoparticles (NPs) are widely used in daily life. However, common utilization of ZnO NPs results in increases in environmental release, and their health hazards have attracted extensive attention. To investigate the cytotoxicity of ZnO NPs and their mechanism in HepG2 cells, a comprehensive analytical system was developed. The internalization, cytotoxic mechanism, death mechanism and elimination behavior of three sizes of ZnO NPs were studied by electrothermal vaporization (ETV)-inductively coupled plasma mass spectrometry (ICP-MS), MTT assays, GSH measurements, ROS measurements and analyses of apoptosis and gene expression. The size-, dose- and time-dependent characteristics of ZnO NPs were determined, and the metabolism of ZnO NPs in cells was discussed. The cytotoxicity of ZnO NPs was confirmed to depend on both the size and concentration and was attributed to the release of Zn2+, induction of oxidative stress and inflammatory response; the death mode of HepG2 cells incubated with ZnO NPs was necrotic rather than programmed cell death.

A novel near-infrared-responsive (NIR-responsive) photothermal therapy (PTT) agent based on perylene-diimide-encapsulated (PDI-encapsulated) PEGylated silica nanocapsules (SNCs) is developed. Dicyclohexylamino-PDI (DCAPDI) with electron-donating cyclohexylamino substitutes at bay positions aggregates into J-aggregation in the core of SNCs, and their electronic coupling interactions are strengthened because of the spatial confinement of SNCs, resulting in strong NIR absorption but negligible fluorescence emission which is crucial for NIR-responsive PTT. Based on our knowledge, this is the first example of generating NIR photothermal conversion by means of molecular aggregation derived from spatial confinement. Unprecedented photostability is achieved with the DCAPDI-encapsulated SNCs in response to more than 60 runs of cyclic NIR exposure with each run exposed to the 808 nm, 1 W cm

Background Soybean oil constitutes an important source of vegetable oil and biofuel. However, high temperature and humidity adversely impacts soybean seed development, yield, and quality during plant development and after harvest. Genetic improvement of soybean tolerance to stress environments is highly desirable. Results Transgenic soybean lines with knockdown of phospholipase Dα1 (PLDα1KD) were generated to study PLDα1′s effects on lipid metabolism and seed vigor under high temperature and humidity conditions. Under such stress, as compared with normal growth conditions, PLDα1KD lines showed an attenuated stress-induced deterioration during soybean seed development, which was associated with elevated expression of reactive oxygen species-scavenging genes when compared with wild-type control. The developing seeds of PLDα1KD had higher levels of unsaturation in triacylglycerol (TAG) and major membrane phospholipids, but lower levels of phosphatidic acid and lysophospholipids compared with control cultivar. Lipid metabolite and gene expression profiling indicates that the increased unsaturation on phosphatidylcholine (PC) and enhanced conversion between PC and diacylglycerol (DAG) by PC:DAG acyltransferase underlie a basis for increased TAG unsaturation in PLDα1KD seeds. Meanwhile, the turnover of PC and phosphatidylethanolamine (PE) into lysoPC and lysoPE was suppressed in PLDα1KD seeds under high temperature and humidity conditions. PLDα1KD developing seeds suffered lighter oxidative stresses than did wild-type developing seeds in the stressful environments. PLDα1KD seeds contain higher oil contents and maintained higher germination rates than the wild-type seeds. Conclusions The study provides insights into the roles of PLDα1 in developing soybean seeds under high temperature and humidity stress. PLDα1KD decreases pre-harvest deterioration and enhances acyl editing in phospholipids and TAGs. The results indicate a way towards improving production of quality soybean seeds as foods and biofuels under increasing environmental stress. As is well-known, arsenite (As(iii)) is a human carcinogen associated with many human cancers. As(iii) can act as a co-carcinogen to induce DNA damage with other carcinogens. Benzo(a)pyrene diol epoxide (BPDE) is one of the most-studied environmental carcinogens, which exists ubiquitously in our daily life. The elucidation of the mechanism of As(iii) as a co-carcinogen with BDPE in cells causing genotoxicity is beneficial for the evaluation of its bioeffect. In this study, a comprehensive analytical system is used for DNA damage evaluation, BPDE-DNA adduct detection, arsenic speciation and gene expression analysis. Based on the experimental results, it can be inferred that BPDE and As(iii) synergistically cause genotoxicity, and the possible mechanism is that BPDE inhibits arsenic methylation, leading to cellular As(iii) enrichment. As(iii) inhibits nucleotide excision repair (NER) of the DNA adduct damage caused by BPDE. The synergistic effect of BPDE and As(iii) causes DNA strand break damage, which further results in carcinogenesis.

In this work, magnetic nanoporous carbons (MNPCs) based on Zn/Co-MOFs was synthesized via a facial one-step carbonization method. The prepared MNPCs exhibited high specific surface areas, good chemical stability and high extraction efficiency for interest organophosphorus pesticides (OPPs). The good extraction performance of the MNPCs for interest OPPs can be primarily attributed to π-π interaction and hydrophobic interaction between MNPCs and OPPs. Based on it, MNPCs-based MSPE-GC-FPD method was established for the quantification of five OPPs in fruit samples. The main experimental conditions for MSPE of five target OPPs were investigated systematically and optimized, such as the desorption solvent and volume, extracted volume, ionic strength, extraction time and desorption time. Under the optimal conditions, the limits of detection (LODs, S/N = 3) were found to be in the range of 0.018 (ethion) -0.045 (malathion) μg/L. The linearity was 0.05-100 μg/L for phorate, diazinon, ethion and 0.1-100 μg/L for malathion, fenthion respectively. The reproducibility of the method was in the range of 3.5-9.7%. The recovery experiments were carried out by spiking target OPPs at three concentration levels to validate the accuracy of the proposed method, and the recoveries for target OPPs were 84-116%. The results show that the proposed method in this work can be successfully used to analyze OPPs residue in fruit sample.

A novel ternary hybrid of carbon nanotubes/graphene oxide/MoS2 was prepared via hydrothermal method, and its microstructure, phase composition, and enhancement effect on the tribological properties of epoxy were investigated systematically. Results revealed EP-CNTs/GO/MoS2 possessed the lowest friction coefficient and wear rate than EP and its other composite coatings reinforced by single filler or binary hybrids. In particular, its friction coefficient and wear rate were 0.042 and 3.44 × 10−5 mm3/Nm, respectively, which were reduced by up to 90% and 95% compared to those of pure-EP. And this was mainly attributed to uniform dispersion of CNTs, GO, MoS2 in CNTs/GO/MoS2 hybrid, and load-carrying capacity of CNTs and GO, self-lubricating effect of MoS2, as well as formation of transfer film onto counterpart surface.

Arsenite (As(III)) has been considered as a human carcinogen associated with many human cancers especially skin cancer. Elucidation of the transformed species of As(III) during its metabolism in cells is beneficial for evaluation of its bioeffect. In this work, a hyphenated method of reversed phase ion pair high performance liquid chromatography - inductively coupled plasma mass spectrometry (RP-IP-HPLC-ICP-MS) equipped with collision/reaction cell technology (CCT) was developed for speciation of As(III) and its metabolites (arsenate [As(V)], monomethylarsonic acid [MMA(V)], and dimethylarsinic acid [DMA(V)]) in SCC-7 cells. The developed analytical method exhibits low limits of detection for interest arsenic species in the range of 14-27 ng/L and wide linear range up to four orders of magnitude, providing a sensitive tool for arsenic metabolites analysis and further understanding the metabolism of As(III) in SCC-7 cells. The effect of exposure time, exposure concentrations and elimination time on the arsenic species and total arsenic in SCC-7 cells incubated by As(III) were systematically studied. At low exposure concentrations (5 μM), large proportion of intracellular As(III) transformed to methylated metabolites, and the final methylated metabolite DMA(V), which could not be completely removed from the cells in the elimination process, is considered to play as the primary carcinogen. While at high exposure concentrations (5 μM), most of intracellular As(III) probably bound to biomacromolecules rather than followed biomethylation process, exhibiting different metabolism.

Accumulating evidence suggests that mitochondrial dysfunction and adipocyte differentiation promote lipid accumulation in the development of obesity and diabetes. Curcumin is an active ingredient extracted from Curcuma longa that has been shown to exhibit antioxidant and anti-inflammatory potency in metabolic disorders. However, the underlying mechanisms of curcumin in adipocytes remain largely unexplored. We studied the effects of curcumin on adipogenic differentiation and mitochondrial oxygen consumption and analysed the possible mechanisms. 3T3-L1 preadipocytes were used to assess the effect of curcumin on differentiation of adipocytes. The Mito Stress Test measured by Seahorse XF Analyzer was applied to investigate the effect of curcumin on mitochondrial oxygen consumption in 3T3-L1 adipocytes. The effect of curcumin on the morphology of both white and brown adipose tissue (WAT and BAT) was evaluated in a high-fat diet-induced obese mice model. We found that curcumin dose-dependently (10, 20 and 35 µM) induced adipogenic differentiation and the intracellular fat droplet accumulation. Additionally, 10 µM curcumin remarkably enhanced mature adipocyte mitochondrial respiratory function, specifically, accelerating basic mitochondrial respiration, ATP production and uncoupling capacity via the regulation of peroxisome proliferator-activated receptor γ (PPARγ) ( p < 0.01). Curcumin administration also attenuated the morphological changes in adipose tissues in high-fat diet-induced obese mice. Moreover, curcumin markedly increased the mRNA and protein expressions of mitochondrial uncoupling protein 1 (UCP1), PPARγ, peroxisome proliferator-activated receptor-γ coactivator-1α (PGC-1α) and PR domain protein 16 (PRDM16) in vivo and in vitro . Collectively, the results demonstrate that curcumin promotes the adipogenic differentiation of preadipocytes and mitochondrial oxygen consumption in 3T3-L1 mature adipocytes by regulating UCP1, PRDM16, PPARγ and PGC-1α expression.

Introduction The present study aims to evaluate the efficacy and safety of apatinib monotherapy or combination therapy for patients with advanced or recurrent biliary tract cancer (BTC). Methods Twenty-eight patients with advanced or recurrent BTC who progressed after prior systemic therapies and treated with apatinib from January 2017 to June 2019 were enrolled in this retrospective and observational study. The primary end point was progression free survival (PFS). Secondary end points included overall survival (OS), objective response rate (ORR), disease control rate (DCR), and toxicity. Results A total of 28 patients with advanced or recurrent BTC who progressed after prior systemic therapies received apatinib monotherapy or combination therapy (with capecitabine, S-1, oxaliplatin, irinotecan or PD-1 inhibitor), including 9 cases of gallbladder cancer and 19 cases of cholangiocarcinoma. Six patients achieved PR, 15 patients had SD and 7 patients had PD. Median progression-free survival (PFS) and overall survival (OS) was 4.3 months (95%CI = 1.8–6.8) and 6.2 months (95% CI = 4.6–7.8) respectively. The ORR and DCR were 21.4% (6/28) and 75.0% (21/28), respectively. Most of the adverse events were grade 1–2 in severity, apatinib treatment was well tolerated. Conclusions Apatinib monotherapy or combination therapy can improve PFS in patients with advanced or recurrent BTC who progressed after prior systemic therapies, and adverse reactions can be well tolerated. Our study support apatinib therapy as a feasible therapeutic strategy in advanced or recurrent BTC.

In recent years, the biological activity of polysaccharides and their derivatives has been widely studied. However, in addition to the natural polysaccharides directly extracted from plants and animals, there are rich polysaccharides in the pulping waste liquor that have not been fully utilized. The extracted polysaccharide from eucalyptus Alkaline Peroxide Mechanical Pulping (APMP) waste liquor was used as a raw material. For the production of carboxymethyl polysaccharide, the effects of temperature (T), the amount of alkali (NaOH) and the amount of etherifying agent (ClCH2COOH) on the degree of substitution (DS) were investigated, the optimal preparation conditions are: reaction time 2 h, temperature 75 °C, and the molar ratio of polysaccharide, NaOH and ClCH2COOH is 1:1:2, the highest DS is 1.47; FT-IR, NMR and GPC were used to characterize the structure and Molecular weight, the results show that the polysaccharide of APMP waste liquor is rich in xylan, and it was proved that the carboxymethyl substitution was successful and the positions of the substituent group were determined. The characterization and biological activity research of xylan polysaccharide (XP) and carboxymethyl xylan polysaccharide (CMXP), such as antioxidation, moisture absorption/retention, bacteriostatic action and cytotoxicity were discussed. CMXP shows better effects compared with XP.

In this study, CNTs were first modified by polydopamine (PDA) to facilitate their surface activation and then Ag nanoparticles were in-situ grown on CNT-PDA to obtain CNT-PDA/Ag. The effects of functionalized CNTs on the tribological behaviors of poly (phthalazinone ether sulfone ketone) (PPESK) were investigated. Results revealed that PDA layer could not only increase the active functional groups on the CNT surface, but also act as an activated platform for the deposition of mono-dispersed Ag nanoparticles. The decoration of Ag nanoparticles decreased the interaction among CNTs, which was conducive to their good dispersion. Besides, by filling 0.3 wt.% CNT-PDA/Ag, the thermal properties of PPESK were significantly improved, and its friction-reduction and anti-wear performances were the best among all PPESK composites, due to the good interfacial interaction between CNTs and PPESK, the synergistic reinforcing and lubricating effect of CNTs and Ag nanoparticles, as well as the lubricating effect of transfer film.

Recent progress of stir bar sorptive extraction (SBSE) in the past six years is reviewed. The preparation methods including electrodeposition, self-assembly, solvent exchange, physical magnetic adsorption and electrostatic spinning, for the coated stir bar are summarized and compared, specifically for a specific material for coatings fabrication, e.g., carbon-based materials and metal organic frameworks. The emerging materials (e.g., graphene, graphene oxide, carbon nanotubes, monolith, metal-organic frameworks and porous organic polymers) applied for coated stir bar fabrication are one of the focus of this review, along with their respective advantages in extraction process and application in trace analysis. The development and application of extraction apparatus of SBSE are also involved. Based on these information, the development status and prospects of SBSE as an efficient sample pretreatment technique in real sample analysis are discussed.

Fast green FCF (FGF) is often used in foods, pharmaceuticals, and cosmetics. However, little is known about the interactions of FGF with amyloid-β protein (Aβ) associated with Alzheimer's disease. In this study, the inhibitory effects of FGF on Aβ fibrillogenesis, the disruption of preformed Aβ fibrils, the reduction of Aβ-induced cytotoxicity, and the attenuation of Aβ-induced learning and memory impairments in mice were investigated. FGF significantly inhibited Aβ fibrillogenesis and disintegrated the mature fibrils as evidenced by thioflavin T fluorescence and atomic force microscopy studies. Co-incubation of Aβ with FGF greatly reduced Aβ-induced cytotoxicity in vitro. Moreover, FGF showed a protective effect against cognitive impairment in Aβ-treated mice. Molecular dynamics simulations further showed that FGF could synergistically interact with the Aβ17-42 pentamer via electrostatic interactions, hydrogen bonds and π-π interactions, which reduced the β-sheet content, and disordered random coils and bend structures of the Aβ17-42 pentamer. This study offers a comprehensive understanding of the inhibitory effects of FGF against Aβ neurotoxicity, which is critical for the search of effective food additives that can combat amyloid-associated disease.

Ti (IV) modified vinyl phosphate magnetic nanoparticles (Fe3 O4 @SiO2 @KH570-PO4 -Ti (IV)) was prepared for simultaneous extraction of multiple arsenic species, followed by high performance liquid chromatography (HPLC) - inductively coupled plasma mass spectrometry (ICP-MS) analysis. Inorganic arsenic (iAs), dimethyl arsenic acid (DMA), monomethyl arsenic acid (MMA), p-amino phenyl arsenic acid (p-ASA), 4-hdroxyphenylarsenic acid (4-OH), phenyl arsenic acid (PAA) and 3-nitro-4-hydroxyphenylarsenic acid (ROX) were investigated as interest analytes. It was found that they were quantitatively adsorbed on Fe3 O4 @SiO2 @KH570-PO4 -Ti (IV) at pH 5, and desorbed completely with 0.1 mol/L sodium hydroxide solution. Enrichment factor of 100-fold was obtained by consuming 100 mL sample solution. Under the optimal conditions, the method combining MSPE with HPLC-ICP-MS presented a linear range of 1-5000 ng/L for seven arsenic species. The limits of detection were 0.39, 0.60, 0.23, 1.85, 0.54, 0.48 and 0.84 ng/L for DMA, MMA, p-ASA, iAs, 4-OH, PAA, ROX, with the relative standard deviations (c = 10 ng/L, n = 7) of 3.6, 3.9, 5.5, 12.4, 6.1, 5.8, 5.0, respectively. The accuracy of the method was validated by analyzing BCR 627 Tuna fish. The application potential of the method was further evaluated by chicken muscle and liver samples. No target arsenic species were detected in these samples, and good recoveries (80.6-123%) were obtained for the spiked samples at low, medium and high concentration levels. ZIF-8 nanoparticles (NPs) has been demonstrated with good potential in drug delivery, which causes an increasing attention on relevant toxicity study. In this work, MTT (3-[4,5-dimethylthiazol-2-yl]-2,5-diphenyltetrazolium bromide), glutathione (GSH), reactive oxygen species (ROS), stain analysis and gene detection assays were performed on ZIF-8 (50, 90 and 200 nm) incubated HepG2 cells. Moreover, time-resolved inductively coupled plasma mass spectrometry (TRA-ICP-MS) was applied for single cell analysis; the variation in cellular zinc amount and the proportion of zinc up-taken cells was investigated as a function of NPs size, incubation concentration/time and elimination. Smaller size of ZIF-8 NPs would lead to higher zinc accumulation and toxicity. The function of ZIF-8 on cells is assumed to be mainly related to zinc intracellular accumulation. The possible action path is presented as high accumulation of zinc in ZIF-8 incubated cells lead to high ROS level and cellular inflammation, ultimately inducing necrocytosis. For better understanding of the bio-effect of ZIF-8, ZnO NPs and Zn2+ incubated HepG2 cells were evaluated in the same way. Higher accumulation of zinc in larger part of the cell population was found in ZIF-8 incubated cells than that in ZnO NPs incubated cells. It demonstrated higher bioavailability for ZIF-8 over ZnO NPs. While, in drug delivery application, the possible risk of the remained intracellular ZIF-8 cannot be ignored.

The growing use of rare-earth doped upconversion nanoparticles (UCNPs) has caused increasing concern about their biosafety. Here, to understand the toxicity of UCNPs and their mechanism in HepG2 cells, we systematically study the cytotoxicity, uptake and elimination behaviors of three types of UCNPs combined multiple cytotoxicity evaluation means with inductively coupled plasma mass spectrometry (ICP-MS) detection. Sodium yttrium fluoride, doped with 18% (molar ratio) ytterbium and 2% erbium (NaYF4: Yb3+, Er3+) was selected as the model UCNPs with two sizes (35 and 55 nm), and the poly(acrylic acid) and polyethylenimine were selected as the representatives of negative and positive surface coating of UCNPs, respectively. UCNPs were found to induce cytotoxicity in time- and dose-dependent manners, which might be mediated by reactive oxygen species generation and oxidative stress. Apoptosis, inflammation, and metabolic process were enhanced after cells exposed to 200 mg/L UCNPs for 48 h. Increase in the protein levels of cleaved caspased-9, cleaved caspase-3 and Bax and decrease in the anti-apoptotic protein, Bcl-2 suggested that the mitochondria mediated pathway was involved in UCNP-induced apoptosis. With the aid of ICP-MS, it demonstrated that the cytotoxicity was associated with internalized amount of UCNPs, which largely relied on their surface properties rather than size in the tested range. By comparing UCNPs with Y3+ ions, it demonstrated that NPs properties played a nonnegligible role in the cytotoxicity of UCNPs. These findings provide new insights for fundamental understanding of cytotoxicity of UCNPs and may contribute to more rational use of these materials in the future.

A melamine-based porous organic polymer (mPMF) was synthesized as solid phase extraction (SPE) adsorbent for inline SPE-high performance liquid chromatography (HPLC)-ultraviolet detector (UV) analysis of five phytohormones. Melamine contributed to the rich π-electron and N-containing triazine structure for mPMF, which could form π-π interaction and intermolecular hydrogen bond with COOH containing phytohormones. The synthesized mPMF adsorbent shows extremely high extraction efficiency for five target analytes (>80%) including salicylic acid, indole-3-acetic acid, abscisic acid, 2,4-dichlorophenoxyacetic acid and 1-naphthalene acetic acid. The factors affecting extraction of the target phytohormones were investigated and the optimized experimental conditions were established. The linear range was 0.2–100 μg/L with the limits of detection in the range of 0.06–0.11 μg/L for five target phytohormones. The developed method of mPMF-inline-SPE-HPLC-UV was applied for the analysis of trace target phytohormones in tomato and grape juice samples, and the recovery in the range of 83.1–116% and 87.2–121% was obtained for the spiked tomato and grape juice, respectively. This method has the advantages of high sensitivity and good reproducibility, and it has good application potential for the analysis of phytohormones in fruit samples. Moreover, inline analysis avoided the problems of sample pollution and sample loss, and provided a sample throughput of 5/h.

A dual column packed with a magnetic metal-organic framework composite (MFC) and mercapto-functionalized MFC nanoparticles (MFC-SH) in microfluidic chip channels is described for array chip-based magnetic solid phase microextraction of arsenic species including arsenite [As(III)], arsenate [As(V)], monomethylarsonous acid (MMA) and dimethylarsinic acid (DMA) in SCC-7 cells. At pH 6, the MFC-SH adsorbs As(III) while MFC quantitatively adsorbs As(V), DMA and MMA. The As(III) adsorbed on the MFC-SH can be desorbed with a 2% solution of thiourea in 0.5 M HNO3. The arsenic species MMA, DMA and As(V) are retained on the MFC but can be desorbed by ammonia. A sequential elution strategy was employed to elute MMA, DMA, As(V) and As(III) one by one for subsequent on-line determination by ICP-MS. The limits of detection are 4.8, 6.3, 3.8 and 7.1 ng L−1 for As(V), DMA, MMA and As(III), respectively. The enrichment factors are between 23 and 25, and the throughput is 7 samples per hour. The arsenic species in (spiked) SCC-7 cell samples were analyzed by the online system with adequate recoveries (89–110%).

Symbiotic rhizobia-legume interactions are energy-demanding processes, and the carbon supply from host cells that is critically required for nodulation and nitrogen fixation is not fully understood. Investigation of the lipidomic and carbohydrate profiles with the transcriptome of developing nodules revealed highly activated glycolysis, fatty acid (FA), 2-monoacylglycerol (2-MAG), and membrane lipid biosynthesis and transport during nodule development. RNA-sequence profiling of metabolic genes in roots and developing nodules highlighted the enhanced expression of genes involved in the biosynthesis and transport of FAs, membrane lipids, and 2-MAG in rhizobia-soybean symbioses via the RAML-WRI-FatM-GPAT-STRL pathway, which is similar to that in legume-arbuscular mycorrhizal fungi symbiosis. The essential roles of the metabolic pathway during soybean nodulation were further supported by analysis of transgenic hairy roots overexpressing soybean GmWRI1b-OE and GmLEC2a-OE. GmLEC2a-OE hairy roots produced fewer nodules, in contrast to GmWRI1b-OE hairy roots. GmLEC2a-OE hairy roots displayed different or even opposite expression patterns of the genes involved in glycolysis and the synthesis of FAs, 2-MAG, TAG, and membrane lipids compared to GmWRI1b-OE hairy roots. Glycolysis, FA and membrane lipid biosynthesis were repressed in GmLEC2a-OE but increased in GmWRI1b-OE hairy roots, which may account for the reduced nodulation in GmLEC2a-OE hairy roots but increased nodulation in GmWRI1b-OE hairy roots. These data show that active FA, 2-MAG and membrane lipid biosynthesis are essential for nodulation and rhizobia-soybean symbioses. These data shed light on essential and complex lipid metabolism for soybean nodulation and nodule development, laying the foundation for the future detailed investigation of soybean nodulation.

The inhibitory effect of brazilin against α-synuclein (α-syn) fibrillogenesis, disruption effect against mature fibrils, and the following cytotoxicity were examined by systematical biochemical, biophysical, cellular biological, and molecular simulation experiments. It is found that brazilin inhibited α-syn fibrillogenesis and disrupted the performed fibrils with a concentration-dependent manner. Moreover, cellular experimental data showed that brazilin effectively reduced the cytotoxicity induced by α-syn aggregates. Finally, molecular dynamics simulations were performed to explore the interactions between brazilin and α-syn pentamer. It is found that brazilin directly interacts with α-syn pentamer, and the hydrophobic interactions are favorable for brazilin binding with the α-syn pentamer, while the electrostatic part provides adverse effects. Three binding regions were identified to inhibit α-syn fibrillogenesis or disrupt the preformed aggregates. Furthermore, six important residues (i.e., G51, V52, A53, E61, V66, and K80) of α-syn were also identified. We expected that brazilin is an effective agent against α-syn fibrillogenesis and associated cytotoxicity.

In this work, poly(1-vinylimidazole-ethyleneglycol dimethacrylate) (poly(VI-EDMA)) monolith coated stir bars were synthesized by one-pot polymerization, and they exhibited higher extraction efficiency and faster extraction dynamics for selected PFAAs than commercial ethylene glycol modified silicone (EG-silicone) and polydimethylsiloxane (PDMS) coated stir bars. Taking eleven PFAAs as target analytes, including C4-C12 perfluoroalkyl carboxylates (PFCAs) and C6, C8 perfluoroalkane sulfonates (PFSAs), a method combining monolith-based stir bar sorptive extraction (SBSE) with high performance liquid chromatography (HPLC) − electrospray tandem mass spectrometry (ESI–MS/MS) was proposed for the determination of multiplex PFAAs in environmental water samples. Under the optimized conditions, low limits of detection (0.06–0.40 ng/L) and wide linear range (0.6–400 ng/L) were obtained for target PFAAs with. The developed method was then applied for the analysis of target PFAAs in environmental water samples, and recoveries of 80.1–117% and 80.3–122% were obtained for target PFAAs in spiked Yangtze River and East Lake water samples respectively.

A switchable hydrophilicity solvent (SHS) based liquid phase microextraction (LPME) method was proposed for the preconcentration of trace Pb (II) and Cd (II) followed by graphite furnace atomic absorption spectrometry (GFAAS) detection. The SHS of protonated triethylamine carbonate (P-TEA-C) was synthesized and applied as the extractant, which is environmentally friendly and low cost. Pb (II) and Cd (II) in ammonium pyrrolidine dithiocarbamate (APDC) complex form was extracted into the triethylamine (TEA) phase by converting P-TEA-C to TEA. The vortex led to the rapid formation of fine droplets of the extractant in the aqueous solution, and enlarged contact surface between both immiscible liquids. The factors affecting the microextraction procedure, including sample pH, APDC concentration, volume of SHS and NaOH, sample volume and matrix effect were investigated. Under the optimal conditions, enrichment factor of 50 was achieved with consumption of 10 mL aqueous sample, and detection limits of 16 and 3.9 ng L−1 were obtained for Pb (II) and Cd (II), respectively. The accuracy of the proposed method was validated by the analysis of Certified Reference Materials of GSBZ50009-88 environmental water, GBW07605 tea and GBW07601(GSH-1) human hair. The method was also successfully applied for determination of Pb (II) and Cd (II) in water, tea and human hair samples.

A method based on ion-pair reversed phase high performance liquid chromatography (HPLC) hyphenated with inductively coupled plasma mass spectrometry (ICP-MS) was developed for arsenic speciation in extract of tree moss. Under the optimal conditions, the limit of detection of eight arsenic species including arsenite (AsIII), arsenate (AsV), monomethylarsonic acid (MMA), dimethylarsonic acid (DMA), trimethylarsinoxide (TMAO), tetramethylarsonium (Tetra), arsenocholine (AsC) and arsenobetaine (AsB) is between 0.04 and 0.07 ng/mL, with a linear range of 0.2 - 500 ng/mL. Three unknown arsenic species (Unk1, Unk2 and Unk3) and six specific arsenic species (AsIII, AsV, DMA, TMAO, Tetra and AsB) were detected in the extract of tree moss. Unk3 was identified as a kind of arsenosugars (2,3-dihydroxypropyl-5-deoxy-5(dimethylarsenoso)furanoside, arsenosugar X) by electrospray ionization quadrupole time-of-flight mass spectrometry (ESI-qTOF-MS).

Amino-decorated Zr-based magnetic Metal-Organic Frameworks composites (Zr-MFCs) were prepared by a facile and efficient strategy. The nano-sized Fe3O4@SiO2 core (about 15 nm) was coated with a shell of Zr-MOFs (about 5 nm) by means of in-situ growth. And, Fe3O4@SiO2@UiO-66 and its amino derivatives (Fe3O4@SiO2@UiO-66-NH2 and Fe3O4@SiO2@UiO-66-Urea) were successfully prepared by using different precursors. The obtained Zr-MFCs were demonstrated to be efficient adsorbents for metal ions/organic dyes removal from aqueous solution, with high adsorption capacity and fast adsorption kinetics. It was found that the amine-decorated MFCs were highly efficient for metal ions/dyes removal compared to raw MFC-O. Among them, MFC-N exhibited the highest capacity for Pb2+ (102 mg g−1) and methylene blue (128 mg g−1), while MFC-O exhibited the highest capacity for methyl orange (219 mg g−1). Moreover, anionic and cationic dyes could be selectively separated and removed from the mixed solution just by adjusting the solution pH with Zr-MFCs as the adsorbents. And these Zr-MFCs materials can be easily regenerated by desorbing metal ions/organic dyes from the sorbents with appropriate eluents, and the adsorption capacity can be remained unchanged after 6 recycles. The obtained results demonstrated the great application potential of the prepared MFCs as fascinating adsorbents for water treatment.

To overcome the poor mechanical properties of biomass aerogels in oil/water separations, three different freeze-casting methods were used including direct freezing, unidirectional freezing, and bidirectional freezing with sodium alginate (SA) aerogels reinforced with cellulose nanofibrils (CN). After chemical crosslinking and silane modification, compression testing revealed that the SA/CN aerogels with parallel lamellar microstructures prepared by bidirectional freezing exhibited super-elasticity with the minimal energy dissipation of ~ 0.04 in each cycle, a maximum compressive stress of 80.4 kPa, and minimal plastic deformation at ~ 4.15%. Additionally, the water and oil contact angle of the surface of the lamellar SA/CN aerogel was 148.7° and 0°, respectively. By combining the super-elasticity and hydrophobicity/superoleophilicity, the lamella SA/CN aerogels could be reused for the separation of oil/water mixture with oil absorption capacities up to 34 times its weight. Furthermore, the lamellar SA/CN aerogel could continuously separate oil/water mixtures with the assistance of a pump. Therefore, the present study offers a simple and environmentally friendly method for fabrication of super-elastic and hydrophobic/superoleophilic biomass aerogels that are applied to continuous removal of oil from water.

Magnetic metal–organic framework composites (MFCs) were prepared in water at room temperature through a simple, low-cost and green strategy only by employing organic salts as anionic linker sources. Amino-functionalized MFCs (MFC-N) with different amino group contents were successfully prepared by varying the ratio of disodium of 2-aminoterephthalic (NH2-Na2BDC)/terephthalic acid (Na2BDC). The prepared nanoscale MFC-N-X (X represents the percentage of NH2-Na2BDC, i.e., X = 0, 50, and 100) showed high surface area, large pore volume, excellent magnetic response, and good water and thermal stability. Fe–Ni-based composites with Ni-coated reduced grapheme oxide (RGO)–MoS2 were fabricated by powder metallurgy technique, and their morphology, phase composition, and tribological properties at different temperatures were investigated systematically. Results showed that Fe–Ni-based composite with Ni-coated RGO–MoS2 possessed much more uniform and denser microstructure, and higher hardness than that with RGO–MoS2. Furthermore, Ni-coated RGO–MoS2 additive greatly allowed increasing the friction-reducing and wear-resistant properties, due to the reinforcing and lubricating effect new phase (Fe3W3C, CrxS1+ x, etc.) and the improvement on interfacial compatibility between Ni-coated RGO–MoS2 and Fe–Ni matrix. In particular, as the content of Ni-coated RGO–MoS2 was 5 wt%, the friction coefficient and wear rate of the corresponding composite was decreased nearly 50% and 75%, respectively. More importantly, Ni-coated RGO–MoS2 reinforced composite kept excellent tribological properties at elevated temperature. Its friction coefficient decreased firstly with the increased temperature from 25℃ to 400℃, then increased slightly when the temperature increased to 600℃. Besides, the wear rate was only 0.73–0.9 × 10−4 mm3/Nm during the whole temperature range. This suggested that Ni-coated RGO–MoS2 was a promising additive of metallic-based composites suitable for high-temperature sliding condition.

Cyanuric chloride and 4,4′-diamino-p-terphenyl were adopted as monomers to synthesize poly (4,4′-diamino-p-terphenyl-triazine) (PDT) covalent organic polymer. PDT coated stir bar was prepared and evaluated for the extraction of five phthalate esters (PAEs) with relatively lower logP values (2.7-4.9), including diethyl phthalate, diallyl phthalate, dipropyl phthalate, benzylbutyl phthalate and dibutyl phthalate. It exhibited higher extraction recovery (> 65%) and faster extraction kinetics (50 min vs 240 min) for target PAEs over commercial polydimethylsiloxane coated stir bar. Based on the superior performance, PDT coated stir bar sorptive extraction was combined with high-performance liquid chromatography-diode array detection for trace analysis of five PAEs plasticizers. The limits of detection for target PAEs were 0.04-0.27 μg/L, with the enrichment factors of 54-80-fold. The potential of the method was demonstrated by detecting five target PAEs in Chinese liquor and mineral water samples. No target analytes were detected in Chinese liquor sample, and recoveries of 85.4-109% were obtained for target analytes in spiked liquor samples; trace diethyl phthalate (1.19-2.98 μg/L) and dibutyl phthalate (0.77-0.91 μg/L) were detected in two mineral water samples, with recoveries of 85.4-117% and 87.4-117% respectively in spiked mineral water samples.

In this study, spherical imine-linked covalent organic frameworks (COFs) were fabricated from 2,5-dimethoxybenzene-1,4-dialdehyde (DMTP) and 1,3,5-tris (4-aminophenyl) benzene (TAPB) and named as TAPB-DMTP-COFs. The resulting powders were coated onto bare glass bars via physical-adhesion to obtain TAPB-DMTP-COFs coated stir bars. The self-made stir bars exhibited higher extraction efficiency (74–85%) and faster dynamics (50 min) towards non-steroidal anti-inflammatory drugs (NSAIDs) over ethylene glycol-Silicone (42–68%, 180 min) and polydimethylsiloxane (3–61%, 180 min) coated stir bars. Fourier transform infrared (FT-IR) spectra, X-ray photoelectron spectroscopy (XPS), zeta potential and water contact angle were employed to provide a comprehensive understanding of the adsorption mechanism between the coating and analytes. The results displayed that methoxy group worked as an adsorption site helping the adsorption of interest NSAIDs onto the TAPB-DMTP-COFs coating and hydrogen bonds formed between the O atoms and the analytes. Additionally, the adsorption mechanisms possibly also involved π-π interaction and hydrophobic interaction. Moreover, TAPB-DMTP-COFs coated stir bars exhibited good stability and could be reused more than 60 times. Subsequently, a method by combining TAPB-DMTP-COFs coated stir bar sorptive extraction (SBSE) with liquid chromatography (HPLC)-ultraviolet detector (UV) was established for the determination of four NSAIDs in environmental waters. Under the optimized conditions, the established method showed a wide linear range of 0.2/1–500 μg/L for interest NSAIDs, the limits of detection varied from 0.039 to 0.312 μg/L. Yangtze River water, East Lake water and Spring water were subjected to the proposed method, the recoveries in spiked samples were 84.7–104%, 81.2–101% and 82.6–97.6%, respectively.

As an amplification tool, DNA walker nanomachine exhibits promising potential in the fields of biosensing. To further improve amplification efficiency of DNA walker, a number of cascade amplification systems have been developed, but they still suffer from separated steps and long reaction time. Herein, by combining entropy-driven catalytic (EDC) amplification with DNAzyme reaction, a highly integrated cascade three-dimensional (3D) DNA walker is constructed, which allows the autonomous DNA circuit to be completed in one step. Further combined with element tag and inductively coupled plasma mass spectrometry (ICP-MS) detection, the cascade walker can be used for quantification of miRNA-21. Specifically, the reactant of EDC (WLT complex) and the substrate of DNAzyme (Substrate-Tb) are both fixed on magnetic beads (MBs) to assemble the track of walker. Once cascade DNA walker is triggered by target miRNA, EDC amplification runs first and keeps circulating to liberate many DNAzyme strands. DNAzyme can repeatedly cleave Substrate-Tb with autonomously walking along the substrate on MBs. As a result, a great number of fragments of Substrate-Tb, as output signal, are generated within 1 h, resulting in elemental signal response in ICP-MS and reflecting the amount of miRNA-21. A good linear relationship was obtained with miRNA-21 concentration in the range of 0.02−1 nmol L−1, and the detection limit of 5 pmol L−1 was achieved. The recoveries of miRNA-21 in human serum were 92.0–107 %. The developed method provided a novel idea for constructing highly integrated and efficient cascade DNA walker with ICP-MS strategy for bioanalysis.

Polydimethylsiloxane (PDMS)/MIL-100(Fe) coated stir bar was prepared by sol gel technique, and good preparation reproducibility was achieved with relative standard deviations (RSDs) ranging from 2.6% to 7.5% (n=7) and 3.6% to 10.8% (n=7) for bar-to-bar and batch-to-batch, respectively. Compared with commercial PDMS coated stir bar (Gerstel) and PEG coated stir bar (Gerstel), the prepared PDMS/MIL-100(Fe) stir bar showed better extraction efficiency for target triazines compounds. It also exhibited relatively fast extraction/desorption kinetics and long lifespan. Based on it, a method of PDMS/MIL-100(Fe) coated stir bar sorptive extraction (SBSE)-high performance liquid chromatography-ultraviolet detector (HPLC-UV) was developed for the determination of six triazines (simazine, atrazine, prometon, ametryn, prometryne and prebane) in environmental water samples. Several parameters affecting SBSE of six target triazines including extraction time, stirring rate, sample pH, ionic strength, desorption solvent and desorption time were investigated. Under the optimal experimental conditions, the limits of detection (LODs, S/N=3) were found to be in the range of 0.021-0.079μgL-1. The repeatability RSDs were in the range of 2.3-6.3% (n=7, c=0.5μgL-1) and the enrichment factors (EFs) ranged from 51.1 to 102-fold (theoretical EF was 200-fold). The proposed method was applied to the analysis of target triazines in environmental water samples, with recoveries of 98.0-118% and 94.0-107% for spiked East Lake water and local pond water samples, respectively.

Magnetic porous organic polymers (MOPs) with abundant thiol groups were synthesized successfully in high yield through a template-free and catalyst-free diazo-coupling reaction. The reaction was conducted under mild conditions in aqueous solution, in which the introduction of magnetism and thiol-functionalization was realized simultaneously, avoiding the use of environment-unfriendly organic solvents. The magnetic nanoparticles (MNPs) were embedded into hierarchical porous network structures of porous organic polymers (POPs) physically and the magnetism of thiol-functionalized MOPs (MOP-SH) was easily controlled by varying the amount of spiked MNPs. In this work, zeolitic imidazolate framework-8 (ZIF-8)/poly (methyl methacrylate-ethyleneglycol dimethacrylate) (MMA-EGDMA) composite monolith was in situ synthesized on stir bar by one-pot polymerization. Compared with the neat monolith, ZIF-8/poly(MMA-EGDMA) composite monolith has larger surface area and pore volume. It also exhibits higher extraction efficiency for target phytohormones than poly(MMA-EGDMA) monolith and commercial polyethylene glycol (PEG) coated stir bar. Based on it, a method of ZIF-8/poly(MMA-EGDMA) monolith coated stir bar sorptive extraction (SBSE)-high performance liquid chromatography-ultraviolet detection (HPLC-UV) was established for the analysis of five phytohormones in apple and pear samples. The developed method exhibited low limits of detection (0.11-0.51μg/L), wide linear range (0.5-500μg/L) and good recoveries (82.7-111%), which demonstrated good application potential of the ZIF-8/monolith coated stir bar in trace analysis of organic compounds.

We prepared a superhydrophilic/underwater superoleophobic graphene oxide (GO)-coated cotton for wastewater purification by a simple approach to overcome the difficulty of simultaneously removing insoluble oily compounds and water-miscible contaminants from water. GO can be attached onto the surface of cotton fibers by means of hydrogen bonds between oxygen-containing groups of GO and cotton, with a mass fraction of 6.9 wt%. Large numbers of oxygen-containing groups, combined with the inherent three-dimensional network structure, lead to a water contact angle of 0° and an underwater oil contact angle of 170° ± 1° on the GO-coated cotton. Various oil-in-water emulsions, dye wastewaters and even mixtures of both can be economically and effectively purified by using GO-coated cotton. Most of oils can be filtered with removal efficiency of over 99% and dyes can be absorbed with removal efficiency of over 97%. Our results show that the GO-coated cotton can be regenerated and maintain this high removal efficiency after 50 separation cycles. We anticipate that underwater superoleophobic GO-coated cotton would be an ideal wastewater purification material for practical applications.

Advanced reduced graphene oxide/ZnS (RGO/ZnS) hybrids were synthesized via a facile and effective hydrothermal method to enhance the tribological properties of epoxy coatings. Physical characterization of the hybrids indicated that the as-prepared products had the high chemical purity and that the ZnS nanoparticles combined well with RGO nanosheets. Meanwhile, ZnS nanoparticles were uniformly dispersed on RGO nanosheets with the diameters of several decade nanometers. The tribological properties of ZnS, RGO, and RGO/ZnS with epoxy coating were systematically investigated. By comparing the result of tribological study, it was found that RGO/ZnS hybrids had better lubricating properties with lower coefficient of friction and higher wear resistance than those of ZnS or RGO. In addition, the test results indicated that RGO/ZnS hybrids showed good stability with increased in applied load. These excellent tribological properties of RGO/ZnS hybrids were attributed to the excellent synergistic effects between ZnS nanoparticles and RGO nanosheets and to the transfer film that formed on the steel ball. This work paved a way to expand the potential applications of nanoparticles in the tribological field.
