To tackle the dissolution problem of boron carbide particles in silicon infiltration process, carbon-coated boron carbide particles were fabricated for the preparation of the reaction-bonded boron carbide composites. The carbon coating can effectively protect the boron carbide from reacting with liquid Si and their dissolution, thus maintaining the irregular shape of boron carbide particles and preventing the growth of boron carbide particles and reaction formed SiC regions. Furthermore, the nano-SiC particles, originated from the reaction of the carbon coating and the infiltrated Si, uniformly coated on the surfaces of boron carbide particles, thus forming a ceramic skeleton of the nano-SiC particles-coated and -bonded boron carbide particles. The Vickers hardness, flexural strength and fracture toughness of the composites can be increased by 26 %, 45 %, and 37 % respectively, by using carbon-coated boron carbide particles as raw materials.

Isomers sometimes play a fundamental role in the formation of coordination architectures. In this article, bifunctional 5-(n-pyridyl)tetrazole-2-isopropanoic acid (n = 2, 3) has been designed and prepared. Then, reactions of Ca(NO3)2·4H2O with isomeric 5-(n-pyridyl)tetrazole-2-isopropanoic acid (denoted as Hn-pytzipa, n = 2, 3) afforded mononuclear [Ca(2-pytzipa)2(H2O)4] (1) and one-dimensional [Ca(3-pytzipa)2(H2O)2]n (2), respectively. In 1, 2-pytzipa is a bidentate ligand which chelates one Ca(II) via the nitrogen atoms of the pyridine and tetrazole rings while in 2, 3-pytzipa bridges adjacent Ca(II) centers in a μ 1,1,3-COO mode. PEG-5000 (poly(ethyleneglycol-5000)) coated [Ca(2-pytzipa)2(H2O)4] (1) and [Ca(3-pytzipa)2(H2O)2]n (2) nanoparticles (NPs) show toxicity towards Hela cells, but the ligands are non-toxic in nature and 2 is superior to 1. Furthermore, cell migration across a 2D artificial gap indicates that both NPs can inhibit the migration of Hela cells.

Cr(VI)-based compounds pollution have attracted global concern due to serious harm to humans and environment. Hence, it is crucial to exploit an effective technique to eliminate Cr(VI) in water. Herein, we in-situ grown BiOI on graphitic carbon nitride to prepare the BiOI/g-C3N4 (BCN) direct Z-scheme heterojunction by solid phase engineering method at room temperature. Experimental result shown the photocatalytic activity of pure BiOI were obviously enhanced by constructing Z-scheme BCN heterostructure, and BCN-3 heterostructure exhibited the optimal photocatalytic degradation of RhB with 98% yield for 2.5 h and reduction of Cr(VI) with more than 99% yield for 1.5 h at pH = 2. Stability test shows BCN-3 still kept more than 98% reduction efficiency after 6 cycles. In addition, we also studied the reduction mechanism that shown the ·O2− radicals essentially helped to reduce the Cr(VI) in aqueous solution under illumination, verified the direct Z-scheme charge transfer path by X-ray photoelectron spectroscopy (XPS) and the free radical trapping experiment. The work open a new way for rationally designing photocatalyst heterostructure to reduce Cr(VI) to Cr(III).

Chemoresistance is a major challenge for the treatment of cancer with metastasis. We investigated the mechanisms of lipid metabolites involved in drug resistance. Here, metastatic cancer cells isolated from mouse models were resistant to paclitaxel treatment in vitro and in vivo when compared with parental cancer cells. FOXM1, an oncogenic transcriptional factor, was highly expressed in metastatic cancer cells, and overexpression of FOXM1 conferred parental cancer cells resistance to paclitaxel. Lipidomic analysis showed that FOXM1 increased unsaturated triglyceride (TG) and phosphatidylcholine (PC) abundance, which are the main components of lipid droplet (LD). Inhibition of LD formation sensitized cells to paclitaxel. Mechanistically, the enzyme phospholipase D1 (PLD1) was identified as a potential effector target of FOXM1. PLD1 promoted LD accumulation, which reduced the level of reactive oxygen species (ROS) and maintained endoplasmic reticulum (ER) homeostasis in resistant cells with the treatment of paclitaxel. Moreover, inhibition of PLD1 reversed FOXM1-conferred paclitaxel resistance in vitro and in vivo. This study, for the first time, reveals the role of FOXM1-mediated PLD1 in LD accumulation and paclitaxel resistance. Targeting PLD1 or LD formation may help reverse chemoresistance in metastatic cancer cells. Generally, our results identified FOXM1 as a driver of paclitaxel resistance via activation of PLD1 to promote of LD accumulation, which contributes to the maintenace of ER homeostasis when metastatic cancer cells are confronted with ROS induced by paclitaxel.

InGaO3(ZnO)m (m = 1, 2, 3, 4, and 5) ceramics are a series of n-type oxide thermoelectric materials with layered structures and low thermal conductivities. Herein, InGaO3(ZnO)m (m = 1, 2, 3, 4, and 5) ceramics were fabricated by spark plasma sintering (SPS). Two different trends in the thermoelectric properties of the InGaO3(ZnO)m (m = 1, 2, 3, 4, and 5) ceramics were observed depending on the odevity of the m value. The InGaO3(ZnO) sample exhibited a relatively high electrical conductivity and was therefore selected for vacuum annealing to further improve the electrical transport performance. Oxygen vacancy defects were introduced to the matrix during the annealing procedure, which improved the thermoelectric performance. A maximum ZT of 0.45 was obtained at 973 K for the InGaO3(ZnO) sample with a 96 h vacuum annealing treatment, which is 30 times higher than that of the pristine sample.

New amphiphilic carbazole-based dyes assemble in water into deep-blue-emitting, highly fluorescent helical aggregates as observed by transmission electron microscopy and atomic force microscopy. Single crystal X-ray diffraction and NMR spectroscopy reveal that self-complementary, antiparallel H-bonding and π-π stacking interactions are the driving forces for the formation of these dye aggregates.

Bromodomain containing protein 4 (BRD4), as an epigenetic reader, can specifically bind to the acetyl lysine residues of histones and has emerged as an attractive therapeutic target for various diseases, including cancer, cardiac remodeling and heart failure. Herein, we described the discovery of hit 5 bearing 4-phenylquinazoline skeleton through a high-throughput virtual screen using 2,003,400 compound library (enamine). Then, structure–activity relationship (SAR) study was performed and 47 new 4-phenylquinazoline derivatives toward BRD4 were further designed, synthesized and evaluated, using HTRF assay set up in our lab. Eventually, we identified compound C-34, which possessed better pharmacokinetic and physicochemical properties as well as lower cytotoxicity against NRCF and NRCM cells, compared to the positive control JQ1. Using computer-based molecular docking and cellular thermal shift assay, we further verified that C-34 could target BRD4 at molecular and cellular levels. Furthermore, treatment with C-34 effectively alleviated fibroblast activation in vitro and cardiac fibrosis in vivo, which was correlated with the decreased expression of BRD4 downstream target c-MYC as well as the depressed TGF-β1/Smad2/3 signaling pathway. Taken together, our findings indicate that novel BRD4 inhibitor C-34 tethering a 4-phenylquinazoline scaffold can serve as a lead compound for further development to treat fibrotic cardiovascular disease., Graphical abstract In current work, we performed high-throughput virtual screen and structure-based optimization targeting BRD4, leading to 47 new 4-phenylquinazoline derivatives with potent inhibitory activity toward BRD4. The glucagon-like peptide-1 receptor (GLP-1R) has broad physiological roles and is a validated target for treatment of metabolic disorders. Despite recent advances in GLP-1R structure elucidation, detailed mechanistic understanding of how different peptides generate profound differences in G protein-mediated signalling is still lacking. Here we combine cryo-electron microscopy, molecular dynamics simulations, receptor mutagenesis and pharmacological assays, to interrogate the mechanism and consequences of GLP-1R binding to four peptide agonists; glucagon-like peptide-1, oxyntomodulin, exendin-4 and exendin-P5. These data reveal that distinctions in peptide N-terminal interactions and dynamics with the GLP-1R transmembrane domain are reciprocally associated with differences in the allosteric coupling to G proteins. In particular, transient interactions with residues at the base of the binding cavity correlate with enhanced kinetics for G protein activation, providing a rationale for differences in G protein-mediated signalling efficacy from distinct agonists., The glucagon-like peptide-1 receptor (GLP-1R) can be targeted in the treatment of diabetes, obesity and other metabolic disorders. Here, the authors assess the molecular mechanisms of peptide agonists binding to GLP-1R and the responses elucidated by these ligands, including distinct kinetics of G protein activation.

Swine acute diarrhea syndrome coronavirus (SADS-CoV) is a newly discovered enteric coronavirus. We have previously shown that the caspase-dependent FASL-mediated and mitochondrion-mediated apoptotic pathways play a central role in SADS-CoV-induced apoptosis, which facilitates viral replication. However, the roles of intracellular signaling pathways in SADS-CoV-mediated cell apoptosis and the relative advantages that such pathways confer on the host or virus remain largely unknown. In this study, we show that SADS-CoV induces the activation of ERK during infection, irrespective of viral biosynthesis. The knockdown or chemical inhibition of ERK1/2 significantly suppressed viral protein expression and viral progeny production. The inhibition of ERK activation also circumvented SADS-CoV-induced apoptosis. Taken together, these data suggest that ERK activation is important for SADS-CoV replication, and contributes to the virus-mediated changes in host cells. Our findings demonstrate the takeover of a particular host signaling mechanism by SADS-CoV and identify a potential approach to inhibiting viral spread.

Polymer dielectrics have drawn great attentions for applications in advanced electronic devices and power grids because of their high breakdown strength, low dielectric loss, and excellent flexibility. However, the low energy density in polymer dielectric capacitors will hinder the continuous miniaturization of electrical systems. In this work, ultraviolet irradiation is demonstrated to greatly enhance the breakdown strength and energy density of polypropylene. Dramatically improved breakdown strength of 867 MV/m and discharged energy density of 8.0 J/cm3, together with the high energy efficiency of >90%, were simultaneously achieved in polypropylene after ultraviolet irradiation. Our research shows that proper ultraviolet irradiation can effectively improve the energy density of polypropylene without sacrificing its high charge‐discharge efficiency, being potential for applications in power electronics and pulse electric systems.

Nanotherapy based on thermochemotherapy has boomed as a promising alternative for oncotherapy due to the enhanced permeability and retention (EPR) effect. However, a lack of self-targeting capacity prevents nanotherapy from efficiently accumulating in tumor tissue and internalizing into tumor cells, resulting in a suboptimal therapeutic effect. To overcome these bottlenecks, a kind of methotrexate (MTX)-soybean phospholipid (SPC) inclusion complex (MTX-SPC)-modified graphene oxide (CGO) nanotherapy (CGO-MTX-SPC) is constructed by CGO nanosheets as a supporter for MTX-SPC, thereby realizing active-targeting and synergistic thermochemotherapy. As an FDA-approved chemotherapeutic drug, MTX can be regarded as a tumor-targeting enhancer against the folate receptor on account of its similar structure to folic acid (FA). The fabricated CGO-MTX-SPC has a sheet shape with a size of ca. 109 nm and tumor microenvironment-responsive on-demand drug release. It is worth noting that the physiological stability of CGO-MTX-SPC is better than that of CGO while displaying an improved photothermal effect. In addition, CGO-MTX-SPC can specifically recognize tumor cells and then achieve on-demand drug burst release by dual stimuli of internal lysosomal acidity and an external laser. Moreover, in vivo experimental results further demonstrate that CGO-MTX-SPC displays significant enrichment at the tumor location by active targeting mechanisms due to the introduction of MTX-SPC, endowing the synergistic thermochemotherapy effect upon 808 nm laser irradiation and almost thorough tumor elimination while significantly erasing undesirable side effects. Taken together, the design idea of our nanotherapy not only provides a potential tumor-targeting therapeutic strategy but also broadens the drug payload method of two-dimensional nanomaterials.

The ever-increasing need for sustainable development requires advanced battery techniques beyond the current generation of lithium ion batteries. Among all candidates being explored, lithium-sulfur batteries are a very promising system to be commercialized in the near future. Towards this end, the development of lithium sulfide (Li2S) nanocrystal-based cathodes has received tremendous effort and witnessed multiple reviews. Differentiated from the focus on performance improvement in previous reviews, this review summarizes the research progress in line with the approaches of materials synthesis and electrode fabrication. The key chemistry, operation procedure, materials characterizations and performance assessments are all given a balanced description. Moreover, all approaches are collectively analyzed along the lines of six criteria towards practical applications, i.e., electrode performance, materials quality, resources sustainability, production cost, operation procedure, and consumed energy. Finally, some perspective viewpoints on the future research directions are offered.

Porphyrin-based photosensitizers are conventional photodynamic agents that are used clinically. However, their clinical applications have been overshadowed by poor water solubility. In addition, they have weak tumor selectivity, which may cause undesirable side effects. The preparation of novel porphyrin derivatives has been explored for potential applications in photodynamic therapy (PDT). To achieve this goal, lactose-conjugated porphyrin nanoparticles (Lac-Por NPs) have been synthesized and characterized. PDT with Lac-Por NPs exhibited tumor-specific cytotoxicity in lactose receptor-overexpressing HepG2 cells in vitro and in vivo. In summary, we designed and synthesized lactose conjugate porphyrins with enhanced water solubility and tumor selectivity. This work expands the application range of porphyrin-based photosensitizers for cancer treatment.

In this study, a fully biodegradable ultra-light poly(lactic acid)/silicon dioxide (PLA/SiO2) aerogel nanocomposite with ultra-low thermal conductivity was successfully fabricated. PLA used was a produced from lactic acid, where the lactic acid has been produced from carbohydrates. The rheological properties of PLA were enhanced by diphenylmethane diisocyanate (MDI). The foaming properties, cell density, cell size uniformity, mechanical properties and thermal conductivity and thermal diffusivity of PLA were further improved by SiO2 aerogel, and finally the ultra-low density foamed material was prepared by supercritical CO2. The density of PLA foam can be as low as 0.02 g/cm3 and the thermal conductivity as low as 0.02628 W/m·K. The PLA-based composites can be used in many fields such as thermal insulation, vibration damping and packaging, and can be fully biodegradable and sustainable throughout their life cycle, which meets the global trend of energy saving and emission reduction.

Design and construction of a membrane that can achieve selective separation of uranium from spent fuel or seawater is a big challenge in the field of separation science. In this work, 1,3,5-benzenetricarboxylic acid (BTC) and three different nitrates (Zn/Ni/Cu) were used to prepare metal-organic frameworks (BTC-MOFs) with different pore sizes, and then, BTC-MOFs were intercalated into the interlayers of graphene oxide (GO) for preparing the composite membranes which presented selective separation of uranium with strong acid resistance. Composite membranes prepared by Zn/Ni/Cu-BTC-MOFs and GO can achieve the separation between ions of different valence states, and their permeability and selectivity depend on the membrane thickness, the acidity of driving solution, and the pore sizes of MOFs. Importantly, Cu-BTC-MOF-intercalated GO membranes can not only achieve the selective separation of Th4+ and UO22+ with a selectivity of ≈6 but also induce the ultra-high selectively separation of UO22+ and Ce3+ because the rejection rate of Ce3+ is about 100%. Moreover, the Zn-BTC-MOF-intercalated GO membrane shows an excellent selectivity of Th4+ and UO22+ with a selectivity of ≈25, and it may also achieve selective separation of uranium from seawater.

The tea plant, Camellia sinensis (L.) O. Kuntze, is an economically important, perennial woody plant rich in catechins. Although catechins have been reported to play an important role in plant defences against microbes, their roles in the defence of tea plants against herbivores remain unknown. In this study, we allowed the larvae of Ectropis grisescens, a leaf-feeding pest, to feed on the plants, and alternatively, we wounded the plants and then treated them with E. grisescens oral secretions (WOS). Both approaches triggered jasmonic acid-, ethylene- and auxin-mediated signalling pathways; as a result, plants accumulated three catechin compounds: (+)-catechin, epicatechin and epigallocatechin. Not only was the mass of E. grisescens larvae fed on plants previously infested with E. grisescens or treated with WOS significantly lower than that of larvae fed on controls, but also artificial diet supplemented with epicatechin, (+)-catechin or epigallocatechin gallate reduced larval growth rates. In addition, the exogenous application of jasmonic acid, ethylene or auxin induced the biosynthesis of the three catechins, which, in turn, enhanced the resistance of tea plants to E. grisescens, leading to the coordination of the three signalling pathways. Our results suggest that the three catechins play an important role in the defences of tea plants against E. grisescens.

Inflammation usually results in high-level reactive oxygen species (ROS) and reactive nitrogen species (RNS) not only in acidic tissue but also in alkaline tissue. However, noninvasively in vivo monitoring reactive species specifically within alkaline tissue remains a huge challenge. Here we introduce a dual activatable fluorescent probe PN910 located in the second near-infrared window (NIR-II, 900-1700 nm), which shows high selectivity toward H 2 O 2 and OONO - at pH beyond 7.4. Then we verified that PN910 could be used for the real-time, specific and accurate monitoring of cystitis and colitis for living animals. This report presents a unique approach to the development of dual activatable probe for in vivo biosensing.

The temperature-dependent (T-dependent) linewidth (ΓG) and frequency shift (ΔωG) of the G mode provide valuable information on the phonon anharmonicity of graphene-based materials. In contrast to the negligible contribution from electron-phonon coupling (EPC) to the linewidth of a Raman mode in semiconductors, ΓG in pristine graphene is dominated by EPC contribution at room temperature due to its semimetallic characteristics. This leads to difficulty in resolving intrinsic contribution from phonon anharmonicity to ΓG. Here, we probed the intrinsic phonon anharmonicity of heavily-doped graphene by T-dependent Raman spectra based on FeCl3-based stage-1 graphite intercalation compound (GIC), in which the EPC contribution is negligible due to the large Fermi level (EF) shift. The ΔωG and ΓG exhibit a nonlinear decrease and noticeable broadening with increasing temperature, respectively, which are both dominated by phonon anharmonicity processes. The contribution of phonon anharmonicity to ΓG of heavily-doped graphene decreases as the EF approaches to the Dirac point. However, the T dependence of ΔωG is almost independent on EF and qualitatively agrees with the theoretical result of pristine graphene. These results provide a deeper understanding of the role of phonon anharmonicity on the Raman spectra of heavily doped graphene.

Background: Cerebral ischemia-reperfusion injury is caused by a blood reperfusion injury in the ischemic brain and usually occurs in the treatment stage of ischemic disease, which can aggravate brain tissue injury. Objective: Curcumin was reported to exert a good therapeutic effect on neural cells against ischemia- reperfusion injury, However, the mechanism is not clear. Methods: In this study, Oxygen-Glucose Deprivation (OGD) model of fetal rat cerebral cortical neurons and the Middle Cerebral Artery Occlusion (MCAO) model of rats were employed to mimic cerebral ischemia-reperfusion injury in vitro and in vivo, respectively. Results: We confirmed that curcumin has a promotive effect on neuronal proliferation and an inhibitory effect on neuronal pyroptosis. Furthermore, we found that curcumin could improve cerebral infarction. The results of western blotting showed that curcumin down-regulated the expression of nucleotide-binding oligomerization domain-containing protein-, leucine-rich repeats-, and pyrin domain-containing protein 1 (NLRP1), cysteinyl aspartate-specific protease 1 (caspase-1), gasdermin D (GSDMD), IL-1β, IL-6, TNF-α, and iNOS proteins in OGD and MCAO models. NLRP1- dependent neuronal pyroptosis played an important role in cerebral ischemia-reperfusion injury. Conclusion: Curcumin could effectively inhibit NLRP1-dependent neuronal pyroptosis by suppressing the p38 MAPK pathway and therefore exerted neuroprotective effects against cerebral ischemia- reperfusion injury.

Although nanometer-sized aluminum hydroxide clusters (i.e., ϵ-Al13 , [Al13 O4 (OH)24 (H2 O)12 ]7+ ) command a central role in aluminum ion speciation and transformations between minerals, measurement of their translational diffusion is often limited to indirect methods. Here, 27 Al pulsed field gradient stimulated echo nuclear magnetic resonance (PFGSTE NMR) spectroscopy has been applied to the AlO4 core of the ϵ-Al13 cluster with complementary theoretical simulations of the diffusion coefficient and corresponding hydrodynamic radii from a boundary element-based calculation. The tetrahedral AlO4 center of the ϵ-Al13 cluster is symmetric and exhibits only weak quadrupolar coupling, which results in favorable T1 and T227 Al NMR relaxation coefficients for 27 Al PFGSTE NMR studies. Stokes-Einstein relationship was used to relate the 27 Al diffusion coefficient of the ϵ-Al13 cluster to the hydrodynamic radius for comparison with theoretical simulations, dynamic light scattering from literature, and previously published 1 H PFGSTE NMR studies of chelated Keggin clusters. This first-of-its-kind observation proves that 27 Al PFGSTE NMR diffusometry can probe symmetric Al environments in polynuclear clusters of greater molecular weight than previously considered.

Furin is one of the nine-member proprotein convertase family. Furin cleaves proteins with polybasic residues, which includes many viral glycoproteins such as SARS-Cov-2 spike protein. The cleavage is required for the activation of the proteins. Currently, the mechanisms that regulate Furin activity remain largely unknown. Here we demonstrated that Furin is a novel heparin/heparan sulfate binding protein by the use of biochemical and genetic assays. The KD is 9.78 nM based on the biolayer interferometry assay. Moreover, we found that sulfation degree, site-specific sulfation (N-sulfation and 3-O-sulfation), and iduronic acid are the major structural determinants for the binding. Furthermore, we found that heparin inhibits the enzymatic activity of Furin when pre-mixes heparin with either Furin or Furin substrate. We also found that the Furin binds with cells of different origin and the binding with the cells of lung origin is the strongest one. These data could advance our understanding of the working mechanism of Furin and will benefit the Furin based drug discovery such as inhibitors targeting the interaction between heparan sulfate and Furin for inhibition of viral infection. Tuberculous meningitis (TBM) is an incurable disease with high mortality. It is an extrapulmonary tuberculosis caused by mycobacterium tuberculosis which penetrated the blood-brain barrier and infected the meninges. Mycobacterium tuberculosis lurking in the body mainly reside in macrophages. Anti-tuberculous drugs usually can not target the blood-brain barrier and macrophages, the drug concentration in the lesion is low, which cannot effectively kill mycobacterium tuberculosis, making TBM difficult to treat. Targeted drug delivery systems can target drugs to specific nidus. In the study, we constructed a drug delivery system, which was a cell penetrate peptide B6 and phosphatidylserine (PS) modified polyethylene glycol (PEG) nanomaterial to target the blood-brain barrier and to target macrophages. This nanomaterial was a combined anti-tuberculosis drug delivery system encapsulating antituberculosis drugs rifampicin and pyrazinamide, designed to target macrophages in the brain and kill mycobacterium tuberculosis lurking in the macrophages. We have physically characterized the drug delivery system, and verified the bactericidal ability at cellular and animal level. Results have shown that the targeted drug delivery system had a remarkable efficacy to treat TBM in mice.

Rubichaetoglobin A (1), a new cytochalasan alkaloid, together with nine closely related known ones (2-10), were isolated from the ethyl acetate extracts of the endophytic fungus Chaetomium tectifimeti S104 harbored in the root of Rubia podantha Diels. Their structures were elucidated based on comprehensive spectroscopic analysis. All isolated compounds were tested for cytotoxic, antibacterial, and nitric oxide inhibitory activities. The results showed that 2, 4, and 5 possessed moderate cytotoxicity against MDA-MB-231 cells with the IC50 values of 19.14, 11.43, and 10.27 μM, respectively.

Aberrant lipid metabolism has recently been recognized as a new hallmark of malignancy, but the characteristics of fatty acid metabolism in breast cancer stem cells (BCSC) and potential interventions targeting this pathway remain to be addressed. Here, by using the in vitro BCSC models, mammosphere-derived MCF-7 cells and HMLE-Twist-ER cells, we found that the cells with stem cell-like properties exhibited a very distinct profile of fatty acid metabolism compared with that of their parental cancer cells, characterized by increased lipogenesis, especially the activity of stearoyl-CoA desaturase 1 (SCD1) responsible for the production of monounsaturated fatty acids, and augmented synthesis and utilization of the omega-6 arachidonic acid (AA). Suppression of SCD1 activity by either enzyme inhibitors or small interfering RNA (siRNA) knockdown strikingly limited self-renewal and growth of the BCSC, suggesting a key role for SCD1 in BCSC proliferation. Furthermore, elevated levels of SCD1 and other lipogenic enzymes were observed in human breast cancer tissues relative to the noncancer tissues from the same patients and correlated with the pathological grades. Interestingly, treatment of BCSC with omega-3 fatty acids, eicosapentaenoic acid and docosahexaenoic acid, effectively downregulated the expression of the lipogenic enzymes and markedly suppressed BCSC self-renewal and growth. Dietary supplementation of nude mice bearing BCSC-derived tumors with omega-3 fatty acids also significantly reduced their tumor load. These findings have demonstrated that increased lipogenesis is critical for self-renewal and growth of BCSC, and that omega-3 fatty acids are effective in targeting this pathway to exert their anticancer effect.

In this work, a new kind of butterfly-like molecules of oxacalix[2]arene[2]pyrazine (OAP) are reported, which exhibit typical characteristics of aggregation-induced emission (AIE) via the restriction of intramolecular vibration (RIV) mechanism. Unlike any of the reported RIV-type AIE molecules, the synthetic procedures of which are complicated and associated high costs, OAP AIEgens can be synthesized in a facile manner by a one-step catalyst-free reaction using commercially available materials. Notably, OAP AIEgens are ideal ligands for constructing metal-organic frameworks (MOFs) due to their built-in coordination sites of pyrazine groups. OAP-based MOFs exhibit multiple potential applications in reversible gas response, encrypted information storage, and construction of white light-emitting devices. This work enriches limited kinds of RIV-type AIEgens, offers additional selections of bridging ligands for constructing luminescent MOFs and provides a visualized prototype to understand the effect of RIV process on the luminescence property of MOFs.

Mucins are the major proteins that distributed on the intestinal mucosa layer and protect the intestine from pathogens infection. The composition of intestinal mucin O-glycans can affect the health of the gastrointestinal tract in pigs. Porcine intestinal mucosa is widely used as the main raw material of heparin extraction. The heparin extraction residues rich in mucins were usually wasted. The structure of mucin derived O-glycans in porcine intestinal mucosa are currently unknown. In this study, we isolated the mucins from the heparin extraction residues and profiled the O-glycans. After heparin extraction, mucin was digested with trypsin, and separated by strong anion exchange chromatography. The mucin derived O-glycans were release by alkaline β elimination, and analyzed by ultra high performance liquid chromatography-porous graphitized carbon-Fourier transform mass spectrometry (UPLC-PGC-FTMS/MS). Thirty five kinds of O-glycans were identified, most of which were Core 3-derived glycans. In particular, the O-glycans containing sialic acid Neu5Ac accounted for 71.93% of the total O-glycans, which were different from that of other species, including mouse intestine, fish intestine, and porcine colon. The high content sialylated mucin may explain its effect in biological processes. Furthermore, the immunological activity results indicated that the porcine intestinal mucin could promote phagocytosis and proliferation without any cytotoxic effects, which may aid in the development of immunomodulators.

YBa2Cu3O7− x (YBCO) is treated as a high-temperature superconductor material for its excellent performance. In this article, we investigated the superconducting properties of YBCO grown on LaAlO3 (LAO) substrate with Eu0.6Sr0.4BiO3 (ESBO) buffer layers. Though the chemical solution-based method, we get ESBO buffer with tetragonal SrO2 particles precipitated on the surface. The cross section of transmission electron microscopy shows more microstructure details of defects of YBCO films introduced by buffer layers and self-assembled SrO2. It is observed that the critical current density Jc of YBCO grown with buffer layers enhanced approximately 40% compared with those on LAO at a low applied magnetic field. The comparison of fitted curves of YBCO/ESBO with different pinning types shows that it is closer to δl pinning and began to lean to δ stress pinning with temperature arise. The values of pinning forces also get improved. Besides, the analysis of normalized pinning force density at 77 K demonstrates the interplay of different flux pinning mechanisms. Finally, all the results show the effectiveness of ESBO buffer layers on enhancing the performance of YBCO.

Cellulose esters (CEs) are promising biodegradable substitutes for traditional petroleum-based plastic materials. Research on structure–property relationships of CEs is necessary to evaluate their suitability for industrial applications such as food packaging. Cellulose esters with different side-chain lengths were synthesized and studied. Their thermal and moisture barrier properties were characterized. Cellulose triheptanoate (CTH) was proved to have an optimal moisture barrier (WVTR = 0.31 g·mil/day/in.2) and was used to blend with poly(3-hydroxybutyrate-co-3-hydroxyvalerate) (PHBV) and polylactic acid (PLA) bioplastics. CTH addition improved the PLA thermal stability, enhanced the ductility, and increased the moisture barrier by 32%, while it decreased the PHBV thermal stability, weakened the ductility, and reduced the moisture barrier by 90%. We demonstrated that by proper choice of the combination of CE and bioplastic, bioplastic blends with unique and useful synergistic properties can be obtained. These blends can potentially be used for commercial applications, such as biodegradable flexible packaging.

The design of efficient nanostructured catalysts is highly desirable for promoting the hydrogenation of 4-carboxybenzaldehyde (4-CBA), which are the key process of hydrorefining of crude terephthalic acid (CTA) to pure terephthalic acid (PTA). Herein, the Pt-FeNi@C, Pt-Fe@C and Pt-Ni@C catalysts were developed and proved to be active for the selective hydrogenation of 4-CBA. As a result, the excellent 4-hydroxymethylbenzoic acid (4-HMBA) selectivity (85%) and benzoic acid (BA) selectivity (88%) were achieved on Pt-FeNi@C and Pt/C, respectively. In comparison with the commercial Pd/C catalyst, the Pt-FeNi@C catalyst exhibits excellent activity (97%) and reduces the occurrence of decarbonylation. Moreover, the Pt-FeNi@C catalyst exhibits excellent recyclability: the catalytic activity did not decrease significantly after four reaction cycles. In combination with detailed characterization, the Pt-FeNi@C catalyst has porous graphene core-shell architecture and remarkable electron transfer capacity. This work may provide some inspirations for the exploration of new PTA hydrofining catalysts.

Iminodipropionitrile (IDPN)-induced dyskinetic syndrome is characterized by abnormal repetitive involuntary movements with abnormalities in the neuro-transmission. This study explored the mechanism of glutamate (Glu)/γ-aminobutyric acid (GABA)-glutamine (Gln) metabolic circuit in rat dyskinetic syndrome and the possible regulation mechanism of “tiapride (Tia).” Male Wistar rats were assigned to the control group, dyskinetic syndrome group, and Tia group. Dyskinetic syndrome was induced by injecting with 3,3′-iminodipropionitrile for 7 days. Tia group was treated with tiapride, while the control and dyskinetic syndrome groups were gavaged with saline. Eventually the Glu, GABA, and Gln concentrations in striatum were detected using UPLC-3QMS, additionally another amino acid neurotransmitters (aspartate, glycine) were also detected. Expressions of glutamine synthetase (GS), glutamate transporter (EAAT2), glutamate decarboxylase (GAD65/67), and γ-aminobutyric acid transporter protein (GAD-T) were observed using Western blot and real-time polymerase chain reaction. In this article, nickel (Ni) coating was constructed on brass substrate by a simple electrodeposition technique, and myristic acid/TiO2 composite (MTC) modifier was used to modify Ni coating. The chemical composition and surface morphology of MTC confirm that myristic acid (MA) was successfully grafted on TiO2 nanoparticles. As a comparison, the structures of the Ni coatings were modified by MTC or MA. We found that both modified Ni coatings exhibit strong durability, self-cleaning and superhydrophobicity ability and have good prospect of decontamination. After Ni coating had been modified by MTC, the water contact angle and sliding angle of MTC-Ni were > 170° and < 5°, respectively. In addition, the MTC-Ni coating shows improved corrosion resistance, which can effectively retard the corrosion of brass. This novel and facile electrodeposition-modification route is believed to be widely applied to preparing superhydrophobic surfaces on metallic materials.

Ultrahigh charge separation was observed in Bi4O5I2/Bi5O7I two-dimensional (2D)/one-dimensional (1D) hierarchical structures (HSs) constructed by selective growth of 2D monocrystalline Bi4O5I2 nanoplates on the electron-accumulating (100) facet of 1D monocrystalline Bi5O7I nanobelts. In addition to the presence of type-II heterojunction between Bi4O5I2 and Bi5O7I elementary entities in 2D/1D HSs, the type-II (100)/(001) surface heterojunction in Bi5O7I nanobelt substrates was also confirmed by means of density functional theory (DFT) calculations and selective photoreduction/oxidation deposition experiments. The synergistic effect of two kinds of heterojunctions in Bi4O5I2/Bi5O7I 2D/1D HSs endowed them with ultrahigh charge carrier separation and transfer characteristics. In contrast with the control sample (BB40-C) constructed by growing Bi4O5I2 nanoplates on whole four sides of Bi5O7I nanobelts, Bi4O5I2/Bi5O7I 2D/1D HSs demonstrated significantly enhanced charge transfer between Bi5O7I nanobelt substrates and Bi4O5I2 nanoplates, owing to respective electron and hole accumulations on (100) and (001) facets of Bi5O7I substrates caused by (100)/(001) surface heterojunction. The enhanced separation behavior was successfully verified by steady/transient-state photoluminescence, electrochemical techniques, and photocatalytic degradation experiments. Based on the above effective charge separation of Bi4O5I2/Bi5O7I 2D/1D HSs as well as the routine advantages for 2D/1D HSs, such as the excellent charge transport in monocrystalline elementary entities, much higher specific surface area, and enhanced light absorption by multiple reflections, the optimal BB40 HSs demonstrated ultrahigh photocatalytic performance than the control samples, whose apparent rates for Rhodamine B [or tetracycline hydrochloride (TC)] degradation were 7.1 (2.9 for TC), 10.3 (4.7 for TC), and 2.2 (1.7 for TC) times those of pristine Bi5O7I nanobelts, Bi4O5I2 nanoplates, and BB40-C, respectively. It is hoped that this crystal facet selection during the heterostructure construction in this work could provide a new strategy or some enlightenment for the exploration of highly active 2D/1D HSs or other-dimensional heterostructure nanomaterials applied in the fields of photocatalysts, solar cells, sensors, and others.

Combination therapy has attracted extensive interest in alleviating the shortcomings of monotherapy and enhancing the treatment efficacy. In this work, hollow mesoporous silica nanoparticles (HMSNs) play the role of nanocarriers in the delivery of Cu(II)-doped polydopamine (PDA), termed as HMSNs@PDA-Cu, for synergistic therapy. PDA acts as a traditional photothermal agent to realize photothermal treatment (PTT). Chemodynamic therapy (CDT) is realized by the reaction of Cu(II) with intracellular glutathione (GSH), and subsequently, the generated Cu(I) reacts with H2O2 to produce toxic hydroxyl radical (•OH) through a Fenton-like reaction. The photothermal performance of PDA is improved after its coordination with Cu(II). On the other hand, PDA exhibits superoxide dismutase (SOD)-mimicking activity. PDA converts O2•- to H2O2 and improves the production of H2O2, which promotes the therapeutic effect of CDT. Moreover, the high temperature caused by PTT further enhances the yield of •OH for CDT. This nanotheranostic platform perfectly applied to the tumor depletion of mice, presenting great potential for cancer metastasis therapy in vitro and in vivo.

A perfect PtCu nanocube with partial hollow structure was prepared by hydrothermal reaction and its electrocatalytic methanol oxidation reaction (MOR) was studied. The appropriate concentration of shape-control additives KI and triblock pluronic copolymers, poly(ethylene oxide)-poly(propylene oxide)-poly(ethylene oxide) (PEO19-PPO69-PEO19) (P123) play crucial roles in the final product morphology. The PtCu nanocubes can be perfectly in situ immobilizedonto graphene under the action of P123 while the structure and cubic morphologyremain unchanged. The electrochemical tests suggest that the obtained PtCu nanocube (PtCu-NCb) exhibits better MOR activity and stability than PtCu hexagon nanosheet (PtCu-NSt), PtCu nanoellipsoid (PtCu-NEs) and commercial Pt/C in alkaline medium. When in situ immobilized onto graphene, the MOR catalytic activity and stability of PtCu cubes are further improved. The markedly enhanced electrocatalytic activity and durability maybe attributed to the special cubic morphology with partial hollow structure enclosed by highly efficient facet and the probably the synergistic effect of PtCu and intermediate state CuI decorated on the surface and graphene.

Particulate matter of 0.3 μm in diameter (PM0.3) poses a serious threat to the environment and human beings. Ultrathin and -light nanofibrous filters with excellent filtration properties can significantly prevent the detrimental effects of these particles. Here, we develop free-standing polyamide PA-66 ultrafine nanofiber papers for PM0.3 filtration using effective and scalable blow and electro-blow spinning techniques. The smallest average fiber diameter is 61.7 nm, which is 2-3 orders of magnitude smaller than that of conventional textiles. Poly(ethylene terephthalate) nonwovens are selected to fabricate free-standing nanofiber papers of various polymers, including polyamide, poly(methyl methacrylate), poly(vinylpyrrolidone), and poly(ethylene oxide) owing to the smooth surfaces of the nonwovens. This underlying principle can be used to create similar free-standing nanofiber papers from other commodity polymers in the future. Mechanisms of capturing particulate matter with different nanofiber morphologies are discussed. Salt and oil particulates are used to characterize the filtration properties. PA-66 papers are promising reusable filters owing to their mechanical particle-capture mechanism. The blow-spun PA-66 papers show filtration performance of 98.75% efficiency and a pressure drop of 125.44 Pa owing to the "slip" effect caused by the ultrasmall diameter. In the electro-blow spinning process, a supplementary voltage supply is conducive to separating nanofiber bundles into random-oriented nanofibers. Electro-blown spun papers possess an ultrahigh efficiency of 99.99% with a reduced areal density of 0.9 g m-2. These PA-66 papers can be used in a variety of applications, such as reusable personal protective equipment, industrial waste gas treatment, and central ventilation purification systems.

The main objective of this paper is to elucidate the influence of drug-carrier compatibility and preparation method on the properties of Paclitaxel (PTX)-loaded lipid liquid crystalline nanoparticles (LLCNs). Here, glyceryl monooleate (GMO), glycerol monolinoleate (GML), glyceryl monolinolenate (GMLO) were selected as the lipids, and Soluplus, Poloxamer 407 (P407), Tween 80 were selected as the stabilizer to prepare LLCNs. First of all, PTX-carrier compatibility was screened by molecular dynamic simulation using Flory-Huggins interaction parameter as the criteria. Thereafter, PTX-loaded LLCNs were prepared under different energy input conditions and were characterized. Influence of lipid type, stabilizer type, drug-lipid ratio and preparation method on properties of the LLCNs was explored. It was found that both lipid and stabilizer type had significant influence on drug encapsulation efficiency. Compared to the LLCNs prepared under high energy condition, PTX-loaded LLCN prepared under low energy input had higher drug encapsulation efficiency, smaller particle size (211.6 nm versus 346.8 nm) and a sustained release behavior. In conclusion, molecular dynamic simulation is an effective tool to select the most appropriate composition of LLCNs for a specific drug substance, and LLCNs prepared using low energy input methods was particularly applicable for industrial manufacture.

Summary Mutations in HPRT1, a gene encoding a rate-limiting enzyme for purine salvage, cause Lesch-Nyhan disease which is characterized by self-injury and motor impairments. We leveraged stem cell and genetic engineering technologies to model the disease in isogenic and patient-derived forebrain and midbrain cell types. Dopaminergic progenitor cells deficient in HPRT showed decreased intensity of all developmental cell-fate markers measured. Metabolic analyses revealed significant loss of all purine derivatives, except hypoxanthine, and impaired glycolysis and oxidative phosphorylation. real-time glucose tracing demonstrated increased shunting to the pentose phosphate pathway for de novo purine synthesis at the expense of ATP production. Purine depletion in dopaminergic progenitor cells resulted in loss of RHEB, impairing mTORC1 activation. Background and aims The apolipoprotein A-I mimetic peptide D-4F, among its anti-atherosclerotic effects, improves vasodilation through mechanisms not fully elucidated yet. Methods Low-density lipoprotein (LDL) receptor null (LDLr−/−) mice were fed Western diet with or without D-4F. We then measured atherosclerotic lesion formation, endothelial nitric oxide synthase (eNOS) phosphorylation and its association with heat shock protein 90 (HSP90), nitric oxide (NO) and superoxide anion (O2•-) production, and tetrahydrobiopterin (BH4) and GTP-cyclohydrolase 1 (GCH-1) concentration in the aorta. Human umbilical vein endothelial cells (HUVECs) and aortas were treated with oxidized LDL (oxLDL) with or without D-4F; subsequently, BH4 and GCH-1 concentration, NO and O2•- production, eNOS association with HSP90, and endothelium-dependent vasodilation were measured. Results Unexpectedly, eNOS phosphorylation, eNOS-HSP90 association, and O2•- production were increased, whereas BH4 and GCH-1 concentration and NO production were reduced in atherosclerosis. D-4F significantly inhibited atherosclerosis, eNOS phosphorylation, eNOS-HSP90 association, and O2•- generation but increased NO production and BH4 and GCH-1 concentration. OxLDL reduced NO production and BH4 and GCH-1 concentration but enhanced O2•- generation and eNOS association with HSP90, and impaired endothelium-dependent vasodilation. D-4F inhibited the overall effects of oxLDL. Conclusions Hypercholesterolemia enhanced uncoupled eNOS activity by decreasing GCH-1 concentration, thereby reducing BH4 levels. D-4F reduced uncoupled eNOS activity by increasing BH4 levels through GCH-1 expression and decreasing eNOS phosphorylation and eNOS-HSP90 association. Our findings elucidate a novel mechanism by which hypercholesterolemia induces atherosclerosis and D-4F inhibits it, providing a potential therapeutic approach.

Purpose This study explored whether gastrodin (Gas) could attenuate the symptoms of Tourette syndrome(TS) via the regulation of glutamate (Glu), its transporters (EAAT1 and EAAT2) and its receptors (NMDAR1, NMDAR2A and NMDAR2B) in rats. Materials and Methods Seventy-five Wistar male rats were randomly divided into five groups (n=15 each): the control, TS, Tia (tiapride, 25mg/kg), Gas60 (gastrodin, 60mg/kg) and Gas120 groups (gastrodin, 120mg/kg). Rats in all groups except the control group received intraperitoneal injection of 3,3′-iminodipropionitrile (IDPN) for 7 consecutive days to establish the TS model. Thereafter, rats in the Tia, Gas60, and Gas120 groups were gavaged with 25mg/kg Tia, 60mg/kg Gas and 120mg/kg Gas for 28 days. Rats in the control and TS groups were gavaged with 0.9% normal saline. Behavioral evaluation was performed by using stereotypy scoring, nodding experiment and autonomic activity test. The Glu level was measured by UPLC-QqQ-MS analysis. The expression of EAAT1, EAAT2, NMDAR1, NMDAR2A and NMDAR2B was measured by Western blot and quantitative real-time PCR (qRT-PCR) analyses. Results The results showed that rats with IDPN-induced TS exhibited an increase in stereotypy score, nodding numbers, number of times to enter the central area and autonomic total distance, which could be improved by Tia and Gas treatments. Furthermore, Tia and Gas treatments significantly decreased the IDPN-induced the increase in Glu levels in rats with TS. Furthermore, the decreased expression of EAAT1 and EAAT2 and increased expression of NMDAR1, NMDAR2A, and NMDAR2B in rats with TS induced by IDPN could be substantially altered by Tia and Gas treatments. Conclusion Gas ameliorated the behavioral dysfunction of rats with TS by maintaining Glu at a normal level, upregulating the expression of EAAT1 and EAAT2, and downregulating the expression of NMDAR1, NMDAR2A and NMDAR2B.

Background Tissue-engineered bone grafts (TEBGs) that undergo vascularization and neurotization evolve into functioning bone tissue. Previously, we verified that implanting sensory nerve tracts into TEBGs promoted osteogenesis. However, the precise mechanisms and interaction between seed cells were not explored. In this study, we hypothesized that neurotization may influence the osteogenesis of TEBGs through vascularization. Methods We cultured rat Schwann cells (SCs), aortic endothelial cells (AECs), and bone marrow-derived mesenchymal stem cells (BM-MSCs) and then obtained BM-MSC-derived induced endothelial cells (IECs) and induced osteoblasts (IOBs). IECs and AECs were cultured in an SC-conditioned medium (SC-CM) to assess proliferation, migration, capillary-like tube formation, and angiogenesis, and the vascular endothelial growth factor (VEGF) levels in the supernatants were detected. We established an indirect coculture model to detect the expression of nestin and VEGF receptors in IECs and tissue inhibitor of metalloproteinase (TIMP)-2 in SCs. Then, SCs, IECs, and IOBs were labeled and loaded into a β-tricalcium phosphate scaffold to induce prevascularization, and the scaffold was implanted into a 6-mm-long defect of rat femurs. Three groups were set up according to the loaded cells: I, SCs, and IECs (coculture for 3 days) plus IOBs; II, IECs (culture for 3 days) plus IOBs; III, IOBs. Nestin and TIMP-2 expression and osteogenesis of TEBGs were evaluated at 12 weeks post-implantation through histological and radiological assessments. Results We found that SC-CM promoted IEC proliferation, migration, capillary-like tube formation, and angiogenesis, but no similar effects were observed for AECs. IECs expressed nestin extensively, while AECs barely expressed nestin, and SC-CM promoted the VEGF secretion of IECs. In the coculture model, SCs promoted nestin and VEGF receptor expression in IECs, and IECs inhibited TIMP-2 expression in SCs. The promotion of prevascularized TEBGs by SCs and IECs in group I augmented new bone formation at 6 and 12 weeks. Nestin expression was higher in group I than in the other groups, while TIMP-2 expression was lower at 12 weeks. Conclusions This study demonstrated that SCs can promote TEBG osteogenesis via IECs and further revealed the related specific characteristics of IECs, providing preliminary cytological evidence for neurotization of TEBGs.

Mulberry pomace (MP) is a rich source of natural phenolic compounds. Ultrasonic-assisted extraction coupled with an orthogonal array design was employed for efficient extraction of the phenolic compounds. The optimized extraction conditions were using a solid (viz. MP) to liquid (75% ethanol, pH 3) ratio of 1: 20, and extracting at 50 °C for 90 min with ultrasonic power of 180 W. The total phenolic content (TPC) and DPPH scavenging capacity (EC50) of the extract were 69.914 ± 0.164 mg GAE/g sample and 0.078 ± 0.006 mg/mL, respectively. The MP extract (MPE) was subsequently encapsulated by extrusion technique in calcium alginate beads and the TPC and encapsulation efficiency for the lyophilized beads which initially stored in concentrated MPE reached 81.456 ± 0.333 mg GAE/g dry weight of beads and 43.40% ± 0.18%, respectively. The in vitro release profiles of phenolic compounds in water from encapsulated wet and lyophilized calcium alginate beads showed prolonged release behavior during 180-min dissolution, which followed Fickian diffusion mechanism. During 30-day storage at 4 °C, lyophilized beads showed favorable stability in the TPC, implying that they played a protective role on the encapsulated phenolic compounds. In conclusion, lyophilized calcium alginate beads loaded with MPE could be used as carriers of natural phenolics and nutraceutical ingredients.

In this work, isatin was employed as the scaffold to design aldose reductase inhibitors with antioxidant activity. Most of the isatin derivatives were proved to be excellent in the inhibition of aldose reductase (ALR2) with IC 50 values at submicromolar level, and (E)-2-(5-(4-methoxystyryl)-2,3-dioxoindolin-1-yl) acetic acid (9g) was identified as the most effective with an IC 50 value of 0.015 μM. Moreover, compounds 9a-h with styryl side chains at the C5 position of isatin showed potent antioxidant activity. Particularly, the phenolic compound 9h demonstrated similar antioxidant activity with the well-known antioxidant Trolox. Structure-activity relationship and molecular docking studies showed that the acetic acid group at N1 and C5 p-hydroxystyryl side chain were the key structures to increase the aldose reductase inhibitory activity and antioxidant activity.
