Migratory connectivity describes the degree to which migratory populations are mixed across space and time. Unlike adults, subadult birds often exhibit distinct migratory patterns and continually refine their migratory behaviour and destinations as they mature. Consequently, the influence of subadult movements on overall migratory connectivity might be different from that of adults. However, current studies on migratory connectivity often overlook population age structures, predominantly focusing on adults. In this study, we investigated the role of subadult movements in shaping population level connectivity by using satellite tracking data from 214 black-necked cranes, Grus nigricollis , in western China. We first assessed the variances in spatial separation in different age cohorts using the continuous temporal Mantel correlation coefficient with data from 17 juveniles tracked in the same year for 3 consecutive years. We then calculated the continuous temporal migratory connectivity for the entire population (comprising various age groups) from 15 September to 15 November and compared the result to that of the family group (consisting of juveniles and adults only). Our results revealed a positive correlation between temporal variation in spatial separation and age after the juveniles separated from the adults, suggesting that subadults may have fine-tuned their migration paths. Moreover, the migratory connectivity of the all-age cohort was moderate (below 0.6) in the winter season, and notably lower than that of the family group during the autumn period. Given the considerable impact of subadults on migratory connectivity, we recommend using data collected from birds across all age categories to improve the precision of population level migratory connectivity estimates. • Black-necked cranes have moderate migratory connectivity. • Spatial separation between individuals varied between age groups. • Individuals of all ages should be included when calculating migratory connectivity. Background: Migratory birds exhibit heterogeneity in foraging strategies during wintering to cope with environmental and migratory pressures, and gut bacteria respond to changes in host diet. However, less is known about the dynamics of diet and gut fungi during the wintering period in black-necked cranes (Grus nigricollis). Results: In this work, we performed amplicon sequencing of the trnL-P6 loop and ITS1 regions to characterize the dietary composition and gut fungal composition of black-necked cranes during wintering. Results indicated that during the wintering period, the plant-based diet of black-necked cranes mainly consisted of families Poaceae, Solanaceae, and Polygonaceae. Among them, the abundance of Solanaceae, Polygonaceae, Fabaceae, and Caryophyllaceae was significantly higher in the late wintering period, which also led to a more even consumption of various food types by black-necked cranes during this period. The diversity of gut fungal communities and the abundance of core fungi were more conserved during the wintering period, primarily dominated by Ascomycota and Basidiomycota. LEfSe analysis (P < 0.05, LDA > 2) found that Pyxidiophora, Pseudopeziza, Sporormiella, Geotrichum, and Papiliotrema were significantly enriched in early winter, Ramularia and Dendryphion were significantly enriched in mid-winter, Barnettozyma was significantly abundant in late winter, and Pleuroascus was significantly abundant in late winter. Finally, mantel test revealed a significant correlation between winter diet and gut fungal. Conclusions: This study revealed the dynamic changes in the food composition and gut fungal community of black-necked cranes during wintering in Dashanbao. In the late wintering period, their response to environmental and migratory pressures was to broaden their diet, increase the intake of non-preferred foods, and promote a more balanced consumption ratio of various foods. Balanced food composition played an important role in stabilizing the structure of the gut fungal community. While gut fungal effectively enhanced the host's food utilization rate, they may also faced potential risks of introducing pathogenic fungi. Additionally, we recongnized the limitations of fecal testing in studying the composition of animal gut fungal, as it cannot effectively distinguished between fungal taxa from food or soil inadvertently ingested and intestines. The black-necked crane is the only species of crane that lives in the high-altitude region of the Tibet Plateau. At present, there is little research on viral diseases of the black-necked crane (Grus nigricollis). In this study, a viral metagenomic approach was employed to investigate the fecal virome of black-necked cranes in Saga County, Shigatse City, Tibet, China. The identified virus families carried by black-necked cranes mainly include Genomoviridae, Parvoviridae, and Picornaviridae. The percentages of sequence reads belonging to these three virus families were 1.6%, 3.1%, and 93.7%, respectively. Among them, one genome was characterized as a novel species in the genus Grusopivirus of the family Picornaviridae, four new parvovirus genomes were obtained and classified into four different novel species within the genus Chaphamaparvovirus of the subfamily Hamaparvovirinae, and four novel genomovirus genomes were also acquired and identified as members of three different species, including Gemykroznavirus haeme1, Gemycircularvirus ptero6, and Gemycircularvirus ptero10. All of these viruses were firstly detected in fecal samples of black-necked cranes. This study provides valuable information for understanding the viral community composition in the digestive tract of black-necked cranes in Tibet, which can be used for monitoring, preventing, and treating potential viral diseases in black-necked cranes. The Guizhou Caohai Wetland plays a crucial role as a wintering site for migratory birds in the Yunnan–Guizhou Plateau and ranks among the largest wintering spots globally for black-necked cranes (Grus nigricollis). To better understand the factors influencing waterbird populations and ecosystem changes, we conducted a comprehensive analysis of historical waterbird population variations over a 30-year period spanning from 1992 to 2022. The current investigation revealed a downward trend in the abundance index curve of the total number of waterbirds during this observation period. Among the five waterbird guilds examined, dabbling ducks, diving ducks, and coots (Fulica atra) experienced declines in their populations, while wading birds and shorebirds saw an increase. Moreover, we observed a rise in species richness within the community over time, accompanied by smaller compositional changes. Additionally, the findings indicated positive growth trends in wintering endangered species such as black-necked cranes and common cranes (Grus grus) in Caohai. Furthermore, we observed an increase in the occurrence and persistence of rare species, such as Eurasian spoonbills (Platalea leucorodia), black-faced spoonbills (Platalea minor), and black storks (Ciconia nigra) wintering in Caohai. These occurrences suggest that the wetland environment provides favorable conditions for a diverse range of species. Despite the rise in species richness, these trends in the abundance and species composition of wintering waterbirds over the past thirty years are still of concern. Habitat suitability assessment is critical for wildlife population conservation and management planning. The MaxEnt model is widely used in species habitat suitability modeling. In order to investigate the habitat status of the black-necked crane in the Zoige grassland wetland ecological function area, this study evaluated the habitat suitability of the black-necked crane using the MaxEnt model with 152 occurrence records and 13 environmental variables. Based on the Akaike information criterion corrected for the small sample size, the best optimal parameter combination (feature class LQPHT, regularization multiplier 3.0) was selected. The results show that the Maxent model had good accuracy with an area under the curve (AUC) value of 0.895. Distance to roads, average summer precipitation, distance to lakes, and altitude are the dominant environmental variables affecting the potential distribution of black-necked cranes, with the contribution rates of 31.6%, 29.8%, 14.3%, and 8.2%, respectively. The suitable habitat area of black-necked cranes is mainly distributed in and around the Zoige Wetland National Nature Reserve, accounting for 46.49% of the Zoige Grassland Wetland National Key Ecological Function Zone. The potential distribution area has a tendency to spread to Hongyuan County in the south of the functional zone, and the unsuitable habitat is mainly distributed in the high-altitude area in the southwest of the functional zone. This study recommends focusing on the distribution area of black-necked cranes around Zoige Wetland National Nature Reserve and Hongyuan County to improve conservation strategies and strengthen protection efforts. Black-necked cranes (Grus nigricollis) are national first-level protected wild animals in China. Artificial breeding has been adopted by many zoos and reserves to achieve ex-situ conservation of black-necked cranes, but the breeding rate of the species in cages is low. This study used non-invasive methods combined with behavioural observations to investigate changes in sex hormones and glucocorticoid metabolites in the droppings of black-necked cranes during the breeding cycle, with the results showing that (i) levels of estradiol and testosterone in black-necked cranes increased significantly when they entered the breeding period, and these levels could be used as an important physiological indicator to effectively monitor the physiological status of females and males during the reproductive period, thus providing a theoretical basis for the timing of semen collection; (ii) the level of progesterone in the mid-reproduction stage was significantly higher than that in other stages in female black-necked cranes after successful mating, and this level could be an effective indicator of the mating status of female black-necked cranes; (iii) droppings' glucocorticoid metabolites in the breeding period showed different dynamics between paired and singly caged black-necked cranes, indicating that the physiological phenomenon of reproduction could result in a certain amount of physiological burden on black-necked cranes. Wintering Black-necked (Grus nigricollis) and Common (G. grus) cranes occur sympatrically in Caohai Wetland, Guizhou, China. Seasonal migration dynamics, daily movement patterns, and diurnal activity budgets of the two crane species were investigated in Caohai Wetland, in order to characterize their wintering activity differences and the mechanism of interspecific coexistence, during October - April 2014 - 2017. Black-necked Cranes wintered in Caohai for 147 ± 8 days, and Common Cranes for 169 ± 8 days. Common Cranes generally arrived in Caohai 11.4 ± 3.28 days earlier and departed 11.00 ± 3.20 days later than Black-necked Cranes. During winter in Caohai Wetland, the behavioral activities of both the Black-necked and Common cranes were strongly influenced by circadian rhythms. However, there were significant differences in daily temporal patterns between them in percent time spent in specific behaviors. Common Cranes left their roosting sites 20 min. earlier than Black-necked Cranes and returned 32 min. later. Common Cranes also spent more time foraging and being vigilant than Black-necked Cranes. Background. The black-necked crane (Grus nigricollis) is a vulnerable species and the only species that lives in the plateau. Five migration routes of different populations have been identified, but for cranes wintering in Nyingchi Prefecture, Tibet, the migration route and breeding/summering area are still unknown. The aim of this study was to investigate the spatio-temporal migration patterns of black-necked cranes in this area and to identify important areas for conservation. Methods. In 2016, we fitted seven black-necked cranes in Nyingchi with GPS-GSM satellite transmitters to record their migration routes. We used ArcGIS 10.2 to visualize important stopover sites and the 'ggplot' function inRto analyze the migration patterns. Results. From March 2016 to May 2019, we recorded nine spring migration and four autumn migration tracks from five individuals. Four individuals spent the breeding/- summering season in Qinghai Lake, while the other spent the breeding/summering season in the Jinzihai Wetland of Dulan County, Qinghai Province. Detailed spatiotemporal information showed that the spring migration lasted 8.7 ± 4.6 days and covered 1,182.5 ± 90.4 km, while the autumn migration lasted 30 ± 10.6 days and covered 1,455.7 ± 138 km. Basom Lake and the Shazhuyu River were the most important stopover sites during the spring and autumn migrations, respectively. The cranes spent 4.4 ± 3.7 days in Basom Lake and 26.3 ± 10.7 days in the Shazhuyu River. The black-necked cranes mainly migrated during the daytime (>85 % of the fly points), and 81 % (17/21) of all stopover and roosting sites were in the valley or at lakeside swamps. Only 17.7% (516 / 2,914) of the data points for stopover and roosting sites were in protected areas. Main conclusions. Our study revealed the breeding/summering areas and migration routes of the black-necked cranes wintering in Nyingchi. The black-necked crane (Grus nigricollis) is a vulnerable species, breeding exclusively on the high-altitude wetlands of the Qinghai–Tibet Plateau. Bird species harbor diverse communities of microorganisms within their gastrointestinal tracts, which have important roles in the health, nutrition, and physiology of birds. Hitherto, virtually nothing was known about the gut microbial communities associated with wild black-necked cranes. For the first time, this study characterized the gut microbial community compositions, diversity, and functions of black-necked cranes from six wintering areas in China using the Illumina Miseq platform. The taxonomic results revealed that Firmicutes, Proteobacteria, Actinobacteria, and Bacteroidetes were the four most abundant phyla in the gut of black-necked cranes. At the genus level, 11 genera including Lactobacillus, Pseudomonas, Carnobacterium, Pantoea, Enterococcus, Erwinia, Turicibacter, Bacillus, Phenylobacterium, Sanguibacter, and Psychrobacter were dominant. The differences in the gut microbial community alpha and the beta diversities of black-necked cranes among the six wintering areas were investigated. Furthermore, the representative microbial taxa and their predicted functions in each wintering location were also determined. These data represent the first analysis of the gut microbiome of black-necked cranes, providing a baseline for further microbiological studies and a foundation for the conservation of this bird. Background: Understanding how overwintering birds choose foraging habitats is very important for conservation management. The overwintering Black-necked Crane (Grus nigricollis) feeds on crop remains in farmlands; thus, reasonable conservation management of this type of farmland that surrounds wetlands is critical for the overwintering populations of the Black-necked Crane; however, it is not clear how the Black-necked Crane chooses the foraging land in the farmland. Methods: A thorough field positioning survey of all foraging sites in farmland areas around the Caohai Wetland and a sampling analysis of habitat selection by the Black-necked Crane were conducted during the winters from 2016‒2017 and 2017‒2018. Results: Multiple factors contributed to the selection of foraging habitat in farmlands, i.e., food factors (crop remains and tillage methods) > human disturbance factors (distance to road and settlement) > topography factors (slope aspect), listed according to the strength of influence. Additionally, Black-necked Cranes tend to choose farmland sites where there was no machine tillage, the crop remains were > 500 g/m2, the distance to residences ranged from 100 to 500 m, the distance to roads ranged from 50 to 100 m, and the slopes exhibited western or eastern aspects. As the winters progressed, the volume of the edible crop remains declined, and the influences of the other main factors also changed, i.e., the factors of human disturbance (distance to road and settlement) became less important, while the effect of the food factor (crop remains) was strengthened. Thus, the foraging sites near the road became more important. Conclusion: The farming area surrounding the Caohai Wetland is very important for the overwintering Black-necked Crane. From July to November 2018, 10 Black-necked Crane (Grus nigricollis) juveniles were tracked using GPS-GSM satellite transmitters to study their migration routes and stopover sites in Yanchiwan Nature Reserve, Gansu Province, China. By the end of the autumn migration in November 2018, more than 25,000 GPS locations had been obtained during tracking. Migration routes, migration distances and stopover sites were determined, and the stopover home range was estimated for each individual. Individuals moved away from Yanchiwan during 2-25 October 2018 and migrated through Da Qaidam, Golmud City, Qumarleb County, Zadoi County, Zhidoi County, and Nagqu City. In mid-November 2018, the birds arrived in Linzhou County, Tibet, China to winter. The migration routes of all individuals were the same, and the average migration distance was 1,500 ± 120 km. Da Qaidam Salt Lake was an important stopover site, with an average stopover duration of 27.11 ± 8.43 d, and the average stopover range of the Black-necked Cranes at Da Qaidam was 27.4 ± 6.92 km2. The black-necked crane (Grus nigricollis) which inhabits high-altitude areas has the largest body size of the world's 15 crane species, and is classified as threatened by the IUCN. To support further studies on population genetics and genomics, we present a high-quality genome assembly based on both Illumina and nanopore sequencing. In total, 54.59 Gb Illumina short reads and 116.5 Gb nanopore long reads were generated. The 1.23 Gb assembled genome has a high contig N50 of 17.89 Mb, and has a longest contig of 87.83 Mb. The completeness (97.7%) of the draft genome was evaluated with single-copy orthologous genes using BUSCO. We identified 17,789 genes and found that 8.11% of the genome is composed of repetitive elements. In total, 84 of the 2,272 one-to-one orthologous genes were under positive selection in the black-necked crane lineage. SNP-based inference indicated two bottlenecks in the recent demographic trajectories of the black-necked crane. The genome information will contribute to future study of crane evolutionary history and provide new insights into the potential adaptation mechanisms of the black-necked crane to its high-altitude habitat. • When aquatic plants lacking, potatoes become the main food supplement of the two cranes. • Black-necked cranes adapt to new food by exchanging gut microbiota with common cranes. • Common cranes strengthen the metabolism of amino acids and lipids to adapt food changes. Wintering migratory birds have to travel thousands of kilometers between their breeding and wintering grounds because of their distinct habits. They experience complex and changing survival pressures throughout the winter. This study is the first to investigate the overwintering strategies of sympatric foraging black-necked cranes and common cranes from the perspectives of food and gut microbiota. Non-invasive sampling technique was used to collect stool samples. Sanger sequencing was used to determine the origin of the crane species identified from the samples. The analysis of plant-based food, the gut microbiota, and their interactions in the faeces was performed using high-throughput sequencing technology. It was found that in environments with limited aquatic vegetation, both the black-necked crane and the common crane increased their consumption of potatoes, accounting for more than 50% of their diets. Black-necked cranes also competed with common cranes for food, and they quickly adapted to changes in diet by horizontally transmitting their gut microbiota. To meet body needs, the common cranes—which could not compete with the black-necked cranes—constantly searched for new food sources. They increase the metabolism of amino acids, lipids and these metabolites to adapt to the competitive pressures and dietary changes. This study advances our knowledge of how diet and gut microbiota interact in plateau wetland birds, examining the competitive tactics of interwintering black-necked and common cranes', and provides recommendations for the re-vegetation in the Caohai wetlands and artificial feeding of the two crane species. Background: The behavior of cranes reflects many of their survival strategies, but little has been known of the incubation strategies of cranes, in which both parents share incubation duties, in response to cold temperatures in alpine environments. The lack of information may restrict the effective conservation of the threatened Black-necked Crane (Grus nigricollis), a biparental bird nesting in high elevation wetlands. Methods: We directly observed and used infrared video cameras from 2014 to 2015 to study the incubation behavior and quantitatively measured the frequency and details of egg turning behavior in the Black-necked Crane at the Yanchiwan National Nature Reserve in western Gansu Province, China. Results: At lower ambient temperatures in the morning, crane parents spent more time on the nest with less recess frequency and prolonged on-bout duration, while at higher temperatures around noon, the parents had more frequent recesses from incubation and shorter periods between nest exchanges. They adjusted the amount of time incubating by varying the recess frequency and the length of on-bout duration. Mean nest attendance and egg turning frequency of females were significantly higher than those of the males. The nest attendance and on-bout duration of females showed a significantly negative relationship with those of males. The two parents responded differently to the change of temperature. Females spent more time on the nest at lower morning temperatures, while males increased their time on the nest at higher temperatures after noon. Higher incubation recess frequency and egg turning frequency were observed at noon, probably because parents spent more time foraging, taking advantage of the lower egg cooling rate. Conclusion: Both Black-necked Crane parents in the alpine environment adjusted their behavior in response to the thermal requirements of eggs and the weather conditions experienced. Our findings demonstrate that parents of this species incubated in different but complementary ways and efficiently enhanced egg care in a dynamic environment, so as to maximize benefits from the warm portion of the day and the intense solar radiation while minimizing the cost of rewarming eggs and the risks of cooling eggs. The article presents the results of a study on how the level of vigilance of Black-necked Cranes (Grus nigricollis) is being affected by flock size and position in flock during the winter season. The subject of this study are the Black-necked Cranes wintering at Dashanbao Nature Reserve in southwestern China. The study found that increase in flock size for Black-necked Cranes in winter resulted to increased number of individuals preferring central positions in flock.

We equipped five Black-necked Cranes (Grus nigricollis) with satellite transmitters between February and November 2009 to investigate their migration routes between breeding areas and wintering area at Napahai Marsh (3,260 m asl), China. We identified the Shahili Mountain region (southwest Sichuan), including Daocheng, Litang, Baiyu, and Xinlong counties as a new breeding area with a mean elevation of 4,330 m asl. Four of five tracked cranes spent the summer in Daocheng County. The fifth crane was there briefly and then moved north to Baiyu and Xinlong counties. The distance between Napahai Marsh and Daocheng County (~180 km) is one of the shortest migration routes among crane species, but covered an elevation increase of ~ 1,200 m. The migration route of the fifth crane was ~400 km in length and occurred over 2 or 5 days in spring 2009 and 2010, respectively, and 19 days in fall 2009 with five stopovers. [ABSTRACT FROM AUTHOR]
