Organic and polymer fluorescent nanomaterials are a frontier research focus. Here in this work, a series of fluorinated zwitterionic random copolymers end‐attached with a quasi‐chromophoric group of pyrene or tetraphenylethylene (TPE) are well synthesized via atom transfer radical polymerization with activators regenerated by electron transfer (ARGET ATRP). Those random copolymers with total degree of polymerization 100 or 200 are able to produce fluorescent single‐chain nanoparticles (SCNPs) through intra‐chain self‐folding assembly with quite uniform diameters in the range of 10–20 nm as characterized by dynamic light scattering and transmission electron microscopy. By virtue of the segregation or confinement effect, both SCNPs functionalized with pyrene or TPE group are capable of emitting fluorescence, with pyrene tethered SCNPs exhibiting stronger fluorescence emission reaching the highest quantum yield ≈20%. Moreover, such kind of fluorescent SCNPs manifest low cytotoxicity and good cell imaging performance for Hela cells. The creation of fluorescent SCNPs through covalently attached one quasi‐chromophore to the end of one fluorinated zwitterionic random copolymer provides an alternative strategy for preparing polymeric luminescence nanomaterials, promisingly serving as a new type of fluorescent nanoprobes for biological imaging applications. Ferric ion (Fe3+) serves as an essential and indispensable inorganic element in various biological process. Excess Fe3+ is harmful for biological systems and human health. Herein, highly sensitive carbon quantum dots (Fe-CQDs) for ferric ions were synthesized by a high-efficiency, one-step hydrothermal route utilizing L-arginine and citric acid. The prepared Fe-CQDs exhibited good water solubility, high photostability, and bright blue florescence with relatively high quantum yields (QYs) over 21.33% and a Stokes shift exceeding 100 nm. The functionalized Fe-CQDs offer rapid, sensitive, and selective recognition of Fe3+ with a linear relationship from 0.6 to 80 μM and a low detection limit of 0.2 μM. Moreover, the synthesized Fe-CQDs illustrated strong anti-interference properties in samples and buffer solutions. The developed sensor demonstrated high biocompatibility and was successfully applied for intracellular Fe3+ determination in SHSY5Y and HT22 cells, illustrating excellent imaging application. PEGylated 2-(2-(6-(dimethylamino)benzooxazol-2-yl)vinyl)quinolin-8-ol (BQ-mPEG) was synthesized, which exhibited strong fluorescence at 610 nm (φf = 0.11) and a large Stoke's shift (Δλ = 202 nm) in the aqueous solution. The fluorophore displayed a two-photon absorption characteristic and a strong red emission (λem = 625 nm) was observed with NIR (950 nm) irradiation. The feasibility of using BQ-mPEG as a fluorescent imaging reagent suggested that the fluorophore targets mitochondria, and with 950 nm excitation, bright fluorescence images in HeLa live cells were observed. Synthesis and photophysical properties of a fluorescent probe HBD is described. Probe upon interaction with metal ions, anions and nucleoside pyrophosphates (NPPs) showed fluorescence quenching with Cu2+ due to chelation enhanced quenching effect (CHEQ). Moreover, interaction of ensemble HBD.Cu2+ with anions and NPPs showed fluorescence “turn‐On” response with ATP selectively. “On‐Off‐On” responses observed with Cu2+ and ATP is attributed to an interplay between ESIPT and TICT processes. Cyclic voltammogram of probe exhibited quasi‐reversible redox behaviour with three oxidation and two reduction potentials and the change in band gaps of probe suggested the interaction with Cu2+ and ATP. The 2 : 1 and 1 : 1 binding stoichiometry for an interaction between probe and Cu2+ (LOD, 62 nM) and ensemble, HBD.Cu2+ with ATP (LOD, 0.4 μM) respectively are realised by Job's plot and HRMS data. Cell imaging studies carried out to detect Cu2+ and ATP in HeLa cells. Also, the output emission observed with Cu2+ and ATP is utilized to construct an implication (IMP) logic gate. Test paper strips showed naked‐eye visible color responses to detect Cu2+ and ATP. In real water samples probe successfully detected copper (0.03 μM) between 5–6.5 ppb level (ICP‐MS method). Monitoring hydrogen sulfide (H2S) in living organisms is very important because H2S acts as a regulator in many physiological and pathological processes. Upregulation of endogenous H2S concentration has been shown to be closely related to the occurrence and development of tumors, atherosclerosis, neurodegenerative diseases and diabetes. Herin, a novel fluorescent probe HND with aggregation-induced emission was designed. Impressively, HND exhibited a high selectivity, fast response (1 min) and low detection limit (0.61 μM) for H2S in PBS buffer (10 mM, pH = 7.42). The deployment of small‐molecule fluorescent agents plays an ever‐growing role in medicine and drug development. Herein, we complement the portfolio of powerful fluorophores, reporting the serendipitous discovery and development of a novel class with an imidazo[1,2‐a]pyridinium triflate core, which we term PyrAtes. These fluorophores are synthesized in a single step from readily available materials (>60 examples) and display Stokes shifts as large as 240 nm, while also reaching NIR−I emissions at λmax as long as 720 nm. Computational studies allow the development of a platform for the prediction of λmax and λEm. Furthermore, we demonstrate the compatibility of these novel fluorophores with live cell imaging in HEK293 cells, suggesting PyrAtes as potent intracellular markers. [ABSTRACT FROM AUTHOR]

Owing to their high significance in fundamental study and diverse applications, stimuli-responsive and fluorescent polymers, particularly those with cluster-triggered emission (CTE) featured by non-conjugated chromophores, have drawn tremendous attention in recent years. In this work, fluorescent and multi-responsive polysiloxane (FRPS) was synthesized by hydrolytic condensation polymerization of 3-aminopropyl methyl diethoxysilane (APMS) with 3-(N-isopropyl propionamide) iminopropyl methyl diethoxysilane (APMS-NIP), which was formed in situ through aza-Michael addition between APMS and N-isopropyl acrylamide. FRPS was not only highly sensitive to temperature, pH and CO2 in water, but also showed an enhanced and stimuli-adjustable fluorescence emission. The effects of monomer feeding, pH and CO2 on its lower critical solution temperature and fluorescent property were investigated. FRPS fluorescence emission was ascribed to CTE mechanism. In addition, FRPS was shown to be highly potential as physiological indicator for cell imaging, and for controlled release and trace detection of doxorubicin. This study provides therefore a type of stimuli-responsive and fluorescent material for potential applications in biomedical fields, and it is also of great significance for understanding of the fluorescence mechanism of polysiloxane-based stimuli-responsive polymers. The development of reliable probe technology for the detection of bisulfite (HSO 3 -) in situ in food and biological samples is contributing significantly to food quality and safety assurance as well as community health. In this work, a responsive probe, EHDI, is developed for ratiometric fluorescence detection of HSO 3 - in aqueous solution, meat samples, and living cells. The probe is designed based on the HSO 3 - triggered 1,4-addition of electron deficit C = C bond of EHDI. As a result of this specific 1,4-addition, the π-conjugation system was destructed, resulting in blue shifts of the emission from 687 to 440 nm and absorption from 577 to 355 nm. The probe has good water solubility, high sensitivity and selectivity, allowing it to be used for imaging of HSO 3 - internalization and production endogenously. The capability of probe EHDI for HSO 3 - was then validated by traditional HPLC technology, enabling accurately detect HSO 3 - in beef samples. In this paper, nitrogen-doped carbon dots (NCDs) were synthesized via a facile hydrothermal method using gum arabic (GA) and tris(hydroxymethyl)methyl aminomethane (tris) as precursors. The resultant NCDs show uniform size distribution and stable optical performance. Then a fluorescent signal quenching probe based on NCDs was proposed for the quantitative detection of mercury ions (Hg2+). The developed fluorescent nanoprobe based on NCDs demonstrated a good linear relationship in the range of 10 to 120 μM (R² = 0.99265) for toxic mercury(II), with a detection limit of 4.87 μM. Additionally, the present assay system could be efficiently used for the detection of Hg2+ in real water samples with a satisfactory recovery rate (96.11-104.04%). The metal-free NCDs, which exhibited little cytotoxicity, were successfully applied to in vitro fluorescence imaging of human triple negative breast cancer (TNBC) cells, achieving visualization of Hg2+ in cells. With good water solubility and biocompatibility, the nitrogen-doped carbon dots synthesized by this economical and ecologically friendly process are suitable for use in analytical testing and biological research. The precise release and characterization of nitroxyl (HNO) gas signaling molecule remain a challenge due to its short lifetime to date. To solve this issue, an azobenzene-based HNO donor (Azo-D1) was proposed as a colorimetric and fluorometric chemosensor for HNO releasing, to release both HNO and an azobenzene fluorescent reporter together. Specifically, the Azo-D1 has an HNO release half-life of ∼68 min under physiological conditions. The characteristic color change from the original orange to the yellow color indicated the decomposition of the donor molecule. In addition, the stoichiometry release of HNO was qualitatively and quantitatively verified through the classical phosphine compound trap. As compared with the donor molecule by itself, the decomposed product demonstrates a maximum fluorescence emission at 424 nm, where the increase of fluorescence intensity by 6.8 times can be applied to infer the real-time concentration of HNO. Moreover, cellular imaging can also be achieved using this Azo-D1 HNO donor through photoexcitation at 405 and 488 nm, where the real-time monitoring of HNO release was achieved without consuming the HNO source. Finally, the Azo-D1 HNO donor would open a new platform in the exploration of the biochemistry and the biology of HNO. A new structurally simple fluorescent CP probe based on chromone was designed and synthesized, and its structure was fully characterized using various analytical techniques. The CP probe displays a high selectivity and sensitivity for sensing Fe3+ with a "turn-off" fluorescence response over other metal ions in a DMSO/H2O (4:1, v/v) solution. The experiment results show that the CP probe is stable over a wide pH range of 2.0–12.0. The detection limit for Fe3+ was calculated to be 0.044 μmol•L−1. The molar ratio method indicated that the binding mode between the CP probe and Fe3+ is a 1:1 complex formation. HR-MS and density functional theory (DFT) calculations were also performed to further confirm the recognition mechanism. Both fluorescence imaging experiments and the MTT assay demonstrated that the CP probe was suitable for detecting intracellular Fe3+ and no significant cytotoxicity in living cells. Standing wave (SW) microscopy is a method that uses an interference pattern to excite fluorescence from labelled cellular structures and produces high‐resolution images of three‐dimensional objects in a two‐dimensional dataset. SW microscopy is performed with high‐magnification, high‐numerical aperture objective lenses, and while this results in high‐resolution images, the field of view is very small. Here we report upscaling of this interference imaging method from the microscale to the mesoscale using the Mesolens, which has the unusual combination of a low‐magnification and high‐numerical aperture. With this method, we produce SW images within a field of view of 4.4 mm × 3.0 mm that can readily accommodate over 16,000 cells in a single dataset. We demonstrate the method using both single‐wavelength excitation and the multi‐wavelength SW method TartanSW. We show application of the method for imaging of fixed and living cells specimens, with the first application of SW imaging to study cells under flow conditions. LAY DESCRIPTION: With a standard microscope, it is possible to interfere two or more beams of light and produce a contour map of cell structures using a technique known as standing wave microscopy. This method is usually applied with high power objective lenses, and only a tiny area can be studied, usually showing structural detail from one or two cells at most. The Mesolens is a unique objective lens that has been designed and manufactured to image large populations of cells in a single image. Two‐dimensional images of cells can be obtained with the Mesolens using regular illumination methods, but the cell topography is lost. Here we have combined standing wave illumination with the Mesolens to perform topographical imaging of large numbers of cells in a single image. We first proved the method with a non‐biological specimen comprised of a glass lens, and then we applied the technique to imaging of fixed and living cells: in the case of red blood cells, we showed that we can study the topography of more than 16,000 cells in a single image. With the much larger area of study afforded by the Mesolens, we also used standing wave illumination for the first time monitor red blood cell topography when the cells are subject to flow environment. Biologically‐active near‐ultraviolet emitting spirooxindole‐pyrrolidine/pyrrolothiazole‐β‐ketothiolester have been synthesised via Chemo‐/regio‐/stereoselective manner. Excited‐state intramolecular proton transfer (ESIPT) of β‐ketothiolester exhibits excitation‐dependent emission with intermolecular hydrogen bonding ability with solvents. The emission could be tunable between 330 nm to 460 nm. Selected fluorescent compounds 4{4,2,1}, 4{12,2,2}, 5{4,2,1} and 5{12,2,2} shows cytotoxicity effect on PC3 cells, where the highest IC50 value (6 μM) was found for the probe 5{12,2,2}. A one-pot, three-component reaction between isatoic anhydride, 1,3-diketones, and amines/arylhydrazines catalyzed by a combination of molecular I2 and Et3N in open-air affords a series of quinazolin-4(3H)-one derivatives in good yield. The protocol offers an operationally simple, facile, and cost-effective pathway utilizing easily accessible reagents under mild reaction conditions with significant substrate scope. The results of steady state and time-resolved absorption and emission spectroscopy indicate the potential of quinazolin-4(3H)-one derivatives as a fluorescence probe. They exhibit unique photophysical responses in different solvent environments. In addition, they can also act as FRET acceptors during interaction with protein lysozyme. Preliminary biological studies indicate that quinazolin-4(3H)-ones are cell permeable and can be used as a fluorescent probe for bio-imaging of human cervical cancer (HeLa) cells. Organic fluorophores with dual-state emission (DSE) are rare or difficult to observe because most of them display either aggregation-induced emission (AIE) or aggregation-caused quenching (ACQ). Amazing works have been accomplished, yet most of the DSE compounds were excited by UV light which limits their wide application in bioimaging. In this work, we achieved a visible-light excited DSE fluorophore and realized its imaging in SKOV-3 cells and zebrafish. The naphtho[2',3':4,5]imidazo[1,2-a]pyridine (NIP) core ensures its emission in dilute solution. Meanwhile, the twisted phenyl ring blocks fluorescence quenching induced by the π-π stacking and leads to the emission of the solid. The fluorescence intensity is steady even after 6 h of continuous intense sunlight. More importantly, photostability of NIP in cells is much better than commercial dye (mitochondrial green). Fluorescence in situ hybridization (FISH) is a powerful cytogenetic method used to precisely detect and localize nucleic acid sequences. This technique is proving to be an invaluable tool in medical diagnostics and has made significant contributions to biology and the life sciences. However, the number of cells is large and the nucleic acid sequences are disorganized in the FISH images taken using the microscope. Processing and analyzing images is a time-consuming and laborious task for researchers, as it can easily tire the human eyes and lead to errors in judgment. In recent years, deep learning has made significant progress in the field of medical imaging, especially the successful application of introducing the attention mechanism. The attention mechanism, as a key component of deep learning, improves the understanding and interpretation of medical images by giving different weights to different regions of the image, enabling the model to focus more on important features. To address the challenges in FISH image analysis, we combined medical imaging with deep learning to develop the SEAM-Unet++ automated cell contour segmentation algorithm with integrated attention mechanism. The significant advantage of this algorithm is that it improves the accuracy of cell contours in FISH images. Heavy metal contamination has become a global environmental problem and currently drawn much attention from researchers in worldwide. High exposure to heavy metals can lead to problems like kidney, liver and brain damage, skin and lung cancer, etc. Rapid and sensitive detection of heavy metals is of immense importance in environmental monitoring. Over the past few decades, enormous efforts have been made to design various sensors to identify and monitor these harmful metal ions. In Comparison with other methods, fluorescence‐based chemosensors have received extensive attention because of the advantages of high sensitivity, low cost, real‐time monitoring, simple operation, etc. The pyrene derivatives are significant polycyclic aromatic hydrocarbons (PAHs) with strong fluorescence. They are widely used as fluorescent probes in many applications due to their easy modification, high fluorescence quantum yield, and strong fluorescence emission in live cells, outstanding cell permeability, and very low cytotoxicity. The SDG 6: Clean water and sanitation alarm a strong strategy, and careful identification of the risk connected to environmental pollutants needs urgent attention in this situation. Therefore, we highlight different types of pyrene‐based fluorophores and the fluorescence mechanisms for the sensitive and selective detection methodology of Arsenic, Mercury, Cadmium, Lead, and Chromium heavy metals. A light controllable two‐photon near‐infrared (NIR) optical probe is conveniently constructed by multivalent supramolecular co‐assembly of amphiphilic sulfonatocalix[4]arene (SC4A), tricationic triphenylamine (TPA), and diarylethene derivative (DE). The obtained results indicate that the formed SC4A/TPA nanoparticle displays assemble‐confined behaviors and enhances the two‐photon excited NIR emission. Benefiting from the reversible ring‐opened and closed behaviors of the DE under 365 and > 600 nm light irradiation, the dicationic DE in the assembly could act as an energy acceptor after 365 nm light irradiation, then the co‐assembly of SC4A/TPA/DE could achieve a NIR fluorescence ON and OFF switch and is successfully applied in HeLa cell imaging, providing a convenient approach to supramolecular two‐photon switch for cell imaging. Detection of correct oxidation state of a metal ion is important to understand its speciation and redox properties. Some of rhodamine derivatives can identify Al3+, Cr3+ and Fe3+ as the trivalent cations. We have synthesized a rhodamine derivative, HL‐qui, from the reaction of N‐(rhodamine‐6G)lactam‐ethylenediamine with quinoline‐2‐carboxaldehyde for the colorimetric as well as fluorogenic detection of all of metal ions with +3 oxidation state such as Al3+, Cr3+, Fe3+, Ga3+, In3+, Tl3+ and Gd3+. In the presence of these trivalent cations, colorless and nonfluorescent HL‐qui turns pink and strongly fluorescent in HEPES buffer (10 mM) in H2O/ethanol=1/9 (v/v) (pH 7.4) as its spirolactam ring gets open to show the changes in color and fluorescence properties. High sensitivity of the probe towards the cations is reflected from the limit of detection values that are in nM range. To the best of our knowledge, there is no such report where fluorescence sensing has been utilized to determine oxidation state. This multi‐ion sensing feature has been used for fabrication of multi input‐output based logic circuit using Boolean algebraic method. New portable hydrogel sensors for Al3+ and Fe3+ detection were designed based on the aggregation-induced emission (AIE) and color change of N-doped carbon dots (N-CDs). N-CDs with yellow fluorescence were prepared by a one-pot hydrothermal method from 2,5-dihydroxyterephthalic acid and acrylamide. The fluorescence of N-CDs was enhanced by Al3+ about 20 times and quenched by Fe3+. It was interesting that although Fe3+ showed obvious quenching on the fluorescence of N-CDs it did not cause a noticeable change in the fluorescence of N-CDs + Al3+. The colorless solution of N-CDs appeared blue in the presence of Fe3+ without the influence of Al3+. Therefore, the turn-on fluorometry and colorimetry systems based on N-CDs were constructed for the simultaneous detection of Al3+ and Fe3+. Furthermore, the portable sensing of Al3+ and Fe3+ was realized with the assistance of hydrogel, filter paper, cellulose acetate, and cellulose nitrate film. Multiplexed cellular imaging typically relies on the sequential application of detection probes, as antibodies or DNA barcodes, which is complex and time-consuming. To address this, we developed here protein nanobarcodes, composed of combinations of epitopes recognized by specific sets of nanobodies. The nanobarcodes are read in a single imaging step, relying on nanobodies conjugated to distinct fluorophores, which enables a precise analysis of large numbers of protein combinations. Fluorescence images from nanobarcodes were used as input images for a deep neural network, which was able to identify proteins with high precision. We thus present an efficient and straightforward protein identification method, which is applicable to relatively complex biological assays. We demonstrate this by a multicell competition assay, in which we successfully used our nanobarcoded proteins together with neurexin and neuroligin isoforms, thereby testing the preferred binding combinations of multiple isoforms, in parallel. Multiplexed cellular imaging typically relies on the sequential application of detection probes, which is complex and time-consuming. This study describes a novel approach to protein labeling, in which individual proteins are tagged with combinations of multiple epitopes. A dual-mode pH sensor based on nitrogen-doped carbon dots (N-CDs) with the source of o-phenylenediamine and tryptophan has been constructed. Under the stimulation of pH, the N-CDs exhibited prominent both color and fluorescence changes, leading to the rarely discovered colorimetric and fluorescent dual-readouts for the evaluation of pH. The mathematic relationship was established between pH and fluorescence intensity of N-CDs, and between pH and the UV–Vis absorbance ratio at 630 nm and 488 nm of N-CDs, respectively, over a quite broad pH range of 2.2 to 12.0. Multiple techniques are used to explore the dual-mode pH-responsive mechanism, and the preliminary explanation is put forward. The experimental results show that the N-CDs have visualized pH sensing applicability for actual samples, including various water samples and HeLa cell. Furthermore, the N-CD ink is developed for successful information encryption and anti-counterfeiting. • The review explores the world of functional chromophores synthesized via MCRs. • MCR-synthesized functional chromophores are reshaping imaging research. • Bioimaging probes synthesized by MCRs with tunable properties. This review aims to provide a comprehensive overview of recent advancements and applications of fluorescence imaging probes synthesized via MCRs (multicomponent reactions). These probes, also known as functional chromophores, belong to a currently investigated class of fluorophores that are presently being successfully applied in bioimaging experiments, especially in various living cell lineages. We describe some of the MCRs that have been employed in the synthesis of these probes and explore their applications in biological imaging, with an emphasis on cellular imaging. The review also discusses the challenges and future perspectives in the field, particularly considering the potential impact of MCR-based fluorescence imaging probes on advancing this field of research in the coming years. Considering that this area of research is relatively new and nearly a decade has passed since the first publication, this review also provides a historical perspective on this class of fluorophores, highlighting the pioneering works published between 2011 and 2016. The fluorescence of functional dyes was generally quenched in aqueous solution, which hindered their application in water‐bearing detections. In this work, a novel strategy based on host‐guest interaction was provided for the purpose of fluorescence enhancement in aqueous solution and cell imaging. Three adamantane‐modified fluorescent dyes (Coum‐Ad, NP‐Ad, NR‐Ad) with coumarin, 1,8‐naphthalimide and Nile Red as fluorophores were initially designed and prepared. The ((adamantan‐1‐yl)methyl)amino group, as the auxochrome of those dyes, complexed with methylated β‐cyclodextrin (M‐β‐CD) via supramolecular interaction, and then fluorescent supramolecular nanoparticles (FSNPs) were formed by self‐assembly in water. The inclusion equilibrium constant (K) could be as high as 3.94×104 M−1. With the addition of M‐β‐CD, fluorescence quantum yields of these dyes were separately improved to 69.8 %, 32.9 % and 41.3 %. Inspired by the above satisfactory results, six adamantane‐modified probes organelle‐NPAds with organelle‐targeting capability were further obtained. As the formation of hydrogen bonds between organelle‐NPAd2 and M‐β‐CD verified by theoretical calculation, K of organelle‐NPAd2 (5.13×104 M−1~4.53×105 M−1) with M‐β‐CD was higher than that of organelle‐NPAd1 (1.15×104 M−1~3.66×104 M−1) and their fluorescence quantum yields increased to 32.8 %~83.6 % in aqueous solution. Phototoxicity is an important issue in fluorescence live imaging of light-sensitive cellular processes such as mitosis. Among several approaches to reduce phototoxicity, the addition of antioxidants to the media has been used as a simple method. Here, we analyzed the impact of phototoxicity on the mitotic progression in fluorescence live imaging of human cells and performed a screen to identify the most efficient antioxidative agents that reduce it. Quantitative analysis shows that high amounts of light illumination cause various mitotic defects such as prolonged mitosis and delays of chromosome alignment and centrosome separation. Among several antioxidants, our screen reveals that ascorbic acid significantly alleviates these phototoxic effects in mitosis. Furthermore, we demonstrate that adding ascorbic acid to the media enables fluorescence imaging of mitotic events at very high temporal resolution without obvious photodamage. Thus, this study provides an optimal method to effectively reduce the phototoxic effects in fluorescence live cell imaging. Cyclodextrin (CD) is an important guest material owing to the water solubility and biocompatibility. In the paper, an organic small molecule was synthesized. According to supramolecular self-assembly, the organic molecule was bounded to the cavity of Poly β-cyclodextrin, which was characterized by IR, SEM and TEM et al. After self-assembly interaction, the morphology has changed obviously comparing with precursors. Simultaneously, the supramolecular self-assembly complex exhibited good water solubility. Moreover, By Gaussian calculation, the high binding activity between organic molecule and cyclodextrin was confirmed. By fluorescence investigation, the supramolecular system showed high fluorescence sensing activity for Zn2+ in pure water environment, which could track the dynamic change of Zn2+ in organisms. Small non-coding RNAs (sncRNA) are involved in many steps of the gene expression cascade and regulate processing and expression of mRNAs by the formation of ribonucleoprotein complexes (RNP) such as the RNA-induced silencing complex (RISC). By analyzing small RNA Seq data sets, we identified a sncRNA annotated as piR-hsa-1254, which is likely derived from the 3′-end of 7SL RNA2 (RN7SL2), herein referred to as snc7SL RNA. The 7SL RNA is an abundant long non-coding RNA polymerase III transcript and serves as structural component of the cytoplasmic signal recognition particle (SRP). To evaluate a potential functional role of snc7SL RNA, we aimed to define its cellular localization by live cell imaging. Therefore, a Molecular Beacon (MB)-based method was established to compare the subcellular localization of snc7SL RNA with its precursor 7SL RNA. We designed and characterized several MBs in vitro and tested those by live cell fluorescence microscopy. Using a multiplex approach, we show that 7SL RNA localizes mainly to the endoplasmic reticulum (ER), as expected for the SRP, whereas snc7SL RNA predominately localizes to the nucleus. Given the important role of carboxylesterase 2 (CES2) in the metabolism of various esters, it is of significance to develop a tool to determine endogenous CES2 activity in a rapid and highly selective manner. In this work, an "off-on" rhodamine-based fluorescent probe is reported as an effective tool for CES2 activity. Based on the substrate recognition preference of CES2 (large alcohol and small acyl groups), the probe with dimethyl carbamate as the recognition group performs well in terms of selectivity and sensitivity to CES2. The fluorescence switching control of the probe for CES2 was achieved by using the spirolactone structure of rhodamine. The probe shows the fast generation of a fluorescence signal at 634 nm upon hydrolysis of CES2 with excitation at 578 nm. The results showed a linear relationship between the fluorescence intensity at 634 nm and the CES2 activity from 0 to 4 μg/mL. The probe reacts rapidly with CES2, and the reaction is stable within 40 minutes. The probe is highly selective for CES2, with a detection limit as low as 0.303 μg/mL. In addition, the probe has currently been successfully utilized to evaluate CES2 activity in living cells. Herein, a Eugenol-derived fluorescence 'turn-on' probe FLHE was synthesized by condensing 2-((3-(trifluoromethyl)phenyl)amino)benzohydrazide with 5-allyl-2-hydroxy-3-methoxybenzaldehyde. FLHE demonstrated very low fluorescence in the studied organic solvents of varying polarities. However, upon titration with Zn2+ in HEPES buffer (pH = 7.4, 50% ACN, v/v), FLHE showed 40-fold higher fluorescence signals indicating the formation of the FLHE–Zn2+ complex. The fluorescence turn-on phenomenon upon FLHE–Zn2+ complex formation results from a chelation-enhanced fluorescence (CHEF) effect. The FLHE–Zn2+ complexation demonstrated a stokes shift of 156 nm (λ ex = 350 nm, λ em = 506 nm) and an about 33-fold increase in the quantum yield (FLHE , Φ = 0.007; FLHE–Zn2+ complex, Φ = 0.23). The binding constant (K a) determined by the Benesi-Hildebrand plot for interaction between FLHE and Zn2+ was 5.33 × 103 M−1. FLHE demonstrated a LOD of 31.8 nM for detecting Zn2+ in the environmental samples without interference from other cations and anions. FLHE -based paper strip (FLHE-PS) assay was developed to quantify the Zn2+ ions in water and the water content of organic solvent. Aza-BODIPY biotin-based nano-self-assembly has been designed with a biotin receptor for selective NIR turn-on fluorescence activity against cancer cells. The nanostructured aza-BODIPY assemblies exhibit distinctive fluorescence characteristics via an avidin-induced partial disintegration process. A stringent DNA probe to profile microRNA (miRNA) expression within a specific cell remains a key challenge in biology. To address this issue, an intracellular ATP-activated Y-DNA probe for accurate imaging of miRNA in living cells was designed. Y-DNA was based on the fabrication of tripartite function modules, which consisted of a folate (FA)-modified targeting module, an ATP aptamer-sealed driver, and a miRNA sensing module. The Y-DNA probe could be specifically activated by ATP after it efficiently internalized into FA-receptor-overexpressed cells based on caveolar-mediated endocytosis, leading to the activation of the miRNA sensing module. The activated Y-DNA probe allowed for the imaging of miRNA in living cells with high sensitivity. In this paper, a novel amino acid surface-functionalized semiconductor CdTe quantum dot fluorescent probe amidated by carboxyl and amide groups was synthesized to detect pyrophosphate ions (P2O74−, PPi). L-Arginine (L-Arg) was grafted onto cysteine modified CdTe quantum dots (Mea-CdTe QDs) to form a new L-Arginine-functionalized quantum dot fluorescent probe (L-Arg@Mea-CdTe). The prepared probe has good optical properties with multiple grafted functional groups on the surface. The guanidine group of the L-Arg@Mea-CdTe fluorescent probe is strongly basic and will be fully protonated under physiological conditions. The resulting hydrogen bonds bound to pyrophosphate lead to significant changes in the fluorescence of CdTe quantum dots. IR and XPS characterization were performed to confirm it. The addition of PPi induces a significant fluorescence quenching of L-Arg@Mea-CdTe in aqueous solution. The fluorescent QDs probe can also detect pyrophosphate with good sensitivity and anti-interference performance. The detection limit of the L-Arg@Mea-CdTe fluorescence probe for PPi is as low as 0.30 μM. The present work highlights the sustainable approach for the transformation of plastic waste into fluorescent carbon dots (CDs) through carbonization and then they were functionalized with L-cysteine and o-phenylenediamine. CDs which were characterized by different analytical techniques such as X-ray diffraction (XRD), thermogravimetric analysis (TGA), Scanning electron microscopy (SEM), and transmission electron microscopy (TEM) are employed to recognize Cu2+, Fe2+, and Hg2+ ions. The results show that the fluorescence emission was considerably quenched, and it is consistent with the interference and Jobs plots. The detection limit was found to be 0.35µM for Cu(II), 1.38 µM for Hg(II), and 0.51µM Fe(III). The interaction of CDs with metal ions enhances the fluorescence intensity detecting histamine successfully. It shows that plastic waste-based CDs can be applied clinically to detect toxic metals and biomolecules. Moreover, the system was employed to develop the cellular images using Saccharomyces cerevisiae cells with the support of a confocal microscope. Furthermore, theoretical studies were performed for the naphthalene layer (AR) as a model for C-dots, then optimized its structure and analyzed by using the molecular orbital. Carbon dots (CDs) have preeminent application prospects as a new star in the nanomaterials field. In this work, a green and facile method to synthesize the blue-emitting CDs was proposed with Melia azedarach leaves as the carbon precursors. Using nature materials without other expensive reagents and instruments, the processes were simple and environmental-friendly. The CDs had high fluorescence quantum yield (11.8%) and excellent luminescence properties. The size of them were among 1.5–2.5 nm and the emission spectrum exhibited a strong peak at 460 nm when excited at 380 nm. Additionally, the CDs were stable in most ions but sensitive to different pH values. As a result, a pH sensor was established for the detection of pH with a linear range of 3–10 pH. Moreover, it was demonstrated that the synthesized CDs had extremely low cytotoxicity. A novel fluorescent probe based on azamonardine (Aza) fluorophore was designed and synthesized for the highly selective detection of cysteine (Cys) in vivo and in vitro. After reacting with acryloyl chloride, the fluorescence of Aza is effectively quenched, resulting in the formation of the Aza-acryl probe. Upon the addition of Cys, the ester bond of Aza-acryl is cleaved, releasing a new compound (Compound 1) with strong fluorescence, thereby achieving fluorescence turn-on detection of Cys. The structure of Aza-acryl was characterized using X-ray crystallography and NMR spectroscopy. Additionally, density functional theory was employed to elucidate the quenching mechanism of the acyl group on the Aza. Aza-acryl exhibits high selectivity towards Cys and distinguishes it from other biothiols such as homocysteine (Hcy) and glutathione (GSH). The mechanism of Aza-acryl for detecting Cys was investigated through HPLC, NMR spectroscopy, high-resolution mass spectrometry, and reaction kinetics experiments. Aza-acryl demonstrates excellent imaging capabilities for Cys in cells and zebrafish, providing a reliable and selectable tool for the detection and imaging of Cys in biological systems. Cysteine (Cys) is a significant biological mercaptan that achieves key roles in several important physiological processes, such as reversible redox homeostasis in living organisms. Abnormal levels of Cys in the human body are directly related to many diseases. In this work, we constructed a sensitive sensor (Cys-NR) by connecting a Cys recognition group to a Nile red derivative. Due to photo-induced electron transfer (PET), the Cys-NR probe showed little fluorescence at 650 nm. With the addition of Cys to the assay solution, the chlorine unit of the probe was substituted by the thiol group of Cys. Further, the amino and sulfhydryl groups in cysteine underwent an intramolecular rearrangement, which led to the Cys-NR probe water solution turning from colorless to pink with an enhancement in fluorescence. The red fluorescence at 650 nm increased about 20 times. Based on the turn-on signal, a selective Cys detection method is developed. The probe signal is not affected by various potential interferences or other competing biothiols and the limit of detection (LOD) is determined to be 0.44 μM. In addition, the probe is further employed for imaging of Cys in live cells, revealing good biological imaging ability that could provide a new way of intracellular Cys detection. Fluorescence and photoacoustic imaging techniques offer valuable insights into cell- and tissue-level processes. However, these optical imaging modalities are limited by scattering and absorption in tissue, resulting in the low-depth penetration of imaging. Contrast-enhanced imaging in the near-infrared window improves imaging penetration by taking advantage of reduced autofluorescence and scattering effects. Current contrast agents for fluorescence and photoacoustic imaging face several limitations from photostability and targeting specificity, highlighting the need for a novel imaging probe development. This review covers a broad range of near-infrared fluorescent and photoacoustic contrast agents, including organic dyes, polymers, and metallic nanostructures, focusing on their optical properties and applications in cellular and animal imaging. Similarly, we explore encapsulation and functionalization technologies toward building targeted, nanoscale imaging probes. Bioimaging applications such as angiography, tumor imaging, and the tracking of specific cell types are discussed. This review sheds light on recent advancements in fluorescent and photoacoustic nanoprobes in the near-infrared window. A series of nitrogen-doped photoluminescent carbon dots (N-doped CDs) with a proper quantum yield was prepared through microwave-assisted pyrolysis of citric acid and urea as the carbon and nitrogen sources, respectively. The obtained N-doped CDs were fully characterized by high-resolution transmission electron microscopy, elemental analysis, Fourier transform infrared spectroscopy, and X-ray diffraction pattern. The average size of the synthesized N-doped CDs was determined using high-resolution transmission electron microscopy analysis was approximately 7 nm and the interlayer distance was 0.29 nm, which was consistent with the results obtained by X-ray diffraction. The optical characteristics of the samples were determined by UV-Vis and photoluminescent spectroscopy. The aqueous solution of N-doped CDs showed a multicolor emission wavelength, in the range of 350 to 650 nm, dependent on the nitrogen-doped percentage. With increasing the nitrogen percentage, the emission spectra, depending on the excitation wavelength, have one or two peaks in the green and blue regions, which can be considered in applications such as special bioimaging and related fields. Imidazo[1,2-a]pyridine derivatives have excellent potential for chelation with transition metal ions. Two new imidazo[1,2-a]pyridine-8-carboxylates were synthesized and characterized by 1H NMR, 13C NMR, HRMS, and single crystal-XRD techniques. Methyl carboxylate (probe 1) turns on fluorescence upon coordination with Zn2+, while sodium carboxylate (probe 2) turns off its fluorescence upon coordination with Co2+ or Cu2+ ions present in aqueous acetonitrile medium. 13C NMR study revealed that the change in metal ion specific binding was due to the involvement of carboxylate anion in complex formation with Co2+ or Cu2+ ions. The carboxylate anion at 8-position also enhanced the sensitivity of detection of probe 2 by an order of magnitude (detection limits: 3.804 × 10–7 M, probe 1/Zn2+; 0.420 × 10–7 M, probe 2/Co2+ and 0.304 × 10–7 M, probe 2/Cu2+). The detection limits of probes 1 and 2 comply well with the World Health Organization (WHO) and US Environmental Protection Agency (US-EPA) guidelines for detection of heavy metal ions present in drinking water and ground water. Both the probes form a 1:1 complex with Zn2+, Co2+ or Cu2+, and the stoichiometry was verified by Job plot and ESI-mass analysis. The sensing mechanism is explained using 13C NMR experiments, ESI-mass analytical data and theoretical DFT calculations. The suitability of probes 1 and 2 for on-site detection and quantitative determination of Zn2+, Co2+ and Cu2+ ions present in biological, environmental and industrial samples is demonstrated. Organic nanomaterials, as nanocarrier platforms, have tremendous potential for biomedical applications. The authors successfully prepared novel two‐dimensional covalent organic nanosheets (CONs) that can be used as efficient in vivo bioimaging probes by condensing 1,3,5‐triformylglucinol (Tp) and 2,7‐diaminopyrene (Py) to produce TpPy covalent organic frameworks (COFs). TpPy COFs are then subjected to a liquid exfoliation process to obtain TpPy CONs (< 200 nm in size and < 1.7 nm in thickness). TpPy CONs disperse well in water to provide a stable, homogeneous colloidal suspension, which shows favorable photoluminescence properties. Cell viability tests using MDA‐MB‐231 and RAW 264.7 cells reveal that TpPy CONs are low in cytotoxicity. Confocal microscopy reveals clear fluorescent cell images after incubation with TpPy CONs for 24 h, without reduction in cell activity or cytosolic aggregation. To investigate the biological behavior of the TpPy CONs, the authors perform an in vivo fluorescence imaging study using MDA‐MB‐231 tumor‐bearing mice. After intravenous injection of TpPy CONs disperse in phosphate‐buffered saline (PBS), persistent and strong fluorescence signals are observed in the tumor region, with low background signals from normal tissues at 1, 3, 12, and 24 h after injection. Developing effective intelligent nanocarriers is highly desirable for fluorescence imaging and therapeutic applications but remains challenging. Using a vinyl-grafted BMMs (bimodal mesoporous SiO2 materials) as a core and PAN ((2-aminoethyl)-6-(dimethylamino)-1H-benzo[de]isoquinoline-1,3(2H)-dione))-dispersed dual pH/thermal-sensitive poly(N-isopropylacrylamide-co-acrylic acid) as a shell, PAN@BMMs with strong fluorescence and good dispersibility were prepared. Their mesoporous features and physicochemical properties were extensively characterized via XRD patterns, N2 adsorption–desorption analysis, SEM/TEM images, TGA profiles, and FT-IR spectra. In particular, their mass fractal dimension (dm) features based on SAXS patterns combined with fluorescence spectra were successfully obtained to evaluate the uniformity of the fluorescence dispersions, showing that the dm values increased from 2.49 to 2.70 with an increase of the AN-additive amount from 0.05 to 1%, along with the red shifting of their fluorescent emission wavelength from 471 to 488 nm. The composite (PAN@BMMs-I-0.1) presented a densification trend and a slight decrease in peak (490 nm) intensity during the shrinking process. Its fluorescent decay profiles confirmed two fluorescence lifetimes of 3.59 and 10.62 ns. The low cytotoxicity obtained via in vitro cell survival assay and the efficient green imaging performed via HeLa cell internalization suggested that the smart PAN@BMM composites are potential carriers for in vivo imaging and therapy. Enterovirus consists of a variety of viruses that could cause a wide range of illness in human. The pathogenesis of these viruses remains incompletely understood and no specific treatment is available. Better methods to study enterovirus infection in live cells will help us better understand the pathogenesis of these viruses and might contribute to antiviral development. Here in this study, we developed fluorescent cell-based reporter systems that allow sensitive distinction of individual cells infected with enterovirus 71 (EV71). More importantly, these systems could be easily used for live-cell imaging by monitoring viral-induced fluorescence translocation after EV71 infection. We further demonstrated that these reporter systems could be used to study other enterovirus-mediated MAVS cleavage and they are sensitive for antiviral activity testing. Therefore, integration of these reporters with modern image-based analysis has the potential to generate new insights into enterovirus infection and facilitate antiviral development. Metal cations such as Zn2+, Al3+, Hg2+, Cd2+, Sn2+, Fe2+, Fe3+ and Cu2+ play important roles in biology, medicine, and the environment. However, when these are not maintained in proper concentration, they can be lethal to life. Therefore, selective sensing of metal cations is of great importance in understanding various metabolic processes, disease diagnosis, checking the purity of environmental samples, and detecting toxic analytes. Schiff base probes have been largely used in designing fluorescent sensors for sensing metal ions because of their easy processing, availability, fast response time, and low detection limit. Herein, an in-depth report on metal ions recognition by some Schiff base fluorescent sensors, their sensing mechanism, their practical applicability in cell imaging, building logic gates, and analysis of real-life samples has been presented. The metal ions having biological, industrial, and environmental significance are targeted. A major challenge in ART is to select high-quality oocytes and embryos. The metabolism of oocytes and embryos has long been linked to their viability, suggesting the potential utility of metabolic measurements to aid in selection. Here, we review recent work on noninvasive metabolic imaging of cumulus cells, oocytes, and embryos. We focus our discussion on fluorescence lifetime imaging microscopy (FLIM) of the autofluorescent coenzymes NAD(P)H and flavine adenine dinucleotide (FAD+), which play central roles in many metabolic pathways. FLIM measurements provide quantitative information on NAD(P)H and FAD+ concentrations and engagement with enzymes, leading to a robust means of characterizing the metabolic state of cells.
