During infancy, orienting and gaze aversion serve as major self-regulatory mechanisms and play an important role in the development of deliberate self-regulation and control. The present study examined the interaction of intrinsic factors (MAOA-uVNTR and 5-HTTLPR gene polymorphisms) and extrinsic factors (maternal sensitivity) on early infant self-regulatory behavior. We assessed 5-HTTLPR (ss + sl versus ll) and MAOA-uVNTR (3 and 4 among boys, and 3/3, 3/4, and 4/4 among girls) polymorphisms, determined maternal sensitivity during mother–child free play, and coded infant self-regulatory behavior (i.e., orienting shifts in a temperament test) in 281 six-month-old infants. We found that infants who experienced a lower level of maternal sensitivity and had the short allele of 5-HTTLPR variants and the 3/3 MAOA-uVNTR polymorphism displayed lower self-regulation capacity than did those infants with a higher level of maternal sensitivity. This finding suggested a modulatory role of maternal sensitivity. Moreover, these findings are consistent with the genetic vulnerability hypothesis, which states that beneficial environmental factors serve as a buffer against harmful genetic predispositions during child development. Self-regulatory behavior in early childhood is an important characteristic that has considerable implications for the development of adaptive and maladaptive functioning. The present study investigated the relations between a functional polymorphism in the upstream region of monoamine oxidase A gene (MAOA) and self-regulatory behavior in a sample of Chinese infants at 6 months of age. Self-regulation was assessed by observing infants' behavior of orienting visual attention away from a threatening event in the laboratory situation. The results indicated that regulatory behavior was associated with the functional MAOA gene polymorphism in girls, but not boys. Girls with 4/4 genotypes displayed significantly higher regulation than girls with 3/3 and 3/4 genotypes. This study aims to assess the role of methotrexate-related gene polymorphisms in children with acute lymphoblastic leukemia (ALL) during high-dose methotrexate (HD-MTX) therapy and to explore their effects on serum metabolites before and after HD-MTX treatment. The MTHFR 677C>T, MTHFR 1298A>C, ABCB1 3435C>T, and GSTP1 313A>G genotypes of 189 children with ALL who received chemotherapy with the CCCG-ALL-2020 regimen from January 2020 to April 2023 were analyzed, and toxic effects were reported according to the Common Terminology Criteria for Adverse Events (CTCAE, version 5.0). Fasting peripheral blood serum samples were collected from 27 children before and after HD-MTX treatment, and plasma metabolites were analyzed by liquid chromatography-tandem mass spectrometry (LC–MS). The results of univariate and multivariate analyses showed that MTHFR 677C>T and ABCB1 3435 C>T gene polymorphisms were associated with the delayed MTX clearance (P < 0.05) and lower platelet count after treatment in children with MTHFR 677 mutation compared with wild-type ones (P < 0.05), and pure mutations in ABCB1 3435 were associated with higher serum creatinine levels (P < 0.05). No significant association was identified between MTHFR 677C>T, MTHFR 1298A>C, ABCB1 3435 C>T, and GSTP1 313A>G genes and hepatotoxicity or nephrotoxicity (P > 0.05). However, the serum metabolomic analysis indicated that the presence of the MTHFR 677C > T gene polymorphism could potentially contribute to delayed MTX clearance by influencing L-phenylalanine metabolism, leading to the occurrence of related toxic side effects. Conclusion: MTHFR 677C>T and ABCB1 3435 C>T predicted the risk of delayed MTX clearance during HD-MTX treatment in children with ALL. Serum L-phenylalanine levels were significantly elevated after HD-MTX treatment in children with the MTHFR 677C>T mutation gene. The present study aimed to test whether monoamine oxidase A ( MAOA) gene polymorphism was a marker of the genetic vulnerability model or the differential susceptibility model by investigating the interaction of MAOA gene polymorphism with maternal parenting on externalizing and internalizing problems and social competence among Chinese children. Participants were 290 children and their parents in China. MAOA genotypes were ascertained by polymerase chain reaction amplification. Mothers reported their parenting stress and engagement in parent-child activities when the child was 6 months old. Children's behavioral problems and social competence were rated by mothers and fathers at 48 months old. The results showed that MAOA genotypes interacted with maternal parenting stress in predicting boys' internalizing problems. Also, MAOA genotypes interacted with maternal engagement in parent-child activities in predicting boys' social competence. The analyses of regions of significance and the proportion of interaction index indicated that boys with the low-activity genotype were at risk for (a) high internalizing problems under high maternal parenting stress and (b) low social competence if mothers displayed a low level of maternal engagement. No significant interaction effects were found among girls. The results suggested that the MAOA gene might be a marker of the genetic vulnerability model among Chinese boys. Abstract: Three-dimensional polyacrylamide gel microarray (3-D gel microarray) has a strong adsorption for hybridzation probes with fluorescent label, resulting in high fluorescent background noise. The background noise has hampered the broad application of the microarray in scientific research. A low-frequency ultrasound of 40kHz has been successfully employed to efficiently remove the nonspecifically bound fluorescent probes from the 3-D gel microarray. The method can significantly enhance the signal to noise ratio of the 3-D gel microarray and make it work better in single-nucleotide polymorphism genotyping. [Copyright &y& Elsevier]
