Background: Alternative insecticidal compounds with mortality effect against mosquito life cycle stages are currently needed. The compounds should be biodegradable and nontoxic to non-targeted insects. Plant based larvicides provide effective control of vector populations. This study explored Cryptomeria japonica leaf essential oil larvicidal potency against Anopheles gambiae sensu stricto., Methods: Essential oils (12.5 to 200 μg/mL) extracted from C. japonica leaves were evaluated against An. gambiae s.s. larvae in both the laboratory and semi field in 6 replicates for each dose. Larval mortality readings were taken at 12, 24, 48, and 72 h post treatment., Results: C. japonica leaf essential oil yield was 17.06 ± 0.56 mL/kg and 1.60 ± 0.33% (w/w). GC-FID and GC-MS analyses revealed 22 constituents. Essential oil was more effective against An. gambiae s.s. larvae in the laboratory than in semi field trials. Mortality increased with increasing dosages (12.5 to 200 μg/mL) in the laboratory (31.75 to 100%) and semi field trials (17.75 to 99.5%), respectively. The LC50 value ranged from 5.55 to 63.92 μg/mL in the laboratory, and 8.22 to 134.84 μg/mL in semi field conditions, LC90 value ranged from 41.34 to 205.93 μg/mL in the laboratory and 50.92 to 213.11 μg/mL in semi field conditions., Conclusion: This study has demonstrated the potential of C. japonica leaf essential oil to cause mortality effects to An. gambiae s. s. larval populations, however, further studies need to be conducted under field conditions and also with individual active compounds of C. japonica essential oil.

Background: Formosan subterranean termite, Coptotermes formosanus Shiraki (Isoptera: Rhinotermitidae), is a major urban pests in many countries throughout the world. In an attempt to find natural products that could be employed in the effective and environmentally friendly control of the termite, this study investigates, for the first time, the insecticidal activities of the wood and leaf essential oils and ethanolic extracts from Cunninghamia konishii Hayata against C. formosanus according to direct contact application., Results: Wood and leaf essential oils and wood ethanolic extract showed the excellent antitermitic activities. Among the four fractions of wood ethanolic extract, the hexane-soluble fraction showed the strongest antitermitic activities. In addition, β-elemol and α-cadinol showed excellent inhibitory action against C. formosanus., Conclusion: The results clearly reveal that the wood and leaf essential oils and ethanolic extract from C. konishii, and their effective constituents, served as potential, eco-friendly termite-control agents., (© 2013 Society of Chemical Industry.)

Background: The increasing status of insecticide resistant mosquitoes in sub-Saharan Africa is a threatening alert to the existing control efforts. All sibling species of An. gambiae complex have evolved insecticide resistance in wild populations for different approved classes of the insecticides currently in use in the field. An alternative compound for vector control is absolutely urgently needed. In this study, the larvicidal activity and chemical composition of the Cinnamomum osmophloeum leaf essential oils were investigated., Methods: C. osmophloeum leaf essential oils were extracted by hydrodistillation in a Clevenger-type apparatus for 6 hours, and their chemical compositions identified using GC-MS. These oils were evaluated against An. gambiae s.s. in both laboratory and semi-field situations. The WHO test procedures for monitoring larvicidal efficacy in malaria vectors were used., Results: The composition of C. osmophloeum leaf essential oil has been found to have 11 active compounds. The most abundant compound was trans-cinnamaldehyde (70.20%) and the least abundant was caryophyllene oxide (0.08%). The larvicidal activity was found to be dosage and time dependant both in laboratory and semi-field environments with mortality ranging from 0% to 100%. The LC50 value was found to vary from 22.18 to 58.15 μg/ml in the laboratory while in semi-field environments it was 11.91 to 63.63 μg/ml. The LC90 value was found to range between 57.71 to 91.54 μg/ml in the laboratory while in semi-field environments was 52.07 to 173.77 μg/ml. Mortality ranged from 13% to 100% in the laboratory while in semi-field environments it ranged between 43% to 100% within mortality recording time intervals of 12, 24, 48, and 72 hours., Conclusions: The larvicidal activity shown by C. osmophloeum leaf essential oil is a promising alternative to existing larvicides or to be incorporated in integrated larval source management compounds for An. gambiae s.s control. The efficacy observed in this study is attributed to both major and minor compounds of the essential oils.

Cryptomeria japonica D. Don, called 'sugi' in Japanese, is an abundant and renewable potential resource of valuable natural products that may serve as natural biocides in Taiwan. Hydrodistillation (HD) and steam distillation (SD) were used for extracting the essential oils from C. japonica leaves in this study. The chemical constituents of the two leaf essential oils were identified by GC/MS analysis, and their antitermitic activities were evaluated. The results from the antitermitic tests against Coptotermes formosanus Shiraki showed that the leaf essential oils extracted by HD and SD had excellent antitermitic activities, with LD(50) values of 1.57 and 1.72 mg/g after 7 d of testing, respectively. Comparison of the chemical composition of the two leaf essential oils revealed that oxygenated diterpenes (kaur-16-ene) and oxygenated sesquiterpenes (β-elemol) were the dominant constituents. In addition, 13 constituents isolated from the C. japonica leaf essential oils were tested individually against C. formosanus. Among these constituents, β-elemol and α-terpineol achieved 100% termite mortality at the dosage of 1 mg/g after 7 d of testing. These results demonstrated that both C. japonica leaf essential oils as well as β-elemol and α-terpineol possessed commendable antitermitic activity., (Copyright © 2012 Verlag Helvetica Chimica Acta AG, Zürich.)

An insecticidal toxin gene, tccC, was cloned from the recently discovered novel species Pseudomonas taiwanensis using degenerate PCR and genomic walking. The DNA sequence of the tccC gene (2,940 bp) has an open reading frame encoding 980 amino acids with a calculated molecular weight of 107.93 kDa. The amino acid sequence alignment showed the highest sequence identity (41.2%) with the insecticidal toxin from Pseudomonas entomophila. To examine the insecticidal functionality of the tccC gene product, TccC was heterologously expressed in Escherichia coli as a recombinant His6 fusion protein and purified by immobilized metal ion-affinity chromatography. The recombinant TccC was fed to Drosophila larvae at a concentration of 350 ppm, which induced about 60% mortality within 72 h. The recombinant TccC was stable at pH 7.0 and at 37 °C. When the pH was less than 5.0 or greater than 9.0, or temperature was greater than 55 °C, less than 20% Drosophila larvae mortality was observed. These results prove that Pseudomonas taiwanensis could be used as a source for developing novel biopesticides.

The purpose of this study is to determine the larvicidal activities of ethanolic extracts from leaves, wood, and bark of black heartwood-type Cryptomeria japonica against fourth-instar larvae of mosquitoes Aedes aegypti and Aedes albopictus. Among three ethanolic extracts from C. japonica, wood extract exhibited the best larvicidal activity against A. aegypti and A. albopictus with LC(50) values of 63.2 and 93.8 microg/ml, respectively. Four major compounds, including ferruginol, epi-cubebol, cubebol, and isopimarol, were isolated from wood extract, and it was demonstrated that cubebol exhibited the best activity against A. aegypti and A. albopictus with LC(50) values of 60.1 and 50.0 microg/ml, respectively. Our findings showed that the wood extract and cubebol from C. japonica have good potential as a source for natural larvicides.

Background: The current study investigates, for the first time, the mosquito larvicidal activities of leaf and twig essential oils from Clausena excavata Burm. f. and their individual constituents against Aedes aegypti L. and Aedes albopictus Skuse larvae. The yields of essential oils obtained from hydrodistillation were compared, and their constituents were determined by GC-MS analyses., Results: The LC(50) values of leaf and twig essential oils against fourth-instar larvae of Ae. aegypti and Ae. albopictus were 37.1-40.1 microg mL(-1) and 41.1-41.2 microg mL(-1) respectively. This study demonstrated that C. excavata leaf and twig essential oils possess mosquito larvicidal activity, inhibiting the growth of mosquito larvae for both species at a low concentration. In addition, results of larvicidal assays showed that the effective constituents in leaf and twig essential oils were limonene, gamma-terpinene, terpinolene, beta-myrcene, 3-carene and p-cymene. The LC(50) values of these constituents against both mosquito larvae were below 50 microg mL(-1). Among these effective constituents, limonene had the best mosquito larvicidal activity, with LC(50) of 19.4 microg mL(-1) and 15.0 microg mL(-1) against Ae. aegypti and Ae. albopictus larvae respectively., Conclusion: The findings suggested that the essential oils from Clausena excavata leaf and twig and their effective constituents may be explored as a potential natural larvicide., (2008 Society of Chemical Industry)

The larvicidal activities of leaf essential oils and their constituents from six chemotypes of indigenous cinnamon (Cinnamomum osmophloeum Kaneh.) trees were evaluated against three mosquito species. Results of larvicidal tests demonstrated that the leaf essential oils of cinnamaldehyde type and cinnamaldehyde/cinnamyl acetate type had an excellent inhibitory effect against Aedes albopictus larvae, and their LC(50) values in 24h were 40.8 microg/ml (LC(90)=81.7 microg/ml) and 46.5 microg/ml (LC(90)=83.3 microg/ml), respectively. Results of the 24-h mosquito larvicidal assays also showed that the effective constituents in leaf essential oils were trans-cinnamaldehyde and benzaldehyde and that the LC(50) values of these constituents against A. albopictus larvae were below 50 mug/ml. In addition, cinnamaldehyde type leaf essential oil and trans-cinnamaldehyde have also exhibited great larvicidal performance against Culex quinquefasciatus and Armigeres subalbatus larvae. Comparisons of mosquito larvicidal activity of trans-cinnamaldehyde congeners revealed that alpha-methyl cinnamaldehyde, benzaldehyde, and trans-cinnamaldehyde exhibited strong mosquito larvicidal activity.

The larvicidal effects of the essential oils extracted from the leaves of Cryptomeria japonica at different ages (58, 42, and 26 years old) against 2 mosquito species, Aedes aegypti and Aedes albopictus, were studied. The analysis of major constituents of these essential oils was also investigated. Results obtained from the larvicidal tests, using essential oil from the leaves of 58-year-old C. japonica was found to be most effective against both A. aegypti and A. albopictus larvae, indicating tree age has significant influence on mosquito larvicidal activity. In addition, the eleven pure constituents from C. japonica leaf essential oil were also tested individually against the two mosquito larvae. Among them, alpha-terpinene, gamma-terpinene, p-cymene, 3-carene, terpinolene, and beta-myrcene shows strong larvicidal effect against the two mosquito larvae. Among these pure constituents, 3-carene exhibits the best larvicidal effect against A. aegypti and terpinolene shows an excellent inhibitory action against A. albopictus larvae. The results of this study show that the leaf essential oil and its effective constituents might be considered as a potent source for the production of fine natural larvicides.

In the current study, the mosquito larvicidal activity of leaf essential oils and their constituents from two eucalyptus species (Eucalyptus camaldulensis and Eucalyptus urophylla) against two mosquito species, Aedes aegypti and Aedes albopictus, was investigated. In addition, the chemical compositions of the leaf essential oils were analyzed using gas chromatography-mass spectrometry. Results from the larvicidal tests revealed that essential oil from the leaves of E. camaldulensis had an excellent inhibitory effect against both A. aegypti and A. albopictus larvae. The 12 pure constituents extracted from the two eucalyptus leaf essential oils were also tested individually against two mosquito larvae. Among the six effective constituents, alpha-terpinene exhibits the best larvicidal effect against both A. aegypti and A. albopictus larvae. Results of this study show that the leaf essential oil of E. camaldulensis and its effective constituents might be considered as a potent source for the production of fine natural larvicides.

Eleven compounds from indigenous cinnamon (Cinnamomum osmophloeum) leaf essential oil were identified by GC-MS and the dominant constituent was trans-cinnamaldehyde (79.85%). The toxicity of leaf essential oil and trans-cinnamaldehyde were then determined to study their effectiveness in controlling the red imported fire ant, Solenopsis invicta Buren. The results of the toxicity tests indicated that both the indigenous cinnamon leaf essential oil and trans-cinnamaldehyde had an excellent inhibitory effect in controlling the red imported fire ant. The LT(50) values for both 2% leaf essential oil and 2% trans-cinnamaldehyde after open exposure were 105.0min and 32.2min; after close exposure were 18.5min and 21.2min, respectively.

Chemical compositions of leaf essential oils from eight provenances of indigenous cinnamon (Cinnamomum osmophloeum Kaneh.) were compared. According to GC-MS and cluster analyses, the leaf essential oils of the eight provenances and their relative contents were classified into five chemotypes-cinnamaldehyde type, linalool type, camphor type, cinnamaldehyde/cinnamyl acetate type, and mixed type. The larvicidal activities of leaf essential oils and their constituents from the five chemotypes of indigenous cinnamon trees were evaluated by mosquito larvicidal assay. Results of larvicidal tests demonstrated that the leaf essential oils of cinnamaldehyde type and cinnamaldehyde/cinnamyl acetate type had an excellent inhibitory effect against the fourth-instar larvae of Aedes aegypti. The LC(50) values for cinnamaldehyde type and cinnamaldehyde/cinnamyl acetate type against A. aegypti larvae in 24 h were 36 ppm (LC(90) = 79 ppm) and 44 ppm (LC(90) = 85 ppm), respectively. Results of the 24-h mosquito larvicidal assays also showed that the effective constituents in leaf essential oils were cinnamaldehyde, eugenol, anethole, and cinnamyl acetate and that the LC(50) values of these constituents against A. aegypti larvae were <50 ppm. Cinnamaldehyde had the best mosquito larvicidal activity, with an LC(50) of 29 ppm (LC(90) = 48 ppm) against A. aegypti. Comparisons of mosquito larvicidal activity of cinnamaldehyde congeners revealed that cinnamaldehyde exhibited the strongest mosquito larvicidal activity.

The antitermitic activities of the essential oils from the leaves of two Cinnamomum osmophloeumclones (A and B) and their chemical ingredients against Coptotermes formosanus Shiraki were investigated according to direct contact application. Results from this experiment have demonstrated that the indigenous cinnamon B leaf essential oil has a more effective antitermitic activity than indigenous cinnamon A leaf essential oil. Furthermore, when cinnamaldehyde, eugenol, and alpha-terpineol are extracted from indigenous cinnamon leaf essential oil and used at the strength of 1 mg/g, their antitermitic effectiveness is much higher than that using indigenous cinnamon leaf essential oil. Among the congeners of cinnamaldehyde examined, cinnamaldehyde has exhibited the strongest termiticidal property.
