Nitrosyl iron complexes are remarkably multifactorial pharmacological agents. These compounds have been proven to be particularly effective in treating cardiovascular and oncological diseases. We evaluated and compared the antioxidant activity of tetranitrosyl iron complexes (TNICs) with thiosulfate ligands and dinitrosyl iron complexes (DNICs) with glutathione (DNIC-GS) or phosphate (DNIC-PO 4 - -butyl hydroperoxide, oxidative modification of Hb, and antioxidant properties of nitrosyl iron complexes. Measuring luminol chemiluminescence revealed that the antioxidant effect of TNICs was higher compared to DNIC-PO tert -butyl hydroperoxide, oxidative modification of Hb, and antioxidant properties of nitrosyl iron complexes. Measuring luminol chemiluminescence revealed that the antioxidant effect of TNICs was higher compared to DNIC-PO 4 - . DNIC-GS either did not exhibit antioxidant activity or exerted prooxidant effects at certain concentrations, which might have resulted from thiyl radical formation. TNICs and DNIC-PO 4 - efficiently protected the Hb heme group from decomposition by organic hydroperoxides. DNIC-GS did not exert any protective effects on the heme group; however, it abolished oxoferrylHb generation. TNICs inhibited the formation of Hb multimeric forms more efficiently than DNICs. Thus, TNICs had more pronounced antioxidant activity than DNICs in Hb-containing systems.

Nitric oxide (NO) mediates diverse physiological processes in living organisms. Small molecular NO donors usually lack stability and have a short half-life in human tissues, limiting the therapeutic application. The anionic tetranitrosyl iron complex with thiosulfate ligands (TNIC) is one of the most promising NO donors. This study shows that bovine serum albumin (BSA) can effectively stabilize the TNIC complex under aerobic (physiological) conditions, which contributes to its prolonged action as NO donor. Our results demonstrated that TNIC-BSA inhibits formation of TBARS - standard biomarker for the lipid peroxidation induced oxidative stress. Also, it was found that TNIC-BSA inhibits the catalytic activity of mitochondrial membrane-bound enzymes: cytochrome c oxidase and monoamine oxidase A. Together, these results demonstrate that, stabilization of TNIC with BSA opens up the possibility of its practical application in chemotherapy of socially significant diseases., (Copyright © 2021 Elsevier Inc. Microbially influenced corrosion (MIC) results in significant damage to metallic materials in many industries. Anaerobic sulfate-reducing bacteria (SRB) have been well studied for their involvement in these processes. Highly corrosive environments are also found in pulp and paper processing, where chloride and thiosulfate lead to the corrosion of stainless steels. Acidithiobacillus ferrooxidans is a critically important chemolithotrophic acidophile exploited in metal biomining operations, and there is interest in using A. ferrooxidans cells for emerging processes such as electronic waste recycling. We explored conditions under which A. ferrooxidans could enable the corrosion of stainless steel. Acidic medium with iron, chloride, low sulfate, and pyrite supplementation created an environment where unstable thiosulfate was continuously generated. When combined with the chloride, acid, and iron, the thiosulfate enabled substantial corrosion of stainless steel (SS304) coupons (mass loss, 5.4 ± 1.1 mg/cm 2 over 13 days), which is an order of magnitude higher than what has been reported for SRB. There results were verified in an abiotic flow reactor, and the importance of mixing was also demonstrated. Overall, these results indicate that A. ferrooxidans and related pyrite-oxidizing bacteria could produce aggressive MIC conditions in certain environmental milieus. IMPORTANCE MIC of industrial equipment, gas pipelines, and military material leads to billions of dollars in damage annually. Thus, there is a clear need to better understand MIC processes and chemistries as efforts are made to ameliorate these effects. Additionally, A. ferrooxidans is a valuable acidophile with high metal tolerance which can continuously generate ferric iron, making it critical to copper and other biomining operations as well as a potential biocatalyst for electronic waste recycling. The antioxidant and antiradical properties of the tetra nitrosyl iron complex with thiosulfate ligands (TNIC) were studied in vitro in mouse brain homogenates. It was found for the first time that TNIC is an effective antioxidant. The effect of TNIC on the catalytic activity of mitochondrial enzymes cytochrome c oxidase and monoamine oxidase A was studied. It was shown for the first time that TNIC is an inhibitor of the catalytic activity of cytochrome c oxidase and monoamine oxidase A in animal brain mitochondria in vitro.

Iron oxide and oxy-hydroxide are commonly used for remediation and rehabilitation of arsenic (As)-contaminated soil and water. However, the stability of As sequestered by iron oxide and oxy-hydroxide under anaerobic conditions is still uncertain. Geochemical properties influence the behavior of As; in addition, microbial activities affect the mobility of sequestered As in soil and water. Microbial-mediated iron reduction can increase the mobility of As by reductive dissolution of Fe oxide; however, microbial-mediated sulfate reduction can decrease the mobility of As by sulfide mineral precipitation. This study investigated the geomicrobial impact on the behavior of As and stability of sequestered As in iron-rich sediment under anaerobic conditions. Increase in Fe(II) concentrations in water was evidence of microbial-mediated iron reduction. Arsenic concentrations increased with Fe(II) concentration; however, the thiosulfate reduction process also induced immobilization of As through the precipitation of AsFeS. Therefore, microbial-mediated iron reduction and thiosulfate reduction have opposite influences on the mobility of As under anaerobic condition.

A column study for hexavalent chromium (Cr(VI)) removal from contaminated soil was performed using calcium polysulfide (CPS) and nanoscale zero-valent iron stabilized with green tea extract (GT-nZVI). Injection of CPS at 12 times the stoichiometric requirement (12×) resulted in quantitative Cr(VI) removal for up to 195 days of equivalent groundwater flow. Solid-bound Cr(VI) was reduced up to >99% (<2mg/kg). Treatment with CPS resulted in a short-term release of high sulfur concentrations. Injections of 12× and 24× stoichiometric GT-nZVI resulted in decrease in leachate pH from 6 to 2.5, which rebounded to 4.5 after the equivalent of 45 days and remained stable for the next equivalent 3 years. Metals concentrations in the effluent (Pb, Cr and Fe) increased following injection and quickly decreased, such that the mass flux was low with respect to the total amounts in the solid. Aqueous Cr(VI) was non-detect for the majority of the monitoring time, but concentrations eventually increased with respect to the control sample. Solid-bound Cr(VI) concentrations decreased by 30% and 66% in the 12× and 24× treatments, respectively. The low efficiency was attributed to increased sorption to iron surfaces at pH 2.5 and slow dissolution of PbCrO4, both of which were identified by micro-X-ray fluorescence and absorption analyses., (Copyright © 2011 Elsevier B.V. The determination of the redox properties of the cofactor in heme proteins provides fundamental insight into the chemical characteristics of this wide-spread class of metalloproteins. For the preparation of the ferroheme state, probably the most widely applied reductant is sodium dithionite, which at neutral pH has a reduction potential well below the reduction potential of most heme centers. In addition to the heme iron, some heme proteins, including the nitrophorins (NPs), contain cysteinecysteine disulfide bonds. In the present study, the effect of dithionite on the disulfides of NP4 and NP7 is addressed. To gain deeper understanding of the disulfide/dithionite reaction, oxidized glutathione (GSSG), as a model system, was incubated with dithionite and the products were characterized by (13)C NMR spectroscopy and reverse phase chromatography in combination with mass spectrometry. This revealed the formation of one equivalent each of thiol (GSH) and glutathione-S-thiosulfate (GSSO(3)(-)). With this background information, the effect of dithionite on the cystines of NP4 and NP7 was studied after trapping of the thiols with para-cloromercurybenzyl sulfonate (p-CMBS) and subsequent matrix assisted laser desorption/ionization time-of-flight mass spectrometry (MALDI-TOF MS) where the heterolytic cleavage of the SS bond appears with only 2molar equivalents of the reductant. Furthermore, prolonged electrochemical reduction of NP4 and NP7 in the presence of electrochemical mediators also leads to disulfide breakage. However, due to sterical shielding of the disulfide bridges in NP4 and NP7, the cystine reduction can be largely prevented by the use of stoichiometric amounts of reductant or limited electrochemical reduction. Chemical phosphorus (P) removal during aerobic wastewater treatment induced by iron salt addition in sewer systems for sulfide control is investigated. Aerobic batch tests with activated sludge fed with wastewater containing iron sulfide precipitates showed that iron sulfide was rapidly reoxidised in aerobic conditions, resulting in phosphate precipitation. The amount of P removed was proportional to the amount of iron salts added, and for the sludge used, ratios of 0.44 and 0.37 mgP/mgFe were obtained for ferric and ferrous dosages, respectively. The hydraulic retention time (HRT) of iron sulfide in sewers was found to have a crucial impact on the settling of iron sulfide precipitates during primary settling, with a shorter HRT resulting in a higher concentration of iron sulfide in the primary effluent and thus enabling higher P removal. A mathematical model was developed to describe iron sulfide oxidation in aerated activated sludge and the subsequent iron phosphate precipitation. The model was used to optimise FeCl(3) dosing in a real wastewater collection and treatment system. Simulation studies revealed that, by moving FeCl(3) dosing from the WWTP, which is the current practice, to a sewer location upstream of the plant, both sulfide control and phosphate removal could be achieved with the current ferric salt consumption. Pyrite oxidation processes by aqueous Cr(VI) were investigated at 25 degrees C under an argon atmosphere. Synthetic pyrite suspensions (6 g L(-1)) were reacted for 20 h with a range of Cr(VI) solutions from 0 to 700 microM and at pH 2-12. The main objective of this work was to investigate the reaction mechanisms by emphasizing the role of sulfur species. Aqueous chemical processes were well illustrated in acidic media where significant amounts of sulfate and iron species were determined. Sulfate anions are the final stable sulfur species involved in the reaction pathway. Experiments showing complete Cr(VI) removal from solution displayed ratios [S(VI)]/[Fe](tot)<2, probably due to a deficit in aqueous sulfur species. Experiments showing incomplete Cr(VI) removal displayed ratios [Cr(VI)](removed)/[S(VI)] close to 1.5. This ratio was found to be consistent with the formation of thiosulfate (S(2)O(2-)(3)). Thiosulfate ions disproportionated into elemental sulfur S(0) and tetrathionate ions (S(4)O(2-)(6)) that were finally oxidized to sulfate anions under acidic conditions. The distribution of the oxidation state of sulfur atoms at the pyrite surface determined by XPS was additional evidence for the multistep sulfur oxidation process. The presence of elemental sulfur in the S(2p) spectra correlated well with the disproportion of thiosulfate under acidic conditions.

Extracellular polymeric substances seem to play a pivotal role in biocorrosion of metals and bioleaching, biocorrosion of metal sulfides for the winning of precious metals as well as acid rock drainage. For better control of both processes, the structure and function of extracellular polymeric substances of corrosion-causing or leaching bacteria are of crucial importance. Our research focused on the extremophilic bacteria Acidithiobacillus ferrooxidans and Leptospirillum ferrooxidans, because of the "simplicity" and knowledge about the interactions of these bacteria with their substrate/substratum and their environment. For this purpose, the composition of the corresponding extracellular polymeric substances and their functions were analyzed. The extracellular polymeric substances of both species consist mainly of neutral sugars and lipids. The functions of the exopolymers seem to be: (i) to mediate attachment to a (metal) sulfide surface, and (ii) to concentrate iron(III) ions by complexation through uronic acids or other residues at the mineral surface, thus, allowing an oxidative attack on the sulfide. Consequently, dissolution of the metal sulfide is enhanced, which may result in an acceleration of 20- to 100-fold of the bioleaching process over chemical leaching. Experiments were performed to elucidate the importance of the iron(III) ions complexed by extracellular polymeric substances for strain-specific differences in oxidative activity for pyrite. Strains of A. ferrooxidans with a high amount of iron(III) ions in their extracellular polymeric substances possess greater oxidation activity than those with fewer iron(III) ions. These data provide insight into the function of and consequently the advantages that extracellular polymeric substances provide to bacteria. The role of extracellular polymeric substances for attachment under the conditions of a space station and resulting effects like biofouling, biocorrosion, malodorous gases, etc. will be discussed.

Fenton's reagent has been shown to be a feasible technique to treat phenolic-type compounds present in a variety of food processing industry wastewaters. A model compound, p-hydroxybenzoic acid was oxidised by continuously pumping two solutions of ferrous iron and hydrogen peroxide. Typical operating variables like reagent feeding concentrations and flowrate, temperature and pH were studied. A mechanism of reactions based on the classical Fenton's chemistry was assumed, and computed concentration profiles of the parent compound, ferrous ion and dihydroxybenzene were compared to experimental results. The model qualitatively predicted the influence of several operating variables, however, calculated results suggested the presence of parallel routes of substrate elimination and/or a initiating rate constant with a higher value. The low efficiency of a well-known hydroxyl radical scavenger (tert-butyl alcohol) also supports the contribution of oxidising species different from the hydroxyl radical to substrate removal. Further evidence of the presence of reactions different from the hydroxyl radical oxidation was observed from comparison of the simultaneous Fenton's or UV/H2O2 oxidations of p-hydroxybenzoic acid, tyrosol and p-coumaric acid.
