For the first time, this paper aimed to evaluate nine ionophore and synthetic coccidiostat residues in poultry muscle samples, obtained from different production types, by solid-liquid extraction followed by liquid chromatography with tandem mass spectrometry (LC-MS/MS). The fully validated methodology was successfully applied to a total of 101 chicken and turkey samples obtained from canteens, supermarkets, and home productions in Portugal. Halofuginone, diclazuril, decoquinate, narasin, lasalocid, and salinomycin were detected in 20.8% of the samples. Home raised samples showed a greater frequency, 47.1%. The synthetic coccidiostats halofuginone, diclazuril, and decoquinate were found in averages of 0.7 μg kg-1,2.9 μg kg-1, and 3.7 μg kg-1, respectively, while averages of 1.2 μg kg-1, 1.6 μg kg-1, and 1.3 μg kg-1 were found regarding the ionophores narasin, lasalocid, and salinomycin. As for the risk assessment, values lower than 8.06% of the acceptable daily intake (ADI) were observed, indicating that exposure to coccidiostats through consumption of poultry meat does not represent risk to consumers.

The occurrence, concentrations, and variables associated with tetracycline, polyether ionophore, and anthelmintic residues in the livers of chickens sold in the informal market in South Africa were determined. Ultrahigh-performance liquid chromatography-tandem mass spectrometry was used to simultaneously analyze for four tetracyclines, five polyether ionophores, and six anthelmintic residues. The study determined the presence of residues in liver samples at both the limit of quantifications (LOQ) and concentrations over the maximum residue limit (MRL), i.e., noncompliant. Doxycycline (tetracycline group) was detected in 24 (24.5%) of 98 chicken livers, and 15 (15.3%) of the 98 were noncompliant. Mean ± standard deviation (SD) concentrations of 919.04 ± 1,081.30 ppb (LOQ) and 1,410.57 ± 108.89 ppb (MRL) were obtained. Maduramicin was detected in 27 (27.6%) of 98 chicken livers, and 19 (19.4%) of 98 were noncompliant. The mean ± SD for LOQ was 117.96 ± 84.56 ppb, and MRL was 153.21 ± 76.29 ppb. The concentrations of residues of doxycycline and maduramicin in chicken livers varied significantly across townships. Lasalocid was found in 31 (31.6%) of 98 samples, of which 5 (5.1%) had concentrations above the MRL. The mean ± SD concentration of lasalocid was 62.90 ± 170.84 ppb for samples in which lasalocid was quantified and 310.16 ± 356.68 ppb for noncompliant samples. Detectable concentrations of praziquantel, closantel, and rafoxanide (anthelmintics) residues were found in 3 (3.1%), 1 (1.0%), and 2 (2.0%) of 98 chicken livers, respectively. The presence of residues of three classes of veterinary drugs in chicken livers poses food safety implications to consumers and indicates a need for enhanced regulatory enforcement in controlling these drugs in South Africa. Moxidectin (MXD), an antiparasitic drug, is effective for a variety of external and internal parasites in companion and farm animals. This study aimed to calculate the withdrawal period by investigating the residue depletion of MXD in swine edible tissues after pouring at the dosage of 2.5 mg/kg B.W. The concentrations of MXD in swine edible tissues were determined by a modified preparation procedure based on HPLC-FLD. The method was validated giving LOD and LOQ of 0.5 μg/kg and 1 μg/kg respectively with measured recoveries ranging from 62.9%-89.2% at three different concentrations and a precision (RSD) of less or equal to 15.7%. The muscle, liver, kidney and fat tissues were collected at 0.5, 5, 10, 20, 25 d after administration. The results showed that fat was the target tissue with the highest concentration for MXD. The withdrawal period was 26 days for the MRL of 500 μg/kg in fat. The results provide fundamental information to ensure food safety and establishment of a rational medication regimen.

Residues of veterinary drugs in poultry meat have serious health effects on humans (e.g., antimicrobial resistance, carcinogenicity, and hypersensitivity), which make the control of veterinary drug residues an important parameter in ensuring consumer protection. This work was performed to quantitatively determine two co-formulated anti-infective veterinary agents, tylosin tartrate (TYT) and doxycycline hydrochloride (DOX) in different tissues of broiler chickens (liver, muscles, and fat) using high performance liquid chromatography. The chicken was treated with the recommended dose of a binary mixture of the drugs (Tydovet). Moreover, the study aimed to estimate the withdrawal time of both drugs in chicken tissues. The analysis was done by solvent extraction and solid-phase extraction for clean-up of samples from the tissue matrix, followed by liquid chromatographic determination of the cited drugs with UV-detection. Residue decline with time was tracked, and both antibiotics were found to be more persistent in liver tissues than other tissues (muscle and fat). The effect of freezing and cooking was investigated on tissue residue levels. While freezing had little effect on the concentration of both antibiotics; cooking, as anticipated, led to a marked decline. Therefore, it is recommended to pay attention to the proper withdrawal periods before marketing to ensure the hygienic suitability of broilers edibles for safe human consumption. PRACTICAL APPLICATION: This novel study measures tylosin and doxycycline residues simultaneously in different tissues (muscle, fat, and liver) after administration of Tydovet powder to the broiler chicken. Residues in fat persisted for a longer time than in muscle in case of TYT, whereas the reverse was noticed in DOX.

It is commonly known that the widespread use of antibiotics has led to their existence in food products as residues and ingestion of these food products may create a selection pressure on bacteria inhabiting the human body. In this study, an optimized method for the analysis of antibiotic residues in different food groups, including cereals, meat, eggs, milk, vegetables, and fruits, was developed using solvent extraction, solid-phase extraction cleanup, and liquid chromatography-mass spectrometry (LC-MS/MS). The limits of detection (LODs) were achieved as 0.007-1.1, 0.008-0.46, 0.002-0.67, 0.007-0.63, 0.001-0.098, and 0.005-0.26 ng/g in ng/g in cereals, meat, eggs, milk, vegetables, and fruits, respectively. The overall average recoveries at three spiking levels of the 81 antibiotics (5, 25, and 50 ng/g dry weight) were 82 ± 26, 77 ± 26, 70 ± 34, 69 ± 31, 73 ± 29, and 62 ± 37% in cereals, meat, eggs, milk, vegetables, and fruits, respectively. The method was then applied to the analysis of the targets in the collected wheat flour, mutton, chicken egg, boxed milk, cabbage, and banana samples, with the total concentration of the antibiotics detected being 4.4, 2.3, 36, 5.5, 2.7, and 14 ng/g, respectively. This work suggests that the developed method provides a time- and cost-effective method to identify and quantify antibiotic residues in common food products.

A simple, rapid and sensitive screening method by liquid chromatography-tandem mass spectrometry (LC-MS/MS) was developed for the identification of 7 macrolides (clarithromycin, erythromycin, oleandomycin, spiramycin, tilmicosin, troleandomycin and tylosin) and 8 quinolones (ciprofloxacin, difloxacin, enrofloxacin, flumequine, moxifloxacin, nalidixic acid, norfloxacin and ofloxacin) in meat and egg-based baby foods. Sample preparation was performed using an alkaline modified QuEChERS (Quick, Easy, Cheap, Effective, Rugged, Safe) extraction method without additional clean-up steps. A simplex-lattice mixture experimental design was used in the optimization of the QuEChERS extraction solvent. The developed method was successfully validated according to the Commission Decision 2002/657/EC and the European Community Reference Laboratories Residues Guidelines regarding the validation of screening methods 20/01/2010, adopting a fixed permited tolerance for relative ion ratio. Samples of baby food (n = 44) commercialized in Rio de Janeiro, Brazil, were analyzed using the validated method and none of them presented residues of the searched macrolides and quinolones, with a screening target value of 5 µg kg

The use of antimicrobials in animals for growth promotion and infection prevention significantly contributes to the development of antimicrobial resistance (AMR), a growing public health threat. While the World Health Organization (WHO), the United Nations (UN) and the European Union (EU) have taken steps towards reducing and restricting the use of antimicrobials in animals, initiatives are insufficient in developing countries where the demands for food animals continue to rise over the years. The inter-sectoral acknowledgment of inextricable link between animal health, human health and the environment (One Health approach) is critical. Concerted and collaborative efforts among all the stakeholders are essential to deal with this complex problem of resistance.

In Brazil, the buffalo milk market has been growing. However, identity and quality standards have not been established for this raw material, nor have proper distinctions between buffalo milk and bovine milk been defined. Currently, the State of Rio Grande do Sul (RS) has only three producers that supply raw material for officially marketed derivatives. The aim of this study was to determine the identity and quality standards of raw buffalo milk in this region. Samples were obtained biweekly from three farm cooling tanks between June 2017 and August 2018, to reach a total of 69 samples. The averages for the results of the physicochemical parameters fat, protein, lactose, total solids, SNF (solids-not-fat), calcium, density, FP, acidity and SCC were 5.5 g/100 g, 4.06 g/100 g, 5.07 g/100 g, 15.5 g/100 g, 9.96 g/100 g, 0.161 g/100 g, 1.034 g/ml, −0.527°C, 16°D and 95 × 103 cells/ml, respectively. With reference to the microbiological parameters, the mean of the Standard Plate Count (SPC) and thermotolerant coliforms were 9,0 × 104 CFU/ml and 1.6 × 102 MPN/ml, respectively. Regarding coagulase-positive staphylococci, 36 samples tested positive (52% of total). Neither Salmonella spp. nor Listeria monocytogenes, nor antibiotic or antiparasitic residues were detected in any sample. In conclusion, the buffalo milk used as raw material for dairy products in southern Brazil demonstrated satisfactory physicochemical and microbiological characteristics, in accordance with recent scientific literature.

A novel, sensitive and rapid high performance liquid chromatography (HPLC) method for the determination of ceftiofur by pre-column derivatization with 1,2-naphthoquinone-4-sulfonate. Analysis was performed within 5 min on a Kinetex C18 column based on core-shell technology. The mobile phase composed of acetonitrile-water (50:50, v/v) pumped isocratically at a flow rate of 1.0 mL/min under UV detection at 254 nm. The factors affecting the derivatization reaction and separation conditions were carefully evaluated and optimized. The method was linear over the concentration range of 45–450 ng/mL with a limit of detection of 3.29 ng/mL and limit of quantitation of 10.97 ng/mL. The new method was successfully applied for the analysis of ceftiofur in the veterinary formulation and honey with average recoveries of 100.78% and 98. 83%, respectively. The present method is suitable and favorable for the analysis of ceftiofur on account of its sensitivity, rapidity and cost-effectiveness. In addition, it could have significant application for the determination of ceftiofur in other food products.

A simplified method is described for reducing the analysis time of nitrofurans (NFs) and nitrofuran metabolites (NFMs) in the aquatic animals. Most existing HPLC-MS/MS methods are intended only for NFMs and are based on their fast metabolic transformations. We optimized a method for simultaneously detecting major NFs and their metabolites, including nitrovin (NV) that imply use of an optimized buffer solution. The novel method was validated by six different aquatic animal matrices (loach, catfish, shrimp, lobster, scallop, and eel) spiked with the analytes at 0.5, 1.0, and 2.0 μg kg−1. Recovery rates and %RSDs (relative standard deviations) of 82–97% and 1–8% were observed for NFMs, respectively, with values of 70–96% and 1–8% obtained for furazolidone, furaltadone, nitrofurazone, nitrofurantoin, and NV, respectively. Linearity was observed in the 0.1–20 μg L−1 range, with correlation coefficients greater than 0.99 recorded for all compounds. The developed method is sensitive, accurate, easier to use, and faster than previous methods when applied to real samples. To the best of our knowledge, this is the first method that can simultaneously determine NFs and their metabolites, as well as NV, using a single-step extraction process.

Antibiotic residues are major contaminants in milk because of their use in agriculture and animal husbandry. In particular, streptomycin, an aminoglycoside antibiotic, is a potential risk to consumers because of its ototoxicity, anaphylaxis, and growth inhibition. Herein, monoclonal antibodies for streptomycin were conjugated with europium microspheres to serve as detection probes for the development of a chromatographic time-resolved fluoroimmunoassay to detect streptomycin residues in milk. The method had a low detection limit of 0.58 µg/kg, a linear range of 0.8 to 6.25 μg/kg, and substantial recovery, from 85.6 to 108.3%. It showed slight cross-reactivity with another aminoglycoside analog. Strong correlations between the results of established chromatographic time-resolved fluoroimmunoassay and ultra-performance liquid chromatography-tandem mass spectrometry indicated that the established fluoroimmunoassay is a reliable method for rapid onsite detection of streptomycin in milk and it has great potential in food safety monitoring.

Marbofloxacin (MBF) is a key class of synthetic fluoroquinolone antibiotic that is commonly used as a veterinary drug. However, excess residue of MBF in animal-derived food samples, such as milk, is harmful to consumers. In this study, 4 mAb against MBF, namely, M4E3, M7A6, M3C7, and M5C6, were produced. Indirect competitive (ic) ELISA revealed that the M4E3 exhibited the highest sensitivity with a half-maximal inhibitory concentration (IC50) of 0.07 ng/mL and a limit of detection of 0.01 ng/mL for detection of MBF. The results of the cross-reactivity experiment revealed that the M4E3 could detect lomefloxacin, ofloxacin, fleroxacin, pefloxacin, danofloxacin, and enrofloxacin sensitively with IC50 of 0.02, 0.07, 0.18, 0.27, 0.30, and 0.32 ng/mL, respectively. Meanwhile, a cross-reactivity experiment showed that the M4E3 had a crossover rate of more than 20% with these fluoroquinolone analogs. A weak crossover rate below 1.11% was observed with 14 other fluoroquinolone analogs. The recovery rate of MBF in milk ranged from 72.28% to 129.19%, which showed that the developed indirect competitive ELISA was effective in detecting MBF in milk. Furthermore, a visual colloidal gold-based immunochromatographic assay was developed for detecting MBF with a cut-off value of 1 ng/mL in both phosphate-buffered saline and a milk sample by using this mAb. Given this sensitive mAb, both indirect competitive ELISA and colloidal gold-based immunochromatographic assay could be effective screening tools for detection of MBF in milk.

An environmentally friendly method for the determination of testosterone and methyltestosterone by acid–base-induced deep eutectic solvents liquid–liquid microextraction (DES-ABLLME) combining with high-performance liquid chromatography was established. The deep eutectic solvent (DES) consisting of menthol:lauric acid:decanoic acid (3:1:1) can act as both hydrogen bond donor and hydrogen bond acceptor. In this approach, ammonia solution (NH3•H2O) is used as an emulsifier to react with DESs in the extraction process to generate salt and form milky white solution, achieving high extraction efficiency. Hydrochloric acid was used as a phase separator to change the emulsification state and promote the separation of extraction agent from water phase. A series of parameters were optimized including the volume of DES and the emulsifying agent, glucose concentration as well as hydrochloric acid volume. The method was linear in the range 0.5–100 μg mL−1 with a correlation coefficient (R) of 0.9999, and the limits of detection were 0.067 and 0.2 μg mL−1 for testosterone and methyltestosterone, respectively. This method was applied to analyze testosterone and methyltestosterone in milk samples, and the recoveries were between 89.2 and 108.2%.

Antibiotic abuse in agricultural products leads to serious food safety issues. To this end, we proposed a mix-and-read and enzyme-free amplified assay for antibiotics based on a dual triple helix-aptamer probe, potentially applicable for on-site monitoring of antibiotic residues. A dual triple helix-aptamer probe can leverage the response toward target molecules without enzyme-based amplification, rendering it sensitive and robust for profiling target molecules. The proposed assay allowed mix-and-read detection of chloramphenicol with a detection limit of 0.18 nM. Besides, it accommodated for specifically resolving chloramphenicol among other antibiotics. Chloramphenicol residual in aquatic products in fish and milk can be precisely determined. Thus, the aptamer probe deems to enrich the toolbox for managing antibiotic use.

The use of veterinary drugs in food-producing animals may result in the presence of low levels of drug residues in these edible, animal-derived foods, with potential dietary exposure to humans. Since therapeutic doses of antibiotics have been shown to affect bacterial populations in the gastrointestinal tract microbiome and can also promote the emergence of antibiotic-resistant bacteria, there is concern that animal drugs at residue level concentrations could also perturb the intestinal microbiome composition and modify the antimicrobial resistance profile of the human intestinal microbiota. This review provides updated information on the VICH GL#36(R2), on evaluating the safety of veterinary drug residues in animal-derived foods and their effects on the human intestinal microbiome; discusses critical research knowledge gaps and challenges in evaluating the impact of drug residues in animal-derived foods on the human intestinal microbiome; and analyzes integrated basic and applied research approaches, currently being conducted at FDA, on studies that specifically address key regulatory science questions. Moreover, this review aims to emphasize future research needs on scientific methodology and provides general recommendations on drug inactivation, bioavailability, and antimicrobial resistance, to improve the safety evaluation and risk assessment of antimicrobial residues and their impact on the gastrointestinal microbiota, with the goal of ensuring food safety.

The study aimed to evaluate the effects of heat on macrocyclic lactone residues in cows' milk. Ivermectin, abamectin, doramectin, eprinomectin and moxidectin were added to raw milk in three concentrations. The milk was then pasteurised (40 seconds at 74°C or 1 minute at 80°C) and boiled (10 minutes at 100°C). The analyses were performed with a validated method: LC-MS/MS. Thermal treatment resulted in a statistically significant decrease in the abamectin, eprinomectin, and moxidectin concentrations in the milk; however, the residues did not completely degrade. Boiling resulted in a greater decrease in the moxidectin concentrations than was observed with pasteurisation. The high pasteurisation and boiling processes had a greater effect on the eprinomectin residues than did the low pasteurisation process. The pasteurisation and boiling processes did not have an effect on the doramectin and ivermectin. The study concluded that the macrocyclic lactones are generally resistant to such processes.

In this short Research Reflection I address and refute the suggestion that oestrogens consumed in milk might contribute in a significant way to endogenous levels and thereby have a physiological action, possibly resulting in adverse consequences including increased breast cancer risk. Quantitative analysis based on published data shows that, even in worst case scenarios, oestrogen consumption in milk is considerably less than regulatory bodies regard as entirely safe.

Testing milk for antibiotics before acceptance into dairies is required by the U.S. Pasteurized Milk Ordinance. Technological advances in tests have reduced screening times and improved detection accuracy. This work describes the validation of the Charm Rapid One Step Assay Beta-Lactam 30 Second Test according to the U.S. Food and Drug Administration Center for Veterinary Medicine protocol for raw commingled milk. Milk is added to the lateral flow test strip in an incubator/reader to deliver a 30 second result. Independent laboratory validation followed sensitivity, interference, and incurred residue protocols. Sensitivity, in parts per billion (ppb = µg/kg), using a probit curve determined 90% percent detection with 95% confidence, which met National Conference of Interstate Milk Shipments (NCIMS) specifications. Six U.S. approved beta-lactam drugs were detected below, but within 50% of, target/tolerance levels for penicillin G 2.9 ppb, ampicillin 5.9 ppb, amoxicillin 5.8 ppb, cephapirin 13 ppb, cloxacillin 8.1 ppb, and ceftiofur metabolites 73 ppb. No interferences were observed from 33 animal drugs at 100 ppb, somatic cells at 1.2 million/mL, or bacterial levels of >300 000 CFU/mL. Incurred residue detection levels were similar to levels determined with the spiked parent compound. The data support NCIMS that the BL30SEC method met U.S. criteria for testing milk for beta-lactams.

It is often difficult to compare studies examining the effects of endectocides on dung fauna because of different experimental approaches, for example, active ingredients (eprinomectin, doramectin, ivermectin, moxidectin) and formulations (injectable, pour-on, spiked). To gain a better understanding, we performed a quantitative meta-analysis using 22 studies to assess the overall effect of endectocide residues on the occurrence (presence or absence) and abundance of aphodiine dung beetles. Our results document a positive effect on the occurrence of adult beetles, indicating that adults tend to be attracted to dung with residues. Conversely, larvae are less likely to occur in the presence of residues. Thus, either adults that colonize dung with residues do not lay eggs or, more likely, the larvae that hatch from these eggs die early in development. Abundance of adult and larval stages was shown to be significantly reduced in dung containing residues. When individual endectocides were compared, only ivermectin demonstrated a significantly negative effect on the abundance of both adults and larvae, possibly owing to a small sample size for other agents. In laboratory studies, only dung "spiked" with endectocides reduced the abundance of larvae, whereas during field research, only pour-on applications were shown to reduce the abundance of larvae. The present study further documents the nontarget effects of endectocide residues on dung-dwelling organisms, provides robust evidence on the consequences of different application methods, and emphasizes the need for standardized methodological techniques in future studies. Clenbuterol is a steroid-type drug used in respiratory treatments in both humans and animals. However, it has a secondary effect related to the hypertrophy process in muscle and fat reduction. The illegal or bad use of clenbuterol has been reported in several countries, but there is scarce information in South America, where the production and consumption of meat are considerable. In this sense, the present study aimed at evaluating the occurrence of clenbuterol in bovine muscle and liver samples from a high cattle production area of Ecuador in 2015 and 2018. For this purpose, 57-58 samples were evaluated in 2015 and 20 samples in 2018 using the Enzyme-Linked Inmuno Sorbent Assay and ultrahigh-performance liquid chromatography-tandem mass spectrometry. The results showed complained results for clenbuterol in meat samples from both years and 23% (2015) and 85% (2018) of the samples of meat complied the maximum residue level defined by CODEX.

The study was carried out to evaluate the pharmacokinetic disposition of enrofloxacin (ENF) with a single dose of 20 mg/kg after oral administration in largemouth bass (Micropterus salmoides) at 28°C. The concentrations of ENF and of its metabolite ciprofloxacin (CIP) in plasma, liver, and muscle plus skin in natural proportions were determined using HPLC. The concentration-time data for ENF in plasma were best described by a two-compartment open model. After oral administration, the maximum ENF concentration (Cmax ) of 10.99 μg/ml was obtained at 0.60 hr. The absorption half-life (T1/2Ka ) of ENF was calculated to be 0.07 hr whereas the elimination half-life (T1/2β ) of the drug was 90.79 hr. The estimates of area under the plasma concentration-time curve (AUC) and apparent volume of distribution (Vd/F) were 1,185.73 μg hr/ml and 2.21 L/kg, respectively. ENF residues were slowly depleted from the liver and muscle plus skin of largemouth bass with the T1/2β of 124.73 and 115.14 hr, respectively. Very low levels of ciprofloxacin were detected in the plasma and tissues. A withdrawal time of 24 days was necessary to ensure that the residues of ENF + CIP in muscle plus skin were less than the maximal residue limit (MRL) of 100 μg/kg established by the European Union.

A qualitative 3 min one-step assay for detecting beta-lactam, sulfonamide, and tetracycline antibiotics was validated following milk screening test guidelines developed by FDA-CVM, AOAC-RI, and IDF. The validated 90% detection levels with 95% confidence were: penicillin G 2 part per billion (ppb); amoxicillin 4 ppb; ampicillin 9 ppb; ceftiofur plus metabolites 50 ppb; cloxacillin 9 ppb; cephapirin 15 ppb; sulfadimethoxine 8 ppb; sulfamethazine 9 ppb; chlortetracycline 34 ppb; oxytetracycline 53 ppb; and tetracycline 42 ppb. Detection levels were lower than U.S. and Canadian allowable limits for milk and were consistent with most European Maximum Residue Limits. Detection of ciprofloxacin residues in milk by sensitive and rapid methods is of great interest due to its use in the treatment of dairy livestock health. Current analytical approaches to antibiotics detection, are laboratory-based methods and they are time-consuming and require trained personnel. To cope this problem, we propose an assay, based on fluorescence polarization principle, able to detect the presence of ciprofloxacin in diluted milk sample without any pre-treatment. The proposed method is based on the use of ciprofloxacin-protein conjugate labeled with near infrared fluorescence dye, which upon binding to specific antibody causes an increase of the fluorescence polarization emission signal. The developed assay allows for the detection of ciprofloxacin at a concentration of 1ppb, which represents an amount lower than the maximum residual limit (MRL) of ciprofloxacin in milk, as set by the European Union regulation (100 ppb).

A rapid analytical method was developed and validated for the analysis of eight bound nitrofurans in animal tissue, shortening laboratory turnaround times from 4 to 2 days. The majority of methodologies for nitrofuran analysis focus on the detection of only four drugs (nitrofurantoin, furazolidone, furaltadone and nitrofurazone), and is time-consuming given the 16-h overnight derivatisation step and a double liquid–liquid extraction. In this study, the narrow scope of analysis was addressed by including further four important nitrofuran drugs (nifursol, nitrofuroxazide, nifuraldezone and nitrovin). Full chromatographic separation was achieved for the metabolites of all eight nitrofurans, using phenyl-hexyl column chemistry and a rigorous optimisation of the mobile phase additives and gradient profile. The conventional, lengthy sample preparation was substantially shortened by replacing the traditional overnight water bath derivatisation with a rapid 2-h microwave-assisted reaction, followed by a modified-QuEChERS (Quick, Easy, Cheap, Effective, Rugged and Safe) extraction. This confirmatory method was fully validated in accordance with the new 2021/808/EC legislation, and was shown to perform satisfactorily when applied to incurred tissues. The decision limit (CCα) for the eight analytes ranged between 0.013 and 0.200 µg kg−1, showing abundant sensitivity given that the current RPA for nitrofurans is 0.5 µg kg−1. This innovative method can play a major role in the surveillance of the illegal use of nitrofuran drugs. To control the drug residues in foods, the national Maximum Residue Limits (MRLs) have been determined by the international organizations; however, no regulation has been complied for MRL in food animals in Iran. This study was a systematic review and meta-analysis about studies of antibiotic residues in slaughtered poultry carcasses, in Iran. The information was first collected and analyzed throughout six international databases, and four Iranian national databases. According to the data, the total prevalence of antibiotic residues was obtained as 39.41%, 5.40%, and 0.30% using four-plate test (FPT), High-Performance Liquid Chromatography (HPLC), and Enzyme-linked immuno_sorbent assay (ELISA) methods, respectively. The highest prevalence of the residues was shown in the liver by both methods of HPLC (47.75%) and ELISA (14.35%). Moreover, the total mean of antibiotics was recorded as 71.03 ppb and 65.86 ppb in different tissues using the HPLC and ELISA method, respectively. Based on this study, we can conclude that the prevalence of antibiotic residue in poultry meat in Iran is high and that this level does not cause health problems for consumers. It is highly recommended to execute tight surveillance strategies from the government in antibiotic monitoring.

International audience; A liquid chromatography with tandem mass spectrometry (LC-MS/MS) method was developed for the simultaneous quantification of residues of spiramycin, a macrolide antibiotic, and its active metabolite neospiramycin in cow's milk as well as in minor species milk, goat and ewe. Spiramycin-d3 was used as internal standard for quantification of both analytes. This analytical method was validated using a global accuracy profile as a graphical decision tool built according to the trueness and the precision of the method. A unique and optimal linear model with logarithm transformation (with a determination coefficient of 0.9991) allowed the measurement of both analytes in the milks of the three animal species, in a wide range from 0.2 to 10 times the Maximal Residue Limit (MRL) (40-2000 µg.kg(-1)). The limits of detection and quantification were 13 µg.kg(-1) and 40 µg.kg(-1), respectively. The accuracy profile was established to get 80% of future measurements in routine assays that will fall within the acceptance limits. Trueness of the method, expressed as relative bias, was comprised between -1.6% and 5.7% over the whole range of concentrations. The mean relative standard deviation for repeatability and intermediate precision were comprised between 1.1% and 2.7%; 2.5 and 4.2%, respectively, in all levels of concentration for the three milks. Moreover, a two-order polynomial function was used to model the relative expanded uncertainty with a determination coefficient of 0.834. This function aimed to determine the uncertainty of the future quantifications within the validated dosing range. Overall, the global accuracy profile highlighted the reliability of the method for the routine assays of spiramycin and neospiramycin even in milk from minor species (goat, ewe) by using the most accessible milk (often from cow), while guaranteeing a very high proportion of samples within the fixed acceptance limits. The applicability of this method was tested during a depletion study of spiramycin and neospiramycin in the milk of cow, goat and ewe. The developed analytical method will be useful to assess the distribution profile of the antibiotic and its metabolite in milk of minor species where few studies are available.

The simultaneous determination of multiple analytes has been an urgent demand in screening of antibiotic residues in food products of animal origin due to its higher analysis efficiency. Five aminoglycoside antibiotics (AGAs) have been monitored in milk, including gentamicin (GEN), kanamycin (KAN), neomycin (NEO), and streptomycin/dihydrostreptomycin (STR/diSTR). A chemiluminescence microarray immunoassay (CLMIA) based on nitrocellulose membrane had been developed for the detection of multiple AGAs, which the LODs for STR, KAN, NEO, and GEN were 4.74 ng/mL, 4.97 ng/mL, 2.99 ng/mL, and 4.42 ng/mL respectively. To improve the sensitivity of immunoassay, single-well carbon tubes (SWCNTs) were utilized as solid support for loading horseradish peroxidase-labelled goat anti-mouse antibody to obtain the multi-enzyme particles. After the optimization of usage of multi-enzyme particles and antibodies, the enhanced CLMIA was established and evaluated. The LODs were 1.25 ng/mL for STR, 0.64 ng/mL for KAN, 0.38 ng/mL for GEN, and 0.39 ng/mL for NEO, which was improved by threefold, sevenfold, 11-fold, and sevenfold compared with the conventional CLMIA developed. These methods presented higher specificity and repeatability. Finally, the enhanced CLMIA based on CNT-assisted multi-enzyme particles was utilized to analyze twenty-five milk samples from local market and dairy farm, which all the results were below the LOD. The enhanced CLMIA showed the great application potential for the detection of multiple targets simultaneously and provided efficient tool for the screening of pollutants in food.

An innovative, rapid and stable method for simultaneous determination of three tetracycline (oxytetracycline, tetracycline and doxycycline) and two fluoroquinolone (ciprofloxacin and enrofloxacin) residues in poultry eggs by ultra-high performance liquid chromatography–fluorescence detection (UPLC-FLD) was established and optimized. The samples were homogenized and extracted with acetonitrile/ultrapure water (90:10, v/v) and then purified by solid-phase extraction (SPE). LC separation was achieved on an ACQUITY UPLC BEH C18 column (1.7 µm, 2.1 mm × 100 mm), and the mobile phase was composed of acetonitrile and a 0.1 mol/L malonic acid solution containing 50 mmol/L magnesium chloride (the pH was adjusted to 5.5 with ammonia). When the five target drugs were spiked at the limit of quantification, 0.5 times the maximum residue limit (MRL), 1.0 MRL and 2.0 MRL, the recoveries were above 83.5% and the precision ranged from 1.99% to 6.24%. These figures of merit complied with the parameter validation regulations of the EU and U.S. FDA. The limits of detection and quantifications of the targets were 0.1–13.4 µg/kg and 0.3–40.1 µg/kg, respectively. The proposed method was easily extended to quantitative analyses of target drug residues in 85 egg samples, thus demonstrating its reliability and applicability.

A recombinant gene CCS consisting of three single genes (CAP-ScFv, CPFX-ScFv and SM2-ScFv) were constructed by PCR with three pairs of primers. The length of CCS gene fragment was 2,260 bp. A recombinant plasmid (pGEX-CCS) was obtained in pGEX-6p-1. pGEX-CCS was induced in E.coli BL21(DE3), and the molecular weight of recombinant fusion protein (GST-CCS) was 108.87 kDa. GST-CCS was successfully applied to analysis of CAP-OVA (CAP-ovalbumin conjugate), CPFX-OVA and SM2-OVA simultaneously. The sensitivity of GST-CCS against three veterinary drugs was tested by indirect ELISA at dilution ratio 1:8000. These findings provide a foundation for the construction of fusion genes with linkers and for the potential development of a rapid screening method for the simultaneous detection of veterinary drug residues.

This study established a rapid and reliable method to determine chloramphenicol (CAP), thiamphenicol (TAP) and florfenicol (FF) residues in Chinese gelatin medicines. CAP, TAP and FF were extracted from medicine samples using 2% (v/v) ammonium hydroxide in acetonitrile. Trypsin was used to eliminate the matrix effect caused by protein components in gelatin medicines, whereas anhydrous sodium sulfate, C18-N and NH2-PSA adsorbents were applied to reduce matrix effect induced by other components. The analytical method of these drugs was optimized on ultra high-performance liquid chromatography-mass spectrometer (UHPLC-MS/MS) through the analysis of their standard linearity and regression. The optimized extraction and analytical method were validated in one Chinese gelatin medicine sample (Colla corii asini, E Jiao) with three fortification levels (2, 5 and 10 μg/kg), and the recoveries of these drug residues ranged of 87.6–102.7%. The limit of detection and quantification of CAP, TAP and FF in the sample were 0.2 and 0.5 μg/kg, 0.4 and 1.5 μg/kg, and 0.5 and 1.5 μg/kg, respectively. A total of 30 Chinese gelatin medicine samples were analyzed using the established method. No drug residues were found in these samples except for one Testudinis Carapacis et Plastri (1.67 μg/kg FF) and one turtle shell glue (2.55 μg/kg FF).

Excessive use of antibiotics in aquatic products is a serious problem for food safety and human health, and on-site detection of antibiotics is highly demanded. Herein, we proposed multivalence aptamer probes, allowing sensitive, label-free, and homogeneous detection of antibiotics in different aquatic products. Compared to commonly used aptamers, multivalence aptamer probes can provide multiple binding sites and a higher affinity for target molecules, and the iterative binding on different binding sites contributes to an amplified recognition effect, sharply increasing the response and sensitivity of aptamer probes. The 2-valence aptamer probes conferred a limit of detection of 0.097 nM for kanamycin detection, where it is estimated that their sensitivity is enhanced 12 times compared to 1-valence aptamer probes. Meanwhile, multivalence aptamer probes allowed us to specifically identify kanamycin among other antibiotics. It could detect kanamycin residual in aquatic products including river eel and puffer fish, as well as tap water with high precision. A multivalence design strategy of aptamer probes would significantly improve the detection performance of aptamers, facilitating the translation of aptamer for food safety control.

By utilizing the coffee-ring effect and microfluidic paper-based analytical devices (µPADs), this study improved the sensitivity of the determination of norfloxacin in four different food matrices. Micro-PADs in this study were fabricated by designing and embedding wax channels onto cellulose-based filter paper through printing and subjecting the paper to heat to allow the wax to penetrate the paper. Determination of norfloxacin concentration in food samples was achieved by monitoring the colorimetric reaction that occurred between norfloxacin and the added iron (III) nitrate nonahydrate in 5 mM ammonia in each reaction chamber. A transition metal hydroxide was formed through this reaction that resulted in the formation of a solid precipitate to enable the antibiotic to bind to the iron molecule via coordination chemistry. This metal ion-antibiotic complex generated a visible color change. Following the colorimetric reaction, images were taken and subsequently analyzed via ImageJ to determine the relative pixel intensity that was used to infer norfloxacin concentration. The analytical sensitivity of this device was determined to be as low as 50 ppm when analyzing the inner-ring reaction, and as low as 5 ppm when analyzing the outer coffee ring thereby allowing for an alternative cheaper, faster, and more user-friendly method to detect norfloxacin than the conventional methods. PRACTICAL APPLICATION: This novel paper-based microfluidic device can achieve the detection of antibiotic residues in agrifoods in a faster, cheaper, and more user-friendly manner.

United States dairy operations use antibiotics (primarily β-lactams and tetracyclines) to manage bacterial diseases in dairy cattle. Antibiotic residues, antibiotic-resistant bacteria (ARB), and antibiotic resistance genes (ARG) can be found in dairy manure and may contribute to the spread of antibiotic resistance (AR). Although β-lactam residues are rarely detected in dairy manure, tetracycline residues are common and perhaps persistent. Generally,15% of bacterial pathogen dairy manure isolates are ARB, although resistance to some antibiotics (e.g., tetracycline) can be higher. Based on available data, the prevalence of medically important ARB on dairy operations is generally static or may be declining for antibiotic-resistant Staphylococcus spp. Over 60 ARG can be found in dairy manure (including β-lactam and tetracycline resistance genes), although correlations with antibiotic usage, residues, and ARB have been inconsistent, possibly because of sampling and analytical limitations. Manure treatment systems have not been specifically designed to mitigate AR, though certain treatments have some capacity to do so. Generally, well-managed aerobic compost treatments reaching higher peak temperatures (60°C) are more effective at mitigating antibiotic residues than static stockpiles, although this depends on the antibiotic residue and their interactions. Similarly, thermophilic anaerobic digesters operating under steady-state conditions may be more effective at mitigating antibiotic residues than mesophilic or irregularly operated digesters or anaerobic lagoons. The number of ARB may decline during composting and digestion or be enriched as the bacterial communities in these systems shift, affecting relative ARG abundance or acquire ARG during treatment. Antibiotic resistance genes often persist through these systems, although optimal management and higher operating temperature may facilitate their mitigation. Less is known about other manure treatments, although separation technologies may be unique in their ability to partition antibiotic residues based on sorption and solubility properties. Needed areas of study include determining natural levels of AR in dairy systems, standardizing and optimizing analytical techniques, and more studies of operating on-farm systems, so that treatment system performance and actual human health risks associated with levels of antibiotic residues, ARB, and ARG found in dairy manure can be accurately assessed.

Imprudent and superfluous use of antibiotics contributes to the selection of resistant bacteria, which is a large threat to human health. Therefore analytical procedures have been implemented in the poultry production sector to check if antibiotic treatments are registered, aiming to achieve more prudent use of antibiotics. These methods rely on the analysis of feathers, a matrix in which antibiotic residues persist. However, other routes besides direct administration, through which poultry feathers could contain antibiotic residues, should also be taken into account. In this research the vertical transmission from parent hen to broiler was investigated through a controlled animal study for the antibiotics enrofloxacin, doxycycline and sulfachlorpyridazine. Vertical transmission was observed for all antibiotics to both egg and egg shell. Also it is demonstrated that the transferred antibiotics from parent hen to chick are subsequently excreted via the chick’s droppings. Through this route, the broilers’ environment is contaminated. If eggs are hatched that were taken during treatment of the parent hen, this indirect route and/or the direct vertical transmission can eventually result in the detection of low concentrations of antibiotic residues in the broilers’ feathers at greater age

The polypeptide antibiotics colistin (COL) and bacitracin (Baci) are extensively used as veterinary drugs and feedstock additives in the livestock industry, which inevitably causes residues in animal-origin food, which can accelerate human tolerance to antibiotics. In this study, a portable lateral flow immunoassay (LFIA) for the simultaneous determination of COL and Baci residues in milk was developed. The replacement of gold nanoparticles used in the traditional LFIA with fluorescent microspheres (FMs) to label monoclonal antibodies (mAbs) allowed qualitative and quantitative analyses within a few minutes. Based on the principle of competitive binding to FM-labelled mAbs between analytes in samples and fixed antigens on the membrane, the assay provided qualitative cut-off values of 100 and 50 ng mL-1 for Baci and COL in milk samples. Furthermore, a strip reader-based semi-quantitative detection system could detect lower limits of 7.85 and 1.89 ng mL-1 for Baci and COL, respectively. In conclusion, the proposed multiplex LFIA immunosensor provides an auxiliary analytical tool for the rapid and simultaneous screening of COL and Baci in large cohorts of samples.

A novel type of enzyme-antibody conjugation using mesoporous silicon nanospheres (MSN) was developed, which amplified the labeling signal and highly increased the sensitivity of enzyme-linked immunosorbent assay (ELISA) for the determination of pesticide and veterinary drug residues in food. First, conjugates were prepared through layer-by-layer immobilization of an enzyme and an antibody on an MSN scaffold. Then the MSN scaffold was employed for labeling and signal amplification to develop a sensitive colorimetric immunoassay through the catalytic oxidation reaction of 5,50-tetramethylbenzidine (TMB). When this MSN-based ELISA was applied to detect chloramphenicol, avermectin, tetracycline and streptomycin in food samples, it provided linear ranges of 0.025 ng ml-1-25 ng ml-1, 0.05 ng ml-1-10 ng ml-1, 0.025 ng ml-1-10 ng ml-1 and 0.05 ng ml-1-25 ng ml-1, respectively, with low detection limits down to 0.011 ng mL-1, 0.134 ng mL-1, 0.015 ng ml-1 and 0.106 ng ml-1, respectively. For avermectin, it provided a 16.7-fold decrease of the limit of detection in contrast to that of standard ELISA without the loss of method specificity and accuracy. This novel immunoassay was hypersensitive, simple and easy-to-use, which made it high potential in applying for the accurate analysis of harmful substances in food.

Matrix certified reference materials (CRMs) are an indispensable part of method validation and have played an important role in ensuring reliable analytical results. To retain similarity to real samples, a new matrix CRM for the mass fraction of ciprofloxacin in whole liquid egg was developed by use of incurred materials with a target value corresponding to residue levels in real sample. The source materials were collected from laying hens following oral administration of ciprofloxacin. An optimized homogenization method and a strict bottling process were applied to bulk whole egg materials to prepare the CRM candidate. The mass fraction of ciprofloxacin in whole liquid egg was certified by a collaborative characterization program with eight accredited participating laboratories. Liquid chromatography coupled with isotope dilution mass spectrometry was studied as a reliable reference method for value assignment and was used by all participating laboratories. The certified value and expanded uncertainty (k = 2, at a confidence level of 95%) was 39.7 ± 5.2 μg/kg for ciprofloxacin in whole liquid egg. Homogeneity, long-term stability at -70 °C for 12 months, and short-term stability at -18 °C, 4 °C, and room temperature were assessed for 9 days. Additionally, uncertainties arising from inhomogeneity, instability, and characterization were analyzed in detail and fully estimated. This CRM would be a useful tool for validation of analytical methods and proficiency testing in ciprofloxacin residue analysis of egg. Graphical Abstract.

In this study, we developed a microbiological inhibition method for the rapid screening of antibiotics in milk with Geobacillus stearothermophilus ATCC12980 as an indicator bacterium and an easy sample pretreatment. We observed that the limits of detection of the kit for 34 common antibiotic residues in milk, including β-lactams (13), aminoglycosides (6), tetracyclines (4), sulfonamides (6), macrolides (4), lincosamides (1), were lower than or close to the maximum residue limits formulated by the European Union and China. Moreover, the false-positive rate was 1% and the false-negative rates were less than 5%. The ruggedness of the method (the reproducibility of detection capability of different batches of medium) met requirements at determined levels and residual limits. The shelf life of the kit was more than 6 mo at 4°C. Additionally, we observed good correlations between the kit results and ultra-high-performance liquid chromatography-tandem mass spectrometry results for incurred milk (samples taken from animals treated with antibiotics according to the pre-slaughter medication data), which indicated that the kit was reliable for screening antibiotics in incurred samples. In conclusion, the kit has a broad application potential with high sensitivity, specificity, and reproducibility, stability, and reliability, combined with simple operation, low cost, and high-throughput capacity.

The previously adopted marker residue for florfenicol (FF) in China was only florfenicol amine (FFA); however, the marker residue has been changed to FF plus FFA since the end of 2017. The previous official withdrawal period determined based on the only concentration of FFA may no longer be suitable. Therefore, the present study aimed to determine the depletion profiles of FF and FFA and further calculate the withdrawal period in the crucian carp (Carassius auratus) based on the new marker residues. Florfenicol was intramuscularly administered at 10 mg/kg bodyweight daily for five consecutive days to crucian carps reared in freshwater at 10°C. After the last dose, plasma and tissue samples were randomly collected from 10 fish at different time points. The FF and FFA concentrations were simultaneously determined by high-performance liquid chromatography (HPLC) with a fluorescence detector and further subjected to noncompartmental analysis. The elimination half-life (h) of FF in different tissues decreased as follows: liver (39.1) > kidney (36.3) > skin plus muscle (34.6) > plasma (31.7), whereas that of FFA decreased as follows: kidney (41.4) > skin plus muscle (39.4) > liver (39.3) > plasma (35.7). Considering a maximum residue limit of 1 μg/g for the total concentration of FF and FFA in the skin plus muscle, a withdrawal period of 6 days was calculated based on the upper limit of the one-sided 95% confidence interval.

Here, magnetic molecularly imprinted polymers were designed for norfloxacin via oil-in-water emulsifier-free emulsion method. It was prepared by simply mixing norfloxacin, methacrylic acid-co-ethylene glycol dimethacrylate copolymer, and Fe3 O4 together at room temperature. Characterized by multiple analytical tools, the particle size, pore size, pore volume, specific surface area, and saturation magnetization of the product were about 30 µm, 10-500 nm, 2.92 mL/g, 105.84 m2 /g, and 3.052 emu/g, respectively. And the adsorption capacity was high at 27.04 mg/g towards norfloxacin. Combined with ultra high performance liquid chromatography, it was used to determine norfloxacin in real samples. Average recoveries were above 77.44% with relative standard deviations between 1.21 and 6.85% at three spiked levels (n = 3) for lake water and pork liver. The determination was achieved for the most complex biosample pork liver spiked with norfloxacin low to 30 ng/g, about 100 times less than the maximum residue limit regulated by Commission of the European Communities. All evidences demonstrated that the magnetic molecularly imprinted polymers can be used in practice for monitoring norfloxacin either in environmental water or meat product with high accuracy and reliability.

Carbofuran residue in vegetables is a concern to human health. Direct competitive enzyme-linked immunosorbent assay (dcELISA) and dipstick immunoassay were developed in the present study. The dcELISA showed a 50% inhibition concentration (IC50 ) and working range of 1.3 and 0.2 to 7.5 ng/mL, respectively, while the cutoff value of dipstick immunoassay was 20 ng/mL. Applying the two immunoassays, we achieved the goal of rapid screening of carbofuran residue in commercial vegetables with a simple sample processing method. Among 46 leek, 39 potato, and 39 sweet potato samples, carbofuran residue was detected in 22% of the leek samples, and two samples exceeded the maximum residue limit of China (0.02 mg/kg). In addition, carbofuran residue was found at less than 2.5 ng/g in one potato and one sweet potato samples. The residual level of carbofuran measured by immunoassays agreed well with those determined by ultra-performance liquid chromatography tandem mass spectrometry. To ensure food safety and human health, it is greatly necessary and meaningful to monitor carbofuran residue in commercial vegetables. PRACTICAL APPLICATION: Rapid monitoring of carbofuran residue in vegetables is very necessary and important for consumers, regulatory agencies, and food industry.

The validity of two microbiological methods: Tube (Premi® Test) and Plate (Three Plate Test) Test for the detection of oxytetracycline (OTC) in poultry was done using Enzyme Linked Immunosorbent Assay (ELISA) immunoassay as gold standard. OTC was administered to two groups of birds: intramuscular drug administration (group A) and oral drug administration (group B). Liver and muscle tissue samples from birds in both groups were tested for the presence of OTCwith the Four Plate Test (FPT), Premi® Test and ELISA. For muscle tissues, FPT had a sensitivity of 71.4% and 60%, while Premi® Test had a sensitivity of 57% and 20% for intramuscular and orally treated birds, respectively. For the liver tissues, FPT had 87.5% and 83.5% while Premi® Test had 37.5% and 16.6% sensitivity for intramuscular and orally treated birds, respectively. The two tests had 100% specificity for OTC in tissues of birds from both treatment groups. There is a strong correlation (r = 0.93) between the inhibition zones of FPT and ELISA concentrations in OTC detection. FPT, therefore, has a higher sensitivity for OTC than Premi® Test.

A method for the simultaneous determination of 27 sulfonamides in poultry feathers using ultra performance liquid chromatography-tandem mass spectrometry (UPLC-MS/MS) was established in this study. The samples were extracted using 0.1 mol/L HCl solutions in a 60 °C water bath for 2 h, purified using hydrophilic-lipophilic balance solid-phase extraction, nitrogen-dried, and then reconstituted for UPLC-MS/MS analysis, which was performed with a CSH-C18 column. Linearity, limit of detection, limit of quantification, recovery, and precision were calculated in accordance with Commission Decision 2002/657/EC. For linearity, all standard curves showed a standard coefficient greater than 0.99, and the recoveries and coefficient of variation were 89-115% and

This Research Communication describes the residue concentration of a dry cow antibiotic in two different milk fractions and describes effects of milk fraction and milk composition on the test performance of a rapid screening and a microbial inhibitor test. Thirteen dry cows were treated with an intramammary dry cow antibiotic containing 150 mg cefquinome. Quarter foremilk and stripping samples were collected on the first 10 d postpartum. All milk samples were analyzed for milk composition by the local Dairy Herd Improvement Association and were tested for antibiotic residues using the rapid screening test Milchtest BL and the microbial inhibitor test Delvotest BR Brilliant Plates. The residue concentration of cefquinome was determined in foremilk and stripping samples from milkings 1, 2, 3, 5, and 7 after calving using high performance liquid chromatography – tandem mass spectrometry. The logarithm of cefquinome concentration (logCef) was higher in foremilk than in stripping samples and higher in milk samples with lower lactose content. Furthermore, logCef decreased with the number of milkings (P < 0.001). The Milchtest BL was more likely to be not evaluated (i.e. no test and control line or no control line appeared) in stripping samples and milk samples with higher protein content. In the Delvotest BR Brilliant Plates milk samples with higher protein content were more likely to have a false positive result (i.e. the screening test result was positive, but the HPLC-MS/MS result was below the detection limit of the screening test). These results indicate that foremilk is the recommended milk fraction to be tested for residues of cefquinome and that a high protein content can be a cause of test failure and false positive results when milk during the first 10 d postpartum is tested for antibiotic residues using screening tests.

In this study, core–shell lincomycin-imprinted polymers were successfully synthesized and their binding properties evaluated. The functional monomers of methacrylamide and acrylamide were used for synthesis of core and shell, respectively. The optimum synthesized core–shell molecularly imprinted polymer (MIP) was applied as a sorbet in solid phase extraction cartridge. Afterwards, the method of core–shell molecularly imprinted solid phase extraction (CSMISPE) was used for pre-concentration and clean-up of lincomycin in the milk matrix prior to analysis via high performance liquid chromatography equipped with UV detector (HPLC-UV). The linear range for analysis of lincomycin in the milk matrix using introduced method was obtained from 0.08 to 2 μg/mL with recovery range of 80%–89%. The limit of detection and limit of quantification were 0.02 μg/mL and 0.08 μg/mL, respectively. Finally, calibrated CSMISPE-HPLC-UV method was used for lincomycin residue checking and quantification in the pasteurized milk samples of Mashhad city market.

The abundant use of antibiotics leads to antibiotic residues in frequently consumed foods. Residual antibiotics in food may have adverse effects on humans by directly causing disease via low-dose exposure and indirect harm via antibiotic resistance. However, the current methods for antibiotic extraction and analysis in food have not yet formed a uniform standard, and only a few data exist regarding the residual antibiotic condition in various types of foods. Hence, we review the literature since 2008 to summarize analytical methods and residue status of antibiotics in food. Then, we discuss the causes of antibiotic residues in food and the possible hazards to human health. We hope that the joint efforts of the scientific community and political circles will lead to the formation of a unified standard for the extraction and analysis of antibiotics in food, to allow for comprehensive monitoring of residual antibiotics and ensure human health.

A rapid and eco-friendly method was developed to extract and pre-concentrate sulfonamides (sulfadiazine, sulfamerazine and sulfadimethoxine) from chicken meat before high-performance liquid chromatographic analysis. The method involved rapid microwave-assisted extraction with hydrochloric acid (pH = 5), further solid phase purification using HLB cartridge and 5% ammonium hydroxide, and homogeneous liquid-liquid microextraction using hexanoic acid as switchable hydrophilicity solvent. The overall time required for sample preparation was 10 min and the volume of organic solvent used for extraction was only 200 μL. The method exhibited good linearity (r2 > 0.9941), low limits of detection (2.5–6.0 ng/g) and allowed quantification of target sulfonamides spiked at levels of 50–400 ng/g in chicken meat with spiked recoveries in the range of 94.1%–112.0%.
