In response to the growing demand for biomolecular diagnostics, metasurface (MS) platforms based on high-Q resonators have demonstrated their capability to detect analytes with smart data processing and image analysis technologies. However, high-Q resonator meta-atom arrays are highly sensitive to the fabrication process and chemical surface functionalization. Thus, spectrum scanning systems are required to monitor the resonant wavelength changes at every step, from fabrication to practical sensing. In this study, we propose an innovative dielectric resonator-independent MS platform that enables spectrometer-less biomolecule detection using artificial intelligence (AI) at a visible wavelength. Functionalizing the focused vortex MS to capture gold nanoparticle (AuNP)-based sandwich immunoassays causes the resulting vortex beam profiles to be significantly affected by the localized surface plasmon resonance (LSPR) occurring between AuNPs and meta-atoms. The convolutional neural network algorithm was carefully trained to accurately classify the AuNP concentration-dependent focused vortex beam, facilitating the determination of the concentration of the targeted diagnostic biomolecule. Driven by their exceptional optical characteristics, robust chemical stability, and facile bioconjugation, gold nanoparticles (AuNPs) have emerged as a preferred material for detection and biosensing applications in scientific research. This study involves the development of a simple, rapid, and cost-effective colorimetric immuno-sensing probe to detect aflatoxin B1 and zearalenone using AuNP antibody (AuNP-mAb) conjugates. Anti-toxin antibodies were attached to the AuNPs by using the physical adsorption method. The colorimetric immunosensor developed operates on the principle that the optical properties of the AuNP are very sensitive to aggregation, which can be induced by a critical high salt concentration. Although the presence of antibodies on the AuNP surface inhibits the aggregation, these antibodies bind to the toxin with higher affinity, which leads to exposure of the surface of AuNPs and aggregation in a salt environment. The aggregation triggers a noticeable but variable alteration in color from red to purple and blueish gray, as a result of a red shift in the surface plasmon resonance band of the AuNPs. The extent of the shift is dependent on the toxin exposure dose and can be quantified using a calibration curve through UV-Visible-NIR spectroscopy. The limit of detection using this assay was determined to be as low as 0.15 ng/mL for both zearalenone and aflatoxin B1. The specificity of the prepared immunoprobe was analyzed for a particular mycotoxin in the presence of other mycotoxins. The developed immunoprobe was evaluated for real-world applicability using artificially spiked samples. This colorimetric immunoprobe based on localized surface plasmon resonance (LSPR) has a reduced detection limit compared to other immunoassays, a rapid readout, low cost, and facile fabrication.

The metastasis of cancer cells is a principal cause of morbidity and mortality in cancer. The combination of a cytosensor and photothermal therapy (PTT) cannot completely eliminate cancer cells at one time. Hence, this study aimed to design a localized surface plasmonic resonance (LSPR)-based aptasensor for a circuit of cytosensing-PTT (COCP). This was achieved by coating a novel sandwich layer of polydopamine/gold nanoparticles/polydopamine (PDA/AuNPs/PDA) around the Ω-shaped fiber-optic (Ω-FO). The short-wavelength peak of the sandwich layer with strong resonance exhibited a high refractive index sensitivity (RIS). The modification with the T-shaped aptamer endowed FO-LSPR with unique characteristics of time-dependent sensitivity enhancement behavior for a sensitive cytosensor with the lowest limit of detection (LOD) of 13 cells/mL. The long-wavelength resonance peak in the sandwich layer appears in the near-infrared region. Hence, the rate of increased localized temperature of FO-LSPR was 160 and 30-fold higher than that of the bare and PDA-coated FO, indicating strong photothermal conversion efficiency. After considering the localized temperature distribution around the FO under the flow environment, the FO-LSPR-enabled aptasensor killed 77.6% of cancer cells in simulated blood circulation after five cycles of COCP. The FO-LSPR-enabled aptasensor improved the efficiency of the cytosensor and PTT to effectively kill cancer cells, showing significant potential for application in inhibiting cancer metastasis.

The translation of silver-based nanotechnology 'from bench to bedside' requires a deep understanding of the molecular aspects of its biological action, which remains controversial at low concentrations and non-spherical morphologies. Here, we present a hemocompatibility approach based on the effect of the distinctive electronic charge distribution in silver nanoparticles (nanosilver) on blood components. According to spectroscopic, volumetric, microscopic, dynamic light scattering measurements, pro-coagulant activity tests, and cellular inspection, we determine that at extremely low nanosilver concentrations (0.125-2.5 μ g ml -1 ), there is a relevant interaction effect on the serum albumin and red blood cells (RBCs). This explanation has its origin in the surface charge distribution of nanosilver particles and their electron-mediated energy transfer mechanism. Prism-shaped nanoparticles, with anisotropic charge distributions, act at the surface level, generating a compaction of the native protein molecule. In contrast, the spherical nanosilver particle, by exhibiting isotropic surface charge, generates a polar environment comparable to the solvent. Both morphologies induce aggregation at NPs/bovine serum albumin ≈ 0.044 molar ratio values without altering the coagulation cascade tests; however, the spherical-shaped nanosilver exerts a negative impact on RBCs. Overall, our results suggest that the electron distributions of nanosilver particles, even at extremely low concentrations, are a critical factor influencing the molecular structure of blood proteins' and RBCs' membranes. Escherichia coli (E. coli) bacteria with varying solution concentrations have been successfully detected using silver nanoparticles (Ag NPs)-based localized surface plasmon resonance (LSPR) biosensors. The Ag NPs were effectively synthesized by a chemical method using trisodium citrate with L-Histidine (L-His) and deposited on the surface of Au thin film-coated half-cylinder BK-7 prisms. He-Ne laser with a wavelength of 632.8 nm was used to generate LSPR phenomena in Kretschmann configuration with prism/Au thin film/His-Ag NPs/E. coli bacteria/air structure arrangements. The variation of E. coli bacteria concentration was carried out to determine the effect of E. coli bacteria concentration on the LSPR curve characteristics. The characterization results showed that the size of Ag NPs was 18.7 nm, and that of His-Ag NPs was 17.9 nm. Selected area electron diffraction results indicated the formation of diffraction rings with the presence of lattice planes (111), (200), (220), and (311), proving the face-centered cubic crystal structure of silver. The absorbance peak of Ag NPs shifted from a wavelength of 421-414 nm with an increase in band gap energy from 2.94 eV to 2.99 eV, along with a decreased average particle size. The functional groups observed in His-Ag NPs showed wavenumbers at 3320 to 3318 cm -1 , 2106 to 2129 cm -1 , and 1635 cm -1, showing the OH, CH, and C CO bonds, respectively. The SPR angle of the prism/Au thin film/air structure is 44.80°. Meanwhile, the LSPR angle for the prism/Au thin film/His-Ag NPs/air structure is 44.92°. There is an increase in the LSPR angle by 0.12°. Moreover, the minimum reflectance increases by 0.02. After detecting E. coli bacteria, the LSPR angle shifted by 0.26°, 0.38°, and 0.49° for concentrations of 6.0 × 10 8 CFU/mL, 6.0 × 10 7 CFU/mL and 6.0 × 10 6 CFU/mL respectively. However, the minimum reflectance rose from 0.09° to 0.14°, 0.20°, and 0.22°. Moreover, SPR testing with the structure of the prism/Au thin film/E. coli bacteria/air was carried out to determine the contribution of His-Ag NPs for detecting E. coli bacteria. The results showed that no angular shift occurs. These results indicate that using Ag NPs encapsulated with L-His is essential in amplifying the SPR signal and detecting E. coli bacteria. Silver nanoparticles (AgNPs) are extensively used as an antibacterial agent, and monitoring the dissolution behavior of AgNPs in native biological environments is critical in both optimizing their performance and regulating their safety. However, current assessment methods rely on sophisticated analytical tools that are off-site and time-consuming with potential underestimations, due to complicated sample preparation. Although localized surface plasmon resonance (LSPR) sensing offers a facile method for the detection of AgNP dissolution, it is limited by low sensitivity and poor nanoparticle stability in native biological environments. Herein, we constructed a highly sensitive and stable LSPR sensor using gold-silver core-shell nanoparticles (Au@AgNPs), in combination with polymeric stabilizing agents, for the direct measurement of the Ag shell dissolution in native biological media. The high sensitivity was attributed to the acute and large LSPR shift generated by bimetallic nanoparticles. The sensor was used for the real-time monitoring of the Ag dissolution of Au@AgNPs during their co-culture with both bacteria and fibroblast cells. The media pH was found to dominate the Ag dissolution process, where Au@AgNPs exhibited bactericidal effects in the bacteria environment with relatively low pH, but they showed little toxicity towards fibroblast cells at pH 7.4. The minimum inhibition concentration of Au@AgNPs for bacterial growth was found similar to that of AgNO 3 in terms of released Ag amount. Gold nanoshells have been actively applied in industries beyond the research stage because of their unique optical properties. Although numerous methods have been reported for gold nanoshell synthesis, the labor-intensive and time-consuming production process is an issue that must be overcome to meet industrial demands. To resolve this, we report a high-throughput synthesis method for nanogap-rich gold nanoshells based on a core silica support (denoted as SiO 2 @Au NS), affording a 50-fold increase in scale by combining it with a dual-channel infusion pump system. By continuously dropping the reactant solution through the pump, nanoshells with closely packed Au nanoparticles were prepared without interparticle aggregation. The thickness of the gold nanoshells was precisely controlled at 2.3-17.2 nm by regulating the volume of the reactant solution added dropwise. Depending on the shell thickness, the plasmonic characteristics of SiO 2 @Au NS prepared by the proposed method could be tuned. Moreover, SiO 2 @Au NS exhibited surface-enhanced Raman scattering activity comparable to that of gold nanoshells prepared by a previously reported low-throughput method at the same reactant ratio. The results indicate that the proposed high-throughput synthesis method involving the use of a dual-channel infusion system will contribute to improving the productivity of SiO 2 @Au NS with tunable plasmonic characteristics.

The purpose of this study is to develop a dual fluorescence-colorimetric sensor for determination of the non-chromophoric drug, tobramycin using fluorescein-modified silver nanoparticles. Fluorescein is adsorbed on the surface of silver nanoparticles resulting in quenching of the fluorescence intensity of fluorescein at 513 nm. Upon addition of tobramycin to fluorescein-bound silver nanoparticles, tobramycin can displace fluorescein from the surface of nanoparticles resulting in nanoparticles aggregation and liberation of free fluorescein restoring its fluorescence. The interaction of tobramycin with fluorescein-bound silver nanoparticles is manifested by a decrease in the surface plasmon resonance band of silver nanoparticles at 395 nm, an increase in the fluorescence intensity of fluorescein at 513 nm and color change of the colloidal solution from yellow to light pink. These spectral effects are directly proportional to the concentration of tobramycin with a linearity range of 0.10 - 0.45 μg mL -1 and 0.05 - 0.45 μg mL -1 for the spectrophotometric and spectrofluorimetric methods, respectively. The proposed methods were applied for determination of tobramycin in Tobrin® ophthalmic solution with mean %recovery ± standard deviation of 99.036 ± 1.737 for the spectrophotometric method and 101.192 ± 1.315 for the spectrofluorimetric method. This review summarizes recent advances in leveraging localized surface plasmon resonance (LSPR) nanotechnology for sensitive cancer biomarker detection. LSPR arising from noble metal nanoparticles under light excitation enables the enhancement of various optical techniques, including surface-enhanced Raman spectroscopy (SERS), dark-field microscopy (DFM), photothermal imaging, and photoacoustic imaging. Nanoparticle engineering strategies are discussed to optimize LSPR for maximum signal amplification. SERS utilizes electromagnetic enhancement from plasmonic nanostructures to boost inherently weak Raman signals, enabling single-molecule sensitivity for detecting proteins, nucleic acids, and exosomes. DFM visualizes LSPR nanoparticles based on scattered light color, allowing for the ultrasensitive detection of cancer cells, microRNAs, and proteins. Photothermal imaging employs LSPR nanoparticles as contrast agents that convert light to heat, producing thermal images that highlight cancerous tissues. Photoacoustic imaging detects ultrasonic waves generated by LSPR nanoparticle photothermal expansion for deep-tissue imaging. The multiplexing capabilities of LSPR techniques and integration with microfluidics and point-of-care devices are reviewed. Remaining challenges, such as toxicity, standardization, and clinical sample analysis, are examined. Overall, LSPR nanotechnology shows tremendous potential for advancing cancer screening, diagnosis, and treatment monitoring through the integration of nanoparticle engineering, optical techniques, and microscale device platforms.

The development of label-free and sensitive detection of pathogenic bacteria is of great significance for disease prevention and public health protection. In this study, an originally bent structure, named as J-shaped optical fiber probe, was first designed to engineer a localized surface plasmon resonance (LSPR) aptamer biosensor for the rapid and ultrasensitive detection of Helicobacter pylori (H. pylori). The J-shaped optical fiber probe exhibited a significant improvement in refractive index sensitivity (RIS) and LSPR signal response. Meantime, the original sequence of aptamer was truncated in order to effectively capture H. pylori on the optical fiber surface. Besides, a spacer nucleic acid with short stem-loop structure was adopted to control the aptamer density on gold nanoparticles (AuNPs) on the surface of the J-shaped optical fiber probe, which displayed a further enhancement in LSPR signal response. Benefitting from these creative designs, the proposed LSPR biosensor can realize label-free and sensitive detection of H. pylori with a detection limit as low as 45 CFU/mL and a wide linear range from 1.0 × 10 2  CFU/mL to 1.0 × 10 8  CFU/mL. At the same time, the sensing strategy can detect the pathogenic bacteria from actual water samples in one step just in 30 min without any sample pretreatment. Alanine aminotransferase (ALT), a type of inactive enzyme largely present in fish liver cells, is essential for the tricarboxylic acid (TCA) cycle. Monitoring ALT activity in the blood/hepatocellular layer has been demonstrated to be a sensitive sign of liver dysfunction and an essential method for determining the health status of fish. This study details the development of a multi-layer material (hybrids of graphene oxide and multi-walled carbon nanotubes (GO/MWCNTs), gold nanoparticles (AuNPs), and glutamate oxidase (GluOx) enzyme) immobilized localized surface plasmon resonance based unique fiber structure biosensor for the quantitative determination of ALT biomolecules at concentrations ranging from 0 to 1000 U/L. For this kind of detection, a novel taper-in-taper with four tapered (TIT4T) structure based on single-mode fiber has been developed. In addition to AuNPs, GO/MWCNTs were immobilized in the probe's sensing region to increase its LSPR efficiency and sensitivity. Synthesis of AuNPs was carried out utilizing the Turkevich method. The selectivity of the sensor is ensured by the effective immobilization of GluOx on the surface treatment. The linearity of sensor is in the range of 0-1000 U/L, whereas the sensitivity, limit of detection, and detection time are individually found at 7.5 p.m./(U/L), 4.84 U/L and 20 min, respectively. After evaluating the prospective applications of the sensors, the sensors' reusability, reproducibility, stability, pH test, and selectivity have all been found to be satisfactory. Deoxyribonuclease-I (DNase-I), a representative endonuclease, is an important biomarker for the diagnosis of infectious diseases and cancer progression. However, enzymatic activity decreases rapidly ex vivo, which highlights the need for precise on-site detection of DNase-I. Here, a localized surface plasmon resonance (LSPR) biosensor that enables the simple and rapid detection of DNase-I is reported. Moreover, a novel technique named electrochemical deposition and mild thermal annealing (EDMIT) is applied to overcome signal variations. By taking advantage of the low adhesion of gold clusters on indium tin oxide substrates, both the uniformity and sphericity of gold nanoparticles are increased under mild thermal annealing conditions via coalescence and Ostwald ripening. This ultimately results in an approximately 15-fold decrease in LSPR signal variations. The linear range of the fabricated sensor is 20-1000 ng mL -1 with a limit of detection (LOD) of 127.25 pg mL -1 , as demonstrated by spectral absorbance analyses. The fabricated LSPR sensor stably measured DNase-I concentrations from samples collected from both an inflammatory bowel disease (IBD) mouse model, as well as human patients with severe COVID-19 symptoms. To achieve reliable and ultrasensitive detection for disease markers in PEC bioanalysis, constructing and nano-engineering of ideal photoelectrodes and signal transduction strategies are of vital importance. Herein, a non-/noble metal coupled plasmonic nanostructure (TiO 2 /r-STO/Au) was tactically designed with high-efficient PEC performance. Evidenced by the DFT and FDTD calculations, the reduced SrTiO 3 (r-STO) was found to support the localized surface plasmon resonance due to the sufficiently increased and delocalized local charge in r-STO. Under the synergistic coupling of plasmonic r-STO and AuNPs, the PEC performance of TiO 2 /r-STO/Au was found remarkably promoted with reduced onset potential. This merit supported TiO 2 /r-STO/Au as a self-powered immunoassay via a proposed oxygen-evolution-reaction mediated signal transduction strategy. With the increase of the target biomolecules (PSA), the catalytic active sites of TiO 2 /r-STO/Au would be blocked and result in the decrease of the oxygen evaluation reaction. Under optimal conditions, the immunoassays exhibited an excellent detection performance with a LOD as low as 1.1 fg/mL. Surface-Enhanced Raman Spectroscopy (SERS) is a powerful surface-sensitive technique for molecular analysis. Its use is limited due to high cost, non-flexible rigid substrates such as silicon, alumina or glass and less reproducibility due to non-uniform surface. Recently, paper-based SERS substrates, a low-cost and highly flexible alternative, received significant attention. We report here a rapid, inexpensive method for chitosan-reduced, in-situ synthesis of gold nanoparticles (GNPs) on paper devices towards direct utilization as SERS substrates. GNPs have been prepared by reducing chloroauric acid with chitosan as a reducing and capping reagent on the cellulose-based paper surface at 100 °C, under the saturated humidity condition (100 % humidity). GNPs thus obtained were uniformly distributed on the surface and had fairly uniform particle size with a diameter of 10 ± 2 nm. Substrate coverage of resulting GNPs directly depended on the precursor's ratio, temperature and reaction time. Techniques such as TEM, SEM, and FE-SEM were utilized to determine the shape, size, and distribution of GNPs on paper substrate. SERS substrate produced by this simple, rapid, reproducible and robust method of chitosan-reduced, in situ synthesis of GNPs, showed exceptional performance and long-term stability, with a detection limit of up to 1 pM concentration of test analyte, R6G. Circulating cell-free DNA (cfDNA) is a promising biomarker of liquid biopsy, but it still faces some difficulties in achieving sensitive and convenient detection. Herein, an Ω-shaped fiber optic localized surface plasmon resonance (FO-LSPR) biosensor based on hybridization chain reaction (HCR) coupled with gold nanoparticles (AuNPs) was developed, and applied in simple and sensitive detection of cfDNA. Specifically, one-base mismatch was designed in HCR hairpins (H1 and H2) to obtain high reaction efficiency, and AuNPs was introduced onto H1 through poly-adenine to construct HCR coupled with AuNPs strategy. Meanwhile, target cfDNA was designed into two domains: one could trigger HCR to generate dsDNA concatemer carrying numerous AuNPs, and the other could hybridize with capture DNA on the surface of Ω-shaped fiber optic (FO) probes. Thus, the presence of target cfDNA would initiate HCR, and bring the formed dsDNA concatemer and AuNPs to approach the probe surface, resulting in dramatically amplified LSPR signal. Besides, HCR required simple isothermal and enzyme-free condition, and Ω-shaped FO probe with high refractive index sensitivity just needed to be immersed into HCR solution directly for signal monitoring. Nanopillars (NPs) are submicron-sized pillars composed of dielectrics, semiconductors, or metals. They have been employed to develop advanced optical components such as solar cells, light-emitting diodes, and biophotonic devices. To integrate localized surface plasmon resonance (LSPR) with NPs, plasmonic NPs consisting of dielectric nanoscale pillars with metal capping have been developed and used for plasmonic optical sensing and imaging applications. In this study, we studied plasmonic NPs in terms of their fabrication techniques and applications in biophotonics. We briefly described three methods for fabricating NPs, namely etching, nanoimprinting, and growing NPs on a substrate. Furthermore, we explored the role of metal capping in plasmonic enhancement. Then, we presented the biophotonic applications of high-sensitivity LSPR sensors, enhanced Raman spectroscopy, and high-resolution plasmonic optical imaging. After exploring plasmonic NPs, we determined that they had sufficient potential for advanced biophotonic instruments and biomedical applications.

Photothermal therapy based on plasmonic gold nanoparticles is considered a promising approach for cancer treatment. Here, we investigate the in vitro photothermal effect of 30-nm gold nanoparticles, optically excited with a 532-nm continuous laser, on the U87MG malignant glioblastoma cells, and demonstrate the role of nanoparticle concentration and exposure power density in achieving its optimum performance. Laser-induced collective oscillation of electrons in plasmonic gold nanoparticles is employed to generate localized heat to denature tumor cells. Optical spectroscopy is used to measure the plasmonic band of nanoparticles and select the excitation laser light. The MTT assay for the IC50 viability assessment is performed to evaluate the live and metabolically active cells after treatment with plasmonic nanoparticles. The quantitative data is statistically analyzed using analysis of variance followed by Tukey's post hoc test. The viability test demonstrates that the metabolic activity of treated U87MG was decreased compared with untreated cells, leading to the determination of the IC50 as 92 [Formula: see text]. A 532-nm laser light was selected for the excitation of gold nanoparticles since the maximum plasmonic band is at 524 nm. The viability tests show that although cells have natural photothermal agents, their absorption efficiency is very low; therefore, the laser-induced plasmonic effect is necessary to observe photothermal effects. The optimal condition was achieved when the nanoparticle concentration was 92 [Formula: see text], and the exposure power density was 96 [Formula: see text]. Thus, it is demonstrated that plasmonic nanoparticle concentration and laser exposure power density are among the key parameters in the photothermal treatment of cancer cells, and higher concentrations of Au-NPs and laser power density lead to less cell viability in the selected range due to the excitation of gold nanoparticles' localized surface plasmon resonance., (© 2023. Rapid, sensitive, and reliable detection of high mobility group box 1 (HMGB1) is essential for medical and diagnostic applications due to its important role as a biomarker of chronic inflammation. Here, we report a facile method for the detection of HMGB1 using carboxymethyl dextran (CM-dextran) as a bridge molecule modified on the surface of gold nanoparticles combined with a fiber optic localized surface plasmon resonance (FOLSPR) biosensor. Under optimal conditions, the results showed that the FOLSPR sensor detected HMGB1 with a wide linear range (10 -10 to 10 -6 g/mL), fast response (less than 10 min), and a low detection limit of 43.4 pg/mL (1.7 pM) and high correlation coefficient values (>0.9928). Furthermore, the accurate quantification and reliable validation of kinetic binding events measured by the currently working biosensors are comparable to surface plasmon resonance sensing systems, providing new insights into direct biomarker detection for clinical applications.

In this study, we developed a biosensor based on the localized surface plasmon resonance (LSPR) phenomenon of gold nanoparticles (AuNPs) to detect the widely used herbicide glyphosate in food samples. To do so, either cysteamine or a specific antibody for glyphosate were conjugated to the surface of the nanoparticles. AuNPs were synthesized using the sodium citrate reduction method and had their concentration determined via inductively plasma coupled mass spectrometry. Their optical properties were analyzed using UV-vis spectroscopy, X-ray diffraction, and transmission electron microscopy. Functionalized AuNPs were further characterized via Fourier-transform infrared spectroscopy, Raman scattering, Zeta potential, and dynamic light scattering. Both conjugates succeeded in detecting the presence of glyphosate in the colloid, although nanoparticles functionalized with cysteamine tended to aggregate at high concentrations of the herbicide. On the other hand, AuNPs functionalized with anti-glyphosate functioned at a broad concentration range and successfully identified the presence of the herbicide in non-organic coffee samples and when it was added to an organic coffee sample. This study demonstrates the potential of AuNP-based biosensors to detect glyphosate in food samples. The low-cost and specificity of these biosensors make them a viable alternative to current methods for detecting glyphosate in foodstuffs.

Silver nanoparticles (AgNPs) have long been overshadowed by gold NPs' success in sensor and point-of-care (POC) applications. However, their unique physical, (electro)chemical, and optical properties make them excellently suited for such use, as long as their inherent higher instability toward oxidation is controlled. Recent advances in this field provide novel strategies that demonstrate that the AgNPs' inherent capabilities improve sensor performance and enable the specific detection of analytes at low concentrations. We provide an overview of these advances by focusing on the nanosized Ag (in the range of 1-100 nm) properties with emphasis on optical and electrochemical biosensors. Furthermore, we critically assess their potential for point-of-care sensors discussing advantages as well as limitations for each detection technique. We can conclude that, indeed, strategies using AgNP are ready for sensitive POC applications; however, research focusing on the simplification of assay procedures is direly needed for AgNPs to make the successful jump into actual applications., (© 2023. Wearable biosensors have the potential for developing individualized health evaluation and detection systems owing to their ability to provide continuous real-time physiological data. Among various wearable biosensors, localized surface plasmon resonance (LSPR)-based wearable sensors can be versatile in various practical applications owing to their sensitive interactions with specific analytes. Understanding and analyzing endocrine responses to stress is particularly crucial for evaluating human performance, diagnosing stress-related diseases, and monitoring mental health, as stress takes a serious toll on physiological health and psychological well-being. Cortisol is an essential biomarker of stress because of the close relationship between cortisol concentration in the human body and stress level. In this study, a flexible LSPR biosensor was manufactured to detect cortisol levels in the human body by depositing gold nanoparticle (AuNP) layers on a 3-aminopropyltriethoxysilane (APTES)-functionalized poly (dimethylsiloxane) (PDMS) substrate. Subsequently, an aptamer was immobilized on the surface of the LSPR substrate, enabling highly sensitive and selective cortisol capture owing to its specific cortisol recognition. The biosensor exhibited excellent detection ability in cortisol solutions of various concentrations ranging from 0.1 to 1000 nM with a detection limit of 0.1 nM. The flexible LSPR biosensor also demonstrated good stability under various mechanical deformations. Furthermore, the cortisol levels of the flexible LSPR biosensor were also measured in the human epidermis before and after exercise as well as in the morning and afternoon. Our biosensors, which combine easily manufactured flexible sensors with sensitive cortisol-detecting molecules to measure human stress levels, could be versatile candidates for human-friendly products.

Human epidermal growth factor receptor 2 (HER2) has been regarded as the considerable biomarker of breast and gastric cancer. Thus, precise detection of HER2 is of significance for the early diagnosis and treatment. Here, a photofuel cell-based self-powered biosensor (PFC-SPB) was constructed for the ultrasensitive HER2 detection, which was composed of a plasmonic gold nanoparticles (Au NPs)/organic semiconductor hybrid photoanode and a cathode with biosensing strategy of electrochemical sandwich structure. The localized surface plasmon resonance effect of Au NPs can obviously enhance the separation efficiency of photo-generated electron/hole pair, which was beneficial to the sensitivity and stability of PFC-SPB. Meanwhile, the cathodic sandwich structure not only was used for the target recognition, but also can guarantee the enrichment of electroactive molecules (molybdophosphate). Consequently, with the open circuit voltage (E OCV ) as the output signal, the PFC-SPB can achieve the HER2 detection in the range of 0.1-500 pg mL -1 with a low detection limit of 0.02 pg mL -1 . Low-cost, instrument-free colorimetric tests were developed to detect SARS-CoV-2 using plasmonic biosensors with Au nanoparticles functionalized with polyclonal antibodies (f-AuNPs). Intense color changes were noted with the naked eye owing to plasmon coupling when f-AuNPs form clusters on the virus, with high sensitivity and a detection limit of 0.28 PFU mL -1 (PFU stands for plaque-forming units) in human saliva. Plasmon coupling was corroborated with computer simulations using the finite-difference time-domain (FDTD) method. The strategies based on preparing plasmonic biosensors with f-AuNPs are robust to permit SARS-CoV-2 detection via dynamic light scattering and UV-vis spectroscopy without interference from other viruses, such as influenza and dengue viruses. The diagnosis was made with a smartphone app after processing the images collected from the smartphone camera, measuring the concentration of SARS-CoV-2. Both image processing and machine learning algorithms were found to provide COVID-19 diagnosis with 100% accuracy for saliva samples. In subsidiary experiments, we observed that the biosensor could be used to detect the virus in river waters without pretreatment. With fast responses and requiring small sample amounts (only 20 μL), these colorimetric tests can be deployed in any location within the point-of-care diagnosis paradigm for epidemiological control.

Heavy metals are important sources of environmental pollution and cause disease in organisms throughout the food chain. A localized surface plasmon resonance sensor was proposed and demonstrated to realize Ni 2+ detection by using ion-imprinted chitosan. Au nanoparticles were coated on the multimode fiber to excite the local surface plasmon resonance, and Ni 2+ -imprinted chitosan was then functionalized by using the dip coating technique. Ethylene diamine tetra-acetic acid was used to release the Ni 2+ ions and hence form countless voids. Ni 2+ was refilled into the voids to increase the refractive index of the sensing material, thus realizing the measurement of Ni 2+ by monitoring the wavelength shift in the localized surface plasmon resonant peak. The coating thickness of the Ni 2+ -chitosan gel was optimized to obtain greater sensitivity. Experimental results show that the proposed Ni 2+ sensor has a sensitivity of 185 pm/μM, and the limit of detection is 0.512 μM. The comparison experiments indicated that the ion-imprinted chitosan has better selectivity than pure chitosan.

A new innovative approach is essential for early and effective diagnosis of gastric cancer, using promoter hypermethylation of the tumor suppressor, SOCS-1, that is frequently inactivated in human cancers. We have developed an amplification-free fiber optic nanoplasmonic biosensor for detecting DNA methylation of the SOCS-1 human genome. The method is based on the fiber optic nanogold-linked sorbent assay of PCR-free DNA from human gastric tumor tissue and cell lines. We designed a specific DNA probe fabricated on the fiber core surface while the other probe is bioconjugated with gold nanoparticles in free form to allow percentage determination and differentiating the methylated and unmethylated cell lines, further demonstrating the SOCS-1 methylation occurs in cancer patients but not in normal cell lines. The observed detection limit is 0.81 fM for methylated DNA, and the detection time is within 15 min. In addition, our data were significantly correlated to the data obtained from PCR-based pyrosequencing, and yet with superior accuracy. The 1064 nm Nd:YAG laser shows a good prospect for the treatment of port-wine stain (PWS), but it is necessary to enhance the blood absorption to laser energy by exogenous chromophore. Owing to the conjunction effect of local surface plasmon resonance (LSPR) by gold nanoparticle and drug delivery as well as lumen blockage abilities by liposome, liposome@Au core-shell nanoparticles are used as exogenous chromophore, and the efficiency of photothermal therapy is studied systematically. In this work, theoretical simulations were conducted to investigate the electric field and solid heat conduction of liposome@Au core-shell nanoparticles with various size and particles distance, aiming to achieve maximum photothermal conversion efficiency during the laser irradiation. Thereafter, liposome@Au core-shell nanoparticles with optimal size and structure were prepared, and in vivo experiments were conducted to evaluate the thermal damage of blood vessels enhanced by liposome@Au core-shell nanoparticles. Theoretical results imply that maximum temperature rise (167 K) is obtained when radius is 45 nm and shell thickness is 5 nm with distance of 4 nm. Liposome@Au core-shell nanoparticles were prepared with diameter of 101 nm and shell thickness of 5 nm according to the finite element simulation of electric field and solid heat conduction. When the molar ratio of chloroauric acid to phospholipid is 2.25, the LSPR absorption peak is about 981 nm, which is close to the wavelength of Nd:YAG laser. In vivo experiments show that injecting liposome@Au core-shell nanoparticles into the blood vessels can effectively reduce the number of laser pulses and the corresponding energy density required for obvious vasoconstriction., (© 2022. The N-terminal pro-brain natriuretic peptide (NT-proBNP) is considered an important blood biomarker for heart failure. Herein, we report about a fiber optic nanogold-linked immunosorbent assay (FONLISA) method for the rapid, sensitive, and low-cost detection of NT-proBNP. The method is based on a sandwich immunoassay approach that uses two monoclonal NT-proBNP antibodies, a capture antibody (Ab C ), and a detection antibody (Ab D ). Ab D is conjugated to a free gold nanoparticle (AuNP) to form the free AuNP@Ab D conjugate, and Ab C is immobilized on an unclad segment of an optical fiber. The detection of analyte (A), in this case NT-proBNP, is based on the signal change due to the formation of an AuNP@Ab D -A-Ab C complex on the fiber core surface, where a green light transmitted through the optical fiber will decrease in intensity due to light absorption by AuNPs via the localized surface plasmon resonance effect. This method provides a wide linear dynamic range of 0.50~5000 pg·mL -1 and a limit of detection of 0.058 pg·mL -1 for NT-proBNP. Finally, the method exhibits good correlation ( r = 0.979) with the commercial central laboratory-based electrochemiluminescent immunoassay method that uses a Roche Cobas e411 instrument. Hence, our method is potentially a suitable tool for point-of-care testing.

Conductive polymers are promising electrode candidates in the nonenzymatic catalytic detection of small molecule metabolites, due to the tunable electronic conductivity and versatile modifiability. However, the complex catalytic reaction pathway of conductive polymers results in lower detection sensitivity and a narrower linear range compared with clinical metal-based and carbon-based electrodes. Localized surface plasmon resonance (LSPR), characterized by deep strong light-matter coupling, has great potential in driving surface catalytic reactions at an ultrafast rate. Here, we constructed a salix argyracea-like polypyrrole nanowires/silver nanoparticles (PPy/AgNPs) heterojunction electrode using polydopamine as a dopant and chelator. Through cyclic voltammetry, the Mott-Schottky curve, and COMSOL simulation, we demonstrated that the LSPR-excited photocarriers enhanced PPy/AgNPs electrode electrocatalysis. Thus, the detection current response and linear range were significantly improved under the LSPR excitation when taking glucose and hydrogen peroxide as models of small molecule metabolites. Furthermore, we discussed the LSPR-enhanced detection mechanism of PPy/AgNPs electrode from the aspects of the Tafel slope, the apparent electron diffusion coefficient, and the charge transfer resistance. This strategy opens a new avenue toward the design of LSPR-enhanced conductive polymer electrodes.

The etching of gold nanorods/nanobipyramid, or silver-coated nanorods/nanobipyramid inducing plasmon changes represents an efficient strategy to improve the performance of enzyme-linked immunosorbent assay (ELISA). However, the effect of shape on the sensitivity was negligible, especially the thickness of coated silver shell. Here, we propose a plasmonic ELISA for multi-colorimetric detection of CRP based on the etching of Ag-coated Au nanobipyramid (Au NBP@Ag). The effect of silver shell thickness on the sensitivity of plasmon peak shifting was investigated by experiments and DDA calculations. The relationship between the Ag shell thickness and the sensitivity of plasmon peak shifting was obtained. Our results reveal that the thickness of coated Ag shell acts as a key factor in the multi-color change of Au NBP@Ag etching. It is found that Au NBP@Ag with medium Ag shell thickness and rod-like shape has the higher sensitivity and is suitable for sensing. At the optimized most sensitive Ag shell, the detection limit of proposed plasmonic ELISA for CRP was determined to be 0.09 ng/mL with a spectrometer in the range from 0.09 ng/mL to 25 ng/mL. Importantly, the visual detection limit was 0.78 ng/mL, which allows the differential diagnosis with the naked eye. Milk is one of the most important multicomponent superfoods owing to its rich macronutrient composition. It requires quality control at all the production stages from the farm to the finished products. A localized surface plasmon resonance optical sensor based on a citrate-capped silver nanoparticle (Cit-AgNP)-coated glass substrate was developed. The fabrication of such sensors involved a single-step synthesis of Cit-AgNPs followed by surface modification of glass slides to be coated with the nanoparticles. The scanning electron microscope micrographs demonstrated that the nanoparticles formed monolayer islands on glass slides. The developed surface-enhanced infrared absorption spectroscopy (SEIRA) sensor was coupled to artificial neural networking (ANN) for the qualitative differentiation between cow, camel, goat, buffalo, and infants' formula powdered milk types. Moreover, it can be used for the quantitative determination of the main milk components such as fat, casein, urea, and lactose in each milk type. The qualitative results showed that the obtained FTIR spectra of cow and buffalo milk have high similarity, whereas camel milk resembled infant formula powdered milk. The most difference in FTIR characteristics was evidenced in the case of goat milk. The developed sensor adds several advantages over the traditional techniques of milk analysis using MilkoScan™ such as less generated waste, elimination of pre-treatment steps, minimal sample volume, low operation time, and on-site analysis., (© 2022. For real-time and high-sensitivity analysis of low-concentration targets, a sandwich immunoassay using second antibody-second gold nanoparticle (2nd Ab-2nd AuNP) conjugates was combined with fiber-optic localized surface plasmon resonance (FO LSPR). An FO LSPR format was constructed by immobilizing AuNPs on a fiber-optic cross-section for compactness, portability, and ease of handling. In addition, it was combined with a microfluidic system to ensure reproducibility and reliability of measurements. A detection limit of 97.6 fg/mL (148 aM) was obtained for thyroglobulin (Tg) without a sandwich assay. The detection limit was enhanced by approximately 15 times (6.6 fg/mL, 10 aM) when a sandwich strategy was performed with a 2nd Ab-2nd AuNP signal amplifier to further improve the responsivity. Additionally, the good selectivity of the proposed method was confirmed against the unpaired antigen. To evaluate its practical applicability in the field, an FO LSPR biosensor boosted with a sandwich assay using antibody-functionalized AuNPs was applied to detect Tg contained in patient serum, and the results were compared and verified with those of a commercial radioimmunoassay kit. Development of rapid, sensitive, and selective method for pathogenic bacteria detection is of great importance for food safety, medical diagnostic, and environmental monitoring. Currently, most techniques for low numbers of bacteria detection require advanced instrumentation or skilled operators. Herein, we present a facile colorimetric detection platform for bacterial detection using Ag nanoplates as chromogenic substrate, which takes advantages of the high specificity and affinity of aptamer and the ability of catalase to hydrolyze H 2 O 2 that can etch Ag nanoplates. By introducing catalase to the sandwich structure composed by dual-aptamer recognition strategy, bacteria detection signal is converted to the peak shift of LSPR and colorimetric change. This proposed method allows a fast naked-eye detection of S. aureus at the concentration of 60 CFU/mL based on the combination of streptavidin-biotin system and inherent sensitivity of plasmonic Ag nanoplates. Herein, a facile and sensitive dual mode sensing strategy for amantadine (AMD) level evaluation by coupling the plasmonic Au nanorod (NR) and supramolecular SH-cyclodextrin (CD) through strong Au-S bond is proposed. Methylene blue (MB) molecules can be inserted into the cavity of CD molecules through the hydrophobic interaction, which would cause the plasmon resonance energy transfer (PRET) process as well as electrochemical signal response due to the spectrum overlap between Au NR and MB molecules and the electrochemical conversion activity of MB molecules. Subsequently, AMD would induce the replacement of MB molecules because of the stronger interaction with CD, resulting the recovery of scattering intensity of Au NR and decrease of the electrooxidation current of MB. On one hand, the increase of Au NR scattering intensity is linearly proportional to AMD with the concentration from 0.4 to 3.0 μM, and reaches a limit of detection (LOD) of 0.28 μM. On the other hand, electrochemical measurement method enlarged the detection range of AMD. The variation of electrochemical oxidation peak current of MB is linearly proportional to the logarithm value of AMD concentration from 2.5 to 375.0 μM, with LOD of 1.9 μM. Subsequently, the proposed dual mode response sensing strategy was successfully employed for the detection of AMD in human serum samples with great selectivity and sensitivity, with a recovery percentage ranged from 92.6 to 112.0%. In the current work, we have reported a fast and easy method based on gold nanoparticles for the determination of uric acid in urine samples. In the first stage, gold nanoparticles were synthesized using the chemical reduction method and then applied as a sensor to measure uric acid concentration based on its strong reducing property. The main parameters affecting response signals such as pH, reagent concentration, and time are optimized using the multivariate method. The development of rapid and sensitive detection methods for pathogenic bacteria is crucial for the therapy and prevention of related diseases. However, the rapid and ultrasensitive assays are difficult to be realized simultaneously. To solve the problem, a sandwich method based on Ω-shaped fiber optic localized surface resonance (Ω-FOLSPR) was constructed, where poly adenine-tailed aptamer (PolyA-apt) and SH modified gold nanoparticles tags (AuNPs tags) were chosen as the capturing aptamer and amplifying tags, respectively. The small AuNPs were modified on the surface of fiber-optic (FO) rapidly, which saved the preparation time. Then, the PolyA-apt was modified on the AuNPs surface to capture the bacteria effectively due to its ability to adjust the density and conformation of aptamer on the AuNPs surface. Finally, the large AuNPs tags were used to generate intense signal enhancement. It is found that the sandwich method enables the unique characteristic of a time-dependent sensitivity enhancement. Specifically, the LOD of 108.0 CFU/mL and 7.4 CFU/mL was achieved with the analysis time of 10 min and 100 min, respectively. Besides, the Ω-FOLSPR sensor exhibits excellent selectivity against the other bacteria and good performance for detecting the spiked and natural samples. MicroRNAs involved in tumor-related tissues at abnormal expression level present tremendous potential in the early diagnosis of cancers. However, their intrinsic shortcomings, for instance, low abundance and high sequence homology, make it challengeable to quantify them with high sensitivity and selectivity. Herein, a highly sensitive platform with great specificity was developed for microRNA-21 based on the produced-I 2 triggered chemical etching of gold nanorods to a smaller size, resulting in a significant blue shift and a great intensity decrease in the localized surface plasmon resonance (LSPR) scattering. The synergism of strand displacement and enzymatic reaction enabled the proposed strategy with a high sensitivity and selectivity toward microRNA-21 in a dynamic range from 0.1 to 10,000 pM and a low limit of detection of 71.22 fM (3σ/k) by dark-field microscope. Additionally, the remarkable discrimination of single nucleotide difference suggested the superior selectivity towards microRNA-21, which presented a satisfactory recovery in human serum samples. Although there is considerable interest in self-assembly of ordered, porous "inverse opal" structures for optical, electronic, and chemical applications, uncontrolled defect formation limits the usefulness of such materials. Herein, we develop a highly ordered and plasmonic enhanced sensing inverse opal photonic crystal (IOPC) material. The co-assembly of the colloidal template with the matrix material avoids the need for liquid penetration into the preassembled colloidal crystals and minimizes the associated rupture and inhomogeneity of the resulting IOPC. Au nanoparticles (Au NPs) not only act as a "bridge" between recognition elements (aptamers) and IOPCs, but also can amplify optical signals. Furthermore, the enhancement mechanism of Au NPs is simulated by COMSOL. During the detection process, the optical signal of the sensing Au-Apt IOPC responds to the Staphylococcal enterotoxin B with a concentration ranging from 10 -2 to 10 3 pg mL -1 , and the limit of detection is 2.820 fg mL -1 . Spiked real sample detection indicates that the as-proposed method possessed good accuracy. The sensing Au-Apt IOPC provides an extensive biosensor platform to detect a variety of toxic and harmful substances through replacing the aptamer by other recognition elements, such as antibodies or receptors.

Inadequate sensitivity and side-effect are the main challenges to develop cytosensors combining with therapeutic potential simultaneously for cancer diagnosis and treatment. Herein, localized surface plasma resonance (LSPR) based on hybridized nanolayer modified Ω-shaped fiber-optic (HN/Ω-FO) was developed to integrate cytosensor and plasmonic photothermal treatment (PPT). On one hand, hybridized nanolayers improve the coverage of nanoparticles and refractive index sensitivity (RIS). Moreover, the hybridized nanoploymers of gold nanorods/gold nanoparticles (AuNRs/AuNPs) also result in intense enhancement in electronic field intensity (I). On the other hand, Ω-shaped fiber-optic (Ω-FO) led to strong bending loss in its bending part. To be specific, a majority of light escaped from fiber will interact with HN. Thus, HN/Ω-FO synergistically enhances the plasmonic, which achieved the goal of ultrasensitive cytosensor and highly-efficient plasmonic photothermal treatment (PPT). The proposed cytosensor exhibits ultrasensitivity for detection of cancer cells with a low limit of detection down to 2.6 cells/mL was realized just in 30 min. HN/Ω-FO-based LSPR exhibits unique characteristics of highly efficient, localized, and geometry-dependent heat distribution, which makes it suitable for PPT to only kill the cancer cells specifically on the surface or surrounding fiber-optic (FO) surface. We here report on a facile method for preparation of gold nanostar-graphene quantum dot (AuNS@GQD) composite, which produces highly active surfactant-free AuNSs. The etching reaction of this composite with Na 2 SO 3 was studied and used as a new sensing strategy for colorimetric detection of nM levels of cysteine. In the presence of Na 2 SO 3 , the shape of AuNSs changes to sphere-like nanoparticles, leading to a distinct color change of solution from light green to indigo. This phenomenon results from the redox reaction of Au atoms at the apexes and sharp corners of the NSs with oxygen which leads to the formation of [Au(SO 3 ) 2 ] 3- . Our studies indicated that the stars with larger sizes show greater activity in etching reaction since they have more branches and sharper tips. Due to the strong coordination between Au and thiols, pre-added cysteine can protect the AuNSs from SO 3 2- etching and so the shape and the color of AuNSs remain unaltered. This anti-etching effect was used for the detection of cysteine with the detection limit as low as 0.35 nM. Fiber optic localized surface plasmon resonance (FO LSPR) sensors capable of portable, real-time, and remote sensing are emerging with the progress of lab-on-fiber technology. However, the small area of the substrate by the optical fiber often restricts the sensitivity of the FO LSPR sensors. To improve the performance of the FO LSPR sensors, it is necessary to enhance the interactions between incident light and plasmonic nanostructures within a defined region. Dimer in which two nanoparticles are arranged with nanometer spacing can effectively increase the light-nanostructure interactions. It is well known that the nanogap made in the assembled nanoparticles significantly enhances the intensity of the electromagnetic field in the confined area by the hot spot effect. We fabricate the dimers of gold nanoparticles on the optical fiber with benzenethiol using a method that reduces the repulsive force between the nanoparticles. In the dimers, the strong plasmonic interaction between the two nanoparticles produces a longitudinal plasmon coupling band, which is compared to the transverse plasmon band by the monomer-based FO LSPR sensor with a similar density of gold nanoparticles. In the proposed sensor, the longitudinal band displays approximately 9.1 times improved sensitivity. When two types of sensors are applied to the biosensor application, the dimer-based FO LSPR sensor also proves an improved limit of detection of about 2.6 times. Biosensors need to meet the rising food industry demand for sensitive, selective, safe, and fast food safety quality control. Disposable colorimetric sensors based on gold nanoparticles (AuNPs) and localized surface plasmon resonance are low-cost and easy-to-perform devices intended for rapid point-of-need measurements. Recent studies demonstrate various facile and versatile AuNPs-based analytical platforms for the detection of bacteria and their toxins in milk, meat, and other foods. The electron density of a nanoparticle is a very important characteristic of the properties of a material. This paper describes the formation of silver nanoparticles (NPs) and the variation in the electronic state of an NP's surface upon the reduction in Ag + ions with oxalate ions, induced by UV irradiation. The calculations were based on optical spectrophotometry data. The NPs were characterized using Transmission electron microscopy and Dynamic light scattering. As ~10 nm nanoparticles are formed, the localized surface plasmon resonance (LSPR) band increases in intensity, decreases in width, and shifts to the UV region from 402 to 383 nm. The interband transitions (IBT) band (≤250 nm) increases in intensity, with the band shape and position remaining unchanged. The change in the shape and position of the LSPR band of silver nanoparticles in the course of their formation is attributable to an increasing concentration of free electrons in the particles as a result of a reduction in Ag + ions on the surface and electron injection by CO2- radicals. The ζ-potential of colloids increases with an increase in electron density in silver nuclei. A quantitative relationship between this shift and electron density on the surface was derived on the basis of the Mie-Drude theory. The observed blue shift (19 nm) corresponds to an approximately 10% increase in the concentration of electrons in silver nanoparticles.

Alkaline phosphatase (ALP) is an enzyme that catalyzes the dephosphorylation of proteins, nucleic acids, and biomolecules. It is a potential biomarker for diverse diseases such as breast cancer, osteopenia, and hepatobiliary. Herein, we developed a colorimetric sensor for the ALP assay based on its enzymatic activity to dephosphorylate the p-aminophenol phosphate (pAPP) into pAP. In a solution containing silver nanoparticles (AgNPs) and Ag + ions prepared using a low concentration of NaBH 4 , pAP mediates the growth of AgNPs by reducing the concentration of Ag + ions to enhance the intensity of localized surface plasmon resonance as the pAPP cannot induce a reduction of the remaining Ag + due to the masking of the hydroxyl with phosphate. The quantitative assay of the ALP was demonstrated via the colorimetric detection of the pAP-mediated growth of AgNPs in the presence of an ALP. The highly sensitive enzymatic growth of AgNPs provided a wider dynamic linear range of 0.5-225 U/L with a lower limit of detection of 0.24 U/L than that previously reported. The use of pAP resulted in excellent selectivity of the sensor for the ALP assay in human serum, yielding a high recovery rate and a high precision of 99.2 ± 1.5 % for the standard addition method., (Copyright © 2021 Elsevier B.V. The regulation of tolerable levels of ochratoxin A in food for human and animal consumption has been defined in some countries. To meet these levels, simpler, more efficient, and faster analytical methods are being developed to facilitate the identification of this dangerous contaminant in food. Here, we combined gold nanoparticles (AuNPs) with anti-ochratoxin A (OTA) IgG to detect elementary levels of OTA based on Localized Surface Plasmon Resonance. AuNPs were prepared with trisodium citrate and characterized by UV-visible spectroscopy, X-ray, dynamic light scattering, and transmission electron microscopy. The conjugation of AuNPs to IgG anti-OTA was confirmed by bathochromic shift (UV-vis) and RAMAN spectroscopy. The sensitivity of the nanosensor was investigated by measuring LSPR band λ max shifts. Our results suggest this assay is highly sensitive, with a lower detection limit of about 0.001 pg mL -1 . The LSPR nanosensor reduced detection limits by roughly 10 times compared to other methods. The social impact of virus spread is immeasurable. Vaccine prophylaxes take considerable time to develop because clinical trials are required. The best initial response to an emerging virus is establishing a virus detection technology adapted by simply preparing virus-specific antibodies. A virus detection system that detects two signals from one analyte has been developed to detect the target virus more sensitively and reliably. Plasmon regions on the surface of nanoparticles are effective in enhancing optical and electrochemical signals. Thus, CdSeTeS quantum dots (QDs) have been used as optical and electrochemical signal-generating materials. In contrast, gold nanoparticle-magnetic nanoparticle-carbon nanotube (AuNP-MNP-CNT) nanocomposites are used for the magnetic separation of the virus from interferences and for signal enhancement. In the presence of the target virus, the QDs optically show a virus concentration-dependent fluorescence enhancement effect due to the localized surface plasmon resonance (LSPR) of AuNPs. Regarding the electrochemical signal, Cd ions eluted by acid degradation of the QDs in solution show a virus concentration-dependent increase in the current peak on an electrode whose electrochemical properties are improved by the deposition of these nanocomposites. Both nanomaterials are conjugated with antibodies specific to influenza virus A (IFV/A), binding this target in a sandwich structure. We are successfully detecting the virus from these two signals during actual virus detection, even when the virus particles are in a human serum matrix. The limit of detection is 2.16 fg/mL for optical detection and 13.66 fg/mL for electrochemical detection.

Colorimetric sensors based on localized surface plasmon resonance (LSPR) have attracted much attention for biosensor and chemical sensor applications. The unique optical effect of LSPR is based on the nanostructure of noble metals (e.g., Au, Ag, and Al) and the refractive index of the environment surrounding these metal nanomaterials. When either the structure or the environment of these nanomaterials is changed, their optical properties change and can be observed by spectroscopic techniques or the naked eye. Colorimetric-probe-based LSPR provides a simple, rapid, real-time, nonlabelled, sensitive biochemical detection and can be used for point-of-care testing as well as rapid screening for the diagnosis of various diseases. Gold and silver nanoparticles, which are the two most widely used plasmonic nanomaterials, demonstrate strong and sensitive LSPR signals that can be used for the selective detection of several chemicals in biochemical compounds provided by the human body (e.g., urine and blood). This information can be used for the diagnosis of several human health conditions. This paper provides information regarding colorimetric probes based on LSPR for the detection of three major chemicals in human urine: creatinine, albumin, and glucose. In addition, the mechanisms of selective detection and quantitative analysis of these chemicals using metal nanoparticles are discussed along with colorimetric-detection-based LSPR for many other specific chemicals that can be detected in urine, such as catecholamine neurotransmitters, thymine, and various medicines. Furthermore, issues regarding the use of portable platforms for health monitoring with colorimetric detection based on LSPR are discussed.

A potential cancer antigen (Ag), protein-phosphatase-1-gamma-2 (PP1γ2), with a restricted expression in testis and sperms has been identified as a biomarker specific to cervical cancer (CaCx). Detection of this cancer biomarker antigen (NCB-Ag) in human urine opens up the possibility of noninvasive detection of CaCx to supplement the dreaded and invasive Pap-smear test. A colorimetric response of an assembly of gold nanoparticles (Au NPs) has been employed for the quantitative, noninvasive, and point-of-care-testing of CaCx in the urine. In order to fabricate the immunosensor, Au NPs of sizes ∼5-20 nm have been chemically modified with a linker, 3,3'-di-thio-di-propionic-acid-di( n -hydroxy-succinimide-ester) (DTSP) to attach the antibody (Ab) specific to the NCB-Ag. Interestingly, the addition of Ag to the composite of Ab-DTSP-Au NPs leads to a significant hypsochromic shift due to a localized surface plasmon resonance phenomenon, which originates from the specific epitope-paratope interaction between the NCB-Ag and Ab-DTSP-Au NPs. The variations in the absorbance and wavelength shift during such attachments of different concentrations of NCB-Ag on the Ab-DTSP-Au NPs composite have been employed as a calibration to identify NCB-Ag in human urine. An in-house prototype has been assembled by integrating a light-emitting diode of a narrow range wavelength in one side of a cuvette in which the reaction has been performed while a sensitive photodetector to the other side to transduce the transmitted signal associated with the loading of NCB-Ag in the Ab-DTSP-Au NPs composite. The proposed immunosensing platform has been tested against other standard proteins to ensure noninterference alongside proving the proof-for-specificity of the NCB detection.

The combination of localized surface plasmon resonance (LSPR) and electrochemiluminescence (ECL) can be an effective way to amplify the signal intensity. In this work, an ECL aptasensor with 3,4,9,10-perylenetetracarboxylic acid-decorated cobalt phosphate (denoted as PTCA/CoP) as the ECL emitter and Au nanoparticles (NPs) as plasma was proposed for diclofenac assay. The prepared PTCA/CoP with special 1D/2D structure exhibited good ability and excellent ECL performance. The diclofenac aptamer acted as a bridge to link the PTCA/CoP and Au NPs; thus, the ECL performance of PTCA/CoP was greatly improved due to the plasma effect of Au NPs. Besides, it was found that the ECL signal of the aptasensor was obviously quenched by the introduction of diclofenac, which might be due to the transformation from the LSPR process to the resonance energy transform (RET) process. Under optimal conditions, the difference of ECL intensity was negatively correlated with the concentration of diclofenac in the range 0.1 pM to 10 μM with a low detection limit of 0.072 pM at the potential of -1.8 V vs. Ag/AgCl (S/N = 3). The aptasensor was proved to be suitable for the detection of diclofenac in real samples, suggesting its great practicability.

The fast measurement of fibrinogen is essential in evaluating life-threatening sepsis and cardiovascular diseases. Here, we aim to utilize biomimetic plasmonic Au nanoparticles using red blood cell membranes (RBCM-AuNPs) and demonstrate nanoscale coagulation-inspired fibrinogen detection via cross-linking between RBCM-AuNPs. The proposed biomimetic RBCM-AuNPs are highly suitable for fibrinogen detection because hemagglutination, occurring in the presence of fibrinogen, induces a shift in the localized surface plasmon resonance of the NPs. Specifically, when the two ends of the fibrinogen protein are bound to receptors on separate RBCM-AuNPs, cross-linking of the RBCM-AuNPs occurs, yielding a corresponding plasmon shift within 10 min. This coagulation-inspired fibrinogen detection method, with a low sample volume, high selectivity, and high speed, could facilitate the diagnosis of sepsis and cardiovascular diseases.

Demand for the detection of estradiol, which is a naturally occurring hormone, has been increasing. Gold nanoparticle-based colorimetric aptasensors have been developed for estradiol detection; however, the long sequence of aptamers due to the formation of the secondary structure likely affects the sensitivity of the aptasensors. Herein, a sensitive colorimetric biosensor is developed for label-free detection of estradiol by using an estradiol-specific split aptamer. The results demonstrate that a superior response is observed when a split aptamer with a high free energy of the secondary structure (ΔG > -3 kcal/mol) is used, in comparison to that observed using a split aptamer with a low free energy of the secondary structure (ΔG < -3 kcal/mol) at 27°C. After selecting the appropriate split aptamer, the standard calibration curve obtained for estradiol has a detection limit of 6.7 nM, with a linear range of 6.7 nM - 66.7 μM in the logarithmic scale. Furthermore, this assay is sensitive, easy-to-operate, inexpensive, and non-time-consuming (provides results within 50 min), thereby showing potential for clinical applications (detection of other small molecular targets).
