The pathogenesis of lung fibrosis involves hyperactivation of innate and adaptive immune pathways that release inflammatory cytokines and growth factors such as tumor growth factor (TGF)β1 and induce aberrant extracellular matrix protein production. During the genesis of pulmonary fibrosis, resident alveolar macrophages are replaced by a population of newly arrived monocyte-derived interstitial macrophages that subsequently transition into alveolar macrophages (Mo-AMs). These transitioning cells initiate fibrosis by releasing profibrotic cytokines and remodeling the matrix. Here, we describe a strategy for leveraging the up-regulation of the mannose receptor CD206 in interstitial macrophages and Mo-AM to treat lung fibrosis. We engineered mannosylated albumin nanoparticles, which were found to be internalized by fibrogenic CD206+ monocyte derived macrophages (Mo-Macs). Mannosylated albumin nanoparticles incorporating TGFβ1 small-interfering RNA (siRNA) targeted the profibrotic subpopulation of CD206+ macrophages and prevented lung fibrosis. The findings point to the potential utility of mannosylated albumin nanoparticles in delivering TGFβ-siRNA into CD206+ profibrotic macrophages as an antilung fibrosis strategy.

Pea-like nanocabins (HA@APT§DOX) were designed for deep tumor inhibition. The AS1411 aptamer (APT) constituted "core shelf" which guaranteed DOX "beans" could be embedded, while the outer HA acted as "pea shell" coating. During the circulation (primary orbit), HA@APT§DOX could autonomously cruise until leak through tumor vasculature. Upon tumor superficial site, the "pea shell" could be degraded by highly expressed hyaluronic acid enzymes (HAase) and peel-off, resulting in orbit changing of released APT§DOX to reach the deep tumor tissue. Furthermore, APT§DOX could be specifically uptaken into A549 tumor cells (secondary orbit). Finally, DOX was released under the acidic environment of lysosome, and delivered into nuclear (targeting orbit) to achieve drug pushing for deep tumor inhibition. More importantly, the in vivo imaging and anti-tumor effects evaluations showed that these nanocabins could effectively enhance drugs accumulation in tumor sites and inhibit tumor growth, with reduced systemic toxicity in 4T1 tumor-bearing mice.

The analysis of heterogeneous subpopulations of circulating tumor cells (CTCs) is critical to enhance our understanding of cancer metastasis and enable non-invasive cancer diagnosis and monitoring. The phenotypic variability and plasticity of these cells – properties closely linked to their clinical behavior – demand techniques that isolate viable, discrete fractions of tumor cells for functional assays of their behavior and detailed analysis of biochemical properties. Here, we introduce the Prism Chip, a high-resolution immunomagnetic profiling and separation chip which harnesses a cobalt-based alloy to separate a flowing stream of nanoparticle-bound tumor cells with differential magnetic loading into ten discrete streams. Using this approach, we achieve exceptional purity (5.7 log white blood cell depletion) of isolated cells. We test the differential profiling function of the integrated device using prostate cancer blood samples from a mouse xenograft model. Using integrated graphene Hall sensors, we demonstrate concurrent automated profiling of single cells and CTC clusters that belong to distinct subpopulations based on protein surface expression.

The current disadvantages (high cost, toxicity, resistance) of chemotherapy for gastric cancer opted people for alternative therapy from natural source. Curcumin (natural product) possess multiple biological activities but low bio-availability limits their uses as therapeutic. The Nano-formulation of curcumin increased the bioavailability and productivity of anti-cancer and anti-bacterial properties. The present study was initiated to determine the anti-cancer and anti-bacterial effect of Nano curcumin against gastric cancer and H. pylori.Curcumin loaded PLGA nanoparticles (CUR-NPs) was prepared by single emulsion solvent evaporation method. The MIC were determined using agar dilution method to find the anti-H. Pylori activity of Nano curcumin. The cytotoxicity of Nano curcumin was evaluated by MTT assay and the apoptotic effect (cell cycle arrest and morphology change) was shown by PI staining and microscopy.The MIC of nanocurcumin and curcumin for all four H. pylori strains were 8 µg/ml and 16 µg/ml respectively. The inhibition rate of gastric cancer cells after treatment with curcumin was increased from 6% to 67% for 24h, from 8% to 75% for 48h, from 10% to 83% for 72h. In case of nanocurcumin, the inhibition rate increased from 7% to 69% for 24h, 11% to 87% for 48h and 16% to 97% for 72h. The IC50 of curcumin and Nano-curcumin were 24.20 µM and 18.78 µM respectively for 72 h. The population of cells in sub-G0 population increased from 4.1% in the control group to 24.5% and 57.8% when treated with curcumin and nanocurcumin respectively. After 72h of treatment with nanocurcumin, the apoptotic cells population increased as compared to native curcumin treated cells.The Nano curcumin might be used as a potential therapeutics against gastric cancer and H. Pylori. There is need of further in vivo study in order to validate CUR-NPs activity.

The emergence of COVID-19 makes imperative the need for safe and potent antiviral drugs. Many metal nanoparticles exhibit significant antiviral potential against many viral diseases. The Ayurvedic system of medicine is the treasure of many metal nanoparticulate drugs termed as Bhasma. Recent findings Gold, silver, copper, zinc and iron oxide nanoparticles are effective against coronavirus. A possible mechanism of action of the metal nanoparticles against coronavirus is a disruption of outer layers of coronavirus. Swarna Bhasma, Rajata Bhasma, Tamra Bhasma and Yashada Bhasma are recommended for COVID-19 treatment due to the ability to reduce the plasma interleukins, interferons and TNFα levels. Summary The Ayurvedic Bhasma preparations are unique metal nanoparticles. These metal nanoparticles are safe, stable in solid state and are having excellent biological activities. Ayurvedic metal nanoparticles, Swarna Bhasma, Rajata Bhasma, Tamra Bhasma and Yashada Bhasma could be proved as novel antiviral agents against SARS-CoV-2 for their anti-inflammatory, immunomodulatory, antiviral and adjuvant activities.

Nerve growth factor (NGF) is a minor and neglected component of snake venom. Present study describes the purification and characterization of a NGF isoform (RVV-NGFa) from Indian Russell’s viper venom (RVV). RVV-NGFa showed a protonated molecular ion [MH+] at m/z 17388.725 Da. The RVV-NGFa induced neuritogenesis in PC-12 cells but did not show cytotoxicity in mammalian cells, hemolytic activity, platelet modulation, and interference in blood coagulation system which are the characteristic pharmacological properties of RVV. Curcumin (CUR), an antioxidant with p-glycoprotein inhibiting activity may be encapsulated with gemcitabine (GEM) as nanosuspension to enhance its anticancer potentiality synergistically. Folate conjugated single (CUR/GEM) and dual (CUR + GEM) drug-loaded nanoformulations were prepared and evaluated for P-glycoprotein-1 (pgy-1) gene resistance, followed by in vitro cellular uptake and cytotoxicity assay in cells. The in vivo biodistribution and scintigraphic imaging was done after radiolabeling the nanoparticles with 99mTechnetium (99mTc). The tumor inhibition study was conducted in nude mice bearing MDA-MB-231 xenografts. The folate conjugated dual drug formulations (FCGNPs) gave better results in suppressing the pgy-1 gene and also showed higher cellular uptake, cytotoxicity, apoptosis, and cell cycle arrest. The radiolabeled nanoformulations were highly stable and FCGNPs showed higher accumulation in the MDA-MB-231 tumor region than folate unconjugated dual drug NPs (CGNPs) as evidenced by scintigraphic imaging and biodistribution studies. The in vivo therapeutic efficacy of FCGNPs was higher compared to unconjugated and respective single-drug formulations. Two drugs in one platform lower breast adenocarcinoma by lowering drug resistance and improving cytotoxic effects.

Polyvinyl-pyrrolidone capped spherical cadmium sulphide quantum dots (CdS-PVP QDs), 2–6 nm in size, were developed as a selective turn-on fluorescence nanosensor for monohydrogen phosphate ion (HPO42−) in aqueous medium. Fluorescence intensity of CdS-PVP QDs significantly increased with addition of HPO42− ions, whereas the other common inorganic ions had very little effect on the fluorescence intensity. The proposed sensor may be efficiently used for the detection of HPO42− ions at a low level of concentration up to 213 nM in real urine sample. Cell imaging study indicates that the CdS-PVP QDs are cell permeable and can detect the intracellular distribution of HPO42− ions under fluorescence microscope. The CdS-PVP QDs showed considerable activity against Staphylococcus aureus also.

Clinical Research Curcumin, a nontoxic bioactive agent of turmeric significantly reduces nicotine-induced toxicity both at cellular and genetic levels. The clinical implication of native curcumin is hindered in the target cells due to its low aqueous solubility, poor bioavailability and poor pharmacokinetics. The problem was tried to overcome by preparing nanocurcumin with a view to improve its aqueous solubility and better therapeutic efficacy against nicotine-induced toxicity. The prepared nanocurcumin was characterized by Ultraviolet-visible spectroscopy; Field emission scanning electron microscopy (FE-SEM); X-ray diffraction (XRD); and Fourier transform infrared spectroscopy (FTIR). Female albino rats of Wistar strain were daily exposed to effective dose of nicotine (2.5 mg/kg, injected subcutaneously) and supplemented with effective dose of curcumin (80 mg/kg body weight orally) or nanocurcumin (4 mg/kg body weight orally) for 21 days. The preventive efficacies of curcumin and nanocurcumin were evaluated against the changes in liver function enzymes, kidney function parameters, lipid profiles, lipid-peroxidation, anti-oxidant status, and tissues damages etc. Results revealed that nanocurcumin more effectively ameliorated the nicotine-induced toxicities at much lower concentration due to its higher aqueous solubility and more bioavailability. The nanocurcumin can be used as a potential therapeutic agent for better efficacy against nicotine-induced toxicities than native curcumin.

A scalable, cost effective synthesis of reduced graphene oxide (RGO)-magnesium oxide (MgO) hybrid nanocomposite (RGOMOHNC) is reported in the present work. The process involves an in-situ sonication method in aqueous medium at room temperature e.g., 30 °C. The as synthesized graphene oxide (GO) and RGOMOHNC powders are characterized by the XRD, FESEM, TEM, FTIR, RS and XPS techniques. The results indicate that in-situ growth of 20–30 nm MgO nanoparticles effectively increases the graphitic nature of the layered RGO microstructure. The experimentally measured cyclic voltammetry (CV) plots show that the specific capacitance of the RGOMOHNC powders is three orders of magnitude higher than that of the GO powders. A mechanism of RGOMOHNC formation has been proposed.

Utilization of nanotechnology and nanomaterials is an integral part of modern life. The increased use of nanomaterials in various consumer products, industries, medical instruments, and information technology and energy sectors has created research interest because of their potential toxicity for the environment . Nanomaterials can also be added directly and indirectly to the soil and water treatment plants to reduce pollutant concentrations. Nanoparticles may enter the aquatic system through runoff and industrial effluent discharge, therefore, potentially contaminate both the aquatic and terrestrial systems. Researchers have used laboratory experiments to understand the effect of nanomaterials and their transformation in water and soil environment. The transformation of nanoparticles in the environment involves various physical and chemical processes, and their degradation/transformation products may introduce further toxicity. Various microparticles or host materials can be used to support or coat nanoparticles in order to reduce their toxicity. Microparticles including clay minerals, polymer , carbon -based materials (biochar ) are popular to support nanoparticles due to their large surface area and improved functional characteristics. This chapter aims to give an overview of the risks associated with using nanoparticles , and how to reduce the possible toxicity related to nanoparticles by using microparticle host materials.

In this study, the silica nanoparticles surface was modified by carboxylic acid with different number of alkyl chain and two different lipases were immobilized on it to increase the glycerolysis activity of the immobilized lipases in organic solvent system. Silica nanoparticles of about 15 nm diameter were grafted with various carboxylic acid modifiers from valeric, caprylic, capric, lauric, stearic and oleic acids. Lipases from Candida antarctica and Candida rugosa were immobilized onto the carboxylic acid, modified silica nanoparticles and used in the mono and diglycerides production through glycerolysis with or without organic solvents. When lipases immobilized on stearic acid-modified silica nanoparticles, both lipases gave higher activity compared to their corresponding free lipases. Immobilized C. rugosa were stable and reused for 11 cycles without loss in activity. The kinetic parameters, K m and V max of free and immobilized lipases were found using Lineweaver–Burk model. Results indicated that the immobilized lipase had a lower K m and V max when compared with the free lipase.

The chemical synthesis of this study targets for development of a bio-safe polymeric nano-vehicle for improvising the solubility of the flavanone naringenin in antidiabetic animal study. Nanoparticles were prepared from two cost-effective carbohydrate biopolymers - chitosan and alginate for successful encapsulation of naringenin. Dual crosslinked nanoparticles were synthesized by using Na2SO4 and CaCl2 as crosslinkers. The nanoparticles were characterized by DLS, FTIR, XRD and SEM. The prepared nano-formulations exhibited significant naringenin entrapment of >90% and pH-responsive slow and sustained release of the flavonoid. In-vivo studies revealed significant hypoglycemic effect after oral delivery of the nanoparticles to streptozotocin-induced diabetic rats. Histopathology and several blood parameters indicated that oral administrations of nanoparticles were free from toxicity. Other studies also suggested that polymeric formulations were quite effective for oral delivery of the flavonoid as a therapeutic agent in the treatment of dyslipidemia, hyperglycemia and haemoglobin iron-mediated oxidative stress in type 1 diabetic model.

The present work provides the first ever report on extraordinarily high antibacterial efficacy of phase pure micro-layered calcium hydroxide nanoparticles (LCHNPs) even under dark condition. The LCHNPs synthesized especially in aqueous medium by a simple, inexpensive method show adequate mechanical properties along with the presence of a unique strain tolerant behaviour. The LCHNPs are characterized by FTIR, Raman spectroscopy, XRD, Rietveld analysis, FE-SEM, TEM, TG-DTA, surface area, particle size distribution, zeta potential analysis and nanoindentation techniques. The LCHNPs have 98.1% phase pure hexagonal Ca(OH)2 as the major phase having micro-layered architecture made up of about ~100-200nm thick individual nano-layers. The nanomechanical properties e.g., nanohardness (H) and Young's modulus (E) of the LCHNPs are found to have a unique load independent behavior. The dielectric responses (e.g., dielectric constant and dielectric loss) and antibacterial properties are evaluated for such LCHNPs. Further, the LCHNPs show much better antibacterial potency against both gram-positive e.g., Staphylococcus aureus (S. aureus) and gram-negative e.g., Pseudomonas putida (P. putida) bacteria even in dark especially, with the lowest ever reported MIC value (e.g., 1 μg ml-1) against the P. putida bacterial strain and exhibit ROS mediated antibacterial proficiency. Finally, such LCHNPs has almost ~8-16% inhibition efficacy towards the development of biofilm of these microorganisms quantified by colorimetric detection process. So, such LCHNPs may find potential applications in the areas of healthcare industry and environmental engineering.

Micron long carbon nanofibers (CNFs) were grown on porous carbon beads to give an active surface for rapid immobilization of guest molecules. The fabrication of nanostructures using a catalytic route involving chemical vapour deposition on a porous substrate was accomplished by the controlled synthesis of iron nanoclusters on the surface of porous carbon beads. The challenge of catalyst nanoparticle diffusion into the porous substrate was addressed by using iron coordinated ligand complexes and optimizing the loading percentage of metal salts onto beads. The effect of using tethered bottom up surface processed CNFs on the porous beads' morphologies was established using structural characterization. The protruding architecture of CNFs on the porous carbon surface was subjected to bacterial colonisation in order to determine the efficiency of cell conjugation onto hairy structures, particularly at a low concentration. The interfaces of immobilized bacteria on the textured surface were studied by varying the pH and external physical stimuli to check the biofilm formation. The strategy of fabricating all carbon porous beads, which had topologically controlled 'nano on micro' geometries, to give fast immobilization of guest molecules could be useful in the future for developing an active disinfectant surface.

Challenges in reliable nucleic acid detection are manifold. The major ones are related to false positive or negative signals due to a lack of target specificity in detection and to low sensitivity, especially when a plethora of background sequences are present that can mask the specific recognition signal. Utilizing designed synthetic nucleic acids that are commonly called xeno nucleic acids could offer potential routes to meeting such challenges. In this article, we present the general framework of nucleic acid detection, especially for nanoscale applications, and discuss how and why the xeno nucleic acids could be truly an alternative to the DNA probes. Two specific cases, locked nucleic acid (LNA) and peptide nucleic acid (PNA), which are nuclease-resistant and can form thermally stable duplexes with DNA, are addressed. It is shown that the relative ease of the conformationally rigid LNA probe to be oriented upright on the substrate surface and of the nonionic PNA probe to result into high probe density assists in their use in nanoscale nucleic acid recognition. It is anticipated that success with these probes may lead to important developments such as PCR-independent approaches where the major aim is to detect a small number of target sequences present in the analyte medium.

Therapeutic angiogenesis is a highly appealing concept for treating tissues that become ischemic due to vascular disease. A major barrier to the clinical translation of angiogenic therapies is that the patients that are in the greatest need of these treatments often have long term disease states and co-morbidities such as diabetes and obesity that make them resistant to angiogenic stimuli. In this study, we identified that human patients with type 2 diabetes have reduced levels of glypican-1 in the blood vessels of their skin. The lack of this key co-receptor in the tissue may make the application of exogenous angiogenic growth factors or cell therapies ineffective. We created a novel therapeutic enhancer for growth factor activity consisting of glypican-1 delivered in a nanoliposomal carrier (a “glypisome”). Here, we demonstrate that glypisomes enhance FGF-2 mediated endothelial cell proliferation, migration and tube formation. In addition, glypisomes enhance FGF-2 trafficking by increasing both uptake and enhancing endosomal processing. We encapsulated FGF-2 or FGF-2 with glypisomes in alginate beads and used these to deliver localized growth factor therapy in a murine hind limb ischemia model. Co-delivery of glypisomes with FGF-2 markedly increased the recovery of perfusion and vessel formation in ischemic hind limbs of wild type and diabetic mice in comparison to mice treated with FGF-2 alone. Together, our findings support that glypisomes are effective means for enhancing growth factor activity and may improve the response to local angiogenic growth factor therapies for ischemia.

The present report describes the catalytic activity of mechanically activated nano quasicrystalline Al65Cu20Fe15 and related nano crystalline Al50Cu28Fe22 for the synthesis of carbon nanotubes (CNTs). CNTs are synthesized by catalytic decomposition of ethanol through nano quasicrystalline Al65Cu20Fe15 and related crystalline Al50Cu28Fe22 alloys as a catalyst. The synthesized multi-walled CNTs exhibits tube diameter ranging from 5 to 25 nm. The synthesized CNTs are characterized by scanning and transmission electron microscopy. It is found that Al65Cu20Fe15 nanoquasicystal shows better catalytic behaviour as compared to nano-crystalline Al50Cu28Fe22 alloys for decomposition of ethanol during the synthesis of multi-walled CNTs.
