Phosvitin (PV) is the main phosphoprotein in egg yolk, with the highest degree of phosphorylation known in nature. The PV and resveratrol (Res) can form a complex, thus effectively improve the solubility of Res. In this work, the interaction between Res and PV was investigated by the fluorescence spectroscopy and molecular docking. The fluorescence emission intensity of PV became weak along with a red shift when it interacted with Res and the antioxidant activity was enhanced. The quenching constants of the interaction systems were 1.12×10 4  M -1 and 9.40×10 3  M -1 at 25°C and 35°C, respectively, which indicated the presence of static quenching phenomena between them. The binding constant was 1.80×10 4  M -1 , and the number of corresponding binding sites was approximately equal to one. The thermodynamic results revealed the combination was spontaneous, and the change of enthalpy and entropy was ∆H = 53.50 kJ/mol, ∆S = 261.00 J/mol·K, respectively. It indicated that the interaction forces between Res and PV were mainly hydrophobic interaction and hydrogen bonding. Molecular docking showed the binding mode, which was consistent with the experiment results. The research on the interaction between Res and PV provided theoretical guidance for the application of Res in food. PRACTICAL APPLICATION: PV is the most highly phosphorylated protein in nature and has pro-calcium absorption effects. Res is a polyphenol with strong antioxidant and anti-inflammatory activity, but its poor solubility limits its application. In this study, the solubility of Res was considerably enhanced by compounding Res and PV, and the antioxidant activity of Res was well retained. This study investigated the effects of phosvitin (PV), one of the major proteins from egg yolk, with different degree of phosphorylation on the physiology of an osteoblast (MC3T3-E1) cell line. The proliferation and differentiation of MC3T3-E1 were analyzed using the CCK-8 and the alkaline phosphatase (ALP) assay, respectively. The effect of PV on the mineralization of MC3T3-E1 was monitored using the Alizarin-red staining. PV at 100 μg/mL increased the ALP activity by 145% of the control after 7 days of incubation. PV also stimulated the proliferation and differentiation of MC3T3-E1 in a phosphorylation level-dependent manner. The RT-PCR reactions indicated that PV stimulated the expression of BMP-2 and OPG mRNA in a phosphorylation-dependent manner, but inhibited RANKL mRNA expression in MC3T3-E1. This result suggested that the phosphate groups in PV not only stimulated the proliferation and differentiation of MC3T3-E1, but also controlled the mineralization by regulating the expression of BMP-2, RANKL and OPG mRNA in the osteoblast cell., (Copyright © 2018 Elsevier B.V. Phosphorylation of phosvitin has been proved to play an important role in the mineralization, but the active region of phosvitin is still not known yet. Four phosvitin phosphopeptides (PPPs) were obtained after separating from ion exchange column and desalting from gel filtration, and named as PPP0, PPP1, PPP3 and PPP4, respectively. The effect of PPP on the mineralization was investigated by pH-stat system, Fourier transform infrared spectroscopy, X-ray diffraction and scanning electron microscope. SDS-PAGE and circular dichroism were applied to study the composition and the structure of PPP. Results showed that the order of promoting mineralization was as follows: PPP3 > phosvitin > PPP4 > PPP1 > control > PPP0. PPP0 and PPP1 was not involved in the mineralization, while the structure of PPP4 was too compact to promote mineralization because of its high β-sheet conformation. PPP3, which contained a 10 kDa peptide, is the most effective promoter for mineralization. Background: Egg yolk phosvitin, one of the most highly phosphorylated extracellular matrix proteins known in nature, has a strong calcium binding and reducing capacity. Here, we investigated the effects of phosvitin on osteoblast differentiation and osteogenic gene expression in cultured mouse osteoblastic MC3T3-E1 cells by using alkaline phosphatase activity analysis, alizarin red S staining and real-time PCR assay., Results: Alkaline phosphatase activity and alizarin red S staining analyses demonstrated no significant difference between differentiating MC3T3-E1 cells cultured in the presence of phosvitin and those cultured in the presence of ascorbic acid after 21 days of differentiation. Our real-time PCR assay also indicated the two groups were similar in the expression of the osteogenic gene markers, collagen type I, osteocalcin, runt-related transcription factor 2, and bone morphogenetic protein-2., Conclusion: Our findings indicate that phosvitin plays a similar role to that of ascorbic acid in osteoblast differentiation and mineralisation. © 2017 Society of Chemical Industry., (© 2017 Society of Chemical Industry.)

Phosvitin is a unique highly phosphorylated protein that plays a role in the regulation of mineralization. This study investigated the role of phosphorylation of phosvitin in the phase transformation of calcium phosphate in the mineralization solution. Partially dephosphorylated phosvitins (T1, T2, T3 and T4) were prepared with 2.98, 19.46, 43.39 and 71.07% of phosphate released from native phosvitin, respectively. And their effect on regulating the phase transformation was investigated, the characterization and composition analysis was performed by circular dichroism, Fourier transform infrared spectroscopy, X-ray diffraction and scanning electron microscopy. Results showed that phosvitin in the acceleration effect of phase transformation was dose-concentration effect by pH-stat titration. With lower dephosphorylation degree of phosvitin (<20%), the acceleration effect was weaker than native phosvitin, since phosphorylation and random structure of phosvitin were reduced. However, with higher dephosphorylation degree of phosvitin (>40%), the acceleration effect was enhanced compared to native phosvitin, in which the β-sheet structure was increased and phosvitin was partially hydrolyzed to phosphopeptides. The acceleration effect of phase transformation was as follows: T4>T3>phosvitin>T1>T2> Control. Phosvitin is a unique highly phosphorylated protein that plays a role in the regulation of calcification. We conducted a comprehensive study of the chemical, thermodynamic and structural aspects of the interaction of phosvitin with calcium ions using a calcium ion selective electrode (ISE), isothermal titration calorimetry (ITC), circular dichroism spectrum (CD) and fluorescence spectroscopy, respectively. The results showed that under neutral and alkaline conditions, distinct high affinity and low affinity binding modes existed in the interaction between phosvitin and calcium. The high affinity association constant was approximately 10(4)mol(-1), while the binding sites contained nearly 30mol of calcium per mole of phosvitin. This reaction was driven by enthalpy. The unordered and β-turn conformations of phosvitin increased, while the β-sheet conformation decreased. The main interaction forces were electrostatic force, hydrogen bonds or van der Waals force. The low affinity association constant and binding sites were not constant, as many calcium ions were sequestered by phosvitin. The binding reaction was driven by entropy, and the β-sheet conformation of phosvitin increased while the unordered conformation decreased. The main interaction force was hydrophobic force. However, under acidic conditions, the interaction between phosvitin and calcium was an entropy-driven endothermic reaction, and the main interaction force was weak hydrophobic force. The aim of this study was to investigate the role of phosvitin in bone formation in chicken embryos. The yolk P content, P/N ratio and secondary structure of phosvitin, alkaline phosphatase (ALP) activity of the tibia, and body length were determined during incubation. A high correlation was found between the phosphate group content of phosvitin and both secondary structure and bone metabolism (ALP activity in the tibia, body length). The ALP activity and body length growth slightly lagged behind changes in the P/N ratio and the secondary structure of phosvitin. The phosphate content of phosvitin decreased, the γ-random coil and β-turn gradually transformed into α-helixes, and the secondary structure of protein tended to become more orderly; these changes mainly occurred on d 13 to 16. Bone formation of the chicken embryos occurred primarily on d 14 to 18, whereas ALP activity and body length growth increased substantially. An attempt was made to develop a new protocol for preparing phosvitin that could be easy scaled up using polyethylene glycol (PEG6000). Influence of PEG6000 concentration and pH of sample solution on phosvitin isolation was investigated. Phosvitin of high purity (99%) was obtained in good yield (47%) with the optimal condition of pH 4.0 and 3% PEG6000 precipitation. In addition, through evaluating different anion-exchange chromatography methods, the DEAE procedure at pH 7.5 was finally selected as the best procedure to obtain metal-free phosvitin that lost the least protein. Furthermore, it is observed that the purity and characterizations of prepared phosvitin were similar to those of the standard phosvitin from Sigma, and the random coil of phosvitin converted into more compact structure after removing metal ion. In a tenfold-concentrated simulated body fluid, a strategy for rapid deposition of a biomimetic calcium phosphate layer on the scaffolds of electrospun collagen nanofiber membranes was developed. The aim of this study was to explore the effects of mineralization conditions and phosvitin (PV) on hydroxyapatite nucleation and growth. The mineralization model, the pH of the environment, and the deposition time were optimized. Scanning electron microscopy (SEM) images demonstrated that homogeneous and well-crystallized inorganic mineral layers were generated in the dynamic mineralization model system after incubating 3 h at pH 5.7. PV, which possesses the highest level of phosphorylation among egg proteins, was used as a model protein to investigate the contribution of PV in the mineralization process. The morphological structure and composition of the collagen/calcium phosphate composite nanofibers were also characterized by energy dispersive spectroscopy, scanning photoelectron spectrometer, X-ray diffraction (XRD), and Fourier transform infrared spectroscopy. XRD results showed the transformation process of mineralization materials from dicalcium phosphate dihydrate (DCPD) to HA through the changes of characteristic peaks at approximately 11° of DCPD and 31.8° of HA. 1.0 mg/mL. Phosvitin significantly promoted the phase transformation from DCPD to hydroxyapatite. High performance liquid chromatography results indicated that PV induced the mineralization rather than being the part of the hydroxyapatite. The minerals formed on electrospun collagen nanofiber membranes were identified to be from hydroxyapatite. These findings extended the potential application field of PV to biomimetic material. Phosvitin has a similar structure and similar properties to the phosphorylated proteins that play an important role in biomineralization, suggesting that phosvitin may have similar regulation properties. This study investigated the effect of phosvitin on regulating the phase transformation of the mineral calcium phosphate in a biomimetic mineralization solution; the characterization techniques used were Fourier transform infrared spectroscopy, X-ray diffraction, scanning electron microscopy, energy dispersive spectroscopy and fluorescence spectroscopy. The results clearly demonstrated that phosvitin significantly promotes the initiation of phase transformation, accelerated the transformation process and shortened the transformation time from 6 to 0.5 h. Phosvitin was involved in the phase transformation and incorporated into or strongly absorbed on the mineral, as evidenced by the protein peaks observed in the FTIR spectra and XRD patterns. The effects of the substrate-addition sequence on the phase transformation demonstrated that the phosvitin-Ca 2+ interaction played a key role in the regulation of mineralization. Compared with those for BSA, the results revealed that the role of phosvitin in mineralization is closely associated with its high level of phosphorylation.
