Perennial ryegrass (Lolium perenne) is a forage and amenity grass species widely cultivated in temperate regions worldwide. As such, perennial ryegrass populations are exposed to a range of environmental conditions and stresses on a seasonal basis and from year to year. One source of potential stress is limitation on water availability. The ability of these perennial grasses to be able to withstand and recover after periods of water limitation or drought can be a key component of grassland performance. Thus, we were interested in looking at changes in patterns of gene expression associated with increasing water stress. Clones of a single genotype of perennial ryegrass were grown under non-flowering growth room conditions in vermiculite supplemented with nutrient solution. Leaf and root tissue was sampled at 4 times in quadruplicate relating to estimated water contents of 35%, 15%, 5% and 1%. RNA was extracted and RNAseq used to generate transcriptome profiles at each sampling point. Transcriptomes were assembled using the published reference genome sequence and differential gene expression analysed using 3 different programmes, DESeq2, edgeR and limma (with the voom transformation), individually and in combination, deriving Early, Middle and Late stage comparisons. Identified differentially expressed genes were then associated with enriched GO terms using BLAST2GO. For the leaf, up-regulated differentially expressed genes were strongly associated with GO terms only during the Early stage and the majority of GO terms were associated with only down-regulated genes at the Middle or Late stages. For the roots, few differentially expressed genes were identified at either Early or Middle stages. Only one replicate at 1% estimated water content produced high quality data for the root, however, this indicated a high level of differential expression. Again the majority of enriched GO terms were associated with down-regulated genes. Abstract Vegetation concrete is an effective material to beautify the landscape, reduce pollution and protect the environment. To further improve the plant compatibility of vegetation concrete, this paper recommends an improved method of adding biochar particles to vegetation concrete. In this research, different masses of biochar were mixed into concrete to study the trend in porosity, permeability and plant compatibility. According to the growth status of three types of common grass species, ryegrass was selected as the best grass species in this study. As the content of biochar increased, the porosity and permeability coefficient of vegetation concrete continued to decrease, while the effect of biochar on plant growth promotion at first showed an increase to the maximum and then a gradual decrease. When the biochar content was 5 kg/m3, ryegrass showed the best growth. Compared with the experimental group of vegetation concrete without biochar, the plant height, root length and germination rate increased by 22.2%, 21.1% and 12%, respectively, on day 25, and the compressive strength also increased slightly. Therefore, adding a suitable amount of biochar can improve the characteristics of vegetation concrete. Furthermore, the optimum mix proportion of biochar-modified vegetation concrete was recommended. Agricultural grasslands are often cultivated as mixtures of grasses and legumes, and an extensive body of literature is available regarding interspecific interactions, and how these relate to yield and agronomic performance. However, knowledge of the impact of intraspecific diversity on grassland functioning is scarce. We investigated these effects during a 4-year field trial established with perennial ryegrass (Lolium perenne) and red clover (Trifolium pratense). We simulated different levels of intraspecific functional diversity by sowing single cultivars or by combining cultivars with contrasting growth habits, in monospecific or bispecific settings (i.e. perennial ryegrass whether or not in combination with red clover). Replicate field plots were established for seven seed compositions. We determined yield parameters and monitored differences in genetic diversity in the ryegrass component among seed compositions, and temporal changes in the genetic composition and genetic diversity at the within plot level. The composition of cultivars of both species affected the yield and species abundance. In general, the presence of clover had a positive effect on the yield. The cultivar composition of the ryegrass component had a significant effect on the yield, both in monoculture, and in combination with clover. For the genetic analyses, we validated empirically that genotyping-by-sequencing of pooled samples (pool-GBS) is a suitable method for accurate measurement of population allele frequencies, and obtained a dataset of 22,324 SNPs with complete data. We present a method to investigate the temporal dynamics of cultivars in seed mixtures grown under field conditions, and show how cultivar abundances vary during subsequent years. Alien chromosome introgression is used for the transfer of beneficial traits in plant breeding. For temperate forage grasses, much of the work in this context has focused on species within the ryegrasses (Lolium spp.) and the closely related fescues (Festuca spp.) particularly with a view to combining high forage quality with reliability and enhanced environmental services. We have analysed a L. perenne (perennial ryegrass) population containing the majority of a F. pratensis (meadow fescue) genome as introgressed chromosome segments to identify a) marker-trait associations for nutrient use and abiotic stress response across the family, and b) to assess the effects of introgression of F. pratensis genomic regions on phenotype. Using container-based assays and a system of flowing solution culture, we looked at phenotype responses, including root growth, to nitrogen and phosphorus status in the growing medium and abiotic stresses within this festulolium family. A number of significant marker/trait associations were identified across the family for root biomass on chromosomes 2, 3 and 5 and for heading date on chromosome 2. Of particular interest was a region on chromosome 2 associated with increased root biomass in phosphorus-limited conditions derived from one of the L. perenne parents. A genotype containing F. pratensis chromosome 4 as a monosomic introgression showed increased tiller number, shoot and root growth and genotypes with F. pratensis chromosome segment introgressions at different ends of chromosome 4 exhibited differential phenotypes across a variety of test conditions. There was also a general negative correlation between the extent of the F. pratensis genome that had been introgressed and root-related trait performances. We conclude that 1) the identification of alleles affecting root growth has potential application in forage grass breeding and, 2) F. pratensis introgressions can enhance quantitative traits, however, introgression can also have more general negative effects. Understanding the stability of chloropyromorphite (CPY) is of considerable benefit for improving risk assessment and remediation strategies in contaminated water and soil. The stability of CPY in the rhizosphere of phosphorus-deficient ryegrass was evaluated to elucidate the role of root-secreted low molecular weight organic acids (LMWOAs) on the dissolution of CPY. Results showed that CPY treatments significantly reduced the ryegrass biomass and rhizosphere pH. The presence of calcium nitrate extractable lead (Pb) and phosphorus (P) suggested that CPY in the rhizosphere could be bioavailable, because P and Pb uptake by ryegrass potentially provided a significant concentration gradient that would promote CPY dissolution. Pb accumulation and translocation in ryegrass was found to be significantly higher in P-sufficient conditions than in P-deficient conditions. CPY treatments significantly enhanced root exudation of LMWOAs irrigated with P-nutrient solution or P-free nutrient solution. Oxalic acid was the dominant species in root-secreted LMWOAs of ryegrass under P-free nutrient solution treatments, suggesting that root-secreted oxalic acid may be the driving force of root-induced dissolution of CPY. Ryegrass (Lolium perenne L.) is a putative turf and forage plant characterized as mainly outcrossing in which the extent of self‐fertilization and its consequences has not been investigated. In this study, selfed and half‐sib family seeds were obtained from ten clonally propagated ryegrass plants among which two were endophyte‐free (EF) and eight were endophyte‐infected (EI). Then, 20 seed populations plus two commercial varieties of Speedy and Vigor were sown and evaluated in a biennial experiment for their turf and forage characteristics. Selfing caused significant inbreeding depression for plant fresh and dry weight and leaf length and width in the first year for both EI and EF genotypes. In the second year, only leaf length remained depressed among these characters. Also, for the most growth, turf and reproductive traits, EI populations showed less depression. The presence of endophyte in self‐pollinated populations made an increasing effect for the number of seed per spike and days to heading, although seed weight was reduced. Some of the populations derived from self‐pollination and infected with fungal endophyte including S9 and S10 were even superior to the two commercial varieties in terms of both seed production and turf characteristics. However, for seed‐related characteristics including number of spikes per plant, seed weight and seed yield, EF populations including Speedy and Vigor were more reproductive than EI populations. It is concluded that selfing may increase the discrimination between EI and EF ryegrass plants because endophyte infection may mitigate the negative consequences of inbreeding depression in prone populations. Proline is an important osmoprotectant in plant in response to osmotic stresses. Delta 1-pyrroline-5-carboxylate synthetase ( P5CS) is a key enzyme in proline biosynthesis. In this study, two P5CS genes ( PrP5CS1 and PrP5CS2) were isolated for the first time from perennial ryegrass. Expression analysis revealed that the transcript of PrP5CS1 in leaves was significantly up-regulated in two ryegrass cultivars exposed to 255 m m NaCl. The up-regulated level of PrP5CS1 was higher in salt-tolerant 'Overdrive' than in sensitive 'Pizzazz'. PrP5CS2 was significantly induced in 'Overdrive' but suppressed in 'Pizzazz' by NaCl treatment. In stems, however, there was no significant transcript change for both genes under salt treatment. The proline accumulation was significantly induced in both cultivars after salt treatment, and it was higher in 'Overdrive' than in 'Pizzazz' after 2 days of salt treatment. Little is known about the effects of NaCl stress on perennial ryegrass (Lolium perenne L.) photosynthesis and carbohydrate flux. The objective of this study was to understand the carbohydrate metabolism and identify the gene expression affected by salinity stress. Seventy-four days old seedlings of two perennial ryegrass accessions (salt-sensitive ‘PI 538976’ and salt-tolerant ‘Overdrive’) were subjected to three levels of salinity stress for 5 days. Turf quality in all tissues (leaves, stems and roots) of both grass accessions negatively and significantly correlated with GFS (Glu+Fru+Suc) content, except for ‘Overdrive’ stems. Relative growth rate (RGR) in leaves negatively and significantly correlated with GFS content in ‘Overdrive’ (P<0.01) and ‘PI 538976’ (P<0.05) under salt stress. ‘Overdrive’ had higher CO2 assimilation and Fv/Fm than ‘PI 538976’. Intercellular CO2 concentration, however, was higher in ‘PI 538976’ treated with 400 mM NaCl relative to that with 200 mM NaCl. GFS content negatively and significantly correlated with RGR in ‘Overdrive’ and ‘PI 538976’ leaves and in ‘PI 538976’ stems and roots under salt stress. In leaves, carbohydrate allocation negatively and significantly correlated with RGR (r2 = 0.83, P<0.01) and turf quality (r2 = 0.88, P<0.01) in salt-tolerant ‘Overdrive’, however, the opposite trend for salt-sensitive ‘PI 538976’ (r2 = 0.71, P<0.05 for RGR; r2 = 0.62, P>0.05 for turf quality). A greater up-regulation in the expression of SPS, SS, SI, 6-SFT gene was observed in ‘Overdrive’ than ‘PI 538976’. A higher level of SPS and SS expression in leaves was found in ‘PI 538976’ relative to ‘Overdrive’. Accumulation of hexoses in roots, stems and leaves can induce a feedback repression to photosynthesis in salt-stressed perennial ryegrass and the salt tolerance may be changed with the carbohydrate allocation in leaves and stems. We propose Pseudomonas coronafaciens sp. nov. as a new species in genus Pseudomonas, which is diverse from P. syringae. We also classified strains from onions which are responsible for yellow bud (YB) disease as P. coronafaciens. Sequencing of 16S rRNA gene and multi-locus sequence analysis (MLSA) of housekeeping genes (gyrB, rpoD, gltA and gap1 genes) for the P. syringae pv. coronafaciens strains along with other strains of P. syringae pathovars resulted in a distinct cluster separate from other P. syringae pathovars. Based on DNA-DNA relatedness, pathotype strain of P. syringae pv. coronafaciens (CFBP 2216PT) exhibited ≤35.5% similarity with the pathotype strains of P. syringae pv. syringae (CFBP 1392PT, 4702T) but exhibited ≥90.6% with the YB strains (YB 12–1, YB 12–4, YB 09–1). Also, the YB strains (YB 12–1, YB 12–4, YB 09–1) were able to infect only onion but not oat, rye and Italian ryegrass (common hosts for P. syrinage pv. coronafaciens). Contrastingly, P. syringae pv. coronafaciens strains (NCPPB 600PT, ATCC 19608, Pcf 83–300) produced typical halo blight symptoms on oat, rye and Italian rye grass but did not produce any symptoms on onion. These results provide evidence that P. syringae pv. coronafaciens should be elevated to a species level and the new YB strains may potentially be a novel pathovar of hereto proposed P. coronafaciens species. Wear tolerance is important in sports turf, but the effects of nitrogen (N) and potassium (K) on wear mechanisms in perennial ryegrass (Lolium perenne L.) are unknown. The objective of this study was to evaluate wear mechanisms in response to N and K in this species. Field studies were conducted in 2006 and 2007 using five N levels (49, 147, 245, 343, and 441 kg ha-1 yr-1) in combination with three rate levels of K (49, 245, and 441 kg ha-1 yr-1). Wear was applied in both years using wear simulators. Ten plant characteristics were measured in field plots. Nitrogen had significant effect on wear tolerance and plant characteristics. Shoot growth accounted for as much as 94% of the variation in wear tolerance (r = -0.97, p ⩽ 0.001). Tissue moisture measured as relative water content and shoot water content covaried with shoot growth. Wear injury, shoot growth rate and tissue moisture increased with increasing N, especially when fertilized in excess of 245 kg N ha-1 yr-1. Tissue moisture explained as much as 66% (r = -0.81, P ⩽ 0.001) of the variation in wear tolerance. Cell wall components, leaf strength, and shoot density were secondary in explaining wear tolerance. Verdure was not important. Tissue K and soil-available K were not correlated with wear tolerance. These results suggest fertilizing at no more than 245 kg N ha-1 y-1 to maintain optimum wear tolerance, shoot growth rate and tissue moisture. [ABSTRACT FROM AUTHOR]

Four perennial ryegrass ( Lolium perenne L.) cultivars were compared for differences in herbage production, nutritive value and herbage intake of dry matter (DM) during the summers of 2002 and 2003. Two paddocks were sown with pure stands of four cultivars in a randomized block design with three replicates. Each plot was subdivided into fourteen subplots (22 m × 6 m) which were grazed by one cow during 24 h. Twelve lactating dairy cows were assigned to one cultivar for a period of 2 weeks in a 4 × 4 Latin square experimental design; the experiment lasted 8 weeks in each year. Sward structure (sward surface height, DM yield, green leaf mass, bulk density and tiller density) and morphological characteristics were measured. The ash, neutral-detergent fibre, acid-detergent lignin, crude protein and water-soluble carbohydrate concentrations, and in vitro digestibility of the herbage were measured. The sward was also examined for infestation by crown rust ( Puccinia coronata f. sp. lolii). Herbage intake of dairy cows was estimated using the n-alkane technique. Cultivar differences for all sward structural characteristics were found except for bulk density and tiller density in 2003. Cultivars differed for proportions of pseudostem, stem (in 2003 only) and dead material. The chemical composition of the herbage was different among cultivars, with the water-soluble carbohydrate concentration showing large variation (>0·35). Cultivars differed in susceptibility to crown rust. Herbage intake differed among cultivars in 2002 (>2 kg DM) but not in 2003. Herbage intake was positively associated with sward height, DM yield and green leaf mass. Canopy morphology did not affect herbage intake. Crown rust affected herbage intake negatively. It was concluded that options for breeders to select for higher intake were limited. High-yielding cultivars and cultivars highly resistant to crown rust were positively related with a high herbage intake. [ABSTRACT FROM AUTHOR]

The objective of this work was to determine if the impact of nitrogen (N) on the release of organic carbon (C) into the soil by roots (rhizodeposition) correlated with the effect of this nutrient on some variables of plant growth.Lolium multiflorumLam. was grown at two levels of N supply, either in sterile sand percolated with nutrient solution or in non-sterile soil. The axenic sand systems allowed continuous quantification of rhizodeposition and accurate analysis of root morphology whilst the soil microcosms allowed the study of14C labelled C flows in physico-chemical and biological conditions relevant to natural soils. In the axenic sand cultures, enhanced N supply strongly increased the plant biomass, the plant N content and the shoot to root ratio. N supply altered the root morphology by increasing the root surface area and the density of apices, both being significantly positively correlated with the rate of organic C release by plant roots before sampling. This observation is consistent with the production of mucilage by root tips and with mechanisms of root exudation reported previously in the literature, i.e. the passive diffusion of roots solutes along the root with increased rate behind the root apex. We proposed a model of root net exudation, based on the number of root apices and on root soluble C that explained 60% of the variability in the rate of C release from roots at harvest. The effects of N on plant growth were less marked in soil, probably related to the relatively high supply of N from non-fertiliser soil-sources. N fertilization increased the shoot N concentration of the plants and the shoot to root ratio. Increased N supply decreased the partitioning of14C to roots. In parallel, N fertilisation increased the root soluble14C and the14C recovered in the soil per unit of root biomass, suggesting a stimulation of root exudation by N supply. However, due to the high concentration of N in our unfertilised plants, this stimulation was assumed to be very weak because no significant effect of N was observed on the microbial C and on the bacterial abundance in the rhizosphere. Considering the difficulties in evaluating rhizodeposition in non sterile soil, it is suggested that the root soluble C, the root surface area and the root apex density are additional relevant variables that should be useful to measure along with the variables that are commonly determined when investigating how plant functioning impacts on the release of C by roots (i.e soil C, C of the microbial biomass, rhizosphere respiration). Perennial ryegrass (Lolium perenneL.) is the most important temperate forage grass species. Unfortunately, the nutritional value of perennial ryegrass declines as maturity progresses, mainly because of a high concentration of poorly digestible compounds in inflorescences. Therefore, the development of forage-type ryegrass varieties with extended vegetative growth is of interest for agriculture. To delay floral transition in perennial ryegrass theArabidopsis ATH1gene driven by the maize ubiquitin promoter, the rice actin promoter or the rice OSH1 promoter, respectively was introduced. InATH1-expressing plants heading was delayed, and in a number of cases the plants never flowered at all. Such non- or late-heading was accompanied by the outgrowth of normally quiescent lateral meristems into extra leaves, resulting in a leafy growth habit. When eventually heading, these plants generally produced a reduced number of inflorescences. The impact of uprooting and covering plants on mortality and growth reduction was investigated in the laboratory using Lolium perenne L. and Lepidium sativum L. (harrowed 3–4 days after emergence) and Chenopodium quinoa Willd. (harrowed at emergence) as model weed species. Although the predominant initial effect of harrowing was to cover the plants, only 1–17% of the non-uprooted covered plants were killed because the depth at which they were buried by the harrow was shallow. Uprooting was more effective (47–61% mortality) but strongly dependent on soil moisture content. It accounted for 93 and 95% of L. sativum and C. quinoa mortality, but for only 60% of L. perenne mortality. In L. perenne, the species most sensitive to burying, a strong positive relationship was observed between the percentage of plants covered by harrowing and the fresh weight reduction of the total population 6 days after harrowing. The fresh weight reduction of the total L. sativum population was best related to the percentage of uprooted plants, but the percentage of covered plants also appeared to be a good predictor because of its correlation with uprooting. Most of the uprooted plants were also buried. The fresh weight reduction of the total C. quinoa population was not related to the covering effect of harrowing and only weakly related to the percentage of uprooted plants. The results indicate that the plant recovery process after harrowing needs further study and that field research methods should be refined so that they can better discern initial and final harrowing effects on weeds. The 15N isotope was used to compare the uptake and the assimilation of NH+4 and NO-3 nitrogen in ryegrass (Lolium perenne L.) during regrowth after cutting. Uptake of nitrate-N, expressed per plant, was at all times greater than ammonium-N uptake. During the first 5-6 d of regrowth, ammonium uptake and assimilation decreased in roots and stubble while its assimilation was maintained at a high level in leaves. It has been suggested that ammonium assimilation is directly related to the availability of carbohydrates in the sink organ (leaves) resulting from their remobilization from the source organs (roots and stubble). Nitrate reduction decreased in all organs, while the uptake of NO-3 was still high. After this first period of regrowth, nitrogen assimilation both from nitrate and ammonium increased in all the plants. Nitrate reduction capacity (expressed in μg NO-3-N reduced per g D.W. per d) is 7.5 and 22.5 times greater in leaves than in stubble and roots, respectively. Genetic control of the capacity to respond to anther culture in perennial ryegrass was studied in F1 offspring from crosses between 11 clones selected for anther-culture response ('inducers') and 10 clones selected from breeding material on the basis of good agronomic performance. Large differences in anther-culture response were observed between the two types of parent, with inducer clones producing an average of 10.7 green plants per 100 anthers, compared with only 0.3 for breeding clones. Hybrid populations produced an average of 2.2 green plants per 100 plated anthers, which is 7.3 times the response of the breeding material. This improvement was mainly due to a 9.4-fold increase in the percentage of green plants regenerated from hybrid populations to 4.7% compared to an average of 0.5% from breeding clones. Most of the GCA (general combining ability) in the experiment was contributed by the breeding material, constituting 55.8, 100.0 and 36.7% of genetic variation among hybrids for embryo formation, regeneration and green plant percentage, respectively. Any remaining genetic variation among hybrids was predominantly due to SCA (specific combining ability) effects, with percentages of 44.2 and 63.3%, respectively, for embryo and green plant formation. These results are discussed with respect to the possible exploitation of genes controlling anther-culture response for haploid production in breeding programmes of perennial ryegrass. Anther-culture response in Lolium perenne was studied in the hybrid populations horn three responsive diploid parent clones to obtain genotypes with superior another-culture response through hybridization. Genetic variation for another-culture response was significant both within and between the four hybrid families and their parents. A reciprocal effect was indicated for embryo formation and regeneration non percentages for one of the cross combinations. Among 55 hybrid clones tested in two environments, 6 clones were identified with an anther- culture response level superior to their parents. While the parents produced one green plant or less per 100 anthers, these superior clones produced from 11 to 59 green plants/100 anthers, mainly as the result of improved embryo formation and improved percentages of regenerated plants being green. A significant correlation (r = 0.397**) was observed between embryo formation and percentage of green plants. Key message : Heuristic genomic inbreeding controls reduce inbreeding in genomic breeding schemes without reducing genetic gain. Abstract: Genomic selection is increasingly being implemented in plant breeding programs to accelerate genetic gain of economically important traits. However, it may cause significant loss of genetic diversity when compared with traditional schemes using phenotypic selection. We propose heuristic strategies to control the rate of inbreeding in outbred plants, which can be categorised into three types: controls during mate allocation, during selection, and simultaneous selection and mate allocation. The proposed mate allocation measure GminF allocates two or more parents for mating in mating groups that minimise coancestry using a genomic relationship matrix. Two types of relationship-adjusted genomic breeding values for parent selection candidates ( $${{\widetilde{\text{GEBV}}}_{\text{P}}}$$ ) and potential offspring ( $${{\widetilde{\text{GEBV}}}_{\text{O}}}$$ ) are devised to control inbreeding during selection and even enabling simultaneous selection and mate allocation. These strategies were tested in a case study using a simulated perennial ryegrass breeding scheme. As compared to the genomic selection scheme without controls, all proposed strategies could significantly decrease inbreeding while achieving comparable genetic gain. In particular, the scenario using $${{\widetilde{\text{GEBV}}}_{\text{O}}}$$ in simultaneous selection and mate allocation reduced inbreeding to one-third of the original genomic selection scheme. A total 568 perennial ryegrass collected from 66 different locations were characterized and evaluated regarding yield and quality. Seeds were firstly sown in plastic trays on October 2010 and then transplanted to field with 50 x 50 distances on March. In first year, there was no observation and plants were cut frequently to avoid seed formation. Observation began the second year and the traits: plant height, main stem diameter, node number, flag leaf length and leaf ratio, plant dry weight, protein, ADF, NDF ratios, and mineral matter (P, K, Ca, Mg,) content were investigated. The data was between 46.2-71.2 cm for plant height, 1.09-6.67 mm for main stem diameter 2.0-5.0 for node number, 1.00-25.20 cm / 0.70-11.13 mm for flag leaf length/width, 5.41-91.03% for leaf ratio, 5.00-192.00 g for plant dry weight, 7.20-21.00% for protein ratio, 21.58-43.90% for ADF, 50.80-82.70% for NDF and 0.32-0.50%, 2.74-5.05%, 0.06-0.678%, 0.09-0.31% for P, K, Ca, Mg content, respectively. The study highlighted the existence of high variability between the populations. Ryegrass (Lolium perenne L.) is a plant species that can express mechanisms of tolerance to copper (Cu) toxicity. Therefore, the agronomical approach of intercropping system with ryegrass may represent a promising tool to limit the onset of Cu toxicity symptoms in the other intercropped plants species, particularly when an inadequate nutrient availability like iron (Fe) shortage is also concurrently present. This study aimed at assessing the mechanisms involved in the mitigation of Cu phytotoxicity and the stress effects on plant growth, root morphology and nutrition of ryegrass fertilized with two different Fe sources. To this purpose, seedlings of ryegrass were hydroponically grown for 14 days in controlled conditions with 4 different levels of Cu (0.2, 5.0, 25 and 50 μM) and with either 100 μM Fe-EDDHA or Fe-EDTA. Results show that high levels of Cu availability enhanced the root content of organic anions as well as the root exudation. Different Fe fertilizations at the condition of 50 μM Cu induced changes in root phenolic compounds, citrate and fumarate contents and the exudation pattern of phenolic compounds. Differences in plant growth were not observed between the two Fe sources, although Cu concentration in plant tissue fed with Fe-EDTA was lower in the condition of 50 μM Cu. The enhanced root exudation of Cu-complexing organic compounds (including phenolics) in ryegrass plants when exposed to excessive Cu availability could be at the basis of the ameliorated edaphic rhizosphere conditions (lower Cu availability). For this reason, from the agronomical point of view ryegrass plants used in intercropping systems with crops like vine plants could represent a promising strategy to control Cu toxicity in vineyard soils. In recent years advances in quantitative trait loci (QTL) mapping of complex metabolomics traits have been made in plants. This study is a two-phase experiment in which we have profiled transesterified non-polar metabolites in a Lolium perenne population consisting of parents, F1 and 325 F2 genotypes. These were grown in a replicated field trial, then analysed in batches by a targeted approach using gas-chromatography mass spectrometry (GC-MS), which identified 17 metabolites. The metabolite data was analysed using a mixed model that included random effects of field replicate, laboratory batch and technical replication to derive genotype means for QTL mapping. Using a linkage map of 326 markers, mainly Diversity Array Technology markers, QTLs were detected for seven of the metabolites. The most heritable metabolite was octacosanol, where a single QTL on linkage group four explained 42 % of the trait variance. [ABSTRACT FROM AUTHOR]

Information on the spring growth status of winter forage crops is crucial for evaluating productivity and nutrient management. This study aimed to determine the spring quick growth stage (QGS) of Italian ryegrass using a groundbased camera system. The camera system, installed in two Italian ryegrass fields at the farm of Hiroshima University, captured images automatically three times per day in red, green and blue channels over the growing season in 2012-13. Four presumed color intensities/indices were fitted using a logistic model to construct smoothed time-series data. Among the color intensities/indices, excess green was suggested to be the best parameter for monitoring seasonal changes. The root mean squared error of the estimated phenology dates against plant height was 7.7 days for the start-QGS and 2.8 days for the end-QGS. These results from a single year should be broadened to examine other methodologies for image processing and extended to multi-year data. [ABSTRACT FROM AUTHOR]

Genomic selection is focused on prediction of breeding values of selection candidates by means of high density of markers. It relies on the assumption that all quantitative trait loci (QTLs) tend to be in strong linkage disequilibrium (LD) with at least one marker. In this context, we present theoretical results regarding the accuracy of genomic selection, i.e., the correlation between predicted and true breeding values. Typically, for individuals (so-called test individuals), breeding values are predicted by means of markers, using marker effects estimated by fitting a ridge regression model to a set of training individuals. We present a theoretical expression for the accuracy; this expression is suitable for any configurations of LD between QTLs and markers. We also introduce a new accuracy proxy that is free of the QTL parameters and easily computable; it outperforms the proxies suggested in the literature, in particular, those based on an estimated effective number of independent loci (Me). The theoretical formula, the new proxy, and existing proxies were compared for simulated data, and the results point to the validity of our approach. The calculations were also illustrated on a new perennial ryegrass set (367 individuals) genotyped for 24,957 single nucleotide polymorphisms (SNPs). Selenium is both essential and toxic for mammals; the range between the two roles is narrow and not only dose-dependent but also related to the chemical species present in foodstuff. Unraveling the metabolism of Se in plants as a function of Se source may thus lead to ways to increase efficiency of fertilization procedures in selenium deficient regions. In this study, stable-isotope tracing was applied for the first time in plants to simultaneously monitor the bio-incorporation of two inorganic Se species commonly used as foodstuff enrichment sources. Occurrence and speciation of Se coming from different Se sources were investigated in root and leaf extracts of ryegrass (Lolium perenne L. ), which had been co-exposed to two labeled Se species (SeIV and SeVI). Although the plant absorbed similar amounts of Se when supplied in the form of selenite or selenate, the results evidenced marked differences in speciation and tissues allocation. Selenite was converted into organic forms incorporated mostly into high molecular weight compounds with limited translocation to leaves, whereas selenate was highly mobile being little assimilated into organic forms. Double-spike isotopic tracer methodology makes it possible to compare the metabolism of two species-specific Se sources simultaneously in a single experiment and to analyze Se behavior in not-hyperaccumulator plants, the ICP-MS sensitivity being improved by the use of enriched isotopes. The greater sensitivity of white clover (Trifolium repens L.) to low temperature compared with perennial ryegrass (Lolium perenne L.) is a major problem in sustaining the relative contributions to yield of the two species in mixed swards. The objectives of this investigation were to examine the dynamics of leaf development of two white clover cultivars, AberHerald and Grasslands Huia, under field conditions, and to determine the significance of leaf area in winter, and of competition by perennial ryegrass, for the overwintering and regrowth of white clover in spring. Undefoliated white clover plants developed 3·8–6·6 new leaves between late autumn and early spring, and stolon dry matter and total non-structural carbohydrates (TNC) content increased by 262 and by 68% respectively. In contrast, white clover plants that were defoliated frequently during the winter showed a 28% decrease in stolon dry matter and an 82% decrease in the content of TNC. Frequent defoliation in winter caused severe reduction in the rates of emergence of nodes (by 60%) and of buds (by 67%), and the rate of death of nodes and buds increased by a factor of 10, leading to small stolon systems of individual plants in spring. Competition had similar, but weaker, effects to those of winter defoliation, presumably caused by shading of white clover leaves. AberHerald had a higher cumulative leaf emergence (by 22%), a higher mean leaf number (by 23%), a higher stolon DM (by 36%) and a higher TNC content per plant (by 115%) than Grasslands Huia. Results demonstrate the crucial positive role of leaf area during winter, the negative effect of grass competition, and the importance of the clover cultivar, for the overwintering and subsequent spring regrowth of white clover. Perennial ryegrass (Lolium perenne) is a perennial crop used in temperate regions as forage. In L. perenne breeding programs, persistency is an important trait. Poor persistency results in sward degradation and associated yield and nutritive value losses. Breeders assess persistency of accessions using visual scoring in field plots during the 2nd or 3rd growing season. This evaluation system is easy and cheap but is not free from human bias. In this study, the correlation between the scoring done by different breeders was only 0.243. As an alternative we have developed a methodology to assess persistency of L. perenne breeding materials based on vegetation indices (VIs) derived from Unmanned Aerial Vehicle (UAV) imagery. The VIs Excess green (ExG2), Green Leaf Index and Normalized green intensity (GCC) were found to provide consistent results for flights carried out under different light conditions and were validated by ground reference information. The correlation between the VIs and the percentage of ground cover extracted from on-ground imagery was 0.885. To test the implementation of the method we compared the ExG2 value based approach to selection with a visual score based selection methodology as applied by two breeders. The breeding decisions of Breeder A agreed well with decisions based on ExG2 values (74.6%), but those of Breeder B displayed a lower agreement (54.0%). In contrast, agreement between decisions based on different flights was very high (91.6%). The methodology was validated for general applicability. In summary, the results demonstrate that basing persistency selection in L. perenne breeding programs on ExG2 values from UAV imagery is likely to be more objective in comparison to the currently-used visual scoring method. To date, knowledge gaps and associated uncertainties remain unaddressed on the effects of nanoparticles (NPs) on plants. This study was focused on revealing some of the physiological effects of magnetite (Fe3O4) NPs on perennial ryegrass ( Lolium perenne L.) and pumpkin ( Cucurbita mixta cv. white cushaw) plants under hydroponic conditions. This study for the first time reports that Fe3O4 NPs often induced more oxidative stress than Fe3O4 bulk particles in the ryegrass and pumpkin roots and shoots as indicated by significantly increased: (i) superoxide dismutase and catalase enzyme activities, and (ii) lipid peroxidation. However, tested Fe3O4 NPs appear unable to be translocated in the ryegrass and pumpkin plants. This was supported by the following data: (i) No magnetization was detected in the shoots of either plant treated with 30, 100 and 500 mg l−−1 Fe3O4 NPs; (ii) Fe K-edge X-ray absorption spectroscopic study confirmed that the coordination environment of Fe in these plant shoots was similar to that of Fe-citrate complexes, but not to that of Fe3O4 NPs; and (iii) total Fe content in the ryegrass and pumpkin shoots treated with Fe3O4 NPs was not significantly increased compared to that in the control shoots. Chemical methods to predict the bioavailable fraction of organic contaminants are usually validated in the literature by comparison with established bioassays. A soil spiked with polycyclic aromatic hydrocarbons (PAHs) was aged over six months and subjected to butanol, cyclodextrin and tenax extractions as well as an exhaustive extraction to determine total PAH concentrations at several time points. Earthworm (Eisenia fetida) and rye grass root (Lolium multiflorum) accumulation bioassays were conducted in parallel. Butanol extractions gave the best relationship with earthworm accumulation (r 2 ≤ 0.54, p ≤ 0.01); cyclodextrin, butanol and acetone–hexane extractions all gave good predictions of accumulation in rye grass roots (r 2 ≤ 0.86, p ≤ 0.01). However, the profile of the PAHs extracted by the different chemical methods was significantly different (p < 0.01) to that accumulated in the organisms. Biota accumulated a higher proportion of the heavier 4-ringed PAHs. It is concluded that bioaccumulation is a complex process that cannot be predicted by measuring the bioavailable fraction alone. The I1781L amino acid substitution in the target ACCase enzyme causes broad resistance to ACCase inhibitor herbicides in several monocotyledenous weeds of agronomic importance. This mutation results from a substitution of an adenine (A) residue by either a thymine (T) or cytosine (C) at position 5341 in Alopecurus myosuroides Huds and at an equivalent position in Lolium species, Avena fatua L. and Setaria viridis (L.) Beauv. Two different procedures, the PCR-based allele-specific assay (ASA) and the derived cleaved amplified polymorphic sequence (dCAPS) method, have previously been described for detecting this mutation. These methods are, however, only amenable to low sample throughput and are used in the analysis of single plants. Here, an alternative high-throughput ARMS/Scorpion real-time quantitative PCR (Q-PCR) method for measuring levels of the I1781L mutation in pools of leaf and seed samples of Lolium populations is presented. The limit of detection for C and T mutant alleles in a background of wild-type A is 0.02 and 0.0003% respectively. In this study, DNA from batches of 24 leaf segments measuring 0.5 cm from different plants or 1000 seeds could be conveniently extracted and accurately analysed. As part of assay validation, the comparative analysis of five geographically distinct Lolium populations with dCAPS and Q-PCR procedures demonstrated the accuracy of the latter method, and the three possible II1781, IL1781 and LL1781 ACCase genotypes being distributed as predicted by the Hardy-Weinberg principle. Given the dominance of the L1781 over the I1781 allele at recommended field rates for most ACCase inhibitors, the frequency of herbicide survivors in the field owing only to the presence of the I1781L mutation is thus predicted to be 2pq + q², where p and q are the frequencies of the I1781 and I1781 alleles as determined by Q-PCR. The Q-PCR assay established allows detection of very low levels of the L1781 ACCase mutation before resistance would normally be discernible in the field. Therefore, it offers the opportunity to tackle resistance at its very onset, potentially avoiding implementation of complicated and often costly weed management practices. [ABSTRACT FROM AUTHOR]

• Costs of resistance are predicted to reduce plant productivity in herbicide-resistant weeds. • Lolium rigidum herbicide-susceptible individuals (S), individuals possessing cytochrome P450-based herbicide metabolism (P450) and multiple resistant individuals possessing a resistant ACCase and enhanced cytochrome P450 metabolism (ACCase/P450) were grown in the absence of mutual plant interaction to estimate plant growth traits. • Both P450 and ACCase/P450 resistant phenotypes produced less above-ground biomass than the S phenotype during the vegetative stage. Reduced biomass production in the resistant phenotypes corresponded to a reduced relative growth rate and a lower net assimilation rate and rate of carbon fixation. There were no significant differences between the two resistant phenotypes, suggesting that costs of resistance are associated with P450 metabolism-based resistance. There were no differences in reproductive output among the three phenotypes, indicating that the cost of P450 resistance during vegetative growth is compensated during the production of reproductive structures. • The P450-based herbicide metabolism is shown to be associated with physiological resistance costs, which may be manipulated by agronomic management to reduce the evolution of herbicide resistance. Large-scale sequencing of disease resistance gene analogs (RGA) in Italian ryegrass was carried out using degenerate primers and a nested polymerase chain reaction (PCR) strategy. A total of 24 000 PCR clones were subjected to single pass sequencing to enable similarity searches of public databases. As a result, 9344 clones showed significant levels of homology to either known resistance genes or RGA derived from other plant species. These clones were grouped into 115 clusters based on nucleotide sequence similarities and representative clones were selected from each cluster. Of 115 representative clones, 62 had a continuous open reading frame and were defined as potentially functional RGA from Italian ryegrass. Specific sequenced tag site (STS) primer sets were designed for these 62 clones and 50 RGA sequences were subsequently amplified from genomic DNA of Italian ryegrass in one step PCR. A subset of these STS primers could also amplify the expected size PCR products from genomic DNA of tall fescue and meadow fescue, both of which are forage crops closely relate to Italian ryegrass. The molecular basis and mode of inheritance of resistance to ACCase-inhibiting herbicides were investigated in a biotype of Lolium rigidum Gaud that has been discovered in Israel. Dose-response experiments at the whole-plant level have revealed that the resistant biotype was more resistant (6.3- to 40-fold) than the susceptible wild-type to ACCase-inhibiting herbicides. A 276-bp genomic DNA encoding the carboxyltransferase domain within the chloroplastic ACCase from resistant and susceptible biotypes were amplified by PCR and analyzed. Sequence comparison revealed that a single isoleucine-to-leucine substitution differentiated ACCases from susceptible and resistant biotypes (corresponds to residue 1769 of wheat ACCase, Ace No AF029895). A PCR amplification of specific alleles (PASA) method was developed to detect the allele composition leading to isoleucine-leucine mutation. ACCase extracted from homozygote resistant, heterozygote and homozygote susceptible plants showed IC50 values of 25.8, 5.6 and 0.6 µM, respectively, suggesting that alteration in the ACCase is governed by a co-dominant gone. The inheritance studies confirmed that the resistance of L rigidum to ACCase-inhibiting herbicides is governed by a single, nuclear and co-dominant gene. [ABSTRACT FROM AUTHOR]

There is ample experimental evidence that, Na, if supplied in separate fertiliser granules or crystals to N, i.e., in blended fertiliser form, can improve both the yield and the recovery of fertiliser N by grassland swards in situations of limited K supply, but not in situations of K abundance. There is some evidence, though, that in K-replete situations, Na, if supplied in the same fertiliser granule as N, i.e. in concentrated complex fertiliser (CCF) form, also improves dry matter production and N recovery by swards whilst lowering the risk of grass tetany in grazing animals. However, the mechanism for the latter effect of Na on N uptake has never been elucidated, nor has it been clarified whether Na stimulates NHand NOuptake by plants or simply NOuptake alone. The aim of the present study was to see if supplying Na in the same fertiliser pellets (NNa-CCF) as NH4NO3 (differentially labelled with15N), or in separate pellets (NNa-blend), had any effect on the recovery of15N-labelled NHand NO-N by perennial ryegrass plants growing in a glasshouse under K-replete conditions. The results of the experiment confirmed that using an NNa-CCF was more beneficial to shoot production than using an NNa-blend. However, the differential in shoot production occurred without any corresponding difference in total N (i.e. NHplus NO-N) recovery in shoot tissue. Instead, Na, in the CCF appears to have stimulated NOuptake at the expense of NHabsorption, thereby altering the balance between NHand NO-nutrition in favour of NO-nutrition, and stimulating shoot production as a consequence. It was concluded that if grassland is already well supplied with K it would be more beneficial in terms of sward production to apply a Na and N-containing CCF than a blend of separate Na and N-containing granules or crystals. A greenhouse study was conducted over a 12-month period to investigate the fate of polycyclic aromatic hydrocarbons (PAHs) in soil using phytoremediation as a secondary treatment. The soil was pretreated by composting for 12 weeks, then planted with tall fescue (Festuca arundinacea), annual ryegrass (Lolium multiflorum), and yellow sweet clover (Melilotus officinalis). Two sets of unvegetated controls also were evaluated, one fertilized and one unfertilized. Total PAH concentrations decreased in the tall fescue, annual ryegrass, and yellow sweet clover treatments by 23.9%, 15.3%, and 9.1%, respectively, whereas the control was reduced by less than 5%. The smaller two- and most of the three-ringed compounds--naphthalene, acenaphthylene, acenaphthene, fluorene, and anthracene--were not found in detectable concentrations in any of the treatments. The most probable number analysis for microbial PAH degraders did not show any statistically significant differences among treatments. There were significant differences among treatments (p<0 05) for the residual concentrations of five of the target PAHs. Root surface area measurements indicated that tall fescue and annual ryegrass both had significantly higher root surface area than yellow sweet clover, although the two species were not significantly different from each other. The tall fescue treatment resulted in the highest root and shoot biomass, followed by annual ryegrass and yellow sweet clover, and also had the highest percent of contaminant removal after 12 months. Although adult Rumex obtusifolius are problematic weeds, their seedlings are poor competitors against Lolium perenne, particularly in established swards. We investigated the possibility of using this weakness to augment control of R. obtusifolius seedlings with combinations of Gastrophysa viridula (Coleoptera: Chrysomelidae) and the rust fungus Uromyces rumicis. Rumex obtusifolius seedlings were grown in competition with L. perenne sown at different rates and times after R. obtusifolius: they competed successfully with L. perenne when sown 21 days before the grass. Sowing both species at the same time resulted in a dominant grass sward, with R. obtusifolius becoming dominant when sown 42 days prior to L. perenne. Grass sowing rate had no effect on R. obtusifolius growth or biomass. A second experiment investigated how competition from L. perenne sown 21 days after R. obtusifolius combined with damage from G. viridula and/or U. rumicis (applied at either the 3–4- or 10–13-leaf stage, or at both stages) affected the growth and final biomass of R. obtusifolius. Beetle grazing at the latter leaf stage was the only treatment that reduced R. obtusifolius biomass, although rust infection at the earlier application led to an increase in shoot and root weight. The characters causing low seed-setting rates in induced tetraploid Lolium perenne L. were investigated. The objective of this study was to determine whether these characters could be used as criteria in the selection of regular meiosis and high seed-setting plants. The seeds of C 1 and C 2 generations of artificially obtained tetraploid perennial ryegrass were used. Cytological studies were done on the root tips and spike samples. A significant positive correlation was found between the frequency of cells showing regular anaphase I and high seed-setting rates. As a result, it was decided that this normal separation in anaphase I could be used as a selection criterion for seed set. The highly significant negative correlation that was found between the number of micronuclei and seed setting showed that this character could also be used as a selection criterion when a large population is under consideration. A defined method for the rapid establishment of cell suspensions releasing small-cell colonies and their regeneration to fertile plants is described Cell suspensions were obtained from 10 mature, lengthwise-divided embryos Five of the 10 suspensions regenerated green plants. Filtration of suspensions through a 150 μm mesh followed by generation was carried out 28 days after initiation of the suspensions, to demonstrate a rapid release of regeneration small cell colonies from cell suspensions using the protocol described. Presents an experiment designed to find a more satisfactory method of weed control in seedling pasture by using these herbicides both pre- and early post-emergence and comparing them with triazines whose effect on Grasslands Manawa ryegrass and Tallarook subterranean clover was not known. The article presents a study that examines the impact of temperature and photoperiod on the growth of the shoot apex in a number of species and strains of genus Lolium, and seeks to relate this behavior to the geographical and ecological origins of species and strains. It highlights the growth of the shoot apex, the materials and methods used for the examination of the development, and the rates of leaf and tiller appearance. A discussion on the growth of leaf and bud of the shoot apex and the breeding systems of Lolium is provided. It shows that the critical photoperiod of many plants is greater than 11 hours.

The success of biofortification and phytoremediation practices, addressing Se deficiency and Se pollution issues, hinges crucially on the fate of selenium in the plant media in response to uptake, translocation and assimilation processes. We investigate the fate of selenium in root and shoot compartments after 3 and 6 weeks of experiment using a total of 128 plants grown in hydroponic solution supplied with 0.2, 2, 5, 20 and 100 mg L −1 of selenium in the form of selenite, selenate and a mixture of both species. Selenate-treated plants exhibited higher root-to-shoot Se translocation and total Se uptake than selenite-treated plants. Plants took advantage of the selenate mobility and presumably of the storage capacity of leaf vacuoles to circumvent selenium toxicity within the plant. Surprisingly, 28% of selenate was found in shoots of selenite-treated plants, questioning the ability of plants to oxidize selenite into selenate. Selenomethionine and methylated organo-selenium amounted to 30% and 8% respectively in shoots and 35% and 9% in roots of the identified Se, suggesting that selenium metabolization occurred concomitantly in root and shoot plant compartments and demonstrating that non-accumulator plants can synthesize notable quantities of precursor compound for volatilization. The present study demonstrated that non-accumulator plants can develop the same strategies as hyper-accumulator plants to limit selenium toxicity. When both selenate and selenite were supplied together, plants used selenate in a storage pathway and selenite in an assimilation pathway. New opportunities for plant breeding using androgenesis in Lolium × Festuca hybrids have been identified. Plants derived by anther culture from a Lolium multiflorum × Festuca arundinacea (5x) hybrid were screened for freezing-tolerance, and their post-freezing recovery compared. The androgenic population showed extreme diversity in freezing-tolerance. While the majority of androgenic plants had inferior freezing-tolerance compared with the freezing-sensitive L. multiflorum parent, 6% of the population were more freezing resistant than the freezing-tolerant Festuca parent. Novel Lolium and Festuca gene combinations resulting from rare meiotic events were recovered within the androgenic population. The two most freezing-tolerant androgenic plants carried virtually the entire F. pratensis subgenome of F. arundinacea. F. pratensis is known to carry genes for freezing-tolerance and would be expected to be the primary source of genes governing this trait within the F. arundinacea genome. The most freezing-tolerant androgenic plants were more freezing-tolerant than the hybrid plant from which they were derived. Consequently, androgenesis was also effective in removing factor(s) reducing the expression of freezing-tolerance within the L. multiflorum × F. arundinacea (5x) hybrid. [ABSTRACT FROM AUTHOR]
