Nanolayer and nanolayer by nanolayer deposition of nanofilms of Ag and C using cold plasma in sequences (Ag, Ag-C, Ag-Ag-C), on porous paper, were used to design three disposable stochastic sensors for the assay of amyloid polypeptide from whole blood. The nanofilms were modified with α-cyclodextrin. The test developed using the nanofilm-based disposable stochastic sensors is used for early detection of diabetes. The wider linear concentration range (1.00 × 10−6–1.00 ng mL−1) and the lower limit of quantification (1.00 × 10−6ng mL−1) were obtained using the disposable stochastic sensors based on Ag-C and Ag-Ag-C, while the highest sensitivity (3.19 × 104 s−1/μg mL−1) was recorded using the disposable stochastic sensor based on Ag-Ag-C. The screening methods were fully validated using whole blood samples from confirmed patients, when the recovery of the islet amyloid polypeptide was higher than 98.00%.

The prediction of plasma rotation is of high interest for fusion research due to the effects of the rotation upon magnetohydrodynamic (MHD) instabilities, impurities, and turbulent transport in general. In this work, an analysis method was studied and validated to reliably extract momentum transport coefficients from neutral beam injection (NBI) modulation experiments. To this end, a set of discharges was created with similar background profiles for the ion and electron temperatures, the heat fluxes, the electron density, and the plasma rotation that, therefore, should exhibit similar momentum transport coefficients. In these discharges, a range of temporal perturbations were imposed by modulating and varying the power deposition of the NBI, electron-cyclotron-resonance heating (ECRH), and ion-cyclotron-resonance heating (ICRH). The transport model including diffusion, convection, and residual stress was implemented within the ASTRA code. The Prandtl number P r = χ φ / χ i was assessed via the GKW code. A convective Coriolis pinch was fitted and the intrinsic torque from the residual stress was estimated. The obtained transport coefficients agree within error bars for sufficiently small imposed temperature perturbations, as would be expected, from the similar background profiles. This successful validation of the methodology opens the door to study the parametric dependence of the diffusive and convective momentum transport of the main ions of the plasma as well as the turbulent intrinsic torque in a future work.

Gas puff modulation experiments are performed at ASDEX Upgrade in L-mode plasmas. We model the discharge with the ASTRA transport code in order to determine transport coefficients outside of a normalized radius of ρ pol = 0.95. The experimental data is consistent with a range of particle diffusivities and pinch velocities of the order of D = (0.20 ± 0.13) m2 s−1 and v = (−1 ± 2) m s−1, respectively. The electron temperature response caused by the gas modulation permits to estimate also that heat diffusivity χ e increases almost linearly when collisionality rises due to fuelling. The fuelling particle flux is amplified by recycling, overcompensating losses.

We describe a laser-plasma platform for photon-photon collision experiments to measure fundamental quantum electrodynamic processes such as the linear Breit-Wheeler process with real photons. The platform has been developed using the Gemini laser facility at the Rutherford Appleton Laboratory. A laser wakefield accelerator and a bremsstrahlung convertor are used to generate a collimated beam of photons with energies of hundreds of MeV, that collide with keV x-ray photons generated by a laser heated plasma target. To detect the pairs generated by the photon-photon collisions, a magnetic transport system has been developed which directs the pairs onto scintillation-based and hybrid silicon pixel single particle detectors. We present commissioning results from an experimental campaign using this laser-plasma platform for photon-photon physics, demonstrating successful generation of both photon sources, characterisation of the magnetic transport system and calibration of the single particle detectors, and discuss the feasibility of this platform for the observation of the Breit-Wheeler process. The design of the platform will also serve as the basis for the investigation of strong-field quantum electrodynamic processes such as the nonlinear Breit-Wheeler and the Trident process, or eventually, photon-photon scattering. While colorimetric-based assays are very convenient to determine biomarkers in point-of-care testing (POCT), they often suffer from pretreatment procedures for separation of plasma or serum from whole blood samples. Here, we report a simple colorimetric paper-based analytical device (c-PAD) that is capable of performing sample-to-answer analysis by directly dropping the whole blood sample on paper. This is accomplished by utilizing sodium alginate hydrogel, which exhibits a nanometer-scale porous structure to effectively prevent the passage of large red blood cells and hemoglobin molecules, to encapsulate enzymes and chromogenic reagents. As the small targets in the blood sample enter the sensing region to trigger a chromogenic reaction, the resulting color signal is recorded by a smartphone. The interference from the red blood to the color signal can be completely avoided without the requirement of any separation process. The analytical performance of the method is evaluated by assaying glucose in real blood samples. The results show that rapid and accurate analysis can be achieved with the limit of detection as low as 0.12 mM. In addition, simultaneous detection of different targets (glucose, cholesterol, and triglycerides) in whole blood can be achieved by fabricating c-PAD with multiple sensing regions. Owing to its several essential advantages including an extremely simple procedure for fabrication, sample-to-answer analysis without tedious pretreatment, and capability to perform high-throughput analysis, the proposed c-PAD will be of great value in POCT applications of whole blood samples.

Laser driven high harmonic generation from relativistically oscillating plasma surfaces is a promising route to isolated attosecond pulses with high peak brightness. Here we investigate a double optical gating scheme to restrict the emission to only a single, intense, attosecond pulse even with a multi-cycle driving laser. This scheme, which uses a second harmonic field, combined with a pair of counter-rotating circularly polarized laser pulses, leads to more efficient attosecond pulse generation with improved temporal isolation when compared to a single colour polarization gating scheme.

Paper-based microfluidic analytical devices (μPADs) have recently attracted attention as a point-of-care test kit because of their low cost and nonrequirement for external forces. To directly detect biomarkers in whole blood, however, they need to be assembled with a filter such as a plasma separation membrane (PSM) because the color of the blood cells interferes with the colorimetric assay. However, this assembly process is rather complicated and cumbersome, and the fluid does not uniformly move to the detection zone when the adhesion between the paper and PSM is not perfect. In this study, we report a simple three-dimensional (3D) printing method for fabricating PSM-integrated 3D-μPADs made of plastics without the need for additional assembly. In detail, PSM was coated with parylene C to prevent its dissolution from organic solvent during 3D printing. Then, the coated PSM was superimposed on the paper. Detection zones and a reservoir were printed on the paper and PSM via liquid photopolymerization, using a digital light processing printer. The limit of detection of the PSM-integrated 3D-μPADs for glucose in whole blood was 0.3 mM, and these devices demonstrated clinically relevant performance on diabetes patient blood samples. Our 3D-μPADs can also simultaneously detect multiple metabolic disease markers including glucose, cholesterol, and triglycerides in whole blood. Our results suggest that our printing method is useful for fabricating 3D-μPADs integrated with PSM for the direct detection of biomarkers in whole blood.

Cellulose is a promising biomass material suitable for high volume applications. Its potential lies in sustainability, which is becoming one of the leading trends in industry. However, there are certain drawbacks of cellulose materials which limit their use, especially their high wettability and low barrier properties, which can be overcome by applying thin coatings. Plasma technologies present a high potential for deposition of thin environmentally friendly and recyclable coatings. In this paper, two different plasma reactors were used for coating two types of cellulose-based substrates with hexamethyldisiloxane (HMDSO). The changes in surface characteristics were measured by atomic force microscopy (AFM), scanning electron microscopy (SEM), surface free energy and contact angles measurements, X-ray photoelectron spectroscopy (XPS), and secondary ion mass spectrometry (SIMS). Successful oleofobization was observed for an industrial scale reactor where pure HMDSO was used in the absence of oxygen.

Significance. One of the modern trends in medical diagnostics is based on metabolomics, an approach allowing determination of metabolites which can be the specific features of disease. High-resolution gas spectroscopy allows investigation of the gas metabolite content of samples of biological origin. We present the elaboration of a method of studying diabetic and non-diabetic biological samples, prepared as pellets, by terahertz (THz) high-resolution spectroscopy. Aim: The main idea of the work is studying the content of thermal decomposition gas products of diabetic and non-diabetic dried blood plasma and kidney tissues for revealing the set of gas-markers that characterized the diabetes by the THz high-resolution spectroscopy method. Approach: We present an approach to study the diabetic and non-diabetic blood plasma (human and rats) and kidney tissues (rats), using high-resolution spectroscopy based on the non-stationary effect of THz frequency range. The methods of preparing the blood and kidney tissue samples as pellets and of vaporizing the samples were developed. Results: The measurements of rotational absorption spectra of vapors at heating the pellets prepared from blood and kidney tissue were carried out in 118 to 178 GHz frequency range. The absorption lines appearing in spectra of the sample vapors were detected and identified. The molecular contents of thermal decomposition products differed for non-diabetic and diabetic samples; e.g., main marker is acetone appearing in the diabetic blood (human and rats) and in the diabetic kidney tissue. Conclusions: Our paper illustrates the potential ability for determining the metabolite content of biological samples for diagnostics and prognosis of diseases for clinical medicine.

Significance: The creation of fundamentally new approaches to storing various biomaterial and estimation parameters, without irreversible loss of any biomaterial, is a pressing challenge in clinical practice. We present a technology for studying samples of diabetic and non-diabetic human blood plasma in the terahertz (THz) frequency range. Aim: The main idea of our study is to propose a method for diagnosis and storing the samples of diabetic and non-diabetic human blood plasma and to study these samples in the THz frequency range. Approach: Venous blood from patients with type 2 diabetes mellitus and conditionally healthy participants was collected. To limit the impact of water in the THz spectra, lyophilization of liquid samples and their pressing into a pellet were performed. These pellets were analyzed using THz time-domain spectroscopy. The differentiation between the THz spectral data was conducted using multivariate statistics to classify non-diabetic and diabetic groups’ spectra. Results: We present the density-normalized absorption and refractive index for diabetic and non-diabetic pellets in the range 0.2 to 1.4 THz. Over the entire THz frequency range, the normalized index of refraction of diabetes pellets exceeds this indicator of non-diabetic pellet on average by 9% to 12%. The non-diabetic and diabetic groups of the THz spectra are spatially separated in the principal component space. Conclusion: We illustrate the potential ability in clinical medicine to construct a predictive rule by supervised learning algorithms after collecting enough experimental data.

Conventional paper lateral flow assays have low sensitivity and suffer from severe interference from complex human fluid sample matrices, which prevents their practical application in the testing of whole blood samples in the point-of-care settings. To solve this problem, gold nanostar@Raman reporter@silica-sandwiched nanoparticles have been developed as the surface-enhanced Raman scattering (SERS) probes for sensing transduction; and a functionalized filter membrane assembly has been designed and constructed in the paper-based lateral flow strip (PLFS) as a built-in plasma separation unit. In this "on-strip" plasma separation unit, three layers of filter membranes are stacked and surface-modified to maximize the separation efficiency and the plasma yield. As a result, the integrated PLFS has been successfully used for the detection of carcinoembryonic antigen (CEA) in 30 μL of whole blood with the assistance of a portable Raman reader, achieving a limit of detection of 1.0 ng mL-1. In short, this report presents an inexpensive, disposable, portable, and field-deployable paper-based device as a general point-of-care testing tool for protein biomarker detection in a drop of whole blood.

Significance: Despite the importance of the cell membrane in regulation of drug activity, the influence of drug treatments on its physical properties is still poorly understood. The combination of fluorescence lifetime imaging microscopy (FLIM) with specific viscosity-sensitive fluorescent molecular rotors allows the quantification of membrane viscosity with high spatiotemporal resolution, down to the individual cell organelles. Aim: The aim of our work was to analyze microviscosity of the plasma membrane of living cancer cells during chemotherapy with cisplatin using FLIM and correlate the observed changes with lipid composition and cell’s response to treatment. Approach: FLIM together with viscosity-sensitive boron dipyrromethene-based fluorescent molecular rotor was used to map the fluidity of the cell’s membrane. Chemical analysis of membrane lipid composition was performed with time-of-flight secondary ion mass spectrometry (ToF-SIMS). Results: We detected a significant steady increase in membrane viscosity in viable cancer cells, both in cell monolayers and tumor spheroids, upon prolonged treatment with cisplatin, as well as in cisplatin-adapted cell line. ToF-SIMS revealed correlative changes in lipid profile of cisplatin-treated cells. Conclusions: These results suggest an involvement of membrane viscosity in the cell adaptation to the drug and in the acquisition of drug resistance.

This study describes a microfluidic paper-based analytical device (μPAD) for separating plasma from whole blood and measuring glucose concentration. A two-dimensional μPAD was fabricated by wax printing, using chromatographic paper and was functionalized by the incorporation of chitosan into the plasma separation zone. Red blood cells were isolated from whole blood in the plasma separation zone consisting of a chitosan polymer structure and a wax barrier. The separated plasma, with buffer, was injected into the test zone to cause a color change. The colorimetric results were digitized with a scanner, and the concentration was calculated using a calibration curve. This μPAD, which separated the plasma from whole blood without a separation membrane, will be useful for constructing point-of-care testing devices that detect analytes in small sample volumes.

Atmospheric pressure plasmas have been ground-breaking for plasma science and technologies, due to their significant application potential in many fields, including medicinal, biological, and environmental applications. This is predominantly due to their efficient production and delivery of chemically reactive species under ambient conditions. One of the challenges in progressing the field is comparing plasma sources and results across the community and the literature. To address this a reference plasma source was established during the ‘biomedical applications of atmospheric pressure plasmas’ EU COST Action MP1101. It is crucial that reference sources are reproducible. Here, we present the reproducibility and variance across multiple sources through examining various characteristics, including: absolute atomic oxygen densities, absolute ozone densities, electrical characteristics, optical emission spectroscopy, temperature measurements, and bactericidal activity. The measurements demonstrate that the tested COST jets are mainly reproducible within the intrinsic uncertainty of each measurement technique.

Significance: Accurate and objective identification of Alzheimer’s disease (AD) and dementia with Lewy bodies (DLB) is of major clinical importance due to the current lack of low-cost and noninvasive diagnostic tools to differentiate between the two. Developing an approach for such identification can have a great impact in the field of dementia diseases as it would offer physicians a routine objective test to support their diagnoses. The problem is especially acute because these two dementias have some common symptoms and characteristics, which can lead to misdiagnosis of DLB as AD and vice versa, mainly at their early stages. Aim: The aim is to evaluate the potential of mid-infrared (IR) spectroscopy in tandem with machine learning algorithms as a sensitive method to detect minor changes in the biochemical structures that accompany the development of AD and DLB based on a simple peripheral blood test, thus improving the diagnostic accuracy of differentiation between DLB and AD. Approach: IR microspectroscopy was used to examine white blood cells and plasma isolated from 56 individuals: 26 controls, 20 AD patients, and 10 DLB patients. The measured spectra were analyzed via machine learning. Results: Our encouraging results show that it is possible to differentiate between dementia (AD and DLB) and controls with an ∼86% success rate and between DLB and AD patients with a success rate of better than 93%. Conclusions: The success of this method makes it possible to suggest a new, simple, and powerful tool for the mental health professional, with the potential to improve the reliability and objectivity of diagnoses of both AD and DLB.

The separation of plasma from whole blood is the first step in many diagnostic tests. Point-of-care tests often rely on integrated plasma filters, but protein retention in such filters limits their performance. Here, we investigate plasma separation on interlocked micropillar scaffolds ("synthetic paper") by the local agglutination of blood cells coupled with the capillary separation of the plasma. We separated clinically relevant volumes of plasma with high efficiency in a separation time on par with that of state of the art techniques. We investigated different covalent and non-covalent surface treatments (PEGMA, HEMA, BSA, O2 plasma) on our blood filter and their effect on protein recovery, and identified O2 plasma treatment and 7.9 μg/cm2 agglutination antibody as most suitable treatments. Using these treatments, we recovered at least 82% of the blood plasma proteins, more than with state-of-the-art filters. This work describes disposable plasma generators made from metallized paper. The fabricated plasma generators with layered and patterned sheets of paper provide a simple and flexible format for dielectric barrier discharge to create atmospheric plasma without an applied vacuum. The porosity of paper allows gas to permeate its bulk volume and fuel plasma, while plasma-induced forced convection cools the substrate. When electrically driven with oscillating peak-to-peak potentials of ±1 to ±10 kV, the paper-based devices produced both volume and surface plasmas capable of killing microbes. The plasma sanitizers deactivated greater than 99% of Saccharomyces cerevisiae and greater than 99.9% of Escherichia coli cells with 30 s of noncontact treatment. Characterization of plasma generated from the sanitizers revealed a detectable level of UV-C (1.9 nW⋅cm−2⋅nm−1), modest surface temperature (60 °C with 60 s of activation), and a high level of ozone (13 ppm with 60 s of activation). These results deliver insights into the mechanisms and suitability of paper-based substrates for active antimicrobial sanitization with scalable, flexible sheets. In addition, this work shows how paper-based generators are conformable to curved surfaces, appropriate for kirigami-like “stretchy” structures, compatible with user interfaces, and suitable for sanitization of microbes aerosolized onto a surface. In general, these disposable plasma generators represent progress toward biodegradable devices based on flexible renewable materials, which may impact the future design of protective garments, skin-like sensors for robots or prosthetics, and user interfaces in contaminated environments.

We use scattering in two foils, that are necessary as vacuum window and laser beam dump, to decrease the betatron function of the incoming electron beam for injection and matching into plasma wakefields driven by a self-modulated proton bunch. We show that, for a total aluminum foil thickness of ~ 280 μm, multiple Coulomb scattering increases the beam emittance by a factor of ~ 10 and decreases the betatron function by a factor of ~ 3. The plasma in the accelerator is created by a ionizing laser pulse, counter-propagating with respect to the electron beam. This allows for the electron bunch to enter the plasma through an “infinitely” sharp vapor-plasma boundary, away from the foils. Plasma HIV viral load testing is the preferred means of monitoring antiretroviral treatment response. Plasma separation cards (PSCs) may overcome these limitations. Health workers collected EDTA whole blood by venipuncture and 140 μl of finger-prick blood by capillary tube from 53 HIV-infected adults. Capillary blood was immediately transferred to PSCs. Additionally, 432 EDTA samples from HIV-infected adults were spotted onto PSCs and analyzed together with the finger-prick samples. Specificity and sensitivity of PSC with paired EDTA-PSC samples tested on a cobas 6800/8800 system with the cobas HIV-1 test (cobas HIV) was determined. LoD (3rd HIV-1 WHO International Standard) and stability at a range of temperatures and storage durations was determined using cobas HIV and cobas AmpliPrep/cobas TaqMan HIV-1 test v2.0 (CAP/CTM). Of 132 specimens with quantitative values for paired EDTA-PSC samples, the mean log10 difference between samples was 0.05 copies/ml (95% confidence interval [CI], −0.01 to 0.11). The LoD for cobas HIV was 790.2 copies/ml and for CAP/CTM was 737.9 copies/ml. At 1,000 copies/ml, PSC sensitivity was 97.0% (128/132) and specificity was 97.2% (343/353). Results correlated well with those from EDTA samples (Deming R2 = 0.90). PSC results were unaffected by temperature and storage conditions. PSC samples correlate well with plasma viral load and have adequate sensitivity and specificity. The improved performance may be as a result of a reduction in contribution from cell-associated viral nucleic acids. The card provides an alternative sample collection technology to DBSs.

An investigation of the influence of an evanescent wave on the dynamics of motion of erythrocytes in blood plasma is presented. Computer simulation of erythrocytes moving in an evanescent field and experimental demonstration of the forecasted motion substantiate the possibility for control of position of red blood cells in a solution. The range of velocities of transversal motion of erythrocytes due to the action of the optical force of the generated evanescent field is determined as a function of the angle of illumination of a cell by a linearly polarized wave with the azimuth of polarization 45 deg.

The Advanced Wakefield Experiment (AWAKE) develops the first plasma wakefield accelerator with a high-energy proton bunch as driver. The 400GeV bunch from CERN Super Proton Synchrotron (SPS) propagates through a 10m long rubidium plasma, ionized by a 4TW laser pulse co-propagating with the proton bunch. The relativistic ionization front seeds a self-modulation process. The seeded self-modulation transforms the bunch into a train of micro-bunches resonantly driving wakefields. We measure the density modulation of the bunch, in time, with a streak camera with picosecond resolution. The observed effect corresponds to alternating focusing and defocusing fields. We present a procedure recovering the charge of the bunch from the experimental streak camera images containing the charge density. Current detection methods for paper-based analytical devices (PADs) rely on spectroscopic and electrochemical properties, which place special requirements on the analyte or need analyte labeling. Here, ion-transmission mass spectrometry (MS) was proposed for coupling with PADs to enable rapid in situ MS analysis of the sample on paper. The sample was analyzed directly on paper via analyte ionization by ions transmitted through the paper, generated by a low-temperature plasma probe. Prior to MS analysis, the sample can be separated by paper electrophoresis or by paper chromatography, among a variety of other features offered by PADs. The versatility of this technique was demonstrated by MS analysis of a paper microarray, a mixture of amino acids, and whole blood doped with drugs on PADs.

We use beam position measurements over the first part of the AWAKE electron beamline, together with beamline modeling, to deduce the beam average momentum and to predict the beam position in the second part of the beamline. Results show that using only the first five beam position monitors leads to much larger differences between predicted and measured positions at the last two monitors than when using the first eight beam position monitors. These last two positions can in principle be used with ballistic calculations to predict the parameters of closest approach of the electron bunch with the proton beam. In external injection experiments of the electron bunch into plasma wakefields driven by the proton bunch, only the first five beam position monitors measurements remain un-affected by the presence of the much higher charge proton bunch. The aim of this study was to improve the electrical resistivity, mechanical properties, and interfacial properties between the carbon fiber (CF) and cellulose of conductive paper containing 10% CF so as to better meet the demands for electric heating wood floors. With waterborne polyurethane (WPU) treatment and nitrogen plasma modification methods, the interfacial properties between CF and cellulose were improved dramatically. Fourier transform infrared spectrometry (FTIR) revealed that the plasma modification method reduced the C-H contents and introduced numerous polar groups onto the CF surface. Scanning electron microscopy (SEM) showed that the CF modified by WPU and plasma had good adhesion with cellulose. The tensile index, tensile energy absorption index, and burst index of the paper were enhanced because of the plasma and WPU coating. The carbon fiber and WPU method presented good synergistic action with respect to mechanical properties and electrical resistivity, and the lowest electrical resistivity of conductive paper was reduced from 0.68 •cm2 to 0.44 •cm2.

This paper is devoted to study a structure of SiC-based materials using ultrasonic tomography method. The SiC-based materials were fabricated from preceramic paper using spark plasma sintering (SPS) method. Also as part of the study the Young's modulus and density of sintered materials were determined and the effect of sintering pressure changing to this parameters value was investigated. The preceramic paper is a composite material including a matrix of organic cellulose fibers and inorganic powder filler (SiC). The sintering temperature and pressure were 2373 K and 20-40 MPa, respectively. The holding time for the sintering process was 10 min. The density of sintered materials was investigated by the hydrostatic weighing method. The refractive index of blood is a key biophysical parameter, which can reflect the physiological state. We measured the refractive index of whole blood and other components, such as serum, plasma, and hemoglobin, based on internal reflection by using a homemade apparatus in the spectral range of 400 to 750 nm. In addition to the hemoglobin solution, which has a Soret band about 420 nm and two Q-bands between 500 and 600 nm, the measurements of other samples are the normal dispersion curve. The results are approximated by the Cauchy equation and Sellmeier equation, and the correlation coefficients are more than 0.997.

Drug levels in patients' bloodstreams vary among individuals and consequently therapeutic drug monitoring (TDM) is fundamental to controlling the effective therapeutic range. For TDM purposes, different analytical approaches have been used, mainly based on immunoassay, liquid chromatography-ultraviolet, liquid chromatography-mass spectrometry and liquid chromatography-tandem mass spectrometry (LC-MS/MS) methods. More recently a matrix-assisted laser desorption/ionisation method has been proposed for the determination of irinotecan levels in the plasma of subjects under therapy and this method has been cross-validated by comparison with data achieved by LC-MS/MS. However, to reach an effective point-of-care monitoring of plasma drug concentrations, a TDM platform technology for fast, accurate, low-cost assays is required. In this frame, recently the use of paper spray mass spectrometry, which is becoming a popular and widely employed MS method, has been proposed. In this paper we report the results obtained by the development of a paper spray-based method for quantitative analysis in plasma samples of imatinib, a new generation of anticancer drug. Preliminary experiments showed that poor sensitivity, reproducibility and linear response were obtained by the “classical” paper spray set-up. In order to achieve better results, it was thought of interest to operate in presence of a higher and more homogeneous electrical field. For this aim, a stainless steel needle connected with the high voltage power supply was mounted below the paper triangle. Furthermore, in order to obtain valid quantitative data, we analysed the role of the different equilibria participating to the phenomena occurring in paper spray experiments, depending either on instrumental parameters or on the chemical nature of analyte and solvents. A calibration curve was obtained by spiking plasma samples containing different amounts of imatinib (1) with known amounts of deuterated imatinib (1d3) as internal standard, with molar ratios [1] / [1d3] in the range 0.00–2.00. A quite good linearity was obtained ( R2 = 0.975) and some experiments performed on spiked plasma samples with known amounts of 1 confirmed the validity of this method.

The quantification of blood cells provides critical information about a patient's health status. Sophisticated analytical equipment, such as hematology analyzers, have been developed to perform these measurements, but limited-resource settings often lack the infrastructure required to purchase, operate, and maintain instrumentation. To address these practical challenges, paper-based microfluidic devices have emerged as a platform to develop diagnostic assays specifically for use at the point-of-care. To date, paper-based microfluidic devices have been used broadly in diagnostic assays that apply immunoassay, clinical chemistry, and electrochemistry techniques. The analysis of cells, however, has been largely overlooked. In this communication, we demonstrate a paper-based microfluidic device that enables the controlled transport of red blood cells (RBCs) and the measurement of the hematocrit—the ratio of RBC packed cell volume to total volume of whole blood. The properties of paper, device treatment, and device geometry affect the overall extent and reproducibility of transport of RBCs. Ultimately, we developed an inexpensive (US$0.03 per device) thermometer-styled device where the distance traveled by RBCs is proportional to the hematocrit. These results provide a foundation for the design of paper-based microfluidic devices that enable the separation and detection of cells in limited-resource settings.

Background: Dried blood spots (DBS) are rapidly gaining a foothold in the pharmaceutical industry. However, applications in exploratory drug discovery are limited mainly owing to method development time. The development of a generic DBS assay is presented with its application in serial microsampling from mice. Results: A generic ‘fit-for-purpose’ assay was developed on FTA® Elute, which allowed 90% of compounds tested to reach sensitivity levels of 1 ng/ml. A ten-time point serial mouse pharmacokinetic study was conducted using 20-µl microsamples and DBS. Application of generic ‘fit-for-purpose’ approach did not compromise data delivery or quality. Conclusions: Serial microsampling and DBS in exploratory mouse pharmacokinetic has been shown to provide superior data quality when compared with traditional plasma-based composite studies.

The wide scale application of dried blood spots (DBS) as a collection tool for low-cost HIV drug resistance testing requires a greater understanding of the accuracy of DBS for genotype analysis and the stability of DBS under various environmental conditions. Analysis of a 50microl DBS via a single amplicon, nested PCR-based in-house assay (the Burnet genotyping assay) showed an average nucleotide concordance of 98.9% with plasma samples, although only 65% of nucleotide mixtures detected in plasma were also detected within DBS. The analysis of three DBS resulted in the detection of a greater number of nucleotide mixtures (72 and 109 mixtures detected within one and three DBS, respectively, n=10). Two DBS extraction protocols (silica particle; NucliSENS, bioMerieux and spin column extraction; High Pure, Roche) were assessed and found to be equivalent (79% and 84% recovery success respectively, n=19). FTA Elute paper (Whatman) was an inferior DBS collection medium compared to Whatman 903 paper. DBS appeared relatively tolerant to multiple freeze/thaw cycles, with 79% of DBS subjected to ten freeze/thaw cycles successfully amplified compared to 93% of DBS defrosted once (n=14). High temperature (37 degrees C) and high humidity (>90%) substantially impaired DBS recovery within two weeks of storage (38%, n=8), whilst storage at -20 degrees C or 4 degrees C adequately preserved DBS for this period (100% recovery, n=8). Therefore, whilst DBS are suitable for HIV drug resistance surveillance, the use of multiple DBS may be required to ensure accurate detection of minor HIV quasispecies and short-term storage of samples at either 4 degrees C or -20 degrees C is recommended.

Pigment-coated, surface-sized, and surface-sized copy papers were treated with conventional corona, experimental pilot-scale plasma and laboratory-scale plasma. All the treatments increased the surface energy and oxidized the surface. The changes in the surface chemistry depended on treatment time and composition of the substrate surface. It seems that plasma especially oxidizes the long polymer chains, such as surfactants and other paper chemicals, on the surface of the paper. The ToF-SIMS distribution maps indicated that the pilot-scale treatment led to an uneven CO+ group distribution, whereas laboratory scale treatment gave a more even distribution of CO+ groups. In addition to chemical changes, topographical changes due to plasma treatment were detected. The RMS roughness increased for pigment-coated paper, whereas plasma treatment induced small nodules between the paper pigment particles with pigmented and surface-sized paper. The increase in roughness was also found to increase the wettability. This serves as a demonstration of roughness-induced increase of surface energy of the samples.

This work concerns the ageing effect of the atmospheric plasma and corona treatments when used to treat paper substrates. Pigment coated and surface sized papers were modified using two types of atmospheric plasma equipment; one at the pilot scale and one at the laboratory scale. In addition, the plasma treatments were compared to conventional corona treatment. Surface energy was estimated by contact angle measurements and surface chemistry by X-ray photoelectron spectroscopy (XPS) as a function of the time during three months. The treatments increased surface energy and oxidation level of surface for both papers. The ageing effect could be detected only in the surface energy, whereas the oxidation level remained stable during the twelve weeks. The decay in surface energy was faster during the first weeks of storage and subsequently leveled off leading to a permanent change. The permanent change was explained as a contribution of oxygen containing polar molecular groups, which were detected by XPS. The ageing effect was suggested to originate from already existing polar molecular groups, which have rotated on the surface by plasma-related process and then rotate back into the material in time. A part of the decay was also explained by the plasma cleaning model, in which the ageing effect occurred through re-contamination. Paper is a multicomponent system, where the constituents that have the lowest surface energy were suggested to migrate to paper surfaces.

We demonstrate capillarity-driven plasma separation from whole blood on simple paper-based H-channels. This methodology, unlike other reported techniques, does not necessitate elaborate and complex instrumentation, and the usage of expensive consumables. We believe that this technique will be ideally suited to be implemented in rapid and portable blood diagnostic devices designed to be operative at locations with limited resources.
