Abstract Maltotetraose amylase (Mta) catalyzes the hydrolysis of amylaceous polysaccharides into maltotetraose, which is an important functional sugar used in the food industry. However, the lack of efficient expression systems for recombinant Mta has hindered its scale-up production and application. In this study, a codon-optimized mta gene from Pseudomonas saccharophila was efficiently produced in Bacillus subtilis by optimizing the regulatory elements. First, a plasmid library containing 173 different signal peptide sequences placed upstream of mta gene was constructed, and transformed into B. subtilis strain WB800N(amyEΔ1) for high-throughput screening. The signal peptide yhcR was found to significantly enhance the secretion of Mta, reaching an activity of 75.4 U/mL in the culture medium. After optimization of the promoters, the Mta activity was further increased to 100.3 U/mL using a dual-promoter PHpaIIPamyE. Finally, the carbon sources and nitrogen sources for recombinant Mta production were optimized, yielding a highest Mta activity of 288.9 U/mL under the optimal culture conditions. The crude enzyme solution containing recombinant Mta produced a highest maltotetraose yield of 70.3% with 200 g/L of maltodextrin as the substrate. Therefore, the present study have demonstrated a high yield of Mta produced in B. subtilis, laying the foundation for large-scale Mta production and application.

​​​​​​​This study compares aerosol mixing-state parameters obtained via simultaneous measurements using DMA–CCNC, H(/V)TDMA, and DMA–SP2, shedding light on the impacts of primary aerosol emissions and secondary aerosol (SA) formation. The analysis reveals significant variations in mixing-state parameters among different techniques, with VTDMA and DMA–SP2 indicating that non-volatile particles mainly stem from black carbon (BC)-containing aerosols, while a substantial proportion of nearly hydrophobic aerosols originates from fossil fuel combustion and biomass-burning emissions. Synthesizing the results, some nearly hydrophobic BC-free particles were found to be cloud condensation nuclei (CCN)-inactive under the measured supersaturated conditions, likely from fossil fuel combustion emissions, while others were CCN-active, linked to biomass-burning emissions. Moreover, BC-containing aerosols emitted from fossil fuel combustion exhibit more external mixing with other aerosol components compared to those from biomass burning. Secondary nitrate and organic aerosol formation significantly affect aerosol mixing states, enhancing aerosol hygroscopicity and volatility while reducing heterogeneity among techniques. The study also highlights distinct physical properties of two resolved secondary organic aerosol factors, hinting at their formation through different mechanisms. These findings underscore the importance of comparing aerosol mixing states from different techniques as a tool for understanding aerosol physical properties from different sources and their responses to SA formation, as well as aiding in the exploration of SA formation mechanisms.

Protein content in food is an important indicator of the nutritional value of food, and it is also related to food safety and many other issues, therefore it is significant to accurately determine the protein content for the valuation of food nutrition and safety. The main elements in food include carbon, nitrogen, hydrogen and sulphur, of which nitrogen mainly comes from protein. The average nitrogen content in protein is 16%, so the conversion factor for “nitrogen-to-protein” is 6.25. Therefore, the protein content in food can be obtained by measuring the nitrogen content of food combined with “nitrogen-to-protein” conversion factor. The traditional methods of protein detection are Kjeldahl method, spectrophotometry, and Dumas combustion. The Kjeldahl method is time-consuming and consumes a large amount of reagents, which can cause environmental pollution. Spectrophotometry is susceptible to interference and the quantitative curve is not easy to draw. Dumas combustion is expensive in terms of consumables. In this paper, a platform of tube furnace-quadrupole mass spectrometer was set up to analyze the protein content in food. Food samples were heated at high temperatures in a tube furnace and combusted in an oxygen-rich environment. After the gas products were ionized, they were directly introduced into a quadrupole mass analyzer for detection, which eliminated the need for processes such as oxidation-reduction, adsorption-desorption by using the selection ion scan function of the mass spectrometer. Five kinds of food samples with different protein contents were selected as research object. Each food sample was weighed at 1 g. The food samples were fully combusted in tube furnace at high temperature (1 300 ℃). The gas products were infused through a drying and filtering tube to remove water and smoke, and then entered the EI ion source through a capillary for ionization. By electronic ionization, nitrogen element characteristic ions, N+, NO+, and NO2+ are generated. The peak intensity of the NO2+ ions were detected by the selection ion scan function of the quadrupole mass analyzer. A standard curve for nitrogen quantification is plotted to be y=115.64x+1 896.9 with the correlation coefficient (R2) of 0.99992 and the relative standard deviation (RSD) of 2.1%-6.1%. The nitrogen content can be calculated by using the peak intensity of NO2+ ions according to the standard curve. The experimental platform of tube furnace-mass spectrometer provides a green, rapid, accurate, and low-cost detection method for quantitative analysis of protein content in food.

Owing to the significant development in graphene, an increasing number of studies have been conducted to identify novel two-dimensional (2D) organic materials with Dirac cones and topological properties. Although a series of toy models based on specific lattice patterns has been proposed and demonstrated to possess a Dirac cone, realistic materials corresponding to the lattice models must be identified to achieve excellent properties for practical applications. To understand factors contributing to the rarity of 2D organic Dirac materials and provide guidance for identifying novel organic Dirac systems, we review recent theoretical studies pertaining to various 2D Dirac models and their corresponding organic Dirac materials, including the Haldane, Kagome, Libe, line-centered honeycomb, and Cairo pentagonal models. Subsequently, the corresponding structural and topological electronic properties are summarized. Additionally, we investigate the relationship between the existence of Dirac cones and their structural features, as well as the manner by which Dirac points emerge and propagate in these systems.

Nitrated aromatic compounds (NACs) and oxygenated derivatives of polycyclic aromatic hydrocarbons (OPAHs) play vital roles within brown carbon (BrC), influencing both climate dynamics and human health to a certain degree. The concentrations of these drug classes were analyzed in PM2.5 from an urban area in Beijing during the autumn and winter of 2017 and 2018. There were four heavy haze episodes during the campaign, two of which happened prior to heating and the other two during heating. During the entire course of sampling, the mean total concentrations of the nine NACs and the eight OPAHs were 1.2–263 and 2.1–234 ng m−3, respectively. The concentrations of both NACs and OPAHs were approximately 2–3 times higher in the heating period than before heating. For NACs, the relative molecular composition did not change significantly before and during heating, with 4-nitrocatechol and 4-nitrophenol demonstrating the highest abundance. For OPAHs, 1-naphthaldehyde was the most abundant species before and during heating, while the relative proportion of anthraquinone increased by more than twice, from 13 % before heating to 31 % during the heating. In Beijing's urban area during autumn and winter, significant sources of NACs and OPAHs have been traced back to automobile emissions and biomass burning activities. Interestingly, it was observed that the contribution from coal combustion increased notably during heating. It is worth noting that the secondary generation of BrC was important throughout the whole sampling period, which was manifested by the photochemical reaction before heating and the aqueous reaction during heating. It was further found that the haze in autumn and winter was nitrate-driven before heating and secondary organic carbon (SOC)-driven during heating, and the secondary formation of BrC increased significantly in pollution events, particularly during heating.

In this research, a pesticide-fertilizer combination (PFC) was fabricated using chlorantraniliprole and biochar. The PFC had a spherical shape, smooth surface, uniform particles, relatively dense structure and good compression resistance. The adsorption propertie of biochar to chlorantraniliprole was investigated. During the preparation of pelleted granules, the types and concentrations of core fillers and adhesives were screened and optimized. Besides, the safety to maize and effectiveness against fall armyworms of PFC had been investigated. Meanwhile, The PFC could promoted growth of maize significantly and control fall armyworm effectively. This work provides a promising approach to slow the release of chlorantraniliprole, which has a potential application to enhance the pesticides efficiency.

Plant growth regulators (PGRs) are chemical substances that imitate the functions of phytohormones to enhance the crop yield and the harvest process. Phenylurea-derived plant growth regulators are known for their excellent efficacy in promoting fruit growth, particularly in kiwifruit, grapes, and melons. Phenylurea derivatives represent one class of the highly efficient and versatile PGRs. Specifically, forchlorfenuron (CPPU, N-(2-chloro-4-pyridinyl)-N′-phenylurea) exhibits similar growth-regulating efficacy to cytokinins and has a significant impact on the plant growth and the crop yield. As a result, there is growing interest in exploring the incorporation of various phenylurea moieties into agrochemicals to enhance their regulatory properties on crops. This review aims to provide a comprehensive overview on representative synthetic approaches for phenylurea derived PGRs. Additionally, we provide our perspective on the future development in this active research field.

The partitioning of semi-volatile organic compounds (SVOCs) between gas and particle phases plays a crucial role in their long-range transport and health risk assessment. However, the accurate prediction of the gas–particle (G–P) partitioning quotient (KP′) remains a challenge, especially for the light-molecular-weight (LMW) SVOCs due to their upward deviation from equilibrium state. In this study, the phenomenon with the influence of gaseous degradation on G–P partitioning was observed. Based on the diurnal study of concentrations and KP′ values for methylated polycyclic aromatic hydrocarbons (Me-PAHs), it was found that the KP′ values of methylated naphthalenes (Me-Naps; one type of LMW SVOC) during the daytime were higher than during the nighttime, and the regression lines of log KP′ versus log KOA (octanol–air partitioning coefficient) for daytime and nighttime were non-overlapping, which were different from other Me-PAHs. Compared with other diurnal influencing factors, the higher gaseous degradation of Me-Naps in the daytime than in the nighttime should partially explain their special diurnal variation in KP′, which provided a new explanation for the non-equilibrium behavior of KP′ for LMW SVOCs. Moreover, the influence of gaseous degradation on the deviation of KP′ from equilibrium state was deeply studied based on the steady-state G–P partitioning model considering particulate proportion in emission (ϕ0). The increasing times of KP′ influenced by the gaseous degradation deviated from equilibrium state can be calculated by 1 + 13.2ϕ0 × kdeg (kdeg, gaseous degradation rate). The increase in KP′ along with the increase in kdeg proved that higher gaseous degradation in the daytime could increase the KP′ value. Furthermore, an amplification in KP′ ranging from 1.11 to 5.58 times (90 % confidence interval: 1.01 to 14.4) under different ϕ0 values (0 to 1) in the temperature range of −50 to 50 °C was estimated by the Monte Carlo analysis. In summary, it can be concluded that the influence of gaseous degradation should also be considered in the G–P partitioning models of SVOCs, especially for the LMW SVOCs, which provided new insights into the related fields.

Abstract In this study, we firstly established and verified a method by ultra performance liquid chromatography tandem mass spectrometry (UPLC-MS/MS) for the analysis of vilazodone and its metabolite M10 in rat plasma, then this method was used to explore the pharmacokinetics of vilazodone and M10 present or absence of 80 mg/kg bergenin in rats. Protein precipitation with acetonitrile was used to prepare the samples in this research. The mobile phase for liquid chromatography was consisted of 0.1% formic acid aqueous solution and acetonitrile. Brexpiprazole was used as the internal standard (IS), and the multiple reaction monitoring (MRM) mode was used for detection. The verification items required by the US Food and Drug Administration (FDA) guidelines such as selectivity, sensitivity, linearity, stability, recovery and matrix effect of this method were all met the standards. Besides, rats were used to explore the drug-drug interaction between vilazodone and bergenin, which were divided into two groups, and separately gavaged with the same-volume of carboxymethyl cellulose sodium (CMC-Na) solution and 80 mg/kg bergenin, respectively. The results showed that bergenin significantly affected the metabolism of vilazodone. It suggested that there was a potential drug-drug interaction between bergenin and vilazodone in rats. In clinical application, we should pay attention to the dose of vilazodone when in combination with bergenin.

It is crucial to explore the earth-abundant, high-efficient and stable electrocatalysts suitable for accelerating water splitting kinetics of oxygen evolution reaction (OER) in alkaline electrolytes. Herein, FexCo1-xS2 (x = 0, 0.25, 0.5, 0.75 and 1) nanoparticles were fabricated via hydrothermal method and sulfidizing. Remarkably, the FeS2 delivers the most outstanding OER activity among prepared catalysts, with the overpotential of 240 mV for 10 mA/cm2, ultra-low Tafel slope of 45 mV/decades and high stability for 16 h. XRD, SEM, HRTEM, Raman and XPS characteristics before and OER process, TOF and electrochemical results indicate that surface reconstruction, strong combination of Fe-S and uniform dispersion of particles endow FeS2 with excellent catalytic property. The strong combination of Fe-S bond and uniform dispersion of FeS2 facilitates electron transfer. And the reconstructed crystalline-amorphous Fe2O3/FeOOH with rough surface may have a low energy barrier for the elementary reaction of O*→OOH*, which is responsible for boosting the generation of O2. In the practical two-electrode overall water splitting system in 1 M KOH, the FeS2/NF//Pt/C/NF system only needs an ultra-low cell voltage of 1.46 and 1.642 V to generate the current densities of 10 and 50 mA/cm2, respectively.

Mosquitoes are the vectors of numerous diseases such as malaria, Dengue and Zika. Controlling mosquito vectors is essential for preventing and managing diseases transmitted by mosquitoes. Larvicides target larvae within their breeding habitats before they mature into adult mosquitoes and disperse. In this study, using α-hexyl cinnamaldehyde (HCA) as a lead compound, three series of derivatives comprising a total of 78 compounds were designed and synthesized. The larvicidal activities of these compounds against Aedes albopictus were evaluated, resulting in the identification of ten promising compounds. Among them, furan-3-ylmethyl cinnamate (compound 2v) and (E)-allyl 3-(4-chlorophenyl)acrylate (compound 4c) exhibited the lowest LC50 values (8.0 ppm and 7.2 ppm respectively). In a 96-h acute toxicity assay conducted on zebrafish, 2v showed no or very low toxicity with an LC50 higher than 100 ppm. To elucidate their mode of action, morphological and histopathologic analyses were performed, and the effects of the compounds on the acetylcholinesterase (AChE) and phosphatase activities were evaluated. Results suggested that the active HCA derivatives induce injury to intestinal epithelial cells by inhibiting phosphatase activity, ultimately resulting in the mortality of mosquito larvae.

ObjectiveThe aim of the present study was to carry out a systematic research on bitterness quantification to provide a reference for scholars and pharmaceutical developers to carry out drug taste masking research. Significance: The bitterness of medications poses a significant concern for clinicians and patients. Scientifically measuring the intensity of drug bitterness is pivotal for enhancing drug palatability and broadening their clinical utility.MethodsThe current study was carried out by conducting a systematic literature review that identified relevant papers from indexed databases. Numerous studies and research are cited and quoted in this article to summarize the features, strengths, and applicability of quantitative bitterness assessment methods.ResultsIn our research, we systematically outlined the classification and key advancements in quantitative research methods for assessing drug bitterness, including in vivo quantification techniques such as traditional human taste panel methods, as well as in vitro quantification methods such as electronic tongue analysis. It focused on the quantitative methods and difficulties of bitterness of natural drugs with complex system characteristics and their difficulties in quantification, and proposes possible future research directions.ConclusionThe quantitative methods of bitterness were summarized, which laid an important foundation for the construction of a comprehensive bitterness quantification standard system and the formulation of accurate, efficient and rich taste masking strategies.

In this work study, a comparative analysis was undertaken to investigate investigation into the cavitation erosion (CE) and corrosion behavior of laser powder bed fusion (LPBF) TC4 and as-cast TC4 in 0.6 mol/L NaCl solution. Relevant results indicated that LPBF TC4 revealed a rectangular checkerboard-like pattern with a more refined grain size compared to as-cast TC4. Meanwhile, LPBF TC4 surpassed its as-cast counterpart in CE resistance, demonstrating approximately 2.25 times lower cumulative mass loss after 8 h CE. The corrosion potential under alternating CE and quiescence conditions demonstrated that both LPBF TC4 and as-cast TC4 underwent a rapid potential decrease at the initial stages of CE, while a consistent negative shift in corrosion potential was observed with the continuously increasing CE time, indicative of a gradual decline in repassivation ability. The initial surge in corrosion potential during the early CE stages was primarily attributed to accelerated oxygen transfer. As CE progressed, the significant reduction in corrosion potential for both LPBF TC4 and as-cast TC4 was attributed to the breakdown of the passive film. The refined and uniform microstructure in LPBF TC4 effectively suppresses both crack formation and propagation, underscoring the potential of LPBF technology in enhancing the CE resistance of titanium alloys. This work can provide important insights into developing high-quality, reliable, and sustainable CE-resistant materials via LPBF technology.

The waterglass or St o¨ ber method is commonly used to synthesize spherical colloidal silica; however, these methods have some disadvantages, such as complicated processes for the removal of sodium ions and expensive and energy-consuming raw materials such as tetraethoxysilane (TEOS). In this study, size-controlled spherical colloidal silica was synthesized from silicon metal at room temperature using an ultrasound process with hydrazine monohydrate as the solvent. Silicon metal dissolves easily in hydrazine monohydrate under ultrasound irradiation, and spherical colloidal silica can be synthesized by adding alcohol to this precursor solution. By changing the concentration or type of alcohol, size-controlled colloidal silica 20–200 nm in size could be easily obtained. In addition, finer and more monodisperse particles were produced by low-frequency ultrasound irradiation, which had a higher stirring effect at the particle formation stage. The present method is effective because size-controlled colloidal silica can be synthesized at room temperature using only silicon metal, hydrazine, and alcohol as raw materials, without complicated processes or expensive and energy-consuming raw materials such as TEOS or tetramethoxysilane (TMOS).

This study aimed to investigate the effects of ultrasonic treatment at different powers on the physicochemical properties, microstructure and quercetin delivery capacity of fermentation-induced soy protein isolate emulsion gel (FSEG). The FSEG was prepared by subjecting soy protein isolate (SPI) emulsion to ultrasonic treatment at various powers (0, 100, 200, 300, and 400 W), followed by lactic acid bacteria fermentation. Compared with the control group (0 W), the FSEG treated with ultrasound had higher hardness, water holding capacity (WHC) and rheological parameters. Cu3VSe4 material has become a new kind of photovoltaic absorber due to its optical bandgap matching light-absorbing, abundant elements in crust, and high light absorption coefficient. Here, we prepared Cu3VSe4 thin films by amine-thiol method and investigated the preparation process of excellent Cu3VSe4 absorption layer for the first time. The effects of various Cu/V ratios on the morphology, structure, and electrical properties of the films are studied. XRD and Raman results show that pure-phase Cu3VSe4 films are obtained with the Cu/V ratios lower than 2.9. SEM results simply adjusting the Cu/V ratio cannot obtain a compact selenized Cu3VSe4 thin films. The best crystallinity of thin film with 2.8 of Cu/V ratio is named as Cu2.8VSe4 and selected as the representative. Though optimizing the selenization conditions, a dense morphology and good electrical properties of the absorption layer can be prepared under 570 °C of the high-temperature process. Our findings can lay the favorable foundation for high-efficiency Cu3VSe4 devices.

Stacking engineering is a popular method to tune the performance of two-dimensional materials for advanced applications. In this work, Jansu MoSSe and WSSe monolayers are constructed as a van der Waals (vdWs) heterostructure by different stacking configurations. Using first-principle calculations, all the relaxed stacking configurations of the MoSSe/WSSe heterostructure present semiconductor properties while the direct type-II band structure can be obtained. Importantly, the Z-scheme charge transfer mode also can be addressed by band alignment, which shows the MoSSe/WSSe heterostructure is an efficient potential photocatalyst for water splitting. In addition, the built-in electric field of the MoSSe/WSSe vdWs heterostructure can be enhanced by the S–Se interface due to further asymmetric structures, which also results in considerable charge transfer comparing with the MoSSe/WSSe vdWs heterostructure built by the S–S interface. Furthermore, the excellent optical performances of the MoSSe/WSSe heterostructure with different stacking configurations are obtained. Our results provide a theoretical guidance for the design and control of the two-dimensional heterostructure as photocatalysts through structural stacking.
