The review presents: a) a brief description of the disease; b) a summary of the most important metabolic targets so far identified in Trypanosome cruzi (T. cruzi) along with corresponding inhibitor compounds; c) the current state of knowledge on the trypanothione reductase system of trypanosomatids with reference to oxidative stress defenses; d) detailed discussions on T. cruzi trypanothione reductase inhibitors such as nitrofuranes, naphthoquinones and phenothiazines. As yet, the chemotherapy of Chagas' disease remains an unsolved problem. Further search for new drugs must continue by means of nucleating existing chemotherapy efforts.

Fenton systems (H2O2/Fe(II) or H2O2/Cu(II)) inhibited Trypanosoma cruzi and Crithidia fasciculata topoisomerase I activity. About 61-71% inactivation was produced by 25 mM Fe(II) or Cu(II) with 3 mM H2O2. Thiol compounds and free radicals scavengers prevented the Fenton systems effects, depending on the topoisomerase assayed. With the T. cruzi enzyme, reduced glutathione, DL-dithiothreitol, cysteine and N-acetyl-L-cysteine entirely prevented the effect of the H2O2/Fe(II) system, mannitol protected 37%, whereas histidine and ethanol were ineffective. With C. fasciculata topoisomerase, reduced glutathione, DL-dithiothreitol and N-acetyl-L-cysteine protected 100%, cysteine, histidine and mannitol protected 28, 34 and 48% respectively, whereas ethanol was ineffective. With the H2O2/Cu(II) system and T. cruzi topoisomerase, DL-dithiothreitol and histidine protected 100% and 60%, respectively but the other assayed protectors were less effective. Similar results were obtained with the C. fasciculata enzyme. Topoisomerase inactivation by H2O2/Fe(II) or H2O2/Cu(II) systems was irreversible since they were not reverted by the more effective enzyme protectors. It is suggested that topoisomerases could act either as scavengers of "reactive oxygen species" (ROS) generated by Fenton systems or bind the corresponding metal ions, whose redox cycling would generate reactive oxygen species "in situ".

Phenothiazine cation radicals (PTZ+*) irreversibly inactivated Trypanosoma cruzi dihydrolipoamide dehydrogenase (LADH). These radicals were obtained by phenothiazine (PTZ) peroxidation with myeloperoxidase (MPO) or horseradish peroxidase (HRP/H2O2) systems. LADH inactivation depended on PTZ structure and incubation time. After 10 min incubation of LADH with the MPO-dependent systems, promazine, trimeprazine and thioridazine were the most effective; after 30 min incubation, chlorpromazine, prochlorperazine and promethazine were similarly effective. HRP-dependent systems were equally or more effective than the corresponding MPO-dependent ones. Chloro, trifluoro, propionyl and nitrile groups at position 2 of the PTZ ring significantly decreased molecular activity, specially with the MPO/H2O2 systems. Comparison of inactivation values for LADH and T. cruzi trypanothione reductase demonstrated a greater sensitivity of LADH to chlorpromazine and perphenazine and a 10-fold lower sensitivity to promazine, thioridazine and trimeprazine. Alkylamino, alkyl-piperidinyl or alkyl-piperazinyl groups at position 10 modulated PTZ activity to a limited degree. Production of PTZ+* radicals was demonstrated by optical and ESR spectroscopy methods. PTZ+* radicals stability depended on their structure as demonstrated by promazine and thioridazine radicals. Thiol compounds such as GSH and N-acetylcysteine, L-tyrosine, L-tryptophan, the corresponding peptides, ascorbate and Trolox, prevented LADH inactivation by the MPO/H2O2/thioridazine system, in close agreement with their action as PTZ+* scavengers. NADH (not NAD+) produced transient protection of LADH against thioridazine and promazine radicals, the protection kinetics being affected by the relatively fast rate of NADH oxidation by these radicals. The role of the observed effects of PTZ radicals for PTZ cytotoxicity is discussed.

Peroxidase/H2O2/phenothiazine systems irreversibly inhibit Trypanosoma cruzi dihydrolipoamide dehydrogenase (LADH). Inactivation of the parasite enzyme depended on (a) phenothiazine structure; (b) peroxidase nature; (c) incubation time and (d) the presence of a cation radical scavenger. With the myeloperoxidase/H2O2/system, promazine, trimeprazine, thioridazine, promethiazine, prochlorperazine, chlorpromazine and perphenazine were the most effective derivatives out of twelve phenothiazines studied. An electronegative substituent at position 2 of the phenothiazine ring such as Cl, or trifluoromethyl, propionyl and nitrile groups decreased or nullified phenothiazine activity. Myeloperoxidase/H2O2/, horseradish peroxidase/H2O2/, and myoglobin/H2O2/systems activated phenothiazines producing the corresponding cation radicals, myeloperoxidase being the most selective one with respect to phenothiazine structure. The myoglobin/H2O2/system activated phenothiazines that were scarcely active or inactivate with the MPO/H2O2/system, such as the trifluoromethyl derivatives. Production of phenothiazine cation radicals was demonstrated by optical spectroscopy. Phenothiazine cation radical stability depended on their structure as illustrated by promazine and thioridazine. Thiol compounds (GSH, N-acetyl-cysteine and penicillamine), aromatic aminoacids (L-tyrosine, L-tryptophan, and the corresponding peptides) and ascorbate scavenged phenothiazine cation radicals, thus preventing LADH inactivation. Comparison of the summarized phenothiazine effects with those of phenothiazines on T. cruzi suggest the role of cation radicals in phenothiazines chemotherapeutic actions.

Peroxidase/H2O2/phenothiazine systems produced irreversible inhibition (inactivation) of Trypanosoma cruzi trypanothione reductase (TR). The enzyme inactivation depended on (a) the incubation time of TR with the peroxidase/H2O2/phenothiazine system; (b) the peroxidase nature and (c) the phenothiazine structure. With the more effective peroxidase/H2O2/phenothiazine systems, TR inactivation kinetics presented a relatively fast initial phase, lasting for about 10 min, in which most of the enzyme activity disappeared. This phase was followed by a slower one and, after 30 min incubation, TR was totally inactivated. Three peroxidases were assayed as catalysts of TR inactivation: the horseradish peroxidase (HRP), leukocyte myeloperoxidase (MPO) and modified myoglobin (Mb). Under comparable experimental conditions, the peroxidase system activity decreased in the given order. With HRP systems, 10 microM Thioridazine (TRDZ), Promazine (PZ), Trimeprazine (TMPZ), Prochlorperazine (PCZ), Propionylpromazine (PPZ), Chlorpromazine (CPZ) and Perphenazine (PFZ), produced 95-100% inactivation of TR. With the MPO/H2O2 systems, PZ. TRDZ and TMPZ were the most effective. Under similar experimental condition, the Mb/H2O2/PZ,/TMPZ, /TRDZ and CPZ systems effectively inactivated TR. The presence of alkylamino, piperazinyl, or piperidinyl groups in PTZ N atom (position 10) and -Cl, -CF3, -SCH3, COCH2CH3 and -CN in position C2 exerted significant influence on phenothiazine activity. Glutathione (GSH) prevented TR inactivation by the HRP/H2O2/PZ and MPO/H2O2/PZ systems. The HRP/H2O2 and MPO/H2O2/phenothiazines systems generated the corresponding cationic radicals (FTZ.+) the stability of which was limited by their conversion into phenothiazine-sulfoxides (PTZ-SO). The latter ones were inactive on TR. GSH rapidly reacted with PTZ+.; thus producing cation radical detoxication. These reactions fit in well with GSH protection of TR against the peroxidase/H2O2/phenothiazine system, as well as with the FTZ.+ role in phenothiazine cytotoxicity.

Dihydrolipoamide dehydrogenase (LADH) from Trypanosoma cruzi was inactivated by treatment with myeloperoxidase (MPO)-dependent systems. With MPO/H2O2/NaCl, LADH lipoamide reductase and diaphorase activities significantly decreased as a function of incubation time. Iodide, bromide, thiocyanide and chloride effectively supplemented the MPO/H2O2 system, KI and NaCl being the most and the least effective supplements, respectively. LADH inactivation by MPO/H2O2/NaCl and by NaOCl was similarly prevented by thiol compounds such as GSH, L-cysteine, N-acetylcysteine, penicillamine and N-(2-mercaptopropionyl-glycine) in agreement with the role of HOCI in LADH inactivation by MPO/H2O2/NaCl. LADH was also inactivated by MPO/NADH/halide, MPO/H2O2/NaNO2 and MPO/NADH/NaNO2 systems. Catalase prevented the action of the NADH-dependent systems, thus supporting H2O2 production by NADH-supplemented LADH. MPO inhibitors (4-aminobenzoic acid hydrazide, and isoniazid), GSH, L-cysteine, L-methionine and L-tryptophan prevented LADH inactivation by MPO/H2O2/NaNO2. Other MPO systems inactivating LADH were (a) MPO/H2O2/chlorpromazine; (b) MPO/H2O2/monophenolic systems, including L-tyrosine, serotonin and acetaminophen and (c) MPO/H2O2/di- and polyphenolic systems, including norepinephrine, catechol, nordihydroguaiaretic acid, caffeic acid, quercetin and catechin. Comparison of the above effects and those previously reported with pig myocardial LADH indicates that both enzymes were similarly affected by the MPO-dependent systems, allowance being made for T. cruzi LADH diaphorase inactivation and the greater sensitivity of its LADH lipoamide reductase activity towards the MPO/H2O2/NaCl system and NaOCl.

Dihydrolipoamide dehydrogenase (LADH) from Trypanosoma cruzi, the causative agent of Chagas' disease, was inactivated by treatment with myeloperoxidase (MPO)-dependent systems. LADH lipoamide reductase and diaphorase activities decreased as a function of incubation time and composition of the MPO/H2O2/halide system, a transient increase preceding the loss of diaphorase activity. Iodide, bromide, thiocyanide and chloride were effective components of MPO/H2O2 or MPO/NADH systems. Catalase prevented LADH inactivation by the MPO/NADH/halide systems in agreement with H2O2 production by NADH-supplemented LADH. Thiol compounds (L-cysteine, N-acetylcysteine, penicillamine, N-(2-mercaptopropionylglycine) and Captopril prevented LADH inactivation by the MPO/H2O2/NaCl system and by NaOCl, thus supporting HOCl as agent of the MPO/H2O2/NaCl system. MPO/H2O2/NaNO2 and MPO/NADH/NaNO2 inactivated LADH, the reaction being prevented by MPO inhibitors and thiol compounds. T. cruzi LADH was affected by MPO-dependent systems like myocardial LADH, allowance being made for the variation of the diaphorase activity and the greater sensitivity of the T. cruzi enzyme to MPO/H2O2/halide systems.

ATP and ADP levels were determined in Crithidia fasciculata and Trypanosoma cruzi. The nucleotide levels in crithidia or epimastigotes at the stationary phase of growth were, in nmol/10(8) cells, 15-40, and 3-7, for ATP and ADP, respectively. Incubation with the lipophilic o-naphthoquinones CG 8-935, CG 9-442 and CG 10-248 or the anti-chagasic nitrofuran nifurtimox, significantly decreased ATP level, with non-significant variations of the ADP level. The kinetics of ATP variation showed an initial 1-2 h lag and the diminution of the ATP level reached maximum value after 4-6 h incubation. Addition of L-glutamate or D-glucose as energy sources produced 2- or 3-fold increase of ATP level, after incubation the protozoa for 4-6 h with the corresponding substrates. Quinones and nifurtimox strongly depressed D-glucose or L-glutamate effects. Buthionine sulfoximine an inhibitor of glutathione biosynthesis, enhanced the effect of nifurtimox on ATP level in Crithidia fasciculata. It is concluded that by inhibiting ATP synthesis, either directly or-through oxygen radicals, the assayed drugs produced their cytotoxic action.

ATP hydrolysis by Crithidia fasciculata mitochondrial ATPase was studied through the use of submitochondrial particles and the soluble enzyme. (a) ATP hydrolysis by these preparations was inhibited by spermine but not by spermidine or putrescine. (b) The ATPase activity inhibited by spermine was partially recovered when excess Mg2+ was added to the reaction mixture. (c) When polyamines were present throughout the preparation of the mitochondrial membranes, the membrane-bound ATPase was irreversibly inactivated. The polyamine effect decreased in the order spermine > putrescine > spermidine. (d) The spermidine or putrescine pre-treated membranes oxidized succinate at a faster rate than the control ones. (e) Trypanosoma cruzi F-ATPase was inhibited by polyamines in the same manner as the C. fasciculata enzyme.

1. A high-affinity (Ca2+ + Mg2+)-ATPase and a low-affinity Mg(2+)-ATPase were identified in the 105,000 g fraction from epimastigote forms of Trypanosoma cruzi, the agent of Chagas' disease (Tulahuen strain). 2. Activities were conserved after enzyme solubilization with deoxycholate. 3. The Ca(2+)-stimulated ATPase activity was (a) lower than that of the Mg(2+)-ATPase; (b) inhibited by p-chloromercurobenzoate and orthovanadate and (c) insensitive to oligomycin. 4. Optimal stimulation by Ca2+ was observed at pH 6.5-6.8 in the presence of 1 mM MgCl2 and 0.1 M KCl. 5. The Mg(2+)-ATPase was insensitive to p-chloromercurobenzoate and orthovanadate and did not require KCl for activity. 6. Kinetic analysis of the (Ca2+ + Mg2+)-ATPase yielded a half-maximal stimulating concentration of 1.1 microM for Ca2+ and a Km of 66 microM for ATP. 7. The (Ca2+ + Mg2+)-ATPase clearly differed from the Ca(2+)- or Mg(2+)-ATPases previously characterized in the same strain of T. cruzi (Frasch et al., 1978; Comp. Biochem. Physiol. 60B, 271-275).

Several 4-(aminomethylisoxazolyl)-1,2-naphthoquinones inhibited growth and DNA synthesis in Trypanosoma cruzi and stimulated O2 uptake and O2-. generation by the parasite epimastigotes and their mitochondrial and microsomal membranes; these results support the idea that oxygen radicals play a role in quinone toxicity. Maximal effects on respiration and O2-. generation were observed with antimycin-inhibited cells. Similar results as well as stimulation of H2O2 production were obtained with Crithidia fasciculata despite the presence of catalase in this organism.

The small circular components (mini-circles) from Trypanosoma cruzi kinetoplast DNA (kDNA) were cloned in the plasmid vector pBR325. These clones have been used before to demonstrate the rapid evolution of mini-circle subpopulations (S anchez , D.O., Frasch , A.C.C., Carrasco , A.E., Gonzalez Cappa , S.M., Isola , E. and Stoppani , A.O.M. (1984) Mol. Biochem. Parasitol ., in the press). We have now analyzed the cloned molecules and used them to study some structural characteristics of T. cruzi mini-circles and their distribution in total kDNA restriction endonuclease digests. Most molecules partially conserved TaqI, HaeIII and HapII site clusters (constant regions) separated by one-quarter of the total mini-circle length, also detected in total kDNA digests. In addition, in one of the cloned mini-circles, the constant region was present only once, instead of four times as expected. Outside the conserved regions, the mini-circles diverged enough so that no cross-hybridization took place even under relaxed conditions. The recombinant molecules were used to probe total kDNA digests from T. cruzi. Some of them hybridized with most restriction endonuclease kDNA fragments, while one cloned mini-circle ( pTck -14) detected only its homologous subpopulation. The mini-circles detected with the latter probe proved to be nearly homogeneous, and were present in the proportion of 1/20 molecules. These results suggest that some of the generated molecules might have acquired a higher replication rate, giving rise to the homogeneous subpopulation detected. Further mutations, insertions and/or deletions, together with recombination between molecules, would bring this process to an end.

Phosphorylating mitochondrial membranes were obtained from Trypanosoma cruzi culture (epimastigote) forms. Using ADP as phosphate acceptor and succinate, sn-glycerol-3-phosphate, L-malate or ascorbate plus tetramethyl-p-phenylenediamine (TMPD) as oxidizable substrates, energy coupling sites II and III were detected, with respiratory control values in the range of 2.8-2.0. Carbonyl cyanide m-chlorophenylhydrazone and sonication uncoupled the respiratory control mechanism. Antimycin and cyanide partially inhibited succinate, sn-glycerol-3-phosphate and L-malate oxidation, while cyanide totally inhibited ascorbate + TMPD oxidation by the mitochondrial preparation. Succinate oxidation was inhibited by malonate and oxalacetate, but this latter was only effective with sonicated mitochondria. At variance with other substrates, NADH oxidation was not controlled by ADP concentration or inhibited by antimycin or cyanide. Rotenone failed to inhibit electron transfer in T. cruzi mitochondria.

Incubation of Trypanosoma cruzi epimastigotes with beta-lapachone (3,4-dihydro-2,2-dimethyl-2H-naphtho[1,2-b]pyran-5,6-dione), a lipophilic o-quinone, produced inhibition of [3H]thymidine, [3H]uridine, and L-[3H]leucine incorporation into DNA, RNA, and protein, respectively. With 1.6 microM beta-lapachone, DNA synthesis was preferentially inhibited. The inhibition was irreversible, and time and concentration dependent. Other effects of beta-lapachone were (a) inhibition of 3H precursor uptake into epimastigotes, (b) exaggerated degradation of DNA, RNA, and protein, (c) increased unscheduled synthesis of DNA, and (d) increased number of strand breaks in nuclear and kinetoplast DNA. DNA damage by 1.6 microM beta-lapachone was repaired by reincubating the drug-treated epimastigotes in fresh medium for 24 h, but with 7.8 microM beta-lapachone DNA damage was irreversible. The p-quinone isomer alpha-lapachone (3,4-dihydro-2,2-dimethyl-2H-naphtho[2,3-b]pyran-5,10-dione), was less effective than beta-lapachone, especially on DNA and RNA synthesis, and did not stimulate unscheduled DNA synthesis. Since beta-lapachone redox cycling in T. cruzi generates oxygen radicals while alpha-lapachone does not (A. Boveris, R. Docampo, J. F. Turrens, and A. O. M. Stoppani (1978) Biochem. J. 175, 431-439), the summarized results support the hypothesis that oxygen radicals contribute to beta-lapachone toxicity in T. cruzi.

Inactivation of Trypanosoma cruzi mitochondrial ATPase by oxygen radicals, generated by redox cycling of the ascorbate-Cu system (Cataldi de Flombaum, M.A. and Stoppani, A.O.M. (1986) Biochem. Int. 12, 785-793), involves oxidation of the enzyme thiols, as indicated by the competitive kinetics obtained with p-chloromercuribenzoate, a selective SH-reagent. Dithiothreitol prevented the ascorbate-Cu effect but did not reactivate the enzyme. Non-competitive kinetics were obtained with ascorbate-Cu and increasing MgATP concentration, or with phenylglyoxal, as second inhibitor. Since phenylglyoxal reacts with arginyl residues at the ATPase hydrolytic site, these results suggest that the oxgen-sensitive thiols were located outside the hydrolytic site.

Antimycin A and 2-heptyl-4-hydroxyquinoline N-oxide, two specific inhibitors of the b-c1 segment of the respiratory chain, affected the respiration of Trypanosoma cruzi epimastigote forms. The half-maximum inhibitory concentrations were about 0.05 and 4.0 micrograms/mg cells (dry wt.), respectively. The maximum effect of antimycin (about 80% inhibition of respiration) was at about 0.1 microgram antimycin/mg cells. Differential spectrophotometry of T. cruzi epimastigotes in the presence of antimycin, cyanide (or sulfide) and uncouplers, revealed the presence of functional cytochromes aa3, b and c558. In the stationary growth phase respiration by T. cruzi was completely inhibited by cyanide and effectively inhibited by sulfide, but in the exponential growth phase respiration was about 20% insensitive to 5 mM cyanide. Cyanide- and antimycin-insensitive respiration was completely inhibited by salicylhydroxamic acid (2 mM). Antimycin inhibited the operation of the tricarboxylic acids cycle in T. cruzi, as shown by the lesser production of 14CO2 and by the modification of 14C distribution in epimastigotes incubated with [1-14C]glucose, [2-14C]acetate or NaH14CO3. The inhibition of electron transport by antimycin increased the rate of the fumarate reductase reaction, an alternative electron pathway for the oxidation of reduced pyridine nucleotides. Addition of carbonyl cyanide 3-chlorophenylhydrazone to epimastigotes increased the rate of respiration and promoted the oxidation of reduced cytochrome b components, thus showing that these components are subject to respiratory (acceptor) control. Pentachlorophenol similarly affected the cytochrome b redox level but did not modify the rate of respiration. The uncouplers released N,N'-dicyclohexylcarbodiimide inhibition of respiration, and uncouplers and cyanide significantly decreased the ATP level in epimastigotes. The combined effects of the assay inhibitors on respiration, cytochrome b redox level, ATP content and energy charge confirmed the operation of oxidative phosphorylation in T. cruzi epimastigotes. Antimycin, uncouplers and N,N'-dicyclohexylcarbodiimide inhibited growth of T. cruzi, thus proving the essential role of oxidative phosphorylation for the parasite.

Nifurtimox and benznidazole have trypanostatic actions in vitro and inhibit the incorporation of [3H] thymidine, [3H] uridine and L-[3H] leucine in T. cruzi macromolecules. The effect of nifurtimox may be explained by (a) direct inhibition of nucleic acid biosynthesis, or (b) generation of the oxygen radicals in T. cruzi and therefore, only mechanism (a) should be valid. In order to obtain more information on the action of these drugs on T. cruzi, in the present study we examined the effect of nifurtimox and benznidazole on DNA, RNA and protein turnover in epimastigote (culture) forms of the parasite. Complementary experiments were performed with beta-lapachone that, like nifurtimox, generates oxygen radicals in T. cruzi. Epimastigotes (Tulahuen strain) at the exponential-phase of growth were cultured with [3H] thymidine, [3H] uridine or L-[3H] leucine to label DNA, RNA and protein, respectively. After incubation, the cells were washed free of radioactive precursor, resuspended in fresh medium and reincubated at 30 degrees C with nifurtimox (10 or 100 microM), benznidazole (38 or 380 microM) or beta-lapachone (1.6 or 7.8 microM), for 1-3 hours. Controls were incubated without drug. At one hour time intervals, sampler were taken, washed free of medium and filtered through 0.45 microns Metricel filters. The filters were washed with 10% trichloroacetic acid to remove the acid soluble material, and after drying, the radioactivity incorporated in DNA, RNA and protein was counted with a scintillation counter. The results show that after elimination of the labelled precursors, 3H activity in DNA, RNA and protein decayed as a function of the time of incubation. Nifurtimox, benznidazole and beta-lapachone, stimulated in all cases they decay of the incorporated radioactivity. Calculation of "half-life" values for DNA, RNA and protein(s) indicated that nifurtimox and beta-lapachone exerted their greatest effects on DNA while benznidazole increased the decay of DNA, RNA and protein to about the same extent. Taking into account the effects of nifurtimox and beta-lapachone on DNA stability, specific lesions (single-strand breaks) were investigated in DNA from control, nifurtimox, benznidazole or beta-lapachone treated epimastigotes. The number of single-strand breaks was (per 10(6)b) 25 with 100 microM nifurtimox, 1.4 with 380 microM benznidazole and 45 with 7.8 microM g-lapachone. Interestingly enough, after reincubation of nifurtimox-damaged epimastigotes in fresh medium for 24 hours, recovery of DNA lesions could be observed.(ABSTRACT TRUNCATED AT 400 WORDS)

Nifurtimox and benznidazole, two nitroheterocyclic drugs, inhibited DNA, RNA and protein synthesis, stimulated macromolecular degradation, and stimulated unscheduled DNA synthesis in Trypanosoma cruzi (Tulahuen strain). Significant differences in the mode of action of these drugs could be established and, in every case, nifurtimox was more active than benznidazole. The inhibition of macromolecular synthesis varied with drug concentration, precursor and incubation time. Nifurtimox effect was time dependent and irreversible. When assayed on macromolecular degradation, nifurtimox was more effective on DNA and protein than on RNA, while benznidazole displayed almost the same activity on DNA, RNA and protein. Labeling of RNA with [3H]uridine in the presence of nifurtimox followed atypical kinetics since, depending on incubation time and concentration, RNA degradation prevailed over RNA synthesis.

Phosphorylating mitochondrial preparations were obtained from T. cruzi culture (epimastigote) forms by grinding the cells with glass - beads and differential centrifugation. Using ADP as phosphate acceptor and succinate, L-malate or ascorbate + tetramethyl-p-phenylenediamine (TMPD) as oxidizable substrates, the respiratory control (R.C.) values were (mean +/- S.D.; n = 4, in parenthesis, the substrate): 2.8 +/- 0.10 (succinate): 2.3 +/- 0.13 (L-malate) and 2.0 +/- 0.12 (ascorbate + TMPD). The ADP:O values were 1.68 +/- 0.08, 1.42 +/- 0.08 and 0.66 +/- 0.66 +/- 0.12, respectively. The uncoupler CCCP stimulated substrate oxidation somewhat more than ADP. Succinate oxidation was by malonate and also by oxaloacetate but the latter was effective only after sonicating the mitochondrial preparation. The mitochondrial membranes oxidized also NADH but this oxidation was not subjected to control by the phosphate acceptor. Our results support the existence of two energy-conserving sites in T. cruzi respiratory chain (sites 2 and 3) and confirm previous observations by Stoppani et al. (Mol. Biochem. Parasitol. 2:3-21, 1980) with intact epimastigotes.

Tingenone and horminone, two natural quinonoid substances, inhibited the in vitro growth of Trypanosoma cruzi, 30 microM drug concentration producing total inhibition of growth. Tingenone inhibited total uptake and incorporation of [3H]thymidine, [3H]uridine, L-[3H]leucine into parasite macromolecules. Other quinonoids assayed were either less effective (abruquinone A) or even quite inactive (visminone B and ferruginin B). Investigation of several mechanisms for the cytotoxic action of tingenone pointed to the interaction with DNA as the most likely factor involved. Tingenone also inhibited the growth of Crithidia fasciculata, but the drug was significantly less active on this organism than on T. cruzi.

Kinetoplast DNA (kDNA) from Trypanosoma cruzi epimastigotes pretreated with the trypanocidal drugs nifurtimox (10 or 100 microM) or benznidazole (38 or 380 microM) showed an increased number of strand-breaks, as revealed by (a) trapping of alkali-denatured kDNA by nitrocellulose filters and (b) electrophoresis in an alkaline 2% agarose gel. Similar damage was observed in nuclear DNA (nDNA), as detected by centrifugation in an alkaline-sucrose gradient. DNA damage was reversible since reincubation in fresh medium for 24 hr restored filtration and electrophoretic and sedimentation patterns to normal.

Several new 1,2,3,4-tetrahydrocarbazole, 5,6-dihydrobenzo(alpha) carbazole, 3-methylindole and substituted benzimidazolyl compounds were synthesized and assayed for their action on growth of Trypanosoma cruzi, cultured in Warren's liquid medium. 6-chloro and 6,8-dichloro-N-(1-ethyl-N-diethylamino)-1,2,3,4-tetrahydrocarbazole++ fumarate resulted the more active in inhibiting growth of the parasite. The new compounds were apparently more effective than those prepared previously by Poliakoff et al. (5).

Nine Trypanosoma cruzi isolates not examined previously for kDNA structure were characterized by (a) endonuclease restriction analysis of mini-circles, followed by agarose gel-electrophoresis of digests, and (b) hybridization of mini- and maxi-circle fragments with four 32P-labeled cloned mini-circles from T. cruzi (pTck-1, 12, 13 and 14) or with 32P-labeled maxi-circles from T. brucei, respectively. The gel electrophoresis patterns demonstrated significant differences between isolates, which were confirmed and extended by the hybridization assay. When using pTck-1 and pTck-12 as probes, widely distributed heterogeneous mini-circle subpopulations were demonstrated in all the examined isolates, despite the occurrence of extensive homologies. pTck-14, assayed under high stringent conditions, detected an almost homogeneous mini-circle subpopulation in only three isolates, although under relaxed conditions, pTck-14 shared sequence homologies with most of the mini-circle subpopulations from all isolates. Rapidly evolving mini-circle regions were also detected using as probe pTck-13, a small mini-circle fragment. Preliminary maxi-circle characterization revealed polymorphic restriction endonuclease sites in the different T. cruzi isolates. These results were consistent with those obtained with mini-circles subjected to the same treatment.

Electron microscopic observation of Trypanosoma cruzi epimastigotes reveals the presence of microbody-like structures (microperoxisomes) in which 3,3'-diaminobenzidine (DAB) is peroxidized to electron-opaque material. The role of peroxidase in DAB peroxidation is supported by the enzyme demonstration in disrupted epimastigotes and the microbody-containing cell fractions.

Miconazole and econazole, two fungicide imidazole derivatives, completely inhibited growth of Trypanosoma cruzi (Tulahuen strain) at concentrations of about 20 muM. Culturing of T. cruzi in the presence of lower doses of imidazole derivatives produced: decrease of 5,7-diene sterol content in epimastigotes (including ergosterol); disappearance of the nuclear chromatin, vacuolization and decrease in the electron density of the cytoplasm; selective surface alterations as revealed by an increased response to wheat-germ- and phytohemagglutinin. At variance with the effect of miconazole on Candida (De Nollin et al. (1977) Antimicrobial. Agents Chemother. 11, 500-513), miconazole and econazole, under the experimental conditions used, did not increase the rate of hydrogen peroxide generation by T. cruzi.

Culturing of Trypanosoma cruzi in the presence of ethidium bromide (EB; 1.0-2.5 microM), for 5 days, affected the biogenesis of the Fo/F1-ATPase complex, presumably by EB interference with the expression of the putative Fo-sector subunits encoded by maxicircle DNA. In vitro, EB inhibited ATP hydrolysis by the Fo/F1-ATPase complex and also by soluble F1, the former preparation being the most strongly affected (EB I50, 250 microM). Analysis of EB effect at increasing MgATP concentrations, as well as combination of EB and p-chloromercuribenzoate inhibitions, indicated that the dye interacts with ATPase at sites other than the catalytic ones. No correlation was found between the in vitro inhibition of ATPase and EB effect on parasite growth.

Steady-state velocity studies using a substrate regenerating system showed that efrapeptin, citreoviridin and aurovertin inhibit both membrane-bound and soluble mitochondrial ATPase (coupling factor F1) from Trypanosoma cruzi. Maximal inhibitions of ATP hydrolysis produced by efrapeptin and citreoviridin were 100-93%, while the maximal inhibition produced by aurovertin was 40%. Half-maximal inhibitory concentrations decreased in the order citreoviridin greater than aurovertin greater than efrapeptin. Dissociation constants (KD) for the inhibitor-F1 complex were 81 nM (efrapeptin), 6.6 muM (aurovertin) and 40 muM (citreoviridin); KD values for the membrane-bound F1 were 2-4 fold higher than for soluble F1. Representation of efrapeptin inhibition data in the Hill form yielded straight lines (n = 1) while the same representation of citreoviridin inhibition yielded concave down plots. In contrast to the immediate effect of citreoviridin and aurovertin, efrapeptin inhibition was time-dependent. The onset of inhibition, which was pseudo-first-order with respect to efrapeptin, indicated that ATP may promote the binding of efrapeptin to the enzyme. The kinetics of ATP hydrolysis by T. cruzi ATPase as a function MgATP concentration could be explained by the presence of two substrate sites on the enzyme, interacting in such a way that the binding and catalytic events at one site were conformationally linked to the events at the other site, as with the mammalian ATPase. When the antibiotics were assayed at increasing substrate concentrations, efrapeptin produced a linear, mixed-type inhibition whereas citreoviridin produced a parabolic noncompetitive-type inhibition. The aurovertin effect was unusual since the extent of inhibition was greater at high substrate concentrations. Maximal concentrations of all the assayed antibiotics linearized the biphasic double reciprocal plot of control ATPase activity. Comparison of T. cruzi and mammalian F1 responses to the assayed antibiotics revealed the operation of similar inhibition mechanisms but the T. cruzi enzyme was significantly less sensitive to inhibitors than its mammalian counterpart.

Differential reduced minus oxidized (Red-Ox) or reduced. CO minus reduced (Red. CO-Red) spectra of Trypanosoma cruzi metacyclic trypomastigotes (Tulahuen strain), revealed the presence of cytochromes aa3, b, c5 5 8, o and possibly d (Fig. 1). Mitochondrial membranes from the epimastigote form of the parasite showed similar cytochrome spectra (Fig. 2A). Extraction of the mitochondrial membranes with guanidine and cholate, and spectroscopy of the cytochrome o -cyanide derivative proved that this cytochrome is an integral constituent of the mitochondrial membranes (Figs. 2B and 4, and Table 1). Contamination of the investigated samples with hemoglobin, peroxidase, catalase, cytochrome P-420, cytochrome P-450 or culture medium hemoproteins was ruled out by filtering the cells and mitochondrial membranes on Sephadex G-50, or by differential spectroscopy of pigments, or by differential centrifugations of samples. Investigation of pyridine-hemochromes revealed hemes A, B, C and D (Fig. 3), thus confirming the presence of the postulated cytochromes. Comparative spectroscopy of a series of T. cruzi stocks (including Tulahuen and Y strains), many of them obtained from acute or chronic forms of Chagas disease, revealed significant variability in the cytochrome content (Table 2). Taking cytochromes o and b as standard for comparison, the epimastigotes samples could be grouped as follows (in parenthesis number of passages through the culture medium): 1) stocks with a relatively high content of cytochromes b and o, prevailing the former (stocks Y (116), RA (114), AF, FN, TN and MG (14 y 16); 2) stocks with a relatively low content of both cytochromes: Y (119), AWP and UP; 3) stocks with a low content of cytochrome b, without cytochrome o: CA-I and CA-I (V); 4) stocks without cytochromes: Y(117 and 118) and RA(113). In some strains (e.g. Y and RA), significant variation of cytochrome content in different stocks of the same isolate was observed (Table 2), but the Tulahuen strain proved to be less variable. Comparison of cytochrome distribution and other properties of parasites, namely, lethality for mice and morphology, did not allow to establish positive correlations.

Addition of beta-lapachone to the epimastigote (culture) form of Trypanosoma cruzi, suspended in saline, buffered-isotonic medium (pH 7.2), determined the appearance of large amounts of H2O2 in the suspension medium, as measured spectrophotometrically by formation of the H2O2 horse radish peroxidase complex. Under similar conditions, alpha-lapachone did not induce H2O2 formmation. Using NADH as electron donor, beta-lapachone (not alpha-lapachone) increased significantly the rate of H2O2 generation by epimastigote homogenates and the same occurred with NADPH, although in a reduced extent. Similar results were obtained with the isolated mitochondrial and microsomal fractions although with the latter NADPH was more effective than NADH as electron donor for beta-lapachone reduction and peroxide generation. The distribution of peroxide generation in epimastigote fractions would indicate that about 92% of the beta-lapachone dependent formation of peroxide occurred in the mitochondria, and 8% in the endoplasmic reticulum. The growth of epimastigotes was inhibited 95% by 1 microgram/ml beta-lapachone, a concentration that determined maximal rate of H2O2 production. Since H2O2 and other intermediates of oxygen reduction such as O2- (superoxide anion) and OH (hydroxyl radical) are lethal to cells and tissues, it is possible that the effect of beta-lapachone on T. cruzi proliferation in vitro was mediated by H2O2 and related free radicals.

Addition of beta-lapachone, an o-naphthoquinone endowed with trypanocidal properties to respiring Trypanosoma cruzi epimastigotes induced the release of O2- and H2O2 from the whole cells to the suspending medium. The same beta-lapachone concentration (4 micron) that released H2O2 at maximal rate completely inhibited T. cruzi growth in a liquid medium. The position isomer, alpha-lapachone, did not stimulate O2- and H2O2 release, and did not inhibit epimastigote growth. beta-Lapachone was able to stimulate H2O2 production by the epimastigote homogenate in the presence of NADH as reductant. The same effect was observed with the mitochondrial fraction supplemented with NADH, where beta-lapachone enhanced the generation of O2- and H2O2 4.5- and 2.5-fold respectively. beta-Lapachone also increased O2- and H2O2 production (2.5 and 2-fold respectively) by the microsomal fraction with NADPH as reductant. Cyanide-insensitive NADH and NADPH oxidation by the mitochondrial and microsomal fractions (quinone reductase activity) was stimulated to about the same extent by beta-lapachone. alpha-Lapachone was unable to increase O2- and H2O2 production and quinone reductase activity of the mitochondrial and microsomal fractions.

Homogenates from T. cruzi epimastigotes produced 3.4 pmoles H2O2/min 10(6) cells, as detected by the cytochrome c peroxidase assay. Addition of NADH or NADPH increased H2O2 production by a factor of 3 and 5, respectively. When supplemented with NADH and NADPH, the mitochondrial, microsomal and supernatant fractions produced H2O2, the soluble fraction and the mitochondrial membranes being apparently the main generators of H2O2. The epimastigote homogenates showed cyanide-sensitive superoxide dismutase activity, equivalent to 0.28 microgram bovine superoxide dismutase per mg homogenate protein.

1. Grinding of epimastigotes of Trypanosoma cruzi with glass powder in a mortar yielded a Mg2+-activated adenosine triphosphatase (ATPase) preparation which was highly sensitive to oligomycin. 2. Chloroform treatment of the particles resulted in the solubilization of an ATPase which was (a) activated by MgCl2; (b) slightly inhibited by CaCl2; (c) activated by sulphite and bisulphite; (d) had an optimum pH of 7.6; and (e) had a Km for ATP of 2.1 mM (in the presence of 4 mM MgCl2). 3. The solubilized enzyme was insensitive to oligomycin and leucinostatin, which inhibited the membrane-bound ATPase, though inhibited by efrapeptin and quercetin. 4. The results indicate that the chloroform-extracted enzyme is a soluble F1-ATPase similar to those isolated from mammalian mitochondria.

Incubation of Trypanosoma cruzi mitochondrial ATPase (Fo-F1) with the xanthine oxidase system (XO), Fenton's reagent (Fe2+ + H2O2) and the ascorbate-Cu system, caused gradual loss of enzyme activity, which increased as a function of incubation time and rate of oxygen radical generation. The essential role of OH. radicals for ATPase inactivation was supported by a) the enzyme protection afforded by superoxide dismutase, catalase and mannitol, when using the XO system; b) the similar effect of mannitol and benzoate with Fenton's reagent; c) the similar effect of catalase, EDTA and histidine with the ascorbate-Cu system; d) the increased rate of ATPase inactivation by 1) the XO system supplemented with chelated iron, and 2) the ascorbate-Cu system supplemented with H2O2. Comparison of oxygen radical generators for their action on membrane-bound (Fo-F1) and soluble F1 revealed that ascorbate-Cu was the most effective one, possibly because of its capability of producing OH. radicals that react preferentially with the enzyme at their formation site.

Incubation of Trypanosoma cruzi culture (epimastigote) forms with nifurtimox (10 or 100 microM), benznidazole (38 or 380 microM) and beta-lapachone (1.6 or 7.8 microM) produced damage of nuclear DNA, as shown by the increased rate of the "unscheduled DNA synthesis" in epimastigotes arrested at phase S (9-, 3-, and 6-fold, respectively). alpha-lapachone, a position isomer of beta-lapachone, was completely ineffective. In order to demonstrate the "unscheduled repair of DNA", the semiconservative replication was inhibited by preincubating the epimastigotes for 16 hours with 10 mM hydroxyurea and 0.3 mM cycloheximide. Kinetoplast DNA (kDNA) extracted from epimastigotes pretreated with the trypanocidal agents revealed an increased number of single-strand breaks. After alkaline agarose-gel electrophoresis, a fast moving DNA fraction was detected in the kDNA from nifurtimox, benznidazole and beta-lapachone-treated parasites, while trapping of alkali-denatured kDNA by nitrocellulose filters, was significantly increased after treating the epimastigotes with the same drugs. Reincubation of these epimastigotes in fresh medium for 24 h, reestablished kDNA electrophoretic and filtration patterns to normality, except with 7.8 microM beta-lapachone, thus proving the reversibility of DNA lesions. Redox-cycling of nifurtimox and beta-lapachone in T. cruzi generates oxygen radicals, and accordingly, the higher effectiveness of these drugs (as compared with benznidazole and alpha-lapachone) supports the role of oxygen radicals for the trypanocidal action.

Despite its epidemiological importance, chemotherapy of Chagas' disease is still an unsolved problem. Many drugs have been assayed for their trypanocidal action on T. cruzi, but, so far, naftoquinone-imines have not been included in drug screenings. Fernández A.E. et al., prepared a series of 4-(aminomethylisoxazolyl)-1,2-naftoquinones. Some of these, namely, IIIA (2-hydroxy-N-(3,4-dimethyl-5-isoxazolyl)-1,4-naftoquinone-4- imine), IIIC hydroxy-N-3,5-dimethyl-4-isoxazolyl)-1,4-naftoquinone-4-i min e, IIIE (2-hydroxy-N-(3-methyl-5-isoxazolyl)-1,4-naftoquinone-4-i min e), and IVD (5-methyl-3-isoxazolyl)-1,2-naftoquinone-4-imine) were assayed on T. cruzi epimastigotes. Parasite growth and DNA synthesis were inhibited 12-36% and 14-49%, respectively, by 18-19 microM quinones or 44-83% and 37-73%, respectively, by 60-78 microM quinones (Table 1). The assayed compounds (37-100 microM concentration) stimulated oxygen uptake (Figs. 2 and 3) and superoxide anion generation by epimastigotes to 0.20-0.70 nmol/min/mg of cell protein (O2- measured by the adrenochrome method, Table 2). In the presence of quinones, T. cruzi mitochondrial fraction supplemented with 0.42 mM NADH produced O2- at a rate of 1.10-1.45 nmol/min/mg of protein (31 and 75 microM quinone, respectively; Table 3) whereas, under the same experimental conditions, the microsomal fraction supplemented with NADPH produced O2- at rate of 0.85 and 1.03, respectively (same units). The kinetics of O2- generation by the mitochondrial fraction supplemented with quinone IIIC and NADH revealed two interacting sites (Fig. 5). The corresponding Km(s) were 5.2 and 17 microM and the Vm, 13 and 17 nmoles O2-/min/mg of protein, respectively. The quinone interaction with the mitochondrial membranes involved a "quinone reductase" located on the substrate site of the antimycin site, as shown by the 3.6-fold increase of the oxygen uptake rate induced by quinone IIIE in Fig. 2. Apparently, the quinone excess inhibited the reductase since by varying the quinone concentration in the 0-500 microM range, the oxygen uptake versus quinone concentration plots showed a maximum at 100-150 microM quinone (Fig. 3). O2- generation by quinones depended on the latter redox-cycling, as shown by a) the decrease of quinone absorption at 480-500 nm after incubation with T. cruzi epimastigotes for the time necessary to exhaust the reaction mixture oxygen, and b) the reappearance of the quinone absorption band after oxygenation of the anaerobic reaction mixture (Fig. 6).
