The rapid development of Lentinula edode industry in China has led to the production of large amounts of waste substrates, which has brought enormous pressure to ecological environment. Thus, it is of great significance to study utilization of spent substrate. In this study, complex enzyme combined with high pressure pretreatment were applied to extract polysaccharide from spent substrate of L. edode in order to improve the utilization efficiency of its residue and explore more biological application of the polysaccharides from spent L. edodes substrate. The optimal extraction conditions were as follows: complex enzyme (cellulase, xylanase, β-glucanase) ratio, 3:2:1; total amount of enzyme, 660 U·g
-1 ; liquid/material ratio, 20 mL/g; temperature, 45 ℃; time, 6 h; The yield of purified polysaccharide (LE-UE) was 5.98% and the content was 51.60%. LE-UE could markedly improve inflammatory damage of LPS-induced macrophages RAW264.7 by inhibiting release of NO and secretion of inflammatory cytokines. Animal experiments showed that LE-UE could markedly improve the histopathological damage and inflammation in ulcerative colitis mice by regulating the level of oxidative stress, and reduce the release of inflammatory cytokines by down-regulating mRNA expression and proteins levels. Therefore, LE-UE has anti-inflammatory activity and may be used as a novel anti-inflammatory agent for treatment. This study could provide scientific evidence for the high-value utilization of spent substrates and also provide theoretical basis for the development of functional feed additives.
