Green tea is popular among consumers because of its high nutritional value and unique flavor. There is often a strong correlation among the type of tea, its quality level and the price. Therefore, the rapid identification of tea types and the judgment of tea quality grades are particularly important. In this work, a novel sensor array based on nanozyme with polyphenol oxidase (PPO) activity is proposed for the identification of tea polyphenols (TPs) and Chinese green tea. The absorption spectra changes of the nanozyme and its substrate in the presence of different TPs were first investigated. The feature spectra were scientifically selected using genetic algorithm (GA), and then a sensor array with 15 sensing units (5 wavelengths × 3 time) was constructed. Combined with the support vector machine (SVM) discriminative model, the discriminative rate of this sensor array was 100% for different concentrations of typical TPs in Chinese green tea with a detection limit of 5 μM. In addition, the identification of different concentrations of the same tea polyphenols and mixed tea polyphenols have also been achieved. Based on the above study, we further developed a facile and efficient new method for the category differentiation and adulteration identification of green tea, and the accuracy of this array was 96.88% and 100% for eight types of green teas and different adulteration ratios of Biluochun, respectively. Amino acids are the basic building blocks of protein that are very important to the nutrition and health of humans and animals, and widely used in feed, food, medicine and daily chemicals. At present, amino acids are mainly produced from renewable raw materials by microbial fermentation, forming one of the important pillar industries of biomanufacturing in China. Amino acid-producing strains are mostly developed through random mutagenesis- and metabolic engineering-enabled strain breeding combined with strain screening. One of the key limitations to further improvement of production level is the lack of efficient, rapid, and accurate strain screening methods. Therefore, the development of high-throughput screening methods for amino acid strains is very important for the mining of key functional elements and the creation and screening of hyper-producing strains. This paper reviews the design of amino acid biosensors and their applications in the high-throughput evolution and screening of functional elements and hyper-producing strains, and the dynamic regulation of metabolic pathways. The challenges of existing amino acid biosensors and strategies for biosensor optimization are discussed. Finally, the importance of developing biosensors for amino acid derivatives is prospected.

Water toxicity detection, as a valuable supplement to conventional water quality measurement, is an important method for evaluating water environmental quality standards. However, the toxicity of composite pollutants is more complicated due to their mixture effects. This study developed a novel, rapid and interference-resistant detection method for water toxicity based on an electrochemical biosensor using peak current from nitrite oxidation as a signal. Toxicants could weaken the characteristic peak current of nitrite to indicate the magnitude of toxicity. The proof-of-concept study was first conducted using a synthetic water sample containing trichloroacetic acid (TCAA), and then the results were compared with those of the traditional toxicity colorimetric method (CCK-8 kit) and laser confocal microscopy (CLSM). The accuracy of the biosensor was further verified with water samples containing individual pollutants such as Cd 2+ (50-150 μg/L), Cr 6+ (20-80 μg/L) mixture, triclosan (TCS; 0.1-1.0 μg/L) and TCAA (10-80 μg/L), or a mixture of the above. The viability of the sensor was further validated with the actual water sample from the Tuojiang River. The results demonstrated that although the concentration of a single conventional pollutant in water did not exceed the discharge standard for surface water, the comprehensive toxicity of natural water should not be ignored. Electrochemiluminescent (ECL) sensors for organophosphorus pesticides (OPs) have received considerable attention, whereas complicated electrode's immobilization, response to single hydrolysate and anodic emission correlated with ECL assays restrict their potential utilization. Herein, we developed a homogeneous dual-response cathodic ECL system for highly sensitive and reliable analysis of OP using CdTe QDs as emitters. CdTe QDs, emitting red light, were fabricated through a hydrothermal reaction and generated anodic and cathodic ECL emission upon stimulation of tripropyl amine and K 2 S 2 O 8 , respectively. Notably, CdTe QDs-K 2 S 2 O 8 showed a simultaneous response to thiol and acidic pH, and were regarded as a ECL sensor for methidathion with limit of detection of 0.016 ng/mL based on hydrolysis of acetylthiocholine into thiocholine and CH 3 COOH by acetylcholinesterase (AChE) and OPs' inhibition on AChE activity. This sensor also exhibited good practicability to detect methidathion in Chinese cabbage. Overcoming the limitations of traditional analytical methods and developing technologies to continuously monitor environments and produce a comprehensive picture of potential endocrine-disrupting chemicals (EDCs) has been an ongoing challenge. Herein, we developed a portable nuclear receptor (NR)-based biosensor within 90 min to perform highly sensitive analyses of a broad range of EDCs in environmental water samples. Based on the specific binding of the fluorescence-labeled NRs with their ligands, the receptors were attached to the EDC-functionalized fiber surface by competing with EDCs in the samples. The biosensor emitted fluorescence due to the evanescent wave excitation, thereby resulting in a turn-off sensing mode. The biosensor showed a detection limit of 5 ng/L E 2 -binding activity equivalent (E 2 -BAE) and 93 ng/L T 3 -BAE. As a case study, the biosensor was used to map the estrogenic binding activities of surface waters obtained from a rural community in the Yellow River basin in China. When the results obtained were compared with those from the traditional yeast two-hybrid bioassay, a high correlation was observed. It is anticipated that the good universality and versatility exhibited by this biosensor for various EDCs, which is achieved by using different NRs, will significantly promote the continuous assessment of global EDCs.

Food safety is a top priority of concern for consumers. To promote growth, as well as to treat diseases which occur in aquaculture, antibiotics, even banned veterinary antibiotics, are widely used. This survey investigated the levels of chloramphenicol (CAP) by LC-MS/MS in 291 aquatic samples (all the products in this survey were local sales, exported products were not included), including shellfish, shrimp and fish, from Shenzhen, South China. The CAP concentrations in these samples (based on wet weight) were: shellfish (-1 , mean: 68.8 µg kg -1 ); shrimp (-1 , mean: 0.5 µg kg -1 ) and fish (-1 , mean: 0.7 µg kg -1 ). The highest concentration of CAP was found in shellfish samples ( Mactra veneriformis , 205 µg kg -1 ), these levels were greater than those reported in aquatic products from other locations in China. The detection rate of CAP was 37.2% in shellfish samples (64/172), 13.6% in shrimp samples (3/22) and 16.5% in fish samples (16/97). The detection rate of CAP in shellfish samples was higher than that in fish and shrimp samples, and therefore food safety in this region cannot be ignored. The results obtained from the current study indicated a high incidence of CAP in aquatic products in this region and argue for stronger market supervision and regulation in China.

An ultra-performance liquid chromatography coupled with electrospray ionisation tandem mass spectrometry (UPLC-ESI-MS/MS) with pre-column derivatisation was developed and validated for the determination of semicarbazide in human urine. Urine samples were derivatised with 2-nitrobenzaldehyde and subsequently extracted with acetonitrile. Extracts were concentrated and then analysed by UPLC-MS/MS. The time per run was 7 min. Good results were observed for the linearity of matrix-matched calibration curves (R 2  > 0.99) in the concentration range of 1-100 µg/L. The absolute recovery ranged from 98.7% to 108.6%, with the relative standard deviations (RSDs) of 2.2%-3.6%. The limit of detection and quantification for the semicarbazide was 0.5 µg/L and 1 µg/L, respectively. The method showed good extraction efficiency, high sensitivity, and good reproducibility. It was suitable for the detection of semicarbazide in human urine. Residues of semicarbazide were between 1.0 and 41.5 μg/L in children's 24-h urine. This work is the first report on the quantitative analysis of SEM in 24-h human urine samples.

Indigo is a plant dye that has been used as an important dye by various ancient civilizations throughout history. Today, due to environmental and health concerns, plant indigo is re-entering the market. Strobilanthes   cusia (Nees) Kuntze is the most widely used species in China for indigo preparation. However, other species under Strobilanthes have a similar feature. In this work, 12 Strobilanthes spp. were analyzed using electrochemical fingerprinting technology. Depending on their electrochemically active molecules, they can be quickly identified by fingerprinting. In addition, the fingerprint obtained under different conditions can be used to produce scattered patter and heatmap. These patterns make plant identification more convenient. Since the electrochemically active components in plants reflect the differences at the gene level to some extent, the obtained electrochemical fingerprints are further used for the discussion of phylogenetics.

Aim: Smart bracelets are popular today. Based on their built-in motion sensors, they can serve as a cost-effective method of gait assessment in home-based care. Few studies have applied smart bracelets in the gait assessment of older Chinese adults. The present study aimed to: (i) establish reference gait parameters of older Chinese adults using smart bracelets under single and dual task; and (ii) explore the differences in gait parameters among non-frail and pre-frail Chinese older adults., Methods: A total of 50 community-dwelling older Chinese adults aged ≥50 years wore a smart bracelet sensor in the L3 region of the back and underwent a 10-m walking test under single- and dual-task conditions. Participants were preliminarily classified into non-frail and pre-frail groups based on the Fatigue, Resistance, Ambulation, Illnesses and Loss of Weight scale. Gait parameters including average walking speed, step frequency, root mean square (RMS), acceleration amplitude variability, step variability, step regularity and step symmetry were calculated based on the data exported from the bracelet., Results: Multivariate analysis of covariance (mancova) analysis showed that older adults had significantly decreased speed and step frequency (P < 0.05) under the dual cognitive task condition. Pre-frail older adults showed significantly decreased speed, mediolateral RMS, vertical RMS, anteroposterior RMS, vertical amplitude variability and vertical step regularity compared with non-frail older adults (P < 0.05)., Conclusions: The present study suggested that the decline in gait parameters as a result of frailty could be detected by the smart bracelet sensor. A highly sensitive electrochemical DNA biosensor made of polyaniline (PANI) and gold nanoparticles (AuNPs) nanocomposite (AuNPs@PANI) has been used for the detection of trace concentration of Ag(+). In the presence of Ag(+), with the interaction of cytosine-Ag(+)-cytosine (C-Ag(+)-C), cytosine-rich DNA sequence immobilized onto the surface of AuNPs@PANI has a self-hybridization and then forms a duplex-like structure. The whole detection procedure of Ag(+) based on the developed biosensor was evaluated by electrochemical impedance spectroscopy. On semi-logarithmic plots of the log Ag(+) concentration versus peak current, the results show that the prepared biosensor can detect silver ions at a wide linear range of 0.01-100 nM (R = 0.9828) with a detection limit of 10 pM (signal/noise = 3). Moreover, the fabricated sensor exhibits good selectivity and repeatability. The interfacial electron transfer of glucose oxidase (GOx) on a poly(glutamic acid)-modified glassy carbon electrode (PGA/GCE) was investigated. The redox peaks measured for GOx and flavin adenine dinucleotide (FAD) are similar, and the anodic peak of GOx does not increase in the presence of glucose in a mediator-free solution. These indicate that the electroactivity of GOx is not the direct electron transfer (DET) between GOx and PGA/GCE and that the observed electroactivity of GOx is ascribed to free FAD that is released from GOx. However, efficient electron transfer occurred if an appropriate mediator was placed in solution, suggesting that GOx is active. The PGA/GCE-based biosensor showed wide linear response in the range of 0.5-5.5 mM with a low detection limit of 0.12 mM and high sensitivity and selectivity for measuring glucose., (Copyright © 2015 Elsevier Inc. In this work, a biosensor based on luminescence resonance energy transfer (LRET) from NaYF4:Yb,Tm upconversion nanoparticles (UCNPs) to SYBR Green I has been developed. The aptamers are covalently linked to UCNPs and hybridized with their complementary strands. The subsequent addition of SYBR Green allows SYBR Green I to insert into the formed double-stranded DNA (dsDNA) duplex and brings the energy donor and acceptor into close proximity, leading to the fluorescence of UCNPs transferred to SYBR Green I. When excited at 980 nm, the UCNPs emit luminescence at 477 nm, and this energy is transferred to SYBR Green I, which emits luminescence at 530 nm. In the presence of oxytetracycline (OTC), the aptamers prefer to bind to its corresponding analyte and dehybridize with the complementary DNA. This dehybridization leads to the liberation of SYBR Green I, which distances SYBR Green I from the UCNPs and recovers the UCNPs' luminescence. Under optimal conditions, a linear calibration is obtained between the ratio of I530 to I477 nm (I530/I477) and the OTC concentration, which ranges from 0.1 to 10 ng/ml with a limit of detection (LOD) of 0.054 ng/ml., (Copyright © 2015 Elsevier Inc. A three-dimensional (3D) continuous and interconnected network graphene foam (GF) was synthesized by chemical vapor deposition using nickel foam as a template. The morphologies of the GF were observed by scanning electron microscopy. X-ray diffraction and Raman spectroscopy were used to investigate the structure of GF. The graphene with few layers and defect free was closely coated on the backbone of the 3D nickel foam. After etching nickel, the GF was transferred onto indium tin oxide (ITO) glass, which acted as an electrode to detect uric acid using cyclic voltammetry (CV) and differential pulse voltammetry (DPV). The GF/ITO electrode showed a high sensitivity for the detection of uric acid: approximately 9.44 mA mM(-1) in the range of 25 nM-0.1 μM and 1.85 mA mM(-1) in the range of 0.1-60 μM. The limit of detection of GF/ITO electrode for uric acid is 3 nM. The GF/ITO electrode also showed a high selectivity for the detection of uric acid in the presence of ascorbic acid. Mycotoxin contamination in agro-food systems has been a serious concern over the last few decades in China, where the Ministry of Health has set maximum limits for mycotoxins in different agro-products. Overall survey data show that aflatoxin contamination in infant cereals, edible oils, raw milk, ginger and its related products are far below Chinese regulatory limits. The absence of aflatoxin M1 contamination in infant milk powders indicates a high standard of control. Aflatoxins in liquorice roots and lotus seeds have been reported for the first time. For deoxynivalenol, high levels were found in wheat grown in the Yangtze Delta region, which is more prone to rainfall, supporting Fusarium infection. The emerging mycotoxins beauvericins and enniatins have been reported in the medicinal herbs in China. Ochratoxin A in wine was below the European Union regulatory limits, but fumonisins in maize need to be monitored and future regulatory control considered. Overall from all the survey data analysed in this review, it can be concluded that 92% of the samples analysed had mycotoxin levels below the Chinese regulatory limits. In terms of detection techniques in recent years, immuno-based assays have been developed largely due to their excellent sensitivity and ease of use. Assays targeting multiple mycotoxins like aflatoxins, ochratoxin A, zearalenone and deoxynivalenol have been reported using microarrays and suspension arrays targeting in particular maize, rice and peanuts. Aptamer-based assays against ochratoxin A and aflatoxins B1 and B2 have been developed involving fluorescence detection; and surface plasmon resonance immunosensors have been developed targeting wine, maize, wheat, wild rye, hay and peanut oil with high sensitivity (> 0.025 ng l(-1)). Commercialisation of these technologies is much needed for wider usage in the coming years.
