Partially selective gold nanoparticle sensors have the sensitivity and selectivity to discriminate and quantify benzene, toluene, ethylbenzene, p-xylene and naphthalene (BTEXN) at concentrations relevant to the US Environmental Protection Agency. In this paper we demonstrate that gold nanoparticle chemiresistors can do so in the presence of 16 other hydrocarbons and that they did not reduce the discriminating power of the array. A two-level full factorial designed experiment was performed on unary, binary, ternary, quaternary, quinary combinations of BTEXN analytes with and without the possibly interfering hydrocarbons. The nominal component concentration of the mixtures was 100 μg L−1, equivalent to approximately 100 parts per billion (ppb). Concentrations predicted with the random forests method had an average root mean square error of 10–20% of the component concentrations. This level of accuracy was achieved regardless of whether or not the 16 possibly interfering hydrocarbons were present. This work shows that the sensitivity and selectivity of gold nanoparticles chemiresistor sensors towards BTEXN analytes are not unduly affected by the other hydrocarbons that are expected to be present at a petroleum remediation site. ABSTRACT Alpha-synuclein (α-Syn) aggregates are the main component of Lewy bodies, which are the characteristic pathological feature in Parkinson's disease (PD) brain. Evidence that α-Syn aggregation can be propagated between neurones has led to the suggestion that this mechanism is responsible for the stepwise progression of PD pathology. Decreasing α-Syn expression is predicted to attenuate this process and is thus an attractive approach to delay or halt PD progression. We have used α-Syn small interfering RNA (siRNA) to reduce total and aggregated α-Syn levels in mouse brains. To achieve widespread delivery of siRNAs to the brain we have peripherally injected modified exosomes expressing Ravies virus glycoprotein loaded with siRNA. Normal mice were analyzed 3 or 7 days after injection. To evaluate whether this approach can decrease α-Syn aggregates, we repeated the treatment using transgenic mice expressing the human phosphorylation-mimic S129D α-Syn, which exhibits aggregation. In normal mice we detected significantly reduced α-Syn messenger RNA (mRNA) and protein levels throughout the brain 3 and 7 days after treatment with RVG-exosomes loaded with siRNA to α-Syn. In S129D α-Syn transgenic mice we found a decreased α-Syn mRNA and protein levels throughout the brain 7 days after injection. This resulted in significant reductions in intraneuronal protein aggregates, including in dopaminergic neurones of the substantia nigra. The 550 km-long KwaZulu-Natal coast broadly divides into a bedrock-framed southern section and northern coastal plain section, each of approximately equal length. Seven types of coastal dune are identified on this coast: estuary barrier dunes; mainland beach foredunes; prograded beachridge dunes; headland bypass dunes; transverse dune fields; parabolic/migrating dunefield; and climbing dunes. This is largely because of the widespread assumption that preservation of buildings is a higher priority than preservation of beaches in response to sea level rise. Continuing beach degradation will be inevitable through active or passive processes. Active degradation means the actual removal of the beach, mostly as a result of shoreline engineering or mining. By far, the most important cause of beach loss in this category will be hard structures, especially seawalls. The incorrectly but widely perceived panacea of each replenishment will become economically impossible because of raised sea levels. Replenishment leads to intensified beachfront development and this ironically and inevitably will increase the future construction of seawalls. Passive degradation refers to reduction of the quality of the beach to the point that human usage drops, along with political support for costly beach preservation by nourishment. Passive degradation includes trash accumulation, oil spills, beach driving and most importantly, pollution, which is increasing rapidly apace with population growth and remains largely unrecognized by the beach-using public. Background Reliable methods of labeling human enteric nervous system ( ENS) stem cells for use in novel cell replacement therapies for enteric neuropathies are lacking. Here, we explore the possibility of using lentiviral vectors expressing fluorescent reporter genes to transduce, label, and trace mouse and human ENS stem cells following transplantation into mouse gut. Methods Enteric nervous system precursors, including ENS stem cells, were isolated from enzymatically dissociated mouse and human gut tissues. Lentivirus containing e GFP or mCherry fluorescent reporter genes was added to gut cell cultures at a multiplicity of infection of 2-5. After fluorescence activated cell sorting for e GFP and subsequent analysis with markers of proliferation and cell phenotype, transduced mouse and human cells were transplanted into the gut of C57 BL/6 and immune deficient Rag2-/gamma chain-/C5 mice, respectively and analyzed up to 60 days later. Key Results Mouse and human transduced cells survived in vitro, maintained intense e GFP expression, proliferated as shown by BrdU incorporation, and formed characteristic neurospheres. When transplanted into mouse gut in vivo and analyzed up to 2 months later, transduced mouse and human cells survived, strongly expressed e GFP and integrated into endogenous ENS networks. Conclusions & Inferences Lentiviral vectors expressing fluorescent reporter genes enable efficient, stable, long-term labeling of ENS stem cells when transplanted into in vivo mouse gut. This lentiviral approach not only addresses the need for a reliable fluorescent marker of human ENS stem cells for preclinical studies, but also raises the possibility of using lentiviruses for other applications, such as gene therapy. Serotonin toxicity results from serotonin excess in the central nervous system from serotonergic drugs. Previous studies suggest an association between T102C polymorphism of the serotonin 2A (5-hydroxytryptamine 2A) receptor gene and serotonergic adverse effects with serotonergic drugs. We aimed to determine whether there is an association between the T102C polymorphism and serotonin toxicity in patients taking serotonergic drug overdoses. Ninety-five patients presenting with serotonergic drug overdoses were examined for serotonin toxicity and had blood collected for DNA analysis. A diagnosis of serotonin toxicity was made in 14 patients (15%) based on the Hunter Serotonin Toxicology Criteria. Four of the 14 patients (29%) with serotonin toxicity had the C/C genotype compared with 20/81 (25%) without serotonin toxicity. There were no differences in age or sex, but the median defined daily dose taken by patients with serotonin toxicity was 27 (14-84) compared with 18 (2-136) in patients without serotonin toxicity (P=0.06). We discuss a recent resonant ultrasound spectroscopy (RUS) study of YBa2Cu3O6+Δ, which infers a line of phase transitions bounding the pseudogap phase and argue that this scenario is not supported by thermodynamic evidence. We show that the anomalies in RUS, heat capacity, and thermal expansion at the superconducting transition temperatures agree well, but there are large discrepancies between RUS and thermodynamic measurements at T× where the pseudogap phase transitions are purported to occur. Moreover, the frequency and temperature dependence of the RUS data for the crystal with Δ = 0.98, interpreted in terms of critical slowing down near an electronic phase transition, is five orders of magnitude smaller than what is expected. Choice feeding is often used to investigate an animal's nutritional requirements and dietary preferences. A problem with this approach is that animals with long gut transit times, such as the horse, may find it difficult to associate a chosen food with its nutritional consequence when alternative foods are presented simultaneously. One solution is to present foods singly for a period of time before a simultaneous choice session to allow the development of learned associations. This method was used to determine if horse's voluntary intake and feeding behavior was influenced by the macronutrient composition of the diet. Seven stabled horses, maintained on a low intensity exercise regimen, were allowed, on an ad libitum basis, haylage and 3 isocaloric forage based diets that were rich in 1 of 3 macronutrients (protein, lipid, and hydrolyzable carbohydrate). Initially, diets were presented as a 3-way choice for 5 d (self-selection a [SSa]), then singly (monadic phase) with exposure to each diet for 2 separate periods of 3 d each, and finally again as a choice for 5 d (self-selection b [SSb]). The total amount of trial diet offered differed with trial phase, with 2 to 2.5% of BW during SSa and the monadic phase, increasing to ad libitum access during SSb. To control differences in the total amount of trial diet offered, 2 measurements of voluntary intake were taken at 4 and 22 h postpresentation. Daily macro-nutrient and energy intakes were estimated from proximate analysis of the trial diets and batches of haylage fed. Feeding behavior was observed over a single 4-h period during both self-selection phases. Horses showed no initial preference after 4 h for any 1 diet during SSa. Following the monadic phase, horses demonstrated a preference for the protein and hydrolyzable carbohydrate rich diets over the lipid rich diet (P < 0.001). Dietary experience modified foraging behavior as the total number of visits to the diets decreased during SSb (P < 0.005). Analysis of 24 -h macronutrient consumption showed that protein and hydrolyzable carbohydrate intake increased during SSb, whereas lipid intake remained constant over both self-selection phases (P < 0.001). These data indicate for perhaps the first time that horses can respond to dietary macronutrient content and that single presentations during choice studies facilitates expression of dietary preferences. An approach to reduce the computational effort required to determine the worst-case gust loads for aircraft is described, specifically when structural modifications are included as part of the design process. The Sherman-Morrison-Woodbury formula is employed to determine the gust response of the modified structure using a reanalysis approach and to undertake an efficient modal reanalysis. The approach is demonstrated successfully using a simple wing and full-scale aircraft finite-element models to determine the worst-case "1-cosine" gust following structural modification. Selective development and preservation of major shoreline complexes on the continental shelf provide geomorphic evidence in support of alternating periods of Holocene sea-level stillstand and rapid rise during meltwater pulses. On a tectonically stable far-field setting (the southeast African shelf), regionally developed, well-preserved shoreline complexes occur at ~-100 m and ~-60 m within an overall shelf sequence dominated by transgressive ravinement (non-preservation of shorelines). The development of geomorphically mature submerged shorelines with equilibrium forms is attributed to extended periods of sea-level stability, and their preservation is the result of early cementation followed by very rapid sea-level rise (several centimeters per year) that caused overstepping. Meltwater Pulses 1A and 1B are recorded in the shelf stratigraphy and geomorphology. Setting aside local influences of topography and sediment supply, we hypothesize that shelf stratigraphies should preferentially preserve shorelines at water depths associated with the stillstands or slowstands immediately preceding meltwater pulses. A reappraisal of shelf stratigraphy and geomorphology, in particular the preservation potential of former shorelines, is required in the light of contemporary understanding of postglacial sea-level history. The duration of shoreline occupation at a given sea-level, coastal response to sea-level change and the controls on preservation of various shoreline elements can be recognized by detailed examination of submerged shorelines on the continental shelf. Using bathymetric and seismic observations, this article documents the evolution and preservation of an incised valley and lithified barrier complex between −65 m and −50 m mean sea-level on a wave-dominated continental shelf. The barrier complex is preserved as a series of aeolianite or beachrock ridges backed by laterally extensive back-barrier sediments. The ridges include prograded cuspate lagoonal shoreline features similar to those found in contemporary lagoons. The incised valley trends shore-parallel behind the barrier complex and records an early phase of valley filling, followed by a phase of extensive lagoonal sedimentation beyond the margins of the incised bedrock valley. Sea-level stability at the outer barrier position ( ca −65 m) enabled accumulation of a substantial coastal barrier that remained intact during a phase of subsequent slow sea-level rise to −58 m when the lagoon formed. These lagoonal sediments are stripped seawards by bay ravinement processes which caused the formation of several prograded marginal cuspate features. An abrupt rise in sea-level to −40 m, correlated with melt-water pulse 1B, enabled the preservation of thick lagoonal sediments at the top of the incised valley fill and preservation on the sea bed of the cemented core of the barriers. This situation is unique to subtropical coastlines where early diagenesis is possible. The overlying sandy sediment from the uncemented upper portion of the barriers is dispersed by ravinement, partly burying the ridges and protecting the underlying sediments. The high degree of barrier or shoreline preservation is attributed to rapid overstepping of the shoreline, early cementation in favourable climatic conditions and the protection of the barrier cores by sand sheet draping. Once a Urolepis rufipes male mounted, the female beat her antennae against his mouth and clypeus. Immediately after he swept his antennae rapidly downward and extruded his mouthparts, her abdomen rose as she opened her genital orifice. Almost simultaneously he backed up for copulation and she folded her antennae against her head. Neither her abdomen rising nor her antennal folding were essential to his backing up as determined from their timing and from experiments in which her abdomen was sealed or her antennae were removed. Females did not open their genital orifice if with a sealed-mouth male; and antennae-removed females did not open even in the few cases where untreated males extruded their mouthparts. Abstract: This paper presents an analysis of decadal (mesoscale) geomorphic change on sandy barrier islands in the fetch-limited environment of Chesapeake Bay. Low energy barrier islands exist in two settings: on the fringe of marshes and in open water and this analysis shows the various types of barrier island to be genetically related. Barrier islands that face the dominant wind and wave direction (E or W) retreat via barrier translation, preserving the barrier island volume. Those that exist in re-entrants are dominated by longshore transport processes, are strongly affected by sediment supply and are subject to disintegration. Marsh fringe barrier islands are perched on or draped over the surface of the underlying marsh. They migrate landwards via barrier translation during periodic high water events accompanied by large waves (hurricanes and northeasters). The underlying marsh surface erodes under all water levels and the rate of retreat of the barrier island and underlying marsh may take place at different rates, leading to various configurations from perched barrier islands several metres landward of the marsh edge, to barrier islands that have a sandy shoreface extending into the subtidal zone. The coastal configuration during landward retreat of marsh fringe barrier islands is subject to strong local control exerted by the underyling marsh topography. As erosion of marsh promontories occurs and marsh creeks are intersected and bypassed, the configuration is subject to rapid change. Periodic sediment influxes cause spits to develop at re-entrants in the marsh. The spits are initiated as extensions of adjacent marsh fringe barrier islands, but as the sediment volume is finite, the initial drift-aligned spits become sediment-starved and begin to develop a series of swash-aligned cells as they strive for morphodynamic equilibrium. The individual cells are stretched until breaches form in the barrier islands, creating inlets with tidal deltas. At this stage the low energy barrier islands closely resemble open ocean barrier islands. Continued reworking leads to widening of the inlets with consequent loss of constriction of tidal flow. The tidal deltas are, thus, no longer maintained and ultimately the island system disintegrates through inlet widening and is transformed to subtidal shoals. Barrier islands at various stages in this evolutionary cycle can be observed around the bay. Mid-bay barrier islands are affected by wave processes from both sides. This helps maintain the barrier island form and enables barrier islands to persist as sediment is exchanged between both sides of the island. Rates of barrier island translation are extremely high (up to 30m/year over a 12year period). This is attributed to the low volume of sand, which facilitates complete rollover in short periods. Accelerated sea level rise is likely to hasten the translation rates of marsh fringe barrier islands. The rapid disintegration of most spits compared to the persistence of marsh fringe barrier islands points to a reliance on the marsh as a stabilising point. The mesoscale behaviour of unusual barrier inlets on the east coast of South Africa is described. The inlets are associated with river-dominated estuaries and are consequently subject to periodic closure during low river flow. They are also located on a prograding barrier coast. Deflection of the estuary channel of a small river in the direction of longshore transport by 1230 m resulted in its capture by the estuarine channel of a larger river. The larger catchment river was not deflected by coastal progradation but maintained its position by periodic breaching of a channel through the barrier during floods. Flood breaches reseal by landward reworking of ephemeral deltas and may undergo limited migration. A tidal inlet at the northern end of the barrier is maintained by a small tidal prism and has a flood-tidal delta. It exhibits limited lateral migration and closes occasionally when wave-generated sediment transport filling the channel overcomes the flushing ability produced by tidal currents and freshwater discharge by the river. Large swell waves and the development of a washover channel cause the occasional breaching of the barrier, creating an ephemeral inlet midway along the barrier. Background and Objectives Evaluation of variant Creutzfeldt- Jakob disease ( vCJD) diagnostic/donor screening tests is made complicated by the very limited supply of blood samples from clinically confirmed cases of vCJD. To determine appropriate access for test developers to rare Creutzfeldt- Jakob disease ( CJD) blood samples, the oversight committee of the NIBSC CJD Resource Centre has developed a process and protocols detailing minimum requirements for both test sensitivity and specificity. This protocol is broadly similar to that outlined in the common technical specification (European Directive 98/79/EC). Materials and Methods Tests are subjected to a stepwise evaluation (step 1). vCJD tissue homogenates spiked into pooled human plasma (step 2). Blood samples from animals known to be incubating (Transmissible spongiform encephalopathy) TSE disease (scrapie/Bovine Spongiform encephalopathy (BSE)-infected sheep, BSE-infected primates) and appropriate controls (step 3). Fresh or frozen plasma from normal UK blood donors and (step 4). Plasma samples from individuals with confirmed clinical stage variant CJD (transfusion transmission) or sporadic CJD (no evidence of blood transmission). Results The assay evaluated performed with good sensitivity with vCJD-spiked tissue homogenates, poor sensitivity for ovine TSE-infected blood samples and failed with plasma from BSE-infected non-human primates and with true vCJD clinical samples. Background: Concern exists that involving vulnerable individuals as participants in research into suicide and self-harm may cause distress and increase suicidal feelings. Actual understanding of participants' experiences is however limited, especially in relation to in-depth qualitative research.Methods: Data were collected from four separate studies focused on self-harm or suicide. These included people with varying levels of past distress, including some who had made nearly lethal suicide attempts. Each involved semi-structured qualitative interviewing. Participants (n=63) were asked to complete a visual analogue scale measuring current emotional state before and after their interview and then comment on how they had experienced the interview, reflecting on any score change.Results: Most participants experienced a change in well-being. Between 50% and 70% across studies reported improvement, many describing the cathartic value of talking. A much smaller group in each study (18-27%) reported lowering of mood as they were reminded of difficult times or forced to focus on current issues. However, most anticipated that their distress would be transient and it was outweighed by a desire to contribute to research. An increase in distress did not therefore necessarily indicate a negative experience.Limitations: There was no follow-up so the long-term effects of participation are unknown. Scores and post interview reflections were collected from participants by the researcher who had conducted the interview, which may have inhibited reporting of negative effects.Conclusions: These findings suggest individuals are more likely to derive benefit from participation than experience harm. There is concern over the sustainability and environmental impact of mineral fertilisers and crop protection inputs used in intensive arable crop production systems. However, replacing mineral with organic fertilisers (animal and green manures) and restricting the use of chemosynthetic crop protection may significantly reduce crop yields. The effects of (a) replacing mineral with composted cattle manure fertiliser input and (b) omitting pesticide-based crop protection on potato ( Solanum tuberosum) tuber yield, leaf and tuber mineral nutrient content and leaf protein profiles were investigated. Switching to organic fertiliser had a greater effect on yield and protein profiles than the omission of chemosynthetic crop protection. Leaf N and P composition were significant drivers of protein expression, particularly proteins involved in photosynthesis such as the large subunit of RuBisCO, RuBisCO activase and the photosystem I reaction centre, which were at higher abundances in potato leaves grown under mineral fertiliser regimes. Proteins known to be induced in response to stress, such as dehydroascorbate reductase and Glutathione S-transferases, were also shown to be up-regulated under mineral fertilisation, possibly associated with higher Cd composition, whereas two proteins known to be involved in biotic stress (1,3-β- d-glucan glucanohydrolase; putative Kunitz-type tuber invertase inhibitor) were more abundant under compost fertilisation. Results showed that switching from mineral to organic fertilisers led to reduced N availability, a significant change in leaf protein expression and lower tuber yield. In contrast, omission of chemosynthetic crop protection inputs had limited effects on protein expression and no significant effect on tuber yield. This study provides information on the effects of changes in nutrient supply on protein expression patterns. Barrier islands commonly occur in coastal plain and deltaic settings, and a variety of behavioral and stratigraphic models exist. In this paper, a 90 km long barrier island system in the Outer Hebrides of Scotland is identified for the first time. Uniquely among barrier islands, the system rests directly on a planar bedrock (gneiss) surface (a strandflat). This bedrock surface has a profound influence on the morphology of the barrier island chain, and exerts the dominant influence on barrier island form and evolution during transgression. Topographical irregularities on the bedrock surface control both plan and profile morphology of the barrier islands. Under sea-level rise, by barrier migration, sediment is redistributed onshore and alongshore in a strongly three-dimensional manner, determined by topographic variability in the bedrock. The barrier islands represent an endpoint in transgressive barrier island geomorphology that is controlled by the bedrock surface: they are at the upper end of the recognized tidal amplitude for barrier islands; the offshore wave regime is very high-energy; they lack a shoreface; and neither ravinement surface nor transgressive sand sheet are developed. Spin injection and transport in solution-processed 6,13-bis(triisopropylsilylethynyl)-pentacene (TIPS-pentacene) are investigated using vertical CoPt/TIPS-pentacene/AlOx/Co spin valve architectures. The morphology and the molecular orientation of TIPS-pentacene spin-coated from a chloroform solution as grown in the spin valve devices are characterized by means of atomic force microscopy and x-ray diffraction, where the π-π stacking is found to be in planes oriented parallel to the substrate plane on CoPt (not L10-ordered). The magnetization hysteresis loops recorded with a superconducting quanmm interference device show an individual switching of the magnetization of the two ferromagnetic layers. The antiparallel state of the relative orientation of CoPt and Co is achieved due to their different coercive fields. A spin valve effect is observed at room temperature. The scaling of the magnetoresistance (MR) with the bulk mobility of TIPS-pentacene as a function of temperature together with the dependence of the MR on the interlayer thickness clearly indicates spin injection and transport in TIPS-pentacene. From organic semiconductor-spacer thickness-dependent MR measurements, we estimate a spin relaxation length of TIPS-pentacene of λs = (24 ± 6) nm and a spin relaxation time of approximately τs = 3.5 μs at r = 300 K, taking the measured bulk mobility of holes into account. The lack of robust chronological frameworks for pre-Columbian archaeology in the Caribbean limits the interpretive potential of ongoing studies of inter-site and inter-island interaction. This paper reviews the dating of 28 wood and shell artefacts from sites in northern Cuba in an attempt to date pre-Columbian activity in the region and develop dating methodologies for the study of pre-Columbian interaction in the Caribbean. This research compares dates from wood and shell artefacts from the waterlogged site of Los Buchillones in north-central Cuba with dates from shell artefacts from sites on eight islands in the Sabana-Camaguey archipelago in the Bahama Channel. In addition to providing a chronological framework based on absolute dates for pre-Columbian activity in northern Cuba, this paper also considers the methodological issues of using shell for dating pre-Columbian activity in the Caribbean. In cell and animal models, telomere erosion promotes chromosomal instability via breakage-fusion-bridge cycles, contributing to the early stages of tumorigenesis. However, evidence involving short telomeres in cancer development in humans is scarce, epidemiological and indirect. Here we directly implicate telomere shortening as a critical molecular event for malignant evolution in aplastic anemia (AA). Patients' telomere lengths at diagnosis of AA, while comparable to age-matched controls, inversely correlated with the probability of developing a cytogenetically abnormal clone. A significantly increased number of telomere signal-free chromosomal ends and chromosomal numerical and structural abnormalities were observed in bone marrow cells of patients with shorter telomeres in comparison with patients with longer telomeres and healthy subjects. The proportion of monosomy-7 cells in the bone marrow at diagnosis of AA inversely correlated with telomere length, years before the emergence of an autonomous and clinically detectable abnormal clone. Marrow cells of clinically healthy individuals carrying loss-of-function telomerase mutations and with extremely short telomeres also showed chromosomal instability in vitro. These results provide the first clinical direct evidence in humans that short telomeres in hematopoietic cells are dysfunctional, mediate chromosomal instability and predispose to malignant transformation in a human disease. The literature on incised river valley sedimentology is dominated by studies of sediment-rich systems in which the valley has been filled during and/or shortly after drowning. In contrast, the Holocene evolution of the Kosi Lagoon, South Africa (an incised coastal plain river valley) took place under very low sedimentation rates which have produced a distinctive stratigraphy and contemporary sedimentary environments. The findings are based on a synthesis of the results of studies of seismic stratigraphy, sediment distribution, morphodynamics and geomorphology. Barrier migration was prevented by a high pre-Holocene dune barrier against which Holocene coastal deposits accumulated in an aggradational sequence. Holocene evolution of the back barrier involved: (i) drowning of the incised valley; (ii) wave-induced modification of the back-barrier shoreline leading to segmentation during the highstand; and (iii) marine sedimentation adjacent to the tidal inlet. Segmentation has divided the estuary into a series of geochemically and sedimentologically distinctive basins connected by channels in the estuarine barriers. The seismic stratigraphy of the back barrier essentially lacks a transgressive systems tract, shoreline modification and deposition having been accomplished during the highstand. The lack of historical geomorphological change suggests that the system has achieved morphological equilibrium with ambient energy conditions and low sediment supply. This study presents a classification for estuarine incised valley fills based on the balance between sea-level rise and sedimentation in which Kosi represents a 'give-up' estuary where much of the relict incised channel form is drowned and preserved. It exhibits a fundamentally different set of evolutionary processes and stratigraphic sequences to those of the better known incised valley systems in which sedimentation either keeps pace with sea-level ('keep-up' estuaries) or occurs after initial drowning ('catch-up' estuaries). By exploiting the correlation properties of ultracold atoms in a multimode interferometer, we show how quantum enhanced measurement precision can be achieved with strong robustness to particle loss. While the potential for enhanced measurement precision is limited for even moderate loss in two-mode schemes, multimode schemes can be more robust. A ring interferometer for sensing rotational motion with noninteracting fermionic atoms can realize an uncertainty scaling of 1 /(N√&eegr;) for N particles with a fraction &eegr; remaining after loss, which undercuts the shot-noise limit of two-mode interferometers. Neurocritical care is a subspecialty of critical care medicine, dedicated to the care and the advancement of care of critically ill patients with neurosurgical or neurological diseases. Neurocritical care patients are heterogeneous, in both their disease process and the therapies they receive, however, several studies demonstrate that care of these patients in dedicated NeuroIntensive Care Units (neuroICUs) by neurointensivists, who coordinate their care is associated with reduced mortality and resource utilization. NeuroICUs foster innovation, and yet despite all the recent advances, much research needs to be undertaken in neurocritical care to better understand the disease pathophysiology and to demonstrate improved outcome with the use of goal-directed therapy based on evolving techniques and therapies. Background and Objectives With four transfusion related transmissions of variant Creutzfeldt-Jakob Disease (vCJD), three of which developed clinical disease and the other died of other causes but was positive for markers of infection, there is an increased urgency to identify and implement a test for blood donor screening. With limited amounts of blood samples from vCJD cases available test evaluation is challenging. Alternative approaches are therefore needed. Control and vCJD tissues homogenates, where levels of markers of infectivity are known, were sequentially diluted in pooled human plasma. Identical sets of samples were provided blind to research groups developing diagnostic tests for vCJD; identical sample sets allows for direct comparisons of sensitivity to be made. Materials and Methods Control and vCJD tissue homogenates were sequentially diluted in pooled human plasma (detergent solvent treated or cryo-depleted) supplied by commercial fractionators. Dilutions of vCJD tissues were within and beyond the limits of detection previously determined by the conformation-dependent immunoassay (Cooper et al.: Vox Sang 2007;92:302-310; Bellon et al.: J Gen Virol 2003; 84: 1921-1925). A number of methods were used for the analysis of the blinded panels; with background signal from the normal prion protein (PrP) being removed by digestion with proteinase, epitope protection or selective capture of PrPtse. Results Assay sensitivities were directly compared using identical sample sets. This approach identified several transmissible spongiform encephalopathies (TSE) diagnostic tests, based on different principles, high in analytical sensitivity that reproducibly detected markers of vCJD infectivity in tissue homogenates. Conclusion The approach outlined has successfully compared in vitro diagnostics assays for their sensitivity and reproducibility and is a first step toward the evaluation of an assay suitable for blood donor screening/diagnosis of vCJD. Quantum entanglement offers considerable advantages in how the sensitivity of interferometric precision scales with the resources available. However, when using entangled states it is important that a balance is found between the phase sensitivity of the state and its robustness to particle loss. Recent work (Dorner et al 2009 Phys. Rev. Lett. 102 040403) has optimized this balance and found the ideal initial state for any given loss rate in a two-path interferometer. Here we describe a route towards achieving precisions close to that of the theoretical optimum by using beam splitters with variable reflectivity. We studied the valency and spin behavior of M = Mn, Fe, Co, Li, and Al in the high-temperature superconducting compound Bi2.15 Sr1.85Ca(Cu1-zMz)2O8+δ (Bi-2212) for small values of z. Mn, Fe, aud Co retain their magnetic moments, and our thermopower and magnetic susceptibility data imply ionization states Mn3+, Fe-2+, and Co2+, while Li and Al are accommodated in the charge reservoir layers. Single-crystal studies show that the susceptibility of Co2+ ions in Bi-2212 is strongly anisotropic, with a weak anisotropy detected for Mn3+ and none for Fe2+. Fits to a pseudogap formula for a pure Bi-2212 crystal suggest that the spin susceptibility of the host compound is more anisotropic than previously realized. Data in the superconducting state allow us to compare the pair-breaking properties of the different impurities. Several aspects of the data, including the stronger suppression of the superconducting transition temperature Tc by Co compared with Fe for underdoped and optimally doped samples, show that the d-level structure of the magnetic ions and multiorbital effects are important. We also find that the temperatures of the magnetization crossing points are equal to the low-field Tc values to within 1% or 2%. Background and Aims: An important part is played by inflammation in intracranial aneurysm formation. The hypothesis that there is an association of the proinflammatory cytokine interleukin-6 (IL-6) genotypes (-572G>C and -174G>C) with intracranial aneurysms was tested.Methods: IL-6 genotypes were determined in 91 Caucasian patients with aneurysms and compared with 2720 healthy UK controls.Results: For both polymorphisms, the distribution of the genotypes and estimated allele frequency were different between the control group and the aneurysm group. For -572G>C, a higher frequency of the C allele (p = 0.001) and more people homozygous for the C allele were found among those with aneurysms than among the controls (4.4% v 0.3%, p = 0.001). For -174G>C, more people homozygous for the C allele were found among the controls than among those with aneurysm (18% v 7%, p = 0.007). The 572C/174G haplotype was associated with an increased risk of aneurysms, with the relative risk compared with the common haplotype being 1.89 and that for the -572G/174C haplotype being 0.58 (p<0.0005).Conclusion: This is the first study to show that IL-6 promoter polymorphisms are associated with intracranial aneurysmal disease. The result will be a virtual data commons, expanding as further data are shared, allowing biology to become more of a ‘big science’. Informatics devices will exploit this ‘big new biology’, revitalizing comparative biology with a broad perspective to reveal previously inaccessible trends and discontinuities, so helping us to reveal unfamiliar biological truths. The collared peccary (Pecari tajacu) is a social species that recently has expanded its historical range, yet little is known of its dispersal behavior at the population or biogeographic level. We used autosomal and cytoplasmic markers to assess whether the genetic variation among 4 populations in Texas retains a signal of historically recent founding events. Genetic differentiation based on mitochondrial DNA (mtDNA) haplotype frequencies (FST) among these 4 populations was substantial. The mtDNA haplotype mismatch distributions indicate that P. tajacu recently has expanded its range across Texas; however, microsatellite-based standard analyses of molecular variance (AMOVAs) and pairwise FST estimates indicated little variation among populations, suggesting that dispersal is accomplished primarily by males. We also tested the alternative power of mtDNA haplotype or microsatellite genotype distribution patterns to detect a sex bias in instantaneous short-distance dispersal. Sex-specific hierarchical AMOVAs of mtDNA haplotype distributions (based on data from the most genetically diverse population) yielded significantly larger female-specific fixation index values compared to males, indicating a male instantaneous dispersal bias; however, variation in microsatellite frequency distribution was not partitioned in a sex-specific manner. Also, when sex-specific pairwise FST/(1 2 FST) among herds was regressed against the log of the distance between each pair of herds, we found no significant effect of distance for either sex and no sex-specific difference using other microsatellite-based analyses (mean sex-specific relatedness, r, or FST). We conclude that sampling scale limits microsatellite-based methods of detecting instantaneous dispersal sex bias, especially when gene flow is substantial and microsatellite polymorphism is modest (,20 alleles/locus). The clinical assessment of patients with peripheral nerve injury is primarily dependent on subjective clinical examination. We aimed to assess whether a thermotropic liquid crystal ring (TLC-ring) could provide the basis for an objective assessment of peripheral nerve injury by detecting temperature changes in the digits innervated by the damaged nerve. A group of patients with known median, ulnar or both digital nerve injuries were compared against a control group. TLC-rings, marketed to the general public as "mood rings", were applied to the affected and unaffected digits and the colour change recorded. Areas with nerve damage showed a statistically significant difference in colour response to those without damage. This study establishes the ability of TLCs to detect cutaneous temperature changes associated with peripheral nerve injuries. Further studies and improvements are needed to refine TLC as an acute assessment tool for peripheral nerve injuries. [ABSTRACT FROM AUTHOR]

Abstract: Objective: To examine all-cause and cardiovascular mortality in patients with severe hypertriglyceridaemia. Methods: 337 patients aged less than 80 years (47 with diabetes, 75 women) with a fasting triglyceride concentration on at least two occasions of >5.0mmol/l were registered by 21 lipid clinics in the United Kingdom and followed prospectively between 1980 and 2008 for 4353 person-years. The standardised mortality ratio (SMR) was calculated by comparison with the general population. Results: The mean untreated total cholesterol concentration was 9.8 (SD 3.6)mmol/l for men and 11.9 (7.2)mmol/l for women and the corresponding geometric mean triglyceride concentration was 12.6 (inter-quartile range 7.3, 21.6) and 15.7 (8.2, 29.2)mmol/l. There were 70 deaths, including 35 from CHD and 7 from stroke. The SMR for CHD was raised at 327 (95% confidence intervals 228, 455; p <0.0001) and remained elevated after excluding patients with diabetes at registration (SMR=287, 95% CI 190, 419; p <0.0001), and after excluding patients with CHD at registration (SMR=259, 95% CI 158, 400; p =0.0003). The increased SMR was most marked in younger men aged 40–59 years (SMR=544, 95% CI 304, 897; p <0.0001). The SMR for stroke for patients aged 20–79 years was raised at 262 (95% CI 105, 540; p =0.04), as was all-cause mortality at 164 (95% CI 129, 208; p <0.001). Conclusion: Severe hypertriglyceridaemia is associated with a substantially increased mortality from cardiovascular disease, even in the absence of diabetes. Abstract: The equilibrium planform concept (EPC) for bayed beaches has achieved wide currency in coastal morphodynamics. The north coast of Ireland comprises a series of discrete headland-embayment beaches within which waves and currents recycle a finite sediment volume. It is therefore an ideal setting in which to explore the applicability of the concept. Application of the approach to 9 embayment beaches on the north coast of Ireland provides some insights into the application of the concept. The planform of some beaches does correspond to that predicted while others do not. Those whose measured planform does not correspond to the predicted planform can be interpreted through, (a) difficulty in identifying the wave diffraction point, (b) disequilibrium on the beach (sediment scarcity or excess), (c) geological control of beach morphology. The subjectivity in selecting the diffraction point renders alternative explanations difficult and reduces the utility of the approach on natural shorelines, where significant irregularities render identification of such points difficult. Recent advances in the statistical analysis of microsatellite data permit calculation of sex-specific dispersal rates through sex- and age-specific comparisons of genetic variation. This approach, developed for the analysis of data derived from co-dominant autosomal markers, should be applicable to a sex-specific marker such as mitochondrial DNA. To test this premise, we amplified a 449 bp control region DNA sequence from the mitochondrial genome of the collared peccary (Pecari tajacu), and estimated intra-class correlations among herds sampled from three Texas populations. Analyses on data partitioned by breeding group showed a clear signal of male-biased dispersal; sex-specific fixation indices associated with genetic variation among social groups within populations yielded values for females (FGP=0.91), which were significantly larger than values for males (FGP=0.24; P=0.0015). The same general pattern emerged when the analyses were conducted on age classes (albeit nonsignificantly), as well as categories of individuals that were predicted a posteriori to be dispersers (adult males) and philopatric (adult females and all immatures). By extending a previously published methodology based on biparentally inherited markers to matrilineally inherited haploid data, we calculated sex-specific rates of contemporary dispersal among social groups within populations (m♂=0.37). The article presents a study which seeks to correlate clinical events such as surgery and death with the growth rate of abdominal aortic aneurysms (AAAs). Several factors that contribute to the prevalence of the disease including hypertension, hypercholesterolaemia, and diabetes are taken into consideration. It mentions that a growth rate of at least 2 millimeters annually posed significant connections with clinical events. Moreover, the study has recorded a bimodal growth pattern for AAA.

A candidate gene study was conducted on 10 established type II diabetes genes and 45 genes associated with autoimmune diseases, including type I diabetes (T1D), in a maximum of 1410 affected sib-pair families assembled by the Type I Diabetes Genetics Consortium. Associations at P values <10−3 were found for three known T1D regions at chromosomes 4q27, 12q13.2 and 12q24.13 (http://www.T1DBase.org). Support was obtained for a newly identified T1D candidate locus on chromosome 12q13.3–12q14.1 (rs1678536/KIF5A: P=8.1 × 10−3; relative risk (RR) for minor allele=0.89, 95% CI=0.82–0.97), which has a separate association from the previously reported T1D candidate locus ERBB3/12q13.2–q13.3. Our new evidence adds to that previously published for the same gene region in a T1D case–control study (rs1678542; P=3.0 × 10−4; odds ratio (OR)=0.92, 95% CI=0.88–0.96). The advent of genome-wide association (GWA) studies has revolutionized the detection of disease loci and provided abundant evidence for previously undetected disease loci that can be pooled together in meta-analysis studies or used to design follow-up studies. A total of 1715 SNPs from the Wellcome Trust Case Control Consortium GWA study of type I diabetes (T1D) were selected and a follow-up study was conducted in 1410 affected sib-pair families assembled by the Type I Diabetes Genetics Consortium. In addition to the support for previously identified loci (PTPN22/1p13; ERBB3/12q13; SH2B3/12q24; CLEC16A/16p13; UBASH3A/21q22), evidence supporting two new and distinct chromosome locations associated with T1D was observed: FHOD3/18q12 (rs2644261, P=5.9 × 10−4) and Xp22 (rs5979785, P=6.8 × 10−3; http://www.T1DBase.org). There was independent support for both SNPs in a GWA meta-analysis of 7514 cases and 9045 controls (P values=5.0 × 10−3 and 6.7 × 10−6, respectively). The chromosome 18q12 region contains four genes, none of which are obvious functional candidate genes. In contrast, the Xp22 SNP is located 30 kb centromeric of the functional candidate genes TLR8 and TLR7 genes. Both TLR8 and TLR7 are functional candidate genes owing to their key roles as pathogen recognition receptors and, in the case of TLR7, overexpression has been associated directly with murine autoimmune disease. Current federal and state legislation for mandated nurse staffing falls short of the ideal plan. Hospital nurse-staffing ratios are inadequate to maintain patient safety and quality nursing care. Mandating fixed nurse-to-patient ratios does not accommodate changes in patients' needs, evolving technology, and experience of nursing staff and will be immediately obsolete. Ideal measurement of nurse staffing incorporates nurse skill level and patient acuity. The Pennsylvania State Nurses Association supports legislation to establish patient acuity-based nurse staffing. Addressing health-related quality of life is considered a desirable component of routine care for patients with multiple sclerosis. However, use of available health-related quality of life surveys is not part of routine multiple sclerosis care, possibly due to administration, scoring, and interpretation challenges presented by available questionnaires. The 25-item Functional Index for Living with Multiple Sclerosis (FILMS) questionnaire was developed and validated to allow providers to monitor and easily apply health-related quality of life information to the patient encounter. The development and pilot test processes and the results of the validation study are reported here. Convergent validity, internal consistency reliability, and test-retest reproducibility were evaluated. Strong correlations of Functional Index for Living with Multiple Sclerosis subscales with control questionnaires were demonstrated by Pearson's correlation coefficients from 0.73 to 0.88. Internal consistency reliabilities ranged from 0.80 to 0.90, demonstrating that items were grouped into the appropriate subscale domains and that the subscale domains and the questionnaire as a whole exhibited good directionality. A biological survey of the Prince Edward Islands took place in December 2008. The survey repeated an earlier survey of the populations of surface-nesting seabirds on both islands and of fur seals (Arctocephalus spp.) and alien plants on Prince Edward Island in December 2001. Observations on burrowing seabirds, macro-invertebrates and plant communities on Prince Edward Island and an oceanographic survey of surrounding waters were also included. The survey confirmed many of the observations made on the earlier survey and permitted an assessment of trends in the abundance and distribution of biota since 2001. [ABSTRACT FROM AUTHOR]

There are more than 15,000 barrier islands in fetch-limited nearshore environments around the world. About half that number are actively evolving (eroding, accreting, migrating) in response to oceanographic processes and are the subject of this study. The remaining half consists of inactive islands protected by surrounding salt marsh or mangroves. Despite their global abundance these islands have not been previously systematically studied or even recognized as a major landform type. More than 70% of fetch-limited barrier islands are found on trailing edge coasts because conditions there are favorable for formation of sheltered waters. Fully 50% are found in the coastal zone of Australia, Mexico, and Russia. We identify eight different types of fetch-limited barrier islands based on genesis and mode of occurrence. Most of the active islands form in estuaries or bays (Spencer Gulf Australia), behind open ocean barrier islands (Pamlico Sound, North Carolina), or on flood tidal deltas of open ocean tidal inlets (Tapora Bank, New Zealand). Others occur on river deltas sheltered by offshore islands (Menderes Delta, Turkey), in sheltered bays with thermokarst topography (Yensei Bay, Russia), and on glacial outwash plains in fjords (Golfo Esteban, Chile). Due to a Holocene sea level drop, some southern hemisphere islands have been stranded above mean sea level and are intermittently active (Maputo Bay, Mozambique); they are only surrounded by water during spring tides and storms. Intermittent islands also form under conditions of high tidal amplitude (Kings Bay, Australia). Fetch-limited barrier islands are much smaller than their open ocean counterparts, averaging roughly 1 km long and 50 m wide and 1 to 2 m maximum elevation. They evolve in similar fashion to ocean barriers except that overwash is almost always the dominant island building process and dune formation is much less important. The two biggest distinctions between open-ocean and fetch-limited barrier islands are (1) complete evolutionary dependence on storms and (2) the important role of salt marsh and mangrove vegetation in controlling the shape and location of fetch limited barrier islands. Stabilized by salt marshes and mangroves, vegetative control is responsible for the irregular shape of some fetchlimited barrier islands and often plays a role in creating the foundation upon which the island evolves. The European Recommendation on Integrated Coastal Zone Management has six core principles. These form two groups, one concerned with strategic goals and one that has a local focus. The principles are presented as a menu of free-standing options, with no prioritization either within or between groups. In the case of coastal erosion management these characteristics result in irreconcilable conflicts, and actively hinder sustainable management of eroding coasts. The principles require clarification, and recognition that they are an indivisible integrated set which should not be used to select principles to advance a particular agenda.
