Medicinal plants are the primary sources of healthcare among the people of developing countries in villages and local towns. Documenting and reporting the traditional knowledge of medicinal plants may contribute to pharmaceutical research development. For this reason, we present our findings on ethnomedicinal plants from Lingchuan County, Shanxi, China, an unexplored area rich in medicinal plant resources. Information of ethnomedicinal plants were collected through questionnaire/semi-structured interviews from 180 informants, including traditional healers. Field surveys were conducted in 53 villages of Lingchuan County from 2017 to 2018. Informed consent was obtained from each participant before conducting the interview process. Quantitative analysis was performed for each recorded species, such as Relative Frequency Citation (RFC), Use Value (UV), and Factor of Informant Consensus (FIC). Diseases were categorized into twelve groups. A total 138 species of medicinal plants were recorded, belonging to 123 genera of 58 families. Asteraceae was the dominant plant family with 19 species, followed by Rosaceae and Fabaceae. Herbs were dominant among plant life-forms with 96 species, followed by shrubs and trees (15 species each). Roots were the most commonly used plant parts with 58 species, followed by whole plants and fruits (28 species each). Most plant species were reported non-toxic (84, 60%), followed by unknown toxicity (35, 25%), poisonous, and less toxic (19, 14%). Quantitative analysis revealed that Forsythia suspensa was with higher (0.33) RFC value, and Scutellaria baicalensis was recorded with a higher (0.91) UV. Treated diseases were categorized in 12 groups and evaluated by their FIC value, in which gynecological diseases have higher (0.93) FIC value followed by urinary system diseases. Most medicinal plants are used to clear away heat and relieve the surface. The present study revealed that local people of Lingchuan County confidently use ethnomedicinal plants for their healthcare needs. The higher indices value of a plant species resulted from quantitative analysis warrants further investigation, which may possess valuable phytochemical compounds that may result in new drugs for treating various human disorders.

Background: Human dental pulp stem cells (hDPSCs) exhibit excellent differentiation potential and are capable of differentiating into several different cellular phenotypes, including neurons. Platelet-rich plasma (PRP) contains numerous growth factors that can stimulate stem cell differentiation. In this study, we investigated the potential stimulatory effects of PRP on neurogenic differentiation and anti-apoptosis of hDPSCs in injured spinal cords., Methods: The unipotential differentiation capacity of hDPSCs was analyzed by cell surface antigen identification and cell cycle analysis. A spinal cord injury rat model composed of 40 Sprague-Dawley (SD) rats was used to facilitate an in vivo study. Rats were divided into four groups: a double-treatment group (receiving both neurogenic-induced hDPSCs and PRP), two single-treatment groups (receiving neurogenic-induced hDPSCs or PRP) and a sham group (receiving normal saline). The Basso, Beattie, Bresnahan Locomotor Rating Scale was subsequently used to evaluate the motor function of the spinal cord. Cell viability and differentiation of hDPSCs in the damaged spinal cords were analyzed and apoptosis of neural cells was evaluated using the terminal uridine nucleotide end labeling (TUNEL) assay., Results: Growth pattern, cell surface marker and cell cycle analyses revealed that hDPSCs have a high degree of multi-directional differentiation potential and can be induced into neurons in vitro . In the rat spinal cord injury model, double-treatment with hDPSC/PRP or single treatment with hDPSCs or PRP significantly improved motor function compared with the sham group (P<0.05). Apoptosis of neural cells was observed to be significantly higher in the sham group compared to any of the treatment groups. Double-treatment with hDPSCs and PRP resulted in the lowest apoptotic rate among the groups analyzed., Conclusions: hDPSCs exhibit differentiation potential and are capable of transforming into neural cells both in vitro and in vivo . Objective: This study aimed to evaluate the relationships between the albumin/globulin ratio (AGR), neutrophil/lymphocyte ratio (NLR), and platelet/lymphocyte ratio (PLR) and clinicopathological information for gastric cancer patients. In addition, the prognostic values of these hematological parameters for resectable gastric cancer patients undergoing a total gastrectomy were determined., Methods: A total of 245 patients with gastric cancer who underwent a total gastrectomy at our hospital between January 1, 2005, and December 30, 2015, were enrolled into this study. The preoperative AGR, NLR, and PLR in the serum samples of the patients were measured. The relationships between the hematological parameters and the disease-free survival (DFS) as well as overall survival (OS) were analyzed by statistical analysis., Results: The cutoff values of AGR, NLR, and PLR were 1.57, 3.5, and 193, respectively. Univariate analyses demonstrated that a low AGR, a high NLR, and a high PLR were significant risk factors for a poor prognosis. According to multivariate analysis, a high PLR was found to be independently associated with a poor survival. Additionally, when age was considered as a stratified factor, univariate analyses demonstrated that a low AGR had the tendency to be correlated with a shorter DFS in nonelderly patients (<65 years old). A low AGR was significantly correlated with a shorter DFS and OS in elderly patients (≥65 years old)., Conclusion: AGR, NLR, and PLR are independent risk factors associated with a poor gastric cancer survival by univariate analysis, and AGR is an independent risk factor for predicting DFS and OS in elderly patients (≥65 years old) with gastric cancer after total gastrectomy., (© 2022. The high electron mobility transistor (HEMT) structures on Si (111) substrates were fabricated with heavily Fe-doped GaN buffer layers by metalorganic chemical vapor deposition (MOCVD). The heavy Fe concentrations employed for the purpose of highly insulating buffer resulted in Fe segregation and 3D island growth, which played the role of a nano-mask. The in situ reflectance measurements revealed a transition from 2D to 3D growth mode during the growth of a heavily Fe-doped GaN:Fe layer. The 3D growth mode of Fe nano-mask can effectively annihilate edge-type threading dislocations and improve transfer properties in the channel layer, and consequently decrease the vertical leakage current by one order of magnitude for the applied voltage of 1000 V. Moreover, the employment of GaN:C film on GaN:Fe buffer can further reduce the buffer leakage-current and effectively suppress Fe diffusion.

For a chemotherapeutic agent to be effective, it must conquer the presence of blood-brain barrier (BBB), which limits the penetration of drugs into the brain. Tumours in the brain compromise the integrity of BBB and result in a highly heterogeneous vasculature, known as blood-brain tumour barrier (BBTB). In this chapter, we firstly highlight the cellular and molecular characteristics of the BBB and BBTB as well as the challenges aroused by BBB/BBTB for drug delivery. Secondly, we discuss the current strategies overcoming the challenges in invasive and non-invasive manners. Finally, we highlight the emerging strategy using focused ultrasound (FUS) with systemic microbubbles to transiently and reversibly enhance the permeability of these barriers for drug delivery., (© 2020. Vascular impairment is a crucial factor associated with chronic muscle pain, but relevant research from the microcirculatory aspect is lacking. Here, we investigated the differences in neck muscle microcirculation detected through laser-doppler flowmetry (LDF) and cervical biomechanics by a videofluoroscopic image in asymptomatic participants and patients with postural neck and shoulder pain. To understand the mechanism behind the effect of myofascial treatment, transverse friction massage (TFM) was applied and the immediate effects of muscular intervention on microcirculation were monitored. In total, 16 asymptomatic participants and 22 patients (mean age = 26.3 ± 2.4 and 25.4 ± 3.2 years, respectively) were recruited. Their neck muscle microcirculation and spinal image sequence were assessed. The differences in the baseline blood flow between the asymptomatic and patient groups were nonsignificant. However, the standard deviations in the measurements of the upper trapezius muscle in the patients were significantly larger ( p < 0.05). Regarding the TFM-induced responses of skin microcirculation, the blood flow ratio was significantly higher in the patients than in the asymptomatic participants ( p < 0.05). In conclusion, postintervention hyperemia determined through noninvasive LDF may be an indicator for the understanding of the mechanism underlying massage therapies and the design of interventions for postural pain.

Objective: To analyze the prognostic factors of breast cancer patients with isolated chest wall recurrence (ICWR) after mastectomy, and investigate the optimal treatment. Methods: A total of 201 breast cancer patients with ICWR after mastectomy who were treated in Cancer Hospital, Chinese Academy of Medical Sciences and the Fifth Medical Center Chinese PLA General Hospital from October 1998 to April 2018 were retrospectively analyzed. The median follow-up was 92.8 months and survival data were obtained. Results: Among 201 patients with ICWR, 103 patients developed subsequent locoregional recurrence (sLRR) and 5-year cumulative sLRR rate was 49.1%; 134 patients developed distant metastasis (DM) and 5-year DM rate was 64.4%; 103 patients died, the median progression-free survival (PFS) was 17.4 months and the 5-year PFS rate was 23.2%; the median overall survival (OS) was 62.5 months and the 5-year OS rate was 52.1%. Multivariate analysis showed that the recurrence interval ( HR =2.17, 95% CI: 1.26-3.73) and the locoregional treatment ( HR =1.59, 95% CI: 1.05-2.40) were the independent prognostic factors for sLRR. The initial HER2 status ( HR =1.60, 95% CI: 1.03-2.48) was the independent prognostic factor for DM. The recurrence interval ( HR =1.99, 95% CI: 1.30-3.04), the locoregional treatment ( HR =1.99, 95% CI: 1.43-2.76) and the treatment modalities after recurrence ( HR =1.70, 95% CI: 1.18-2.46) were the independent prognostic factors for PFS. The initial HER2 status ( HR =1.69, 95% CI: 1.02-2.81), the recurrence interval ( HR =1.85, 95% CI: 1.15-2.98) and the treatment modalities after recurrence ( HR =2.48, 95% CI: 1.56-3.96) were the independent prognostic factors for OS. Conclusions: Breast cancer patients after ICWR have an optimistic OS until now, but the risk of sLRR and DM is high. Comprehensive treatment modalities including surgery, radiotherapy and systemic therapy improve the outcome of breast cancer patients with ICWR after mastectomy.

Hydrostatically pressurized studies using diamond anvil cells on the structural phase transition of the free-standing screw-dislocation-driven (SDD) GaSe thin film synthesized by molecular beam epitaxy have been demonstrated via in-situ angle-dispersive synchrotron X-ray diffraction and Raman spectroscopy. The early pressure-driven hexagonal-to-rock salt transition at approximately ~ 20 GPa as well as the outstandingly structural-phase memory after depressurization in the SDD-GaSe film was recognized, attributed to the screw dislocation-assisted mechanism. Note that, the reversible pressure-induced structural transition was not evidenced from the GaSe bulk, which has a layer-by-layer stacking structure. In addition, a remarkable 1.7 times higher in bulk modulus of the SDD-GaSe film in comparison to bulk counterpart was observed, which was mainly contributed by its four times higher in the incompressibility along c-axis. This is well-correlated to the slower shifting slopes of out-of-plane phonon-vibration modes in the SDD-GaSe film, especially at low-pressure range (< 5 GPa). As a final point, we recommend that the intense density of screw dislocation cores in the SDD-GaSe lattice structure plays a crucial role in these novel phenomena., (© 2021. The Author(s).)

The effect of Mg doping on the electrical and optical properties of the p-GaN/AlGaN structures on a Si substrate grown by metal organic chemical vapor deposition was investigated. The Hall measurement showed that the activation efficiency of the sample with a 450 sccm Cp 2 Mg flow rate reached a maximum value of 2.22%. No reversion of the hole concentration was observed due to the existence of stress in the designed sample structures. This is attributed to the higher Mg-to-Ga incorporation rate resulting from the restriction of self-compensation under compressive strain. In addition, by using an AlN interlayer (IL) at the interface of p-GaN/AlGaN, the activation rate can be further improved after the doping concentration reaches saturation, and the diffusion of Mg atoms can also be effectively suppressed. A high hole concentration of about 1.3 × 10 18 cm -3 can be achieved in the p-GaN/AlN-IL/AlGaN structure.

Renal ischemia-reperfusion injury (IRI) is an important cause of setting off acute kidney injury. Neutrophil-mediated immunomodulation has a pivotal role in the evolving of IRI. The HMGB1-TLR4-IL-23-IL-17A axis gives rise to neutrophil activation. Therefore, in the study, the role of the HMGB1-TLR4-IL-23-IL-17A axis in IRI was evaluated. Cell viability, inflammation, apoptosis, oxidative stress, survival, renal function and pathology, and the activation of macrophages and neutrophils were measured. Moreover, we evaluated the acetylation, translocation, and secretion of HMGB1 as well as levels of TLR-4, IL-23, IL-17A, and neutrophil chemokines (KC, LIX, and MIP-2). In vivo, anti-HMGB1 antibody decreased the acetylation, translocation, and secretion of HMGB1, reduced the expression of TLR-4, IL-23, IL-17A, KC, LIX, and MIP-2, alleviated the activation of macrophages and neutrophils, improved the survival rate and renal dysfunction, and decreased inflammation, apoptosis, oxidative stress, and pathological injury of the kidney. However, the intervention with recombinant HMGB1(R-HMGB1) significantly abolish the above effect of anti-HMGB1 in IRI. Neutralization IL-23 or IL-17A can alleviated the neutrophils mediated renal dysfunction by suppressing inflammation, apoptosis, and oxidative stress in IRI. In vitro, we confirmed that hypoxic/deoxygenation (H/R) induces the secretion of HMGB1 though acetylation on HK-2 and HMGB1 promotes the secretion of IL-23 in a HMGB1/TLR-4-dependent manner on macrophages. Together, these results implied that the HMGB1-TLR4-IL-23-IL-17A axis regulates inflammation, oxidative stress, apoptosis, and renal injury in IRI by promoting the recruitment and migration of neutrophils., (Copyright © 2021 Elsevier B.V. Human Polycomb Repressive Complex 2 (PRC2) catalysis of histone H3 lysine 27 methylation at certain loci depends on long noncoding RNAs (lncRNAs). Yet, in apparent contradiction, RNA is a potent catalytic inhibitor of PRC2. Here, we show that intermolecular RNA-RNA interactions between the lncRNA HOTAIR and its targets can relieve RNA inhibition of PRC2. RNA bridging is promoted by heterogeneous nuclear ribonucleoprotein B1, which uses multiple protein domains to bind HOTAIR regions via multivalent protein-RNA interactions. Chemical probing demonstrates that establishing RNA-RNA interactions changes HOTAIR structure. Genome-wide HOTAIR/PRC2 activity occurs at genes whose transcripts can make favorable RNA-RNA interactions with HOTAIR. We demonstrate that RNA-RNA matches of HOTAIR with target gene RNAs can relieve the inhibitory effect of a single lncRNA for PRC2 activity after B1 dissociation. Focused ultrasound (FUS) offers an attractive tool for non-invasive neuromodulation, addressing a clinical need to develop more minimally invasive approaches that are safer, more tolerable and versatile. In combination with a cavitation agent, the effects of ultrasound can be amplified and localized for therapy. Using c-Fos expression mapping, we show how ultrasound-sensitive nanodroplets can be used to induce either neurosuppression or neurostimulation, without disrupting the blood-brain barrier in rats. By repurposing a commercial ultrasound contrast agent, Definity, lipid-shell decafluorobutane-core nanodroplets of 212.5 ± 2.0 nm were fabricated and loaded with or without pentobarbital. FUS was delivered with an atlas-based targeting system at 1.66 MHz to the motor cortex of rats, using a feedback-controller to detect successful nanodroplet vaporization and drug release. Neuromodulation was quantified through changes in sensorimotor function and c-Fos expression. Following FUS-triggered delivery, sham nanodroplets induced a 22.6 ± 21% increase in local c-Fos expression, whereas pentobarbital-loaded nanodroplets induced a 21.7 ± 13% decrease (n = 6). Background and Purpose: Optimal radiation target volumes for breast cancer patients with their first isolated chest wall recurrence (ICWR) after mastectomy are controversial. We aimed to analyze the regional failure patterns and to investigate the role of prophylactic regional nodal irradiation (RNI) for ICWR., Materials and Methods: Altogether 205 patients with ICWR after mastectomy were retrospectively analyzed. Post-recurrence progression-free survival (PFS) and overall survival (OS) rates were calculated by Kaplan-Meier method and the differences were compared with Log-rank test. Competing risk model was used to estimate the subsequent regional recurrence (sRR) and locoregional recurrence (sLRR) rates, and the differences were compared with Gray test., Results: The 5-year sRR rate was 25.2% with median follow-up of 88.6 months. Of the 52 patients with sRR, 30 (57.7%) recurred in the axilla, 29 (55.8%) in supraclavicular fossa (SC), and five (9.6%) in internal mammary nodes. Surgery plus radiotherapy was independently associated with better sLRR and PFS rates (p<0.001). The ICWR interval of ≤ 4 years was associated with unfavorable sRR (p=0.062), sLRR (p=0.014), PFS (p=0.001), and OS (p=0.005). Among the 157 patients who received radiotherapy after ICWR, chest wall plus RNI significantly improved PFS (p=0.004) and OS (p=0.021) compared with chest wall irradiation alone. In the 166 patients whose ICWR interval was ≤ 4 years, chest wall plus RNI provided the best PFS (p<0.001) and OS (p=0.022) compared with chest wall irradiation alone or no radiotherapy., Conclusion: Patients with ICWR have a high-risk of sRR in SC and axilla. Objective: To evaluate the effect of nutritional status on clinical and pathological data for stage Ⅰ-Ⅳ gastric cancer patients from the cancer survival investigation information. Methods: A database of 302 consecutive gastric cancer patients underwent radical gastrectomy was enrolled in this study. The clinical and pathological information of them were corrected and the relationship between the nutritional index and the patients survival time were analyzed by a Cox regression model. Results: The clinical data analysis of 302 patients with gastric cancer who received total gastric resection indicated that the nutritional status was related to the stage of tumor patients, suggesting that the later the stage was, the more necessary the nutritional therapy intervention was. Univariate analysis showed that Ⅲ+Ⅳ of TNM staging ( HR =4.417, 95% CI :2.483-6.351; P =0.029), patient age of 65 and above ( HR =2.217, 95% CI :0.522-3.912; P =0.038), lymph node metastasis positive ( HR =2.517, 95% CI :0.516-4.518; P =0.036), poor tumor differentiation ( HR =3.626, 95% CI :0.721-6.531; P =0.021) and low PNI ( HR =2.612, 95% CI : 0.712-4.512; P =0.029) is an important risk factor for poor prognosis. In the multivariate analysis, Ⅲ+Ⅳ of TNM staging ( HR =3.821, 95% CI :1.923-5.719; P =0.014), patient age of 65 and above ( HR =1.168, 95% CI :0.321-2.015; P =0.036) and low PNI ( HR =2.435, 95% CI: 1.024-3.846; P =0.039) was independently correlated with poor survival time; When age was used as a stratification factor, the correlation between CONUT recurrence and survival in patients with gastric cancer ≥65 years old after total gastric resection was analyzed and compared. For disease-free survival, the CONUT high group (>3) was 25.2 months, while the CONUT low group (≤3) was 30.9 months, (χ 2 =3.763, P =0.029), showing a significant difference. For the overall survival, the CONUT high(>3) group was 30.3 months, compared with the CONUT low(≤3) group at 34.5 months, (χ 2 =4.924, P =0.042), and the difference was also statistically significant. Conclusions: High controlling nutritional status is an independent risk factor associated with poor gastric cancer survival and it is an independent risk factor in predicting overall survival (OS) in elderly (≥65) gastric cancer after radical gastrectomy.

Background: Skin, the largest organ of the human body by weight, hosts a diversity of microorganisms that can influence health. The microbial residents of the skin are now appreciated for their roles in host immune interactions, wound healing, colonization resistance, and various skin disorders. Still, much remains to be discovered in terms of the host pathways influenced by skin microorganisms, as well as the higher-level skin properties impacted through these microbe-host interactions. Towards this direction, recent efforts using mouse models point to pronounced changes in the transcriptional profiles of the skin in response to the presence of a microbial community. However, there is a need to quantify the roles of microorganisms at both the individual and community-level in healthy human skin. In this study, we utilize human skin equivalents to study the effects of individual taxa and a microbial community in a precisely controlled context. Through transcriptomics analysis, we identify key genes and pathways influenced by skin microbes, and we also characterize higher-level impacts on skin processes and properties through histological analyses., Results: The presence of a microbiome on a 3D skin tissue model led to significantly altered patterns of gene expression, influencing genes involved in the regulation of apoptosis, proliferation, and the extracellular matrix (among others). Moreover, microbiome treatment influenced the thickness of the epidermal layer, reduced the number of actively proliferating cells, and increased filaggrin expression. Many of these findings were evident upon treatment with the mixed community, but either not detected or less pronounced in treatments by single microorganisms, underscoring the impact that a diverse skin microbiome has on the host., Conclusions: This work contributes to the understanding of how microbiome constituents individually and collectively influence human skin processes and properties. The results show that, while it is important to understand the effect of individual microbes on the host, a full community of microbes has unique and pronounced effects on the skin. Thus, in its impacts on the host, the skin microbiome is more than the sum of its parts. Video abstract.

The presence of blood-brain barrier (BBB) and/or blood-brain-tumor barriers (BBTB) is one of the main obstacles to effectively deliver therapeutics to our central nervous system (CNS); hence, the outcomes following treatment of malignant brain tumors remain unsatisfactory. Although some approaches regarding BBB disruption or drug modifications have been explored, none of them reach the criteria of success. Convention-enhanced delivery (CED) directly infuses drugs to the brain tumor and surrounding tumor infiltrating area over a long period of time using special catheters. Focused ultrasound (FUS) now provides a non-invasive method to achieve this goal via combining with systemically circulating microbubbles to locally enhance the vascular permeability. In this review, different approaches of delivering therapeutic agents to the brain tumors will be discussed as well as the characterization of BBB and BBTB. We also highlight the mechanism of FUS-induced BBB modulation and the current progress of this technology in both pre-clinical and clinical studies.

Beyond being the product of gene expression, RNA can also influence the regulation of chromatin. The majority of the human genome has the capacity to be transcribed and the majority of the non-protein-coding transcripts made by RNA Polymerase II are enriched in the nucleus. Many chromatin regulators can bind to these ncRNAs in the nucleus; in some cases, there are clear examples of direct RNA-mediated chromatin regulation mechanisms stemming from these interactions, while others have yet to be determined. Recent studies have highlighted examples of chromatin regulation via RNA matchmaking, a term we use broadly here to describe intermolecular base-pairing interactions between one RNA molecule and an RNA or DNA match. Aim: Novel coronavirus pneumonia (COVID-19) has become pandemic. It brings serious threat to hemodialysis (HD) patients. Therefore, we carried out a study on the clinical characteristics of HD patients with COVID-19., Methods: We retrospectively analyzed the data of 31 HD patients with COVID-19. The clinical features of patients include epidemiology, clinical symptoms, laboratory and imaging test, treatment and prognosis., Results: 61.3% were severe, and 38.7% were mild. 83.9% had a close contact history with COVID-19 patients. The average age was 62.3 years comprising of 58.1% men and 41.9% women. Ninety percent had chronic diseases except ESRD. Clinical symptoms include cough (85%), fever (43%), and shortness of breath (48.4%), etc. Complications included ARDS (25.8%), AHF (22.6%), and septic shock (16.1%), etc. 64.5% of patients had remission, and 35.5% of patients had no remission with 6.5% deaths. Compared with the baseline before infection, HD patients with COVID-19 had lower lymphocytes, albumin and glucose, and higher D-dimer, albumin, phosphorus, lactate dehydrogenase, and CRP. There was no significant correlation between the neutrophils/lymphocytes ratio and the severity of the disease., Conclusions: Compared with the reported general population, the HD patients are susceptible to COVID-19 infection, especially older men and those with other underlying diseases. Moreover, HD patients have more severe infection and inflammation with less symptoms and worse outcome. COVID-19 infection can cause dialysis patients lower immunity, stronger inflammation, malnutrition, and internal environment disorder. Neutrophils/lymphocytes ratio does not reflect the severity of the HD patients with COVID-19.

Purpose of Review: This review provides an update on chronic cluster headache (CH) focusing on clinical features, pathophysiology, and management as well as comparisons between Eastern and Western populations., Recent Findings: Chronic CH in Eastern populations was relatively rare, compared to that in Western populations. Lacrimation and/or conjunctival injection is the most frequently reported cranial autonomic symptom, and visual aura is predominant in chronic CH patients. Neuroimaging evidence in both ethnic groups suggests that CH pathophysiology involves the hypothalamus and pain-modulatory areas, with dynamic alternations between CH episodes. Recent evidence indicates that midbrain dopaminergic systems may participate in CH chronicity. Noteworthy advances have emerged in neuromodulatory therapies for chronic CH, but treatment with calcitonin gene-related peptide (CGRP) monoclonal antibodies has been unsuccessful. Recent evidence shows divergence of chronic CH between Eastern and Western populations. Neuromodulatory therapies but not CGRP inhibition is effective in this intractable patient group.

Thermosensitive liposomes represent an important paradigm in oncology, where hyperthermia-mediated release coupled with thermal bioeffects enhance the effectiveness of chemotherapy. Their widespread clinical adoption hinges upon performing controlled targeted hyperthermia, and a leading candidate to achieve this is temperature-based magnetic resonance imaging (MRI)-guided focused ultrasound (MRgFUS). However, the current approach to hyperthermia involves exposures lasting tens of minutes to hours, which is not possible to achieve in many circumstances because of blood vessel cooling and respiratory motion. Here, we investigate a novel approach to overcome these limitations: to use fractionated ultrashort (~30 s) thermal exposures (~41° to 45°C) to release doxorubicin from a thermosensitive liposome. This is first demonstrated in a dorsal chamber tumor model using two-photon microscopy. Thermal exposures were then conducted with a rabbit tumor model using a custom MRgFUS system incorporating temperature feedback control. Two-dimensional (2D) layered GaSe films were grown on GaAs (001), GaN/Sapphire, and Mica substrates by molecular beam epitaxy (MBE). The in situ reflective high-energy electron diffraction monitoring reveals randomly in-plane orientations of nucleated GaSe layers grown on hexagonal GaN/Sapphire and Mica substrates, whereas single-orientation GaSe domain is predominant in the GaSe/GaAs (001) sample. Strong red-shifts in the frequency of in-plane [Formula: see text] vibration modes and bound exciton emissions observed from Raman scattering and photoluminescence spectra in all samples are attributed to the unintentionally biaxial in-plane tensile strains, induced by the dissimilarity of symmetrical surface structure between the 2D-GaSe layers and the substrates during the epitaxial growth. The results in this study provide an important understanding of the MBE-growth process of 2D-GaSe on 2D/3D hybrid-heterostructures and pave the way in strain engineering and optical manipulation of 2D layered GaSe materials for novel optoelectronic integrated technologies.

Objective: To explore whether femoral plasty can improve the fracture resistance of osteoporotic femoral specimens and prevent hip fracture, and to compare the difference of mechanical strength changes between two different femoral plasty methods in osteoporotic femoral specimens, so as to determine the best strengthening area of the plasty., Methods: Eighteen pairs of fresh osteoporotic femur specimens were collected and divided into two groups, A and B, 9 pairs in each group. Nine fresh osteoporotic femur specimens in each group were randomly selected for enhancement, and the corresponding contralateral specimens were used as control group. In group A1, the enhancement areas were femoral head, femoral neck, femoral trochanter and subtrochantericregion. And in group B1, the enhancement areas were femoral head, femoral neck and femoral trochanter region. The amount of cement injected into the femoral neck was recorded and the surface temperature of the femoral neck was measured. All specimens were biomechanically tested under simulated falls. Load-displacement curves, final loads were recorded. The final energy and stiffness of specimens were calculated. The biomechanical differences between the specimens of the enhancement group and those of the corresponding control group were compared, and the mechanical changes of the specimens by two different enhancement methods were compared., Results: Compared with the control group, the ultimate load and energy of the specimens in the enhanced group increased significantly, but the stiffness did not change significantly. There was no significant difference in final load and energy between specimens strengthened by two different methods., Conclusion: Femoral plasty has the advantages of minimally invasive, simple operationand remarkable effect. It can be used as a new method to prevent osteoporotic hip fracture.

The in-situ anomalous small-angle X-ray scattering (ASAXS) technique was used to investigate the strain glass transition (SGT) in as-quenched Ti 48.7 Ni 51.3 shape memory alloy during a thermal cycle of 30 °C to the SGT temperature T g (-50 °C) and then to 30 °C again. The Ni atoms play a critical role as point defects in the SGT mechanism and are very difficult to characterize using conventional tools. ASAXS identified the distribution of Ni atoms in nanodomains, which have a disk-like core-shell configuration with a Ni-rich shell and a highly Ni-rich core. Moreover, the morphological evolution, growth and shrinkage of the highly Ni-rich core domains during the thermal cycle through T g are demonstrated. The enhancement and reversible behavior of the local lamellar ordering arrangement of nanodomains during the SGT process at T g are revealed. The structural evolution and local ordering arrangement of nanodomains can play a role in hindering martensitic transformation. The ASAXS results provide new knowledge about the SGT beyond that from current simulation works. However, this corresponding structure of the nanodomains was destroyed when the specimen was heated to 250 °C.

Objectives: To characterize the genetic environment of the carbapenem resistance determinant in Proteus vulgaris of swine origin., Methods: The carbapenem-resistant P. vulgaris strain BC22 was isolated from a faecal swab from a diseased pig with diarrhoea in Sichuan Province of China in 2018. The presence of carbapenemase genes was screened by PCR. WGS and bioinformatics analysis were performed to analyse the genetic environment of the carbapenem resistance determinant., Results: P. vulgaris strain BC22 was found to harbour the carbapenemase gene blaNDM-1. WGS data revealed that blaNDM-1 was located in a truncated ISAba125 composite transposon. The carbapenem resistance gene blaNDM-1 and 20 other resistance genes, including the multiresistance gene cfr and the bifunctional aminoglycoside/quinolone resistance gene aac(6')-lb-cr, were located in a novel SXT/R391 integrative and conjugative element (ICE). This new SXT/R391 ICE of 148.7 kb was chromosomally located, and could be transferred to Escherichia coli., Conclusions: Here, we report a carbapenemase gene, blaNDM-1, integrated into an SXT/R391 ICE. Our study highlights that this SXT/R391 ICE may facilitate the dissemination of clinically important resistance genes such as blaNDM-1, cfr and aac(6')-lb-cr., (© The Author(s) 2020. While a large body of studies have examined polybrominated diphenyl ethers (PBDEs) and alternative brominated flame retardants (ABFRs) in wildlife, information on the bioaccumulation of these contaminants in reptiles in general, and snakes in particular, are scarce. We investigated the bioaccumulation characteristics of PBDEs and several ABFRs including decabromodiphenyl ethane (DBDPE), 1,2-bis(2,4,6-tribromophenoxy) ethane (BTBPE), hexabromobenzene (HBB) and pentabromotoluene (PBT) in a frog-eating snake, the striped keelback snake (Amphiesma stolata), from an e-waste recycling site in South China. The concentrations of ∑PBDEs and ∑ABFRs in the snakes ranged 53-5200 and 3.1-87 ng/g lipid weight, respectively; with higher levels in males than females. Additionally, the concentrations of BDE-28, -47, and -66 were positively correlated with snake size (snout-vent length and body mass), while negative correlations were found for most of the higher brominated PBDE congeners and HBB, PBT and BTBPE. The biomagnification factors (BMFs) estimated in the snake/frog relationship indicated a mild to moderate biomagnification of BDE-28, -47, -66, -100, -153 and -154 (with mean BMFs of 1.1-5.3), while a lack of magnification for the other PBDE congeners and all the ABFRs. Riparian zones, the critical ecological interfaces between terrestrial and aquatic ecosystems, are species rich habitats. However, riparian zones are seriously threatened by human activities in the world. Riparian invertebrates represent a large proportion of riparian biodiversity, perform various ecological functions, and provide an essential link between aquatic and terrestrial ecosystems. Although many studies have investigated the riparian invertebrate communities, there is lacking a comprehensive summary of maintaining mechanisms underlying riparian invertebrate diversity. This review discussed seven characteristics of riparian zones that might support high riparian invertebrate diversity: flood and drought, nutrient, microhabitat diversity, riparian vegetation, microclimate gradients, food resources and river spatial gradients. Further, we summarized the maintaining mechanisms of riparian invertebrate diversity. Disturbances of periodic flood and drought trigger the reproduction and migration of invertebrates, increase the turnover of invertebrate communities, and create suitable conditions for riparian invertebrates. Adequate nutrients support a high invertebrate diversity by increasing primary productivity of riparian habitats. Elevated microhabitat diversity provides a variety of niche space for specialist riparian invertebrates. Strong microclimate gradients provide complex and diverse habitats and thus facilitate the coexistence of aquatic and terrestrial invertebrates in riparian zones. Cross-ecosystem resource subsidies increase food availability and contribute unique food sources to riparian invertebrates. The differentiation of these factors along river longitudinal and lateral gradients provides conditions for the diversification of riparian invertebrates at a larger scale. Understanding the maintaining mechanisms of riparian invertebrate diversity is important for conservation of riparian biodiversity and integrated management of river ecosystems.

The crude electronic waste (e-waste) recycling has caused severe contamination of polychlorinated biphenyls (PCBs) and polybrominated diphenyl ethers (PBDEs) in the local environment, begging the question of whether wildlife like birds living at e-waste sites are suffering from adverse effects. We examined several oxidative status markers and their relationships with hepatic concentrations of PCBs and PBDEs in common kingfisher (Alcedo atthis) that inhabit an e-waste site in South China. The results showed that the mean concentrations of ∑PCBs (19100 ng/g) and ∑PBDEs (507 ng/g) in kingfishers from e-waste site were several orders of magnitude higher than those in the species from a reference site. Correspondingly, hepatic concentrations of malondialdehyde (MDA) and reactive oxygen species (ROS) in kingfishers from the e-waste site were significantly higher than those detected in the reference population, suggesting oxidative distress in the birds breeding at the e-waste site. The activities of superoxide dismutase (SOD) and catalase (CAT) in the liver from the exposed group were significantly lower compared with the reference group, while the opposite trend was observed for glutathione peroxidase (GPx). Significantly positive correlations were observed between PCB or PBDE concentrations and the levels of MDA and ROS; while negative correlations were found for enzymatic activities of SOD and CAT. Overall, our results may suggest a potential linkage between exposure to e-waste-derived pollutants and elevated oxidative stress, thereby indicating a potential oxidative stress-related health effects in common kingfisher breeding at the e-waste site., (Copyright © 2019 Elsevier Inc. Objective: Depression is the most common mental disorder in patients with chronic kidney disease (CKD), and previous studies have found that: (a) depression can accelerate the progression of CKD; and (b) depression is an independent risk factor for hospitalization and death among patients with CKD. Therefore, the objective of the current study was to investigate the prevalence of depression in Chinese patients with CKD, and to identify variables associated with depression., Methods: The study analyzed baseline data from a multicenter prospective cohort study of Chinese patients with chronic kidney disease (the C-STRIDE study). In all, 2995 participants in CKD stages 1 to 4 who completed a survey of depressive symptoms were included in the analyses. Depressive symptoms were assessed by the Zung Self-Rating Depression Scale (ZSDS). A ZSDS ≥50 was used as the cut-off score for the presence of depressive symptoms. Multivariable logistic regression was used to identify variables associated with depression., Results: The mean estimated glomerular filtration rate (eGFR) in the study sample was 51.59±29.49 ml/min/1.73 m 2 . The prevalence of depressive symptoms was 37.8% and increased significantly with CKD stage. Being female, a higher level of education, a low income, a larger economic impact of disease cost, comorbid cardiovascular disease, anemia, and impaired physical ability were independently associated with depressive symptoms., Conclusion: Our study revealed that depressive symptoms were common among patients with CKD in China. Regardless of the dissimilarity in the crystal symmetry, the two-dimensional GaSe materials grown on GaAs(001) substrates by molecular beam epitaxy reveal a screw-dislocation-driven growth mechanism. The spiral-pyramidal structure of GaSe multi-layers was typically observed with the majority in ε-phase. Comprehensive investigations on temperature-dependent photoluminescence, Raman scattering, and X-ray diffraction indicated that the structure has been suffered an amount of strain, resulted from the screw-dislocation-driven growth mechanism as well as the stacking disorders between monolayer at the boundaries of the GaSe nanoflakes. In addition, Raman spectra under various wavelength laser excitations explored that the common ε-phase of 2D GaSe grown directly on GaAs can be transformed into the β-phase by introducing a Se-pretreatment period at the initial growth process. This work provides an understanding of molecular beam epitaxy growth of 2D materials on three-dimensional substrates and paves the way to realize future electronic and optoelectronic heterogeneous integrated technology as well as second harmonic generation applications.

Objective: This study aims to investigate the association of filamin A with the function and morphology of prostate cancer (PCa) cells, and explore the role of filamin A in the development of PCa, in order to analyze its significance in the evolvement of PCa., Materials and Methods: A stably transfected cell line, in which filamin A expression was suppressed by RNA interference, was first established. Then, the effects of the suppression of filamin A gene expression on the biological characteristics of human PCa LNCaP cells were observed through cell morphology, in vitro cell growth curve, soft agar cloning assay, and scratch test., Results: A cell line model with a low expression of filamin A was successfully constructed on the basis of LNCaP cells. The morphology of cells transfected with plasmid pSilencer-filamin A was the following: Cells were loosely arranged, had less connection with each other, had fewer tentacles, and presented a fibrous look. The growth rate of LNCap cells was faster than cells transfected with plasmid pSilencer-filamin A (P<0.05). The clones of LNCap cells in the soft agar cloning assay was significantly fewer than that of cells stably transfected with plasmid pSilencer-filamin A (P<0.05). Cells stably transfected with plasmid pSilencer-filamin A presented with a stronger healing and migration ability compared to LNCap cells (healing rate was 32.2% and 12.1%, respectively; P<0.05)., Conclusion: The expression of the filamin A gene inhibited the malignant development of LNCap cells. In TiNi-based shape memory alloys (SMAs), the effects of thermal cycling on the transformation peak temperatures of B2 ↔ B19', B2 ↔ R, B2 ↔ B19, B2 ↔ R ↔ B19', and B2 ↔ B19 ↔ B19' one-stage and two-stage transformations have been investigated and compared. Experimental results of the differential scanning calorimeter and hardness tests indicate that the alloy's intrinsic hardness and the shear strain, s , associated with martensitic transformation, are two important factors, due to their relation to the ease of introducing dislocations during cycling. The temperature decrease by cycling for one-stage transformation was in the order of B2 ↔ B19' > B2 ↔ B19 > B2 ↔ R according to the orders of magnitude of their s values. This phenomenon also affected the suppression of B19 ↔ B19' and R ↔ B19' transformation peak temperatures in two-stage transformation. Both Ti 50 Ni 48 Fe 2 and Ti 48.7 Ni 51.3 SMAs aged at 450 °C for 4 h exhibited B2 ↔ R ↔ B19' transformation, but the hardness of the latter was much higher than that of the former due to the precipitation hardening of the Ti 3 Ni 4 precipitates. This causesd the decrease of the R ↔ B19' transformation peak temperature in the Ti 50 Ni 48 Fe 2 SMA to be much higher than that in Ti 48.7 Ni 51.3 SMAs aged at 450 °C for 4 h, which directly affected the sequential B2 ↔ R transformation of Ti 50 Ni 48 Fe 2 SMA in the next thermal cycle and decreased this transformation peak temperature. The Ti 48 Ni 52 SMA aged at 600 °C for 150 h underwent B2 ↔ B19' transformation and then B2 → R → B19'/B19' → B2 transformation as the cycle number increased, in which the B2 ↔ R transformation peak temperature raised slightly by cycling. This characteristic is uncommon and may have resulted from the strain field around the thermal-cycled dislocations favoring the formation of the R-phase.

Glioblastoma multiforme (GBM) continues to have a dismal prognosis and significant efforts are being made to develop more effective treatment methods. Sonodynamic therapy (SDT) is an emerging modality for cancer treatment which combines ultrasound with sonosensitizers to produce a localized cytotoxic effect. It has long been known that ultrasound exposure can cause both thermal and non-thermal bioeffects and it remains an open question to what degree does temperature impact the efficacy of SDT. In order to optimize the ultrasound parameters of SDT, transcranial MRI-guided focused ultrasound (MRgFUS) and real-time MRI thermometry were used to monitor the therapy in a rat brain tumor model. Experiments were performed using a C6 intracranial glioma tumor model in 37 male Sprague Dawley rats. Treatments were performed about 7 days following tumor implantation when the tumor reached 1-3 mm in diameter as determined by MRI. 5-aminolevulinic acid (5-ALA) was injected at a dose of 60 mg/kg six hours before sonication. MRgFUS at 1.06 MHz was delivered continuously at an in situ spatial-peak temporal-average intensity of 5.5 W/cm 2 for 20 min. MR thermometry was acquired to monitor the temperature change in the brain during sonication. The tumor growth response for animals receiving 5-ALA alone, FUS alone, 5-ALA + FUS and a sham control group were evaluated with MRI every week following treatment. During 20 min of MRgFUS at 5.5 W/cm 2 , the temperature within the targeted brain tumor was elevated from 32.3 ± 0.5 °C and 37.2 ± 0.7 °C to 33.2 ± 0.9 °C and 38.4 ± 1.1 °C, respectively. Both the tumor growth inhibition and survival were significantly improved in the 5-ALA + FUS group with 32 °C or 37 °C as the starting core body (rectal) temperature. 5-ALA alone and FUS alone did not improve survival. These promising results indicate that relatively low power continuous wave transcranial MRgFUS in conjunction with 5-ALA can produce an inhibitory effect on rat brain tumor growth in the absence of thermal dose. Further investigation of the ultrasound parameters is needed to improve the therapeutic efficacy of MRgFUS and 5-ALA.

Information on biomagnification of alternative brominated flame retardants (ABFRs) is limited and results are inconclusive, due in part to uncertainty in the understanding of predator/prey relationships. In the present study, a predatory fish, Channa argus, and several forage fish species were obtained from an ABFR contaminated site. The predator/prey relationships were identified based on fatty acid (FA) signatures in the predator and prey. Biomagnification factors (BMFs) for several ABFRs including decabromodiphenyl ethane (DBDPE), 1,2‑bis(2,4,6‑tribromophenoxy) ethane (BTBPE), hexabromobenzene (HBB), pentabromotoluene (PBT), and pentabromoethylbenzene (PBEB) were estimated based on the identified predator/prey relationships. The results showed that crucian carp was the main prey of C. argus, contributing to 71%-100% to its total diet. The mean BMFs for DBDPE, BTBPE, and HBB were 0.06, 0.40, and 0.91, respectively, indicating trophic dilution of these ABFRs. However, biomagnification of PBT and PBEB, with BMFs of 2.09 and 2.13, respectively, was observed. The BMFs for PBT, PBEB and HBB were comparable to or even higher than those for some polybrominated diphenyl ether (PBDE) congeners estimated in the same individual predator, indicating that these emerging pollutants may pose significant environmental risks. The BMFs for ABFRs and PBDEs were significantly and negatively correlated to the log K OW s of these chemicals, suggesting that the biomagnification of these chemicals was depressed due to their superhydrophobic nature., (Copyright © 2019 The Authors. RhoGTPase activation of non-muscle myosin II regulates cell division, extrusion, adhesion, migration, and tissue morphogenesis. However, the regulation of myosin II and mechanotransduction is not straightforward. Increasingly, the role of myosin II on the feedback regulation of RhoGTPase signaling is emerging. Indeed, myosin II controls RhoGTPase signaling through multiple mechanisms, namely contractility driven advection, scaffolding, and sequestration of signaling molecules. Here we discuss these mechanisms by which myosin II regulates RhoGTPase signaling in cell adhesion, migration, and tissue morphogenesis.

The wettability, microstructures, and bonding strength of infrared brazing Ti-15-3 and Ti 50 Ni 50 shape memory alloy using 72Ag-28Cu (wt.%) and 68.8Ag-26.7Cu-4.5Ti (wt.%) filler metals have been investigated. Only Ticusil ® active braze readily wets both Ti 50 Ni 50 and Ti-15-3 substrates. Wetting of eutectic 72Ag-28Cu melt on Ti 50 Ni 50 base metal is greatly ameliorated by adding 4.5 wt.% Ti into the alloy. The brazed Ti-15-3/BAg-8/Ti 50 Ni 50 joint consists of Cu-Ti intermetallics in the Ag-rich matrix. The formation of interfacial Cu(Ti,V) and (Cu x Ni 1-x ) 2 Ti intermetallics next to Ti-15-3 and Ti 50 Ni 50 substrates, respectively, is attributed to the wetting of both substrates. The brazed Ti-15-3/Ticusil ® /Ti 50 Ni 50 joint shows a vigorous reaction, which results in the formation of a large amount of Ti 2 Ni intermetallics in the joint. The maximum joint strengths using BAg-8 and Ticusil ® filler metals are 197 MPa and 230 MPa, respectively.

Severe pollution of organohalogen compounds (OHCs) including PBDEs, PCBs and DDTs is demonstrated in e-waste recycling sites and metropolis in South China, but relatively little is known of their impacts on wildlife that inhabits nature reserves in this region, especially those located at the neighborhood areas of e-waste recycling sites. In the present study, PBDEs, PCBs and DDTs as well as liver histological changes were examined in common kingfisher breeding at a nature reserve (Shimentai National Nature Reserve, SNNR) near a notorious e-waste recycling site in South China. Mean ∑PBDEs (84.9 ng/g lipid weight), ∑PCBs (397 ng/g) and ∑TEQs (total toxic equivalent of coplanar PCBs, 2.68 ng/g) concentrations in kingfishers from SNNR were approximately 2-, 5-, and 4-fold higher than those detected in a reference population, respectively; suggesting contamination of the e-waste-derived OHCs in SNNR. Mean ∑DDTs concentration (2150 ng/g) in kingfishers from SNNR was also higher (~2-fold) than that from the reference samples. While ∑DDTs dominated the composition of the OHCs at both sampling sites, ∑PCBs was also important in kingfishers from SNNR, averaging 15% of ∑OHCs. Histological examination of the liver showed steatosis occurred in 60% of the kingfishers from SNNR. Birds with hepatic steatosis had significantly (p = 0.03) higher ∑TEQs than those without steatosis. Similar trends were also found for ∑PCBs and ∑DDTs, although it is not statistically significant. It therefore seems likely that the hepatic steatosis were associated with the relatively high body-burden of OHCs, although the possibility of other factors resulting in hepatic steatosis cannot be ruled out., (Copyright © 2019 Elsevier B.V. The recrystallization behavior, grain growth kinetics, and corresponding hardness variation of homogenized and 80% cold-rolled FeCoNiCrPd, FeCoNiCrMn, and their quaternary/ternary FCC-structured high/medium entropy alloys (H/MEAs) annealed under different conditions were investigated. Experimental results indicate that the grain size and hardness of these H/MEAs follow the Hall-Petch equation, with the Hall-Petch coefficient K H value being mainly dominated by the alloy's stacking fault energy and shear modulus. The FeCoNiCrPd alloy exhibits the highest hardness of the H/MEAs at the same grain size due to the largest Young's modulus difference between Cr and Pd. The grain growth exponent n , kinetic constant k , and activation energy for grain growth Q G of all H/MEAs are calculated. The k can be expressed by the Arrhenius equation with Q G , which is attributed to the diffusion rate. The results demonstrate that the Q G values of these H/MEAs are much higher than those of conventional alloys; most notable is FeCoNiCrPd HEA, which has an unusually lattice distortion effect that hinders grain growth.

Infrared vacuum brazing of CoCrFeMnNi high entropy alloy (HEA) using BNi-2 and MBF601 fillers has been investigated. Both brazes show poor wettability at temperatures only 20 °C above their liquidus temperatures. However, the wettability of BNi-2 and MBF601 fillers on CoCrFeMnNi HEA is greatly improved with increasing the test temperatures, 50 °C above their liquidus temperatures. The BNi-2 brazed joints are dominated by Ni-rich matrix with huge CrB and a few tiny boride precipitates. Average shear strengths of joints increase with increasing brazing temperature and/or time, and fracture location changes from blocky CrB in the brazed zone to grain boundary boride in the substrate. The MBF601 brazed joints are composed of CoCrFeMnNi-based matrix, particles of B/Co/Cr/Fe/Mn/Ni/P compounds, and some phosphides form along the grain boundaries of the substrate. The specimen brazed with MBF601 filler foil at 1050 °C for 600 s has the highest average shear strength of 321 MPa, while that brazed at 1080 °C for 600 s has a lower average shear strength of 271 MPa due to the presence of solidification shrinkage voids.

This study examines three different types of laser carriers, including the phosphors in silicone (PiS), the phosphor in glass (PiG), and alumina-based ceramic binders (CP), for laser-phosphor lighting characterizations via a laser projector light source module. The thermal influence of heat spreading on these phosphor materials and their luminescence performance is also investigated. The conversion efficiency of PiS, PiG and CP was found to be 29.7%, 34.6% and 31.8%, with their corresponding maximum laser power operations of 3.9 W, 7.9 W, and 17.2 W, respectively. This work further correlates the maximum laser operation power with the thermal conductivity of luminescence material. From the optical engine perspective, it was found that CP exhibits the superior thermal conductivity of 17.0 W/m⋅K for slight hot-spot IR observation and higher laser power operation. This work finally delivers a CP device for 50.2W maximum laser operation with the operating temperature below 100 °C. The simulation is also carried out in order to examine spreading resistance's influence, when subject to convective boundary condition. CP's response sensitivity to heat transfer coefficient was found to be rather small.

Growing evidence demonstrates a possible response of specific microRNA (miRNA) to environmental pollutant stimuli in multiple biological processes. We previously reported that a persistent organic pollutant, decabromodiphenyl ether (BDE-209), can enhance Toll-like receptor 4 (TLR4)-dependent lipid uptake in THP-1 macrophages; whether miRNAs are involved in this process remains unclear. In the present study, we investigated the levels of several miRNAs related to TLR4 signaling, including miRs-9, -21, -27b, -125b, -132, -146a, -147, -155, and -let-7e, in THP-1 macrophages after stimulation by BDE-209 and oxidized low-density lipoprotein. The results showed that the levels of miR-21 were significantly suppressed by BDE-209 at concentrations of 6.25, 12.5 and 25 μM, in a dose-dependent manner; whereas there was no significant changes for the other miRNAs investigated. Moreover, the suppression of miR-21 was accompanied by an upregulated TLR4 expression, at both mRNA and protein levels. Further analysis showed that the up-regulated TLR4 induced by BDE-209 was inhibited in macrophages transfected with miR-21 mimic; meanwhile opposite results were exhibited when an anti-miR-21 inhibitor was transfected to the macrophages. Additionally, transfection with miR-21 mimic effectively attenuated BDE-209-induced lipid accumulation in macrophages. Together, these data illustrate that miR-21 inhibits BDE-209-triggered lipid accumulation in macrophages through down-regulating TLR4 expression., (Copyright © 2018 Elsevier Ltd. The rudimentary recycling of electronic waste (e-waste) has been banned in China since the late 2000s, leaving many abandoned e-waste sites. However, knowledge is limited on the concentrations and fates of the e-waste derived contaminants such as polychlorinated biphenyls (PCBs) in these abandoned sites. In this work, we assessed the temporal trend of PCB concentrations in the year 2010, 2012, and 2016 at an abandoned e-waste site in South China, using apple snail as a bioindicator. The mean ∑PCBs concentrations in apple snails sampled in 2016 (53.2 ng/g dry weight) was approximately 11-fold higher than that (4.68 ng/g dry weight) in apple snails from a reference site. The result suggested that the abandoned e-waste recycling site was still heavily polluted by PCBs, despite of the fact that crude e-waste recycling processes have been prohibited for nearly 10 years. The concentrations of ∑PCBs were significantly decreased in 2016 compared to those in 2010 (mean: 115 ng/g dry weight) and 2012 (mean: 92.3 ng/g dry weight), but there were no significant differences in the concentrations between 2010 and 2012. Regarding the congener profiles, the contributions of lower chlorinated congeners (tri- and tetra-PCBs) in the snails tended to be higher over the years. The ∑PCBs in snails were significantly correlated with those in soils. Additionally, PCB profiles in snails resembled those in soils. These results suggested that apple snails can be used as an ideal bioindicator for PCBs in the paddy soils. Field determined biota-soil accumulation factors (BSAFs) for PCBs ranged from 0.31 to 1.9, with most of the values being 1-2; indicating that theoretical BSAFs can be used to predict the bioaccumulation of PCBs in the snails with a reasonable degree of certainty., (Copyright © 2018 Elsevier B.V. There is growing evidence that exposure to persistent organic pollutants (POPs) is statistically associated with incidence of cardiovascular disease (CVD) or its risk factors. Decarbromodiphenyl ether (BDE-209) is a new POP which exists extensively in human tissues, but its potential effects on CVD have so far received less focus. The adhesion of circulating monocytes to endothelial cells is one of the critical underlying steps in the initiation and development of CVD. In the present study, we investigated the effect of BDE-209 on the adhesion of THP-1 monocytes to human aortic endothelial cells (HAECs) and identified the molecular mechanisms involved. Our results showed that 6.25, 12.5 and 25 µM of BDE-209 exposures caused significant increases in monocyte-endothelial cell adhesion, in a dose-dependent manner. Mechanistically, BDE-209 exposure increased the expression of intercellular adhesion molecule-1 (ICAM-1). Moreover, the up-regulation of ICAM-1 was accompanied by a decrease in the expression of microRNA-141 (miR-141). Furthermore, the up-regulation of ICAM-1 and the increased adhesion induced by BDE-209 could be reversed by miR-141 supplement. Objectives: To explore the feasibility of preoperative prediction of vascular invasion (VI) in breast cancer patients using nomogram based on multiparametric MRI and pathological reports., Methods: We retrospectively collected 200 patients with confirmed breast cancer between January 2016 and January 2018. All patients underwent MRI examinations before the surgery. VI was identified by postoperative pathology. The 200 patients were randomly divided into training (n = 100) and validation datasets (n = 100) at a ratio of 1:1. Least absolute shrinkage and selection operator (LASSO) regression was used to select predictors most associated with VI of breast cancer. A nomogram was constructed to calculate the area under the curve (AUC) of receiver operating characteristics, sensitivity, specificity, accuracy, positive prediction value (PPV) and negative prediction value (NPV). We bootstrapped the data for 2000 times without setting the random seed to obtain corrected results., Results: VI was observed in 79 patients (39.5%). LASSO selected 10 predictors associated with VI. In the training dataset, the AUC for nomogram was 0.94 (95% confidence interval [CI]: 0.89-0.99, the sensitivity was 78.9% (95%CI: 72.4%-89.1%), the specificity was 95.3% (95%CI: 89.1%-100.0%), the accuracy was 86.0% (95%CI: 82.0%-92.0%), the PPV was 95.7% (95%CI: 90.0%-100.0%), and the NPV was 77.4% (95%CI: 67.8%-87.0%). In the validation dataset, the AUC for nomogram was 0.89 (95%CI: 0.83-0.95), the sensitivity was 70.3% (95%CI: 60.7%-79.2%), the specificity was 88.9% (95%CI: 80.0%-97.1%), the accuracy was 77.0% (95%CI: 70.0%-83.0%), the PPV was 91.8% (95%CI: 85.3%-98.0%), and the NPV was 62.7% (95%CI: 51.7%-74.0%). The nomogram calibration curve shows good agreement between the predicted probability and the actual probability., Conclusion: The proposed nomogram could be used to predict VI in breast cancer patients, which was helpful for clinical decision-making., (Copyright © 2018 Elsevier B.V. Changes in bone mineral density and bone metabolic indexes in a model of ankylosing spondylitis (AS) mice complicated with osteoporosis (OP) were investigated. BLAB/c mice were used as the subjects. AS was induced using proteoglycan, and OP was induced using tail suspension method. The mice were randomly divided into four groups: AS group, OP group, AS + OP group and negative control group. Changes in bone mineral density, bone strength, serum calcium (Ca), phosphorus, alkaline phosphatase (ALP) and tartrate-resistant acid phosphatase (TRACP), in mice of each group were detected and compared. There were statistically significant differences in bone mineral density and bone strength among groups. Compared with the negative control group, bone mineral density and bone strength in the AS, the OP and the AS + OP groups were significantly decreased, and the lowest bone mineral density and bone strength were found in the AS + OP group (P<0.05). There were no significant differences in bone mineral density and bone strength between the AS group and the OP group. Significant differences in serum Ca, ALP and TRACP but not in serum phosphorus were found among groups. Compared with the control group, serum levels of Ca and TRACP in the AS, the OP and the AS + OP groups were significantly increased, while levels of ALP were obviously decreased (P<0.05). Bone destruction in AS mice complicated with osteoarthritis was more serious than that in mice with simple AS.

Leptomeningeal metastases (LM) are a serious complication of cancer in the central nervous system (CNS) and are diagnosed in approximately 5% of patients with solid tumors. Effective treatment using systemically administered therapeutics is hindered by the barriers of the CNS. Ultrasound can mediate delivery of drugs through these barriers. The goal of this study was to test the feasibility of using ultrasound-mediated drug delivery to improve the treatment of LM. LM was induced in the spinal cord of athymic rats by injecting HER2-expressing breast cancer cells into the subarachnoid space of the thoracic spine. Animals were divided into three groups: no treatment (n = 5), trastuzumab only (n = 6) or trastuzumab + focused ultrasound + microbubbles (FUS + MBs) (n = 7). Animals in groups 2 and 3 were treated weekly with intravenous trastuzumab +/- FUS + MBs for three weeks. Suppression in tumor growth was qualitatively observed by MRI in the group receiving ultrasound, and was confirmed by a significant difference in the tumor volume measured from the histology data (25 ± 17 mm 3 vs 8 ± 5 mm 3 , p = 0.04 in the trastuzumab-only vs trastuzumab + FUS + MBs). This pilot study demonstrates the potential of ultrasound-mediated drug delivery as a novel treatment for LM. Future studies will extend this work to larger cohorts and the investigation of LM arising from other cancers.
