Mesenchymal stromal cells (MSCs) are self-renewing, culture-expandable adult stem cells that have been isolated from a variety of tissues, and possess multipotent differentiation capacity, immunomodulatory properties, and are relatively non-immunogenic. Due to this unique set of characteristics, these cells have attracted great interest in the field of regenerative medicine and have been shown to possess pronounced therapeutic potential in many different pathologies. MSCs' mode of action involves a strong paracrine component resulting from the high levels of bioactive molecules they secrete in response to the local microenvironment. For this reason, MSCs' secretome is currently being explored in several clinical contexts, either using MSC-conditioned media (CM) or purified MSC-derived extracellular vesicles (EVs) to modulate tissue response to a wide array of injuries. Rather than being a constant mixture of molecular factors, MSCs' secretome is known to be dependent on the diverse stimuli present in the microenvironment that MSCs encounter. As such, the composition of the MSCs' secretome can be modulated by preconditioning the MSCs during in vitro culture. This manuscript reviews the existent literature on how preconditioning of MSCs affects the therapeutic potential of their secretome, focusing on MSCs' immunomodulatory and regenerative features, thereby providing new insights for the therapeutic use of MSCs' secretome.

Irreversible myocardial damage happens in the presence of prolonged and severe ischemia. Several phenomena protect the heart against myocardial infarction and other adverse outcomes of ischemia and reperfusion (IR), namely: hibernation related to stunned myocardium, ischemic preconditioning (IPC), ischemic post-conditioning, and their pharmacological surrogates. Ischemic preconditioning consists in the induction of a brief IR to reduce damage of the tissue caused by prolonged and severe ischemia. Nitric oxide (NO) signaling plays an essential role in IPC. Nitric oxide-sensitive guanylate cyclase/cyclic guanosine-3′,5′-monophosphate (cGMP)-dependent protein kinase type I-signaling pathway protects against the IR injury during myocardial infarction. Mitochondrial ATP-sensitive and Ca2+-activated K+ channels are involved in NO-mediated signaling in IPC. Independently of the cGMP-mediated induction of NO production, S-nitrosation represents a regulatory molecular mechanism similar to phosphorylation and is essential for IPC. Unlike conditioning phenomena, the mechanistic basis of myocardial stunning and hibernation remains poorly understood. In this review article, we hypothesize that the disruption of electrical syncytium of the myocardium may underly myocardial stunning and hibernation. Considering that the connexins are the building blocks of gap junctions which represent primary structural basis of electrical syncytium, we discuss data on the involvement of connexins into myocardial conditioning, stunning, and hibernation. We also show how NO-mediated signaling is involved in myocardial stunning and hibernation. Connexins represent an essential element of adaptation phenomena of the heart at the level of both the cardio- myocytes and the mitochondria. Nitric oxide targets mitochondrial connexins which may affect electrical syncytium continuum in the heart. Mitochondrial connexins may play an essential role in NO-dependent mechanisms of myocardial adaptation to ischemia.

The tetrazolium test is considered a promising alternative for quick evaluation of viability and vigor of seeds. The adequatio of tetrazolium test methodology for crambe seeds, specie that presents high potential for biodiesel production, may accelerate the determination of quality of different lots. With the aim to verify the ideal conditions for the tetrazolium test for crambe seeds, five lots of cultivar FMS Brilhante were submitted to the imbibition between paper in water at 30°C for three hours and after the longitudinal slicing, seeds were soaked in tetrazolium solution at concentrations of 0.075%; 0.5% and 1.0% at 30°C for 18 h and 24 h. It was observed that in the concentration of 0.75% seeds got an adequate coloration and with 0.5% and 1.0% the coloration was too intense. The imbibition of seeds between paper for 3 hours, followed by the longitudinal slicing and the utilization of 0.075% concentration for 18h or 24 hours was efficient to the evaluation of viability of crambe seeds.

Purpose: To determine and compare the effects of pre-conditioning and post-conditioning towards gamma radiation responses in human cancer cells and keratinocytes. Material and methods: The clonogenic survival of glioblastoma cells (T98G), keratinocytes (HaCaT), and colorectal carcinoma cells (HCT116 p53+/+ and p53−/−) was assessed following gamma ray exposure from a Cs-137 source. The priming dose preceded the challenge dose in pre-conditioning whereas the priming dose followed the challenge dose in post-conditioning. The priming dose was either 5 mGy or 0.1 Gy. The challenge dose was 0.5-5 Gy. Results: In both pre- and post-conditioning where the priming dose was 0.1 Gy and the challenge dose was 4 Gy, RAR developed in T98G but not in HaCaT cells. In HCT116 p53+/+, pre-conditioning had either no effect or a radiosensitizing effect and whereas post-conditioning induced either radiosensitizing or radioadaptive effect. The different observed outcomes were dependent on dose, the time interval between the priming and challenge dose, and the time before the first irradiation. Post-conditioning effects could occur with a priming dose as low as 5 mGy in HCT116 p53+/+ cells. When HCT116 cells had no p53 protein expression, the radiosensitizing or radioadaptive response by the conditioning effect was abolished. Conclusions: The results suggest that radiation conditioning responses are complex and depend on at least the following factors: the magnitude of priming/challenge dose, the time interval between priming and challenge dose, p53 status, cell seeding time prior to the first radiation treatment. This work is the first parallel comparison demonstrating the potential outcomes of pre- and post-conditioning in different human cell types using environmentally and medically relevant radiation doses. The enzymatic hydrolysis still stands as a challenge regarding the use of renewable feedstocks, as well as of lignocellulosic materials. The present work reports optimization of swollenin addition to pretreated sugarcane bagasse, in a step prior to the enzymatic hydrolysis, named pre-conditioning. Also, it describes further fermentation of the hydrolyzed material to D-lactic acid. Trichoderma harzianum enzymatic cocktail was produced from pretreated sugarcane bagasse, while swollenin was produced using Aspergillus niger as a host for recombinant protein expression. For pre-conditioning optimization, a Central Composite Rotatable Design was applied and validated. The optimized conditions were used to produce the hydrolysate that was fermented to D-lactic acid by Lactobacillus coryniformis subsp. torquens. It could be observed that pre-conditioning effectively leads to improvements in glucose release: 62% of hydrolysis efficiency. Besides, it was possible to use the produced hydrolysate to obtain D-lactic acid, with a yield of 1.34 g/g and a productivity of 1.59 g/L h−1. [ABSTRACT FROM AUTHOR]

Before cracking, subjecting water chestnuts to pre-conditioning is a common practice. However, the existing practice of pre-conditioning has no scientific rationale and therefore leads to low flour recovery and poor quality. Thus, the aim of the present research work was to optimize the pre-conditioning process for water chestnuts and to analyze the efficacy of optimized process as a means to increase the resistant starch content and reduce the glycemic index in water chestnut flour. Central composite rotatable design was used to determine the effect of water chestnut:water ratio, water temperature and pre-conditioning time on yield and brightness (L* value) of flour. Optimization of pre-conditioning process was done by response surface of desirability function. Regression models for both the product responses were highly significant (p < 0.0001) with high coefficient of determination (R2 > 0.95). Scanning electron microscopy (SEM) showed compact, smooth and oval shaped starch granules with crystalline network in preconditioned water chestnut flour. FTIR studies also indicated higher absorbance ratio of 1047/1022 cm−1 for preconditioned water chestnut flour which further authenticated the SEM findings. Subjecting water chestnuts to optimized conditions of pre-conditioning (i.e., water chestnut:water ratio 1:1.5; water temperature 87.85̊C and pre-condition time 45 min), also increased the resistant starch content and lowered the glycemic index and glycemic load of the water chestnut flour. Abstract The main objective of the full waveform inversion (FWI) is to deliver a velocity model which produces accurate and high resolution depth images. An integral part of FWI is the iterative optimization process aiming at minimizing an objective function describing the misfit between the observed and modelled data with respect to the velocity model parameters. However, FWI commonly suffers from local minima due to errors in data fitting and the starting velocity model. In this study, we develop a methodology to enhance convergence and mitigate the risk of having solutions falling into local minima. We employ optical flow to decompose the source and residual receiver wavefields and map them into the subsurface angle domain. Having evaluated the propagation vectors of the respective cleaner wavefields, subsurface reflection and azimuth angles can be robustly computed. Using such information we can apply angle-dependent preconditioning within the inversion algorithm. During a conventional FWI workflow, larger angles (i.e., transmission wavepaths) are mainly responsible for long wavelength velocity anomalies whereas narrow angles (i.e., reflection wavepaths) influence short wavelength velocity anomalies. In the proposed FWI methodology, we apply an optimized weighting function based on these estimated subsurface reflection angles in order to weight certain angle directions needed for gradient estimation using in parameter updating. Following a hierarchical approach, allowing for larger angles at the early stage of the FWI application, we initially solve for the smooth and longer wavelength velocity updates. Relaxing the opening angle from large to mid to narrow, the gradient direction will first estimate long wavelength velocity model updates (transmission-based wavepaths), following by short wavelength velocity model updates (reflection-based wavepaths). Moreover, angle-dependent-based gradient preconditioning can reduce cycle skips and improve convergence during FWI iterations. Structure innovation and optimization of polymer electrolyte fuel cell (PEFC) catalyst layers is instrumental in improving catalyst utilization and effectiveness. Electrospinning technique has been recently used to fabricate nanofiber electrodes for PEFCs with improved performance compared to electrosprayed electrodes. However, the origins for the performance improvement are yet poorly understood. This study examined the evolution of the electrochemically active surface area (ECSA) and electrochemical impedance spectroscopy (EIS) during the pre‐conditioning process, and compared the performance of electrospun nanofiber electrodes with electrosprayed electrodes within a wide range of operating conditions. ECSA increases, and both mass transfer resistance and charge transfer resistance decrease monotonically during the pre‐conditioning process of electrospun nanofiber electrodes. Such effects are attributed to the dissolution of poly(acrylic acid) (PAA) and the ensuing change of the interface between Pt and the ionomer. After pre‐conditioning, the electrospun nanofiber electrodes outperformed the electrosprayed electrodes with the same Pt‐loadings at 80 °C, whereas the electrosprayed electrodes have higher cell voltages at high current densities at 60 °C and 70 °C in high relative humidity (RH) range. The change in the relative performance of the two types of electrodes is attributed to the competing effects of electrode thickness and pore sizes on cathode gas transport. [ABSTRACT FROM AUTHOR]

Abstract: We previously demonstrated that pre‐conditioning with CpG oligonucleotide (ODN) 1668 induces quick up‐regulation of gene expression 3 hours post‐murine myocardial ischaemia/reperfusion (I/R) injury, terminating inflammatory processes that sustain I/R injury. Now, performing comprehensive microarray and biocomputational analyses, we sought to further enlighten the “black box” beyond these first 3 hours. C57BL/6 mice were pretreated with either CpG 1668 or with control ODN 1612, respectively. Sixteen hours later, myocardial ischaemia was induced for 1 hour in a closed‐chest model, followed by reperfusion for 24 hours. RNA was extracted from hearts, and labelled cDNA was hybridized to gene microarrays. Data analysis was performed with BRB ArrayTools and Ingenuity Pathway Analysis. Functional groups mediating restoration of cellular integrity were among the top up‐regulated categories. Genes known to influence cardiomyocyte survival were strongly induced 24 hours post‐I/R. In contrast, proinflammatory pathways were down‐regulated. Interleukin‐10, an upstream regulator, suppressed specifically selected proinflammatory target genes at 24 hours compared to 3 hours post‐I/R. The IL1 complex is supposed to be one regulator of a network increasing cardiovascular angiogenesis. The up‐regulation of numerous protective pathways and the suppression of proinflammatory activity are supposed to be the genetic correlate of the cardioprotective effects of CpG 1668 pre‐conditioning. An alternative control method to the use of chemical insecticides against soil dwelling insect pests is the application of entomopathogenic nematodes formulated in alginate beads for enhanced shelf life. The aim was to compare the benefit on nematode survival and infectivity of: (i) pre-conditioning of juveniles, and (ii) coating of alginate beads. The nematodes Steinernema glaseri, Steinernema carpocapsae, and Heterorhabditis bacteriophora were reproduced in last instar larvae of the wax moth Galleria mellonella and the infective juveniles emerged were subjected to two capture treatments: white traps and plaster of Paris, the latter was utilized as a pre-conditioning treatment. A total of 1,000 infective juveniles were formulated in each sodium alginate bead with or without an alginate coating. The beads were stored at 23 ± 3°C and a bidistilled water suspension of nematodes was set as a control. The survivorship of these formulates and their infectivity on Galleria mellonella larvae were evaluated every 7 d post-formulation (dpf). In beads without pre-conditioning juveniles nor coating, Steinernema carpocapsae had the higher survival rate (58.8%), the longest survival time (28 dpf) and the higher infectivity (100%) as well. Pre-conditioning improved the survival and infectivity of Steinernema glaseri by 22.5% at 21 dpf and 70.0% at 14 dpf, respectively. Alginate coating increased survival (21.7%) and infectivity (95%) of Heterorhabditis bacteriophora for up to 28 dpf. The combination of pre-conditioning and coating slightly favored the survival (10% for up to 14 dpf) of Steinernema glaseri and infectivity (100% for up to 35 dpf) of Steinernema carpocapsae. Non pre-conditioned Steinernema carpocapsae formulated in uncoated beads was the combination with better performance in survivorship (58.8%) and infectivity (100%) at 28 dpf. Despite the widespread use of chemical and biological nano-silver are in industry, their side effects on hepatocytes have been less studied. On the other hand, different types of physical activities may increase liver resistance to toxins. Therefore, the aim of this study was to evaluate the resistance of hepatocytes to receiving chemical versus biological silver nanoparticles in aerobic and anaerobic pre-conditioned rats. 45 male Wistar rats with similar average range of age (8–12 weeks) and weight (180–220 g), were randomly divided normally into 9 groups, including Control (C), Aerobic (A), Anaerobic (AN), Biological nano-silver (BNS), Chemical nano-silver (CNS), Biological nano-silver + Aerobic (BNS+A), Biological nano-silver + Anaerobic (BNS+AN), Chemical nano-silver + Aerobic (CNS+A) and Chemical nano-silver + Anaerobes (CNS+AN). Prior to injection, rats trained on a rodent treadmill, 10 weeks, 3 sessions per week, according to aerobic and anaerobic protocols.Then, 48 h after the last training session, the rats received 10 % of their body weight, chemical and biological nanosilver intraperitoneally. Liver enzymes (ALT, AST, and ALP) and liver tissue were sent to the relevant laboratories for further evaluation. Results showed that the weight of rats in all groups of physical pre-conditioning, decreased comparison to the control and non-exercise groups, and this decrease was much greater in the anaerobic group (p -value=0.045). Also, the distance traveled in the progressive endurance running a test on a rodent treadmill, increased significantly in the training groups compared to the nano-exercise and control groups (p -value=0.001). Also, the results showed that the level of ALT in chemical nano-silver (p -value=0.004) and biological nano-silver (p -value=0.044), increased significantly compared to other groups. Also, histopathological results showed that nano-silver injection affects the structure of the liver of male Wistar rats and causes inflammation, hyperemia and destruction of liver cells, especially in chemical nano-silver. The results of the present study showed that chemical silver nanoparticles cause liver damage more than comparison biological ones. Also, physical pre-conditioning increases hepatocyte resistance to toxic nanoparticle doses and aerobic preparation appears to be more effective than anaerobic. [ABSTRACT FROM AUTHOR]

Team-sport athletes and coaches use varied strategies to enhance repeated-sprint ability (RSA). Aside from physical training, a well-conducted warm-up enhances RSA via increased oxidative metabolism. Strategies that impede blood flow could potentiate the effects of a warm-up due to their effects on the endothelial and metabolic functions. This study investigated whether performing a warm-up combined with blood-flow restriction (WFR) induces ergogenic changes in blood volume, muscle oxygenation, and RSA. In a pair-matched, single-blind, pre-post parallel group design, 15 American football players completed an RSA test (12 × 20 m, 20 s rest), preceded by WFR or a regular warm-up (SHAM). Pressure was applied on the athletes’ upper thighs for ≈15 min using elastic bands. Both legs were wrapped at a perceived pressure of 7 and 3 out of 10 in WFR and SHAM, respectively. Changes in gastrocnemius muscle oxygen saturation (SmO2) and total hemoglobin concentration ([THb]) were monitored with near-infrared spectroscopy. Cohen’s effect sizes (ES) were used to estimate the impact of WFR. WFR did not clearly alter best sprint time (ES −0.25), average speed (ES 0.25), total time (ES −0.12), and percent decrement score (ES 0.39). While WFR did not meaningfully alter average SmO2 and [THb], the intervention clearly increased the maximum [THb] and the minimum and maximum SmO2 during some of the 12 sprint/recovery periods (ES 0.34−1.43). Results indicate that WFR positively alters skeletal muscle hemodynamics during an RSA test. These physiological changes did not improve short-term RSA, but could be beneficial to players during longer activities such as games.

Pharmacologically-induced pre- and post-conditioning represent attractive therapeutic strategies to reduce ischaemia/reperfusion injury during cardiac surgery and following myocardial infarction. We have previously reported that transglutaminase 2 (TG2) activity is modulated by the A 1 adenosine receptor and β 2 -adrenoceptor in H9c2 cardiomyoblasts. The primary aim of this study was to determine the role of TG2 in A 1 adenosine receptor and β 2 -adrenoceptor-induced pharmacological pre- and post-conditioning in the H9c2 cells. H9c2 cells were exposed to 8 h hypoxia (1% O 2 ) followed by 18 h reoxygenation, after which cell viability was assessed by monitoring mitochondrial reduction of MTT, lactate dehydrogenase release and caspase-3 activation. N 6 -cyclopentyladenosine (CPA; A 1 adenosine receptor agonist), formoterol (β 2 -adrenoceptor agonist) or isoprenaline (non-selective β-adrenoceptor agonist) were added before hypoxia/reoxygenation (pre-conditioning) or at the start of reoxygenation following hypoxia (post-conditioning). Pharmacological pre- and post-conditioning with CPA and isoprenaline significantly reduced hypoxia/reoxygenation-induced cell death. In contrast, formoterol did not elicit protection. Pre-treatment with pertussis toxin (G i/o -protein inhibitor), DPCPX (A 1 adenosine receptor antagonist) or TG2 inhibitors (Z-DON and R283) attenuated the A 1 adenosine receptor-induced pharmacological pre- and post-conditioning. Similarly, pertussis toxin, ICI 118,551 (β 2 -adrenoceptor antagonist) or TG2 inhibition attenuated the isoprenaline-induced cell survival. Knockdown of TG2 using small interfering RNA (siRNA) attenuated CPA and isoprenaline-induced pharmacological pre- and post-conditioning. Finally, proteomic analysis following isoprenaline treatment identified known (e.g. protein S100-A6) and novel (e.g. adenine phosphoribosyltransferase) protein substrates for TG2. These results have shown that A 1 adenosine receptor and β 2 -adrenoceptor-induced protection against simulated hypoxia/reoxygenation occurs in a TG2 and G i/o -protein dependent manner in H9c2 cardiomyoblasts. One of the exciting advances in modern medicine and life science is cell-based neurovascular regeneration of damaged brain tissues and repair of neuronal structures. The progress in stem cell biology and creation of adult induced pluripotent stem (iPS) cells has significantly improved basic and pre-clinical research in disease mechanisms and generated enthusiasm for potential applications in the treatment of central nervous system (CNS) diseases including stroke. Endogenous neural stem cells and cultured stem cells are capable of self-renewal and give rise to virtually all types of cells essential for the makeup of neuronal structures. Meanwhile, stem cells and neural progenitor cells are well-known for their potential for trophic support after transplantation into the ischemic brain. Thus, stem cell-based therapies provide an attractive future for protecting and repairing damaged brain tissues after injury and in various disease states. Moreover, basic research on naïve and differentiated stem cells including iPS cells has markedly improved our understanding of cellular and molecular mechanisms of neurological disorders, and provides a platform for the discovery of novel drug targets. The latest advances indicate that combinatorial approaches using cell based therapy with additional treatments such as protective reagents, preconditioning strategies and rehabilitation therapy can significantly improve therapeutic benefits. In this review, we will discuss the characteristics of cell therapy in different ischemic models and the application of stem cells and progenitor cells as regenerative medicine for the treatment of stroke. This paper presents and discusses measurements performed to study the interaction mechanism of calcium and sulfate with galena and their effect on xanthate adsorption. The results of collector adsorption show that regardless of the solution pH (5.5, 7.5 and 9.5), the presence of calcium and sulfate during the mineral pre-conditioning, decreases the adsorption of isopropyl xanthate onto galena. Microflotation measurements carried out at pH 9.5 show that at low concentrations of calcium and sulfate (below about 0.005 mol/L), the recovery of galena is not affected. However, as the concentration of both ions increases, flotation recovery substantially decreases. Contact angle measurements corroborate this behavior: a relatively large contact angle in the absence of calcium and sulfate (i.e., 64°), and a significant decrease when these species are present (i.e., ca. 45°). The observed behavior suggests that calcium ion probably has a certain chemical affinity for the active sites of the galena, which hinders the occurrence of ion exchange collector-sulfate or collector-hydroxyl to form lead xanthate (PbX 2 ), which is the species responsible for the enhanced hydrophobicity of the mineral. On the other hand, the adverse effect of the sulfate is probably due to the formation of a layer of lead sulfate on the galena surface, which acts as a barrier that hinders the interaction of the collector with the metallic sites of the mineral lattice, thus negatively affecting the chemisorption mechanism. Likewise, the decreasing of lead concentration in solution, in equilibrium with the precipitated PbSO 4 and the sulfate, limits the precipitation/coagulation of lead xanthate in the vicinity of the interface. The recent rise in power conversion efficiencies reported for perovskite solar cells has been a remarkable development in photovoltaics research. It is now pressing that the technology transitions from a research phenomenon to a real-world deployable device: this will require both robust methods for efficiency measurement, and accurate models for performance variation at different conditions. However, the generally slow response of perovskite solar cells to changes in voltage bias and irradiance, and the susceptibility of these cells to degradation, presents significant challenges. In this paper, we investigate current and voltage stabilisation of planar CH3NH3PbI3 perovskite solar cells and observe remarkably large variations in stabilisation time depending on exposure history. To address this, we demonstrate a dynamic approach that continues device pre-conditioning until pre-determined stability criteria are met. This approach is then employed to obtain measurements of short-circuit current and open-circuit voltage temperature coefficients under quasi-steady-state conditions for perovskite devices and a control monocrystalline silicon cell. The obtained open-circuit voltage temperature coefficient for the perovskite is −2700 ppm/°C, which interestingly, is similar to typically reported values for crystalline silicon devices. It is shown that the implemented approach can successfully differentiate between transient responses to the onset of illumination and true temperature related changes. We also find new manifestations of the complex transient processes that occur in perovskite devices. These observations highlight the importance of sophisticated characterisation approaches for correct characterisation of the performance of perovskite solar cells. Helium has been classified as a kind of inert gas that is not effortless to spark chemical reactions with other substances in the past decades. Nevertheless, the cognition of scientists has gradually changed accompanied with a variety of studies revealing the potential molecular mechanism underlying organ-protection induced by helium. Especially, as a non-anesthetic gas which is deficient of relevant cardiopulmonary side effects, helium conditioning is recognized as an emerging and promising approach to exert favorable effects by mimicking the cardioprotection of anesthetic gases or xenon. Aim The isolated, retrogradely perfused heart (modified Langendorff model) is a widely used method in experimental heart research. The presence of an intraventricular balloon is necessary to get functional measurements. We have previously shown that the balloon induces phosphorylation of some suggested cardioprotective mitogen-activated protein kinases ( MAPK): P38- MAPK, ERK 1/2 and JNK. We hypothesized that the balloon could influence cardioprotection, protect against ischaemia reperfusion injury and interfere with coronary flow. Methods and Results Isolated mouse hearts were perfused for 5, 10, 20, 40 and 60 min with a balloon in the left ventricle. We found a wavelike phosphorylation of all MAPK while AKT displayed a gradual dephosphorylation when compared to non-perfused hearts. Hearts were subjected to 20 min of stabilization with or without the balloon, followed by 35 min of ischaemia and 120 min of reperfusion. Although the MAPK were phosphorylated, the infarcts were larger in the balloon group. When the balloon was present during the entire protocol, compared to removal at the end of ischaemia, the infarct size was also larger, especially in the endocardial layer. The balloon reduced post-ischaemic endocardial coronary flow, despite a higher average flow, indicating a hyperperfused epicard. Blocking the balloon-induced ERK 1/2 phosphorylation during stabilization did not affect infarct size. The effect of post-conditioning was influenced by the balloon, showing reduced infarct size when the balloon was present. Objectives Basic military training is physically and psychologically demanding placing recruits at high risk of injury and premature discharge. This study aimed to identify risk factors for lower leg, ankle and foot injury in Maltese military recruits during basic training. Design This was a prospective cohort study. Setting An armed forces barracks. Participants 127 recruits commencing one basic military training course agreed to participate in the study. The cohort comprised 114 males and 13 females with a mean age of 21.7 ± 2.4 years. Main outcome measures All injuries to the lower leg, ankle and foot were recorded using the Orchard Sports Injury Classification System. Injuries were analysed for associations with fitness scores, smoking status, body mass index and foot type. Results A total of 34 (26.2%) recruits sustained at least one injury, with 10 recruits (7.9%) terminating their training prematurely (three due to musculoskeletal injury). Smoking history, body mass index and foot type were not associated with injury risk. Lower fitness levels at the commencement of basic military training compared with fitness levels measured six months prior, were associated with higher injury risk. Conclusions Lower fitness at the commencement of basic training was associated with higher injury risk in army recruits. Thus, conditioning programmes aimed at improving recruit fitness should be considered within an injury prevention strategy. [ABSTRACT FROM AUTHOR]

Rapidly rising atmospheric CO2 concentrations are driving acidification in parallel with warming of the oceans. Future ocean acidification scenarios have the potential to impact coral growth and associated reef function, although reports suggest such affects could be reduced in adjacent seagrass habitats as a result of physio-chemical buffering. To-date, it remains unknown whether these habitats can actually support the metabolic function of a diverse range of corals. Similarly, whether mangroves provide the same ecological buffering service remains unclear. We examine whether reef-associated habitat sites (seagrass and mangroves) can act as potential refugia to future climate change by maintaining favorable chemical conditions (elevated pH and aragonite saturation state relative to the open-ocean), but by also assessing whether the metabolic function (photosynthesis, respiration and calcification) of important reef-building corals are sustained. We investigated three sites in the Atlantic, Indian and Pacific Oceans and consistently observed that seagrass beds experience an overall elevation in mean pH (8.15 ± 0.01) relative to the adjacent outer-reef (8.12 ± 0.03), but with periods of high and low pH. Corals in the seagrass habitats either sustained calcification or experienced an average reduction of 17.0 ± 6.1 % relative to the outer-reef. In contrast, mangrove habitats were characterized by a low mean pH (8.04 ± 0.01) and a relatively moderate pH range. Corals within mangrove-dominated habitats were thus pre-conditioned to low pH but with significant suppression to calcification (70.0 ± 7.3 % reduction relative to the outer-reef). Both habitats also experienced more variable temperatures (diel range up to 2.5°C) relative to the outer-reef (diel range less than 0.7°C), which did not correspond with changes in calcification rates. Here we report, for the first time, the biological costs for corals living in reef-associated habitats and characterize the environmental services these habitats may play in potentially mitigating the local effects of future ocean acidification.

Nephrotoxicity is the main side effect of antibiotics such as gentamicin. Preconditioning has been reported to protect against injuries as ischemia/reperfusion. The objective of the present study was to determine the effect of preconditioning with gentamicin on LLC-PK1 cells. Preconditioning was induced in LLC-PK1 cells by 24-h exposure to 2.0 mM gentamicin (G/IU). After 4 or 15 days of preconditioning, cells were again exposed to gentamicin (2.0 mM) and compared to untreated control or G/IU cells. Necrosis and apoptosis were assessed by acridine orange and HOESCHT 33346. Nitric oxide (NO) and endothelin-1 were assessed by the Griess method and available kit. Heat shock proteins were analyzed by Western blotting. After 15 days of preconditioning, LLC-PK1 cells exhibited a significant decrease in necrosis (23.5 ± 4.3 to 6.5 ± 0.3%) and apoptosis (23.5 ± 4.3 to 6.5 ± 2.1%) and an increase in cell proliferation compared to G/IU. NO (0.177 ± 0.05 to 0.368 ± 0.073 µg/mg protein) and endothelin-1 (1.88 ± 0.47 to 2.75 ± 0.53 pg/mL) production significantly increased after 15 days of preconditioning compared to G/IU. No difference in inducible HSP 70, constitutive HSC 70 or HSP 90 synthesis in tubular cells was observed after preconditioning with gentamicin. The present data suggest that preconditioning with gentamicin has protective effects on proximal tubular cells, that involved NO synthesis but not reduction of endothelin-1 or production of HSP 70, HSC 70, or HSP 90. We conclude that preconditioning could be a useful tool to prevent the nephrotoxicity induced by gentamicin.

Microwave pre-conditioning is considered to be the most promising complementary hard rock cutting method, with a strong potential to improve the advance rate and profitability of future mechanized tunnel boring and mining equipment. This technique is based on selective heating and differential volumetric expansion of minerals causing micro- and macrofracturing in rock. Over 700 samples consisting of granite and three different strength grades of concrete were irradiated and underwent a comprehensive geotechnical testing program. Besides longitudinal wave velocity, porosity, specific heat capacity, density, and temperature difference, additional geotechnical parameters are influenced by material-specific heating intervals: Unconfined compressive strength (UCS), Brazilian tensile strength (BTS), Point load index (PLI), CERCHAR abrasivity index (CAI), the LCPC abrasivity coefficient (LAK) and its breakability coefficient (LBK). Comparisons of thin sections of irradiated and non-irradiated materials show microscopic structural changes. A slight reduction of the UCS was observed after longer irradiation intervals for granite and concrete, while the weakening effect on the BTS and PLI is more pronounced. Rock strength was reduced by up to 48% for granite and 40–48% for concrete, which potentially leads to a substantial increase in penetration rate of a tunnel boring machine and reduced wear. Furthermore, we found a linear correlation between granite strength, sample temperature, and specific energy, resulting in a reduction of the PLI by 0.12% K−1. The tested concrete types showed strength reduction ratios between 0.21 and 0.28% K−1. By calculating efficiencies of the deployed energy, we showed that pre-conditioning with microwaves can be an effective and efficient tool to artificially reduce the strength of rocks in order to increase the advance rate of tunnel boring and mining machines. Protective responses are pivotal in aiding organismal persistence in complex, multi-stressor environments. Multiple-stressor research has traditionally focused on the deleterious effects of exposure to concurrent stressors. However, encountering one stressor can sometimes confer heightened tolerance to a second stressor, a phenomenon termed 'cross-protection'. Cross-protection has been documented in a wide diversity of taxa (spanning the bacteria, fungi, plant and animal kingdoms) and habitats (intertidal, freshwater, rainforests and polar zones) in response to many stressors (e.g. hypoxia, predation, desiccation, pathogens, crowding, salinity, food limitation). Remarkably, cross-protection benefits have also been shown among emerging, anthropogenic stressors, such as heatwaves and microplastics. In this Commentary, we discuss the mechanistic basis and adaptive significance of cross-protection, and put forth the idea that cross-protection will act as a 'pre-adaptation' to a changing world. We highlight the critical role that experimental biology has played in disentangling stressor interactions and provide advice for enhancing the ecological realism of laboratory studies. Moving forward, research will benefit from a greater focus on quantifying the longevity of cross-protection responses and the costs associated with this protective response. Although beneficial in determined contexts, eccentric exercise-induced muscle damage (EIMD) might be unwanted during training regimens, competitions and daily activities. There are a vast number of studies investigating strategies to attenuate EIMD response after damaging exercise bouts. Many of them consist of performing exercises that induce EIMD, consuming supplements or using equipment that are not accessible for most people. It appears that performing maximal isometric contractions (ISOs) 2-4 days prior to damaging bouts promotes significant attenuation of EIMD symptoms that are not related to muscle function. It has been shown that the volume of ISOs, muscle length in which they are performed, and interval between them and the damaging bout influence the magnitude of this protection. Additionally, it appears that this protection is not long-lived, lasting no longer than 4 days. Although no particular mechanisms for these adaptations were identified, professionals should consider applying this non-damaging stimulus before submitting their patients to unaccustomed exercised. However, it seems not to be the best option for athletes or relatively trained individuals. Future studies should focus on establishing if ISOs protect other populations (i.e., trained individuals) or muscle groups (i.e., knee extensors) against EIMD, as well as investigate different mechanisms for ISO-induced protection.

Background:Liver transplantation is the second most common transplant procedure in the United States. A leading cause of post-transplantation organ dysfunction is I/R injury. During I/R injury, the serine/threonine kinase Akt is activated, stimulating downstream mediators to promote cellular survival. Due to the cellular effects of Akt, therapeutic manipulation of the Akt pathway can help reduce cellular damage during hepatic I/R that occurs during liver transplantation.Objective:A full description of therapeutic options available that target Akt to reduce hepatic I/R injury has not been addressed within the literature. The purpose of this review is to illuminate advances in the manipulation of Akt that can be used to therapeutically target I/R injury in the liver.Methods:An in depth literature review was performed using the Scopus and PubMed databases. A total of 75 published articles were utilized for this manuscript. Terminology searched includes a combination of “hepatic ischemia/reperfusion injury”, “Akt/PKB”, “preconditioning” and “postconditioning.”Results:Four principal methods that reduce I/R injury include hepatic pre- and postconditioning, pharmacological intervention and future miRNA/gene therapy. Discussed therapies used serum alanine aminotransferase levels, liver histology and phosphorylation of downstream mediators to confirm the Akt protective effect.Conclusion:The activation of Akt from the reviewed therapies has resulted in predictable reduction in hepatocyte damage using the previously mentioned measurements. In a clinical setting, these therapies could potentially be used in combination to achieve better outcomes in hepatic transplant patients. Background As an indispensable clinical inhalation anesthetic, sevoflurane is widely used for peri-operative sedation. The neuroprotective effect of sevoflurane pre-conditioning against cerebral ischemia/reperfusion has been gradually realized, but the underlying mechanism during the early reperfusion period has not been established. Method Primary cultured cortical neurons were treated with 2% sevoflurane pre-conditioning for 30 min, exposed to oxygen-glucose deprivation for 90 min, and followed by 60 min of reperfusion (OGD/R). Additionally, neuronal cells were treated with an inhibitor of extracellular signal–related kinases 1 and 2 (Erk1/2) phosphorylation (PD98059), a mPTP opener (atractyloside), or a mPTP opening inhibitor (cyclosporine A) before sevoflurane pre-conditioning. Result Sevoflurane pre-conditioning decreased neuronal apoptosis (assessed by TUNEL), oxidative stress (assessed by malondialdehyde [MDA], superoxide dismutase [SOD], and heme oxygenase [HO]-1), and opening of mitochondrial permeability transition pores [mPTPs] (assessed by calcein-cobalt), but increased neuronal viability (assessed by MTT) and mitochondrial membrane potential (assessed by JC-1) after OGD/R exposure compared with OGD/R treatment alone. Pre-treatment with the mPTP opener and inhibitor of Erk1/2 phosphorylation abolished the protective effect induced by sevoflurane pre-conditioning. Pre-treatment with the mPTP opener attenuated the phosphorylation of Erk1/2 in mitochondria of neuronal cultures exposed to OGD/R induced by sevoflurane pre-conditioning. The mPTP opening inhibitor, like sevoflurane pre-conditioning, increased phosphorylation of Erk1/2 after OGD/R exposure, while PD98059 failed to reverse inhibition of mPTP opening in cultures exposed to OGD/R induced by sevoflurane pre-conditioning. Conclusion The neuroprotective mechanism of sevoflurane pre-conditioning might be associated with increased Erk1/2 phosphorylation in mitochondria via inhibition of mPTP opening in the early reperfusion period. Isoflurane, a volatile and inhalational anesthetic, has been extensively used in perioperative period for several decades. A large amount of experimental studies have indicated that isoflurane exhibits neuroprotective properties when it is administrated before or after (preconditioning and post-conditioning) neurodegenerative diseases (e.g., hypoxic ischemia, stroke and trauma). Multiple mechanisms are involved in isoflurane induced neuroprotection, including activation of glycine and γ-aminobutyric acid receptors, antagonism of ionic channels and alteration of the function and activity of other cellular proteins. Although neuroprotection provided by isoflurane is observed in many animal studies, convincing evidence is lacking in human trials. Acute brain injury is a critical and emergent condition in clinical settings, which needs to be addressed urgently. Commonly acute brain injuries include traumatic brain injury, ischemic and hemorrhagic strokes. Oxidative stress is a key contributor to the subsequent injuries and impedes the reparative process after acute brain injury; therefore, facilitating an anti-oxidative approach is important in the care of those diseases. Readiness to deliver and permeability to blood brain barrier are essential for the use of this purpose. Inhaled anesthetic gases are a group of such agents. In recent years, hyperbaric oxygen (HBO) has been used in the treatment of a lot of diseases such as decompression sickness, arterial gas embolism, carbon dioxide poisoning, soft tissue infection, refractory osteomyelitis, and problematic wound, but little is known about its application in liver transplantation. Although several studies have been conducted to investigate the protective effects of HBO on liver transplantation and liver preservation, there are still some controversies on this issue, especially its immunomodulatory effect. Rubber-like materials as many soft tissues can be described as incompressible and hyper-elastic materials. Their comparable elastic behavior, up to a certain extent, has been exploited to develop and test experimental methodologies to be then applied to soft biological tissues such as aortic wall. Hence, theoretical and experimental simulation of aortic tissue, and more generally blood vessel tissue, has been often conducted using rubbers. Despite all the efforts in characterizing such materials, a clear and comprehensive testing procedure is still missing. In particular, the influence of pre-conditioning in the mechanical response of hyper-elastic materials has been often neglected. In this paper, the importance of pre-conditioning is demonstrated by: (i) exploring the effect of stretching frequency applied before the uniaxial tensile test; (ii) recognizing the role of specimen geometry and strain amplitude; (iii) verifying the impact of experimental data acquisition on finite element predictions. It was found that stress-strain relationship shows a statistical difference between some frequencies of pre-conditioning and its absence. Only certain pre-conditioning frequencies were able to generate repeatable experimental data for strip or dumb-bell shapes. A novel immersed boundary (IB) method based on an implicit direct forcing (IDF) scheme is developed for incompressible viscous flows. The key idea for the present IDF method is to use a block LU decomposition technique in momentum equations with Taylor series expansion to construct the implicit IB forcing in a recurrence form, which imposes more accurate no-slip boundary conditions on the IB surface. To accelerate the IB forcing convergence during the iterative procedure, a pre-conditioner matrix is introduced in the recurrence formulation of the IB forcing. A Jacobi-type parameter is determined in the pre-conditioner matrix by minimizing the Frobenius norm of the matrix function representing the difference between the IB forcing solution matrix and the pre-conditioner matrix. In addition, the pre-conditioning parameter is restricted due to the numerical stability in the recurrence formulation. Consequently, the present pre-conditioned IDF (PIDF) enables accurate calculation of the IB forcing within a few iterations. We perform numerical simulations of two-dimensional flows around a circular cylinder and three-dimensional flows around a sphere for low and moderate Reynolds numbers. The result shows that PIDF yields a better imposition of no-slip boundary conditions on the IB surfaces for low Reynolds number with a fairly larger time step than IB methods with different direct forcing schemes due to the implicit treatment of the diffusion term for determining the IB forcing. Finally, we demonstrate the robustness of the present PIDF scheme by numerical simulations of flow around a circular array of cylinders, flows around a falling sphere, and two sedimenting spheres in gravity. Transient ischaemia leads to tolerance to subsequent protracted ischaemia. This 'ischaemia pre-conditioning' results from the induction of numerous protective genes, involved in cell metabolism, proliferation and survival, in antioxidant capacity, angiogenesis, vascular tone and erythropoiesis. Hypoxia-inducible factors ( HIF) play a pivotal role in this transcriptional adaptive response. HIF prolyl hydroxylases ( PHDs), serving as oxygen sensors, control HIF α degradation. HIF-mediated ischaemic pre-conditioning can be achieved with the administration of PHD inhibitors, with the attenuation of organ injury under various hypoxic and toxic insults. Clinical trials are currently under way, evaluating PHD inhibitors as inducers of erythropoietin. Once their safety is established, their potential use might be further tested in clinical trials in various forms of acute ischaemic and toxic organ damage. Repeated transient limb ischaemia was also found to attenuate ischaemic injury in remote organs. This 'remote ischaemic pre-conditioning' phenomenon ( RIP) has been extensively studied recently in small clinical trials, preceding, or in parallel with an abrupt insult, such as myocardial infarction, cardiac surgery or radiocontrast administration. Initial results are promising, suggesting organ protection. Large-scale multi-centre studies are currently under way, evaluating the protective potential of RIP in cardiac surgery, in the management of myocardial infarction and in organ transplantation. The mechanisms of organ protection provided by RIP are poorly understood, but HIF seemingly play a role as well. Thus, Inhibition of HIF degradation with PHD inhibitors, as well as RIP (in part through HIF), might develop into novel clinical interventions in organ protection in the near future. Generalist herbivores can face many challenges when choosing their host plant. This can be particularly difficult if their choice and performance are affected by host experience. Greenhouse whitefly, Trialeurodes vaporariorum Westwood ( Hemiptera: Aleyrodidae), is an invasive generalist herbivore, which has established in year-round greenhouses at northern latitudes where it cannot overwinter outdoors. It mainly uses crops such as cucumber ( Cucumis sativus L.), tomato ( Solanum lycopersicum L.), and ornamentals as host plants. However, every summer the insect escapes greenhouses and is exposed to natural vegetation. We evaluated the performance of T. vaporariorum on diverse vegetation outside greenhouses after prolonged experience of greenhouse crops. First, we surveyed the vegetation near infested greenhouses. Development success of the insect differed among wild hosts. We identified five new hosts among 12 plant species that bore pupae and were thus considered suitable as the insect's host plants. Members of the Urticaceae and Onagraceae were the most preferred and frequently inhabited by all insect life stages. The highest abundance of insects occurred in plots with low plant species richness, independent of plant family in these habitats. We then studied experimentally the impact of 1 year of preconditioning to one of three common greenhouse crops, cucumber, tomato, or poinsettia ( Euphorbia pulcherrima Willd. ex Klotzsch), on the performance of the preconditioned adults and their progeny on four wild plants. Adults from tomato and poinsettia preferred the novel host species over the species to which they were preconditioned. The whitefly population preconditioned to cucumber was the most fecund on all offered hosts. We conclude that generalist herbivores can have large variation in performance, despite polyphagy, on novel hosts as shown by the variable abundance of T. vaporariorum pupae among outdoor hosts. Furthermore, performance of whiteflies on natural vegetation was affected by experience on greenhouse crops. Based on our observations, we provide insights and recommendations for pest management.
