Mucuna pruriens, Ashwagandha, and Tribulus terrestris are known as the enhancers for sexual health, functional activities, vitality, and longevity. These herbs had been widely used in the Ayurveda medicine as aphrodisiacs through the ages, and their efficacy was also verified separately in our previous publication. Therefore, the aim of this study was to determine the effects of Mucuna, Ashwagandha, and Tribulus complexes on sexual function in rats. Twenty-eight male rats allocated to four groups as follows: (i) negative control (C); (ii) positive control or sildenafil citrate treated group (5 mg/kg) (S); (iii) MAT1 (combination of 10 mg Mucuna (M) + 10 mg Ashwagandha (A) + 10 mg Tribulus (T)/kg BW); (iv) MAT 2 (20 mg Mucuna + 20 mg Ashwagandha + 20 mg Tribulus/kg BW). There was no significant difference found between the MAT1 and MAT2 groups while they showed significantly increased testosterone, follicle-stimulating hormone (FSH), and luteinizing hormone (LH) levels when compared to the negative control. Significant increases in Nrf2/HO1 levels and decreases in NF-κB were detected in MAT groups similar to the decrease in serum and testis malondialdehyde (MDA) levels as compared to both controls. The sperm motility, count, and rate also significantly improved in both MAT groups, while ALT, AST, creatinine, ALP, and urea levels did not change in any of the groups. Oral consumption of MATs combination in male rats resulted in inhibition of NF-κB and MDA and also increased sex hormones with Nrf2-mediated HO-1 induction. MAT combinations may improve sexual functions by increasing levels of sexual hormones and regulation of NF-κB and Nrf2/HO-1 signaling pathways.

Graphical abstract, Highlights • Capsaicinoids (CAPs) are active compounds in Capsicum fruits. • CAPs have anti-inflammatory and antioxidant properties. • CAPs with regular exercise may enhance lipid metabolism. • CAPs down-regulate muscle SREBP-1c, LXRs, ACLY, FAS in exercised rats., The purpose of this study was to investigate the effects of capsaicinoids (CAPs) on lipid metabolism, inflammation, antioxidant status and the changes in gene products involved in these metabolic functions in exercised rats. A total of 28 male Wistar albino rats were randomly divided into four groups (n = 7) (i) No exercise and no CAPs, (ii) No exercise + CAPs (iii) Regular exercise, (iv) Regular exercise + CAPs. Rats were administered as 0.2 mg capsaicinoids from 10 mg/kg BW/day Capsimax® daily for 8 weeks. A significant decrease in lactate and malondialdehyde (MDA) levels and increase in activities of antioxidant enzymes were observed in the combination of regular exercise and CAPs group (P < 0.0001). Regular exercise + CAPs treated rats had greater nuclear factor-E2-related factor-2 (Nrf2) and heme oxygenase-1 (HO-1) levels in muscle than regular exercise and no exercise rats (P < 0.001). Nevertheless, regular exercise + CAPs treated had lower nuclear factor kappa B (NF-κB) and IL-10 levels in muscle than regular exercise and control rats (P < 0.001). Muscle sterol regulatory element-binding protein 1c (SREBP-1c), liver X receptors (LXR), ATP citrate lyase (ACLY) and fatty acid synthase (FAS) levels in the regular exercise + CAPs group were lower than all groups (P < 0.05). However, muscle PPAR-γ level was higher in the regular exercise and CAPs alone than the no exercise rats. These results suggest CAPs with regular exercise may enhance lipid metabolism by regulation of gene products involved in lipid and antioxidant metabolism including SREBP-1c, PPAR-γ, and Nrf2 pathways in rats.

Background and aim It has been reported that intestinal ischemia-reperfusion (I/R) injury results from oxidative stress caused by increased reactive oxygen species. Dexpanthenol (Dxp) is an alcohol analogue with epitelization, anti-inflammatory, antioxidant, and increasing peristalsis activities. In the present study, the aim was to investigate protective and therapeutic effects of Dxp against intestinal I/R injury. Materials and methods Overall, 40 rats were assigned into five groups including one control, one alone Dxp, and three I/R groups (40-min ischemia; followed by 2-h reperfusion). In two I/R groups, Dxp (500mg/kg, i.m.) was given before or during ischemia. The histopathological findings including apoptotic changes, and also tissue and serum biochemical parameters levels, were determined. Oxidative stress and ileum damage were assessed by biochemical and histological examination. In the control (n=8) and alone Dxp (n=8; 500mg/kg, i.m. of Dxp was given at least 30min before recording), groups were incised via laparotomy, and electrical activity was recorded from their intestines. In this experiment, the effect of Dxp on the motility of the intestine was examined by analyzing electrical activity. Results In ileum, oxidant levels were found to be higher, while antioxidant levels were found to be lower in I/R groups when compared with controls. Dxp approximated high levels of oxidants than those in the control group, while it increased antioxidant values compared with I/R groups. Histopathological changes caused by intestinal I/R injury and histological improvements were observed in both groups given Dxp. In the Dxp group, electrical signal activity markedly increased compared with the control group. Conclusions Here, it was seen that Dxp had protective and therapeutic effects on intestinal I/R injury and gastrointestinal system peristaltism.

Introduction In this study, we aimed to determine the protective effects of hesperidin, a citrus flavonoid, in a model of testicular ischemia/reperfusion injury in rats. Material and methods Forty-two pubertal male Wistar-Albino rats were divided into six groups: group 1 – control; group 2 – 50 mg/kg hesperidin (low dose hesperidin) used without torsion (LH group); group 3 – 100 mg/kg hesperidin without torsion (HH group); group 4 – torsion/detorsion group (T/D); group 5 – T/D + 50 mg/kg hesperidin treatment group (T/D + LH); and group 6 – T/D + 100 mg/kg hesperidin treatment group (T/D + HH). Hesperidin was given to the treatment groups 30 min before detorsion. After the fourth hour of reperfusion, orchiectomy was performed on the rats under anesthesia. The tissue samples were examined histologically and biochemically. Results In the T/D group testicular malondialdehyde (MDA) levels were increased significantly (p < 0.001) whereas superoxide dismutase (SOD), catalase (CAT) and glutathione (GSH) levels were decreased compared to the control and other groups. However, hesperidin caused the effect of T/D to become closer to normal biochemical values. In addition, the histological examinations showed that T/D caused damage in the testis but hesperidin reduced this effect. The effects of hesperidin were found to be dose dependent. Thus, applying high doses would generate greater therapeutic effects. Conclusions In a rat testicular T/D model we observed biochemical and histological damage due to ischemia. However, high and low dose applications of hesperidin were shown to have protective effects against this damage. Therefore, the aforementioned citrus flavonoid may provide positive results in cases of testicular torsion.

The aim of this study is to investigate effects of selenium and enlighten the possible mechanism of action in a rat transverse musculocutaneous flap model following ischemia-reperfusion injury.In this study, an experimental model, which mimicked free tissue transfer, was applied. Twenty-four male Wistar Albino rats were divided into a control group (N = 12), and a selenium treated group (N = 12). A superiorly based transverse rectus abdominis musculocutaneous (TRAM) flap was elevated and an ischemic insult for 4 hours was given. In selenium treated group (Group 2), sodium selenite (0.625 mg/kg) was injected intraperitoneally (i.p), 2 hours before the induction of ischemia. Six rats from each group were sacrificed at 24 hours after the operation and malonyldialdehyde (MDA), nitric oxide (NO), and glutathione (GSH) levels were measured biochemically, whereas the intensity of neutrophil infiltration was evaluated. For the rest of the rats in Group 2, sodium selenite was injected at the same dose everyday to the postoperative 10th day, in which the remaining 6 rats from each group were sacrificed. On postoperative 10th day, flap viability was assessed along with the evaluation of intensity of neovascularization.In Group 1, MDA levels were higher significantly (P0.05) when compared with Group 2. No statistical difference, however, was found for NO (P0.05), and GSH (P0.05) levels among Group 1 and 2. Neutrophil infiltration was more intense in Group 1, when compared with Group 2 whereas neovascularization was more abundant in samples of Group 2. Group 2 shows higher average flap surface areas when compared with Group 1 (P0.05).The results of this study demonstrated the preventive effect of selenium against ischemia-reperfusion injury by reducing tissue necrosis in muscle flaps possibly by decreasing MDA, increasing neovascularization, and decreasing neutrophil infiltration, thus suppressing inflammation.

Background and Aims Despite it being a highly potent antineoplastic drug, cisplatin has important toxic adverse effects limiting its use such as nephrotoxicity, neurotoxicity and ototoxicity. It is thought that cisplatin-induced hepatotoxicity is caused by oxidative stress resulting from increased reactive oxygen species (ROS). Apocynin (APO) exerts its antioxidant effect by reducing ROS production via inhibition of NADPH oxidase. The present study intended to demonstrate effects of cisplatin on hepatic pro-oxidant/antioxidant systems and to investigate protective effects of APO against cisplatin-induced hepatotoxicity. Methods Rats were randomly assigned into four groups (n = 8 each): a) control group; b) single dose of cisplatin (5 mg/kg); c) APO group (20 mg/kg on three consecutive days; i.p.); and d) APO plus cisplatin group. Liver tissue was assessed in all groups by biochemical and histopathological means. Also, serum alanine transaminase (ALT), aspartate transaminase (AST) and alkaline phosphatase levels were studied in all groups. Results When cisplatin group was compared to controls, it was seen that lipid peroxidation product, total oxidant status and ALT levels were markedly increased, whereas superoxide dismutase and glutathione peroxidase levels were overtly decreased. APO therapy markedly prevented cisplatin-induced harmful changes in liver. Our histopathological findings such as central vein dilatation, perivenuler and periportal sinusoidal dilatation, parenchymal inflammation, vacuolar changes in hepatocytes, biliary duct proliferation and caspase-3 positive hepatocytes were in accordance with the biochemical changes. Conclusion In light of these results, it is our thought that APO has a protective role against cisplatin-induced hepatotoxicity at both biochemical and histopathological levels.

Free iron leads to the formation of pro-oxidant reactive oxygen species (ROS). Humic acids (HAs) enhance permeability of cellular wall and act as a chelator through electron transferring. This study was designed to test chelator effect of HA on iron as well as its anti-oxidant effect against the iron-induced hepatotoxicity and cardiotoxicity. The rats used were randomly divided into four groups (n = 8/group): group I (the control group); group II (the HA group), humic acid (562 mg/kg) was given over 10 days by oral gavage; group III (the iron group), iron III hydroxide polymaltose (250 mg/kg) was given over 10 days by intraperitoneal route; and group IV (the HA plus iron group), received the iron (similar to group II) plus humic acid (similar to those in groups II and III) group. Blood and two tissue samples both from liver and heart were obtained for biochemical and histopathological evaluations. Iron deposition, the iron-induced hepatotoxicity, and cardiotoxicity were demonstrated by histopathological and biochemical manner. However, no significant differences were observed in the serum biochemical values and the histopathological results among the iron and the HA plus iron groups in the liver tissue but not in the heart tissue. The protective effects of humic acid against iron-induced cardiotoxicity were shown but not against hepatotoxicity in our study.

Renal cell carcinoma (RCC) is the most frequent upper urinary tract cancer in humans and accounts for 80–85% of malignant renal tumors. Eker rat represents a unique animal model to study RCC since these rats develop spontaneous renal tumors and leiomyoma, which may be due to tuberous sclerosis 2 (TSC2) mutation resulting in the activation of the mammalian target of rapamycin (mTOR) pathway. This study examines the role of a lycopene-rich diet in the development of RCC in the TSC2 mutant Eker rat model. Ten-week old female Eker rats (n = 90) were assigned in equal numbers to receive 0,100 or 200 mg/kg of lycopene as part of their daily diet. After 18 months the rats were sacrificed and the kidneys were removed. Immunohistochemical staining with antibodies against mTOR, phospho-S6 and EGFR were performed, as well as hematoxylin–eosin staining for histologic examination of the tumors. Tumors were counted and measured in individual kidneys. Presence of tumor decreased from 94% in control animals to 65% in the experimental group, but the difference was not statistically significant (P< 0.12). However, mean numbers of renal carcinomas were statistically significantly decreased in the lycopene-treated rats (P< 0.008) when compared to untreated controls. In the lycopene group, tumor numbers decreased (P< 0.002) and the numbers tended to decrease linearly (P< 0.003) as supplemental lycopene increased from 0 to 200. An experiment was conducted to evaluate the effects of a histidine complex of chromium (chromium histidinate, CrHis) on egg production, lipid peroxidation and the expression of hepatic nuclear factor kappa-light-chain-enhancer of activated B cells (NF-κB) and heat-shock proteins (HSPs) in Japanese quails (Coturnix coturnix japonica) exposed to heat stress (HS). A total of 180 5-week-old female quails were reared either at 22°C for 24 h/d (thermoneutral, TN) or 34°C for 8 h/d (heat stress, HS) for 12 weeks. Birds in both environments were randomly given one of three diets: basal diet and basal diet supplemented with 400 or 800 µg of elemental Cr as CrHis per kg of diet. Blood, egg yolk and liver samples collected at the end of the trial were analysed to determine concentrations of cholesterol and malondialdehyde (MDA) and expressions of transcription and heat-shock proteins. Exposure to HS caused reductions in feed intake (-8.1%) and egg production (-15.8%), elevations in serum (14.8%) and egg-yolk (29.0%) cholesterol concentrations, decreases in serum (113%) and egg-yolk (73.0%) MDA concentrations and increases in the expressions of hepatic NF-κB (52.3%) and HSPs (averaging 53.6%). The effects of increasing supplemental CrHis on the response variables were more notable in the HS environment than in the TN environment. There were considerable improvements in feed intake and egg production, decreases in serum and egg-yolk cholesterol concentrations and suppressions in the expressions of hepatic nuclear protein and HSPs in response to increasing supplemental CrHis concentration in the diet of quails reared under the HS environment. In conclusion, supplemental CrHis improves productivity through alleviating oxidative stress and modulating the expressions of hepatic NF-κB and HSPs in heat-stressed quails.

Lichen myxedematosus is a chronic, inflammatory, systemic dermatose characterized by dermal musin deposition and increased fibroblasts in the absence of thyroid dysfunction. It is usually seen together with paraproteinemia. It is clinically classified as scleromyxedema (papular mucinosis), localized lichen myxedematosus, and atypical lichen myxedematosus. Etiopathogenesis of the disease which is very difficult to treat is still unknown. Herein, we present a case of atypical scleromyxedema without monoclonal gammopathy mimicking angioedema, rapidly developing and responding to steroid treatment.

Background Dietary intake of lycopene has been associated with a reduced risk of ovarian cancer, suggesting its chemopreventive potential against ovarian carcinogenesis. Lycopene's molecular mechanisms of action in ovarian cancer have not been fully understood. Therefore, in the present study, we investigated the effects of lycopene on the ovarian cancer formation using the laying hen model, a biologically relevant animal model of spontaneous ovarian carcinogenesis due to high incidence rates similar to humans. Methods In this study, a total of 150 laying hens at age of 102 weeks were randomized into groups of 50: a control group (0 mg of lycopene per kg of diet) and two treatment groups (200 mg or 400 mg of lycopene per kg of diet, or ~26 and 52 mg/d/hen, respectively). At the end of 12 months, blood, ovarian tissues and tumors were collected. Results We observed that lycopene supplementation significantly reduced the overall ovarian tumor incidence (P < 0.01) as well as the number and the size of the tumors (P < 0.004 and P < 0.005, respectively). Lycopene also significantly decreased the rate of adenocarcinoma, including serous and mucinous subtypes (P < 0.006). Moreover, we also found that the serum level of oxidative stress marker malondialdehyde was significantly lower in lycopene-fed hens compared to control birds (P < 0.001). Molecular analysis of the ovarian tumors revealed that lycopene reduced the expression of NF-κB while increasing the expression of nuclear factor erythroid 2 and its major target protein, heme oxygenase 1. In addition, lycopene supplementation decreased the expression of STAT3 by inducing the protein inhibitor of activated STAT3 expression in the ovarian tissues. Conclusions Taken together, our findings strongly support the potential of lycopene in the chemoprevention of ovarian cancer through antioxidant and anti-inflammatory mechanisms.

Background: Cinnamon polyphenol extract is a traditional spice commonly used in different areas of the world for the treatment of different disease conditions which are associated with inflammation and oxidative stress. Despite many preclinical studies showing the anti-oxidative and antiinflammatory effects of cinnamon, the underlying mechanisms in signaling pathways via which cinnamon protects the brain after brain trauma remained largely unknown. However, there is still no preclinical study delineating the possible molecular mechanism of neuroprotective effects cinnamon polyphenol extract in Traumatic Brain Injury (TBI). The primary aim of the current study was to test the hypothesis that cinnamon polyphenol extract administration would improve the histopathological outcomes and exert neuroprotective activity through its antioxidative and anti-inflammatory properties following TBI. Methods: To investigate the effects of cinnamon, we induced brain injury using a cold trauma model in male mice that were treated with cinnamon polyphenol extract (10 mg/kg) or vehicle via intraperitoneal administration just after TBI. Mice were divided into two groups: TBI+vehicle group and TBI+ cinnamon polyphenol extract group. Brain samples were collected 24 h later for analysis. Results: We have shown that cinnamon polyphenol extract effectively reduced infarct and edema formation which were associated with significant alterations in inflammatory and oxidative parameters, including nuclear factor-κB, interleukin 1-beta, interleukin 6, nuclear factor erythroid 2-related factor 2, glial fibrillary acidic protein, neural cell adhesion molecule, malondialdehyde, superoxide dismutase, catalase and glutathione peroxidase. Conclusion: Our results identify an important neuroprotective role of cinnamon polyphenol extract in TBI which is mediated by its capability to suppress the inflammation and oxidative injury. Further, specially designed experimental studies to understand the molecular cross-talk between signaling pathways would provide valuable evidence for the therapeutic role of cinnamon in TBI and other TBI related conditions.

Background/aim: A novel complex of a nutritional supplement (CDB) contains chromium picolinate (CrPic), phosphatidylserine (PS), docosahexaenoic acid (DHA), and boron (B). The present study aimed to investigate the effects of CDB on the metabolic profile and memory acquisition in rats fed a high-fat diet (HFD). Materials and methods: Male Wistar rats were divided into six groups and received either a regular diet or HFD supplemented with or without different levels of CDB (0, 11, or 22 mg/kg BW). Results: Rats fed the HFD had greater glucose, insulin, lipid profile, and serum malondialdehyde concentrations, but lower serotonin and tryptophan in the serum and brain and lower Cr concentrations in serum, kidney, brain, and liver (P < 0.0001). CDB complex supplementation reversed all the effects, and the reversal effect was more pronounced with HFD for some parameters. Latency was less (P < 0.05) but probe was greater (P < 0.0001) for rats fed a regular diet. Increasing CDB complex levels in the diets resulted in a linear decrease in latency (P < 0.0002) but a linear increase in probe (P < 0.0002). Conclusion: Findings of the present work indicate that the CDB complex could be considered as an alternative treatment for preventing certain metabolic diseases and improving neurological functions, such as learning and memory.

In recent years, two meta‐analyses of chromium (Cr) supplementation have shown beneficial effects on glucose metabolism. Chromium histidinate (CrHis) reduces serum glucose levels in rats fed a high‐fat diet but no study has been conducted on cats until now. The aim of this study was to examine the effects of CrHis on glucose and lipid metabolism in cats. To challenge the glucose metabolism, 16 cats were fed a high‐carbohydrate high‐fat diet for three months. One group (n = 8) received 800 ug CrHis per day for two months, while the other group (n = 8) served as control group. An oral glucose tolerance test was conducted, blood samples were taken, and biochemical parameters and oxidative stress were measured. This experiment was conducted to compare the effect of the supplemental chromium (Cr) form on performance, egg quality, and metabolic profile in laying hens exposed to heat stress (HS). Laying hens (n = 1800; 16-wk-old; Lohmann LSL-Lite) were kept in cages in temperature-controlled rooms at either 22 ± 2°C for 24 h/d (thermoneutral, TN) or 34 ± 2°C for 8 h/d, from 08:00 to 17:00 h, followed by 22°C for 16 h (HS) for 12 wks. Hens reared under both environmental conditions were fed 1 of 3 diets: a basal diet and the basal diet supplemented with either 1.600 mg of chromium-picolinate (CrPic, 12.43% Cr) or 0.788 mg of chromium-histidinate (CrHis, 25.22% Cr) per kg of diet, delivering 200 μg elemental Cr per kg diet. Data were analyzed by 2-way ANOVA. Exposure to HS caused decreases in feed intake (P < 0.0001), egg production (P < 0.0001), egg weight (P < 0.0001), eggshell weight (P < 0.0009), eggshell thickness (P < 0.0001), eggshell strength (P < 0.0001), and Haugh unit (P < 0.0001), deterioration in feed conversion ratio (P < 0.0001), increases in serum glucose and cholesterol concentrations (P < 0.0001 for both), decreases in serum and egg yolk Cr concentrations (P < 0.0001 for both), and decreases in serum Na (P < 0.002) and K (P < 0.01) concentrations. Both Cr sources were equally effective in alleviating performance variables under the HS condition. However, neither Cr sources alleviated deteriorations in egg quality parameters and serum electrolytes. Both Cr sources decreased serum glucose and cholesterol concentrations and increased serum and egg yolk Cr concentrations under the HS condition. In conclusion, HS adversely affected laying performance, egg quality, and metabolic profile. Both CrPic and CrHis partially alleviated the adverse effect of HS on these parameters. Inclusion of either Cr source could be a part of nutritional management strategies to overcome the adverse effects of HS performance and metabolic profile in laying hens.

The effects of supplementation of arginine-silicate-inositol complex (ASI; 49.5-8.2-25 g/kg, respectively) to laying hens were investigated with respect to eggshell quality, calcium (Ca) balance, and expression of duodenal proteins related to Ca metabolism (calbindin and tight junction proteins). A total of 360 laying hens, 25 weeks old, were divided into 3 groups consisting of 6 replicate of cages, 20 birds per cage. The groups were fed a basal diet and the basal diet supplemented with 500 or 1000 mg ASI complex per kilogram for 90 days. Data were analyzed by ANCOVA using data during the first week of the adaptation period as covariates. As the ASI complex supplementation level increased, there were increases in feed intake (P < 0.0001), egg production (P < 0.001), egg weight (P < 0.0001) and eggshell weight (P < 0.001) weight, and shell thickness (P < 0.001) and decreases in feed conversion ratio and cracked egg percentage (P < 0.0001 for both). Concentrations of serum osteocalcin (P < 0.0001), vitamin D (P < 0.0001), calcium (P < 0.001), phosphorus (P < 0.001), and alkaline phosphatase (P < 0.008) as well as amounts of calcium retention (P < 0.0001) and eggshell calcium deposition (P < 0.001), and Ca balance (P < 0.0001) increased, whereas amount of calcium excretion (P < 0.001) decreased linearly in a dose-dependent manner. The ASI complex supplementation increased expressions of calcium transporters (calbindin-D28k, N sodium-calcium exchanger, plasma membrane calcium ATPase, and vitamin D receptor) and tight junction proteins (zonula occludens-1 and occludin) in the duodenum in a linear fashion (P < 0.0001 for all). In conclusion, provision of dietary ASI complex to laying hens during the peak laying period improved eggshell quality through improving calcium utilization as reflected by upregulation of genes related to the calcium metabolism. Further studies are needed to elucidate the contribution of each of the ASI complex ingredients.

The aim of this experiment was to determine the effects of β-cryptoxanthin (BCX) on the cardiometabolic health risk factors and NF-κB and Nrf2 pathway in insulin resistance induced by high-fat diet (HFD) in rodents. Twenty-eight Sprague-Dawley rats were allocated into four groups: (1) Control, rats fed a standard diet for 12 weeks; (2) BCX, rats fed a standard diet and supplemented with BCX (2.5 mg/kg BW) for 12 weeks; (3) HFD, rats fed a HFD for 12 weeks, (4) HFD + BCX, rats fed a HFD and supplemented with BCX for 12 weeks. BCX reduced cardio-metabolic health markers and decreased inflammatory markers (P < 0.001). Rats fed a HFD had the lower total antioxidant capacity and antioxidant enzymes activities and higher MDA concentration than control rats (P < 0.001 for all). Comparing with the HFD group, BCX in combination with HFD inhibited liver NF-κB and TNF-α expression by 22% and 14% and enhanced liver Nrf2, HO-1, PPAR-α, and p-IRS-1 by 1.43, 1.41, 3.53, and 1.33 fold, respectively (P < 0.001). Furthermore, in adipose tissue, BCX up-regulated Nrf2, HO-1, PPAR-α, and p-IRS-1 expression, whereas, down-regulated NF-κB and TNF-α expression. In conclusion, BCX decreased visceral fat and cardiometabolic health risk factors through modulating expressions of nuclear transcription factors.

This experiment was conducted to investigate effects of the organic complex form of supplemental chromium (Cr) on performance, oxidative stress markers, and serum profile in broilers exposed to heat stress (HS). A total of 1,200 10-day-old boilers (Ross-308) was divided into one of the 6 treatments (2 environmental temperatures x 3 diets with different Cr forms). The birds were kept in temperature-controlled rooms at either 22 ± 2°C 24 h/d (thermoneutral, TN group) or 34 ± 2°C for 8 h/d, 08:00 to 17:00 h, followed by 22°C for 16 h (HS group) and fed either a basal diet (C) or the basal diet supplemented with Cr (200 μg/kg) through 1.600 mg of CrPic (12.43% Cr) and 0.788 mg of CrHis (25.22% Cr). Feed intake and body weight were recorded weekly. After cervical dislocation, liver samples were harvested to analyze Cr concentration and glucose transporter-2,4 (GLUT-2,4) expression. The breast meat also was sampled for the concentration of Cr and expressions of nuclear factor erythroid 2-related factor 2 (Nrf2) and nuclear factor kappa B (NF-κB). Data were analyzed by 2-way ANOVA. Heat stress caused depressions in feed intake (12.1%) and weight gain (21.1%) as well as elevations in feed conversion (11.2%) and abdominal fat (32.8%). It was also associated with depletion of Cr reserves and increases in serum concentrations of glucose, cholesterol, creatine, and enzymes. Exposure to HS was accompanied by suppression of the expressions of Nrf2 and GLUT-2 in muscle and GLUT-4 in the liver and amplification of the expression of NF-κB in muscle. Both Cr sources partially alleviated detrimental effects of HS on performance and metabolic profile. The efficacy of Cr as CrHis was more notable than Cr as CrPic, which could be attributed to higher bioavailability. In conclusion, CrHis can be added into the diet of broilers during hot seasons to overcome deteriorations in performance and wellbeing related to oxidative stress.

Objectives: CNM (CNM) polyphenol has antioxidant and anti-inflammatory properties. Therefore, we hypothesized CNM decrease heart disease risk factors and may enhance anti- inflammatory properties in rats fed a diet containing CNM. In this study, we evaluated the effects of CNM polyphenol on insulin resistance (IR), hyperlipidemia, hepatic transcription factors expressions [sterol regulatory element-binding protein1c (SREBP-1c), liver X receptor-α (LXR-α), nuclear factor kappa B p65 (NF-κB p65), nuclear factor-E2-related factor-2 (Nrf2)] in rats fed high fat diet (HFD). Method: Twenty-eight Wistar rats were allocated into four groups; (i) normal control; animals fed with normal chow (C ) (ii) CNM (C+CNM 100 mg/kg b.wt.), (iii) HFD (42% of calories as fat, high fat diet [HFD]), and (iv) HFD + CNM for 12 weeks. Blood analysis for triglycerides (TG) and cholesterol (CHOL), glucose, insulin, malonaldehyde (MDA a marker of oxidative stress [OS]) were estimated. Body weight, visceral fat and liver weight recorded and liver MDA assessed. SREBP-1c, LXR-α, ATP-citrate lyase (ACLY), fatty acid synthase (FAS), NF-κB p65 expressions and decreased the PPAR-α, p-IRS-1, Nrf2, HO-1 proteins were evaluated by Western blotting. Results: HFD rats of the liver had increased SREBP-1c, LXR-α, ATP-citrate lyase (ACLY), fatty acid synthase (FAS), NF-κB p65 expressions and decreased the PPAR-α, p-IRS-1, Nrf2, HO-1 expressions compared to control group. CNM supplementation decreased body weight (8.4%), visceral fat (36.6%), liver weight (17.7%), serum glucose and insulin concentrations, lipid profile ( P < 0.05), and serum and liver MDA (23.3% and 25.4 %) concentration compared to HFD rats ( P < 0.05). CNM decreased hepatic SREBP-1c (18.1 %), LXRα (27.9%), ATP-citrate lyase (ACLY, 22.7 %), fatty acid synthase (FAS, 15.8 %), NF-κB p65 (23.3%) and enhanced the PPAR-α, IRS-1 (72.7 %), Nrf2 (111.7 %) and HO-1 (72.1 %) proteins in HFD rat livers ( P < 0.05). Discussion: These results suggest CNM supplementation reduces hyperlipidemia, inflammation, and oxidative stress through activating transcription factors (SREBP-1c, LXR-α, NF-κB, and Nrf2) and anti-oxidative defense signaling pathway.

Fascioliasis is a rarely encountered parasitic infection in Turkey that mainly affects the liver and bile ducts. Other defined localizations of the parasite are the lungs, gastrointestinal system, and subcutaneous fatty tissue. Two cases of female patients who presented to the hospital with abdominal pain and whose physical examination and laboratory findings were normal except peripheral eosinophilia, were detected to have liver masses with necrotic areas. Segmental hepatectomies were performed in both cases with the preliminary diagnosis of liver tumors. Upon microscopic examinations of the resection materials, necrotic granulomatous inflammation with eosinophilic reaction at the periphery and the parasite (Fasciola hepatica) were seen. Both cases were reported to be fascioliasis according to these findings. Two cases of fascioliasis mimicking malignancy in the liver are presented here together with literature findings.

The purpose of this study was to describe the effect of lycopene on antioxidant status, heat shock protein 70 (Hsp70), nuclear factor (erythroid-derived 2)-like 2 (Nrf2) and heme-oxygenase 1(HO-1) levels in the muscle of rainbow trout ( Oncorhynchus mykiss ). The fish were kept at low (20 kg m 3 − 1 ) or high (100 kg m 3 − 1 ) stocking densities till the end of study and fed a basal diet or the basal diet supplemented with either 200 or 400 mg of lycopene kg − 1 of diet. The protein, lipid and ash contents in the muscle were not changed by the supplementation of lycopene in rainbow trout kept under high or low stocking densities. In response to increasing supplemental lycopene level, there were linear increases in lycopene levels, superoxide dismutase (SOD), catalase (CAT) and glutathione peroxidase (GSH-Px) activities ( P P 0.001). In conclusion, lycopene supplementation to fish reduces the detrimental effects of oxidative stress and lipid peroxidation resulting from stocking densities via activating host defence system at cellular level.

OBJECTIVES: Lutein/zeaxanthin isomers (L/Zi), the major carotenoids, have demonstrated potent antioxidant and antiinflammation. This study was conducted to clarify the effects of L/Zi on brain transcription and brain-derived neurotrophic factors and synaptic proteins combined with exercise training in rats. METHODS: Wistar rats (age: 8 wks) were allocated into four groups: (i) Control: no treatments (ii) L/Zi: Rats treated with L/Zi (100 mg L/Zi/kg BW); (iii) Exercise: Rats with regular exercise only, (iv) Exercise + L/Zi: Rats with combined treatment of L/Zi (100 mg L/Zi/kg BW) and regular exercise. The exercise practice was carried out on a motor-driven rodent treadmill at 25 m/min, 45 min/day, 5 d/week for 8 wks. RESULTS: Rats with combined treatment of L/Zi (100 mg L/Zi/kg BW) and regular exercise. Brain nuclear factor (erythroid-derived 2)-like 2 (Nrf2), and heme oxygenase-1(HO-1) levels increased and nuclear factor (NF-κB) reduced in the combined group. In addition, L/Zi supplementation increased cerebral cortex brain-derived neurotrophic factor (BDNF), synapsin I, synaptophysin (SYP) and growth-associated protein-43 (GAP-43) levels both control and exercised rats (P < 0.001 for all). The highest cerebral cortex BDNF, synapsin I, SYP and GAP-43 levels were detected in the combined group. CONCLUSIONS: These results suggest that regular exercise training with L/Zi may improve brain function by regulating transcription and brain-derived neurotrophic factors and synaptic proteins in rats. Proliferating trichilemmal cyst is slowly growing benign cutaneous adnexal tumor of the hair follicle. Proliferating trichilemmal cysts are seen usually on the scalp and in elderly women. We present pathological findings of 43 years-old female patient with scalp nodular lesion with the literature.

Lycopene, the major carotenoid found in tomato, exhibits health beneficial effects by virtue of its antioxidant activity. As stocking density is known as a stress factor in fish, the effects of dietary lycopene supplementation (0, 200 or 400 mg kg − 1 ) on growth performance and antioxidant status in rainbow trout under normal (20 kg m 3 − 1 ) and high (100 kg m 3 − 1 ) stocking density were studied. Glutathione peroxidase (GSH-Px), superoxide dismutase (SOD) and catalase (CAT) activities were evaluated, as well as lipid peroxidation measured as malondialdehyde (MDA). Nuclear factor kappa-light-chain-enhancer of activated B cells (NF-κB), erythroid-derived 2-like 2 (Nrf2) and heme-oxygenase 1 (HO-1) levels were also assayed as an indicator of detoxifying/antioxidant defense mechanisms to cope with oxidative stress. Results showed that exposure to high stocking density depressed feed intake by 6.8% and body weight gain by 12.1%, increased plasma and hepatic MDA level by 88.5 and 124.3% and decreased hepatic SOD, CAT and GSH-Px activities by 22.1, 43.4 and 51.7%, respectively. In fish under crowded conditions, an expression of hepatic NF-κB was increased by 71.5%, being suppressed in Nrf2 and HO-1 by 40.6 and 42.7%, respectively. Lycopene supplementation increased linearly fish growth performance and antioxidant enzyme activity, and decreased linearly MDA concentration in both low and high stocking densities. NF-κB levels decreased hepatic Nrf2 and HO-1 levels increased linearly with lycopene supplementation at a greater extent in fish under low stocking condition. In conclusion, dietary supplementation of lycopene to fish reduces the detrimental effects of high stocking density on growth performance and modulates oxidative status via activating host defense system at cellular level. It appears that lycopene can be added up to 400 mg kg − 1 to rainbow trout diets to improve meat quality.

Aims This experiment investigated if chromium (Cr) as Cr-histidinate (CrHis) and Cr29 picolinate (CrPic) have a protective role in rats with hypoglycemia-induced brain injury, assessed by neuronal plasticity and regeneration potential. Main methods Male Sprague–Dawley rats were prospectively divided into 2 groups: control and hypoglycemic (induced by insulin administration, 15 U/kg, i.p., n = 56). Hypoglycemic rats were then received randomly 1) none, 2) dextrose (on the day of sampling), 3) CrHis, or 4) CrPic. Cr-chelates were delivered via drinking water (providing 8 μg elemental Cr per day) for one week prior to the hypoglycemia induction. The expressions of neuroplasticity markers [neural cell adhesion molecule (NCAM), growth-associated protein-43 (GAP-43), glial fibrillary acidic protein (GFAP)], glucose transporters (GLUT), and nuclear transcription proteins [nuclear factor-kappa (NF-κB), nuclear factor (erythroid-derived 2)-like 2 (Nrf2), and 4-hydroxyl nonenal (HNE)] were determined using Western blot. Key findings Hypoglycemia caused increases in the expressions of GLUT-1, GLUT-3, GFAP, NF-κB and HNE and decreases in the expression of NCAM's, GAP-43 and Nrf2 in the hippocampus, cerebellum, and cortex. Cr-chelates suppressed expressions of GLUTs, GFAP, NF-κB and HNE expressions and enhanced expressions of NCAM, GAP-43 and Nrf2, which were more notable for CrHis than for CrPic. Significance In conclusion, hypoglycemia leads to cerebral injury and Cr-chelates, particularly CrHis have protective and regeneration potential in cerebral tissues through modulating neuroplasticity markers and nuclear transcription proteins as well as facilitating glucose transporters.

The objective of this study was to analyze the clinical benefit of histopathologic analysis of appendectomy specimens from patients with an initial diagnosis of acute appendicitis. We retrospectively analyzed the demographic and histopathologic data of 1255 patients (712 males, 543 females; age range, 17–85 years) who underwent appendectomy to treat an initial diagnosis of acute appendicitis. Patients who underwent incidental appendectomy during other surgeries were excluded from the study. Histopathologic findings of the appendectomy specimens were used to confirm the initial diagnosis. Ninety-four percent of the appendectomy specimens were positive for appendicitis. Of those, 880 were phlegmonous appendicitis, 148 were gangrenous appendicitis with perforation, and the remaining 88 showed unusual histopathologic findings. In the 88 specimens with unusual pathology, fibrous obliteration was observed in 57 specimens, carcinoid tumor in 11, Encheliophis vermicularis parasite infection in 8, granulatomous inflammation in 6, appendiceal endometriosis in 2, and 1 specimen each showed mucocele, eosinophilic infiltration, Taenia saginata parasite infection, and appendicular diverticulitis. All carcinoid tumors were located in the distal appendix. Six of the 11 carcinoid tumors were defined by histopathology as involving tubular cells, and the other 5 as involving enterochromaffin cells. Six patients had muscularis propria invasion, 2 patients had submucosa invasion, 2 patients had mesoappendix invasion, and 1 patient had serosal invasion. All patients with tumors remained disease free during the follow-up (range, 1–27 months). We conclude that when the ratio of unusual pathologic findings for appendectomy specimens is considered, it is evident that all surgical specimens should be subjected to careful histologic examination.

1. The herb Creeping Thistle, Cirsium arvense (C. arvense), has been used in folk medicine due to its antioxidant and anti-inflammatory effects. 2. The aim of this study was to investigate the effects of dietary C. arvense extract supplementation on performance, egg quality, nutrient digestibility and antioxidant status in quail. 3. Quails (n = 150) were allocated randomly to one of the three dietary treatments: basal diet and basal diet enriched with 100 and 200 mg C. arvense extract per kg diet. 4. Dietary enrichment with C. arvense extract altered neither performance and egg quality parameters nor nutrient digestibility. Although there were no changes in concentrations of vitamin A and E in serum, liver and egg yolk, supplemental C. arvense extract decreased MDA concentrations in serum, liver and egg yolk by 39.3, 40.5 and 51.5%, respectively, in a dose-response manner. As supplemental C. arvense extract increased to 200 mg/kg, the activity of hepatic SOD, CAT and GSH-Px increased by 14.5, 17.4 and 35.5%, respectively. 5. Addition of C. arvense extract up to 200 mg to per kg diet enhanced antioxidant status in laying quail and their eggs, without affecting performance and other egg quality parameters. 6. Future studies are needed to elucidate the mechanism behind the antioxidant effects of C. arvense extract.

We evaluated the effects of cinnamon polyphenol extract on hepatic transcription factors expressions including SREBP-1c and LXR-α in rats fed high fat diet (HFD). Twenty-eight Wistar rats were allocated into four groups: (i) normal control: animals fed with normal chow; (ii) cinnamon: animals supplemented with cinnamon polyphenol; (iii) HFD: animals fed a high-fat diet; and (iv) HFD + cinnamon: animals fed a high-fat diet and treated with cinnamon polyphenol. Obesity was linked to hyperglycemia, hyperlipidemia, and oxidative stress as imitated by elevated serum glucose, lipid profile, and serum and liver malondialdehyde (MDA) concentrations. Cinnamon polyphenol decreased body weight, visceral fat, liver weight and serum glucose and insulin concentrations, liver antioxidant enzymes, and lipid profile (P<0.05) and reduced serum and liver MDA concentration compared to HFD rats (P<0.05). Cinnamon polyphenol also suppressed the hepatic SREBP-1c, LXR-α, ACLY, FAS, and NF-κB p65 expressions and enhanced the PPAR-α, IRS-1, Nrf2, and HO-1 expressions in the HFD rat livers (P<0.05). In conclusion, cinnamon polyphenol reduces the hyperlipidemia, inflammation, and oxidative stress through activating transcription factors and antioxidative defense signaling pathway in HFD rat liver.

We investigated the effect of daily dietary curcumin intake on the development and progression of spontaneous ovarian cancer in a galline (hen) model, as the chicken is the only nonhuman animal in which ovarian cancer spontaneously develops with a high prevalence. At the end of 12 months, ovarian cancer had spontaneously developed in 39% (35/90) of control hens not fed curcumin (n = 90). In comparison, it spontaneously developed in 27% (24/90) and 17% (15/90) of hens given curcumin at 25.8 (n = 90) and 53.0 mg/day (n = 90), respectively (P = 0.004). This represented significant dose-dependent reductions in overall ovarian cancer incidence in the 25.8 and 53.0 mg/day curcumin-fed groups (31% and 57%, respectively). Daily curcumin intake also reduced ovarian tumor sizes (P = 0.04) and number of tumors (P = 0.006). Evaluation of the molecular mechanisms underlying the chemopreventive and antitumor effects of curcumin revealed that NF-κB and STAT3 signaling pathways were significantly inhibited but that the nuclear factor erythroid 2/heme oxygenase 1 antioxidant pathway was induced by curcumin intake in a dose-dependent manner in ovarian tissues (P < 0.05). Sequencing of the Ras family genes (KRAS, NRAS, and HRAS) revealed less frequent KRAS and HRAS mutations in ovarian tumors in the curcumin-fed animals. In conclusion, our results demonstrated for the first time that daily curcumin intake leads to a significant and dose-dependent reduction in spontaneous ovarian cancer incidence and tumor growth, indicating a tremendous role for curcumin as a chemopreventive strategy for ovarian cancer. To examine the molecular mechanism of capsaicinoid supplementation from capsicum extract, laying Japanese quail (n = 180, 5 weeks old) were reared either at 22°C for 24 h/d (thermoneutral, TN) or at 34°C for 8 h/d (heat stress, HS) and fed on one of three diets containing 0, 25 or 50 mg of capsaicinoids per kilogram for 12 weeks (2 × 3 factorial arrangement). 2. The results revealed that exposure to HS decreased feed consumption by 10.7% and egg production by 13.6%, increased serum and ovary malondialdehyde (MDA) levels by 66.9% and 88.1%, respectively, and reduced ovary superoxide dismutase (SOD), catalase (CAT) and glutathione peroxidase (GSH-Px) activities by 28.3%, 48.7% and 43.8%, respectively. 3. There were magnifications in the ovary nuclear factor kappa-light-chain-enhancer of activated B cell (NF-κB) levels by 42.4% and suppressions in nuclear factor (erythroid-derived 2)-like 2 (Nrf2), protein kinase B (Akt) and haem-oxygenase 1 (HO-1) levels by 29.2%, 38.2% and 30.7%, respectively, in heat-stressed quail. 4. With increasing supplemental capsaicinoids, there were linear increases in egg production, antioxidant enzyme activity, linear decreases in ovary MDA and NF-κB levels and linear increases in ovary Nrf2, Akt and HO-1 levels at a greater extent in quail reared under TN condition than those reared under HS condition. Two-way treatment interactions showed that the degree of restorations in all response variables was more notable under the HS environment than under the TN environment as supplemental capsaicinoid level was increased. 5. In conclusion, capsaicinoid supplementation alleviates oxidative stress through regulating the ovary nuclear transcription factors in heat-stressed quail.

Background Mucuna pruriens, Tribulus terrestris and Ashwagandha (Withania somnifera) are widely known as antioxidant effective herbals and have been reported to possess aphrodisiac activities in traditional usages. In this study, we determined the effects of these herbals on sexual functions, serum biochemical parameters, oxidative stress and levels of NF-κB, Nrf2, and HO-1 in reproductive tissues. Methods Thirty-five male rats were divided into five groups: the control group, sildenafil-treated group (5 mg/kg/d), Mucuna, Tribulus and Ashwagandha groups. The extract groups were treated orally either with Mucuna, Tribulus or Ashwagandha (300 mg/kg b.w.) for 8 weeks. Results All of the extracts were found to be significantly effective in sexual functioning and antioxidant capacity and Tribulus showed the highest effectiveness. Serum testosterone levels significantly increased in Tribulus and Ashwagandha groups in comparison to control group. Tribulus was able to reduce the levels of NF-κB and increase the levels of Nrf2 and HO-1 to a much greater extent than Mucuna and Ashwagandha. Conclusions These results demonstrate for the first time that Mucuna, Tribulus and Ashwagandha supplementation improves sexual function in male rats via activating Nrf2/ HO-1 pathway while inhibiting the NF-κB levels. Moreover, Tribulus terrestris extract was found to be more bioavailable from Ashwagandha extract followed by Mucuna extract. Mesozeaxanthin (MZ) is able to protect against chronic and cumulative eye damage and neutralize free radicals produced by oxidative stress. The objective of the present study was to evaluate the protective potential of MZ against retinal oxidative damage and growth and transcription factors of the retina in rats fed with high-fat diet (HFD).Twenty-eight Sprague Dawley rats were randomly divided into the following 4 groups: (1) Control, (2) MZ (100 mg/kg bw/d), (3) HFD (42% of calories as fat), and (4) HFD+MZ (100 mg/kg bw/d) group rats were administered daily as supplement for 12 weeks.Consumption of HFD was associated with hyperglycemia and oxidative stress as reflected by increased serum MDA concentration (P 0.001). No measurable zeaxanthin (Z)+MZ and lutein (L) could be detected in the serum of control and HFD rats, whereas they were observed in the serum of MZ-administered rats. Retinal antioxidant enzyme [superoxide dismutase (SOD) and catalase (CAT)] activities were significantly decreased in the HFD group compared to the normal group (P 0.01). However, retinal antioxidant enzymes were restored close to normal levels in HFD+MZ-treated rats (P 0.05). The retina of rats fed with HFD had increased levels of vascular endothelial growth factor (VEGF), inducible nitric oxide (iNOS), intercellular adhesion molecule-1 (ICAM-1), and nuclear factor-kappa B (NF-κB) levels and decreased nuclear factor erythroid 2-related factor 2 (Nrf2) and heme-oxygenase 1(HO-1) levels compared to the healthy rat retina (P 0.001). Rats treated with MZ partially alleviated the inflammation as reflected by suppressed VEGF, iNOS, ICAM, and NF-κB levels and increased Nrf2 and HO-1 levels in the retina of rats fed (P 0.05).Results from the present study suggest that MZ has protective effects on the retina and the ability to modulate oxidative stress of retina in rats fed an HFD by suppressing retinal lipid peroxidation and regulating growth and transcription factors.

Arginine silicate inositol (ASI) complex is a composition of arginine, silicon, and inositol that has been shown to have beneficial effects on vascular health. This study reports the effects of an ASI ointment on wound healing in rats. A full-thickness excision wound was created by using a disposable 5 mm diameter skin punch biopsy tool. In this placebo-controlled study, the treatment group’s wound areas were covered by 4 or 10 % ASI ointments twice a day for 5, 10, or 15 days. The rats were sacrificed either 5, 10, or 15 days after the wounds were created, and biopsy samples were taken for biochemical and histopathological analysis. Pleural Malignant Mesothelioma (MM) is a fatal disease that has been associated with asbestos exposure. Differential diagnosis between the pleural infiltration of pulmonary carcinomas and MM is rather difficult particularly for epitheloid type mesothelioma. We aimed to investigate the utility of CD90, a cancer stem cell marker, in the differential diagnosis of MM and lung carcinoma, its prognostic significance and compare its value with that of Calretinin. Ninety pathology specimens including MM (n:30), pulmonary adenocarcinoma (n:30) and pulmonary squamous cell carcinoma (n:30) were used in this study. Immunohistochemical comparision of CD 90 and Calretinin was made in all groups. Calretinin was positive in 20 cases with MM (64.5 %), and was negative in 10 (32.3 %). CD 90 was positive in 25 of these cases (80 %) and negative in 5 (16 %). On the other hand pulmonary adenocarcinomas and squamous cell carcinomas showed positivity with CD90, 63,6 % and 73 %, respectively. We think that CD 90 has no place in the differential diagnosis between mesothelioma and pulmonary carcinoma because of the low specificity in spite of the high sensitivity.

Objective: Capsaicinoids (CAPs) are the major pungent, naturally occurring active compounds in capsicum fruits such as hot chili peppers. We recently demonstrated that CAPs inhibits pancreatic lipase and down regulates C/EBPα and PPARγ. Therefore, we hypothesized that CAPS may enhance exercise by effective utilization of lipids in rats. In this study, we observed changes in protein expressions levels of NFkB, IL-10, Nrf2, pAMPK and HO-1 in rats fed a diet containing CAPs. Methods: Eight week old male Wistar rats were treated in four treatment groups (i) control [no Ex, Group I (CTL)] (ii) CTL + CAPs [no Ex, 0.2 mg capsaicinoids from 10 mg/kg BW/day Capsimax ® , Group II] (iii) CTL + Ex [Group III] and (iv) CTL + Ex+ CAPs [0.2 mg capsaicinoids from 10 mg/kg BW/day Capsimax ® ] for 8 wks. The Ex protocols were performed on a motor-driven rodent treadmill (TMR). All animal procedures were approved by the Animal Experimentation Ethics Committee of Inonu University (Malatya, Turkey; 2014/A-55). Blood analysis for triglycerides (TG) and cholesterol (CHOL), muscle analysis for lactate, superoxide dismutase (SOD) and glutathione peroxidase (GPx) and malonaldehyde (MDA a marker of oxidative stress [OS]) were estimated. Phosphorylated AMP-activated protein kinase (pAMPK), Nuclear factor (erythroid-derived 2)-like 2 (Nrf2), heme oxygenase-1 ( HO1), nuclear factor (NF-κB) and Interleukin-10 (IL-10) proteins in the gastrocnemius muscle was evaluated by Western blotting. Results: As shown in Table, Group IV significantly increased running performance and exhaustion time and a significant decrease in TG and CHOL was observed compared with other treatments. A significant decrease in lactate and MDA and increase in SOD and GPx were observed in Group IV. An increase in protein levels of pAMPK, Nrf2, and HO1 and a decrease in NF-κB and IL-10 levels were observed in Group IV. Discussion: These results suggest CAPs with Ex may enhance the effect of exercise by effective utilization of lipids by increasing pAMPK and Nrf2.

Curcumin, a natural polyphenol in the spice turmeric, exhibits antioxidant and antiinflammatory properties. This study was conducted to elucidate the action mode of curcumin alleviation of oxidative stress in heat-stressed quail. A total of 180 birds (10 d old) were assigned randomly to be reared at either 22°C (Thermoneutral) or 34°C (Heat stress) for 8 h/d (0900-1700) until the age of 42 d. Birds in both environments were randomly fed 1 of 3 diets: basal diet and basal diet added with 0, 200 or 400 mg of curcumin per kg of diet. Each of the 2×3 factorially arranged experimental groups was replicated in 10 cages, each containing three birds. In response to increasing supplemental curcumin level, there were linear increases in cumulative feed intake, final body weight, and weight gain, and nuclear factor erythroid 2-related factor two level and heme oxygenase one level; linear decreases in feed efficiency, serum, muscle and liver malondialdehyde level, respectively and inflammatory transcription factor, nuclear factor-κB and heat shock proteins 70 level (P0.0001 for all). The results indicated that curcumin alleviates oxidative stress through modulating the hepatic nuclear transcription factors and heat shock proteins 70 in heat-stressed quails.

The effects of tomato powder supplementation on performance, egg quality, serum and egg yolk carotenoids, vitamins and malondialdehyde (MDA) concentrations in were investigated in laying hens in mid-lay. 2. A total of 90 laying hens, 49 weeks old, were divided into 3 groups consisting of 6 replicate cages, 5 birds per cage. Birds were randomly fed on one of three diets: basal diet and basal diet added with 5 or 10 g tomato powder per kg diet. 3. As tomato powder concentration increased, there were linear increases in feed intake, egg production, egg weight and yolk colour and a linear decrease in feed conversion. Shell weight, shell thickness and Haugh unit remained unchanged in response to dietary treatments. 4. Concentrations of serum and egg yolk lycopene, β-carotene, lutein and vitamin A increased for both diets including tomato powder, whereas MDA decreased linearly with increasing supplemental tomato powder concentration. 5. Tomato powder supplementation increased egg production persistency and increased carotenoids and vitamin A contents in egg yolk, accompanied by reduced yolk lipid peroxidation.

Rearing birds at temperatures higher than thermoneutral temperature might impair cellular signal transduction pathways, influencing the expression of genes coding for antioxidant proteins, and affect the laying performance of the bird. Cisplatin-induced nephrotoxicity is related to an increase in oxidative stress in the kidney. Lycopene, a carotenoid found in tomatoes, is a potent dietary antioxidant. In the present study, we investigated the effect of the tomato lycopene complex against cisplatin-induced lipid peroxidation and nephrotoxicity in rats. Male Wistar rats (n = 28, 8 wk old, between 200-215 g) were divided into 4 groups: (a) control, (b) tomato lycopene complex (6 mg/kg, daily; consisting of 6% lycopene, 1.5% tocopherols, 1% phytoene and phytofluene, and 0.2% β-carotene), (c) cisplatin (7 mg/kg i.p., single dose), and (d) cisplatin + tomato lycopene complex. Cisplatin administration increased serum urea-N (171 vs. 37 mg/dl) and creatinine (1.80 vs. 0.42 mg/dl) and decreased body weight in comparison with the control rats (P0.001). Serum creatinine and urea-N levels were lower in rats treated with tomato lycopene complex + cisplatin compared with rats treated with cisplatin alone (P0.001). The renal tissue from the cisplatin-treated rats had greater malondialdehyde (MDA; 172 vs. 93 nmol/g) and 8-isoprostane levels (1810 vs. 610 pg/g) than that from the control rats (P0.001). Tomato lycopene complex prevented the rise of MDA and 8-isoprostane (P0.001). No measurable lycopene could be detected in the serum of the control and cisplatin-treated rats, whereas lycopene was observed in the serum of rats supplemented with tomato lycopene complex. Renal Bax protein expression was significantly higher in the cisplatin-treated rats than in the control rats, and tomato lycopene complex treatment significantly reduced Bax expression (P0.001). The expression of Bcl-2 was higher in tomato lycopene complex/cisplatin-treated rats than in the cisplatin-injected rats (P0.05). The expression of renal HSP60 and HSP70 was significantly lower in tomato lycopene complex + cisplatin-treated rats than in rats treated with cisplatin alone (P0.001). These results suggest that tomato lycopene complex has protective effects against cisplatin-induced nephrotoxicity and lipid peroxidation in rats.

In the present study, the effects of dietary resveratrol on the induction of heat shock proteins, transcription factors and antioxidative enzyme system in liver of quails under heat stress were investigated. A total of 180 (55-day-old) female Japanese quails (Coturnix coturnix japonica) were reared either at 22 °C for 24 h/day (thermoneutral, TN) or 34 °C for 8 h/day (heat stress, HS; 09:00-17:00 hours) for 12 weeks. Birds in both environments were randomly fed one of three diets: basal diet and basal diet added with either 200 or 400 mg of resveratrol per kg of diet. The results showed that exposure to high ambient temperature induced decreases in feed intake, egg production, and hepatic superoxide dismutase (SOD), catalase (CAT), and glutathione peroxidase (GSH-Px) activities but increases in hepatic malondialdehyde (MDA) concentrations (p < 0.001). Liver Hsp70, Hsp90 and NF-κB expression was greater while Nrf2 expression was lower for quails reared under the heat stress than for those reared under the TN environment (p < 0.0001). There were linear increases in feed intake, egg production, hepatic SOD, CAT, and GSH-Px activities as well as Nrf2 expression, but linear decreases in hepatic MDA concentrations and Hsp70, Hsp90, and NF-κB expressions with increasing supplemental resveratrol level (p < 0.0001). Two-way treatment interactions revealed that the degree of restorations in all response variables was more notable under the high ambient temperature than that of the TN environment as dietary resveratrol concentration was increased. The results of the present study suggest that supplemental resveratrol reduces oxidative stress in heat-stressed quails through modulating the hepatic heat shock proteins and nuclear transcription factors.

Conditions in which glucose metabolism is impaired due to insulin resistance are associated with memory impairment. It was hypothesized that supplemental chromium (Cr) may alleviate insulin resistance in type 2 diabetes and consequently improve memory acquisition, depending upon its source and level. In a complete randomized design experiment, male Wistar rats (n=60; weighing 200-220 g) were fed either normal (8%, normal diet (ND)) or high-fat (40%, high-fat diet (HFD)) diet and supplemented with Cr as either chromium-glycinate (CrGly) or chromium-acetate (CrAc) at doses of 0, 40, or 80 μg/kg body weight (BW) via drinking water from 8 to 20 weeks of age. Feeding HFD induced type 2 diabetes, as reflected by greater glucose/insulin ratio (2.98 vs. 2.74) comparing to feeding ND. Moreover, HFD rats had greater BW (314 vs. 279 g) and less serum (53 vs. 68 μg/L) and brain (14 vs. 24 ng/g) Cr concentrations than ND rats. High-fat diet caused a 32% reduction in expressions of glucose transporters 1 and 3 (GLUTs) in brain tissue and a 27% reduction in mean percentage time spent in the target quadrant and a 38% increase in spatial memory acquisition phase (SMAP) compared with ND. Compared with supplemental Cr as CrAc, CrGly was more effective to ameliorate response variables (i.e., restoration of tissue Cr concentration, enhancement of cerebral GLUTs expressions, and reduction of the glucose/insulin ratio and SMAP) in a dose-response manner, especially in rats fed HFD. Supplemental Cr as CrGly may have therapeutic potential to enhance insulin action and alleviate memory acquisition in a dose-dependent manner, through restoring tissue Cr reserve and enhancing cerebral GLUTs expressions.

Antineoplastic agents induce the production of free radicals and other reactive oxygen species (ROS) in biological systems. Cytotoxic effects of antineoplastic drugs depend on rapid proliferation of cancer cells. Oxidative stress occurring during chemotherapy may potentially interfere with these effects through reducing the rate of cell proliferation. ROS may also contribute to side effects that occur with chemotherapeutic agents such as doxorubicin-induced cardiotoxicity, cisplatin-induced nephrotoxicity, and bleomycin-induced pulmonary fibrosis. Lycopene is a major carotenoid present in tomatoes, and it is a potent antioxidant that may provide protection against cellular damage caused by ROS. Lycopene may reduce or prevent the side effects of chemotherapy due to its antioxidant and anti-inflammatory properties. This review focuses on the protective effects of lycopene against antineoplastic agent induced toxicity.

Epidemiologic studies suggest that zinc deficiency may be associated with increased risk of cancer. We investigated the effects of zinc picolinate supplementation on the development of leiomyomas, malondialdehyde (MDA), 8-isoprostane, 4-hydroxyalkenal (HAE), and 8-hydroxy-2'-deoxyguanosine (8-OHdG) levels, and heat shock protein 70 (Hsp70) expression in Japanese quails. One hundred fifty quails (6 months old) were assigned to three treatment groups consisting of 50 birds in each group. Birds were fed either a basal diet or the basal diet supplemented with 30 mg or 60 mg of zinc/kg of diet. The animals were sacrificed after 350 days, and the tumors were identified. Zinc picolinate supplementation did not affect the number of leiomyomas compared to control birds (P > .05). However, the tumors in zinc-fed birds were smaller than those found in control birds (P = .01) Serum MDA, 8-isoprostane, and HAE levels were lower in the treatment groups than in the control group: MDA, 1.95 versus 0.93 micromol/L; 8-isoprostane, 108 versus 85 pg/mL; HAE, 1.55 versus 0.96 micromol/L (P = .01 for all three parameters). The concentrations of serum 8-OHdG, which is a marker of oxidative damage, in the groups were 28.5, 23.6, and 20.1 ng/mL, respectively (P = .01). Hsp70 expression was significantly decreased in zinc-treated birds (P < .01). The results indicate that dietary zinc picolinate supplementation reduces the growth of spontaneously occurring leiomyomas of the oviduct in the Japanese quail. Clinical trials should be conducted to investigate the efficacy of zinc supplementation in the prevention and treatment of uterine leiomyoma in humans.

In the present study, the effects of different sources of selenium (Se; sodium selenite or selenomethionine) supplementation on the growth and serum concentrations of oxidative stress markers [malondialdehyde (MDA), 8-isoprostane, glutathione peroxidase (GSH-Px) activity] and muscle Se, MDA and heat shock protein 70 (Hsp70) levels in rainbow trouts were evaluated. The fish (n = 360; 0 + years old) with initial average weight of 20 ± 0.8 g were randomly assigned to 12 treatment groups consisting of 3 replicates of 10 fish each in a 2 × 2 × 3 factorial arrangement of treatments (stocking densities, Se sources, Se levels). The fish were kept at low (25 kg m−3) or high (100 kg m−3) stocking densities and fed a basal (control) diet or the basal diet supplemented with either 0.15 or 0.30 mg of Se kg−1 of diet from two different forms: sodium selenite or selenomethionine. High stocking density decreased weight gain, feed intake and feed conversion ratio (FCR) when basal diet was fed (P = 0.001). A linear increase in feed intake and weight gain and improvement in FCR were found in sodium selenite (P = 0.01)- or selenomethionine (P = 0.001)-supplemented fish reared under crowding conditions.
