No. of bitstreams: 0 Previous issue date: 2011-07-01 Fundação de Amparo à Pesquisa do Estado de São Paulo (FAPESP) Objective: The variety of methodologies used to investigate ovariectomized female rats shows different results, which makes a clinical application of these results difficult. The purpose of this work was to evaluate the effect of masticatory effort reduction and of low-calcium diet on maxillary bone turnover of ovariectomized female rats.Study Design: Eighty-four female rats were divided into four groups of 21 animals each as follows: SHAM - sham-operated; OVZ - ovariectomized and fed a standard commercial diet; LCD - fed a low calcium diet, and GCD - fed a grind commercial diet. The inferior first molars were extracted bilaterally 15 days after the ovariectomy, and the animals were euthanized 3, 5 and 8 weeks after ovaries removal. The maxillae were embedded in methylmetacrilate. The results were submitted to analysis of variance.Results: The daily mineral apposition rate lowered with time and was not different between SHAM and OVZ groups. The trabecular bone volume of SHAM and OVZ animals was similar and decreased with time. The GCD animals presented the lowest means and the LCD the highest in comparison to the OVZ group.Conclusion: It was concluded that ovariectomy and a low calcium diet did not cause significant maxillary bone loss in the first molar region, and even in the absence of the antagonist tooth, they did not cause maxillary bone turnover. The grind commercial diet is a good alternative for the study of maxillary bone loss in ovariectomized female rats. No. of bitstreams: 0 Previous issue date: 2010-10-01 A porous material for bone ingrowth with adequate pore structure and appropriate mechanical properties has long been sought as the ideal bone-implant interface. This study aimed to assess in vivo the influence of three types of porous titanium implant on the new bone ingrowth. The implants were produced by means of a powder metallurgy technique with different porosities and pore sizes: Group 1 = 30% and 180 μm; Group 2 = 30% and 300 μm; and Group 3 = 40% and 180 μm. Six rabbits received one implant of each type in the right and left tibiae and were sacrificed 8 weeks after surgery for histological and histomor-phometric analyses. Histological analysis confirmed new bone in contact with the implant, formed in direction of pores. Histomorphometric evaluation demonstrated that the new bone formation was statistically significantly lower in the group G1 than in group G3, (P = 0.023). Based on these results, increased porosity and pore size were concluded to have a positive effect on the amount of bone ingrowth. A proposta deste trabalho foi estudar morfologicamente em ratas os efeitos da ovariectomia na atrofia e regeneraçãoda parótida. Foram utilizadas 56 ratas divididas em dois grupos: grupo controle A (ratas não ovariectomizadas)e grupo experimental B (ratas ovariectomizadas). Aos três meses de idade, as ratas do grupo B foramovariectomizadas e as do grupo A sofreram falsa-ovariectomia. A atrofia e regeneração da parótida foram estudadasem ambos os grupos, sendo a atrofia obtida através da ligadura do ducto excretor e a regeneração, atravésda desobstrução ductal. Os animais foram sacrificados um mês após falsa-ovariectomia ou ovariectomia e nosperíodos de sete dias de atrofia glandular e de sete e 14 dias de regeneração glandular. As parótidas removidascirurgicamente foram submetidas às colorações de HE e PAS. Após falsa-ovariectomia e ovariectomia, as ratasdos grupos A e B, respectivamente, apresentaram os mesmos aspectos morfológicos. Durante a atrofia glandular,houve redução do parênquima, assim como inflamação, fibrose e diminuição de grânulos de secreção nos gruposA e B. Durante a regeneração glandular, houve o restabelecimento do parênquima, redução da inflamação e dafibrose e aumento dos grânulos de secreção em ambos os grupos. Porém, aos sete dias de regeneração glandular,as parótidas das ratas do grupo B apresentaram menor grau de regeneração, o qual foi restabelecido aos 14 dias.Conclui-se, portanto, que a ovariectomia não altera morfologicamente a parótida normal e não influencia oprocesso de atrofia glandular, porém retarda o processo regenerativo glandular em sua fase inicial.

Tissue healing after periodontal therapy includes epithelial and connective tissue repair by producing biocompatible root surface. The objective of this study was to investigate whether etching solutions such as Tetracycline-hydrochloride (TTC-HCl) or citric acid might enhance the healing after periodontal therapy in rats. Periodontal defects were induced on the left maxillary 2nd molar in 36 Wistar rats. The periodontal attachment was reduced using a ligature model. The experimental sites were mechanically treated and delineated by blindly preparing a notch at the most apical aspect of the pocket using a Hedstroen endodontic file. The experimental sites were irrigated with a 10% tetracycline hidrocloride solution, an 8% citric acid solution, or a 0,9% NaCl solution (control) thus forming 3 groups of 12 animals each. The animals were euthanized at 3, 7 and 21 days post-treatment when block sections of the experimental sites were collected for histometric analysis. The histological observations describe normal wound maturation without regeneration in the defective site. Using a grid point counting method and two-way analysis of variance, it was shown that tissue fill in the notch did not differ significantly between sites irrigated with the acid solutions. However, tissue fill in these sites was significantly smaller than in the control sites. The results also showed that the area occupied by inflamed connective tissue was wider in the control compared to sites treated with the acid solutions. Based on these results it was concluded that irrigation with an acid solution could enhance healing following periodontal therapy

Dentre os cimentos de hidróxido de cálcio presentes no mercado, o Sealapex® apresenta grande biocompatibilidade e selamento marginal apical, mas não possui a radiopacidade desejada, motivo pelo qual muitos profissionais passaram a agregar algumas substâncias Dentre os cimentos de hidróxido de cálcio presentes no mercado, o Sealapex® apresenta grande biocompatibilidade e selamento marginal apical, mas não possui a radiopacidade desejada, motivo pelo qual muitos profissionais passaram a agregar algumas substâncias radiopacas ao cimento. O propósito deste trabalho foi avaliar a resposta tecidual ao Sealapex® puro e acrescido de iodofórmio ou óxido de zinco, em tecido conjuntivo subcutâneo de ratos. Utilizaram-se trinta animais, nos quais os três materiais, no interior de tubos de polietileno, foram implantados. Após períodos de observação de 14 ou noventa dias, realizou-se o sacrifício e os espécimes foram encaminhados para preparação histológica de rotina. As respostas do tecido conjuntivo em contato com o cimento puro e com as combinações testadas foram avaliadas de forma descritiva e quantitativa através de escores, analisando-se a intensidade da inflamação e do fibrosamento. Os dados obtidos foram analisados estatisticamente utilizando-se o teste de Friedman ou o teste de Mann-Whitney (????=0,05). Reação inflamatória com presença de macrófagos e células gigantes multinucleadas, muitas vezes contendo partículas dos materiais no seu interior e fibrosamento capsular foram encontrados na maioria dos espécimes, em ambos os períodos de observação. O acréscimo de iodofórmio ao cimento Sealapex® praticamente não alterou as propriedades biológicas do cimento puro, enquanto que o acréscimo de óxido de zinco foi capaz de melhorá-las, diminuindo a intensidade do infiltrado inflamatório.

This study histologically evaluated the behavior of bone tissue of rats submitted to osteotomy with conventional diamond burs in high speed and a new ultrasonic diamond tips system (CVD – Chemical Vapor Deposition), at different study periods. The study was conducted on 24 Wistar rats. Osteotomy was performed on the posterior paws of each rat, with utilization of diamond burs in high speed under thorough water cooling at the right paw, and CVD tips at the left paw. Animals were killed at 0, 7, 14 and 21 days after treatment, followed by removal of connective tissue and bone at the area of osteotomy. Specimens were fixated, demineralized, dehydrated and sectioned for histological analysis by polarized light microscopy (25, 100 and 200x magnification) by two examiners. The results demonstrated that there were no differences in the repair of bone defects between the two different materials employed for osteotomy.

Objetivos: O objetivo deste trabalho foi avaliar os efeitos da administração sistêmica do chumbo, preparado de acordo com a farmacotécnica homeopática Plumbum metallicum 30CH, na reparação óssea. Material e métodos: foram utilizados trinta ratos machos com três meses de idade, nos quais foram confeccionados defeitos cirúrgicos trans-ósseos com broca de aço nº 06 na região do ângulo da mandíbula do lado direito, sem utilização de barreira mecânica. Cada animal do grupo experimental recebeu três gotas diárias de Plumbum metallicum 30CH acrescidas à mamadeira de água. Os animais do grupo controle receberam apenas água. Três animais foram sacrificados nos seguintes períodos experimentais: um, sete, 14, 21 e 28 dias após a cirurgia. Após o sacrifício, as mandíbulas foram removidas, descalcificadas e submetidas ao preparo histológico de rotina. Foram submetidas a cortes semi-seriados e coradas pelos métodos de hematoxilina-eosina e tricômico de Masson para análise histológica em microscópio de luz. Resultados: os resultados obtidos neste trabalho mostraram que o medicamento foi eficiente na estimulação da reparação óssea. A análise histológica qualitativa mostrou evidência de efeito favorável do Plumbum mettalicum 30 CH. Apesar da interferência do tecido muscular, os animais mostraram, já a partir do período de 14 dias, neoformação óssea em praticamente toda a extensão do defeito. Conclusão: a utilização do Plumbum metallicum 30CH favoreceu a reparação de defeitos ósseos em mandíbula de ratos machos, sem a utilização de barreira mecânica.

O objetivo deste trabalho foi estudar a atrofia unilateral da glândula parótida de ratos, após a ligadura do ducto excretor,e a regeneração glandular, através de ligadura ductal seguida pela remoção da obstrução após sete dias, analisando as alterações morfológicas glandulares e o potencial proliferativo dos ductos intercalares. Análise morfológica foi também realizada na parótida contralateral. Os ratos foram sacrificados e suas glândulas foram processadas e submetidas às colorações histológicas (HE e PAS) e histoquímica (AgNOR). Durante a atrofia glandular, houve degeneração e redução do parênquima, assim como inflamação e fibrose. Durante a regeneração glandular, houve o restabelecimento do parênquima e redução da inflamação e da fibrose. Nas glândulas contralaterais, houve desorganização e degeneração teciduais. Não foi observada alteração do potencial proliferativo dos ductos intercalares durante o experimento.

Neste trabalho os autores realizaram estudo histológico da reparação óssea de defeitos realizados em mandíbulas de ratos. Foram utilizados 18 animais, nos quais realizaram-se perfurações trans-ósseas de 4,0 mm de diâmetro na região de ângulo. Os animais foram sacrificados em grupos de três em períodos pós-operatórios de 1, 3, 5, 7, 14 e 28 dias. Após a fixação, os fragmentos das mandíbulas contendo a área da perfuração foram incluídos em parafina e corados pelo HE. Os resultados mostraram neoformação óssea nas margens do defeito a partir de 3 dias, bem como ao redor de algumas esquírolas ósseas. No período final de observação, de 28 dias, não houve total preenchimento do defeito por tecido ósseo.

A reparação óssea, principalmente em decorrência dos implantes osseintegrados, tem sido objeto de estudos em diferentes condições como, por exemplo, as alterações hormonais decorrentes do climatério. Neste trabalho foi estudado a ação da calcitonina no processo de reparação óssea em tíbias de ratas ovariectomizadas. Doze ratas com aproximadamente sessenta dias foram ovariectomizadas e, um mês após esta cirurgia, um defeito ósseo na tíbia foi realizado, através de processo cirúrgico utilizando broca esférica n.º 6, sob irrigação constante de soro fisiológico. Imediatamente após a cirurgia, iniciou-se a administração de calcitonina 2 UI/kg, i.m., três vezes por semana em nove animais. Estes foram sacrificados após sete, 14 e 21 dias, sendo sua tíbias removidas e encaminhadas para preparação histológica de rotina. As ratas do grupo controle (3) foram submetidas ao mesmo procedimento, recebendo apenas soro fisiológico. Todos os espécimes foram analisados histológicamente.

There are no previous studies on the comparative virulence of Candida dubliniensis with other non-albicans species. The aim of this study was to compare the virulence and infection kinetics of C. dubliniensis and other species. Candida albicans, C. dubliniensis, Candida tropicalis and Candida krusei (reference strains) were inoculated intravenously in mice. For infection kinetics evaluation, a group of five animals were sacrificed after 6 h, 3, 7, 14 and 21 days. Microbiological evaluations (liver, spleen, kidneys, lungs and brain) and histopathological examination of the kidney were performed. The results of virulence evaluation were analysed using Kaplan-Meier survival analysis (5%). Candida dubliniensis-inoculated mice survived for longer periods compared with those with C. albicans (P = 0.005). No differences were detected in relation to C. tropicalis (P = 0.326) and C. krusei (P = 0.317). Most of the organs were persistently colonised by C. albicans and C. dubliniensis even by day 21. Tendency of C. krusei clearance was observed in all organs. Fungal masses and renal lesions were observed after inoculation of C. albicans, C. dubliniensis and C. tropicalis. Within the limits of the study, data on survival rate and dissemination capacity suggest that C. dubliniensis is less virulent than C. albicans.

The objective of this study was to evaluate the effect of estrogen deficiency, estrogen therapy, and soy isoflavones on the salivary glands in female rats. Ninety-six animals were ovariectomized, and 24 were sham-operated. Among the ovariectomized rats, 24 received 17β-estradiol; 24 received isoflavone extract; 24 received a combination therapy of both; and 24 received water as placebo. The submandibular glands were histomorphometrically analyzed. As a result, the ANOVA test revealed that the hormonal deficiency affected the acini and the ducts of ovariectomized rats, reducing their percentage compared to the sham group. All treatments caused an increase in ducts and acini compared to the placebo group. It was concluded that the estrogen deficiency may be related to salivary gland function due to a reduction in the quantity of salivary acini and ducts secondary to ovariectomy. The estrogen therapy, soy isoflavone therapy, and the combination of both are effective in reducing the effects of ovariectomy on the salivary glands.

No. of bitstreams: 0 Previous issue date: 2010-03-01 Objective: The aim of this study was to carry out an in vivo assessment of bone ingrowth in two different types of porous titanium -the first being completely porous, and the second with a porous surface and dense nucleus, manufactured by powder metallurgy- and to evaluate their mechanical properties. Study design: Ten scaffolds from each group were submitted to metallographic analysis and compression tests. Next, two scaffolds of each type were inserted into 14 rabbits, which were sacrificed 8 weeks after surgery. The samples were submitted for histological examination. Results: Metallographic analysis revealed interconnected pores, and the average interconnected pore diameter was about 360 mm, with 36% total porosity. The totally porous titanium samples and the titanium samples with porous surface and dense nucleus showed an average compressive strength of 16.19 MPa and 69.27 MPa, respectively. After 8 weeks, the animals showed bone ingrowth, even into the most internal pores. Conclusions: The pore morphology was effective in permitting bone ingrowth in both groups. Titanium scaffolds with a porous surface and dense nucleus showed the best mechanical properties and most adequate interface. No. of bitstreams: 0 Previous issue date: 2010-01-01 Objective: The present study evaluated morphometrically bone loss percentages in experimental periodontitis in rats, comparing different locations (lingual mandible, palatal maxilla and buccal maxilla) and two evaluation methods (distance and area methods). Material and Methods: Ligatures were placed around the maxillary right second molar and around the mandibular right first molar in 14 female Wistar rats. The contralateral molars served as intragroup controls. After 4 weeks, the rats were sacrificed and their mandible and maxilla were removed. The specimens were dissected and stained with methylene blue dye. Bone loss was evaluated by two different methods on the surfaces of the defleshed jaw. In the first method, the distance from the cementoenamel junction (CEJ) to the alveolar bone crest was measured in the roots of teeth associated with ligature. In the second method, the area of bone loss was determined using the alveolar tissue bone, CEJ and the proximal region of roots associated with the ligature as reference. The data were converted to bone loss percentages caused by ligature: (ligated - unligated) × 100/ligated. Results: When comparing the distance and area methods, no statistically significant difference was observed (p>0.05). The expression of proteins related to cell proliferation and apoptosis in oral lichen planus and epithelial dysplasia was analyzed to evaluate the true potential for malignant transformation of this disease. Twenty-four cases of each lesion were subjected to the streptoavidin-biotin technique for identifying the immunohistochemical expression of PCNA, p53, bax, and bcl-2 proteins. Of the 24 cases of oral lichen planus, 14 (58.33%) were positive for PCNA, 10 (41.67%) for p53, 4 (16.67%) for bcl-2 and 12 (50%) for bax, whereas of the 24 cases of epithelial dysplasia, 20 (83.33%) were positive for PCNA, 10 (41.67%) for p53, 6 (25%) for bcl-2, and 20 (83.33%) for bax. Chi-squared test showed no statistically significant differences between the expression of p53 and bcl-2 in oral lichen planus and epithelial dysplasia, regardless of the grade (P > 0.05). However, the expression of PCNA and bax was significantly increased in epithelial dysplasia (P < 0.05). The results of this study showed that alterations in expression of these proteins are observed in oral lichen planus and epithelial dysplasia, suggesting the potential for malignant transformation in both lesions.

No. of bitstreams: 0 Previous issue date: 2009-11-01 Background: Although oral lichen planus has been classified by the World Health Organization (WHO) as a potentially malignant disorder, such classification is still the target of much controversy. Aim: To evaluate the cell proliferation rate in oral lichen planus, comparing it to the rate observed in epithelial dysplasia and oral squamous cell carcinoma, aiming at indications which might indicate the potential for malignant transformation. Material and Methods: Twenty-four cases of each lesion were submitted to the streptoavidin-biotin and AgNOR technique to evaluate the immunohistochemical expression of PCNA and the mean NORs/ nucleus, respectively. Results: Positivity for PCNA was observed in 58.33% of oral lichen planus cases, 83.33% of epithelial dysplasia cases and 91.67% of oral squamous cell carcinoma cases. Chi-squared test showed that the number of positive cases for PCNA was significantly lower in oral lichen planus than in oral squamous cell carcinoma (p0.05) and between the epithelial dysplasia and oral squamous cell carcinoma (p>0.05) was observed. The mean NORs/ nucleus in oral lichen planus, epithelial dysplasia and oral squamous cell carcinoma were 1.74 +/- 0.32, 2.42 +/- 0.62 e 2.41 +/- 0.61, respectively. No. of bitstreams: 0 Previous issue date: 2009 Lymphoblastic lymphoma is a malignant neoplasia that originates from B or T lymphocyte precursors and rarely occurs in the mouth. The authors report a rare case of B-cell lymphoblastic lymphoma in the maxilla of a child. Clinical examination revealed facial asymmetry with a swelling of the right maxilla, covered by healthy mucosa and painful to palpation. Radiographic examination revealed a poorly defined radiolucent lesion. Based on the hypothesis of malignant neoplasia of hematopoietic origin, an incisional biopsy was performed. Histological examination revealed malignant neoplasia with proliferation of monomorphic, lymphoid cells. Immunohistochemical staining was positive for leucocyte common antigen (LCA), CD 10, CD20, CD79, and terminal deoxynucleotidyl transferase (TdT). After the diagnosis of B-cell lymphoblastic lymphoma, the patient underwent chemotherapy, but died of leukoencephalopathy and demyelinization caused by high doses of methotrexate. [Cavalcante, A. S. No. of bitstreams: 0 Previous issue date: 2009-12-01 Ameloblastic fibro-odontoma (AFO) is a rare, benign, slow-growing odontogenic tumor, generally asymptomatic and more prevalent in children and adolescents. We report a case of AFO in the mandible of an eight-year-old Caucasian male patient, and review the literature. Intraoral examination revealed a swelling extending from the deciduous second molar to the retromolar triangle, covered with normal mucosa. A panoramic radiograph showed a large, well-demarcated radiolucency with radiopaque areas. The provisional diagnosis was of AFO, and so an incisional biopsy was performed. Histologically, the lesion was composed of connective tissue resembling the dental papilla, with epithelial strands or islands, as well as denticles and amorphous masses of enamel and dentin consistent with a diagnosis of AFO. Surgical excision and curettage of the lesion were performed. The patient has been monitored for eight years and the lesion has not recurred. The purpose of this article was to describe the clinical and microscopic features of an intraosseous foreign-body granuloma in the mandible that developed after the traumatic implantation of metal fragments during a work-related accident. A 65-year-old male patient had a severe pain in the body of mandible. Clinical examination showed facial asymmetry and a scar, extending to the left mental region. Intraoral examination revealed a soft mass involving the left alveolar bone with normal appearance of the mucosa surface. Panoramic radiographs showed a radiolucent lesion along the mandible extending from the central incisive to the first molar. Computed tomography revealed an osteolytic mass in the same area. His medical history included a work-related accident twenty years prior to evaluation. During the biopsy an important amount of bright metal-like pieces surrounded by soft tissue were found. A microscopic examination showed a foreign body associated with an aggregation of multinucleated giant cells. The final diagnosis was a foreign body granuloma. Even though foreign-body granulomas in the mandible are rare lesions, dentists should be familiar with their features and include them in the differential diagnosis of tissue masses.

No. of bitstreams: 0 Previous issue date: 2008-07-01 Porous titanium scaffolds are promising materials for biomedical applications such as prosthetic anchors, fillers and bone reconstruction. This study evaluated the bone/titanium interface of scaffolds with interconnected pores prepared by powder metallurgy, using scanning electron microscopy (SEM) and energy dispersive spectroscopy (EDS). Porous scaffolds and dense samples were implanted in the tibia of rabbits, which were subsequently killed 1, 4, and 8 weeks after surgery. Initial bone neoformation was observed one week after implantation. Bone ingrowth in pores and the Ca/P ratio at the interface were remarkably enhanced at 4 and 8 weeks. The results showed that the interconnected pores of the titanium scaffolds promoted bone ingrowth, which increased over time. The powder metallurgy technique thus proved effective in producing porous scaffolds and dense titanium for biomedical applications, allowing for adequate control of pore size and porosity and promoting bone ingrowth. The aim of the present study was to analyze the main clinical and histopathological features of autoimmune diseases with oral manifestations such as oral lichen planus (OLP); mucous membrane pemphigoid (MMP); pemphigus vulgaris (PV) and erythema multiforme (EM).Retrospective review of 5770 files from the Oral Pathology Laboratory of São José dos Campos Dental School, São Paulo State University (UNESP) comprising a 27-year period from 1974 to 2000.The cases accounted for 64 (1.10%) of 5770 anatomopathological examinations performed over the study period. Among the autoimmune diseases diagnosed, 49 (76.56%) were OLP, 6 (9.37%) were MMP, 5 (7.82%) were EM and 4 (6.25%) were PV. Descriptive statistical analysis was used.The initial manifestations of most autoimmune diseases occur in the oral mucosa, an earlier diagnosis and proper therapeutic protocol will delay the dissemination of the lesions, thus greatly contributing to a better prognosis and quality of life of the patient.

OBJETIVOS: Relatar dois casos com história clínica semelhante, confrontando seus aspectos com aqueles encontrados na literatura. RELATO DOS CASOS: Dois casos de mulheres na quinta e sexta décadas de vida apresentando nódulo submucoso em ventre lingual. Após exame microscópico que revelou numerosos vasos sangüíneos entremeados a células fusiformes, com núcleos ovalados ou alongados, e exame imuno-histoquímico com positividade para actina de músculo liso nos dois casos, o diagnóstico final foi leiomioma vascular. CONCLUSÃO: O estudo do leiomioma vascular bucal é de extrema importância devido à raridade e semelhança desse com outras lesões da cavidade bucal. A análise imuno-histoquímica é importante para o diagnóstico final do leiomioma. INTRODUCTION: The leiomyomas are benign tumors of smooth muscle origin, which are more frequently found in genitourinary and gastrointestinal tracts. However, leiomyomas rarely arise in the oral cavity, and when it happens, the majority of them is originated by smooth muscle of blood vessels. AIMS: Report two cases of oral vascular leiomyoma with similar clinical characteristics, and discuss their aspects with the literature. CASE REPORT: Two cases of vascular leiomyoma in women that presented a nodular mass in the tongue ventral surface were described. The microscopic analysis revealed numerous blood vessels surrounded by spindle cells with ovoid or elongated nuclei. The two cases presented immunopositivity for smooth muscle actin, confirming the diagnosis. CONCLUSION: The study of vascular leiomyoma is required due to its rarity and similar aspects with other oral common lesions. The immunohistochemical analysis is a fundamental tool for the final diagnosis of leiomyoma. The aim of this study was to report an unusual case of mucoepidermoid carcinoma (MEC) in a 39-year-old woman. The tumor showed a prominent population of clear and intermediate basal cells. Clear cells rarely predominate over other cell types. Such cases are called clear cell variant of MEC. The case also revealed a variable amount of calcified material in the tumor mass. Calcifications are rare in clear cell MEC. These structures were periodic acid-Schiff positive and diastase resistant, excluding glycogen origin. Immunohistochemistry was performed, and the epidermoid component was positive for cytokeratin (CK)7, CK13, CK14, and CK19. The mucous and clear cells presented mild staining for CK7. Cytokeratins 7, 13, and 19 stained luminal cells, and intermediate cells exhibited positivity for CK7, CK14, and vimentin. The origin of the calcifications is speculated to be the result of dystrophic calcification of the amorphous eosinophilic material secreted by intermediate basal cells.

The aim of this study was to investigate, by means of histological and histomorphometric analysis, the effects of 24% ethylenediaminetetraacetic acid (EDTA) gel in periodontal tissue when used in combination with conventional periodontal treatment. Periodontitis was induced in the 2nd upper left permanent molars of 45 male Wistar rats by means of ligature. After 5 weeks, this was removed and debridement was performed. The animals were then randomly divided into 3 groups; group 1: mechanical treatment, group 2: mechanical treatment and EDTA gel application for 2 min, and group 3: mechanical treatment and placebo gel application for 2 min. After the treatment, rinsing was done with 0.9% saline solution for 1 min in all cases, followed by root notching in the deepest part of the pocket. After 4, 10, and 28 days the animals were sacrificed. The averages obtained were evaluated by means of test two-way analysis of variance (ANOVA) and Tukey statistical tests (P < 0.05). The results showed that with respect to the type of treatment employed, there were no statistically significant differences in the vitality of the periodontal tissue. It was concluded that 24% EDTA gel did not interfere with periodontal tissue repair when used in combination with conventional periodontal treatment. (J. Oral Sci. 48, 207-214, 2006)

Sjögren's syndrome is an autoimmune disease characterized by sialoadenitis and elevated titers of autoantibodies. To assess whether it is possible to induce inflammatory changes in salivary gland tissues, a series of immunizations in Balb/c mice have been undertaken, using salivary gland extract, modified or not, added to several adjuvants. Mice's humoral immune response to salivary gland antigens was monitored by ELISA. Inflammatory cells infiltrating gland tissue were seen 3 months after immunization with salivary gland extract modified with pepsin (AgGp) and metaperiodate (AgGMp). Although pathological progression was not observed, the histopathological picture was similar to the initial phase of Sjögren's syndrome. In addition, a monoclonal antibody reactive with 3 gland polypeptides and anhydrase carbonic II was rescued among B cells from immunized mice. Thus, immunizations with modified autoantigens were able to initiate pathological damage to glandular tissue and stimulate the proliferation of auto-reactive B cells. A Síndrome de Sjögren é uma doença auto-imune caracterizada por desenvolvimento de sialoadenite e títulos elevados de auto-anticorpos. Com o objetivo de induzir alterações inflamatórias no tecido das glândulas salivares foram realizadas várias imunizações em camundongos BALB/c utilizando extratos de glândulas salivares, modificados ou não, em vários adjuvantes. A resposta humoral para antígenos salivares foi monitorada por ELISA. Células inflamatórias infiltrando o tecido glandular foram vistas 3 meses pós-imunização com extrato de glândula salivar modificado com pepsina (AgGp) e metaperiodato (AgGMp). Embora a evolução patológica não tenha sido observada, o quadro histopatológico foi semelhante à fase inicial da Síndrome de Sjögren. Também foi possível notar, a partir das células B dos animais imunizados, a produção de anticorpos monoclonais reativos com 3 polipeptídeos glandulares e anidrase carbônica II. Assim, a imunização com auto-antígenos glandulares modificados foi capaz de iniciar o processo patológico no tecido glandular e induzir a proliferação de células B produtoras de auto-anticorpos.

The fixation and the bone ingrowth at the interface of porous cylindrical implants (total porosity of 37% and average pores diameter of 480 μm) were compared in vivo to rough cylindrical implants (R a = 5.3 μm), both of commercially pure titanium, made by powder metallurgy. The implants were inserted into the tibias of 20 rabbits and the animals were sacrificed 4 and 8 weeks after surgery. The percentage of bone–implant contact observed in porous implant was significantly larger than in the rough ones for all of sacrifice periods, respectively, 57% vs. 46% after 4 weeks, and 59% vs. 50% after 8 weeks. The mechanical tests showed a significant increase in the shear strength of the porous implants for the two analyzed periods, 4 and 8 weeks (14 and 20 MPa), when compared with rough ones (4 and 13 MPa). These results suggest that porous implants improve the contact at the implant–bone interface and increase the fixation to the bone, improving the osseointegration. Thus, the porous implant might be an alternative to dental implant in less favorable conditions, and appear to be better fixed to bone, offering promising alternatives.

Polymorphous low-grade adenocarcinoma (PLGA) is a subtype of salivary gland carcinoma with a marked predilection for the minor salivary glands. The diagnosis of this lesion can be a challenge in incisional biopsies because of its similarity to other salivary neoplasms and the difficulty of analyzing its infiltrative nature and neurotropism. Diagnosis can be facilitated, however, by immunohistochemistry, a technique that has become a fundamental tool to the pathologist. The purpose of this study was analyze the immunohistochemical aspects of 4 cases that were examined in the São José dos Campos School of Dentistry. The patients included 4 women, ages 32 to 57 years (mean, 50 years), who reported an intraoral painless lesion, ranging in size from 1 to 3 cm (mean, 1.75 cm) with a variable evolution period. Histologically, the lesions showed similar aspects, characterized by polymorphous growth pattern, including solid, cribriform, trabecular, papillary, and "single-file" arrays. The tumors showed positivity to cytokeratins (Cks) 7 and 8 and vimentin, variable expression for Ck 14, and a negative reaction to Cks 13 and 19 and muscle-specific actin. PLGA is a tumor of the salivary gland that must be distinguished from other neoplasms for therapeutic and prognostic considerations, and the panel of antibodies employed in this study was effective for its diagnosis.

Essa expansão da pesquisa deve-se, em parte, ao reconhecimento da osteoporose como importante problema na velhice. A osteoporose constitui uma das osteopatias mais comuns, caracterizando-se pela redução da massa óssea, determinada, por sua vez, pelo desequilíbrio entre reabsorção e neoformação. OBJETIVO: Apresentar uma revisão da literatura sobre os principais aspectos da remodelação e da reparação associados à deficiência estrogênica. Remodelação óssea: O osso apresenta processo contínuo de remodelação, entretanto anormalidades nesse processo ocorrem em algumas doenças, entre elas a osteoporose, sendo que a deficiência estrogênica parece ter o papel principal na sua gênese. Reparação óssea: Tal processo envolve uma cascata complexa de respostas biológicase, assim como a remodelação, é afetado por fatores locais e externos e regulado pela interação de diferentes mecanismos. Portanto, o aumento ou o decréscimo da capacidade de reparação óssea têm sido relacionados a alterações ocorridas na remodelação. Deficiência estrogênica e metabolismo ósseo: A maioria dos autores sugere uma redução na capacidade de remodelação e de reparação do tecido. DISCUSSÃO: Ainda não está determinado qual estágio da reparação é mais alterado, se a fase inicial de formação do calo ósseo, se a de mineralização ou se a fase tardia da reparação, a remodelação óssea. CONCLUSÃO: Como os mecanismos fisiológicos e a patogênese das alterações ósseas causadas pela deficiência estrogênica não estão completamente estabelecidos, novas pesquisas ainda são necessárias. INTRODUCTION: In the past few years, it was recognized the great complexity of the mechanisms that influence bone cellular activity and several studies focus on the factors able to modulate the bone functions. The increase of bone research is, in part, due to the establishment of osteoporosis as a healthy problem common in elderly. Osteoporosis is one of the most important osteopathy, characterized by the bone mass reduction, resulted from disequilibrium between bone resorption and bone formation. OBJECTIVE: Based on the relationship between estrogen and bone metabolism, the aim of this study is present a review of literature about the principal aspects of bone turnover and bone repair associated to estrogen deficiency. Bone turnover: Bone tissue is in continuous turnover, however, changes in this process can result in some disorders, such as osteoporosis. Bone repair: Involves a sequence of biological events. It is affected by local and external factors and regulated by interaction of several mechanisms, like bone turnover. Estrogen deficiency and bone metabolism: The capacity to repair has been associated to changes in bone turnover and repair. DISCUSSION: It is not known which bone repair stage is modified: the bone formation, the mineralization or the resorption stage. CONCLUSION: The pathophysiology of bone changes caused by estrogen deficiency are not completely clear, so, new studies are still necessary. Os implantes foram inseridos em tíbias de coelhos. Foram utilizados neste estudo 7 coelhos machos, sendo que cada um recebeu 3 implantes de superfície porosa na tíbia esquerda e 3 implantes de superfície rugosa na tíbia direita. Os animais foram sacrificados 4 semanas após a cirurgia e os fragmentos das tíbias contendo os implantes foram submetidos à análise histológica e histomorfométrica, visando analisar a neoformação óssea na interface osso-implante. As médias (%) obtidas na análise histomorfométrica foram avaliadas por meio do teste estatístico t-student de amostras pareadas com nível de significância de 5%. Os resultados da análise histológica mostraram que a osseointegração foi obtida nos dois tipos de implantes com similar qualidade de tecido ósseo. Na análise histomorfométrica, verificaram-se médias de neoformação óssea na interface osso-implante de 79,69% ± 1,00 e 65,05 ± 1,23 para os implantes de superfície porosa e rugosa, respectivamente, e foi observada diferença estatisticamente significante entre os dois tipos de implantes com relação à quantidade de neoformação óssea. Concluiu-se que os implantes de superfície porosa contribuíram para a osseointegração devido à sua maior superfície de contato na interface osso-implante. The purpose of this study was to analyze the bone repair around commercially pure titanium implants with rough and porous surface, fabricated using powder metallurgy technique, after their insertion in tibiae of rabbits. Seven male rabbits were used. Each animal received 3 porous-surface implants in the left tibia and 3 rough-surface implants in the right tibia. The rabbits were sacrificed 4 weeks after surgery and fragments of the tibiae containing the implants were submitted to histological and histomorphometric analyses to evaluate new bone formation at the implant-bone interface. Means (%) of bone neoformation obtained in the histomorphometric analysis were compared by Student's t-test for paired samples at 5% significance level.. The results of the histological analysis showed that osseointegration occurred for both types of implants with similar quality of bone tissue. The histomorphometric analysis revealed means of new bone formation at implant-bone interface of 79.69 ± 1.00% and 65.05 ± 1.23% for the porous- and rough-surface implants, respectively. The aim of this study was to evaluate the bone healing in spontaneously hypertensive rats (SHR) and compare the results with normotensive rats, evaluating male and female animals.A bone drill defect was created in the left femur of 24 SHR (12 males and 12 females) and 24 normotensive rats (12 males and 12 females). The animals were divided into two groups and sacrificed 7 and 21 days after the surgical procedure. After the routine laboratory processing, histological and histometric analysis were carried out and data were submitted to ANOVA and Tukeys test (5%).Males and females from the same group had similar histological characteristics. After seven days, all animals presented irregular bone trabeculae. The periosteal osteoblasts were flattened in SHR, and presented a cuboid shape in normotensive animals. After 21 days, the bone defects of all specimens showed a linear closure in all the superficial extension. In addition, SHR presented flattened osteoblasts surrounding the bone trabeculae, while normotensive ones showed cuboidal cells. Statistical analysis of the histometric data indicated similar means between the male and female groups, except for normotensive rats on day 7. In addition, a larger amount of new bone formation was observed in hypertensive when compared to normotensive rats on day 21, in males as well as females.We conclude that bone healing in SHR was more significant than in normotensive ones, as shown by the histological and histometric evaluation 21 days after surgery.

The purpose of this study was to evaluate the effects of simvastatin, by oral or subcutaneous administration, on tibial defects regeneration and blood cholesterol level in rats. A surgical defect was made on the right tibia of 40 male animals assigned to 4 groups (n=10), based on two routes of administration and on the use or not of simvastatin: subcutaneous injection of simvastatin (7 mg/kg) (group AT) or only the vehicle of drug suspension (group AC), above the defect area, for 5 days; and 20 mg/kg of simvastatin macerated on water (group BT) or only water (group BC), orally, daily, during the whole observation period. The animals were sacrificed after 15 or 30 days, when blood samples were analyzed to check plasma cholesterol levels. Tibiae were removed and, after decalcification and routine laboratorial processing, histological and histomorphometrical analyses were carried out. ANOVA was used for statistical analysis at 5% signficance level. The histological and histomorphometrical analyses showed significant differences only between the experimental periods (p0.05) for blood cholesterol levels between the groups. In conclusion, simvastatin administration either orally or subcutaneously did not improve bone repair of experimental tibial defects and did not alter blood cholesterol levels in rats. Este estudo avaliou a influência da sinvastatina, administrada por via oral ou subcutânea, na reparação de defeitos ósseos em tíbia e nos níveis de colesterol sangüíneo, em ratos. Foram realizados defeitos cirúrgicos nas tíbias direitas de 40 ratos machos, distribuídos em 4 grupos (n=10), tomando-se como base duas vias de administração e o uso ou não de sinvastatina: injeção subcutânea de sinvastatina (7 mg/kg) (grupo AT) ou apenas do veículo de suspensão da droga (grupo AC), sobre a região do defeito, durante 5 dias; 20 mg/kg de sinvastatina (grupo BT) ou água filtrada (grupo BC) via oral, diariamente, durante todo o período de observação. Os animais foram sacrificados após 15 ou 30 dias, quando amostras sangüíneas foram colhidas para análise do nível de colesterol. As tíbias foram removidas e, após descalcificação e procedimentos laboratoriais de rotina, procedeu-se à análise histológica e histomorfométrica. Para avaliação estatística utilizou-se ANOVA com nível de significância de 5%. As análises histomorfométrica e histológica mostraram diferença entre os grupos apenas com relação ao período experimental (p0,05). Concluiu-se que, nas condições utilizadas, a sinvastatina, administrada via oral ou subcutânea, não exerceu efeito estimulador sobre o reparo ósseo de defeitos experimentais em tíbias de ratos e não alterou os níveis de colesterol sangüíneo.

Cento e vinte e quatro ratos originalmente negativos para Candida spp. na cavidade oral foram divididos em dois grupos: ovariectomizados e sham ‐ ovariectomizados. Cinqüenta e oito ratos ovariectomizados e a mesma quantidade de ratos sham ‐ ovariectomizados foram inoculados com C. albicanspara o estudo do desenvolvimento da candidose e recuperação de leveduras. Quatro animais de cada grupo não foram inoculados com suspensão de leveduras e submetidos à análise morfológica do dorso da língua por microscopia óptica e eletrônica de varredura. O desenvolvimento de candidose no dorso da língua foi observado por microscopia óptica e eletrônica de varredura nos períodos de 6 horas, 24 horas, 7 dias e 15 dias após a última inoculação. A recuperação de C. albicans foi realizada por amostras orais em placas de ágar Sabouraud após 1, 2, 5 e 7 dias e progressivamente a cada intervalo de 15 dias, até a obtenção de culturas negativas para leveduras. Os resultados foram analisados ​​pelos testes t de Mann ‐ Whitney e Student . Dorso da língua de ratos sham ‐ ovariectomizados e ovariectomizados, não infectados por Candida, apresentou aspecto normal. Entre os ratos infectados, o grupo ovariectomizado apresentou menor ocorrência de lesões de candidose e menor recuperação de C. albicans da cavidade oral em relação ao grupo simulado ovariectomizado. Pode-se concluir que a candidose foi menos frequente nas cavidades orais de ratos ovariectomizados em relação aos simulados ovariectomizados. The aim of this study was to analyze the development of candidosis and the recovery of C albicans from the oral cavity of ovariectomized and sham-ovariectomized rats. One hundred and twenty-four rats originally negative for Candida spp. in the oral cavity were divided into two groups: ovariectomized and sham-ovariectomized. Fifty-eight ovariectomized and the same quantity of sham-ovariectomized rats were inoculated with C albicans for the study of candidosis development and recovery of yeast. Four animals from each group were not inoculated with yeast suspension and were submitted to tongue dorsum morphologic analysis by optical and scanning electron microscopy. The development of candidosis in the tongue dorsum was observed by optical and scanning electron microscopy in the periods of 6 hr, 24 hr, 7 days and 15 days after the last inoculation. Recovery of C albicans was performed by oral samples plating on Sabouraud agar after 1, 2, 5 and 7 days and progressively at each 15-day interval until negative cultures for yeasts were obtained. The results were analyzed by Mann-Whitney and Student's t tests. The tongue dorsum of sham-ovariectomized and ovariectomized rats, not infected by Candida, presented normal aspect. Among the infected rats, the ovariectomized group showed less occurrence of candidosis lesions and lower recovery of C albicans from the oral cavity in relation to the sham-ovariectomized group. Dezesseis ratos da raça Wistar, pesando 300g, foram divididos em dois grupos: PerioGlass (PG) (n=8) e Biogran (BG) (n=8). Defeitos ósseos unicorticais de 3 mm de diâmetro foram realizados na tíbia dos animais e preenchidos com os dois tipos de partículas cerâmicas. Os animais foram sacrificados aos 7, 14, 30 e 60 dias no pós-operatório. O material preparado para processamento e exame histológico em microscopia óptica foi corado com hematoxilina-eosina e tricrômio de Mallory. Aos 7 dias ambos os grupos apresentaram septos ósseos neoformados, mais intensos no grupo PG. Isto também foi observado aos 14 dias. Nos períodos posteriores (30 e 60 dias), ambos os grupos apresentaram tecido ósseo mais maduro ao redor das partículas cerâmicas. As trabéculas ósseas formadas em todos os períodos experimentais foram justapostas com as partículas. Podemos concluir que ambos os materiais promoveram o preenchimento ósseo em toda a extensão do defeito independente de suas granulações comprovando suas propriedades osteocondutivas. The main goal of this study was to histologically evaluate the healing of surgically created defects on the tibiae of adult male rats after implantation of two types of bioactive glass. Sixteen adult Wistar rats (body weight of 300g) were divided into two groups: PerioGlas (PG) (n=8) and BioGran (BG) (n=8). Unicortical bone defects with 3-mm diameter were performed in both tibiae of the animals and filled with two types of glass particles. The rats were then sacrificed at 7, 14, 30 and 60 days, and the tissues were prepared for histological processing, sectioning, and staining with hematoxylin and eosin, as well as Mallory trichrome, and analyzed under light microscope. Within 7-14 days, both groups presented connective tissue septa with new bone formation, more intense in the PG group. In the subsequent periods (30 and 60 days), both groups presented more mature bone tissue around the glass particles. Bone trabeculae formed in all experimental periods were juxtaposed to the glass particles. It can be concluded that both materials promoted comparable bone formation over the entire extension of the defect, independently of the size of the granules, thus confirming their biological osteoconductive property. Immunohistochemistry is an important tool when dealing with salivary gland neoplasms. Canalicular adenoma and polymorphous low-grade adenocarcinoma may share some histologic characteristics that can cause difficulties in their separation. In the present study, cases of polymorphous low-grade adenocarcinoma and canalicular adenoma were submitted to a panel of antibodies to evaluate the differences in their immunoprofiles. The results obtained showed that, while vimentin is only expressed by polymorphous low-grade adenocarcinoma, CK7 and CK8 are present in both neoplasms. Therefore, vimentin is the best marker to differentiate between these tumors.

As características do biomaterial e o desenho da membrana empregados na regeneração tecidual guiada desempenham um papel importante na obtenção de bons resultados. O objetivo deste estudo experimental histológico foi comparar o uso de dois tipos de barreiras físicas na regeneração óssea guiada em defeitos criados na tíbia de ratos. Quinze animais foram divididos em três grupos: grupo I (barreira não-porosa de politetrafluoretileno), grupo II (blocos de hidroxiapatita de coral) e grupo III (controle que não recebeu nenhuma barreira). A análise histológica demonstrou várias quantidades de osso neoformado com ambos os tipos de barreiras. A barreira de politetrafluoretileno mostrou melhores resultados do que a hidroxiapatita. Os resultados deste estudo sugerem que a regeneração óssea pode ser conseguida com a técnica de submersão da barreira física. Regeneration of periodontal and alveolar ridge defects utilizing membranes is a well-established procedure in reconstructive surgery. Biomaterial characteristics and membrane design employed in guided tissue regeneration (GTR) techniques play an important role in good results. The purpose of this histologic experimental study in rats was to compare the use of two physical barriers in the osteopromotion by using GTR principles in bone defects created in tibias. Fifteen animals divided into 3groups were used: group I (non-porous polytetrafluoroethylene (PTFE) barrier), group II (coral hydroxyapatite (HA) blocks), and group III (defects that received no physical barrier). Histological examination showed varied amounts of newly formed bone beneath both types of barriers. The non-porous PTFE barrier showed better results than the HA group. The results of this study suggest that bone regeneration can be successfully enhanced by a submerged membrane technique. A panel of antibodies composed of the cytokeratins (CKs), vimentin, and actin was applied to 114 minor salivary gland tumors to evaluate its diagnostic value. The results revealed that luminal cells of intercalated duct-like structures, such as those seen in pleomorphic adenoma, basal cell adenoma, adenoid cystic carcinoma, and epithelial-myoepithelial carcinoma, expressed CKs 7, 8, 14, and 19. The outer cells of these structures exhibited vimentin or vimentin plus muscle-specific actin, but rarely CK14, which is seen particularly in pleomorphic adenoma, in the tubular type of basal cell adenoma, and seldom in the tubular type of adenoid cystic carcinoma. Modified myoepithelial cells of pleomorphic adenoma and myoepithelioma exhibited a variable immunoprofile. CKs 7 and 8 were also observed in acinar cell adenocarcinoma and polymorphous low-grade adenocarcinoma with vimentin in the latter. CK13 was expressed only by canalicular adenoma and mucoepidermoid carcinoma cells. This study showed that the panel of antibodies employed is effective in distinguishing among salivary gland tumors.

Necrotizing sialometaplasia is a benign, self-limiting and rare inflammatory disease which, on clinical and histological examination, mimics malignant neoplasms. Case report. We report the case of a healthy 25-year-old Caucasian woman with a three-week history of a painless lump on her hard palate. Oral examination revealed a nodule consisting of two lobules on the right side that measured 2.5 cm. Her mucosa was normal in color and a fluctuant area was detected in the posterior region upon palpation. Our patient was submitted to incisional biopsy and histopathological examination. The histological diagnosis was necrotizing sialometaplasia. The lesion had healed spontaneously after 30 days, with observed signs of involution of the nodule. Conclusion: Histopathological examination is necessary for the diagnosis of necrotizing sialometaplasia because the clinical features of this condition can mimic other diseases, particularly malignant neoplasms. © 2011 Oliveira Alves et al; licensee BioMed Central Ltd. Full lips provide a youthful, healthy, feminine and sensual appearance. There are many techniques and materials used to recover the loss of contour that occurs with age. Case presentation. An unusual case of fat tissue accumulation following cosmetic upper lip augmentation in a 61 year-old female was reported. Surgical treatment was performed for esthetic concerns. Microscopically, the tissue removed was composed of muscle fibers and mature adipocytes. Diversos estudos têm demonstrado que os implantes de titânio são altamente biocompatíveis e passíveis de osseointegrar, contudo os mecanismos moleculares que atuam por trás da osseointegração permanecem largamente inexplorados. Uma das possibilidades é que o implante exposto ao sangue do paciente durante a cirurgia absorve proteínas relacionadas com a adesão celular, como a fibronectina e vitronectina presentes no plasma. Células como osteoblastos podem então aderir a estas proteínas através dos mecanismos mediadas pelas integrinas. No presente estudo, utilizamos a imunofluorescência para marcação dos anticorpos contra integrinas α2, α3, α5, α6, αv e β1 em células OsA-CL cultivadas sobre lamínulas de vidro e superfície de titânio modificada por radiação à laser no período de 1h à 24 horas. As células aderidas na superfície lisa das lamínulas de vidro tiveram um maior espalhamento durante as primeiras 3 horas, porém nos outros períodos estudados o espalhamento ocorreu de maneira similar em ambas as superfícies. A expressão de integrinas na superfície rugosa dos implantes manteve-se uniforme em todos os períodos estudados, contudo no grupo controle a expressão de integrinas diminuiu na avaliação de 24 horas. Concluiu-se que as características de superfície dos diferentes biomateriais podem afetar o comportamento celular durante a interação inicial das células com a superfície de material utilizado como substrato. Many studies have been demonstrate that titanium implants are highly biocompatible, however the molecular mechanisms that are behind of the osseointegration process are still largely unexplored. One of the possibilities is that the implant exposed to the patient blood during the surgery, adsorb proteins related to the cellular adhesion. Cells like the osteoblasts can adhere to these proteins trough the mechanisms mediated by integrins. In the present study, we used the immunofluorescence for marking antibodies against α2, α3, α5, α6,, αv and β1 integrins on culture of OsA-CL cells on laminulas of glass and modified titanium surface modified by laser radiation in the period of 1h to 24 hs. Cells adhered in the smooth surface of laminulas of glass had a bigger spreading during the first 3 hours, however in the other studied periods the spreading occurred in a similar way in both surfaces. The expression of integrins in the roughness surface of the implants remained uniform in all the studied periods, but on the control group the integrins expression decreased in the 24-hours evaluation. We concluded that the characteristics of surface of the different biomaterials can affect the cellular behavior during the initial interaction of the cells with the surface of the used material as substratum.

A matriz extracelular (MEC) pode ser definida como um complexo de proteínas e glicoproteínas que envolve as células nos mais diversos tecidos e tem papel importante na diferenciação e atividade celular, bem como no processo de mineralização e nos processos neoplásicos. Os componentes não colágenos da MEC têm sido abundantemente estudados, visando conhecer os minuciosos detalhes da biologia dos tecidos e assim entender os mecanismos envolvidos em suas patologias. Neste contexto, o presente trabalho tem como objetivo estudar a expressão e distribuição dos seguintes componentes não colágenos da MEC dos tecidos dentais: biglican, decorin, fibromodulin, osteonectina (ONC), osteopontina (OPN), sialoproteína óssea (BSP) e osteocalcina (OCC) no ameloblastoma e no tumor odontogênico cístico calcificante (cisto de Gorlin). Para ta nto foi utilizada a técnica da imunoistoquímica, com o método da estreptavidina-biotina-peroxidase, e anticorpos contra as proteínas anteriormente citadas. Os resultados mostraram que o biglican, o decorin e a BSP foram expressos somente nas células epiteliais metaplásicas, nas células fantasmas e células fantasmas em processo de calcificação, no estroma dos ameloblastomas e no ectomesênquima neoplásico do tumor odontogênico cístico calcificante. Já o fibromodulin e a OC foram predominantemente negativos no componente epitelial e no mesenquimal, com exceção para as células fantasmas, células fantasmas em processo de calcificação e áreas de hialinização próximas ao epitélio. A ONC foi positiva na maioria das células epiteliais, com exceção das células estrelárias dos ameloblastomas folicular e acantomatoso, e também no componente mesenquimal de ambas neoplasias. Já a OPN apresentou positividade somente nos focos de calcificação presentes no tumor odontogênico cístico calcificante. As proteínas estudadas apresentaram distribuição semelhante em neoplasias caracterizadas por padrões de crescimento diferentes, levando a crer que, apesar de participarem ativamente do mecanismo de crescimento neoplásico intra-ósseo, isoladamente não exercem papel decisivo na determinação do tipo de padrão de crescimento. Outro fato digno de relevância é a baixa expressão dessas proteínas nas células epiteliais neoplásicas quando comparada com a expressão no estroma e ectomesênquima, levando-nos a crer que as células epiteliais atuem principalmente como estimuladores da expressão dessas proteínas, que, por sua vez, podem atuar de forma agonista ou antagonista ao crescimento neoplásico. The extracellular matrix (ECM) can be defined as a complex of proteins and glycoproteins that involves the cells in all tissues. It has a key role in cell differentiation and activity, as well as in mineralization and neoplastic processes. The non-collagenous components of the ECM have been abundantly studied to know the details of the biology of tissues and thus to understand the mechanisms involved in its pathologies. The aim of the present work is to study the expression and distribution of the following noncollagenous components of the ECM of dental tissues: biglycan, decorin, fibromodulin, osteonectin (ONC), osteopontin (OPN), bone sialoprotein (BSP) and osteocalcin (OCC) in ameloblastoma and the calcifying cystic odontogenic tumour. The streptavidin-biotinperoxidase method of immunohistochemistry was used with antibodies against the antigens previously cited. The results show that biglican, decorin and BSP had been expressed only in metaplastic epithelial cells, in ghost cells and ghost cells in calcification process, stroma of ameloblastomas and neoplastic ectomesenchyma of the calcifying cystic odontogenic tumour. The fibromodulin and the OCC showed predominantly negative expression in the epithelial and mesenchymal components, with exception for the ghost cells, ghost cells in calcification process and hyalinization areas next to the epithelium. The ONC was positive in the majority of the epithelial cells, with exception of the central cells of follicular and acanthomatous ameloblastomas, and also in the mesenchymal component of both tumours. OPN presented positivity only in the calcification focus of the calcifying cystic odontogenic tumour. The proteins studied presented similar distribution in tumours characterized by different patterns of growth, leading to believe that although they participate actively of the mechanism of intraosseous growth, separately they do not exert a key role in the determination of the type of growth pattern. Another relevance fact is the low expression of these proteins in the neoplastic epithelial cells when compared to the expression in stroma and ectomesenchyma, which make us believe that the epithelial cells act mainly as stimulators of the expression of these proteins, which in turn can act as agonist or antagonist to the tumour growth.
