ObjectiveThe surgical management of lesions involving the lateral area of the suprasellar region, including the lateral aspect of the planum sphenoidale and the tight junction region of the optic canal (OC), anterior clinoid process (ACP), and internal carotid artery (ICA) and its dural rings, is extremely challenging. Here, these regions, namely, the “parasuprasellar” area, are described from the endonasal perspective. Moreover, the authors introduce two novels endoscopic endonasal supraoptic (EESO) and endoscopic endonasal infraoptic (EEIO) approaches to access the parasuprasellar area.MethodsSurgical simulation of the EESO and EEIO approaches to the parasuprasellar area was conducted in 5 silicon-injected specimens. The same techniques were applied in 12 patients with lesions involving the parasuprasellar area.ResultsThe EESO approach provided excellent surgical access to the lateral region of the planum sphenoidale, which corresponds to the orbital gyrus of the frontal lobe. With stepwise bone (OC, optic strut and ACP) removal, dissociation of the ophthalmic artery (OA) and optic nerve (ON), the EEIO approach enables access to the lateral region of the supraclinoidal ICA. These approaches can be used independently or in combination, but are more often employed as a complement to the endoscopic endonasal midline and transcavernous approaches. In clinical application, the EESO and EEIO approaches were successfully performed in 12 patients harboring tumors as well as multiple aneurysms involving the parasuprasellar area. Gross total and subtotal tumor resection were achieved in 9 patients and 1 patient, respectively. For two patients with multiple aneurysms, the lesions were clipped selectively according to location and size. Visual acuity improved in 7 patients, remained stable in 4, and deteriorated in only 1. No postoperative intracranial infection or ICA injury occurred in this series.ConclusionsThe EESO and EEIO approaches offer original treatment options for well-selected lesions involving the parasuprasellar area. They can be combined with the endoscopic endonasal midline and transcavernous approaches to remove extensive pathologies involving the intrasellar, suprasellar, sphenoid, and cavernous sinuses and even the bifurcation of the ICA. This work for the first time pushes the boundary of the endoscopic endonasal approach lateral to the supraclinoidal ICA and ON.

PurposeTo identify novel genetic causes of febrile seizures (FS) and epilepsy with febrile seizures plus (EFS+).MethodsWe performed whole-exome sequencing in a cohort of 32 families, in which at least two individuals were affected by FS or EFS+. The probands, their parents, and available family members were recruited to ascertain whether the genetic variants were co-segregation. Genes with repetitively identified variants with segregations were selected for further studies to define the gene-disease association.ResultsWe identified two heterozygous ATP6V0C mutations (c.64G > A/p.Ala22Thr and c.361_373del/p.Thr121Profs*7) in two unrelated families with six individuals affected by FS or EFS+. The missense mutation was located in the proteolipid c-ring that cooperated with a-subunit forming the hemichannel for proton transferring. It also affected the hydrogen bonds with surround residues and the protein stability, implying a damaging effect. The frameshift mutation resulted in a loss of function by yielding a premature termination of 28 residues at the C-terminus of the protein. The frequencies of ATP6V0C mutations identified in this cohort were significantly higher than that in the control populations. All the six affected individuals suffered from their first FS at the age of 7–8 months. The two probands later manifested afebrile seizures including myoclonic seizures that responded well to lamotrigine. They all displayed favorable outcomes without intellectual or developmental abnormalities, although afebrile seizures or frequent seizures occurred.ConclusionThis study suggests that ATP6V0C is potentially a candidate pathogenic gene of FS and EFS+. Screening for ATP6V0C mutations would help differentiating patients with Dravet syndrome caused by SCN1A mutations, which presented similar clinical manifestation but different responses to antiepileptic treatment.

ObjectivePostoperative hypopituitarism associated with increased risks of premature mobility and mortality is often encountered in craniopharyngioma patients. The aim of our study is to construct nomograms related to injury types of the hypothalamus–pituitary axis (HPA) to predict hypopituitarism 1 year after surgery.MethodsCraniopharyngioma patients undergoing initial endoscopic endonasal surgery between December 2012 and March 2021 in our center were retrospectively reviewed, and injury types of the HPA were categorized according to intraoperative endoscopic observation. Included patients were randomly divided into a training group and a validation group. Nomograms were established based on the results of multivariate logistic analysis. The predictive performance of the nomograms was evaluated in the training and validation groups.ResultsA total of 183 patients with craniopharyngioma were enrolled, and seven injury types of the HPA were summarized. Relative to intact HPA, exclusive hypothalamus injury significantly increased the risk of anterior (OR, 194.174; 95% CI, 21.311–1769.253; p < 0.001) and posterior pituitary dysfunction (OR, 31.393; 95% CI, 6.319–155.964; p < 0.001) 1 year after surgery, while exclusively sacrificing stalk infiltrated by tumors did not significantly increase the risk of anterior (OR, 5.633; 95% CI, 0.753–42.133; p = 0.092) and posterior pituitary dysfunction (OR, 1.580; 95% CI, 0.257–9.707; p = 0.621) 1 year after surgery. In the training group, the AUCs of nomograms predicting anterior and posterior pituitary dysfunction 1 year after surgery were 0.921 and 0.885, respectively, compared with 0.921 and 0.880 in the validation group.ConclusionsIntact hypothalamus structure is critical in maintaining pituitary function. Moreover, our preliminary study suggests that the pituitary stalk infiltrated by craniopharyngioma could be sacrificed to achieve radical resection, without substantially rendering significantly worse endocrinological efficiency 1 year after surgery. The user-friendly nomograms can be used to predict hypopituitarism 1 year after surgery.

Background: Neurocognitive (NC) impairment in people with HIV (PWH) is associated with important adverse outcomes, but no markers exist to predict long-term NC decline. We evaluated depressed mood and markers of persistent inflammation, oxidative stress and altered amyloid processing (all common in PWH) as predictors of NC worsening over 12 years. Methods: PWH were enrolled and followed longitudinally in the CNS HIV Antiretroviral Effects Research (CHARTER) study at six US sites. At entry we quantified biomarkers in blood of inflammation: (interleukin-6 [IL-6], C-reactive protein [CRP], monocyte chemoattractant protein type 1 [MCP-1], D-dimer, soluble sCD14 (sCD14), soluble tumor necrosis factor receptor – type II [sTNFR-II], neopterin, and soluble CD40 ligand [sCD40L], oxidative stress (protein carbonyls, 8-oxo-2'-deoxyguanosine [8-oxo-dG]) and altered amyloid processing [amyloid beta (Aβ)-42, soluble amyloid precursor protein-α (sAPPα)] using commercial immunoassays. The Beck Depression Inventory-II (BDI-II) assessed depressed mood at entry. NC decline over 12 years was evaluated using the published and validated summary (global) regression-based change score (sRBCS). A factor analysis reduced dimensionality of the biomarkers. Univariable and multiple regression models tested the relationship between baseline predictors and the outcome of neurocognitive decline. Results: Participants were 191 PWH, 37 (19.4%) women, 46.6% African American, 43.5% non-Hispanic white, 8.83% Hispanic, 15.7% white, 1.6% other; at study entry mean (SD) age 43.6 (8.06) years, estimated duration of HIV infection (median, IQR) 9.82 [4.44, 14.5] years, nadir CD4 104/μL (19,205), current CD4 568/μL (356, 817), and 80.1% had plasma HIV RNA

Neutron imaging is an effective nondestructive testing (NDT) technique widely applied to detect structural defects and the enrichment of nuclear fuel elements due to its high penetration and nuclide-sensitive properties. Since the fuel element pellet is sealed in the cladding, the transmission imaging result is a superposition of the two parts. Therefore, the attenuation of neutrons by the cladding is interference that must be considered in the enrichment analysis. It is necessary to extract and separate cladding and pellets using an edge extraction method. However, the low neutron cross-section of the cladding material (e.g., aluminum and zirconium) leads to poor grayscale contrast at the cladding edge in the imaging result, and the intensity of the cladding edge is significantly lower than that of the pellet edge. In addition, affected by the noise from the imaging environment, the boundaries of targets are further blurred, making edge detection more challenging. Traditional detection algorithms extract the weak edges of cladding incompletely, and the results are discontinuous, with obvious edge breaks and missing areas. This paper proposes a method to extract edges in neutron images based on phase congruency (PC). This study utilized the classical perceptual field model to improve contrast at weak edges. The enriched edge map was generated using our PC model from six directions, allowing more weak edges to be detected accurately. The non-maximum suppression ensured precise localization and avoided edge breaks. Furthermore, the edge results were optimized by eliminating noise through morphological operations. The experimental results demonstrate that the proposed method effectively detects the weak edges of the cladding, is superior in accuracy and integrity to traditional detection, and is able to obtain stable and reliable results with different materials of neutron images. The edge integrity improved by 64.1%, and the edge localization accuracy reached 94.3%. The extracted edge information is useful in the next stage of the high-precision enrichment analysis.

Currently, coaxial electrohydrodynamic jet (CE-Jet) printing is used as a promising technique for the alternative fabrication of drop-on-demand micro- and nanoscale structures without using a template. Therefore, this paper presents numerical simulation of the DoD CE-Jet process based on a phase field model. Titanium lead zirconate (PZT) and silicone oil were used to verify the numerical simulation and the experiments. The optimized working parameters (i.e., inner liquid flow velocity 150 m/s, pulse voltage 8.0 kV, external fluid velocity 250 m/s, print height 16 cm) were used to control the stability of the CE-Jet, avoiding the bulging effect during experimental study. Consequently, different sized microdroplets with a minimum diameter of ~5.5 µm were directly printed after the removal of the outer solution. The model is considered the easiest to implement and is powerful for the application of flexible printed electronics in advanced manufacturing technology.

In this paper, we propose a method for cracking the key parameters of an electro-optic self-feedback temporal optical phase encryption system and experimentally demonstrate the feasibility of the scheme. By scanning a tunable dispersion compensation (TDC) module at the receiver, the time delay signature (TDS) of an encrypted signal can be exposed, making it possible to extract other key parameters of the system and reconstruct a decryption setup. The TDS characteristics for three typical modulation formats are investigated, revealing that while such an encryption system is secure against power detection attack, there is a risk of TDS leakage. The findings can guide the design of advanced optical encryption schemes with TDS suppression for security enhancement.

Metamaterials with actively tunable functionalities are highly desirable for applications of advanced optoelectronic devices. In this paper, we theoretically present a metamaterial with diversified functionalities by availing of the phase transition characteristics of vanadium dioxide (VO2) in terahertz frequency regions. The research results demonstrate that the function of the designed metamaterial can be switched from giant circular dichroism (CD) to a reflecting broadband half-wave plate (HWP) and a quarter-wave plate (QWP). When VO2 is in the isolating state, the metamaterial exhibits a quite distinct transmission efficiency for circularly polarized lights, thus resulting in a maximum CD value ~0.97 at the resonant frequency. When VO2 is operating in the metallic state, the metamaterial performs like a broadband HWP, in which the nearly perfect linear polarization conversion can be achieved at the frequency range from 3 to 7 THz. Moreover, the structure can play a role of a high-efficiency QWP that can simultaneously convert the incident linear polarized light to left-handed and right-handed circularly polarized light. The calculated ellipticity indicates a good polarization conversion at the frequency of 2.4 THz and 7.4 THz, respectively. The physical mechanism of the discussed features and effects can be explained by exploring the electric field distributions. Furthermore, the structural parameters also exert great influences for achieving giant CD and HWP as well as QWP. The proposed metamaterial may offer a new approach for designing metamaterial devices with multi-functions in THz regions.

In wireless distributed computing systems, worker nodes connect to a master node wirelessly and perform large-scale computational tasks that are parallelized across them. However, the common phenomenon of straggling (i.e., worker nodes often experience unpredictable slowdown during computation and communication) and packet losses due to severe channel fading can significantly increase the latency of computational tasks. In this paper, we consider a heterogeneous, wireless, distributed computing system performing large-scale matrix multiplications which form the core of many machine learning applications. To address the aforementioned challenges, we first propose a random linear network coding (RLNC) approach that leverages the linearity of matrix multiplication, which has many salient properties, including ratelessness, maximum straggler tolerance and near-ideal load balancing. We then theoretically demonstrate that its latency converges to the optimum in probability when the matrix size grows to infinity. To combat the high encoding and decoding overheads of the RLNC approach, we further propose a practical variation based on batched sparse (BATS) code. The effectiveness of our proposed approaches is demonstrated by numerical simulations.

Polyketides are an important class of structurally diverse natural products derived from a precursor molecule consisting of a chain of alternating ketone and methylene groups. These compounds have attracted the worldwide attention of pharmaceutical researchers since they are endowed with a wide array of biological properties. As one of the most common filamentous fungi in nature, Aspergillus spp. is well known as an excellent producer of polyketide compounds with therapeutic potential. By extensive literature search and data analysis, this review comprehensively summarizes Aspergillus-derived polyketides for the first time, regarding their occurrences, chemical structures and bioactivities as well as biosynthetic logics.

High-resolution neutron radiography provides novel and stirring opportunities to investigate the structures of light elements encased by heavy elements. For this study, a series of Gd2O2S:Tb, F particles were prepared using a high-temperature solid phase method and then used as a scintillation screen. Upon reaching 293 nm excitation, a bright green emission originated from the Tb3+ luminescence center. The level of F doping affected the fluorescence intensity. When the F doping level was 8 mol%, the fluorescence intensity was at its highest. The absolute quantum yield of the synthesized particles reached as high as 77.21%. Gd2O2S:Tb, F particles were applied to the scintillation screen, showing a resolution on the neutron radiograph as high as 12 μm. These results suggest that the highly efficient Gd2O2S:Tb, F particles are promising scintillators for the purposes of cold neutron radiography.

Background: It remains unclear what B cell and humoral responses are mounted by chronic kidney disease (CKD) patients in response to recombinant and inactivated SARS-CoV-2 vaccines. In this study, we aimed to explore the cellular and humoral responses, and the safety of recombinant and inactivated SARS-CoV-2 vaccines in CKD patients. Methods: 79 CKD and 420 non-CKD individuals, who completed a full course of vaccination, were enrolled in the study. Adverse events (AEs) were collected via a questionnaire. Cellular and humoral responses were detected at 1, 3, and 6 months, including IgG antibody against the receptor-binding domain (RBD) of the SARS-CoV-2 spike protein (anti-RBD-IgG), neutralizing antibodies (NAbs), the positive rate of NAbs and anti-RBD-IgG, RBD-atypical memory B cells (MBCs) (CD3 − CD19 + RBD + CD21 − CD27−), RBD-activated MBCs (CD3 − CD19 + RBD + CD21 − CD27+), RBD-resting MBCs (CD3 − CD19 + RBD + CD21 + CD27+), and RBD-intermediate MBCs (CD3 − CD19 + RBD + CD21 + CD27−). Results: We found no differences in the positivity rates of NAbs (70.89% vs. 79.49%, p = 0.212) and anti-RBD IgG (72.15% vs. 83.33%, p = 0.092) between the CKD and control groups. A total of 22 CKD individuals completed the full follow-up (1, 3, and 6 months). Significant and sustained declines were found at 3 months in anti-RBD IgG (26.64 BAU/mL vs. 9.08 BAU/mL, p < 0.001) and NAbs (161.60 IU/mL vs. 68.45 IU/mL p < 0.001), and at 6 months in anti-RBD IgG (9.08 BAU/mL vs. 5.40 BAU/mL, p = 0.064) and NAbs (68.45 IU/mL vs. 51.03 IU/mL, p = 0.001). Significant differences were identified in MBC subgroups between CKD patients and healthy controls, including RBD-specific atypical MBCs (60.5% vs. 17.9%, p < 0.001), RBD-specific activated MBCs (36.3% vs. 14.8%, p < 0.001), RBD-specific intermediate MBCs (1.24% vs. 42.6%, p < 0.001), and resting MBCs (1.34% vs. 22.4%, p < 0.001). Most AEs in CKD patients were mild (grade 1 and 2) and self-limiting. One patient with CKD presented with a recurrence of nephrotic syndrome after vaccination. Conclusions: The recombinant and inactivated SARS-CoV-2 vaccine was well-tolerated and showed a good response in the CKD cohort. Our study also revealed differences in MBC subtypes after SARS-CoV-2 vaccination between CKD patients and healthy controls.

This paper presents an implementation scheme and experimental evaluation of a high-linearity closed-loop capacitive accelerometer based on ring-diode capacitance detection. By deducing the capacitance detection model of the ring-diode considering the influence of the diode, the existing theoretical model error of the ring-diode is corrected and a closed-loop scheme of reusing the detection electrode and the control electrode of the MEMS die is designed to apply this detection scheme to the parallel-plate accelerometer, which only has three independent electrodes. After analyzing the non-linear problems in the existing closed-loop control schemes, a theoretically absolute linear closed-loop control scheme is proposed, and an integrated closed-loop accelerometer is realized by combining the closed-loop diode detection. The experimental results of the ring-diode detection model are in agreement with the theoretical formula. The non-linearity of the accelerometer within ±1 g after the closed-loop is 130 ppm, compared with 1500 ppm when the open-loop is used.

Abstract With the continuous decreasing in sizes of functional materials and devices, people are being asked to perform a flexible, accurate, in-situ and non-thermal welding of nanowires at the nanoscale. In this work, a well deliberated procedure including three typical stages: sharpening, hooking and welding, was carried out in sequence by in-situ TEM to realize the high demand welding of SiOx nanowire to macroporous carbon film. It was found that the brittle SiOx nanowire was non-thermally softened under energetic e-beam irradiation, and the flexibility and accuracy of welding could be achieved by adjusting the beam spot size, irradiation location and irradiation time. It was demonstrated that the nanocurvature effect of SiOx nanowire and the ultra-fast energy deposition effect induced by energetic e-beam irradiation dominated the diffusion, evaporation and plastic flow of atoms and the resulting nanowire re-shaping and nanowelding processes. In contrast, the traditional knock-on mechanism and e-beam heating effect are inadequate to explain these phenomena. Therefore, such a study is crucial not only to the flexible technical controlling but also to the profound fundamental understanding of energetic e-beam-induced nanowire re-shaping and nanowelding.

Qin et al. show the involvement of Kindlin-2 in osteocyte mechanotransduction. They show Kindlin-2 mediates skeletal responses to mechanical stimulation through Smad 2/3 mediated inhibition of Sclerostin.

Abstract Glutamine:fructose-6-phosphate aminotransferase (GFAT) and phosphofructokinase (PFK) are enzymes related to chitin metabolism. RNA interference (RNAi) technology was used to explore the role of these two enzyme genes in chitin metabolism. In this study, we found that GFAT and PFK were highly expressed in the wing bud of Nilaparvata lugens and were increased significantly during molting. RNAi of GFAT and PFK both caused severe malformation rates and mortality rates in N. lugens. GFAT inhibition also downregulated GFAT, GNPNA, PGM1, PGM2, UAP, CHS1, CHS1a, CHS1b, Cht1-10, and ENGase. PFK inhibition significantly downregulated GFAT; upregulated GNPNA, PGM2, UAP, Cht2-4, Cht6-7 at 48 h and then downregulated them at 72 h; upregulated Cht5, Cht8, Cht10, and ENGase; downregulated Cht9 at 48 h and then upregulated it at 72 h; and upregulated CHS1, CHS1a, and CHS1b. In conclusion, GFAT and PFK regulated chitin degradation and remodeling by regulating the expression of genes related to the chitin metabolism and exert opposite effects on these genes. These results may be beneficial to develop new chitin synthesis inhibitors for pest control.

Abstract Background The association between long-term cannabis use and balance disturbances has not been investigated in people living with HIV (PWH). We hypothesized that long-term cannabis use in PWH might be associated with more deleterious effects on balance than in HIV seronegative individuals due to potential neurotoxic interactions between HIV and cannabis. Methods Three thousand six-hundred and forty-eight participants with and without HIV completed an interviewer-administered timeline follow-back assessment to assess lifetime days and quantity of cannabis use and other cannabis use characteristics. A structured clinical interview was used to collect any history of balance disturbance. Comparisons between HIV+ vs the HIV− groups and moderate-severe vs. no or minimal imbalance in participant characteristics (demographics, cannabis use, medication currently used, and neurological disease) were performed using Student t tests for continuous variables and Fisher’s exact test for binary and categorical variables. Multivariate logistic regression was applied to determine the interaction effect of total quantity of cannabis use with HIV status on balance disturbance. Age, gender, cDSPN symptoms, gait ataxia, opioid medications, and sedatives were included as covariates in the adjusted model after variable selection. The effect sizes are presented as Cohen’s d or odds ratios. Results On average, participants were 45.4 years old (SD = 11 years), primarily male (77.7%), and non-Hispanic white (48.1%). A majority of participants were HIV+ (79.1%). Four hundred thirty (11.9%) of the participants reported balance disturbances within the past 10 years. PWH were more likely to have balance disturbances than demographically matched HIV-uninfected participants (odds ratio [OR] 2.66, 95% CI 1.91–3.7). Participants with moderate-severe balance disturbances did not differ from those with no or minimal imbalance in the proportion who had ever used cannabis (73.8% vs. 74.4%; p = 0.8) (OR 1.03, 95% CI 0.80–1.32) neither did they have a higher total amount of cannabis use (4871 vs. 4648; p = 0.3) (Cohen’s d 0.11, 95% CI 0.01–0.14). In the HIV− population, those with balance disturbances reported more total amount of cannabis use as compared to those with normal balance (11316 vs 4154; p = 0.007). In the HIV+ population on the other hand, there was no significant association (4379 vs 4773; p = 0.6). Conclusions We found unexpectedly that while long-term cannabis use in HIV− individuals was associated with more severe balance disturbances, there were no associations in HIV+ individuals. This suggests that cannabis use in HIV is safe with respect to balance disturbances. Given that HIV is related to persistent inflammation despite virologic suppression on antiretroviral therapy, future mechanistic studies are needed to determine whether HIV-associated inflammation contributes to the higher prevalence of balance disturbance in HIV+ individuals and whether cannabinoids have anti-inflammatory effects that mitigate HIV-associated balance disturbance.

Background: Trichinellosis is a serious food-borne parasitic zoonosis, thus finding high quality antigens is the key to serodiagnosis of trichinosis. This article reports the characterization and sensitivity of four recombinant proteins expressed by four genes (Wn10, Zh68, T668, and Wm5) from different developmental stages of Trichinella spiralis for the diagnosis of trichinellosis in mice. Methods: This study was conducted in Jilin University and National Institute of Parasitic Diseases of Chinese Center for Disease Control and Prevention in 2017-2018. The structures and functions of the proteins encoded by four genes were predicted by bioinformatics analysis. The four genes were cloned and expressed, and the recombinant proteins were purified. Anti-Trichinella IgM and IgG antibodies in the sera of mice infected with T. spiralis from 1-45 d post-infection (dpi) were evaluated by ELISA. Results: The optimal antigen epitopes of four proteins (P1, P2, P3, and P4) encoded by the four genes from T- and B-cells were predicted, and four purified recombinant proteins (r-P1, r-P2, r-P3, and r-P4) were successfully produced. For IgM, the antibody levels detected by the four recombinant antigens were approximately equal to the cut-off value. Anti-Trichinella IgG antibodies were first detected by r-P1 at 8 dpi, followed by r-P2, r-P3, and r-P4 at 10 dpi, 14 dpi, and 16 dpi, respectively, and the antibody levels remained high until 45 dpi. Conclusion: The recombinant antigens r-P1, r-P2, r-P3, and r-P4 could be antigens that react with antibodies, they showed high sensitivity in the detection of anti-Trichinella IgG antibodies in mice. Among these proteins, r-P1 may be a candidate antigen for the detection of anti-Trichinella IgG antibodies in the early infection phase and exhibited the best sensitivity among the antigens.

Background: Probiotic and low fermentable oligosaccharide, disaccharide, monosaccharide, and polyol (FODMAP) diet are two commonly used management approaches for patients with irritable bowel syndrome (IBS). We aimed to evaluate the most effective combinations and components among different probiotics or low FODMAP diet through component network meta-analysis (NMA).Methods: We searched Embase, Ovid Medline, and Web of Science from inception to 21 January 2021. Randomized controlled trials (RCTs) examining the efficacy of probiotics and low FODMAP diet for IBS were included, with placebo, sham diet, or conventional treatments as controls. Binary outcomes were compared among treatments using the relative ratio (RR). A minimally contextualized framework recommended by the GRADE group was used to evaluate the certainty of evidence. The primary efficacy outcome was the relief of global IBS symptoms, and the secondary efficacy outcome was the reduction in IBS symptom scores or abdominal pain scores.Key Results: We included 76 RCTs (n = 8058) after screening 1940 articles. Eight RCTs were classified as low risk of bias. Standard network meta-analysis (NMA) showed that Lactobacillus (RR 1.74, 95% CI 1.22–2.48) and Bifidobacterium (RR 1.76, 95% CI 1.01–3.07) were the most effective for the primary efficacy outcome (high certainty evidence); component NMA showed that Bacillus (RR 5.67, 95% CI 1.88 to 17.08, p = 0.002) and Lactobacillus (RR 1.42, 95% CI 1.07 to 1.91, p = 0.017) were among the most effective components. The results of standard NMA and CNMA analysis of the improvement of overall IBS symptom scores or abdominal pain scores were consistent with this finding.Conclusion:Lactobacillus was the most effective component for the relief of IBS symptoms; Bifidobacterium and Bacillus were possibly effective and need further verification.Systematic Review Registration: website, identifier registration number.

ObjectiveNaturally occurring in-frame deletion is a unique type of genetic variations, causing the loss of one or more amino acids of proteins. A number of in-frame deletion variants in an epilepsy-associated gene SCN1A, encoding voltage gated sodium channel alpha unit 1.1 (Nav1.1), have been reported in public database. In contrast to the missense and truncation variants, the in-frame deletions in SCN1A remains largely uncharacterized.MethodsWe summarized the basic information of forty-four SCN1A in-frame deletion variants and performed further analysis on six variants identified in our cases with epilepsy. Mutants of the six in-frame deletions and one truncating variant used as comparison were generated and co-transfected with beta-1 and -2 subunits in tsA201 cells, followed by patch clamp recordings.ResultsReviewing all the in-frame deletions showed that they spread over the entire Nav1.1 protein, without obvious “hot spots.” The dominant type (54%) was single residue loss. There was no obvious relationship between the length or locations of deletions and their clinical phenotypes. The six in-frame deletions were two single residue deletions (p.M400del and p.I1772del), one microdeletion (p.S128_F130del) and three macrodeletions (p.T303_R322del, p.T160_Y202del, and p.V1335_V1428del). They scatter and affect different functional domains, including transmembrane helices, pore region, and P-loop. Electrophysiological recordings revealed no measurable sodium current in all of the six mutants. In contrast, the truncating mutant p.M1619Ifs*7 that loses a long stretch of peptides retains partial function.SignificanceThe complete loss-of-function in these shortened, abnormal mutants indicates that Nav1.1 protein is a highly accurate structure, and many of the residues have no redundancy to ion conductance. In-frame deletions caused particularly deleterious effect on protein function possibly due to the disruption of ordered residues.

Summary: Deep brain stimulation (DBS) entails neurosurgery to implant electrodes in specific brain structures to modulate the behavior of a particular neural circuit. DBS is best known for treating advanced Parkinson disease and can potentially be applicable to other motor and even cognitive dysfunctions. Here, we describe a detailed protocol allowing for electrode preparation, surgical procedures, stimulation delivery, and field potential recordings in both anesthetized and behaving mice, and the benefit evaluation of DBS at the fimbria-fornix by using a fear conditioning test.For complete details on the use and execution of this protocol, please refer to Hao et al. Trichinellosis caused by Trichinella spiralis is a worldwide food-borne parasitic zoonosis. Several approaches have been performed to control T. spiralis infection, including veterinary vaccines, which contribute to improving animal health and increasing public health by preventing the transmission of trichinellosis from animals to humans. In the past several decades, many vaccine studies have been performed in effort to control T. spiralis infection by reducing the muscle larvae and adult worms burden. Various candidate antigens, selected from excretory-secretory (ES) products and different functional proteins involved in the process of establishing infection have been investigated in rodent or swine models to explore their protective effect against T. spiralis infection. Moreover, different types of vaccines have been developed to improve the protective effect against T. spiralis infection in rodent or swine models, such as live attenuated vaccines, natural antigen vaccines, recombinant protein vaccines, DNA vaccines, and synthesized epitope vaccines. However, few studies of T. spiralis vaccines have been performed in pigs, and future research should focus on exploring the protective effect of different types of vaccines in swine models. Here, we present an overview of the strategies for the development of effective T. spiralis vaccines and summarize the factors of influencing the effectiveness of vaccines. We also discuss several propositions in improving the effectiveness of vaccines and may provide a route map for future T. spiralis vaccines development.

Background: Emergence agitation (EA) is a common problem often observed in children after sevoflurane anesthesia, which can be prevented by dexmedetomidine and alfentanil. This study aims to compare the effectiveness of dexmedetomidine alone and with different doses of alfentanil in preventing EA in children under sevoflurane anesthesia.Materials and Methods: In a double-blind trial, 80 children (ASA I or II, 3–7 years old) undergoing tonsillectomy alone and adenotonsillectomy with sevoflurane anesthesia were randomly assigned into four groups: the control group, dexmedetomidine (DEX) group, dexmedetomidine plus 10 μg/kg alfentanil group (DEX + Alf1), and dexmedetomidine plus 20 μg/kg alfentanil group (DEX + ALf2). The incidence of EA was assessed with the Aono’s scale, and the severity of EA was evaluated with the Pediatric Anesthesia Emergence Delirium (PAED) scale. The time of tracheal extubation and time of wake were recorded. Postoperative pain and complications such as nausea and vomiting, cough, laryngospasm, and bradycardia were recorded.Results: The incidence of EA was 50% in the control group, 25% in the DEX group, and 5% in the DEX + Alf1 group, and it never happened in the DEX + Alf2 group. The Aono’s scale, the PAED scale, and the FLACC scale in the control group and the DEX group were significantly more than those in the DEX + Alf1 group and the DEX + Alf2 group after the tracheal extubation (p < 0.05). The time of tracheal extubation of the control group and the DEX group were significantly shorter than those in the DEX + Alf1 group and the DEX + Alf2 group (p < 0.05). The awakening time of the DEX + Alf2 group is significantly longer than those in other groups (p < 0.05). The case of postoperative nausea and vomiting in the DEX + Alf1 group was fewer than those in the other groups (p < 0.05). And, the cases of cough and laryngospasm and bronchospasm in the DEX + Alf1 group and the DEX + Alf2 group were significantly less than those in the control group and the DEX group after the tracheal extubation (p < 0.05).Conclusion: The combined administration of alfentanil and dexmedetomidine can reduce EA in children undergoing tonsillectomy alone and adenotonsillectomy with sevoflurane anesthesia. Dexmedetomidine plus 10 μg/kg alfentanil seems to be more appropriate than other dose combinations as it reduced EA and postoperative nausea and vomiting but did not prolong the time to awake.

Abstract Future projection of temperature extremes in the “Earth's three poles” (the Arctic, Antarctica, and Third Pole‐Tibetan Plateau [TP]) is of importance to risk assessment and policymaking owing to the high sensitivity to climate change in these regions. In this study, future projections of four extreme temperature indices were constructed after the application of a bias correction method in models of Phase 6 of the Coupled Model Intercomparison Project (CMIP6). The reduced intensification of temperature extremes in the Earth's three poles if warming can be limited to 1.5°C instead of 2°C above the pre‐industrial level was examined. Results showed that all the extreme temperature indices show significant increasing trends under both the SSP2–4.5 and SSP5–8.5 scenarios over the Earth's three poles (SSP: Shared Socioeconomic Pathway). For the coldest night (TNn), warmest night (TNx), and warmest day (TXx), the greatest increase by the end of the 21st century under SSP5–8.5 occurs in the Arctic, followed by the TP and finally Antarctica. For the coldest day (TXn), the greatest increase occurs in the Arctic, followed by Antarctica and finally the TP. If global warming can be limited to 1.5°C rather than 2°C, the intensification of TNn, TNx, TXn, and TXx in the Arctic (Antarctica/TP) under SSP5–8.5 is projected to reduce by 66% (21.7%/44.26%), 50.31% (54.79%/60.52%), 71.58% (12.91%/65.81%), and 41.73% (81.3%/57.34%), respectively, and the results are similar for SSP2–4.5. Therefore, keeping a lower warming target is essential for reducing the risk of extreme events in the Earth's three poles.

Sr0.7Bi0.2TiO3 (SBT) by increasing the proportion and size of polar nano-region. Meanwhile, the BDS remains a high level with x ≤ 0.38 attributed to the addition of KBT with a large band gap. As a result, the 0.62SBT-0.38KBT exhibits a high energy storage density of 2.21 J/cm3 with high η of 91.4% at 220 kV/cm and superior temperature stability (−55 ∼ 150 °C), frequency stability (10 ∼ 500 Hz) and fatigue resistance (105 cycles). Moreover, high pulsed discharge energy density (1.81 J/cm3), high power density (49.5 MW/cm3) and great thermal stability (20 ∼ 160 °C) are achieved in 0.62SBT-0.38KBT. Based on these excellent properties, the 0.62SBT-0.38KBT are suitable for pulsed power systems. This work provides a novel strategy and systematic study for improving energy storage properties of SBT.

Xiaolei Liu, Yayan Feng, et al. report an annotated draft reference genome of the non-encapsulated Trichinella pseudospiralis, and perform comparative multi-omics analyses with encapsulated T. spiralis. Overexpression of TP12446, a key excretory/secretory gene, in a mouse C2C12 skeletal muscle cell line could induce inhibition of myotube formation and differentiation. These results provide insight into the mechanism of nurse cell formation and immunoevasion, as well as the identification of potential pharmacological and diagnostic targets for trichinellosis.

Abstract. Background. Natural killer (NK) cells play a critical role in suppressing human immunodeficiency virus-1 (HIV-1) infection, but knowledge on whether and how NK cells affect immune reconstitution in HIV-1-infected individuals who receive antiretroviral therapy (ART) is limited. Methods. We performed a case-control study with 35 healthy individuals and 66 HIV-1-infected patients including 32 immunological non-responders (INRs) with poor CD4+ T-cell recovery (500 cells/μL after 4 years of ART). NK cell phenotype, receptor repertoire, and early activation in INRs and IRs were investigated by flow cytometry. Results. A significantly higher proportion of CD56dimCD16dim/- NK cells was observed in INRs than IRs before ART and after 4 years of ART. The number of CD56dimCD16dim/- NK cells was inversely correlated with CD4+ T-cell counts in INRs before ART (r = –0.344, P = 0.050). The more CD69-expressing NK cells there were, the lower the CD4+ T-cell counts and ΔCD4, and these correlations were observed in INRs after ART (r = –0.416, P = 0.019; r = –0.509, P = 0.003, respectively). Additionally, CD69-expressing CD56dimCD16dim/- NK cells were more abundant in INRs than those in IRs (P = 0.018) after ART, both of which had an inverse association trend towards significance with CD4+ T-cell counts. Industrial Cyber-Physical Systems (ICPS) connect intelligent manufacturing equipment equipped with sensors, wireless and RFID communication technologies through data interaction, which makes the interior of the factory, even between factories, become a whole. However, intelligent factories will suffer information leakage and equipment damage when being attacked by ICPS intrusion. Therefore, the network security of ICPS cannot be ignored, and researchers have conducted in-depth research on network intrusion detection for ICPS. Though machine learning and deep learning methods are often used for network intrusion detection, the problem of data imbalance can cause the model to pay attention to the misclassification cost of the prevalent class, but ignore that of the rare class, which seriously affects the classification performance of network intrusion detection models. Considering the powerful generative power of the diffusion model, we propose an ICPS Intrusion Detection system based on the Diffusion model (IDD). Firstly, data corresponding to the rare class is generated by the diffusion model, which makes the training dataset of different classes balanced. Then, the improved BiLSTM classification network is trained on the balanced training set. Extensive experiments are conducted to show that the IDD method outperforms the existing baseline method on several available datasets.

The meta-analysis was performed to assess the prevalence of T. spiralis in domestic pigs in China. The potential studies from seven databases (Pubmed, Web of science, Scopus, Google Scholar, CNKI, Wanfang, CBM) were searched. I2, Cochran’s Q statistic and the funnel plot and Egger’s test were used to assess heterogeneity and publication bias, respectively. In this study, a total of 179 articles were captured in the initially screened. Of these, we finally obtained 39 significant articles (including 43 studies involving in 551,097 pigs) for the final analysis. We calculated using a random-effects model, and we found the overall infection rate was 0.04 (95% CI 0.03–0.06). The highest prevalence region was Guangxi. The funnel plot and Egger’s test showed no publication bias in our meta-analysis. In addition, this high heterogeneity index was suggestive of potential variations which could be due to regions, quality scores, detection methods, publication years, or samplings. These results indicated that T. spiralis were still prevalent in some areas in China. This highlights the need for an increased focus on implementing affordable, appropriate control programs to reduce economic losses and T. spiralis infection in domestic pigs in China.

Starch is a major component of crop grains, and its content affects food quality and taste. Tartary buckwheat is a traditional pseudo-cereal used in food as well as medicine. Starch content, granule morphology, and physicochemical properties have been extensively studied in Tartary buckwheat. However, the complex regulatory network related to its starch biosynthesis needs to be elucidated. Here, we performed RNA-seq analyses using seven Tartary buckwheat varieties differing in starch content and combined the RNA-seq data with starch content by weighted correlation network analysis (WGCNA). As a result, 10,873 differentially expressed genes (DEGs) were identified and were functionally clustered to six hierarchical clusters. Fifteen starch biosynthesis genes had higher expression level in seeds. Four trait-specific modules and 3131 hub genes were identified by WGCNA, with the lightcyan and brown modules positively correlated with starch-related traits. Furthermore, two potential gene regulatory networks were proposed, including the co-expression of FtNAC70, FtPUL, and FtGBSS1-3 in the lightcyan module and FtbHLH5, C3H, FtBE2, FtISA3, FtSS3-5, and FtSS1 in the brown. All the above genes were preferentially expressed in seeds, further suggesting their role in seed starch biosynthesis. These results provide crucial guidance for further research on starch biosynthesis and its regulatory network in Tartary buckwheat.

To investigate the effect of moderate thermal modification (TM) on wood properties, American alder (Alnus rubra) wood was treated at 140 °C for 4 h, 8 h and 13 h, the physical and mechanical properties, dimensional stability and color changes of wood were compared and studied. The results showed that the absolute dry density of moderate-TM wood decreased significantly with time except for the 4 h treatment. Moderate TM can significantly reduce the residual stress of wood up to 90.3%. There were no significant differences in MOR and MOE between most moderate TM wood and the control group; moderate TM decreased the moisture absorption and water up-taking of wood significantly; compared to the control group, the swelling of TM wood for 13 h decreased by 24.2% and 16.0% in the tangential and radial direction, respectively, showing a significant improvement in dimensional stability. There were almost no color changes even when wood endured 140 °C and 13 h TM. The moderate TM at 140 °C for 13 h can efficiently improve wood dimensional stability and retains the natural color of wood while causing almost no damage to the wood’s mechanical strength.

Excessive zinc (Zn) is toxic to plants, but the effect of zinc-stressed plants on herbivorous insects is still unclear. Hence, we assessed the effect of zinc-stressed rice plants on its feeding pest, Nilaparvata lugens. The soil–rice–N. lugens system was treated with Zn2+ solution. Sugar content in rice was measured, and the reproduction and trehalose metabolism in N. lugens were assessed. The trehalase activity in rice significantly decreased at 100 mg·kg−1 Zn2+, and the trehalose content increased. The glucose and starch content increased at higher Zn2+ concentrations. The fecundity and trehalose content of N. lugens decreased after feeding on zinc-stressed rice, and the glucose content in the high Zn2+ group was significantly higher than that in the low Zn2+ group. In addition, the soluble trehalase activity of N. lugens significantly decreased under the 125 mg·kg−1 treatment, while the activity of membrane-bound trehalase significantly increased under the 150 mg·kg−1 treatment. Quantitative RT-PCR indicated significantly lower expressions of NlTre1-1, NlTre2, and NITps after Zn treatment. In conclusion, Zn2+ treatment significantly altered the sugar content in rice plants; it also decreased the fecundity of N. lugens, which may be mediated by alterations in trehalose metabolism.

Spodoptera frugiperda is one of the world’s major agricultural pests and it occurs in many countries around the world. In Lepidoptera, the peritrophic membrane in the midgut protects the midgut epithelial cells and facilitates the digestion and absorption of food. Its main components are chitin and protein. Chitin synthase (CHS), as the last enzyme in the chitin biosynthesis pathway, is very important. Here, the expression of chitin synthase B (CHSB) gene identified from midgut was inhibited by RNA interference to explore its function in the metabolism of trehalose and chitin of S. frugiperda larvae. The results were as follows: SfCHSB was highly expressed in actively feeding larvae. Second, knockdown of SfCHSB resulted in decreased expression of most genes involved in chitin metabolism, including chitinase, and abnormal phenotypes of S. frugiperda were observed. In addition, membrane-bound trehalase activity and glucose content increased, and trehalose content decreased at 24 h after dsSfCHSB injection. Trehalase activities increased significantly and the glycogen content decreased at 48 h after dsSfCHSB injection. Finally, the activity of chitinase decreased and the content of chitin increased significantly. Our results indicate that SfCHSB expression is specific, SfCHSB has a large role in regulating chitin metabolism, and it has broad application prospects in the biocontrol of S. frugiperda.

This paper presents a 10-bit successive approximation register analog-to-digital converter with energy-efficient low-complexity switching scheme, automatic ON/OFF comparator and automatic ON/OFF SAR logic for biomedical applications. The energy-efficient switching scheme achieves an average digital-to-analog converter switching energy of 63.56 CVref2, achieving a reduction of 95.34% compared with the conventional capacitor switching scheme for CDACs. With the switching scheme, the ADC can lower the dependency on the accuracy of Vcm and complexity of DAC control logic and DAC driver circuit. Moreover, dynamic circuits and automatic ON/OFF technology are used to reduce power consumption of comparator and SAR logic. The prototype is designed and fabricated in a 180 nm CMOS with a core size of 500 μm × 300 μm (0.15 mm2). It consumes 7.6 nW at 1 kS/s sampling rate and 1.8-V supply with an achieved signal-to-noise-and distortion ratio of 45.90 dB and a resulting figure of merit of 51.7 fJ/conv.-step.

Purpose: To compare the safety and efficacy of percutaneous paricalcitol injection with intravenously administered paricalcitol in treating parathyroid hyperplasia in patients with secondary hyperparathyroidism (SHPT). Methods: This study was approved by the Ethics Committee of our institution. We retrospectively collected data on patients who received percutaneous paricalcitol injection (24 patients) and intravenously administered paricalcitol (22 patients) based on their intact parathyroid hormone (iPTH) level. Serum iPTH, calcium, phosphorus, and the volume of the parathyroid gland were measured at several indicated time points after treatment, and adverse events associated with the two treatments were evaluated. Results: After 6 months of follow-up, we found that patients from the percutaneous injection group had significantly decreased levels of iPTH (from 1887.81 ± 726.81 pg/mL to 631.06 ± 393.06 pg/mL), phosphate (from 1.94 ± 0.36 mmol/L to 1.71 ± 0.34 mmol/L), and volume of the parathyroid gland (from 0.87 ± 0.50 cm3 to 0.60 ± 0.36 cm3), with relief from ostealgia within 48–72 h. In the intravenously administered group, the levels of iPTH decreased from 686.87 ± 260.44 pg/mL to 388.47 ± 167.36 pg/mL; while there was no significant change in phosphate levels, the volume of the parathyroid gland and ostealgia relief were observed at the end of follow-up. The serum calcium level did not significantly change, and no severe complications were observed in both groups. In vitro fluorescence-activated single cell sorting (FACS) analysis indicated that paricalcitol induced parathyroid cell apoptosis in a dose-dependent manner. Conclusions: Percutaneous paricalcitol injection is a selective treatment for SHPT in ESRD.

A DAC switching scheme that combines energy efficiency and resolution reconfigurability is proposed. Compared with the conventional switching scheme, the proposed scheme achieves 93.8%, 96.1%, and 97.3% switching energy saving in 8-bit, 9-bit, and 10-bit modes, respectively. Based on the proposed switching scheme, an 8–10-bit resolution-reconfigurable SAR ADC for biosensor applications is designed. The ADC consists of resolution-reconfigurable binary-weighted capacitive DAC, a two-stage full dynamic comparator, sampling switch, and the resolution-control SAR logic. Simulated in 180 nm CMOS process and 100 kS/s sampling rate, the ADC achieves the 46.80/53.89/60.14 dB signal-to-noise and distortion ratio (SNDR), the 55.22/62.51/73.09 dB spurious-free dynamic range (SFDR) and the 0.81/0.91/1.01 μW power consumption in 8/9/10-bit mode, respectively.

BackgroundResearch suggests that physical activity (PA) has both acute and chronic beneficial effects on cognitive function in laboratory settings and under supervised conditions. Mobile health technologies make it possible to reliably measure PA and cognition in free-living environments, thus increasing generalizability and reach. Research is needed to determine whether the benefits of PA on cognitive function extend from the laboratory to real-world contexts. ObjectiveThis observational study aims to examine the association between daily fluctuations in PA and cognitive performance using mobile health technologies in free-living environments. MethodsA total of 90 adults (mean age 59, SD 6.3 years; 65/90, 72% men) with various comorbidities (eg, cardiovascular risk and HIV) and different levels of baseline cognition (ranging from cognitively normal to impaired) completed ecological momentary cognitive tests (EMCTs) on a smartphone twice daily while wearing an accelerometer to capture PA levels for 14 days. Linear mixed-effects models examined the daily associations of PA with executive function and verbal learning EMCTs. Moderation analyses investigated whether the relationship between daily PA and daily performance on EMCTs changed as a function of baseline cognition, cardiovascular risk, and functional status (independent vs dependent). ResultsDays with greater PA were associated with better (faster) performance on an executive function EMCT after covariate adjustment (estimate −0.013; β=−.16; P=.04). Moderation analyses (estimate 0.048; β=.58; P=.001) indicated that days with greater PA were associated with better (faster) executive function performance in individuals who were functionally dependent (effect size −0.53; P

Aquatic animals cannot synthesize carotenoids, thus they must come from diet or from symbionts. Previous studies have found that orange-muscle abalones are rich in carotenoids, but the effects of different diets on the accumulation of carotenoids are unknown. In this study, the effects of macroalgae (Gracilaria lemaneiformis), fresh and dried kelp (Laminaria japonica) on the contents of the predominant carotenoids, including zeaxanthin, β-carotene, and fucoxanthin, between the common and orange-muscle mutant of Haliotis gigantea were compared through a one-year culture experiment. Our study confirmed that carotenoids in abalones mainly come from diets, and the carotenoid types in the muscles were similar to their diets. We also found that feeding on G.lemaneiformis played an important role in maintaining a stable carotenoid content over time, especially zeaxanthin in H.gigantea. Our data also provided that abalones had a good growth performance under the feeding conditions of G.lemaneiformis. Objective: An overwhelming majority of the genetic variants associated with genetic disorders are missense. The association between the nature of substitution and the functional alteration, which is critical in determining the pathogenicity of variants, remains largely unknown. With a novel missense variant (E1623A) identified from two epileptic cases, which occurs in the extracellular S3-S4 loop of Nav1.1, we studied functional changes of all latent mutations at residue E1623, aiming to understand the relationship between substitution nature and functional alteration.Methods: Six latent mutants with amino acid substitutions at E1623 were generated, followed by measurements of their electrophysiological alterations. Different computational analyses were used to parameterize the residue alterations.Results: Structural modeling indicated that the E1623 was located in the peripheral region far from the central pore, and contributed to the tight turn of the S3-S4 loop. The E1623 residue exhibited low functional tolerance to the substitutions with the most remarkable loss-of-function found in E1623A, including reduced current density, less steady-state availability of activation and inactivation, and slower recovery from fast inactivation. Correlation analysis between electrophysiological parameters and the parameterized physicochemical properties of different residues suggested that hydrophilicity of side-chain at E1623 might be a crucial contributor for voltage-dependent kinetics. However, none of the established algorithms on the physicochemical variations of residues could well predict changes in the channel conductance property indicated by peak current density.Significance: The results established the important role of the extracellular S3-S4 loop in Nav1.1 channel gating and proposed a possible effect of local conformational loop flexibility on channel conductance and kinetics. Site-specific knowledge of protein will be a fundamental task for future bioinformatics.

Psoriasis is a chronic and refractory inflammatory and autoimmune-mediated cutaneous disease affecting approximately 2%–3% of the global population. Most of the current therapies could relieve symptoms rapidly, while the side effects cannot be negligible. Hence, it is urgent to explore much safer and more effective treatments. In the current work, we evaluated the potential beneficial effect of Punica granatum peel polysaccharides (PPPs) in an imiquimod-elicited psoriasis-like mouse model and unraveled their mechanism of action. Firstly, PPPs were isolated from P. granatum peels, and then the molecular weight was determined and monosaccharide analysis was performed. The results revealed that PPPs significantly ameliorated psoriasis-like skin lesions and reduced the Psoriasis Area and Severity Index (PASI) scores and transepidermal water loss (TEWL). PPPs also attenuated the expressions of CD3 and Ki67 in psoriasis-like mouse skin and suppressed the serum or skin levels of pro-inflammatory cytokines, such as tumor necrosis factor alpha (TNF-α), interleukin 6 (IL-6), IL-1β, IL-8, IL-17, and IL-23. Moreover, PPPs were able to upregulate the mRNA and protein expressions of aquaporin-3 (AQP3) and filaggrin (FLG) in the skin of mice. In addition, PPPs inhibited the NF-κB and STAT3 signaling pathways. Overall, these results indicated that PPPs ameliorated the symptoms of psoriasis through inhibition of the inflammatory cytokines by suppressing the NF-κB and STAT3 signaling pathways and improved skin barrier protection via enhancing AQP3 and FLG. These observations potentially contribute to providing theoretical and experimental evidence for the clinical application of PPPs for psoriasis.

Objectives: In breast radiotherapy, the proximity of the target to sensitive structures together with the uncertainty introduced by respiratory movement, make this treatment one of the most studied to increase its effectiveness. Dosimetric and physical variables play an important role and the study of their correlation and impact on treatment is fundamental. This retrospective study aims to highlight the dosimetric differences of 2 different clinical data sets of patients receiving left-sided breast irradiation in free breathing (FB) or breath hold (BH). Methods: A total of 155 left breast carcinoma patients receiving whole-breast irradiation in FB (73 patients) and BH (82 patients) were enrolled in this study. The dosimetric parameters of the target, heart, left and right lung and right breast were evaluated and compared, and possible correlations were studied in both groups. Limited water resources and rapid socioeconomic development pose new challenges to watershed water resource management. By integrating the perspectives of stakeholders and decision-makers, this study aims to identify cases and approaches to achieve sustainable water resources management. It improves and expands the experience of previous project research. The comparative evaluation provides an analytical basis to verify the importance of stakeholder participation in water policy interactions. The results show that if an effective demand management policy is not implemented, the Guanzhong area will not meet water demand in the future. Through the combination of water-saving policies, water transfer projects and other measures, the available water resources will continue into the future. Optimizing management measures, improving the ecological environment, and encouraging stakeholder participation will help change this situation, although supply-side limitations and future uncertainties likely cause unsustainable water. Therefore, decision-makers should pay attention to the application potential of water-saving and other measures to reduce dependence on external water sources. In addition, the three sustainable development decision-making principles identified in this paper can promote the fairness and stability of water policy.

New observations and updated models now suggest terrestrial ecosystems are net sink of atmospheric mercury (Hg), and the critical constrained process to identify the strengths of terrestrial sink is whether the large amount of Hg stored in vegetation originates from the soil as well as from the atmosphere. In this study, field open top chambers (OTCs) experiments reveal that rice plant can assimilate gaseous elemental mercury (GEM, Hg0) from the atmosphere through stomata, and Hg concentrations in rice leaves, upper and bottom stalks and grains increased with Hg0 levels in air, showing significantly quadratic linear relationships. Coupling field stable isotope soil amendment experiments, atmospheric source of Hg in rice plant is quantified with more than 90% of Hg accumulation in rice aboveground biomass from air and approximately 80% of rice root Hg from soil. Furthermore, elevated atmospheric carbon dioxide (CO2) exposure led to lower Hg concentration in rice tissues through reduction stomatal conductance of rice leaf, and subsequently impact the capacity of Hg storage in rice aboveground parts from the atmosphere. The findings from experiments provide a foundation for future quantification of atmospheric sink of crops in local and larger scales and comprehensive evaluation atmosphere - terrestrial processes and exposure risks in the global Hg cycling.

The pandemic caused by the severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) has been basically under control in China since March 2020, but the import of domestic SARS-CoV-2 has begun to increase. This study reported the first case of asymptomatic SARS-CoV-2 infection imported from Spain into Sichuan Province, China, on March 11, 2020. The infected male had a body temperature of 37.5°C, normal blood oxygen saturation levels, and a computed tomography (CT) examination showed that his lungs had no shadows. However, a throat swab from the subject tested positive for SARS-CoV-2 using qPCR assay. In this study, we conducted transcriptome sequencing on respiratory throat swabs from the subject and found that the dominant SARS-CoV-2 sequence (Gene Bank ID: MW301121) was a spike protein D614G mutant strain, which is currently popular throughout world. We downloaded and analyzed SARS-CoV-2 sequences collected from cases in China and Spain for comparison and tracing purposes. After March 11, 2020, the Chinese domestic clade was naturally divided into the imported SARS-CoV-2 D614G mutant strain and evolutionarily-related similar sequences and that of sequences collected in the original Wuhan area. The sequence reported in this study was located on a small branch, far from the evolution of Wuhan sequences. As expected, the identified sequence was closely related to the evolution of the SARS-CoV-2 D614G mutant strain circulating in Spain.

Abstract Background Helicobacter pylori (H. pylori) infection is a serious human health threat. The empiric H. pylori treatment paradigm guided by traditional testing technologies has led to antibiotic resistance. Here, we improved the qPCR method to provide technical support for precision H. pylori diagnosis and treatment. Methods Two pairs of primers and probes targeting the glmM gene were designed to detect H. pylori, and a multiplex qPCR method was established for virulence factor detection. Then, a rapid urease test (RUT), culturing and qPCR were performed on 141 specimens collected from Xinqiao Hospital of China in 2017 to evaluate the qPCR detection capability. Finally, the H. pylori infectious amount and virulence genes were detected by qPCR. Results 1. The improved qPCR method which used two pairs of primers had a higher detection rate (100%) and better accuracy (p = 0.000), compared with the qPCR using a pair of primers. A data identifier (DID) is an essential tag or label in all kinds of databases—particularly those related to integrated computational materials engineering (ICME), inheritable integrated intelligent manufacturing (I3M), and the Industrial Internet of Things. With the guidance and quick acceleration of the development of advanced materials, as envisioned by official documents worldwide, more investigations are required to construct relative numerical standards for material informatics. This work proposes a universal DID format consisting of a set of build chains, which aligns with the classical form of identifier in both international and national standards, such as ISO/IEC 29168-1:2000, GB/T 27766–2011, GA/T 543.2–2011, GM/T 0006–2012, GJB 7365–2011, SL 325–2014, SL 607–2018, WS 363.2–2011, and QX/T 39–2005. Each build chain is made up of capital letters and numbers, with no symbols. Moreover, the total length of each build chain is not restricted, which follows the formation of the Universal Coded Character Set in the international standard of ISO/IEC 10646. Based on these rules, the proposed DID is flexible and convenient for extending and sharing in and between various cloud-based platforms. Accordingly, classical two-dimensional (2D) codes, including the Hanxin Code, Lots Perception Matrix (LP) Code, Quick Response (QR) code, Grid Matrix (GM) code, and Data Matrix (DM) Code, can be constructed and precisely recognized and/or decoded by either smart phones or specific machines. By utilizing these 2D codes as the fingerprints of a set of data linked with its cloud-based platforms, progress and updates in the composition–processing–structure–property–performance workflow process can be tracked spontaneously, paving a path to accelerate the discovery and manufacture of advanced materials and enhance research productivity, performance, and collaboration.

In this work, Ti6Al4V alloy was treated by a duplex treatment of double glow plasma surface molybdenizing-laser surface texturing (LST) aiming to realize surface strengthening and manipulate its tribological behavior. The double glow plasma surface alloying (DGPSA) with molybdenum (Mo) was firstly conducted on the Ti6Al4V alloy and followed by LST to prepare round dimple-shape texture with a density of 5%, 7%, 11%. The results showed that Mo and Mo/Ti solid solution phases were detected in the Mo-layer by X-ray diffraction (XRD), the received Mo-layer with a thickness of about 13 μm was uniform and compact. Double glow plasma surface molybdenizing significantly improved the surface hardness and elastic modulus of Ti6Al4V alloy by nanoindentation test. The gradient distributed Mo-layer was tightly adhered to the Ti6Al4V alloy matrix through scratching and indentation tests. Duplex treated Ti6Al4V alloy with a 5% texture density suggested better wear resistance in comparison to the untreated Ti6Al4V alloy, which was confirmed by a 48.7% lower specific wear rate than that of Ti6Al4V alloy. Taking advantages of DGPSA and LST, duplex treatment of double glow plasma surface molybdenizing-laser surface texturing was established to create on the working surface of Ti6Al4V alloy with promisingly improved tribological behavior.

The nonlocal crystal plasticity finite element (CPFE) simulations and extreme value statistics were combined to study the effects of microstructure on the high cycle fatigue (HCF) behavior of dual-phase Ti alloy. A modified Armstrong-Frederick nonlinear kinematic hardening equation accounting for cyclic softening effect was employed in the crystal plasticity constitutive model. Three-dimensional equiaxed microstructure models and two-dimensional duplex microstructure models with real lamellar structure were generated based on Voronoi method, serving as statistical volume elements (SVEs). The effects of morphological and crystallographic features, including grain size, grain orientation, phase volume fraction and lamellae width, on the fatigue performance were investigated. By simulating multiple SVEs, extreme value distributions of the driving force for fatigue crack formation were predicted using Fatemi-Socie (FS) parameter as a fatigue indicator parameter (FIP). Meanwhile, whether the extreme values of geometrically necessary dislocation (GND) density can be taken as a FIP was discussed with compared to FS FIP. The GND density, related to local stress and strain gradient, has considerable potential as a FIP to estimate the fatigue performance of titanium alloys. Based on simulated results, it is suggested that microstructure with small grain size, low volume fraction of primary α grains, and thinner α lamellae width have the lowest probability of crack formation during HCF.

In recent years, as for the frequent occurrence of failure accidents of high-grade steel pipelines caused by girth weld cracking, great attention has been drawn to the quality of girth weld and defect repair of high-grade steel pipelines. In terms of the types of girth weld defects of high-grade steel pipelines, the recommended treatment methods for girth weld defects in domestic and foreign repair standards for high-grade steel pipes were introduced, and the research results of the existing welding and non-welding repair technologies were systematically combed. As for welding repair technology, Type B sleeve is the preferred repair technology for repairing high-grade steel pipe girth weld defects. At present, the full-automatic welding technology for horizontal straight welds and circumferential fillet welds for open tee, guard plate and Type B sleeve has basically met the conditions for popularization and application, and is expected to greatly improve the welding efficiency and welding quality. Non-welding repair technology divides into three types: composite material, epoxy sleeve and steel lining composite material repair technology. A series of verification tests were conducted abroad to repair the defects (50% incomplete fusion) of X42 pipeline girth welds with composite materials and steel lining composite materials. Compared with composite materials, steel-lined composite materials perform better in axial tensile load test. In China, steel epoxy sleeve was used to reinforce the girth weld, which was inspected and evaluated to be unnecessary to repair but has defects, and the composite steel lining repair technology was designed based on the composite material and epoxy sleeve repair technology. Finally, the solution and key research directions were put forward to solve the problem of girth weld defect repair of high-grade pipelines, and supposed to provide technical reserves for the safe operation of China–Russia Eastern Gas Pipeline Project and similar high-grade pipelines.

Abstract Background It is known that Fas ligand (FasL) is involved in the development of intervertebral disc degeneration (IDD). A recent study reported that lncRNA MAGI2-AS3 up-regulated the expression of FasL to promote breast cancer. Therefore, we investigated the roles that lncRNA MAGI2-AS3 might play in IDD. Methods A total of 66 IDD patients (IDD group) and 58 healthy volunteers (Control group) were recruited in this study. Quantitative real-time PCR (qRT-PCR) and western blot were used to investigate gene expression levels. Cell transfections were carried out to analyze gene interactions. The diagnostic value of lncRNA MAGI2-AS3 for IDD was assessed by ROC curve analysis. Results The expression levels of plasma lncRNA MAGI2-AS3 were lower in IDD patients compared to that in the control group. Down-regulation of lncRNA MAGI2-AS3 effectively distinguished IDD patients from the control group. The expression levels of plasma lncRNA MAGI2-AS3 were significantly increased after the treatments. Over-expression of lncRNA MAGI2-AS3 inhibited the expression of FasL, while the silencing of lncRNA MAGI2-AS3 promoted the expression of FasL in nucleus pulposus (NP) cells. Conclusions Therefore, lncRNA MAGI2-AS3 is down-regulated in IDD and participates in the regulation of FasL expression in nucleus pulposus (NP) cells.
