Objectives. This study aimed to develop an initial deep-learning (DL) model based on computerized tomography (CT) scans for diagnosing lumbar spinal stenosis. Summary of Background Data. Magnetic resonance imaging is commonly used for diagnosing lumbar spinal stenosis due to its high soft tissue resolution, but CT is more portable, cost-effective, and has wider regional coverage. Using DL models to improve the accuracy of CT diagnosis can effectively reduce missed diagnoses and misdiagnoses in clinical practice. Materials and Methods. Axial lumbar spine CT scans obtained between March 2022 and September 2023 were included. The data set was divided into a training set (62.3%), a validation set (22.9%), and a control set (14.8%). All data were labeled by two spine surgeons using the widely accepted grading system for lumbar spinal stenosis. The training and validation sets were used to annotate the regions of interest by the two spine surgeons. First, a region of interest detection model and a convolutional neural network classifier were trained using the training set. After training, the model was preliminarily evaluated using a validation set. Finally, the performance of the DL model was evaluated on the control set, and a comparison was made between the model and the classification performance of specialists with varying levels of experience. Results. The central stenosis grading accuracies of DL Model Version 1 and DL Model Version 2 were 88% and 83%, respectively. The lateral recess grading accuracies of DL Model Version 1 and DL Model Version 2 were 75% and 71%, respectively. Conclusions. Our preliminarily developed DL system for assessing the degree of lumbar spinal stenosis in CT, including the central canal and lateral recess, has shown similar accuracy to experienced specialist physicians. The thermodynamic stable phase of cerium metal in the intermediate pressure regime (5.0-13.0 GPa) is studied in detail by the newly developed local-density approximation (LDA) + Gutzwiller method, which can include the strong correlation effect among the 4f electrons in cerium metal properly. Our numerical results show that the α" phase, which has the distorted body-centered-tetragonal structure, is the thermodynamic stable phase in the intermediate pressure regime and all the other phases including the α' phase (α-U structure), α phase (fcc structure), and bct phases are either metastable or unstable. The article presents a study that assesses the significance of the purification and characterization of levansucrases (LS) on the synthesis of levan and fructoologisaccharides. It notes that the intra and extracellular activities of LS produced by Bacillus amyloliquefaciens were promoted by supplementing the sucrose medium with peptone and yeast as nitrogen sources. It further concludes that the intra and extracellular LSs have demonstrated slight differences in their catalytic properties.

Studies revealed that navigation systems that provided intraoperative assistance might improve pedicle screw insertion accuracy, and also implied that different systems provided different pedicle screw insertion accuracy. A systematic review and meta-analysis was conducted to focus on the pedicle screw insertion accuracy with or without the assistance of image-guided system, and the variance among the different navigation systems. Comparative studies were searched on pedicle screw insertion accuracy between conventional and navigated method, and among different navigation systems. A total of 43 papers, including 28 clinical, 14 cadaveric and 1 model studies, were included in the current study. For clinical articles, there were 3 randomized clinical trials, 4 prospective comparative studies and 21 retrospective comparative studies. The incidence of pedicle violation among computer tomography-based navigation method group was statistically significantly less than that observed among the conventional group (OR 95% CI, in vivo: 0.32-0.60; in vitro: 0.24-0.75 P < 0.01). Two-dimensional fluoroscopy-based navigation system (OR 95% CI, in vivo: 0.27-0.48; in vitro: 0.43-0.88 P < 0.01) and three-dimension fluoroscopy-based navigation system (OR 95% CI, in vivo: 0.09-0.38; in vitro: 0.09-0.36 P < 0.01) also obtained significant reduced screw deviation rate over traditional methods. Between navigated approaches, statistically insignificant individual and pooled RR values were observed for all in vivo subgroups. Pooled estimate of in vitro studies show that computer tomography-based and three-dimension fluoroscopy-based navigation system provided more accurate pedicle screw insertion over two-dimension fluoroscopy-based navigation system. Our review showed that navigation provided a higher accuracy in the placement of pedicle screws compared with conventional methods. The superiority of navigation systems was obvious when they were applied to abnormal spinal structure. Although no strong in vivo evidence has detected significantly different pedicle screw placement accuracy among the three major navigation systems, meta-analysis revealed the variance in pedicle screw insertion accuracy with different navigation methods. On March 23, 2010, we successfully treated a boy with synpolydactyly who had a total of 31 fingers and toes. Although there was bone syndactyly both the hands of the boy, one-step correction of four extremities was successful , this operation lasted 5 h and 20 min and intraoperative bleeding was about 50 ml. US maize yield has increased eight-fold in the past 80 years, with half of the gain attributed to selection by breeders. During this time, changes in maize leaf angle and size have altered plant architecture, allowing more efficient light capture as planting density has increased. Through a genome-wide association study (GWAS) of the maize nested association mapping panel, we determined the genetic basis of important leaf architecture traits and identified some of the key genes. Overall, we demonstrate that the genetic architecture of the leaf traits is dominated by small effects, with little epistasis, environmental interaction or pleiotropy. In particular, GWAS results show that variations at the liguleless genes have contributed to more upright leaves. These results demonstrate that the use of GWAS with specially designed mapping populations is effective in uncovering the basis of key agronomic traits. [ABSTRACT FROM AUTHOR]

Abstract: Background: Diabetic patients are characterised by poorer prognosis and more cardiovascular complications compared with non-diabetic patients, which may be due to metabolic abnormalities and atherosclerotic plaque characteristics. Methods: Patients with unstable angina pectoris were enrolled in the study and divided into diabetes mellitus (DM) (patients, n =25; plaques, n =42) and non-DM (patients, n =53; plaques, n =65) groups according to their DM history. Optical coherence tomography (OCT) examinations were performed on all patients, and images were analysed by two independent investigators. Fibrous cap thickness was measured at the thinnest point of each plaque. The presence of plaque disruption, dissection, erosion, thrombosis and calcification were also noted. Results: Calcified plaques in the DM group were significantly greater than those in the non-DM group (42.9% vs. 23.1%; p =0.03). Thin-cap fibroatheroma (TCFA) were detected, and no significant difference was found in the frequencies (42.9% vs. 52.3%; p =0.34) and fibrous cap thickness (57.08±6.20μm vs. 56.11±9.23μm, p =0.74) between the DM and non-DM groups. Thrombus and plaque erosion were similar in the two groups, but the frequency of dissection in the DM group was greater than that in the non-DM group (21.4% vs. 7.7%, p =0.04). The high sensitivity C-reactive protein between the two groups was similar (0.44±0.20mg/dl vs. 0.46±0.15mg/dl, p =0.83). Conclusion: Higher calcification and dissection were detected in diabetic patients with unstable angina pectoris, and the difference in coronary plaque characteristics can explain the difference in clinical prognoses between DM and non-DM patients. Transcriptional regulation of the gene encoding brain-derived neurotrophic factor (BDNF) has been widely studied. However, an understanding of mechanisms modifying chromatin, events that are essential for controlling transcription, is rudimentary. We focused on two activation-dependent regions of the Bdnf gene physically linked to known transcription sites for exons 1 and 4. Using chromatin immunoprecipitation assays, we determined that N-methyl- d-aspartate (NMDA) receptor activation derepressed promoters 1 and 4-mediated transcription. This derepression correlated with reduced occupancy by histone deacetylase 1 and methyl cytosine-binding protein 2 of each promoter region near known transcription start sites in cultured hippocampal neurons. These changes did not occur at all sites upstream of transcription initiation. Taken together, these findings suggest that histone and other DNA-binding proteins are involved in remodeling of chromatin at some, but not all sites, within Bdnf promoters 1 and 4 and are associated with NMDA receptor-dependent increases in transcription. Giardia species are flagellated parasites of vertebrates and belong to the diplomonads, most of which have two nuclei. These organisms were classified among the earliest branching eukaryotes on the basis of small subunit rDNA sequences and their lack of many canonical organelles. However, some of these organelles have subsequently been identified in rudimentary form, such as Golgi that become apparent during encystation. One of the "missing" organelles has been the nucleolus, the site of rRNA synthesis, since it was not identified in earlier ultrastructural studies. In the current study, we visualized in-vitro-grown Giardia lamblia trophozoites by transmission electron microscopy and laser scanning confocal microscopy. We found that each of the two nuclei contains a single small and deeply stained granular nucleolus, thus demonstrating that Giardia does indeed have nucleoli. [ABSTRACT FROM AUTHOR]

Abstract: The lipopolysaccharide (LPS)-Toll-like receptor 4 (TLR4) signaling pathway in alveolar epithelial cells plays an important role in many pathologic processes such as acute lung injury (ALI). The single immunoglobulin IL-1 receptor-related protein (SIGIRR) is an inhibitor of LPS-TLR4 signaling, but its expression and function in alveolar epithelial cells are still unknown. In this study, we examined the expression of SIGIRR in normal human lung tissue using immunohistochemistry, reverse transcription-PCR (RT-PCR) and Western blot and found that SIGIRR was expressed in alveolar epithelial cells. Treatment of an alveolar epithelial cell line, A549, with LPS and we observed a downregulation of SIGIRR mRNA, which returned to normal levels 24h after LPS exposure. A549 cells were then transfected with a SIGIRR eukaryotic expression vector to over-express SIGIRR or, as a control, with an empty vector. Following LPS exposure, the transcriptional activity of NF-κB was measured using a dual-luciferase reporter assay system, and the concentration of IL-1β, TNF-α and IL-6 was determined by ELISA, and cell proliferation was measured by MTT. In A549 cells that over-expressed SIGIRR, LPS treatment resulted in a significant decrease in the transcriptional activity of NF-κB and cell growth inhibition ratio, as well as lower levels of secreted IL-1β, TNF-α and IL-6. In conclusion, SIGIRR in A549 cells inhibits the transcriptional activity of NF-κB and reduces the amount cytokines produced, protecting these cells from acute LPS-induced damage. Maize domestication is one of the greatest feats of artificial selection and evolution, wherein a weedy plant in Central Mexico was converted through human-mediated selection into the most productive crop in the world. In fact, the changes were so astounding that it took much of the last century to identify modern maize's true ancestor. Through modern genetic studies, the molecular basis of this evolution is being unraveled. Maize's new morphology and adaptation to diverse environments required selection at thou- sands of loci, and we are beginning to understand the magnitude and rates of these genetic changes. Most of the known major genes have experienced strong selection, but only small regions sur- rounding the selected genes exhibit substantially reduced genetic diversity. Here, we report the discovery of a large region on chromosome 10 involved in adaptation or domestication that has been the target of strong selection during maize domestication. Unlike previously described regions in the maize genome, 1.1 Mb and >15 genes lost genetic diversity during selection at this region. To determine the epigenetic events associated with NMDA receptor-mediated activation of brain-derived neurotrophic factor gene ( Bdnf) promoter 1 by hippocampal neurons in culture, we screened 12 loci across 4.5 kb of genomic DNA 5′ of the transcription start site (TSS) of rat Bdnf for specific changes in histone modification and transcription factor binding following NMDA receptor stimulation. Chromatin immunoprecipitation (ChIP) assays showed that NMDA receptor stimulation produced a durable, time-dependent decrease in histone H3 at lysine 9 dimethylation (H3K9me2), within 3 h after NMDA treatment across multiple loci. Concomitant increases in H3K4me2 and H3K9/14 acetylation (H3AcK9/14) were associated with transcriptional activation, but occurred at fewer sites within the promoter. The decrease in H3K9me2 was associated with release of HDAC1, MBD1, MeCP2, and REST from specific locations within promoter 1, although with different kinetics. In addition, occupancy of sites proximal to and distal to the TSS by the transcription factors NF-κB, CREB-binding protein (CBP), and cAMP-response element-binding protein were correlated with increased occupancy of RNA polymerase II at two loci proximal to the TSS following NMDA receptor stimulation. These temporal changes in promoter occupancy could occur thousands of base pairs 5′ of the TSS, suggesting a mechanism that produces waves of Bdnf transcription. [ABSTRACT FROM AUTHOR]

AbstractMitochondrial DNA (mtDNA) damage has been hypothesized to be responsible for aging and various neurological diseases. Abnormalities in 8-oxoguanine DNA glycosylase (OGG1) function can promote DNA oxidative damage, especially in the mitochondria. Here we report changes in the expression of OGG1 targeting to the nucleus, cytosol, and mitochondria in both accelerated senescence mice (SAM-P/8) and normal counterpart SAM-R/1 mice during brain aging. Our results showed that mRNA and protein levels of OGG1, especially OGG1 targeting to mitochondria, and the expression level of cytochrome coxidase subunit III (COX III) in the brain of both SAM-P/8 mice and SAM-R/1 mice, decreased with age. However, such an age-dependent decrease in SAM-P/8 mice was larger than that in normal SAM-R/1 mice. These findings support the concept that down-regulation of OGG1, especially mitochondrial OGG1(mtOGG1) in SAM-P/8 mice, may promote brain aging by its effect on imbalance in the mtDNA damage repair systems, which leads to accumulation of mtDNA damage and oxidative phosphorylation-related protein dysfunction. Application of the pressure controlled isothermal heating vertical deposition method to the fabrication of colloidal photonic crystals is systematically investigated in this paper. The fabricated samples are characterized by scanning electron microscope and transmission spectrum. High-quality samples with large transmissions in the pass bands and the sharp band edges are obtained and the optimum growth condition is determined. For the best sample, the transmission in the pass bands approaches 0.9 while that in the band gap reaches 0.1. More importantly, the maximum differential transmission as high as 0.1/nm is achieved. In addition, it is found that the number of stacking layers does not increase linearly with concentration of PS spheres in a solution, and a gradual saturation occurs when the concentration of PS spheres exceeds 1.5 wt.%. The uniformity of the fabricated samples is examined by transmission measurements on areas with different sizes. Finally, the tolerance of the fabricated samples to baking was studied. • Beijing haze is strongly related to enhanced atmospheric oxidizing capacity. • Two regimes separated by [O 3 ] of 80 μg m−3: AOC-sufficient and -deficient regimes. • Increasing oxidation capacity considerably weakens the emission mitigation effort. Although strict mitigation measures have been implemented since 2013 in Beijing-Tianjin-Hebei (BTH), China, air pollution still frequently occurs. Observations reveal that during pollution episodes in autumn, fine particulate matter (PM 2.5) concentrations have not decreased, and particularly, ozone (O 3) concentrations have increased remarkably from 2013 to 2015 in Beijing. Additionally, a concurrence of O 3 and particulate pollution with high secondary aerosol contributions has been observed frequently, indicating high atmospheric oxidizing capacity (AOC) during particulate pollution. The WRF-Chem model simulations show elevated O 3 concentrations and high fractions of oxygenated secondary aerosols (OSA) in PM 2.5 (0.53–0.73) during the severe pollution period. During daytime there exhibits an AOC-sufficient regime with the persistently high OSA fraction and an AOC-deficient regime with varied OSA fractions, separated by the O 3 level of 80 μg m−3. Scallop-derived plasmalogen (sPlas) has both anti-oxidative and anti-inflammation activities, but its efficacy has not been investigated in ischemic stroke models where oxidative stress, inflammation, and neurovascular unit (NVU) damage accelerates pathophysiological progression. Therefore, in the present study, we aimed to assess the neuroprotective effects of sPlas in ischemic stroke by using a transient middle cerebral artery occlusion (tMCAO) mouse model. After the pretreatment of vehicle or sPlas (10 mg/kg/day) for 14 days, adult male mice were subjected to tMCAO for 60 min, then continuously treated with vehicle or sPlas during reperfusion and for an additional 5 days. Background: Concerning the judgments bias and cue utilization in basketball athletes, previous shot anticipation tasks were hard to examine in regards to whether the experts' judgement bias relies more on the cue of the player's body or the ball trajectory. Methods: Four types of body–ball cues shots were employed: IN–IN, IN–OUT, OUT–IN, and OUT–OUT. Four temporal stages (i.e., shooting, rising, high point, and falling) were divided during a shot. Forty-two participants predicted the fate of the ball after watching the shot videos. Results: The results suggested that for the shooting, rising, and high point phase, compared to the non-athletes, the experts provided superior predictions for IN–IN condition and OUT–IN condition but fewer accurate predictions for IN–OUT condition and OUT–OUT condition. Moreover, a higher bias toward predicting the shots as "in" for the athletes than the non-athletes under early temporal conditions was confirmed. Conclusions: These findings strengthen the idea that the IN cues from both body information and ball trajectory could elicit the experts' judgement bias for made shots and then influence their response, thus rendered two distinct (e.g., impeding and facilitating) effects for the incongruent body–ball cues, respectively. Background: The establishment of new methods to detect human genetic variations, such as single nucleotide polymorphisms, is of importance in hereditary disease diagnosis and pharmacogenomics. Several single nucleotide polymorphism genotyping technologies have been presented in recent years. However, techniques which would allow accurate, fast and cheap allelic determination and multiple single nucleotide polymorphism detection in parallel are still in great need. Methods: Here, we present a new genotyping technique based on gap ligase chain reaction and a fluorescent polystyrene microsphere measurement platform. We chose the human polymorphism rs3730386 as our candidate to establish this method. Probes for gap ligase chain reaction were designed to recognize the target alleles sensitively and specifically and to produce templates for amplifying the correspondent target fragments used in the following hybridization. The genotypes were finally determined by a hybridization process based on the Luminex fluorescent polystyrene microspheres measurement platform. Results: This method was successfully applied for the detection of selected single nucleotide polymorphisms with high sensitivity and specificity. The genotypes were validated by DNA sequencing. Conclusions: The new genotyping method benefited from the high sensitivity and specificity of gap ligase chain reaction and the detection platform of Luminex. Gliosis is a pathological hallmark of posttraumatic epileptic foci, but little is known about these reactive astrocytes beyond their high glial fibrillary acidic protein (GFAP) expression. Using diolistic labeling, we show that cortical astrocytes lost their nonoverlapping domain organization in three mouse models of epilepsy: posttraumatic injury, genetic susceptibility, and systemic kainate exposure. Neighboring astrocytes in epileptic mice showed a 10-fold increase in overlap of processes. Concurrently, spine density was increased on dendrites of excitatory neurons. Suppression of seizures by the common antiepileptic, valproate, reduced the overlap of astrocytic processes. Astrocytic domain organization was also preserved in APP transgenic mice expressing a mutant variant of human amyloid precursor protein despite a marked upregulation of GFAP. Our data suggest that loss of astrocytic domains was not universally associated with gliosis, but restricted to seizure pathologies. Brain-derived neurotrophic factor (BDNF), via activation of TrkB receptors, mediates vital physiological functions in the brain, ranging from neuronal survival to synaptic plasticity, and has been implicated in the pathophysiology of neurodegenerative disorders. Although transcriptional regulation of the BDNF gene (Bdnf) has been extensively studied, much remains to be understood. We discovered a sequence within Bdnf promoter 4 that binds the basic helix-loop-helix protein BHLHB2 and is a target for BHLHB2-mediated transcriptional repression. NMDA receptor activation de-repressed promoter 4-mediated transcription and correlated with reduced occupancy of the promoter by BHLHB2 in cultured hippocampal neurons. Bhlhb2 gene-/-mice showed increased hippocampal exon 4-specific Bdnf mRNA levels compared with +/+ littermates under basal and activity-dependent conditions. Bhlhb2 knock-out mice also showed increased status epilepticus susceptibility, suggesting that BHLHB2 alters neuronal excitability. Adiponectin is a peptide hormone secreted exclusively by adipocytes, and obesity is an established risk factor for breast cancer. We have, thus, evaluated the associations of anthropometric measures of adiposity and adiponectin with the development of breast cancer in a case–control study. Questionnaire information, anthropometric measures, and blood samples were taken before treatment from 244 incident cases with breast cancer, including 141 premenopausal and 103 postmenopausal cases, and 244 controls admitted for health examination at the Tri-Service General Hospital, Taipei between 2004 and 2005. Plasma levels of adiponectin were measured by RIA. The relationship between anthropometric measures of adiposity and breast cancer risk was modified by menopausal status, with a significant increase in risk observed in postmenopausal but not premenopausal women. Moreover, a fairly robust inverse association of adiponectin with the risk was observed only in postmenopausal women (adjusted odds ratio (OR), 0.55; 95% confidence interval (CI), 0.23–0.97), but not in premenopausal women. Additionally, the plasma adiponectin levels tended to be inversely associated with estrogen receptor (ER)-positive (adjusted OR, 0.53; 95% CI, 0.27–0.98) but not ER-negative breast tumors. Furthermore, the associations of adiponectin with breast cancer risk overall and by menopausal and ER status remained after adjustment for obesity indices. These results suggest that adiponectin may have an independent role in breast carcinogenesis, particularly in the postmenopausal and ER-positive breast cancer risk. [ABSTRACT FROM AUTHOR]

Cortical spreading depression (CSD) is a self-propagating wave of cellular depolarization that has been implicated in migraine and in progressive neuronal injury after stroke and head trauma. Using two-photon microscopic NADH imaging and oxygen sensor microelectrodes in live mouse cortex, we find that CSD is linked to severe hypoxia and marked neuronal swelling that can last up to several minutes. Changes in dendritic structures and loss of spines during CSD are comparable to those during anoxic depolarization. Increasing O2 availability shortens the duration of CSD and improves local redox state. Our results indicate that tissue hypoxia associated with CSD is caused by a transient increase in O2 demand exceeding vascular O2 supply. [ABSTRACT FROM AUTHOR]

Background: The study of microRNAs (miRNAs) is attracting great considerations. Recent studies revealed that miRNAs play as important regulators of gene expression and some even as cancer players or inhibitors. Many studies try to discover new miRNAs and reveal the miRNA expression profile in cancer using a SAGE-based total RNA clone method. However, the data processing of this method is labor-intensive with several different biological databases and more than ten data processing steps involved. Results: With miRAS, miRNAs and possible miRNA candidates contained in the submitted sequencing data were obtained together with their expression profile. The functions of known and predicted miRNAs were then analyzed by miRNA target prediction followed by target gene annotations. Finally, to extract the biological significance of the miRNAs in the samples, further annotations of the known miRNA and target genes were performed by collecting the public expression datasets of miRNA and target genes in normal and cancer tissues. Conclusion: We introduce a web-based analysis platform called miRNA Analysis System (miRAS), for processing and analyzing of the sequence data obtained from the total RNA clone method. This letter proposes a novel converter topology that interfaces three power ports: a source, a bidirectional storage port, and an isolated load port. The proposed converter is based on a modified version of the isolated half-bridge converter topology that utilizes three basic modes of operation within a constant-frequency switching cycle to provide two independent control variables. This allows tight control over two of the converter ports, while the third port provides the power balance in the system. The switching sequence ensures a clamping path for the energy of the leakage inductance of the transformer at all times. This energy is further utilized to achieve zero-voltage switching for all primary switches for a wide range of source and load conditions. Basic steady-state analysis of the proposed converter is included, together with a suggested structure for feedback control. Key experimental results are presented that validate the converter operation and confirm its ability to achieve tight independent control over two power processing paths. This topology promises significant savings in component count and losses for power-harvesting systems. As a major complication after percutaneous coronary intervention (PCI) in patients who suffer from coronary artery disease, instent restenosis (ISR) poses a significant challenge for clinical management. A miRNA-mRNA regulatory network of ISR can be constructed to better reveal the occurrence of ISR. The relevant data set from the Gene Expression Omnibus (GEO) database was downloaded, and 284 differentially expressed miRNAs (DE-miRNAs) and 849 differentially expressed mRNAs (DE-mRNAs) were identified. As predicted by online tools, 65 final functional genes (FmRNAs) were overlapping DE-mRNAs and DE-miRNAs target genes. In the biological process (BP) terms of gene ontology (GO) functional analysis, the FmRNAs were mainly enriched in the cellular response to peptide, epithelial cell proliferation, and response to peptide hormone. In the Kyoto Encyclopedia of Genes and Genomes (KEGG) analysis, the FmRNAs were mainly enriched in breast cancer, endocrine resistance, and Cushing syndrome. Jun proto-oncogene, activator protein-1 (AP-1) transcription factor subunit (JUN), insulin-like growth factor 1 receptor (IGF1R), member RAS oncogene family (RAB14), specificity protein 1 (SP1), protein tyrosine phosphatase nonreceptor type 1 (PTPN1), DDB1 and CUL4 associated factor 10 (DCAF10), retinoblastoma-binding protein 5 (RBBP5), and eukaryotic initiation factor 4A-I (EIF4A1) were hub genes in the protein-protein interaction network (PPI network). The miRNA-mRNA network containing DE-miRNAs and hub genes was built. Hsa-miR-139-5p-JUN, hsa-miR-324-5p-SP1 axis pairs were found in the miRNA-mRNA network, which could promote ISR development. The aforementioned results indicate that the miRNA-mRNA network constructed in ISR has a regulatory role in the development of ISR and may provide new approaches for clinical treatment and experimental development. SIL040, an introgression line (IL) developed by introgressing chromosomal segments from an accession of Oryza rufipogon into an indica cultivar Guichao 2, showed significantly less grains per panicle than the recurrent parent Guichao 2. Quantitative trait locus (QTL) analysis in F2 and F3 generations derived from the cross between SIL040 and Guichao 2 revealed that gpa7, a QTL located on the short arm of chromosome 7, was responsible of this variation. Alleles from O. rufipogon decreased grains per panicle. To fine mapping of gpa7, a high-resolution map with 1,966 F2 plants derived from the cross between SIL040 and Guichao 2 using markers flanking gpa7 was constructed, and detailed quantitative evaluation of the structure of main panicle of each of F3 families derived from recombinants screened was performed. By two-step substitution mapping, gpa7 was finally narrowed down to a 35-kb region that contains five predicted genes in cultivated rice. The fact that QTLs for five panicle traits (length of panicle, primary branches per panicle, secondary branches per panicle, grains on primary branches and grains on secondary branches) were all mapped in the same interval as that for gpa7 suggested that this locus was associated with panicle structure, showing pleiotropic effects. The characterizing of panicle structure of IL SIL040 further revealed that, during the domestication from common wild allele to cultivated rice one at gpa7, not only the number of branches and grains per panicle increased significantly, more importantly, but also the ratio of secondary branches per panicle to total branches per panicle and the ratio of grains on secondary branches per panicle to total grains per panicle increased significantly. All these results reinforced the idea that gpa7 might play an important role in the regulation of grain number per panicle and the ratio of secondary branches per panicle during the domestication of rice panicle. Introgression lines (ILs) are useful tools for precise mapping of quantitative trait loci (QTLs) and the evaluation of gene action or interaction in theoretical studies. A set of 159 ILs carrying variant introgressed segments from Chinese common wild rice ( Oryza rufipogon Griff.), collected from Dongxiang county, Jiangxi Province, in the background of Indica cultivar ( Oryza sativa L.), Guichao 2, was developed using 126 polymorphic simple sequence repeats (SSR) loci. The 159 ILs represented 67.5% of the genome of O. rufipogon. All the ILs have the proportions of the recurrent parent ranging from 92.4 to 99.9%, with an average of 97.4%. The average proportion of the donor genome for the BC4F4 population was about 2.2%. The mean numbers of homozygous and heterozygous donor segments were 2 (ranging 0–8) and 1 (ranging 0–7), respectively, and the majority of these segments had sizes less than 10 cM. QTL analysis was conducted based on evaluation of yield-related traits of the 159 ILs at two sites, in Beijing and Hainan. For 6 out of 17 QTLs identified at two sites corresponding to three traits (panicles per plant, grains per panicle and filled grains per plant, respectively), the QTLs derived from O. rufipogon were usually associated with an improvement of the target trait, although the overall phenotypic characters of O. rufipogon were inferior to that of the recurrent parent. Of the 17 QTLs, 5 specific QTLs strongly associated with more than one trait were observed. Further analysis of the high-yielding and low-yielding ILs revealed that the high-yielding ILs contained relatively less introgressed segments than the low-yielding ILs, and that the yield increase or decrease was mainly due to the number of grain. On the other hand, low-yielding ILs contained more negative QTLs or disharmonious interactions between QTLs which masked trait-enchancing QTLs. Local increase in blood flow during neural activity forms the basis for functional brain imaging, but its mechanism remains poorly defined. Here we show that cortical astrocytes in vivo possess a powerful mechanism for rapid vasodilation. We imaged the activity of astrocytes labeled with the calcium (Ca2+)-sensitive indicator rhod-2 in somatosensory cortex of adult mice. Photolysis of caged Ca2+ in astrocytic endfeet ensheathing the vessel wall was associated with an 18% increase in arterial cross-section area that corresponded to a 37% increase in blood flow. Vasodilation occurred with a latency of only 1–2 s, and both indomethacin and the cyclooxygenase-1 inhibitor SC-560 blocked the photolysis-induced hyperemia. These observations implicate astrocytes in the control of local microcirculation and suggest that one of their physiological roles is to mediate vasodilation in response to increased neural activity. Hypersynchronous neuronal firing is a hallmark of epilepsy, but the mechanisms underlying simultaneous activation of multiple neurons remains unknown. Epileptic discharges are in part initiated by a local depolarization shift that drives groups of neurons into synchronous bursting. In an attempt to define the cellular basis for hypersynchronous bursting activity, we studied the occurrence of paroxysmal depolarization shifts after suppressing synaptic activity using tetrodotoxin (TTX) and voltage-gated Ca2+ channel blockers. Here we report that paroxysmal depolarization shifts can be initiated by release of glutamate from extrasynaptic sources or by photolysis of caged Ca2+ in astrocytes. Two-photon imaging of live exposed cortex showed that several antiepileptic agents, including valproate, gabapentin and phenytoin, reduced the ability of astrocytes to transmit Ca2+ signaling. Our results show an unanticipated key role for astrocytes in seizure activity. As such, these findings identify astrocytes as a proximal target for the treatment of epileptic disorders. The neuroprotective effect and molecular mechanisms underlying preconditioning with N-methyl-D-aspartate (NMDA) in cultured hippocampal neurons have not been described. Pre-incubation with subtoxic concentrations of the endogenous neurotransmitter glutamate protects vulnerable neurons against NMDA receptor-mediated excitotoxicity. As a result of physiological preconditioning, NMDA significantly antagonizes the neurotoxicity resulting from subsequent exposure to an excitotoxic concentration of glutamate. The protective effect of glutamate or NMDA is time- and concentration-dependent, suggesting that sufficient agonist and time are required to establish an intracellular neuroprotective state. In these cells, the TrkB ligand, brain-derived neurotrophic factor (BDNF) attenuates glutamate toxicity. Therefore, we tested the hypothesis that NMDA protects neurons via a BDNF-dependent mechanism. Exposure of hippocampal cultures to a neuroprotective concentration of NMDA (50 μ m) evoked the release of BDNF within 2 min without attendant changes in BDNF protein or gene expression. The accumulated increase of BDNF in the medium is followed by an increase in the phosphorylation (activation) of TrkB receptors and a later increase in exon 4-specific BDNF mRNA. The neuroprotective effect of NMDA was attenuated by pre-incubation with a BDNF-blocking antibody and TrkB-IgG, a fusion protein known to inhibit the activity of extracellular BDNF, suggesting that BDNF plays a major role in NMDA-mediated survival. These results demonstrate that low level stimulation of NMDA receptors protect neurons against glutamate excitotoxicity via a BDNF autocrine loop in hippocampal neurons and suggest that activation of neurotrophin signaling pathways plays a key role in the neuroprotection of NMDA. High-voltage amplifiers as a means of amplifying the low-output voltage signals of the feedback controllers to the suspension voltages typically in the kilovolts range are often required for electrostatic force generation in electrostatic levitation. This paper proposes a high-voltage dc amplifier including an amplitude modulator, a power amplifier, a step-up transformer, a pair of peak detectors, and a voltage feedback channel to stabilize the amplifier outputs in an effort to provide high suspension voltage and fast dynamic response. Since the various carrier frequencies have virtually no effect on the power consumption of the dc amplifier by filtering out the high-frequency carrier components with peak detectors while keeping the input signal unaffected, satisfactory dynamic performance can be achieved by choosing a sufficiently high carrier frequency. The operating principle of the dc amplifier is analyzed, followed by an experimental performance evaluation and discussion for electrostatic levitation applications. The experimental results demonstrate the superiority of the high-voltage dc amplifier over classical ac amplifiers in terms of dynamic response, force-voltage coefficient, voltage ripple, power consumption, and long-time stability using a carrier frequency of 30 kHz and the closed-loop control scheme. The viability of 1,3-dipolar cycloadditions of a series of 1,3-dipolar molecules (azomethine ylide, ozone, nitrone, nitrile imine, nitrile ylide, nitrile oxide, diazomethane, and methyl azide) onto the sidewalls of carbon nanotubes has been assessed theoretically by means of a two-layered ONIOM approach. The theoretical calculations predict the following: (i) other than the 18-valence-electron azomethine ylide and ozone, the 16-valence-electron nitrile ylide and nitrile imine are the best candidates for experimentalists to try; (ii) upon 1,3-dipolar cycloaddition, a 1,3-diople molecule is di-σ-bonded to a pair of carbon atoms on the sidewall of SWNT, forming a five-membered ring surface species; (iii) the as-formed 1,3-dipole-SWNT bonding is much weaker than that in the products of the molecular 1,3-DC reactions and can be plausibly broken by heating at elevated temperatures; (iv) the sidewalls of the armchair (n,n) SWNTs (n = 5-10) are subject to the 1,3-DCs of ozone and azomethine ylides; (v) both the 1,3-DC reactivity and retro-1,3-DC reactivity are moderately dependent on the diameters of SWNTs, implying the feasibility of making use of the heterogeneous 1,3-DC chemistry to purify and separate SWNTs diameter-specifically. Noncoherent two- and three-phase two-way relaying (TWR) techniques are bandwidth efficient and entail no pilot symbol–related rate loss. However, noncoherent two-phase TWR (2P-TWR) has not been compared with three-phase TWR (3P-TWR), and several unanswered questions remain. In this paper, we focus on the noncoherent detection of the relay with the decode-and-forward technique. For the noncoherent maximum-likelihood detector at the relay, we propose a distance measure for the transmitted signals of the two terminal nodes. For 2P-TWR, we propose that the signal constellation of one terminal node is the signal constellation of the other terminal node rotated by an angle. Using the proposed distance measure, we obtain the optimal angles and thus resolve those unanswered questions pertaining to 2P-TWR. Finally, noncoherent 3P-TWR was found to significantly outperform noncoherent 2P-TWR in simulations and distance analyses. [ABSTRACT FROM AUTHOR]

Background: Recent studies have revealed that atrial fibrillation (AF) patients have a high risk of developing cognitive impairment, vascular dementia, and Alzheimer's disease (AD). Some reports suggest that the application of oral anticoagulant with an appropriate dose may have a preventive effect on AD. However, which oral anticoagulant drug is more appropriate for preventing AD and the underlying mechanism(s) is still unknown.Objective: The aim of the present study was to assess the treatment effect of rivaroxaban administration as well as investigate the roles of PAR-1 and PAR-2 in the AD + CAA mice model.Methods: In the present study, we compared a traditional oral anticoagulant, warfarin, and a direct oral anticoagulant (DOAC), rivaroxaban, via long-term administration to an AD with cerebral amyloid angiopathy (CAA) mice model.Results: Rivaroxaban treatment attenuated neuroinflammation, blood-brain barrier dysfunction, memory deficits, and amyloid-β deposition through PAR-1/PAR-2 inhibition in the AD + CAA mice model compared with warfarin and no-treatment groups.Conclusion: The present study demonstrates that rivaroxaban can attenuate AD progress and can be a potential choice to prevent AD. • The rainflow distribution was divided into three regions based on the value of zero-order spectral moment of power spectral density curve. • Each region of the rainflow distribution was fitted using Rayleigh or exponential functions, respectively. • A probability density function combining two Rayleigh functions and an exponential function was proposed. The power spectral density curves of the random vibration response of a plate specimen with different spectral width coefficients were obtained by selecting different frequency ranges, and the probability distributions of the stress amplitudes were obtained by applying Monte Carlo method and rainflow counting method. Each probability distribution was divided into three segments by the corresponding zero-order spectral moment value, and each segment was fitted using either a Rayleigh function or an exponential function. Finally, a probability density function of stress amplitude was proposed by combining two Rayleigh functions and an exponential function. The proposed probability density function can be used to obtain the stress amplitude probability distribution for the power spectral density curves of structural random vibration response with any bandwidth, and the accuracy of the proposed function was verified based on the response data of a specimen of 7075-T651 aluminum alloy material under the action of six typical power spectral curves. Implementation of strict emission mitigation measures since 2013 has significantly changed air pollutants in the Beijing-Tianjin-Hebei region (BTH), China. Observations show that ozone (O 3) concentrations have increased by 62.40% (27.84%) and SO 2 concentrations have decreased by 56.42% (35.07%) during particulate pollution episodes in Beijing (BTH) in the autumn from 2013 to 2015. The measured nitrate concentration in Beijing has increased markedly, which to a large degree offsets the sulfate decrease caused by SO 2 emission mitigation. Using the WRF-Chem model, we demonstrate that the enhanced nitrate formation is primarily attributed to increasing atmospheric oxidizing capacity (AOC) and decreasing sulfate competition for base ions. A 9.41–46.24% (7.58–40.97%) decrease in OH radical (O 3) concentrations in October 2015 reduces nitrate and fine particulate matters (PM 2.5) concentrations by 2.51–18.18% and 3.15–18.90% in Beijing, respectively. Based on the scenario in October 2015, if the SO 2 emission increases by 20.00–100.00%, the PM 2.5 concentration increases by 3.02–11.21%, but the nitrate level decreases by 2.48–21.87% simultaneously. Our results suggest that the nitrate aerosol has become a dominant contributor to particulate pollution in Beijing and that decreasing AOC is critical to mitigate nitrate and PM 2.5 concentrations. ● Increasing nitrate concentration largely offsets the sulfate decrease since 2013. ● Increasing nitrate is mainly due to enhanced AOC and weakened sulfate competition. ● Decreasing AOC is beneficial to mitigate nitrate and PM 2.5 concentrations. The observed near-surface ozone (O 3) concentration has been remarkably increasing during recent years in winter in the Guanzhong basin, central China, showing a continuous enhancement of the atmospheric oxidizing capacity (AOC). The impact of such a change in the AOC on secondary aerosol formation, however, has not yet been assessed. In this study, we simulate the formation of O 3 and airborne particles in the atmosphere using the WRF-Chem model, in which the AOC is calculated quantitatively, to understand the responses of secondary aerosols to the AOC increase. Meteorological observations, air pollutants including O 3 , NO 2 , SO 2 , CO, and PM 2.5 concentrations at ambient monitoring sites, and the main compositions of submicron particulates measured using ACSM are used to constrain the model simulation. The model result shows that the population hourly and postmeridian O x (=O 3 + NO 2) concentrations are good indicators for the wintertime AOC in the basin, suggested by the significantly positive correlations between them. Sensitivity experiments present that the AOC changes may exert important influences on fine particle (PM 2.5) concentration with an average rate of 1.94 (μg m−3)/(106 cm−3 s−1) for Δ(PM 2.5)/Δ(AOC), which is mostly caused by the mass changes in secondary organic aerosol (43%) and nitrate aerosol (40%) and less attributed to the ammonium (11%) and sulfate (6%) components. A strong relationship between Alzheimer's disease (AD) and vascular dysfunction has been the focus of increasing attention in aging societies. In the present study, we examined the long-term effect of scallop-derived plasmalogen (sPlas) on vascular remodeling-related proteins in the brain of an AD with cerebral hypoperfusion (HP) mouse model. We demonstrated, for the first time, that cerebral HP activated the axis of the receptor for advanced glycation endproducts (RAGE)/phosphorylated signal transducer and activator of transcription 3 (pSTAT3)/provirus integration site for Moloney murine leukemia virus 1 (PIM1)/nuclear factor of activated T cells 1 (NFATc1), accounting for such cerebral vascular remodeling. Moreover, we also found that cerebral HP accelerated pSTAT3-mediated astrogliosis and activation of the nucleotide-binding domain and leucine-rich repeat protein 3 (NLRP3) inflammasome, probably leading to cognitive decline. On the other hand, sPlas treatment attenuated the activation of the pSTAT3/PIM1/NFATc1 axis independent of RAGE and significantly suppressed NLRP3 inflammasome activation, demonstrating the beneficial effect on AD. Based on the CNN-LSTM fusion deep neural network, this paper proposes a seismic velocity model building method that can simultaneously estimate the root mean square (RMS) velocity and interval velocity from the common-midpoint (CMP) gather. In the proposed method, a convolutional neural network (CNN) Encoder and two long short-term memory networks (LSTMs) are used to extract spatial and temporal features from seismic signals, respectively, and a CNN Decoder is used to recover RMS velocity and interval velocity of underground media from various feature vectors. To address the problems of unstable gradients and easily fall into a local minimum in the deep neural network training process, we propose to use Kaiming normal initialization with zero negative slopes of rectified units and to adjust the network learning process by optimizing the mean square error (MSE) loss function with the introduction of a freezing factor. The experiments on testing dataset show that CNN-LSTM fusion deep neural network can predict RMS velocity as well as interval velocity more accurately, and its inversion accuracy is superior to that of single neural network models. The predictions on the complex structures and Marmousi model are consistent with the true velocity variation trends, and the predictions on field data can effectively correct the phase axis, improve the lateral continuity of phase axis and quality of stack section, indicating the effectiveness and decent generalization capability of the proposed method.
