Reactive oxygen species (ROS) play a crucial role in the pathogenesis of acute and chronic respiratory diseases. Antioxidants have been found to ameliorate airway inflammation and hyperresponsiveness in animal models employing short-term exposure to allergen. However, little data are available on the effect of antioxidants on airway remodeling and signaling pathways in chronic asthma. In the present study, we used a long-term exposure murine model of allergic airway disease to evaluate the effects of an antioxidant, L-2-oxothiazolidine-4-carboxylic acid (OTC) or α-lipoic acid (LA) on airway remodeling, focusing on the ROS-related hypoxia-inducible signaling. Long-term challenge of ovalbumin (OVA) increased ROS production, airway inflammation, and airway hyperresponsiveness, and developed features of airway remodeling such as excessive mucus secretion, subepithelial fibrosis, and thickening of the peribronchial smooth muscle layer. Administration of OTC or LA reduced these features of asthma, including airway remodeling, which was accompanied by suppression of transforming growth factor-β1, vascular endothelial growth factor, and T-helper 2 cytokines. In addition, OVA-induced activation of nuclear factor-κB (NF-κB), nuclear factor erythroid 2p45-related factor-2 (Nrf2), hypoxia-inducible factor (HIF)-1α, and HIF-2α was reduced by OTC or LA. Our results also showed that OTC or LA down-regulated phosphoinositide 3-kinase activity and decreased phosphorylation of p38 mitogen-activated protein kinase but not extracellular signal-regulated kinase 1/2 or c-Jun N-terminal kinase. These findings demonstrate that OTC and LA can inhibit activation of NF-κB, Nrf2, and HIF, leading to attenuate allergen-induced airway remodeling. [ABSTRACT FROM AUTHOR]

D2-40 is a recently developed monoclonal antibody that reacts with a 40 kDa O-linked sialoglycoprotein and has been used for the assessment of lymphatic invasion in tumor specimens. We have evaluated the diagnostic usefulness of D2-40 and association of its immunopositivity with clinicopathological parameters in adenocarcinoma and squamous cell carcinoma of the lung. We investigated 97 cases of surgically resected adenocarcinoma or squamous cell carcinoma of the lung for the determination of D2-40 positivity in tumor cells and peritumoral lymphatic vessel density (LVD) using an immunostaining method. D2-40 immunoreactivity in tumor cells was invariably negative in adenocarcinoma but 47% of squamous cell carcinomas were positive. D2-40 positivity in the tumor was significantly associated with high LVD in squamous cell carcinoma ( P < 0.006). There was no significant association between peritumoral LVD and clinicopathologic parameters, including lymphatic vessel invasion, lymph node metastasis, and survival in adenocarcinoma and squamous cell carcinoma. These results suggest that D2-40 immunoreactivity in tumor cells can be used for distinguishing between adenocarcinoma and squamous cell carcinoma and that the reactivity of tumor cells with D2-40 is positively correlated with LVD in squamous cell carcinoma but not with lymph node metastasis in adenocarcinoma and squamous cell carcinoma. A solid-state reference electrode (SSRE) was fabricated by layering a silicone rubber (SR) film containing KCl on an AgCl surface, then a perfluorinated ionomer film, and finally a polyurethane-based membrane containing an ionophore, a lipophilic ionic additive, and a plasticizer, respectively. The addition of SiCl4 to the polyurethane-based membrane layer enhanced the strength of the membrane in an aqueous solution. The morphologies of the membranes were studied separately by SEM. The fabrication of the Ag/AgCl electrode through this layer-by-layer polymer coating improved the electrode stability enormously. In addition, the potential drift of the SSRE according to the pH of the medium was minimized by introducing a H+-ion-selective ionophore (tridodecylamine; TDDA) into the outmost polymer membrane. The cyclic voltammetric and potentiometric responses using the SSRE and a conventional reference electrode, respectively, were consistent. The SSRE exhibited little potential variation even in the case of the addition of very high concentrations of various salts, such as Na salicylate, LiCl, KCl, CaCl2, MgCl2, KNO3, NaCl, and NaHCO3. The practicability of the proposed SSRE was tested for the determination of blood pH and pCO2 in a flow cell system. The SSRE fabricated in the present study was stable over two years. (1−x)Pb(Zr0.53Ti0.47)O3−xBiYO3 (PZT–x·BY) ceramics were synthesized by conventional bulk ceramic processing technology with various BiYO3 (BY) doping contents. The maximum values of the piezoelectric properties were obtained in the PZT–0.01BY samples sintered at 1160 °C. After optimizing the composition of the PZT–0.01BY piezoelectrics, the sintering temperature could be lowered down to 1000 °C by applying a nano-powder technique. The maximum values of the piezoelectric properties achieved by this novel process were: d33 of 580 pC/N, kp of 67%, and g33 of 95 mV · m N−1. Finally we fabricated a thick-film piezo-speaker with optimized PZT–0.01BY nano-powders using multi-layered ceramic capacitor processes. The sound pressure level (SPL) of the speaker was analyzed at a distance of 50 cm with a driving voltage of 5 Vrms in the frequency range of 100 Hz–11 kHz. Background: This study investigated the effect of increased job stress, caused by musculoskeletal disease (MSD) among firefighters, on a firefighter's intention to leave the profession, henceforth referred to as “turnover intention,” and verified the moderating effect of mindfulness on such a relationship. Methods: A survey involving a total of 549 Korean male firefighters as participants was conducted herein, and the following results were obtained: the mediation effect of the MSD to turnover intention through job stress was confirmed, and the indirect effect of job stress was verified. Results: We verified the moderated mediation effect of mindfulness on the relation:MSD, job stress, and turnover intention. The conditional indirect effect for middle and high levels of mindfulness is significant. Conclusion: The result of this study is supported by proofs of the relationship between a firefighter's MSD, job stress, and turnover intention, and these case studies reveal the moderated mediation effect of dispositional mindfulness.
