Jasmonate sind eine wichtige Gruppe von Phytohormonen, die neben pflanzlichen Abwehrreaktionen z.B. Wachstum und Mechanotransduktion regulieren. Lipoxygenase (LOX), Allen Oxid Synthase (AOS) und Allen Oxid Zyklase (AOC) sind an den plastidären Schritten der Jasmonatbiosynthese beteiligt. Der Schwerpunkt dieser Arbeit lag auf der Untersuchung des katalytischen Mechanismus der Allen Oxid Zyklase über die Strukturbestimmung von Allen Oxid Zyklase 2 aus \(\textit {A.thaliana}\) (AtAOC2) mit gebundenen Liganden, sowie von AtAOC2 Punktmutanten. Außerdem wurden die AOC2-AOS Interaktion und der Oligomerisierungszustand von AtAOC2 in Lösung analysiert. Rubisco (Ribulose-1,5,-Biphosphat-Karboxylase/Oxygenase) spielt eine wichtige Rolle in der Natur, da es anorganische und organische Kohlenstoffquellen verknüpft. Das Ziel dieser Arbeit war die Analyse von Rubisco aus dem Cyanobakterium \(\textit {Th.elongatus}\) und die Suche nach strukturellen Eigenschaften von Rubisco, die die hohe Spezifität verursachen. The steps of jasmonate biosynthesis localized in chloroplasts involve the action of lipoxygenase (LOX), allene oxide synthase (AOS) and allene oxide cyclase (AOC). This work focused on getting new insights into the catalytic mechanism of allene oxide cyclase by the structural analysis of point-mutants and ligand-bound complexes of AOC2 from \(\textit {A.thaliana}\) (AtAOC2). In addition, the interactions between AOC2 and AOS from \(\textit {A.thaliana}\) and the oligomerization state of AtAOC2 in solution were analyzed. Rubisco, an enzyme involved in the dark phase of photosynthesis, is the most abundant protein on earth, linking the pools of inorganic and organic carbon in environment. The aim of this work was to analyse the cyanobacterial Rubisco from \(\textit {Th.elongatus}\) in order to find structural factors, which determine its high specificity.

Das Interferon-\(\gamma\)-induzierte humane Guanylat-bindende Protein 1 (hGBP1) gehört zu den großen GTPase und ist Mitglied der Dynamin-Superfamilie. Die Kristallstruktur des monomeren Proteins und des Dimers der isolierten G-Domänen steht zur Verfügung. Die durch GTP-Bindung induzierte Oligomerisierung ist mit erheblichen Konformationsänderungen verbunden. In dieser Doktorarbeit wurde die Bildung von hGBP1-Homokomplexen mittels Förster-Resonanz-Energie-Transfer (FRET) u.a. unter Einzelmolekülbedingungen und Elektronen-Spin-Resonanz (ESR) untersucht. Eine selektive Fluoreszenzmarkierung ermöglichte die Bestimmung von intra- und intermolekularen Distanzänderungen im Verlauf der GTP-Hydrolyse. Unter Vorgabe der experimentell bestimmten Distanzen wurden auf Basis der monomeren Kristallstrukturkoordinaten molekular-dynamische Simulationen durchgeführt.

Linear α-olefins (LAOs) are crucial importance for the chemical and petrochemical industry as they are highly valuable feedstock for a variety of process mainly as co-monomer for the production of different grade of polyethylene (1-butene, 1-hexene and 1-octene). Some systems and predominantly chromium and titanium systems have performed selective dimerization, trimerization and tetramerization of ethylene to higher α-olefins. For toxicity reasons, industrial and academic research focus more and more on titanium-based systems.Our objective in this context was the study of potential “tridentate phenoxy” systems of Ti(IV) for selective trimerization of ethylene to 1-hexene. Numerous systems with general formula [(ArO-X-L)TiCl3] (X=O,P,N and L=O,P,N) were described and characterized. The study of electronic and steric properties and their consequences on catalytic performances in oligomerization/polymerization of ethylene were discussed. They showed the criticity of all electronic and structural parameters of ligands. Only one system of this family is able to produce selectively 1-hexene, others give mainly polyethylene.DFT study associated with an experimental study of redox properties on catalytic precursors or the hemilabile behavior of ligands involved, led us to propose a mechanism identifying the activation phase as the key to switch between polymerization or selective trimerization of ethylene., Les α-oléfines jouent un rôle très important en tant qu'intermédiaires réactionnels dans l'industrie chimique et pétrochimique. Un seul système de cette famille est capable de produire sélectivement de l’hexène-1, les autres conduisant à la polymérisation de l’éthylène.Une étude DFT associée à une étude expérimentale des propriétés redox des précurseurs catalytiques ou encore du comportement hémilabile des ligands mis en jeu, nous a conduit à proposer un mécanisme identifiant la phase d’activation comme l’élément clé permettant d’orienter les systèmes étudiés vers la polymérisation ou la production sélective d’hexène-1.

In a study from 2011, the protein muskelin was described as a central coordinator of the retrograde transport of GABA(A) receptors in neurons. As muskelin governs the transport along actin filaments as well as microtubules, it might be the first representative of a novel class of regulators, which coordinate cargo transport across the borders of these two independent systems of transport paths and their associated motorproteins. To establish a basis for understanding the mode of operation of muskelin, the aim of this thesis was an in-depth biochemical and structural characterization of muskelin and its interaction with the GABA(A) receptor. One focus of the work was the analysis of the oligomerization of muskelin. As could be demonstrated, the oligomerization is based on two independent interactions mediated by different domains of the protein: a known interaction of the N-terminal discoidin domain with the C-terminal portion, termed head-to-tail interaction, and a dimerization of the LisH motif in muskelin that was so far neglected in the literature. For the detailed studies of both binding events, the solution of a crystal structure of a fragment of muskelin, comprising the Discoidin domain and the LisH motif, was an important basis. The fragment crystallized as a dimer, with dimerization being mediated solely by the LisH motif. Biochemical analysis corroborated that the LisH motif in muskelin serves as a dimerization element, and, moreover, showed that the C-terminal domain of the protein substantially stabilizes this dimerization. In addition, the crystal structure revealed the molecular composition of the surface of the head in the head-to-tail interaction, namely the discoidin domain. This information enabled to map the amino acids contributing to binding, which showed that the binding site of the head-to-tail interaction coincides with the generic ligand binding site of the discoidin domain. As part of the analyses, residues that are critical for LisH-dimerization and the head-to-tail binding, respectively, were identified, whose mutation specifically interfered with each of the interactions separately. These mutations allowed to investigate the interplay of these interactions during oligomerization. It could be shown that recombinant muskelin assembles into a tetramer to which both interactions, the LisH-dimerization and the head-to-tail binding, contribute independently. When one of the two interactions was disturbed, only a dimer mediated via the respective other interaction could be formed; when both interactions were disturbed, the protein was present as monomer. Furthermore, Frank Heisler in the group of Matthias Kneussel was able to show the drastic impact of an impaired LisH-dimerization on muskelin in cells using these mutations. Disturbing the LisH-dimerization led to a complete redistribution of the originally cytoplasmic muskelin to the nucleus which was accompanied by a severe impairment of its function during GABA(A) receptor transport. Following up on these results in an analysis of muskelin variants, for which alterations of the subcellular localization had been published earlier, the crucial influence of LisH-dimerization to the subcellular localization and thereby the role of muskelin in the cell was confirmed. The biochemical studies of the interaction of muskelin and the alpha1 subunit of the GABA(A) receptor demonstrated a direct binding with an affinity in the low micromolar range, which is mediated primarily by the kelch repeat domain in muskelin. For the binding site on the GABA(A) receptor, it was confirmed that the thirteen most C-terminal residues of the intracellular domain are critical for the binding of muskelin. In accordance with the strong conservation of these residues among the alpha subunits of the GABA(A) receptor, it could be shown that an interaction with muskelin in vitro is also possible for the alpha2 and alpha5 subunits. Based on the comparison of the binding sites between the homologous subunits, tentative conclusions can be drawn about the details of the binding, which may serve as a starting point for follow-up studies. This thesis thereby makes valuable contributions to the understanding of muskelin, in particular the significance of its oligomerization. It furthermore provides an experimental framework for future studies that address related topics, such as the characterization of other muskelin interaction partners, or the questions raised in this work., Das Protein Muskelin wurde in einer Studie aus dem Jahr 2011 als zentraler Koordinator des retrograden Transports von GABA(A)-Rezeptoren in Neuronen beschrieben. Da Muskelin den Transport des Rezeptors sowohl entlang von Aktinfilamenten als auch Mikrotubuli steuert, könnte es der erste bekannte Vertreter einer neuen Klasse von Regulatoren sein, die den Transport einer Fracht über die Grenzen dieser beiden unabhängigen Systeme von Transportwegen und der damit assoziierten Motorproteine hinweg koordinieren. Um Grundlagen für das Verständnis der Wirkungsweise von Muskelin zu schaffen, war das Ziel dieser Arbeit die biochemische und strukturelle Charakterisierung von Muskelin und seiner Interaktion mit dem GABA(A)-Rezeptor. Ein Schwerpunkt der Arbeit lag dabei auf der Analyse der Oligomerisierung von Muskelin. Wie gezeigt werden konnte, beruht die Oligomerisierung auf zwei unabhängigen, von verschiedenen Domänen des Proteins vermittelten Bindungen: zum Einen auf einer bereits bekannten Wechselwirkung der N-terminalen Discoidin-Domäne und des C-terminalen Teils (anschaulich als Head-to-tail, englisch für Kopf-an-Schwanz, bezeichnet), zum Anderen auf einer in der Literatur bisher außer Acht gelassenen Dimerisierung des LisH-Motivs in Muskelin. Für die detaillierten Studien beider Bindungen lieferte die Aufklärung der Kristallstruktur eines Teilstücks von Muskelin, das die Discoidin-Domäne und das LisH-Motiv umfasst, eine wichtige Grundlage. Das Teilstück kristallisierte als Dimer, wobei die Dimerisierung ausschließlich über das LisH-Motiv vermittelt wurde. Die biochemischen Analysen bestätigten, dass das LisH-Motiv in Muskelin als Dimerisierungselement wirkt, und zeigten darüber hinaus, dass die C-terminale Domäne des Proteins diese Dimerisierung wesentlich stabilisiert. Zudem offenbarte die Kristallstruktur den molekularen Aufbau der Oberfläche des Kopfes in der Head-to-tail-Bindung, der Discoidin-Domäne. Die Kartierung der zur Bindung beitragenden Aminosäuren belegte, dass die Bindungsstelle der Head-to-tail-Interaktion mit der generischen Ligandenbindungsstelle der Discoidin-Domäne zusammenfällt. Als Teil der Analysen wurden zur LisH-Dimerisierung oder zur Head-to-tail-Bindung kritisch beitragende Aminosäurenseitenketten identifiziert, durch deren Mutationen Ispezifisch jeweils eine der beiden Interaktionen unterbunden werden konnte. Diese Mutationen ermöglichten es, das Zusammenspiel der Bindungen in der Oligomerisierung zu untersuchen. Es konnte gezeigt werden, dass rekombinantes Muskelin ein Tetramer bildet, wozu beide Interaktionen, die LisH-Dimerisierung und die Head-to-tail-Bindung, unabhängig beitragen. Wurde jeweils eine der beiden Interaktionen durch Mutation gestört, konnte nur noch ein über die jeweils andere Interaktion vermitteltes Dimer gebildet werden, bei gleichzeitiger Störung beider Interaktionen lag das Protein als Monomer vor. Darüber hinaus konnte Frank Heisler in der Arbeitsgruppe von Matthias Kneussel mit Hilfe dieser Mutationen zeigen, dass eine Störung der LisH-Dimerisierung drastische Auswirkungen auf Muskelin in Zellen hat. Die Störung der LisH-Dimerisierung führte zu einer vollständigen Umverteilung des sonst im Zytoplasma lokalisierten Muskelins in den Kern, begleitet von einer starken Beeinträchtigung seiner Funktion im Transport des GABA(A)-Rezeptors. Auf diesen Ergebnissen aufbauend wurde durch Analysen der oligomeren Zustände von Muskelin-Varianten, für die in der Literatur eine veränderte Lokalisation beschrieben worden war, die entscheidende Bedeutung der LisH-Dimerisierung für die subzelluläre Verteilung und damit die Rolle von Muskelin in der Zelle bekräftigt. Die biochemischen Studien der Interaktion von Muskelin und der alpha1-Untereinheit des GABA(A)-Rezeptors demonstrierten eine direkte Bindung mit mikromolarer Affinität, die in Muskelin vorwiegend durch die Kelch-repeat-Domäne vermittelt wird. Für die Bindungsstelle auf Seite des GABA(A)-Rezeptors wurde bestätigt, dass die dreizehn C-terminalen Reste der intrazellulären Domäne entscheidend sind. In Übereinstimmung mit der starken Konservierung dieser Reste in verschiedenen alpha-Untereinheiten des GABA(A)-Rezeptors, konnte gezeigt werden, dass in vitro eine Bindung von Muskelin auch an die intrazelluläre Domäne der alpha2- und alpha5-Untereinheit möglich ist. Anhand des Vergleichs der Bindungsstelle zwischen den homologen Untereinheiten lassen sich erste Rückschlüsse auf die Details der Interaktion ziehen, die als Anknüpfungspunkt für kommende Studien dienen können. Diese Arbeit liefert damit wesentliche Beiträge zum Verständnis von Muskelin, insbesondere der Bedeutung seiner Oligomerisierung. Sie bietet zudem ein experimentelles Rahmenwerk für zukünftige Studien, die sich verwandten Themen, wie der Charakterisierung weiterer Interaktionen von Muskelin, oder den in dieser Arbeit aufgeworfenen Fragen widmen.

The demand for short linear alpha olefins is constantly increasing and motivates the development of robust and selective catalysts. In this thesis, several libraries of phosphorus ligands with the capacity to form dissymmetric or supramolecular assemblies were synthesized. The variability observed within the aminophosphine libraries, clearly reflected by the various tautomeric equilibrium of the ligand, was also observed in the nickel complexes as a single ligand could generate several complexes with different structures. Sulphonyliminobisphosphine were then introduced as a new class of ligands. These precursors rearrange in the presence of nickel to generate diphosphinamine nickel complexes. Activated by MAO, these complexes are active in the reaction of ethylene oligomerisation and produce short chain olefins. A new approach that forms stable supramolecular nickel complexes was developed by combining two phosphorus ligands with Ni(0). These complexes stabilised by hydrogen bonding are directly active in the reaction of ethylene oligomerisation with some catalysts leading to high selectivity to 1-butene (up to 84%). To understand the origin of that selectivity, the scope of complexes was extended to ligands with different steric and electronic properties. Their evaluation in the reaction of ethylene oligomerisation evidenced a relation between the catalyst structure and the selectivity of the reaction. Mechanistic studies, under an ethylene atmosphere, reveals that cationic complexes rearrange to neutral complexes, which are likely, the active species.; Les alpha oléfines linéaires courtes sont des molécules de base en pétrochimie donc le marché est en constante augmentation notamment pour les oléfines légères (butène-1, hexène-1, octène-1). Ces oléfines, utilisées massivement dans l’industrie des plastiques, sont produites par la réaction catalytique d’oligomerisation de l’éthylène. IFPEN a contribué à développer plusieurs procédés homogènes d’oligomérisation de l’éthylène (AlphaButol, AlphaHexol, AlphaSelect) à base des complexes de titane, chrome ou zirconium. A travers une collaboration avec l’Université d’Amsterdam de nouvelles stratégies de développement de ligands ont été entreprises afin de rendre les catalyseurs à base de nickel sélectifs pour cette transformation. Ainsi, une approche supramoléculaire basée sur des interactions par liaison hydrogène, jusqu’alors décrite pour les métaux nobles, a été développée et appliquée aux complexes de nickel. Des complexes organométalliques originaux ont pu être générés et les interactions supramoléculaires ont été caractérisées par diffraction aux rayons X notamment. Ces complexes se présentent sous forme zwitterionique et sont formés par la combinaison de deux ligands simples donneurs et/ou accepteurs d’hydrogène. Ces complexes se sont avérés très actifs vis-à-vis de l’éthylène et ne nécessite pas l’ajout d’activateur du fait de la présence d’une liaison nickel-carbone réactive. Ils ont permis d’accéder à des sélectivités très importantes en butène-1. Appuyé par des expériences in situ et l’évaluation en catalyse de plusieurs complexes aux propriétés électroniques et stériques variées, cette approche a permis d’identifier l’espèce active et de mesurer l’impact de plusieurs descripteurs permettant de moduler la sélectivité et l’activité de la réaction catalytique en profondeur.

TRAIL (TNF- Related Apoptosis Inducing Ligand) and CD40Ligand, members of the superfamily (SF) of TNF (Tumor Necrosis Factor), appear as attractive targets for cancer therapy. A common feature of members of the TNF-SF is the homotrimeric structure that oligomerized their receptors to induce cell signaling. This multivalent interaction between ligands and receptors is crucial for apoptosis induction.The aims of this thesis is to i) develop multivalent ligands of TRAIL -R2 and CD40 to enhance their antitumor efficacy and to associate them in innovative strategies and ii) to characterize the interactions between ligands and receptors at cell surface to understand the impact of oligomerization on the initiation of apoptosis at membrane level.We used cyclic peptides specific of TRAIL-R2 (TRAILmim/DR5) that we multimerized on innovative chemical scaffolds. We also interested in the state of receptor oligomerization before their contact with the ligand in order to better understand the multivalent interactions. Finally, we characterized using the technique of surface plasmon resonance and biosensor based fluorescence interactions between multivalent ligands and TRAIL- R2.In this work we showed the need to analyze the interactions between ligands and receptors directly on the surface of the cell to improve the development of effective pro -apoptotic ligands., TRAIL (TNF-Related Apoptosis Inducing Ligand) et CD40 Ligand, membre de la superfamille (SF) du TNF (Tumor Necrosis Factor) apparaissent comme des cibles attractives pour la thérapie ciblée du cancer. Une caractéristique commune des membres de la SF-TNF est la structure homotrimérique du ligand qui oligomérise les récepteurs pour induire la signalisation cellulaire. Cette interaction multivalente entre ligands et récepteurs est déterminante dans l’induction de la réponse cellulaire. L’objectif de cette thèse est de i) développer des ligands multivalents de TRAIL-R2 et CD40 afin d’améliorer leur efficacité anti-tumorale puis de les associer dans des stratégies anti-tumorales innovantes et ii) de caractériser les interactions entre ces ligands et les récepteurs à la surface de la cellule pour comprendre l’impact de l’oligomerisation sur l’initiation de l’apoptose au niveau membranaire.Ainsi nous avons utilisé des peptides cycliques spécifiques du récepteur de TRAIL-R2 (TRAILmim/DR5) que nous avons multimérisés sur des plateformes chimiques innovantes. Nous nous sommes également intéressés à l’état d’oligomérisation des récepteurs avant leur contact avec le ligand dans le but de mieux comprendre les interactions multivalentes. Enfin, nous avons caractérisé à l’aide de la technique de résonance plasmonique de surface et d’un biocapteur basé sur la fluorescence les interactions entre ces ligands multivalents et le récepteur TRAIL-R2. Ces travaux nous ont donc montré la nécessité d’analyser les interactions entre ligands et récepteurs directement à la surface de la cellule pour améliorer le développement de ligands pro apoptotiques efficaces.

This work describes the synthesis and the structural study of linear and cyclic mixed oligomers alternating N-aminoamide and amide bonds, named 1:1-[a/a-N-amino]mers. The first chapter is a bibliographic study on cyclization methods of peptides and pseudopeptides (backbone modified peptides) and their applications. It has been described that the incorporation of potential structural elements, like introduction of changes to peptide backbone, could be facilitating the cyclization of linear oligomers. With this in mind, the use of previous work in our laboratory, discussed in the second chapter, gives access to new phtaloylated 1:1-[a/a-N-amino]mers and also to the first protected cyclo-1:1-[a/a-N-amino]mers. We describe the synthesis of a range of novel iminobisphosphine ligands based on a sulfonamido moiety [(R1SON)-N-2=P(R-2)(2)-P(R-3)(2)]. These molecules rearrange in the presence of nickel by metal-induced breakage of the P-P bond to yield symmetrical and nonsymmetrical diphosphinoamine nickel complexes of general formula Ni{[P(R-2)(2)]N(SO2R1)P(R-3)(2)}Br-2. The complexes can be isolated and are very stable. Upon activation by MAO, these complexes oligomerise ethylene to small chain oligomers (mainly C-4-C-8) with high productivity. Surprisingly fast codimerisation reactions of ethylene with butenes is observed, leading to a high content of branched C-6 products.

This work describes the synthesis and the structural study of linear and cyclic mixed oligomers alternating N-aminoamide and amide bonds, named 1:1-[a/a-N-amino]mers. The first chapter is a bibliographic study on cyclization methods of peptides and pseudopeptides (backbone modified peptides) and their applications. Svrha ovog rada je prikazati svojstva i djelovanje aluminija kao mogućeg etiološkog čimbenika u patofiziologiji Alzheimerove bolesti, s naglaskom na epidemiološke studije vode za piće i biokemijske i stanične pokazatelje dobivene kao rezultat animalnih i in vitro postupaka. Aluminij se prirodno nalazi u hrani. U industrijski prerađenoj hrani se nalazi u obliku aditiva, a u biljnoj apsorbiran iz tla gdje se prirodno nalazi. Aluminij se nalazi i u vodi za piće gdje dolazi kao prirodno otopljeni, ili kao dodatak pri pročišćavanju vode. Količina aluminija u vodi za piće čini do 5% ukupnog dnevnog unosa pa je moguće da u vodi postoje drugi čimbenici koji sprečavaju ili čak potiču povećanu apsorpciju aluminija. Mnogi dokazi upućuju na važnost oligomerizacije beta amiloidnog proteina i neurotoksičnosti u patofiziologiji Alzheimerove bolesti., The purpose of this paper is to show properties and activity of aluminium as a potential etiological factor in the pathophysiology of Alzheimer's disease, with a focus on the epidemiological studies on drinking water and biochemical and cellular parameters which are the result of animal and in vitro experiments. Aluminium is a content of food. In the processed foods it is used as an additive, while in the plant foods it is absorbed from soil, where it is naturally present. Aluminium is also a content of drinking water as a naturally dissolved element, or as an additive in water purification. The amount of aluminum in drinking water makes up to 5% of the total daily intake, so it is possible that in the water are other factors that hinder or even encourage increased absorption of aluminum. Water with dissolved silicates and fluoride reduces the absorption of aluminum in the digestive system. Some cosmetics contain aluminum, as well as vaccines. Antacids and buffered aspirin may contain aluminium. Although being enviromentally abundant, aluminium is not essential for life. It is a widely recognised neurotoxin that inhibits biological systems and causes various adverse effects. The relationship between aluminium exposure and Alzheimer's disease has been the subject of scientific debate for several decades. However, the complex characteristics of aluminium bioavailability make it difficult to evaluate and therefore, the relationship remains to be established. Mounting evidence has suggested that significance of oligomerization of beta-amyloid protein and neurotoxicity in the Alzheimer's disease pathophysiology.

Surfactant protein D (SP-D) is part of the innate immune system involved in lung homeostasis. SP-D knockout mice show accumulations of foamy alveolar macrophages, alveolar lipoproteinosis and pulmonary emphysema. Three single nucleotide polymorphisms (SNPs) have been described in the coding sequence of the human SP-D gene SFTPD. Clinical studies showed that the SNP SFTPD with a nucleotide change from A to C resulting in a Met to Thr substitution at position 11 in the protein (Met(11)Thr), is relevant. This study set out to create a humanised mouse model of the Met(11)Thr SNP. Transgenic mice lines expressing either Met(11) or Thr(11) SP-D under the control of the ubiquitously expressed pROSA26 promoter in C57Bl/6 SP-D deficient mice (DKO) was created. Both Met(11) (142 ± 52 ng mL(-1) ) and Thr(11) (228 ± 76 ng mL(-1) ) mice lines expressed human SP-D at almost similar levels. According to the literature this was within the range of SP-D levels found in wildtype (WT) mice (253 ± 22 ng mL(-1) ). Met(11) or Thr(11) SP-D in serum from transgenic mice bound maltose in a calcium-dependent manner, and binding was inhibited in the presence of EDTA or maltose. Bronchoalveolar lavage showed for both transgenic mice lines complementation of the DKO phenotype by restoring cell counts, phospholipid levels and protein content back to WT levels. Cytospins of BAL pellet cells showed a resemblance to WT but both mice lines showed some foamy alveolar macrophages. The stereological analysis showed for none of the mice lines a complete abrogation of emphysematous alterations. However, both Met(11) and Thr(11) mice lines were partially reverted back to a WT phenotype when compared with DKO mice, indicating important effects on surfactant metabolism in vivo.

Das Nukleoprotein der Arenaviren erfüllt zentrale Aufgaben im viralen Replikationszyklus. Dazu gehört die Enkapsidierung des viralen Genoms und die Regulation von Transkription und Replikation sowie die Interaktion mit dem Matrixprotein und die Inhibition der Typ I Interferon-Antwort. Über die Homo-Oligomerisierung des Nukleoproteins, ihre Bedeutung für diese vier Aufgaben sowie die hierfür notwendigen Domänen innerhalb des Nukleoproteins war bisher nichts bekannt. In der vorliegenden Arbeit wurde gezeigt, dass sich das Nukleoprotein in Abwesenheit viraler RNA zu symmetrischen, ringförmigen Trimeren zusammenlagert. Die Bildung dieser Trimere ist unabhängig von posttranslationalen Modifikationen und ist innerhalb der Arenaviren konserviert. Mutationsanalysen ergaben, dass Bereiche innerhalb der N- und der C-terminalen Domäne kritisch für die Trimerisierung des Nukleoproteins sind. Darüber hinaus wurde gezeigt, dass die Oligomerisierung des Nukleoproteins eine Grundvorraussetzung für seine Funktion bei der Replikation und Transkription des viralen Genoms ist. Im Gegensatz dazu spielt sie bei der Interaktion des Nukleoproteins mit dem Matrixprotein und für die Fähigkeit des Nukleoproteins zur Inhibition der Typ I Interferon-Antwort keine Rolle. Vermutlich wird das Nukleoprotein der Arenaviren durch Wirtszell-spezifische Proteasen verkürzt. Die dabei entstehenden, großen Spaltfragmente liegen als Monomere vor und könnten in der infizierten Zelle die Funktion als Interferon-Antagonist übernehmen, während ungespaltenes Nukleoprotein für die Replikation, Transkription und Enkapsidierung des viralen Genoms verwendet wird. Das Nukleoprotein sorgt durch seine Interaktion mit dem Matrixprotein für den Einbau des viralen Genoms in neu entstehende Virionen. In dieser Arbeit wurden die dafür verantwortlichen Aminosäurebereiche auf kurze Bereiche innerhalb der C-terminalen Proteindomäne des Nukleoproteins eingegrenzt. Anhand dieser Untersuchungen wurde ein Modell für die Interaktion zwischen dem Nukleoprotein und dem Matrixprotein an der Virusmembran aufgestellt. Vergleichende Studien zwischen den Nukleoproteinen und Matrixproteinen verschiedener Arenaviren ergaben zudem, dass heterotypische Interaktionen nur zwischen dem Nukleoprotein und dem Matrixprotein nahe verwandeter Arenaviren möglich sind. Die Interaktion zwischen dem Nukleoprotein und dem Matrixprotein wurde somit als Faktor identifiziert, der die die Bildung von Arenavirus-Reassortanten stark einschränkt. Insgesamt konnte in dieser Arbeit der Zusammenhang zwischen der Homo-Oligomerisierung und den biologischen Funktionen des Nukleoproteins aufgezeigt werden. Darüber hinaus wurden funktionelle Domänen für die NP-NP-Interaktion und die Interaktion zwischen dem Nukleoprotein und dem Matrixprotein innerhalb des Nukleoproteins identifiziert.

Rutin and esculin have been polymerized by laccase. Five fractions with between 2127.42 and 8331.85 g/mol for oligorutins, and between 688.12 and 6973 g/mol for oligoesculins, were obtained. Fourier transformed infrared analysis showed that oligorutins were formed through C-C, C-O and C=O linkages, while oligoesculins were obtained through C-C linkages. Oligorutins and oligoesculins show a higher solubility in water and a lower solubility in ethanol compared to their monomers. The oligomerization of rutin decrease its antiradical capacity, while oligoesculin fractions demonstrated a high antiradical activity compared to monomeric esculin. Oligomer fractions showed a better iron chelating power, xanthine oxidase inhibition, copper reducing power (CUPRAC), antigenotoxic activity, and splenocytes stimulator activity compared to their monomers. Oligorutin and oligoesculin exhibited an important anti-inflammatory capacity through the nitric oxide inhibition. Moreover, oligorutin fractions demonstrated an immunostimulatory effect proportional to their degree of oligomerization, while oligoesculin fractions showed an immunostimulatory effect inversely proportional to their degree of oligomerization, L'oligomérisation enzymatique de la rutine et esculine a donné lieu à cinq fractions d'oligomères de masse moléculaire moyenne entre 2127,42 et 8331,85 g/mol pour la rutine et 688,12 et 6973 g/mol pour l'esculine. L'analyse de ces fractions par FTIR montre que les fractions d'oligorutines sont obtenues à travers des liaisons C-C, C-O et C=O. Une diminution de l'activité antiradicalaire vis-à-vis de DPPH., ABTS+. et OH. proportionnelle à la masse moléculaire moyenne des fractions d'oligorutines a été observé, contrairement aux fractions d'oligoesculines qui montrent un important pouvoir chélateur de ces mêmes radicaux comparé à leurs monomère. Une augmentation du pouvoir chélateur de fer, inhibiteur de la xanthine oxydase, réducteur du cuivre (CUPRAC), de l'activité antigénotoxique, ainsi que de l'activité stimulatrice de la prolifération des splénocytes, et des lymphocytes (B et T) proportionnelle au degré d'oligomérisation des oligomères étudiées a été noté. L'effet des fractions d'oligorutines et oligoesculines étudiées sur les macrophages en suivant la production de monoxyde d'azote (NO) montre un pouvoir anti-inflammatoire comparé à leurs monomères. L'étude de l'activité lysosomale induite par les fractions d'oligorutine révèle un pouvoir immunostimulateur proportionnelle à la masse moléculaire moyenne des oligorutines, et inversement proportionnelle à celle-ci pour les oligoesculines

X-linked Adrenoleukodystrophy (X-ALD) is a rare neurodegenerative disease caused by deficiency of the peroxisomal half-transporter ABCD1, implicated in very long chain fatty acids import. Two additional half-transporters are located in the peroxisomal membrane: ABCD2 and ABCD3. Over-expression of ABCD2 is known to compensate for ABCD1 deficiency, making ABCD2 a therapeutic target for X-ALD treatment. In this context, the main objective of my thesis was to investigate the function and the structure of ABCD2, and more broadly, of peroxisomal ABC transporters.Half-transporters must at least dimerize to form a functional transporter. Alternative dimerization could modulate substrate specificity. In order to test this hypothesis, we engineered plasmidic constructs encoding chimeric ABCD dimers, whose functionality has been evaluated by transient transfection in two cell models (human fibroblasts and yeasts). Our results show that, ABCD1 and ABCD2 are functional whatever their dimeric organization. Besides, like other ABC transporters, peroxisomal ABC transporters could oligomerize. By using a multi-technical approach (co-immunoprecipitation, velocity sucrose gradient and native polyacrylamide gel electrophoresis experiments) on stably transfected hepatoma cells expressing ABCD2-EGFP, we demonstrate that ABCD2-EGFP interacts with ABCD1 and ABCD3, and that peroxisomal ABC transporters oligomerize. The perspectives will consist in determining which factors control the oligomerization process and understanding the functional value of these interactions., L’Adrénoleucodystrophie liée à l’X est une maladie neurodégénérative rare due à des mutations dans le gène ABCD1. Ce gène code un demi-transporteur ABC peroxysomal, impliqué dans l’importation d’acides gras à très longue chaîne. Deux autres demi-transporteurs sont localisés dans la membrane peroxysomale : ABCD2 et ABCD3. La surexpression d’ABCD2 permet de compenser la déficience en ABCD1, ouvrant ainsi des perspectives thérapeutiques. Dans cette optique, l’objectif principal de ma thèse était d’étudier la fonction et la structure d’ABCD2, et plus largement des transporteurs ABC peroxysomaux.Les demi-transporteurs doivent au minimum se dimériser pour constituer un transporteur fonctionnel. Leur dimérisation alternative pourrait moduler leur spécificité de substrat. Afin de tester cette hypothèse, nous avons réalisé des constructions plasmidiques codant différents dimères chimériques, dont la fonctionnalité a été vérifiée par transfection transitoire dans deux modèles cellulaires (fibroblastes humains et levures). D’après nos résultats, ABCD1 et ABCD2 seraient fonctionnels quel que soit leur agencement dimérique. De plus, comme d’autres transporteurs ABC, les transporteurs ABC peroxysomaux pourraient s’oligomériser. En utilisant différentes techniques biochimiques (co-immunoprécipitation, sédimentation sur gradient de sucrose et électrophorèse en conditions natives), sur un modèle cellulaire surexprimant ABCD2-EGFP, nous démontrons qu’ABCD2-EGFP interagit avec ABCD1 et ABCD3, et que les transporteurs ABC peroxysomaux sont capables de s’oligomériser. Il reste désormais à déterminer les facteurs qui contrôlent cette oligomérisation et comprendre la valeur fonctionnelle de ces interactions.

Expression profiling of the three homodimeric (proto-type) chicken galectins (CG-1A, CG-1B and CG-2) has raised evidence for distinct functionalities, explaining the interest in a detailed crystallographic analysis of CG-2. As revealed here, marked differences are found in the ligand-binding site and in the contact pattern within the homodimer interface, underlying a characteristic orientation of the two subunits. Notably, a distinctive trimer of dimers that is unique in all galectin crystal structures reported to date forms the core unit of the crystallographic assembly. Combination with spectroscopic and thermodynamic measurements, and comparisons with CG-1A and CG-1B, identifies differential changes in circular-dichroism spectra in the presence of lactose, reflecting the far-reaching impact of the ligand on hydrodynamic behavior, and inter-galectin differences in both the entropy and the enthalpy of binding. Summary The integration of transmembrane (TM)-spanning regions of many channels and ion transporters is potentially compromised by the presence of polar and charged residues required for biological function. Although the two TMs of the ATP-gated ion channel subunit P2X2 each contain charged/polar amino acids, we found that each TM is efficiently membrane inserted when it is analysed in isolation, and uncovered no evidence for cooperativity between these two TMs during P2X2 integration. However, using minimal N-glycosylation distance mapping, we find that the positioning of TM2 in newly synthesized P2X2 monomers is distinct from that seen in subunits of the high-resolution structures of assembled homologous trimers. We conclude that P2X2 monomers are initially synthesised at the endoplasmic reticulum in a distinct conformation, where the extent of the TM-spanning regions is primarily defined by the thermodynamic cost of their membrane integration at the Sec61 translocon. In this model, TM2 of P2X2 subsequently undergoes a process of positional editing within the membrane that correlates with trimerisation of the monomer, a process requiring specific polar/charged residues in both TM1 and TM2. We postulate that the assembly process offsets any energetic cost of relocating TM2, and find evidence that positional editing of TM2 in the acid-sensing ion channel (ASIC1a) is even more pronounced than that observed for P2X2. Taken together, these data further underline the potential complexities involved in accurately predicting TM domains. We propose that the orchestrated repositioning of TM segments during subunit oligomerisation plays an important role in generating the functional architecture of active ion channels, and suggest that the regulation of this underappreciated biosynthetic step may provide an elegant mechanism for maintaining ER homeostasis.

This thesis describes the development of new catalytic systems based upon iron complexes and their reactivity toward ethylene. First, we focused our interest on the synthesis of iron(III) precursors chelated by monoanionic ligand. Those complexes were obtained either by reaction of the monoanionic ligand with FeCl3 or through oxidation of the iron(II) complex. The second reaction led to binuclear complexes. Then, another aim of the thesis was to design new well-defined cocatalysts for the activation of iron complexes. The study of the reaction between an alcohol and the trimethylaluminum allowed us to reach this aim. Aluminum complexes adopted either a binuclear framework or a trinuclear one, depending on the nature of alcohol reagent. Besides this work, new iron(II) and nickel(II) complexes chelated by imino-imidazole ligands bearing a pendant donor function L were synthesized. All complexes have been evaluated for the oligomerization of ethylene in the presence of EtAlCl2 or MAO as cocatalyst. Only nickel complexes were active toward ethylene transformation.; Cette thèse décrit le développement de nouveaux systèmes catalytiques à base de fer ainsi que l'étude de leur réactivité vis-à-vis de l'éthylène. Dans un premier temps, nous nous sommes intéressés au développement de précurseurs de fer(III) associés à des ligands monoanioniques tridentes. Deux voies de synthèse ont été envisagées. La première décrit la complexation d'un ligand anionique sur le précurseur FeCl3 et la seconde passe par l’oxydation d'un complexe de fer(II) associé à un ligand neutre conduisant à une espèce binucléaire. Activés par le MAO, ces catalyseurs de fer(III) constituent les premiers complexes du genre permettant l’oligomérisation de l'éthylène. L’accent a également été porté sur la recherche de nouveaux activateurs. Des complexes d’aluminium répondant à nos attentes ont été obtenus par réaction entre un alcool et le triméthylaluminium. Selon la nature de l'alcool, la structure des activateurs peut être soit binucléaire ou trinucléaire. Enfin, des complexes de fer et de nickel associés à des ligands imino-imidazoles possédant un bras hémilabile ont été synthétisés. Une fois activés, les systèmes à base de nickel ont montré de bonnes activités en catalyse.

Genome sequencing data has revealed genes encoding uncharacterized protein family UPF0079 which are exclusively found in bacteria; broadly distributed in this kingdom and possess an ATP-binding motif in their sequences. Biochemical characterization and physiological role elucidation of UPF0079 will undoubtedly increase our fundamental biology knowledge, and also remain a prerequisite towards the development of new antimicrobial compounds. Our investigation on YdiB, an archetype of this family in Bacillus subtilis revealed both autophosphorylating and protein phosphotransferase activities. The dual-specificity Ser/Thr and Tyr kinase activity of YdiB seems to require oligomerization is upregulated by basic molecule activators such as natural polyamines or poly-L-lysine. The 10 most conserved residues were studied to gain insights into molecular mechanism of the kinase YdiB. To characterize the function of phosphorylation events linked to YdiB, starting with the B. subtilis ydiA-B-C-D-E operon we showed that YdiB and YdiC function as cognate protein kinase/phosphatase towards two ribosome-related protein substrates YdiD and YdiE. Some co-localization between YdiB and ribosomes were observed. Furthermore, YdiB is capable of phosphorylating both ribosomal 50S and 30S subunits as well as two ribosome-binding GTPases EngA and EngB. We also demonstrated that phosphorylated EngA by YdiB is an in vitro substrate of the phosphatase YdiC. Finally, based on the phosphoproteome pf Bacillus subtilis, peptides mimicking the in vivo phosphorylation sites were used. Some of them were found to be phosphorylated in vitro by YdiB, including two peptides which belongs to the superoxide dismutase SodA. The activity of purified SodA was then shown to be upregulated via phosphorylation by YdiB. We furthermore found that B. subtilis cells lacking ydiB become more sensitive to oxidative stress-causing agents such as paraquat or norfloxacin. We propose that in vivo, YdiB functions as a protein kinase involved in ribosome function in normal condition; and in protecting cells from oxidative stress damage., Les données de séquençage des génomes ont révélé une nouvelle famille de protéines UPF0079, comprenant des protéines de fonction inconnue qui sont exclusivement et largement présentes chez les bactéries et qui possèdent un motif A de Walker dans leur séquence. La caractérisation biochimique et l'élucidation du rôle physiologique de cette famille contribueront à élargir nos connaissances en biologie fondamentale, et sont également un préalable vers le développement de nouveaux composés antimicrobiens. Notre étude sur YdiB, un archétype de cette famille chez Bacillus subtilis a révélé à la fois l‟autophosphorylation de YdiB et son activité de protéine kinase. L‟activité kinase de double spécificité Ser/ Thr et Tyr de YdiB semble nécessiter son oligomérisation et semble être stimulée par des molécules basiques telles que des polyamines naturelles ou la poly-L-lysine. Les 10 résidus les plus conservés chez cette famille ont été étudiés afin de mieux comprendre le mécanisme moléculaire de YdiB. Concernant la caractérisation fonctionnelle de la phosphorylation liée à YdiB, l‟étude de l‟opéron ydiA-B-C-D-E de B. subtilis nous a permis de montrer que YdiB et YdiC fonctionnent comme un couple de protéine kinase/phosphatase de deux protéines substrats dont les fonctions seraient liées aux ribosomes, YdiD et YdiE. Une co-localisation partielle entre YdiB et les ribosomes a été observée. En outre, YdiB est capable de phosphoryler des protéines ribosomiques appartennant aux deux sous-unités 50S et 30S, ainsi que deux GTPases impliquées dans la biogénèse des ribosomes, EngA et EngB. Nous avons également démontré que EngA phosphorylée par YdiB est un substrat in vitro de la phosphatase YdiC. Enfin, basé sur le phosphoprotéome de Bacillus subtilis, des peptides mimant des sites de phosphorylation in vivo ont été utilisés. Certains entre eux sont phosphorylés in vitro par YdiB. Deux de ces peptides appartiennent à la superoxyde dismutase, SodA, dont l'activité in vitro et après purification est régulée positivement via la phosphorylation par YdiB. Nous avons ensuite constaté que les cellules de B. subtilis dépourvues du gène ydiB sont plus sensibles aux agents oxidants tels que le paraquat ou la norfloxacine. Nous proposons que, in vivo, YdiB fonctionne comme une protéine kinase impliquée dans l‟activité et/ou la stabilité des ribosomes dans des conditions physiologiques normales, et YdiB contribuerait à protéger les cellules contre les dommages du stress oxydatif.

This thesis describes the development of new catalytic systems based upon iron complexes and their reactivity toward ethylene. First, we focused our interest on the synthesis of iron(III) precursors chelated by monoanionic ligand. Those complexes were obtained either by reaction of the monoanionic ligand with FeCl3 or through oxidation of the iron(II) complex. The second reaction led to binuclear complexes. Then, another aim of the thesis was to design new well-defined cocatalysts for the activation of iron complexes. The study of the reaction between an alcohol and the trimethylaluminum allowed us to reach this aim. Aluminum complexes adopted either a binuclear framework or a trinuclear one, depending on the nature of alcohol reagent. Besides this work, new iron(II) and nickel(II) complexes chelated by imino-imidazole ligands bearing a pendant donor function L were synthesized. All complexes have been evaluated for the oligomerization of ethylene in the presence of EtAlCl2 or MAO as cocatalyst. Only nickel complexes were active toward ethylene transformation., Cette thèse décrit le développement de nouveaux systèmes catalytiques à base de fer ainsi que l'étude de leur réactivité vis-à-vis de l'éthylène. Dans un premier temps, nous nous sommes intéressés au développement de précurseurs de fer(III) associés à des ligands monoanioniques tridentes. Deux voies de synthèse ont été envisagées. La première décrit la complexation d'un ligand anionique sur le précurseur FeCl3 et la seconde passe par l’oxydation d'un complexe de fer(II) associé à un ligand neutre conduisant à une espèce binucléaire. Activés par le MAO, ces catalyseurs de fer(III) constituent les premiers complexes du genre permettant l’oligomérisation de l'éthylène. L’accent a également été porté sur la recherche de nouveaux activateurs. Des complexes d’aluminium répondant à nos attentes ont été obtenus par réaction entre un alcool et le triméthylaluminium. Selon la nature de l'alcool, la structure des activateurs peut être soit binucléaire ou trinucléaire. Enfin, des complexes de fer et de nickel associés à des ligands imino-imidazoles possédant un bras hémilabile ont été synthétisés. Une fois activés, les systèmes à base de nickel ont montré de bonnes activités en catalyse.

Bacteria use a communication mode named quorum sensing to regulate gene expression depending on the population density and thus to control processes such as sporulation, competence or virulence in a multicellular manner. In Gram-positive bacteria, the quorum sensing relies mostly on the production, the secretion and the detection of small signaling peptides. The project focuses on the study of the quorum sensing system NprR/NprX in Bacillus cereus, where NprR is the effector, which recognizes specifically the signaling peptide NprX. NprR is a bi-functional protein. In the absence of peptide, it acts as a sporulation inhibitor while in complex with NprX, it acts as transcription factor implicated in virulence. NprR belongs to a family of quorum sensing effectors named RNPP (for the first identified members: Rap, NprR, PlcR and PrgX) still not well characterized at a structural level. My PhD project consisted to perform the structure/function analysis of the NprR/NprX system. To understand the functional regulation of NprR by NprX, I carried out different studies in solution (SEC-MALS and DLS). These results allowed me to highlight a molecular switch based on a changing of the oligomerisation state of the protein. NprX binding switches NprR from an Apo dimeric conformation to a tetrameric complex. The structural study by crystallography led to the resolution of the tetrameric NprR/NprX complex structure. The analysis of this tetramer suggests the recognition of 2 DNA binding sites. The structural study of the dimeric conformation of Apo NprR by SAXS, allowed me to propose a model similar to that of the Rap dimers (members of RNPP family). This model is supported by a directed mutagenesis study of interface residues. The characterization by ITC of the NprR/NprX interaction with different forms of the peptide, as well as the analysis of the binding pocket in the complex, led to a better understanding of the specificity of the interaction. Two key residues of the effector were highlighted: Asn275, essential to peptide binding and Arg126, essential to the activation of the transcription factor function. These results have contributed to a better understanding of the NprR/NprX quorum sensing system thanks to the elucidation of the molecular switch controlled by NprX but also in a better knowledge of the RNPP effectors family., Les bactéries utilisent un mode de communication appelé quorum sensing pour régulerl’expression des gènes en fonction de la densité de population et contrôler ainsi de façonmulticellulaire des processus tels que la sporulation, la compétence ou la virulence. International audience; G-protein-coupled receptor homo-oligomerization has been increasingly reported. However, little is known regarding the relationship between activation of the receptor and its associa- tion/conformationalstates.Themammalianolfactoryreceptors (ORs) belong to the G protein-coupled receptor superfamily. In this study, the homo-oligomerization status of the human OR1740 receptor and its involvement in receptor activation upon odorant ligand binding were addressed by co-immuno- precipitationandbioluminescenceresonanceenergytransfer approaches using crude membranes or membranes from dif- ferent cellular compartments. For the first time, our data clearly show that mammalian ORs constitutively self-associ- ate into homodimers at the plasma membrane level. This study also demonstrates that ligand binding mediates a con- formational change and promotes an inactive state of the OR dimers at high ligand concentrations. These findings support andvalidateourpreviouslyproposedmodelofORactivation/ inactivation based on the tripartite odorant-binding protein- odorant-OR partnership.

Dans l’étude des GPCRs, il a longtemps été considéré que ceux-ci étaient présents à la membrane sous forme de monomères et que chaque monomère pouvait être activé par un ligand et transmettre le signal grâce à une protéine G. Certains résultats comme des courbes de compétition ou d’activation à deux phases étaient difficilement explicables avec ce modèle 1 ligand, 1 récepteur, 1 protéine G.Ces dernières années, il a été montré via différentes techniques que les GPCRs étaient présents à la membrane non pas sous forme de monomères mais plutôt de dimères, de rangées de dimères voire de multimères. Cette disposition permet plus facilement d’expliquer certains résultats mais complique considérablement le modèle. Un nombre important de nouvelles questions sont posées quant au fonctionnement d’une telle formation de récepteurs. Quelle est l’unité fonctionnelle ?Y a-t-il une communication entre les récepteurs qui forment ces oligomères ?Est-ce qu’un ligand peut activer plusieurs récepteurs, plusieurs protéines G ?Que se passe-t-il quand l’oligomérisation se fait entre des récepteurs différents ?Est-ce qu’un ligand d’un récepteur peut en activer un autre ?Est-ce que la voie d’activation change en fonction du ligand ou du nombre de molécules liées sur un homomère ou un hétéromère? Toutes ces questions sont à prendre en considération lors de l’étude pharmacologique de n’importe quelle drogue car si le récepteur cible hétéromérise avec un autre, la drogue étudiée ne pourrait plus avoir le même effet. Prendre en considération tous ces paramètres peut compliquer considérablement l’étude d’une drogue mais elle pourrait en prévoir précisément certains effets secondaires.Dans cette thèse nous étudions la question de la communication par la coopérativité négative (effet négatif sur l’affinité d’un site de liaison lors de la liaison du ligand sur un l’autre site du dimère). Nous mettons en évidence un lien entre l’activité constitutive d’un récepteur et le niveau de coopérativité négative grâce à des expériences d’accélération de dissociation de ligand traceur. La coopérativité négative observée sur les mutants du récepteur à la TSH les plus constitutifs est quasiment nulle. Nous montrons que cette perte de coopérativité négative provoque un changement de stœchiométrie ligand/récepteur et qu’au lieu de lier un seul ligand par dimère, les mutants les plus constitutifs sont capables d’en lier deux.Malgré nos efforts pour identifier le ou les domaines de communication entre les récepteurs nous n’avons pu les déterminer. Die Bildung infektiöser Viruspartikel des humanen Zytomegalievirus (HCMV) ist ein mehr-stufiger Prozess. Sie beginnt mit der Verpackung der DNA in die Kapside im Kern, gefolgt von weiterer Reifung während des Transports durch das Zytoplasma und der abschließenden Freisetzung aus der Zelle. Im Zuge dieser Arbeit wurden zwei Proteine, die Einfluss auf die ebengenannten Prozesse haben, analysiert. Der erste Teil der Arbeit befasst sich mit der funktionellen Charakterisierung des HCMV Pro-teins pUL77. Es ist bekannt, dass das homologe Protein pUL25 in alpha-Herpesvirinae essentiell für die DNA-Verpackung ist. Zunächst konnte das Protein als Kapsid-assoziiertes strukturelles Protein identifiziert werden. Es wurden Interaktionen von pUL77 mit DNA-Verpackungs- und Kapsidproteinen gezeigt. Weiterhin wurde die DNA-Bindungsfähigkeit von pUL77 in verschiedenen „in vitro“-Experimenten untersucht. Zusammengefasst weisen unsere Ergebnisse auf eine Funktion von HCMV pUL77 bei der DNA-Verpackung hin. Im zweiten Teil der Arbeit wurde das HCMV Protein pUL71 charakterisiert, das in allen Herpesviren konserviert vorkommt, dessen Funktion jedoch nicht charakterisiert ist. Zunächst wurde das Protein als strukturelles Tegumentprotein mit “earlylate“ Expressionskinetik klassifiziert. Weiterhin wurden die subzelluläre Lokalisation sowie virale und zelluläre Interaktionspartner untersucht. Die Ergebnisse weisen auf eine Funktion von HCMV pUL71 bei der Reifung und beim Transport der Virionen im Zytoplasma hin. „In silico“-Vorhersagen zeigten ein „Leuzin Zipper“-Motiv in pUL71, das als mögliche Oligomerisationsdomäne dienen könnte. Mutationen wurden in dieses Motiv eingebracht und die resultierenden Proteine auf ihre Oligomerisationsfähigkeit mit „in vitro“-Methoden und in rekombinanten Viren untersucht. Zusammenfassend konnten wir zeigen, dass das „Leuzin Zipper“-Motiv wichtig für die Funktion von pUL71 ist und diese mit einer unbeeinträchtigten Oligomerisation des Proteins zusammen hängt. The morphogenesis of Human cytomegalovirus (HCMV) virions starts with the capsid assem-bly and DNA insertion in the nucleus followed by maturation during transport through the cytoplasm prior to release of virus progeny. In this study we are functionally characterising two proteins that are involved in those steps. The function of essential HCMV protein pUL77 is characterised in the first part of the study. HCMV pUL77 was shown to be a structural protein associated with capsids. Furthermore, our experiments demonstrated that HCMV pUL77 interacts with DNA packaging motor compo-nents and capsid proteins. The ability of HCMV pUL77 to bind double-stranded DNA was studied in “in vitro” assays designed for this study. The homologue α-Herpesvirinae protein pUL25 is described to be involved in processes connected with DNA packaging. Data ob-tained in this study demonstrates that HCMV pUL77 might serve a similar function. In the second part of the study HCMV pUL71, conserved throughout the Herpesvirus family but to date unclassified, was functionally characterised. HCMV pUL71 was defined a struc-tural tegument protein with early-late expression kinetics. We studied the sub-cellular local-isation and interactions of pUL71 with a subset of cellular and viral proteins. Thereby we could show that HCMV pUL71 function might be connected with processes of viral egress. By in silico analyses we identified a leucine zipper motif in pUL71 that might serve as a puta-tive oligomerisation domain. In order to investigate the function of the leucine zipper motif, we performed in vitro assays and investigated the alterations of the motif in the viral context. Taken together we can conclude that (i) an intact leucine zipper motif is crucial for the func-tion of pUL71 and (ii) this function is dependent upon undisturbed oligomerisation of the pro-tein.

Das HIV-1-Protein p6 ist im C-terminalen Bereich von Gag lokalisiert und besitzt trotz seiner geringen Größe von nur 52 Aminosäuren eine komplexe strukturelle und funktionelle Organisation. Es bindet mindestens zwei zelluläre Buddingfaktoren (Tsg101 und ALIX) sowie zwei virale Proteine (Vpr und Env) und übt eine Vielzahl biologischer Funktionen aus. Außerdem besitzt p6 sowohl C- als auch N-terminal jeweils eine L-Domäne, welche unter anderem den letzten Schritt in der Abschnürung des naszierenden Virions von der Zellmembran, in einem bis heute noch nicht vollständig geklärten Mechanismus, regulieren. Neben den Modifikationen der Ubiquitinylierung und Sumoylierung fungiert p6 außerdem als Substrat für verschiedene Kinasen: Im Jahr 2002 wurde p6 als Hauptphosphoprotein in HIV-1-Partikeln identifiziert und es konnte gezeigt werden, dass p6 durch die MAP-Kinase ERK-2 an der Position Thr-23 phosphoryliert wird. Weiter gibt es Hinweise, dass die genannte Kinase durch die Phosphorylierung an Thr-23 von p6 den Viruszusammenbau sowie die Virusfreisetzung reguliert. Zur Identifikation weiterer potentieller Kinasen wurde zu Beginn dieser Arbeit eine in silico Analyse durchgeführt und schließlich mittels in vitro Experimenten mit synthetischem und viralem p6-Peptid die Proteinkinase C, welche spezifisch Ser-Reste an Position 14, 25 und 40 in p6 phosphoryliert, identifiziert. Vergleicht man die Konsensussequenzen verschiedener HIV-1-Isolate der M-Hauptgruppe, so ist eine absolute Konservierung von Ser-40 festzustellen, was auf eine mögliche biologische Funktion dieser Stelle hindeutet. Zur Untersuchung der biologischen Funktion der Phosphorylierungsstellen in p6 wurden Konstrukte kloniert, die an den identifizierten Ser-Resten mutiert wurden. Die Substitution aller Ser-Reste einzeln zeigte in Infektionsstudien, dass nur die Mutation von Ser-40 zu einer verminderten Replikationskapazität in T-Zellen sowie in primären humanen Zellen führte. Die Mutation von Ser-14 und Ser-25 zeigte keinen Effekt auf die Replikation von HIV-1. Die Mutation aller drei PKC-Stellen zusammen führte hingegen zu einem kompletten Verlust der Replikationsfähigkeit. Obwohl die Mutation der Phosphoakzeptorstellen die Freisetzung der Viruspartikel nicht beeinflusste, sind die produzierten Virionen der Ser-40-Mutante weniger infektiös und die Dreifachmutation führte sogar zu einem kompletten Verlust der Infektiösität. Als molekularer Mechanismus hinter diesem Phänomen konnte in Pulse-Chase Experimenten beobachtet werden, dass die CA-Prozessierung von p25 zu p24 im Fall der Ser-40-Mutante gestört ist. Elektronenmikroskopische Aufnahmen zeigten, dass der Defekt in der CA-Prozessierung zu einer irregulären Core-Morphologie und zur Bildung einer elektronendichten Struktur außerhalb des Cores führt. Die erhaltenen Ergebnisse belegen eine neue, bis heute unbekannte Funktion von p6, die nicht die Virusfreisetzung, sondern die Gag-Prozessierung betrifft. Das konservierte Ser-40 reguliert auf noch unbekannte Weise, womöglich durch PKC-Phosphorylierung, die CA-Reifung und somit die Infektiösität von HIV-1. Neben den verschiedenen Bindungsstellen für virale und zelluläre Proteine sowie den Modifikationsstellen besitzt p6 die inhärente Fähigkeit zur Ausbildung oligomerer Formen. Diese wurden durch die Behandlung von sp6 und vp6 mit chemischen Crosslinkern und anschließender Western Blot Analyse untersucht. Es zeigte sich, dass das C-terminale Fragment von p6 oligomere Strukturen ausbildet. Somit konnte der in silico Befund, dass p6 eine im C-terminalen Bereich lokalisierte Oligomerisierungsdomäne besitzt, bestätigt werden. Ob diese Oligomerisierungsdomäne in p6 zur Gag-Gag-Multimerisierung, welcher eine wichtige Rolle in der Gag-Assemblierung und der Produktion von Viruspartikeln zukommt, beiträgt, sie eine ganz andere funktionelle Relevanz hat oder gar funktionslos ist, ist bislang nicht bekannt. The HIV-1 protein is located at the C-terminus of Gag and despite its small size of 52 amino acids it comprises a quite complex structural and functional organization: it binds at least two cellular budding factors (Tsg101 and ALIX) as well as two viral proteins (Vpr and Env) and it exerts a variety of biological functions. Among others, it regulates the final abscission step of the nascent virion from the cell membrane by the action of two different l domains, one located at the C- and one located at the N-terminus of the peptide, in a yet not completely elucidated mechanism. Besides the ubiquitinylation and sumoylation p6 acts as a substrate for distinct kinases: 2002 p6 was identified as the major phosphoprotein found in HIV-1 particles and it could be shown that phosphorylation of p6 by MAP-kinase ERK-2 at position Thr-23 regulates the assembly and release of progeny virions. To identify further potential kinases an in silico analysis was performed and finally in vitro experiments with synthetic and viral p6 identified PKC to specifically phosphorylate Ser residues in position 14, 25 and 40 in p6. When the consensus sequences derived from various HIV-1 group M isolates were compared, one can see that Ser-40 is absolutely conserved. Therefore, the biological function of this site has to be identified. To analyze the biological function of the identified phosphorylation sites, different constructs carrying mutations of the identified Ser residues were generated. Individual substitutions of the Ser residues revealed that only mutation of Ser-40 reduced virus replication in T-cell lines and in primary human cells, while mutation of Ser-14 and Ser-25 had no effect on the replication. In contrast, mutation of all three PKC-sites completely abrogated replication. Although mutation of PKC sites had no influence on virus release, the infectivity was reduced for the Ser-40 mutant and completely lost for the triple mutant. As molecular mechanism behind this phenomenon pulse-chase experiments illustrated that the efficiency in final processing of CA from p25 to p24 was dependent on the integrity of Ser-40. Electron micrographs showed that the defect in CA processing led to an irregular morphology of the virus core and the formation of an electron dense extra core structure. Thus, the obtained results support a novel, till this day unknown function of p6, that does not affect virus release but concerns Gag processing. The conserved Ser-40 regulates in a yet unknown manner CA maturation and thereby infectivity of HIV-1 in a process that might involve PKC phosphorylation. In addition to the different binding sites for viral and cellular proteins and the modification sites p6 displays the inherent ability to undergo oligomerization. This observation was analyzed by incubating sp6 and vp6 with chemical crosslinkers and western blotting. It turned out that the C-terminal fragment of p6 has the propensity to form oligomeric structures. This is consistent with in silico analysis, which predicts an oligomerization domain in the C-terminus of p6. Whether this oligomerization domain in p6 contributes to the Gag-Gag multimerization, which has an important role in Gag assembly and virus particle production, or whether it has a complete different functional relevance or it is totally out of any function is previously not known.

The classical NF-kappaB-signaling is not only one uniform pathway but consists of several distinct signaling pathways which differ regarding their inducing agents, the intracellular proteins involved in signal transmission, the kind of activated NF-kappaB dimers and the set of induced target genes. However, the precise molecular mechanisms by which these differential NF-kappaB activation patterns are determined still remain largely unknown. The key regulator within the classical NF-kappaB-signaling cascades is the IKK-complex. Since structural alterations in the IKK-complex are thought to participate in the regulation of its kinase activity, protein-protein-interactions within the IKK-complex were characterized. The IBD in NEMO has been shown to mediate the interaction with the IKKs. However, our studies reveal that the IBD additionally constitutes an independent oligomerization interface. Interestingly, the IBD possesses several phosphorylation sites. Yet, the physiological functions are still largely unknown. IKKß-mediated phosphorylation of Ser68 in the IBD caused a reduced NEMO-IKKß interaction and NEMO oligomerization. The negative effect of the Ser68-phosphorylation on the classical NF-kappaB-signaling pathways underline the importance of phosphatases and specific dephosphorylation events for the classical NF-kappaB activation. Therefore the yet poorly defined role of the protein phosphatase calcineurin in the TCR-induced NF-kappaB activation was analyzed. Our results point out that calcineurin acts as a BCL10 phosphatase that removes inhibitory phosphate groups from BCL10 in the early activation state. Therefore a molecular basis of how calcium influences the TCR-induced NF-kiappaB-signaling pathway is the calcineurin-mediated regulation of the CBM-complex. In summary these data underscore the importance of negatively regulating phosphorylations, which are necessary to avoid an unwanted chronic NF-kappaB activation and highlight the relevance of phosphatases in the process of IKK activation.

International audience; Rotavirus is one of the leading agents of gastroenteritis worldwide. During infection, viral factories (viroplasms) are formed. The rotavirus nonstructural proteins NSP5 and NSP2 are the major building blocks of viroplasms; however, NSP5 function and organisation remain elusive. In this report, we present a structural characterisation of NSP5. Multi-angle laser light scattering, sedimentation velocity and equilibrium sedimentation experiments demonstrate that recombinant full-length NSP5 forms a decamer in solution. Far-Western, pull-down and multi-angle laser light scattering experiments show that NSP5 has two oligomerisation regions. The first region, residues 103–146, is involved in NSP5 dimerisation, whereas the second region, residues 189–198, is responsible for NSP5 decamerisation. Circular dichroism analyses of full-length and truncated forms of NSP5 reveal that the decamerisation region is helical, whereas the dimerisation region involves β-sheets. From these circular dichroism experiments, we also show that the NSP5 protomers contain two α-helices, a disordered N-terminal half and a C-terminal half that is primarily composed of β-sheet folds. This extensive structural characterisation of NSP5 led us to propose a model for its quaternary organisation. Finally, co-expression of NSP5 fragments and NSP2 in uninfected cells shows that the NSP5 decamerisation region is required for viroplasm-like structure formation. However, in vitro, the NSP5 decamerisation region partially inhibits the NSP2–NSP5 interaction. Our NSP5 model suggests that steric hindrance prevents NSP2 from binding to all NSP5 protomers. Some protomers may thus be free to interact with other NSP5 binding partners, such as viral RNAs and the viral polymerase VP1, to perform functions other than viroplasm organisation.

Diese Arbeit befasste sich mit der Fragestellung, wie durch gezielte Modifikation der Proteinfaltung die virale Replikation beeinflusst werden kann. Durch Jahrmillionen der Koevolution entwickelten Viren eine Abhängigkeit von Mechanismen der Wirtszellen. Zelluläre Proteine stellen auf Grund ihrer geringen Mutationsrate interessante Ansatzpunkte zur antiviralen Therapie dar. So wurde für die Assemblierung, Knospung und Reifung diverser Retroviren eine Abhängigkeit gegenüber einem funktionalen Ubiquitin-Proteasom-System in den Zielzellen beschrieben. Diese Effekte wurden bisher allein auf die Proteasom-Inhibitor (PI)-bedingte Depletion des freien zellulären Ubiquitins zurückgeführt. Im ersten Schwerpunkt dieser Arbeit wurde gezeigt, dass ein auf dem chemischen Chaperon Glycerol basierendes Medium, welches Zellen optimierte Bedingungen zur Proteinfaltung bietet, die Prozesse der viralen Freisetzung positiv beeinflusst. Weiterhin war es möglich, den PI-bedingten Freisetzungsdefekt durch die Applikation des Glycerolmediums abzuschwächen. Dies liegt an der Verminderung der durch die Hemmung der Proteasom-Aktivität bedingten Akkumulation fehlgefalteter, polyubiquitinierter Gag-Moleküle, den sogenannten Gag-DRiPs, welche in einer mechanistisch den Prionen ähnlichen Art und Weise die Freisetzung viraler Partikel beeinflussen. Diese in den Replikationsstudien beobachteten Effekte traten in einer derart kurzen Zeitspanne auf, dass sie in starkem Kontrast zu der erst deutlich später detektierbaren Abnahme des freien Ubiquitins stehen. Aus diesen Daten geht hervor, dass die PI-bedingte Hemmung der viralen Freisetzung nicht allein auf einer verringerten zellulären Ubiquitinmenge beruhen kann, sondern vielmehr auch durch Gag-DRiPs bedingt wird. Ein weiterer Schwerpunkt lag in der spezifischen Hemmung des zentralen zellulären Hitzeschockproteins Hsp90 und deren Auswirkung auf die Replikation von HIV-1. Die selektive Hemmung von Faltungsprozessen führt zur Akkumulation polyubiquitinierter Proteine, welche wiederum mit der geordneten Assemblierung von Gag-Molekülen interferieren können. Sowohl in ex vivo kultivierten tonsillärem Gewebe als auch in PBMCs konnte eine deutlich dosisabhängige Hemmung der HIV-1 Replikation durch den Hsp90-Inhibitor 17-AAG nachgewiesen werden. Die hierbei verwendeten Substanzmengen lagen weit unter der maximal tolerierbaren Konzentration. Weiterhin zeigte die Kombination von 17-AAG mit einem PI (PS-341) im niedrigen nanomolaren Bereich synergistische Effekte auf die Hemmung der Replikation von HIV-1, was für einen günstigen therapeutischen Index in vivo spricht. Diese Ergebnisse charakterisieren die selektive Hemmung von Hsp90 durch 17-AAG in Kombination mit Proteasom-Inhibitoren als vielversprechenden therapeutischen Ansatz zur Einflussnahme auf die Replikation von HIV-1. Ein alternativer Mechanismus, wie ein korrekt gefaltetes virales Protein auf die Virusfreisetzung einwirkt, wurde in einem dritten Schwerpunkt aufgezeigt. Das elfte IAV-Protein PB1-F2 wurde auf dessen inhärente Fähigkeit zur Oligomerisierung untersucht. Durch Studien synthetischer Peptide zeigte sich, dass PB1-F2 Oligomere ausbildet, die durch chemische cross linker stabilisiert und nachgewiesen werden konnten. In Übereinstimmung mit in silico generierten Vorhersagen konnten der N- und C-terminale Bereich des Proteins als Oligomerisierungsdomänen charakterisiert werden. Unterstützt durch die Ausprägung von Disulfidbindungen über das Cystein in Position 42, befindet sich die Hauptoligomerisierungsdomäne im C-Terminus von PB1-F2. In Kombination mit dem Vorhandensein einer mitochondrialen Lokalisationssequenz wird deutlich, dass die PB1-F2 vermittelte Apoptose auf der Ausbildung von PB1-F2-Ionenkanälen beruht, was durch eine spätere Publikation unseres Labors bestätigt werden konnte. Folglich könnte PB1-F2, ähnlich wie das M2-Protein der Orthomyxoviridae und das Vpu-Protein der Retroviridae zu den Viroporinen gezählt werden und über Multimerisierung zur Ausbildung von Ionenkanälen in der mitochondrialen Membran zur Pathogenität des IAV beitragen. This study dealt with the influence on viral replication by a systematic modification of protein folding. During millions of years of co-evolution viruses generated a dependency on host cells mechanism. Cellular proteins are interesting starting points for antiviral therapy due to their marginal mutation rate. Therefore a dependency of retroviral assembly, budding, and maturation to a functional ubiquitin-proteasome system of target cells was described. These effects were led back to proteasome inhibitor (PI) caused depletion of free cellular ubiquitin so far. The first emphasis of this work was to demonstrate that a medium based on the chemical chaperone glycerol provides optimized conditions for protein folding, as well as affects the release of viral particles positively. Furthermore the PI-caused defect of virus release was attenuated by application of glycerol substituted medium. This is caused by the decrease of accumulated, misfolded, polyubiquitinated Gag-molecules followed by inhibition of proteasomal activity, so called Gag-DRiPs, which act in a prion-like manner to influence the release of viral particles. In replication studies these effects were detected in such a short period of time that they were in strong contrast to the decrease of free ubiquitin detectable broadly later. These data demonstrate that a PI-caused decrease of virus release is not only caused by a diminished quantity of cellular ubiquitin but through the influence of Gag-DRiPs. A further emphasis was the specific inhibition of the central cellular heat shock protein Hsp90 and the consequences on HIV-1 replication. The selective inhibition of folding processes results in the accumulation of polyubiquitinated proteins which may interfere with the well-regulated assembly of Gag-molecules. As well as in ex vivo cultivated tonsillary tissues as in PBMCs a clear concentration dependency of inhibition of HIV-1 replication was detectable by Hsp90 inhibitor 17-AAG. The applied amounts of the inhibitor were far below maximum tolerated concentration. In addition a combination of 17-AAG with a PI (PS-341) in low nanomolar concentrations demonstrated synergistic effects on inhibition of HIV-1 replication which indicates a beneficial therapeutic index in vivo. These results characterize the selective inhibition of Hsp90 by 17-AAG in combination with proteasome inhibitors as a promising therapeutic approach to influence HIV-1 replication. A third emphasis based on an alternative mechanism which pointed out how a correct folded viral protein is able to influence virus release. The 11th IAV-protein PB1-F2 was characterized concerning its inherent capability to oligomerize. By using synthetic proteins it was shown that PB1-F2 forms detectable oligomers which can be stabilized via a chemical cross linker. In accordance to in silico generated predictions the N- and C-terminus of PB1-F2 were characterized as domains of oligomerization. The main oligomerization domain of PB1-F2 is located in its C-terminus supported by the occurrence of disulfide bonds via the cysteine in position 42. This in combination with the existence of a mitochondrial localization sequence corroborates that the PB1-F2 mediated apoptosis is based on the formation of PB1-F2 ion channels. This fact was published by our laboratory recently. Accordingly to this PB1-F2 may be dedicated to the viroporins, as well as the M2-protein of orthomyxoviridae and the vpu-protein of retroviridae, and support pathogenicity of IAV by its multimerization and formation of ion channels into the mitochondrial membrane.

In dieser Arbeit wurde die Komplexbildung von hGBP1 bei der Hydrolyse von GTP mit Dynamischer Lichtstreuung und Deuteriumaustausch untersucht. Dabei wurden berechnete hydrodynamische Radien aus modellierten Kristallstrukturen mit denen aus den Messungen verglichen, um so die Struktur des Komplexes zu ermitteln. Mit GppNHP (ein nicht hydrolysierbares GTP-Analogon) ergab sich ein über die G-Domänen assoziierender Dimer-Komplex und mit \(GDP/AlF_{x}\) (ein nicht hydrolysierbares GTP-Übergangszustands-Analogon) ein über die Subdomäne \(\alpha\)12/13 gebildeter Tetramer-Komplex. Im zweiten Teil wurden hGBP1 und seine Komplexe an der Oberfläche deuteriert und dann deren Peptide mit Maldi-TOF untersucht. Durch veränderte Deuterierungsgerade, die durch Berechnungen ergänzt wurden, konnten Strukturänderungen nachgewiesen werden. Unter anderem wurde gezeigt, dass bei der Hydrolyse die Subdomäne \(\alpha\)12/13 herausklappt und so das Teramer gebildet wird.

International audience; The Pycnoporus fungi are white-rot basidiomycetes listed as food- and cosmetic-grade microorganisms. Three high redox potential laccases from Pycnoporus coccineus and Pycnoporus sanguineus were tested and compared, with the commercial Suberase® as reference, for their ability to synthesise natural active oligomers from rutin (quercetin-3-rutinoside, one of the best-known naturally occurring flavonoid glycosides). The aim of this work was to develop a process with technical parameters (solvent, temperature, reaction time and raw materials) that were easy to scale up for industrial production and compatible with cosmetic and pharmaceutical formulation guidelines. The aqueous mixture of glycerol/ethanol/buffer described in this study met this requirement and allowed the solubilisation of rutin and its oxidative bioconversion into oligomers. The four flavonoid oligomer mixtures synthesised using laccases as catalysts were analysed by high performance liquid chromatography–diode array detection–negative electrospray ionisation–multistage mass spectrometry. Their chromatographic elution profiles were compared and 16 compounds were characterised and identified as dimers and trimers of rutin. The oligorutins were different in Suberase® and Pycnoporus laccase reaction mixtures. They were evaluated for their antioxidant, anti-inflammatory and anti-ageing activities on specific enzymatic targets such as cyclooxygenase (COX-2) and human matrix metalloproteinase 3 (MMP-3). Expressed in terms of IC50, the flavonoid oligomers displayed a 2.5- to 3-fold higher superoxide scavenging activity than monomeric rutin. Pycnoporus laccase and Suberase® oligorutins led to an inhibition of COX-2 of about 35% and 70%, respectively, while monomeric rutin showed a near-negligible inhibition effect, less than about 10%. The best results on MMP-3 activity were obtained with rutin oligomers from P. sanguineus IMB W006-2 laccase and Suberase® with about 70–75% inhibition

The discovery that class C G protein-coupled receptors (GPCRs) function as obligatory dimeric entities has generated major interest in GPCR oligomerization. Oligomerization now appears to be a common feature among all GPCR classes. However, the functional significance of this process remains unclear because, in vitro, some monomeric GPCRs, such as rhodopsin and β2-adrenergic receptors, activate G proteins. By using wild type and mutant serotonin type 4 receptors (5-HT4Rs) (including a 5-HT4-RASSL) expressed in COS-7 cells as models of class A GPCRs, we show that activation of one protomer in a dimer was sufficient to stimulate G proteins. However, coupling efficiency was 2 times higher when both protomers were activated. Expression of combinations of 5-HT4, in which both protomers were able to bind to agonists but only one could couple to G proteins, suggested that upon agonist occupancy, protomers did not independently couple to G proteins but rather that only one G protein was activated. Coupling of a single heterotrimeric Gs protein to a receptor dimer was further confirmed in vitro, using the purified recombinant WT RASSL 5-HT4R obligatory heterodimer. These results, together with previous findings, demonstrate that, differently from class C GPCR dimers, class A GPCR dimers have pleiotropic activation mechanisms.

International audience; The development of a highly convenient and efficient protocol for iterative solution-phase synthesis of shorter oligomers of para- and meta-arylopeptoids is described. Peptide coupling methods for accessing longer oligomers were studied: use of the new coupling reagent COMU was found to be the most efficient for creation of the tertiary benzamide bonds. The cis/trans isomerism of arylopeptoid backbones was studied by NMR and was found to be highly dependent on the nature of the side chains. Increasing bulkiness of theside chains favored the cis amide bond conformation; arylopeptoids possessing tert-butyl side chains contained exclusively cis amide bonds

Their coordination chemistry was studied and some isolated complexes were used in catalysis (for example in ethylene oligomerization and transfer hydrogenation).; Au cours de ce travail de thèse, la synthèse de nouveaux ligands mixtes combinant des fonctions de propriétés électroniques différentes (phosphores, amines, alcène, amidure, iminophosphorane) a été abordée. Leur chimie de coordination a été étudiée et certains complexes isolés ont été utilisés en catalyse (oligomérisation d'éthylène, hydrogénation par transfert d'hydrure).

The basis of the present study represents the aim in bringing a contribution to the elucidation of certain thermodynamic and molecular aspects characterizing protein-cosolvent and protein-protein interactions and how these two types of interactions are interrelated. The investigation of protein-cosolvent systems was mainly based on ionic species and, in particular, on a special class of organic salts known under the general name of "ionic liquids". By investigating a multitude of ionic liquids, it was possible to rank them in a Hofmeister series, based on the extent of stabilizing/destabilizing effect on the proteins. More, the ionic liquids show the capacity to prevent protein aggregation. The unfolding - dissociation mechanism and the thermodynamic equilibrium between the oligomerization domains of MST1 and Nore1 (the SARAH domains) - having a role in the Ras-induced apoptosis pathway was also investigated.

In this thesis it was succeeded to establish an expression system for BDV-glycoprotein. This system yielded a BDV-glycoprotein, which correlates with the wild type glycoprotein in all characteristics. It is cloven at Amino acid position 246-249 by Furin. The transport to the cell surface and the therewith associated glycosylation conform to the natural glycoprotein. Therefore, it was possible to make the further analysis with the help of a vectorialy expressed glycoprotein, which complies with the glycoprotein of infected cells. As it is fact, that many proteins which are involved in cell fusion are dimmers or trimers, so this was investigated for the BDV-glycoprotein, too. Superior, oligomeric forms of the BDV glycoprotein could be demonstrated, in all probability, dimers and trimers of this protein. By means of this new expression system it was possible to show, that the BDV glycoprotein is able to arrange the cell-cell-fusion on its own after the subsidence of the pH-value. For this reaction it is essential that the BDV-glycoprotein has been cloven before. Compared to the amount in infected cells, the increased fusion activity of BDV-glycoprotein stable cell line could be explained by the higher expression level and the more efficient transport to the cell surface. An inhibition of the fusion of about 50% could be attained by the treatment of the cells with Heparin before lowering the pH-value. Through substitution in the amino acid region 296-305 in the c-terminal subunit of BDV-glycoprotein it could be shown that this region plays an important role during the fusion process. As a conclusion it can be stated, that the fusion peptide is located in this domain. In order to precisely characterise the fusion peptide, further investigations have to be carried out., In dieser Arbeit ist es gelungen, ein Expressionssystem für das BDV-Glykoprotein zu etablieren, welches ein solitäres BDV-Glykoprotein hervorbringt, das vollständig funktionell ist und in allen seinen Eigenschaften denen des Wildtyp-Glykoproteins entspricht. Es wird ebenso wie das Wildtyp-Glykoprotein an Aminosäure-Position 246-249 durch Furin gespalten. Der Transport zur Oberfläche und die damit verbundene Glykosylierung entsprechen der des natürlichen Glykoproteins. Somit konnten nun alle weiteren Untersuchungen an einem vektoriell exprimierten Glykoprotein durchgeführt werden, welches dem aus infizierten Zellen entspricht. Da bekannt ist, dass viele Proteine, die an der Zellfusion beteiligt sind, meist als Dimere oder Trimere vorliegen, wurde hier dieses auch für BDV untersucht. Dargestellt werden konnten höher oligomere Formen des BDV-Glykoproteins, wobei es sich wahrscheinlich um dimere und trimere Formen des BDV-Glykoproteins handelt. Mit Hilfe des Expressionssystems konnte gezeigt werden, dass das BDV- Glykoprotein alleine in der Lage ist, nach einer Erniedrigung des pH-Werts eine Zell-Zell-Fusion zu vermitteln. Essentiell für die Fusion ist, dass das BDV-Glykoprotein gespalten vorliegt. Die erhöhte Fusionsaktivität der stabil Glykoprotein exprimierenden Zelllinie gegenüber infizierten Zellen liegt wahrscheinlich an der erhöhten Expressionsrate und dem effizienteren Transport an die Oberflächen. Eine Inhibition der Fusion um 50% konnte durch eine Behandlung der Zellen mit gelöstem Heparin erreicht werden, bevor der pH-Wert herabgesetzt wurde. Durch Substitutions-Mutationen konnte gezeigt werden, dass der Aminosäurebereich 296-305 in der C-terminalen Untereinheit des BDV-Glykoprotein eine wichtige Rolle bei der Fusion spielt. Es kann daher gesagt werden, dass ein Fusionspeptid in diesem Bereich des BDV-Glykoprotein liegt. Für eine genauere Charakterisierung sind weitere Untersuchungen nötig.

The plasma membrane is composed of several different lipids and can interact directly with the actin cytoskeleton via specific lipids and linker proteins. Among these is the ERM (Ezrin, Radixin, Moesin) family of proteins, which is involved in the direct linkage of the membrane to the actin cytoskeleton via a phosphatidylinositol (4,5) biphosphate (PIP2) lipid binding site. Our aim is to understand the interactions between these proteins and PIP2 using in vitro simplifed biomimetic systems like giant unilamellar vesicles (GUV) containing PIP2. We showed that a conformational change of ezrin occur when the protein binds to PIP2, this conformational change allowing ezrin to bind to actin filaments. We have characterized quantitatively the incorporation of PIP2 in the membrane of giant vesicles, and showed that the interaction of ezrin with GUV induce a partitioning of the lipid within the membrane as well as ezrin aggregates on the membrane. Likewise, ezrin oligomers were observed only in the presence of PIP2. A better understanding of the interplay between ezrin, PIP2-containing membranes and actin will help to get a better view of the role of ezrin in cellulo., La membrane plasmique de la cellule est composée de lipides et interagit notamment avec le squelette de la cellule (le cytosquelette), par l'intermédiaire de protéines d'ancrages et de lipides clefs qui jouent un rôle spécifique dans certains types d'interactions. Parmi les protéines intervenant dans l'ancrage direct de la membrane plasmique au cytosquelette, des protéines de la famille des ERM (Ezrine, Radixine, Moésine) peuvent interagir spécifiquement avec un lipide, le phosphatidylinositol (4,5) biphosphate (PIP2), d'une part et avec l'actine du cytosquelette d'autre part. Dans le but de comprendre les interactions entre membrane plasmique et cytosquelette, nous avons réalisé des expériences in vitro sur des systèmes comportant un nombre minimal de constituants : des vésicules géantes (GUV) contenant du PIP2, de l'ezrine recombinante et de l'actine purifiée. Nous avons mis en évidence que la liaison au PIP2 induit des changements conformationnels de l'ezrine. L'ezrine est alors capable d'interagir avec les filaments d'actine. Nous avons caractérisé quantitativement l'incorporation de PIP2 dans la membrane de vésicule géantes, et montré que l'interaction de l'ezrine avec les vésicule géante contenant du PIP2, induit un partitionnement du lipide dans la bicouche lipidique et conduit à la formation d'agrégats de PIP2 et d'ezrine sur la membrane. La connaissance des effets de l'ezrine sur les membranes contenant du PIP2 et la connaissance des différents mécanismes se produisant lors des interactions permettra de définir plus précisément le rôle de l'ezrine in cellulo.

This work describes the synthesis and the conformational study of new mixed oligomers. In the first chapter, using previous work on the synthesis of N-aminodipeptids, we were obtained mixed oligomers alternating amid and N-aminoamid bond named [[alpha]/[alpha]-N-amino]mers. The oligomerization of N-aminopeptids in liquid phase was achieved through an acid fluorid coupling from a building block with the protections Boc (N-terminus), Bn (C-terminus) and phtaloyl (N-side). The second chapter presents the results obtained by different conformational spectroscopic methods (NMR, IR and DC) and molecular modeling on the various oligomers synthesized in Chapter 1. This work has allowed to highlight a original repetitive folding by a C8 hydrogen bond involving the carbonyl group of phthalimid and a amid proton, Ce travail décrit la synthèse et l'étude conformationnelle de nouveaux oligomères mixtes. Dans le premier chapitre, en exploitant des travaux antérieurs concernant la synthèse des N-aminodipeptides, nous avons obtenu des oligomères mixtes, alternant des liens amides et N-aminoamides nommés [[allpha]/[alpha]-N-amino]mères. L'oligomérisation des N-aminopeptides en phase liquide est réalisable grâce à un couplage au fluorure d'acide à partir d'une unité de base possédant les protections Boc (extrémité N-terminale), Bn (extrémité C-terminale) et phtaloyle (azote latéral). Le deuxième chapitre présente les résultats obtenus par différentes méthodes spectroscopiques (RMN, IR et DC) et modélisation moléculaire sur les différents oligomères synthétisés dans le chapitre 1. Ces travaux ont permis de mettre en évidence un repliement répétitif original par une liaison hydrogène de type C8 impliquant un groupement carbonyle du phtalimide et un proton amidique

Iron-bis(imino)pyridine catalysts are highly active in olefins oligo- or polymerization reactions. IFP notably wishes to study these catalysts to transform ethyleneinto linear olefins with specified chain length. However, the precursor activation step (usually with the MAO cocatalyst) generates an activated species in which the precise nature of the iron metal ion remains largely unknown. Our DFT calculations show that the most reactive activated species likely has an oxidation state +III and quartet spin state. The fact that our DFT calculations in combination with a microkinetic model describe accurately the experimental results of the oligomerization of 1-butene, i.e., the principal formation of linear octenes, furnishes additional support for the retained active species. Moreover, using this molecular representation, a powerful relation has been established between the Schulz-Flory coefficient, which informs of experimental oligo- or polymerization degree for a catalyst, and the difference in the Gibbs free activation energy of a typical insertion and termination reaction. This relation has allowed us to study and better understand the particular behaviour of iron-bis(arylimino)pyridine catalysts with respect to substituents changes. Moreover, its strong universal potential has been shown with the extension of its application domain to other catalysts (other ligands, other metals). Finally, the agostic bonds in bis(arylimino)pyridine complexes have been studied by QTAIM calculations, because this bond type is often a prelude to termination reactions in catalytic cycles.; Les catalyseurs bis(imino)pyridine au fer sont hautement actifs en oligo- ou polymérisation des oléfines. L'IFP souhaite étudier ces catalyseurs pour transformer l'éthylène en oléfines linéaires de tailles maîtrisées. Cependant, l'étape d'activation du précurseur (usuellement avec le cocatalyseur MAO) génère une espèce activée dont la nature reste à ce jour inconnue. Nos calculs DFT montrent que l'espèce activée la plus réactive est dans un état d'oxydation +III et de spin quadruplet. Le fait que nos calculs, en combinaison avec un modèle microcinétique, décrivent précisément les résultats expérimentaux d'oligomérisation du butène-1, c'est-à-dire la formation majoritaire d'octènes linéaires, fournit un support additionnel à la pertinence de l'espèce activée retenue. Par ailleurs, l'utilisation de ce modèle moléculaire a permis d'établir une puissante corrélation entre le coefficient Schulz-Flory, qui informe du degré d'oligo- ou de polymérisation expérimental d'un catalyseur, et la différence des enthalpies libres d'activation d'une réaction d'insertion et de terminaison particulières. Cette relation nous a permis d'étudier et de mieux comprendre le comportement des catalyseurs bis(arylimino)pyridine au fer vis-à- vis des changements de substituants. En outre, son fort potentiel universel est montré grâce à l'extension de son domaine d'application vers d'autres catalyseurs (autres ligands, autres métaux). Enfin, les liaisons agostiques dans les complexes bis(arylimino)pyridine ont été étudiées par des calculs QTAIM parce qu'elles sont souvent un prélude aux réactions de terminaison dans les cycles catalytiques.

Les récepteurs couplés aux protéines G (RCPGs) constituent la plus grande classe de récepteurs membranaires impliqués dans la transmission des signaux extracellulaires. Traditionnellement, la transmission de la signalisation par les RCPGs implique l’activation d’une protéine G hétéro-trimérique qui pourra à son tour moduler l’activité de divers effecteurs intracellulaires. Ce schéma classique de signalisation s’est complexifié au fils des années et l’on sait maintenant qu’en plus d’interagir avec les protéines G, les RCPGs s’associent avec une panoplie d’autres protéines afin de transmettre adéquatement les signaux extracellulaires. En particulier, la découverte d’une famille de protéines transmembranaires modulant la fonction des RCPGs, baptisées protéines modifiant l’activité des récepteurs (« receptor activity-modifying proteins » ; RAMPs), a changé la façon de concevoir la signalisation par certains RCPGs. Dans le cas du récepteur similaire au récepteur de la calcitonine (« calcitonin-like receptor » ; CLR), l’association avec les RAMPs permet l’acheminement à la surface cellulaire du récepteur tout en modulant ses propriétés pharmacologiques. Lorsqu’il est associé avec RAMP1, le CLR fonctionne comme un récepteur du peptide relié au gène de la calcitonine (« calcitonin gene-related peptide » ; CGRP), alors qu’il devient un récepteur de l’adrénomedulline lorsqu’il interagit avec RAMP2 ou RAMP3. D’autre part, en plus d’interagir avec des protéines accessoires transmembranaires telles les RAMPs, les RCPGs peuvent aussi s’associer entre eux pour former des oligomères de récepteurs. Dans cette thèse, nous nous sommes penchés sur les interactions entre les RCPGs et les RAMPs, et plus particulièrement sur l’interrelation entre ce type d’association RCPG/RAMP et l’assemblage en oligomères de récepteurs, en utilisant le récepteur du CGRP comme modèle d’étude. Une première étude nous a tout d’abord permis de confirmer l’interaction entre le récepteur CLR et RAMP1, dans un contexte de cellules vivantes. Nous avons démontré que ce complexe CLR/RAMP1 active la protéine G et recrute la protéine de signalisation -arrestine suite à une stimulation par le CGRP. Ensuite, nous avons déterminé que même s’il doit obligatoirement former un hétéro-oligomère avec les RAMPs pour être actif, le CLR conserve malgré tout sa capacité à interagir avec d’autres RCPGs. En plus d’observer la présence d’homo-oligomère de CLR, nous avons constaté que tout comme les RCPGs, les RAMPs peuvent eux-aussi s’associer entre eux pour former des complexes oligomériques pouvant comprendre différents sous-types (RAMP1/RAMP2 et RAMP1/RAMP3). Cette observation de la présence d’homo-oligomères de CLR et de RAMP1, nous a amené à nous questionner sur la stœchiométrie d’interaction du complexe CLR/RAMP1. Dans une deuxième étude ayant pour but d’établir la composition moléculaire du récepteur CGRP1 in vivo, nous avons développé une nouvelle approche permettant l’étude de l’interaction entre trois protéines dans un contexte de cellules vivantes. Cette technique baptisée BRET/BiFC, est basée sur le transfert d’énergie de résonance de bioluminescence entre un donneur luminescent, la Renilla luciférase, et un accepteur fluorescent, la protéine fluorescente jaune (YFP), reconstituée suite au ré-assemblage de ces deux fragments. En utilisant cette approche, nous avons pu déterminer que le récepteur CGRP1 est constitué d’un homo-oligomère de CLR interagissant avec un monomère de RAMP1. En démontrant un assemblage oligomérique asymétrique pour le récepteur CGRP1 à partir d’une nouvelle approche biophysique, nous croyons que les travaux présentés dans cette thèse ont contribué à élargir nos connaissances sur le fonctionnement de la grande famille des RCPGs, et seront utile à la poursuite des recherches sur les complexes protéiques impliqués dans la signalisation., G protein coupled receptors (GPCRs) constitute the largest family of membrane receptors involved in signal transduction. Traditionally, signal transduction by GPCRs involves the activation of a hetero-trimeric G protein which will then modulate the activity of several intracellular effectors. We can now appreciate the fact that in addition to their interaction with G proteins, GPCRs also associate with several other proteins, in order to allow proper signal transduction. In particular, the discovery of a family of proteins called receptor activity-modifying proteins (RAMPs) has challenged the traditional views of signal transduction by some GPCRs. In the case of the calcitonin-like receptor (CLR), the association with RAMPs allows the proper cell surface targeting of the receptor in addition to modulate it’s pharmacological properties. Co-expression of CLR with RAMP1 leads to a calcitonin gene-related peptide (CGRP) receptor, whereas CLR association with RAMP2 or RAMP3 promotes the formation of an adrenomedullin receptor. In addition to their interaction with transmembrane accessory proteins such as RAMPs, GPCRs can also interact with other receptors to form receptors oligomers. In this thesis, we were interested in the interactions between GPCRs and RAMPs, and particularly, in the link between these GPCR/RAMP interactions and the assembly of receptor oligomers, using CGRP1 receptor as a model. We first confirmed the interaction between CLR and RAMP1 in living cells. We showed that this CLR/RAMP1 complex activates G proteins and recruits the signalling protein -arrestin upon CGRP stimulation. Next, we demonstrated that even if the CLR requires hetero-oligomeric assembly with RAMPs in order to be active, this receptor can still interact with other GPCRs. In addition to CLR homo-oligomers, we observed that RAMPs can also self-associate to form oligomeric complexes which can involve different subtypes (RAMP1/RAMP2 and RAMP1/RAMP3). This observation of the presence of CLR and RAMP1 homo-oligomers raised the question of the stoiechiometry of interaction of the CLR/RAMP1 complex. In order to establish the molecular composition of the CGRP1 receptor in vivo, we developed a novel approach allowing the detection of the interaction between three proteins in living cells. This method called BRET/BiFC is based on the bioluminescence resonance energy transfer between a luminescent energy donor, Renilla luciferase, and a fluorescent energy acceptor, the yellow fluorescent protein (YFP), reconstituted after the re-association of its two fragments. Using this approach, we showed that the CGRP1 receptor consist of a homo-oligomer of CLR interacting with a monomer of RAMP1. By demonstrating the asymmetrical organization of the CGRP1 receptor complex using a novel biophysical approach, we believe that the results presented herein have contributed to increase our knowledge of the mechanisms of function of the large family of GPCRs and will be useful for the pursuit of research on protein complexes involved in signalling pathways.

Butadiene as a bulk chemical achieves through its polymers a high industrial importance. This work describes the upscaling of an oligomerisation of 1,3-butadiene to mid size oligobutadienes from lab scale to pilot scale. A pilot plant is planned, constructed and operated based on data recieved by thermodynamic calculations and reaction calorimetrie. The influence of different reaction parameters and the workup procedure on product quality is examined after applying different product workups. Wihtin an addiational reaction step it is shown how to functionalize the previously synthesized oligobiatdienes in carbon dioxide as reaction media by hydroformylation to the corresponding oligoaldehydes. Afterwards the homogeneous catalyst system is extracted by carbon dioxide from the product. In the third part of the work a system is introduced which allows the selective hydroformylation of certain olefines out of olefin mixtures with different chain lenghts. The problem is solved by using a two phase system, where the catalyst is immobilized in a carbon dioxide phase and the liquid product are converted into the aldehydes dependent on the pressure of carbon dioxide.

Von TRAIL, FasL und APRIL, drei Mitgliedern der TNF-Liganden-Familie, ist bekannt, dass Trimerstabilität und Oligomerisierungsstatus maßgeblich das Rezeptoraktivierungspotential dieser Liganden beeinflussen. Für die immunstimulatorischen TNF-Liganden CD27L, CD40L, OX40L, 41BBL und GITRL war hingegen vor der Durchführung dieser Arbeit praktisch nicht bekannt, inwieweit Trimerbildung, Stabilisierung und Oligomerisierung wichtig für deren Aktitvität sind. Dies wurde in dieser Arbeit systematisch untersucht. CD40L besaß bereits als trimeres Molekül eine hohe Aktivität, die durch sekundäre Oligomerisierung nur wenig gesteigert wurde. Die spezifische Aktivität konnte durch Stabilisierung mit Hilfe der Tenascin-C (TNC)-Trimerisierungsdomäne nur geringfügig gesteigert werden. CD27L war als lösliches Flag-markiertes sowie als hexameres Fc-Protein selbst nach Quervernetzen nicht in der Lage, seinen Rezeptor CD27 zu binden und zu aktivieren. Die TNC-stabilisierte trimere Form des CD27L hingegen induzierte nach Oligomerisierung mit einem anti-Flag-Antikörper ein starkes Signal. Trimerer OX40L und trimerer 41BBL konnten nur in oligomerisierter Form ihre Rezeptoren aktivieren, wobei die Aktivität der TNC-stabilisierten Form signifikant stärker ausgeprägt war. GITRL aktivierte seinen Rezeptor bereits als stabilisiertes Trimer und Hexamer, die Aktivität konnte durch Quervernetzen nur gering gesteigert werden. Zusammenfassend kann man sagen, dass CD27L, OX40L und 41BBL zu der Untergruppe der TNF-Ligandenfamilie gehört, für die eine Stabilisierung des trimeren Moleküls und dessen Oligomerisierung nötig sind, um eine starke Rezeptoraktivierung zu ermöglichen. Im Gegensatz dazu zeigten CD40L und GITRL bereits oligomerisierungsunabhängig eine hohe Aktivität. GITRL benötigte allerdings die Stabilisierung des trimeren Moleküls durch die TNC-Domäne, um gute Aktivität zu zeigen. Im Weiteren wurden Antikörperfragment (scFv-)-TNF-Ligand-Fusionsproteine konstruiert und untersucht, die ein Zelloberflächenantigen binden. Eine starke Zelloberflächenantigen-spezifische Aktivierung des jeweiligen Rezeptors konnte für scFv-41BBL und für scFv-OX40L gezeigt werden, wohingegen scFv-CD40L und scFv-GITRL bereits auf antigennegativen Zellen stark aktiv waren. scFv-CD27L war selbst auf antigenpositiven Zellen inaktiv. Verwendet man an Stelle des Antikörperfragments eine extrazelluläre Proteinbindedomäne, z.B. die eines TNF-Rezeptors, erhält man Fusionsproteine, die zum einen eine selektive Aktivierung der TNF-Ligandendomäne und somit die Aktivierung des korrespondierenden Rezeptors auf der Zielzelle ermöglichen, zum anderen aber durch die Bindung an den membranständigen Liganden dessen Aktitvät neutralisieren können. Für CD40-, RANK- und B7-2-FasL konnte der immobilisationabhängige Aktivierungseffekt auf entsprechenden Zelloberflächenmolekül-exprimierenden Zellen gezeigt werden. Anhand von T47D-Zellen, die durch eine autokrine CD40L-CD40-Signalschleife vor Apoptose geschützt sind, konnte gezeigt werden, dass durch die Bindung von CD40-FasL an membranständigen CD40L die CD40L-CD40-Interaktion gestört und gleichzeitig Apoptose verstärkt induziert werden kann. Das Prinzip der antigenabhängigen Aktivierung von TNF-Liganden könnte Anwendung in der Tumortherapie finden, da bei Verwendung entsprechender selektiv exprimierter Marker eine lokale Rezeptoraktivierung erreicht und so Nebenwirkungen minimiert werden können., Trimer stability and oligomerization status of TRAIL, FasL and APRIL, three members of the TNF ligand family, critically determine their receptor activating potential. However, detailed information for the immunostimmulatory ligands CD27L, CD40L, OX40L, 41BBL and GITRL regarding the importance of trimer formation, stabilization and oligomerization for ligand activity was lacking. These aspects were investigated systematically in this work. CD40L was highly active as a trimeric molecule. Secondary oligomerization and/or stabilization via the tenascin-C (TNC) trimerization domain slightly enhanced its specific activity. As soluble Flag-tagged and as hexameric Fc protein CD27L failed to bind and activate its cognate receptor CD27, even after crosslinking. However, the TNC stabilized form of CD27L induced a strong signal after oligomerization with anti-Flag antibody. Receptor signaling was only activated by oligomerized molecules of trimeric OX40L and 41BBL whereas the respective TNC fusion protein showed significant stronger activity. Stabilized GITRL trimers and hexamers already activated their receptor whereas oligomerization of GITRL just slightly enhanced the specific activity. Taken together, CD27L, OX40L and 41BBL belong to a TNF ligand family subgroup which requires oligomerization and stabilization of the trimeric molecule to ensure strong receptor activation. In contrast, CD40L and GITRL already display high oligomerization-independent activity, though the latter needs stabilization by the TNC domain. Furthermore, antibody fragment (scFv)-ligand fusion proteins targeting specific cell surface antigens were designed and analyzed. Strong cell surface antigen-selective TNF receptor activation was achieved for scFv-41BBL and scFv-OX40L whereas scFv-CD40L and scFv-GITRL already induced signaling in the absence of antigen-positive cells. scFv-CD27L lacked activity even on antigen-positive cells. Using an extracellular protein binding domain for example the ligand binding domain of a TNF receptor instead of an antibody fragment resulted in fusion proteins that on the one hand activate the TNF ligand domain and thus the corresponding receptor on target cells and on the other hand neutralize membrane ligand activity by binding. The effect of cell surface immobilization-mediated activation of these fusion proteins on cells expressing the corresponding target molecule was shown here for CD40-, RANK- and B7-2-FasL. The CD40-FasL fusion protein simultaneously blocked CD40L-CD40 interaction and induced strong apoptosis in T47D cells displaying an antiapoptotic autocrine CD40L-CD40 signaling loop. The principle of antigen-dependent activation of TNF ligands could be of use in tumor treatment due to the fact that tumor specific marker targeting leads to locally restricted receptor activation on antigen positive cells, promising a reduction in potential off target effects.

International audience; To understand the mechanism involved in the apical sorting of glycosylphosphatidylinositol (GPI)-anchored proteins (GPI-APs) we fused to the C-terminus of GFP the GPI-anchor-attachment signal of the folate receptor (FR) or of the prion protein (PrP), two native GPI-anchored proteins that are sorted apically or basolaterally, respectively, in MDCK cells. We investigated the behaviour of the resulting fusion proteins GFP-FR and GFP-PrP by analysing three parameters: their association with DRMs, their oligomerisation and their apical sorting. Strikingly, we found that different GPI-attachment signals differently modulate the ability of the resulting GFP-fusion protein to oligomerise and to be apically sorted. This is probably owing to differences in the GPI anchor and/or in the surrounding lipid microenvironment. Accordingly, we show that addition of cholesterol to the cells is necessary and sufficient to drive the oligomerisation and consequent apical sorting of GFP-PrP, which under control conditions does not oligomerise and is basolaterally sorted.

Der molekulare Schalter Ras ist zusammen mit Nore1 und der MST1-Kinase in der Lage apoptische Wirkung zu vermitteln. In dieser Arbeit wurde die Wirkung der Assemblierung von MST1 und Nore1 auf den Ras-vermittelten Zelltod mit Hilfe biophysikalischer Methoden charakterisiert. Mit Hilfe von analytischer Gelfiltration, chemischer Vernetzung und dynamischen Lichtstreuung wurde der Grad der Oligomerizierung bestimmt. Durch zeitlich aufgelöste Fluoreszenzspektroskopie konnte die Dynamik der Homo- und Hetero- Oligomerisierung charakterisiert werden und in ein Modell der Signalkaskade integriert werden. Desweiteren wurde die Wirkung der Interaktion mit Nore1 auf die Aktivität des MST1 untersucht. Dabei wurde Nore1 als ein neues Substrat des MST1 definiert. Durch Interaktion von Nore1 mit Ras wird die Phosphorylierung inhibiert. Nore1 kann dementsprechend entweder als ein Adaptorprotein für MST1 oder als Regulator des Zellzyklus dienen.
